A new differentiation screening system employing a human neuroblastoma cell line. NB-1, was used to isolate staurosporine as an inducer obtained from the culture broth of Streptomyces actuosus. Staurosporine at a concentration of 20 nm induced elongation of neurites and cell enlargement one hour after treatment of NB-1. In addition, the agent had a cytotoxic effect against NB-1 at a concentration of more than 0.21 μm.  相似文献

Phaseic acid (PA) and dihydrophaseic acid (DPA) are the major metabolites observed when (S)-2-14C-abscisic acid (ABA) is fed to 14-day excised primary bean leaves (Phaseolus vulgaris L. cv. Red Kidney). The distribution of 14C in leaves which were wilted after feeding ABA appears to be the same as that observed in unwilted leaves. A reduction in the relative specific radioactivities of the two metabolites after wilting, compared with the specific radioactivities measured in unwilted plants, indicated that these metabolites continue to be formed endogenously after wilting. Estimates of the endogenous ABA levels showed that they rose from 0.04 μg to approximately 0.5 μg/g fresh weight within 4 hours after the beginning of a 10% wilt and remained at that level during a subsequent 20 hours of wilt. In unwilted leaves, the levels of PA and DPA were 5 times and 20 times higher than that of ABA, respectively. Both PA and DPA levels rose throughout the wilt period. PA rose from 0.20 μg to 1.0 μg and DPA from 0.8 μg to over 3 μg/g fresh weight. From these data, we calculated the rate of ABA synthesis to be at least 0.15 μg/hr.g fresh weight during this period. We have interpreted these results to mean that in wilted leaves an elevated level of ABA is maintained because the rate of synthesis and metabolism are both elevated and approximately equal.  相似文献

The metabolism and transport of (±)-l2-14Clabscisic acidand its metabolites was investigated during the period of graindevelopment in wheat. Forty-five hours after feeding the hormoneinto the flag leaf blade, or after injection into the grains,nine metabolites could be extracted with acetone. Four of thesecompounds have been identified. They are phaseic acid, dihydrophaseicacid, abscisyl-ß-D-glucopyranoside and the polar metabolite.As well as the acetone-soluble metabolites a number of othershave been found which are insoluble in acetone. These appearto be conjugated to lipids, ‘gluten-like’ proteins,and carbohydrates. ABA and its metabolites were transported to all parts of theplant above the flag leaf node when the radioactive hormonewas introduced into the blade. However, when it was injectedinto the grain the radioactivity remained there until the pointwhen dry matter accumulation ceased, after which it was foundonly in the peduncle. The results suggest that the increasing level of ABA duringthe period of dry matter accumulation is due to biosynthesiswithin the grain and that the decrease after dry matter accumulationceases is due to both metabolism and redistribution within theplant.  相似文献

The facultative halophyte, Mesembryanthemum crystallinum, shifts its mode of carbon assimilation from the C3 pathway to Crassulacean acid metabolism (CAM) in response to water stress. In this study, exogenously applied abscisic acid (ABA), at micromolar concentrations, could partially substitute for water stress in induction of CAM in this species. ABA at concentrations of 5 to 10 micromolar, when applied to leaves or to the roots in hydroponic culture or in soil, induced the expression of CAM within days (as indicated by the nocturnal accumulation of total titratable acidity and malate). After applying ABA there was also an increase in phosphoenolpyruvate carboxylase and NADP-malic enzyme activities. The degree and time course of induction by ABA were comparable to those induced by salt and water stress. Electrophoretic analyses of leaf soluble protein indicate that the increases in phosphoenolpyruvate carboxylase activity during the induction by ABA, salt, and water stress are due to an increase in the quantity of the enzyme protein. ABA may be a factor in the stress-induced expression of CAM in M. crystallinum, serving as a functional link between stress and biochemical adaptation.  相似文献

