The present study was performed to assess the HPLC‐DAD analysis as well as antioxidant and protective effects of Tunisian Rhanterium suaveolens (Rs) against acetamiprid (ACT) induced oxidative stress on mice erythrocytes. The in vitro assays showed that the methanolic extract of Rs has an impressive antioxidant effect proved by testing the total antioxidant and scavenging activities using BCB, DPPH and ABTS assays, respectively. Moreover, qualitative and quantitative analysis using HPLC‐DAD revealed the richness of Rs in polyphenols where p‐Coumaric, Apigenin‐7‐glucoside and Ferulic acid were detected as the most abundant polyphenols. In the in vivo experiment, ACT, used as a toxicity model, was given to mice at a dose of 20 mg/kg. The latter was the origin of hemolytic anemia characterized by a significant decrease in red blood cells, hemoglobin and hematocrit levels and an increase in bilirubin, LDH, osmotic fragility, reticulocytes and white blood cells number. Characteristic erythrocyte morphological alterations were also determined as spherocytosis, schistocytosis and dacryocystitis. The oxidative status of ACT‐treated mice was also altered manifested by a significant increase in MDA and GSH levels and a decrease in SOD, CAT and GPx activities. When receiving the Rs methanolic extract at a dose of 300 mg/kg, all the parameters cited above were restored in mice. These remarkable corrections could only confirm the important antioxidant effect and the noticeable protective properties that possess Rs owing to its broad range of secondary bioactive metabolites. 相似文献

The purpose of this study was to evaluate the ability of indolinic and quinolinic nitroxide radicals to protect trout (Salmo irideus) erythrocytes against oxidative stress. By using laurdan as a fluorescence probe, it was observed that the nitroxides inhibited the shift towards a gel phase of liposomes prepared with phospholipids extracted from trout erythrocyte membranes prior to the hemolytic event. In addition, the presence of 100 μM nitroxides in these liposomes protected the latter against lipid peroxidation determined by monitoring conjugated diene formation. However, the short chain analogue of the indolinic nitroxide and the quinolinic nitroxide had a negative effect on trout hemolysis, contrary to what has already been observed in previous studies on human RBCs (red blood cells). The half-time (t1/2) of the hemolytic process was 174 ± 4.02 min for the former and 184 ± 4.30 min for the latter compared to the control, 283 ± 5.05 min. Furthermore, the nitroxides remarkably increased the autoxidation rate of both trout and human hemoglobin to met-Hb. Even though protection at the membrane level is conferred by the nitroxides during the early stages of lipid peroxidation, their antioxidative ability might be overwhelmed at a later stage by other mechanisms such as the increased autoxidation of hemoglobin in the presence of the nitroxides, thus giving a possible explanation for the early induction of hemolysis induced by the nitroxides. The iron content and the ratio of bivalent to total iron in the labile acid-soluble fraction of iron were studied in sublittoral sands of Vostok Bay, Sea of Japan. Iron oxidation and reduction rates in sand sediment were measured in the laboratory. Trivalent iron almost always prevailed in the acid-extractable fraction in the natural environment. The most oxidized state of iron occurred in spring and was associated with a low water temperature and a high seawater oxygen content; the most reduced iron was found in fall during periods of low hydrodynamics and low oxygen concentration. In sand, iron is mainly reduced by bacteria; this process is slow and can be inhibited by adding chloroform. Oxidation of iron is mainly a chemical process and cannot be stopped by chloroform. In sand, the content of redox equivalents such as trivalent iron is much greater than in dissolved oxygen of pore water. It is assumed that labile iron in sands acts as a redox buffer, is oxidized by dissolved pore water oxygen at the periods of advective mixing, and is slowly reduced by benthic bacteria during anaerobic conditions. 相似文献

超氧化物歧化酶(E、C、1.15.1.1)是催化超氧阴离子起歧化反应的金属酶类,血红细胞中的SOD属Cu Zn—SOD。本文详细报道了从鸭血分离纯化SOD的改进方法(同时用猪血作对比研究)。设计了热变性、(NH_4)_2SO_4分级监析、低浓乙醇—氯仿短时去除残留血红蛋白、凝胶层析四步纯化方案,获得高产率电泳纯SOD。用紫外扫描,PAGE电泳后考马斯亮蓝和SOD活性杂色,SDS—电泳,HPLC分离和各电泳带组分的N—端测定等技术检测纯度,并进行性质研究。证明所得SOD是均一的;N—端为Ala;分子量和亚分子量各为32,359和16,500dt;并与猪血SOD对比研究其某些性质;对HPLC出现的多个活力峰及PAGE电泳显现的多条活性带进行了讨论。 相似文献

