The influence of novel biologically active substance potassium 4-toluenethiosulfonate in concentration of 4 x 10(-5) M on the changes of electrophysiological parameters of embryonic cells in early development of fish (Misgurnus fossilis L.) was investigated including the changes of membrane potential (TMP) and enzyme activity of plasmatic membranes of loach embryos during the period of synchronous division of blastomers in the early period of development. The evaluation of influence of these matters was studied and aperiodic changes of the level of TMP were shown. The diminishing of amplitude in every period by 7/12 mV and diminishing of growth of maximal values of vibrations of TMP by 39 mV in comparison to control was noticed. It was related to inhibition of some biosynthetic processes and results in the decline of activity of membrane enzyme (Na+, K(+)-ATPase) by 75.5 +/- 4.1% and 78.4 +/- 10.4% both at the action of high (4 x 10(-3) M) and low (4 x 10(-9) M) concentrations, accordingly, at first hours of development with subsequent renewal of its activity to the level of control only for the actions of low concentration.  相似文献

The activity of aldolase was determined in different parts of the loach (Misgurnus fossilis L.) embryo at the stages from the formation of axial organs till the beginning of embryonic movements (from 19 till 38 hrs of development at 21.5 degrees). In all parts of the embryo, the activity of aldolase at first decreased (21-23 hrs) and then increased. The region of somites is characterized by the highest absolute and specific activity at all developmental stages. The increase in the number of somites is accompanied by the fall and subsequent rise of aldolase activity. In the somites of different degree of differentiation, the enzyme activity changes in a similar way. Hence, there is no correlation between the morphological and biochemical differentiation of somites. Differences in the specific aldolase activity between the anterior and posterior halves of the embryo and the regions of head, somites and tail were found at the stages of 19-23 hrs of development. The maternal aldolase only is present at these stages, as was shown earlier. It means that the early stages of biochemical differentiation may be realized not by means of differential activation of genes controlling the enzyme, but by means of regylation of translation on the templates stored in oogenesis.  相似文献

Human DNA polymerase iota (Pol ι) is a Y-family DNA polymerase with unusual biochemical properties and not fully understood functions. Pol ι preferentially incorporates dGTP opposite template thymine. This property can be used to monitor Pol ι activity in the presence of other DNA polymerases, e.g. in cell extracts of tissues and tumors. We have now confirmed the specificity and sensitivity of the method of Pol ι activity detection in cell extracts using an animal model of loach Misgurnus fossilis embryos transiently expressing human Pol ι. The overexpression of Pol ι was shown to be accompanied by an increase in abnormalities in development and the frequency of pycnotic nuclei in fish embryos. Further analysis of fish embryos with constitutive or regulated Pol ι expression may provide insights into Pol ι functions in vertebrate animals.  相似文献

rRNA synthesis was compared in the loach haploid (In) and diploid (2n) embryos. The relative intensity of synthesis was evaluated by 14C-uridine incorporation in 27S and 18S rRNA isolated from ribosomes taking into account label incorporation into total acid-soluble fraction and phosphrylated uridine derivatives. Label incorporation into rRNA, in reference with DNA content in 1n and 2n embryos, suggests that the level of rRNA synthesis per DNA unit in haploids is twice that in diploids whereas, in reference per cell, the same amount of ribosomes is synthesized both in haploids and diploids. The data obtained show that the amount of rRNAs synthesized in the loach embryogenesis does not depend on ploidy.  相似文献

Tetracycline tagging in coregonid embryos and larvae   总被引:3,自引:0,他引:3  
Coregonus peled (Gmelin) embryos at the eyed stage were immersed in tetracycline hydrochloride (TC) solution (600 mg l−1) and fluorescent marks on the otoliths from fish were identified under a UV light microscope. Three weeks after treatment, when the larval fish had hatched, multiple primordia of the sagitta had fluorescent bands whose locations suggest that calcification begins before formation of the otolith's core. Upon hatching, increments were few or absent, and daily increments started forming after hatching. In coregonid larvae immersed in TC solution immediately after hatching, the fluorescent band was only 1 day's growth increment wide in the otolith. The age of larvae based on ring counts after marking corresponded to the actual age, but further studies are required. Triple marks due to 35-h immersions in TC solution separated by 5–7 day feeding periods were clearly readable in the otoliths. One month after treatment the fluorescent marks were identified under UV light on otoliths from every fish. In treated embryos, 38% of fish had marks 3 months after TC immersion, whereas in treated larvae 60–80% of fish grown up to 38.7 mm were marked.  相似文献

The effects of sodium selenite on the ribonucleic acid synthesis in normal diploid embryos and isolated nuclei of the loach Misgurnus fossilis were studied. The relative rate of synthesis was determined from incorporation of [3H]uridine into the total RNA, taking into consideration the incorporation of the label into the total acid-soluble fraction and into phosphorylated derivatives of uridine. It was shown that sodium selenite inhibits the RNA synthesis in loach embryos at the gastrula stage. It was also found that sodium selenite exerts an inhibiting effect on the activity of form I of DNA-dependent RNA-polymerase in isolated loach nuclei without affecting the activity of the enzyme form II.  相似文献

Loach eggs are stated to possess some quantities of insulin or insulin-like substances with a positive reaction to antiinsulin antiserum. The level of these components is five times as high 30 minutes after fertilization, and does not change for the next 20 hours of the embryo development. No 17-oxycorticosteroids were detected in loach eggs and embryos during first four hours of the development; they appeared only at the blastula stage and at the state of gastrula their level increases considerably. It is supposed that insulin or insulin-like substances participate in metabolism regulation and cell differentiation of fish embryos during early stages of embryogenesis.  相似文献

The value of ATPase activity of the myofibril preparations and the value and duration of actomyosin superprecipitation were estimated for different muscles during the chick embryonic development. The ATPase level increases during embryogenesis 4.5-fold, in the leg muscle this change takes place distinctly earlier than in the leg muscle. The value and rate of actomyosin superprecipitation also markedly increase, to a lesser extent for m. soleus than for m. pectoralis. It is suggested that these changes and differences are mainly due to the delay in synthesis of certain types of the embryonic myosin light chains.  相似文献

The activity of "satellite" enzymes related to gluconeogenesis has been measured in the oocytes and embryos at the early stages of loach (Misgurnus fossilis L.) embryogenesis. The activity of pyruvate dehydrogenase increase during oocyte maturation by 30%, remains constant at the cleavage and blastula stages and decreased on the onset of gastrulation. In the both oocytes and embryos pyruvate dehydrogenase has been found only in the active form. The activity of citrate synthase, malate dehydrogenase and pyruvate carboxylase remained constant during oocyte maturation and et all early stage of embrional development. Citrate lyase and "malic"-enzyme were not found, Oocyte maturation is followed by a considerable increase in the malate and oxalacetate content, the level of pyruvate and acetyl-CoA being found invariable.  相似文献