Recently there have been breakthroughs on a number of fronts in abscisic acid (ABA) biology research that have advanced the field significantly, including discovery of genes involved in ABA metabolism, along with progress in understanding of ABA signaling (Finkelstein and others 2002; Kushiro and others 2004; Lim and others 2005; Saito and others 2004). At the same time, the chemistry of ABA has advanced. New analytical methods have been developed for profiling ABA and catabolites (Ross and others 2004; Zaharia and others 2005). Novel bioactive catabolites have been discovered from feeding studies with deuterated ABA and catabolites (Zaharia and others 2004; Zhou and others 2004). This review covers recent advances and prospects in natural products chemistry, analysis of ABA catabolism, and applications of ABA analogs for biochemical studies and horticultural uses.  相似文献

The effect of abscisic acid (ABA) and of gibberellic acid (GA2)on the metabolism of kinetin in detached leaves was investigated.Kinetin labelled in either the purine ring or the N-6 substitutedside chain was fed into leaves of Rumex pulcher. The leaveswere extracted 4 or 20 h later and the extract chromatographedon paper which was subsequently scanned for radioactivity. Fourmajor radioactive peaks were revealed. Two peaks were tentativelyidentified, one kinetin and its riboside, and the other adenine.ABA effected a significant decrease in the transformation ofkinetin to adenine while GA3 had no consistent effect. The effects of abscisic acid (ABA) on individual plants of Lemna minor L. were studied. The effects on growth and metabolism of the roots were the most noticeable and the most desirable to measure. Two mg/1 of ABA inhibited the root growth rate by 60% and this was accompanied by a 60% deceleration in the rate of uridine incorporation. The uptake of uridine and leucine and the incorporation of leucine were not affected by ABA. The latent period of root growth inhibition was 1 hour, whereas the inhibition of ribonucleic acid (RNA) synthesis occurred 2 to 4 hours after application. The growth inhibition caused an accumulation of starch in the peripheral, differentiated cell layer of the cortex. Apparently, the growth inhibition by ABA was not entirely due to an inhibition of RNA synthesis, and other plausible mechanisms of growth inhibition are discussed.  相似文献

Mefluidide, N-(2,4-dimethyl-5[([trifluoromethyl]sulfonyl) amino] phenyl)acetamide, a synthetic plant growth regulator, was capable of triggering an increase in endogenous free abscisic acid content when corn (Zea mays L.) plants were grown in a nonstress, day/night, temperature regime (26°C) with sufficient moisture supply. The relevance of such an abscisic acid increase prior to chilling exposure and the water relations during chilling are discussed in reference to the mefluidide protection of the chilled corn plants.  相似文献

A cell-free enzyme system capable of metabolizing abscisic acid has been obtained from Eastern Wild Cucumber (Echinocystis lobata Michx.) liquid endosperm. The reaction products were determined to be phaseic acid (PA) and dihydrophaseic acid (DPA) by co-chromatography on thin layer chromatograms as the free acids, methyl esters, and their respective oxidation or reduction products. The crude enzyme preparation was separated by centrifugation into a particulate abscisic acid (ABA)-hydroxylating activity and a soluble PA-reducing activity. The particulate ABA-hydroxylating enzyme showed a requirement for O2 and NADPH, inhibition by CO, and high substrate specificity for (+)-ABA. Acetylation of short term incubation mixtures gave evidence for the presence of 6′-hydroxymethyl-ABA as an intermediate in PA formation. Determinations of endogenous ABA and DPA concentrations suggest that the ABA-hydroxylating and PA-reducing enzymes are extensively metabolizing ABA in the intact E. lobata seed.  相似文献

A Chiralcel OJ column was used to determine the absolute configuration of naturally occurring α-ionylideneacetic acid from Cercospora rosicola and γ-ionylideneacetic acid from C. cruenta as (R) enantiomers in accordance with their biosynthetic product, (S)-ABA. Both enantiomers of [1, 2-13C2]-α and γ-ionylideneacetic acids were prepared and fed to C. rosicola and C. cruenta. Six combinations of feeding experiments comparatively and unequivocally demonstrated stereoselectivity in the biosynthetic conversions, including stepwise hydroxylation at C-1′ and 4′. Enzymatic isomerization from the γ to α-intermediate was suggested not to be involved in ABA biosynthesis in C. rosicola.  相似文献