Female Porton rats have been treated with either parenteral iron (intraperitoneal red cells) or dietary iron (carbonyl iron) for up to 12 months or 22 months respectively. In the parenteral iron loaded animals, the liver iron concentration rose from approximately 2 mg g-1 dry wt at 2 months to 21 mg g-1 dry wt at 12 months, while for the dietary iron loaded animals, this value rose from 14 to 48 mg g-1 dry wt at 12 months to over 60 mg g-1 dry wt after 22 months. In contrast, splenic iron concentrations rose more in the parenterally loaded animals (up to 66 mg g-1 dry wt after 12 months) than in the dietary loaded animals (approx. 34 mg g-1 dry wt after 24 months). This study yielded hepatic iron concentrations comparable to those seen in human thalassaemia patients with comparative low hepatotoxicity. Splenic iron concentrations in the parenteral iron loaded group generally exceeded those reported in thalassaemia. Iron concentrations derived from computer assisted morphometry of liver iron deposits correlated well ( r = 0.88, p < 0.001) with chemical analysis data. The fraction of iron in the non-parenchymal cells correlated positively with the duration of iron loading (r = 0.86, p < 0.001). 相似文献

Super oxide is produced during the authorization of hemoglobin. Authorization of hemoglobin is, however, facilitated under hypoxic conditions where hemoglobin is only partially oxygenated.We have recently found that the erythrocyte superoxide dismutase does not fully react with the additional superoxide produced under hypoxic conditions. A leakage of superoxide from the erythrocyte is thus detected, resulting in a potential source for oxyradical damage to tissues.Detailed studies on intact erythrocytes as a function of oxygen pressure have now been performed. These studies further delineate the hypoxic stress on erythrocytes and the mechanism for the leakage of superoxide. By centrifugation of samples under various oxygen pressures it was possible to show an enhanced rate of lysis at reduced oxygen pressures with a maximum rate in the region of 25 mm Hg. At much lower pressures where the hemoglobin is mostly deoxygenated the rate of lysis was dramatically decreased with almost no lysis detected even after three days. Lysis is shown to be associated with superoxide membrane damage. The formation of superoxide which does not react with endogenous SOD reaches a maximum value at much lower pressures where most of the hemoglobin is deoxygenated. It is suggested that the leakage at low pressure is associated with the formation of superoxide by oxidation of hemoglobin associated with the membrane. 相似文献

为研究大鼠红细胞对葡萄糖利用的异头物选择性及其作用机制,应用大鼠红细胞,对葡萄糖的两种异头物作了异构化速率、乳酸生成量、内流速度和大鼠红细胞已糖激酶作用下的磷酸化速度等进行了测定.结果指出,37℃时大鼠红细胞的D-葡萄糖β-异头物和α-异头物代谢成乳酸的速度分别是0.27μmol/gHb(3min)和0.21μmol/gHb(3min),即前者快于后者30%.同时β-D-葡萄糖向红细胞内转运速度也快于后者:分别是5.0和3.5μmol/gHb(3min).大鼠红细胞已糖激酶的葡萄糖磷酸化速率实验结果指出:β-异头物比α-异头物快30%;对于该两种异头物已糖激酶的Km值均为53μmol/L.红细胞与α-和β-D-葡萄糖保温1min后,其葡萄糖浓度均达到1mmol/L左右,说明至少在1min内对于已糖激酶的磷酸化此两种异头物的葡萄糖浓度均已饱和.这些结果提示,大鼠红细胞葡萄糖利用的β-异头物优选性主要与其磷酸化速度有关,而与其转运速度关系不大. 相似文献

process. AD in particular is associated with accumulation of numerous markers of oxidative stress; moreover, oxidative stress
has been shown to precede hallmark neuropathological lesions early in the disease process, and such lesions, once present,
further accumulate iron, among other markers of oxidative stress. In this review, we discuss the role of iron in the progression
of AD.

动态观察烧伤兔血红细胞内游离氨基酸的代谢变化。以日立 835 - 5 0型氨基酸自动分析仪测定 30 %TBSA兔烧伤后 1、3、7、15和 2 0d血红细胞内游离氨基酸的含量。结果显示 ,烧伤后多数测试点红细胞内游离氨基酸不同程度降低 ;伤后 1天降低最显著 ;苏、丙、缬、组、精和脯氨酸伤后各个时间点降低 ,多数时间点显著降低 (P <0 .0 5 - 0 .0 1)。结果提示烧伤后红细胞内游离氨基酸依靠易化扩散参与全身氨基酸的再分布 相似文献

采用联合亲和层析法从人小脑及红细胞膜中纯化了AChE,纯化的人脑及红细胞AChE在SDS-PAGE上呈一主带,分子量约为66000。人脑AChE制备酯酶与酰胺酶比活性分别为1299与143U/mg,人红细胞AChE制备分别为4584与747U/mg。人脑及红细胞AChE制备的酯酶与酰胺酶活性最适pH较接近,在pH7.5-8.0之间,酯酶活性底物ATCh对其芳基酰胺酶活性有抑制作用。IC_(50)分别为10.2×10~(-3)及3×10~(-3)mol/L。梭曼对其酯酶及酰胺酶活性均有明显抑制作用,说明二者均需活性中心丝氨酸参与。 相似文献

机械应力在心血管系统的正常生理和病理中都起着重要的作用。实验中观察到当提高红细胞悬浮液的旋转速度时会导致葡萄糖跨膜输入的速率增加。改变溶液渗透压及用使细胞膜曲率变化的药物(氯丙嗪)是对红细胞作用的另二种力学因素。研究发现它们同样也能对葡萄糖和阴离子的运输有影响,根据运输速率的温度特性给出了这些力学因素作用下葡萄糖、阴离子运输时活化能的变化,活化能的减小和运输速率的增加有很好的对应关系;活化能减小使膜上运输蛋白在运输过程中的构象变化更为容易,对红细胞血影的内禀荧光淬灭测量量表明,机械应力是通过影响膜上葡萄糖运输蛋白(GLUT1)和阴离子交换蛋白(带3蛋白)物构象起作用的,当用抑制剂抑制了阴离子的运输后。观察到此时葡萄糖跨膜运输对机械应力的响应发生改变,这再次表明在红细胞膜上GLUT1和带3蛋白之间存在着信号连接。 相似文献

动态测定烧伤患者血浆及红细胞内游离氨基酸的含量 ,探讨输入外源性氨基酸后对血及红细胞内游离氨基酸的影响。以日立 835— 5 0型氨基酸自动分析仪测定烧伤患者血浆及红细胞内游离氨基酸含量。结果发现烧伤患者血浆总游离氨基酸浓度从伤后到 2 1天均显著降低 (P <0 .0 5~ 0 .0 1) ;赖、苯丙和苯丙 酪氨酸比值显著升高 (P <0 .0 5~ 0 .0 1) ;色、组、精、丙、甘、苏、脯和丝氨酸比值显著降低 (P <0 .0 5~ 0 .0 1) ;缬、亮、异亮、酪、胱和支链氨基酸伤后早期降低。烧伤患者红细胞内总游离氨基酸含量不同程度降低 ,其中 1、3、7天降低显著 (P <0 .0 5~ 0 .0 1) ;红细胞内苯丙和苯丙 酪氨酸比值未见显著性升高 ;色、蛋、精、脯氨酸含量很低或基本未测出。输注复合氨基酸注射液后未能显著改善患者血及红细胞内游离氨基酸含量。结果提示烧伤患者红细胞内游离氨基酸含量的变化趋势与血浆游离氨基酸变化趋势基本一致 ;烧伤后红细胞内苯丙氨酸及苯丙 酪氨酸比值有别于血浆变化。本研究条件下补充外源性氨基酸未能显著改变烧伤患者血浆及红细胞内游离氨基酸的含量 相似文献