Experiments were conducted to investigate indole-3-acetic acid (IAA) and abscisic acid (ABA) metabolism associated with postanthesis senescence of ovaries from nonpollinated muskmelon (Cucumis melo L.) flowers. Flowers attached to the vine were allowed to senesce for 4 days after anthesis or were harvested at full anthesis and aged for the same time interval with or without access to water. The IAA ester, amide-linked forms of IAA, free ABA, and ABA esters increased in senescent ovaries from flowers left attached to the vine. Detaching flowers from the vine resulted in an accumulation of free and amide-linked IAA in the senescing ovary but suppressed accumulation of ester IAA. Free ABA failed to increase in ovaries detached from the vine. Subjecting detached flowers to water stress had no effect on the endogenous level of free ABA but resulted in the accumulation of ABA ester and suppression of any increase in free IAA. However, detached flowers treated with 0.1 millimolar ABA accumulated 75% less free IAA and initiated the synthesis of ester IAA. Detached flowers treated with ABA also accumulated high levels of ester ABA. These results suggest that the metabolism of free IAA in muskmelon ovary tissue is regulated in situ and not the consequence of external synthesis and importation. ABA appears to be transported into the senescing ovary from an external source and alters the IAA metabolism in such a manner as to suppress the level of free IAA while stimulating accumulation of the ester IAA.  相似文献

Birch seedlings ceased extension growth when transferred fromlong days (LD) to short days (SD). The cessation of growth underSD was associated with enhanced rates of photosynthesis, probablydue to a lowered diffusive resistance to gas exchange. Endogenouslevels of ABA were lowered under SD but the degradation of 2-14C(±)ABAwas not disturbed. It is concluded that the photoperiodic controlof extension growth in Betula lutea is not mediated by ABA,although hormonal changes following transfer to SD may findsome expression in an altered stomatal physiology.  相似文献

The Selective Effect of Abscisic Acid on Ribonucleic Acid Components   总被引:1,自引:0,他引:1  
As determined by methylated albumin kieselguhr (MAK) fractionation, GA3 (gibberellic acid) significantly increased and ABA (abscisic acid) decreased RNA levels. In the case of ABA this effect was selective, the ribosomal RNA manifesting the typical decrease; while the sRNA peak was markedly increased. The pattern of labelled uridine incorporation into RNA resembles the MAK absorbancy profile and here as well, ABA although causing an overall decrease, increases labelling in the sRNA peak. The results are interpreted as a possible selective effect of ABA or alternatively as an accumulation in the sRNA peak of rRNA breakdown products. From in vitro experiments it was furthermore evident that ABA mediated RNA hydrolysis probably does not involve a direct activation of RNase by ABA. The in vivo effect would probably be devious.  相似文献

It was found earlier that depodding and girdling treatments which obstruct translocation, result in increased leaf AbA levels and partial stomatal closure. In the present work (±) [2-14C]abscisic acid (AbA) was introduced into leaves and the mass, and radioactivity of AbA and AbA-metabolites were analyzed following translocation obstruction to determine whether the increased AbA was due to higher rates of synthesis, or lower rates of catabolism or export. The (±) [2-14C]AbA was introduced into soybean (Merr.) leaves by injection into the petiole region. AbA and AbA-metabolites (phaseic acid [PA], dihydrophaseic acid [DPA], AbA-conjugate, and an unknown metabolite) were separated with preparative high performance liquid chromatography. Methyl esters of AbA (free and that released after hydrolysis of AbA-conjugate), PA and DPA were determined with gas chromatography using electron capture detection.  相似文献