Synopsis. Trypanosoma congolense Broden, an intravascular parasite, binds to vessel walls and erythrocytes of infected hosts. In an attempt to characterize T. congolense adhesion to host cells, an in vitro assay was devised. It was shown in the in vitro experiments that T. congolense binds to bovine, sheep, and goat erythrocytes, but not always to erythrocytes of rats, mice, rabbits, horses or humans. Only the anterior part of live trypanosomes adheres to erythrocytes, and the attachment site on the trypanosomes is destroyed by trypsin and chymotrypsin. Trypanosomes did not adhere to bovine erythrocytes that had been incubated with neuraminidase, sodium periodate and poly-L-lysine. The foregoing experiments suggest that the surface of T. congolense contains a protein-associated site which binds to sialic acid of some host cells. This surface site is most likely responsible for attachment to blood vessels in vivo. 相似文献

Mouse carrier erythrocytes containing 125I-interleukin 3 have been prepared and treated with band 3 crosslinking reagents. The incorporation of interleukin 3 by hypotonic treatment into mouse erythrocytes reached levels of about 15% of the interleukin 3 added to the medium being predominantly present in the cytosolic fraction (73%). Uptake fell to about 7.4% when using the same conditions but omitting hypotonic shock. The interaction of band 3 crosslinked interleukin 3 loaded erythrocytes with macrophages was also studied. A high level of incorporation of interleukin 3 into macrophages was observed either from band 3 crosslinked, interleukin 3-loaded erythrocytes or from interleukin 3 loaded erythrocytes. The observations encourage the view that the system may be able to deliver and target cytokines and other growth factors to macrophages. 相似文献

Cigarette smoking is an adverse prognostic factor for health. Its damaging effects on many enzymatic and cellular activities are well known. The present study was carried out to evaluate whether there is a difference in the binding of bilirubin to the erythrocytes from healthy smokers and non-smokers. Oxyhaemoglobins from erythrocytes of different animals including fish, amphibians, reptiles, birds, mammals and human beings
have been isolated by ion-exchange chromatography over phosphocellulose and the comparative rates of autoxidation of oxyhaemoglobin
studied. The mechanism of autoxidationin vitro has been elucidated using toad as well as human oxyhaemoglobin. Autoxidation is markedly inhibited by carbon monoxide as
well as by anion ligands, namely, potassium cyanide, sodium azide and potassium thiocyanate. The inhibition by anions is in
the same order as their strength as nucleophiles, indicating that it is the oxyhaemoglobin and not the ligand-bound deoxy
species which undergoes autoxidation. The structure of oxyhaemoglobin is considered to be mainly

Several acute brain pathological conditions are characterized by the presence of excess glutamate in brain interstitial fluid. We have previously shown that decreasing blood glutamate levels increases the driving force for an enhanced brain-to-blood efflux of glutamate. The present study investigated the glutamate pumping ability of glutamate-depleted erythrocytes both in vitro and in vivo to determine whether the latter could potentially be used in a blood exchange procedure for neuroprotection. We have observed that glutamate is taken up in red blood cells only via a passive diffusive process with a diffusion constant of 0.144/h. When glutamate-depleted blood cells resuspended in 6% hetastarch were injected into recipient rats, using a blood exchange protocol, a decrease of blood glutamate was observed but attributed to plasma dilution. These observations are discussed in light of a novel neuroprotective strategy based on blood glutamate scavenging. 相似文献

用低渗透析 -等渗重封的方法制备了包埋超氧化物歧化酶 ( SOD)的大鼠载体红细胞 ,并从细胞水平上研究了透析条件对大鼠红细胞包埋 SOD的影响与载体红细胞的部分性质 .流式细胞计( FCM)研究表明 ,随透析时间延长和透析液渗透压降低 ,包埋 SOD的载体红细胞百分率升高 ,但载体细胞平均包埋 SOD的量无明显变化 ;SOD浓度对载体细胞百分率无明显影响 ,但与载体细胞平均包埋 SOD的量成线性关系 ;载体红细胞前向角散射 ( FLS)明显下降 ,但显微镜下观察到的载体细胞的大小无明显变化 ,当载体细胞反注射到大鼠体内后 FLS能迅速恢复 ;载体红细胞密度下降 ,其原因是低渗透析时红细胞膨胀未能完全恢复 ;载体红细胞未暴露与自身 Ig G结合的抗原位点 .激光扫描共聚焦显微镜 ( LSCM)分析表明 ,SOD在细胞内呈从细胞中心到细胞膜浓度逐步下降的辐射分布特征 . 相似文献
