Thin layer chromatography was used to analyze the glucose and maltose concentrations of the digestive gland–gonad complex (DGG) of uninfected-estivated Biomphalaria glabrata snails and estivated B. glabrata patently infected with Schistosoma mansoni. All snails were estivated in a most chamber at a relative humidity of 98 ± 1% and a temperature of 23 ± 1 °C for 14 days. Carbohydrates were extracted from the DGG with 70% aqueous ethanol, and extracts were analyzed on silica gel preadsorbent plates using ethyl acetate–glacial acetic acid–methanol–water (60:15:15:10) mobile phase, α-naphthol–sulfuric acid detection reagent, and quantification by densitometry. The concentrations of glucose and maltose were significantly reduced in both uninfected-estivated snails and infected-estivated snails.  相似文献

Snail surveys were carried out in various parts of Mali. All areas surveyed are part of the Niger basin being either affluents or irrigation schemes fed by this river. The snail species present varied greatly between areas. The following potential hosts of schistosomes were recorded: Biomphalaria pfeifferi, Bulinus truncatus, B. globosus, B. umbilicatus, B. forskalii and B. senegalensis. In the large irrigation schemes, i.e. ‘Office du Niger’ and Baguinéda, only B. pfeifferi and B. truncatus appear to be intermediate hosts. Snail distribution appeared to some extent to be focal and high snail densities appeared to be associated with human water contact activities, which apparently create favourable biotopes for the snails. This is probably due to an alteration of the vegetation and an increase of the trophic status of the site by contamination with food remnants and other debris. The larger irrigation canals or lakes in these schemes play an important role in the transmission of human schistosomes and transmission appears to be very focal in these habitats. Infected snails are almost exclusively found in well-defined human water contact sites (WCS). Local infection rates with schistosomes were often high (i.e. up to 27% in B. pfeifferi). In urban areas (i.e. Bamako), transmission patterns are more variable. In Bamako schistosome-infected B. truncatus were found in the Niger river. A number of smaller semi-permanent or permanent streams are very important transmission sites, and schistosome infections were recorded from B. pfeifferi, B. truncatus and B. globosus. Schistosome infection rates in B. pfeifferi were often high (up to 30%). In a new lake at Sélingué, B. truncatus was found to be widely distributed only about a year and a half after the dam was constructed, and in some sites schistosome infections were recorded.  相似文献

Cryptosporidium sp. cervine genotype could possibly emerge as an important human pathogen because current evidence suggests this genotype has a wide host range and zoonotic potential. However, there is confusion in the taxonomy of the Cryptosporidium sp. cervine genotype because different names have been used to refer to this genotype in previous studies and in sequences submitted to GenBank. Consequently, we lack a clear understanding of the epidemiology of this genotype. In the present study, the Cryptosporidium sp. cervine genotype was identified in sheep, was characterized using 3 genes (18S rDNA, COWP, and HSP-70), and was compared with data from all previous reports. Intragenotypic variations were found at the 18S rDNA gene in Cryptosporidium sp. cervine genotype with 3 different subtypes (cervine 1-3). No sequence variation at HSP-70 and COWP genes were observed. Sheep should be considered as an important reservoir for this zoonotic genotype of Cryptosporidium sp.  相似文献

In Lake Albert, an ecological study was conducted, between June 2000 and May 2003, which assessed snail population dynamics, parasite infection patterns and interplay of environmental factors upon Biomphalaria. Monthly sampling surveys were conducted at 29 sites monitoring populations of Biomphalaria stanleyi and Biomphalaria sudanica. Altogether, a total of 21,715 B. stanleyi and 8452 B. sudanica were collected during the period. Both species could be found infected with Schistosoma mansoni although infection prevalence was significantly higher (p < 0.01**) in B. stanleyi (4.4%) than in B. sudanica (3.5%). Each species occupied slightly different aquatic niches with B. stanleyi preferring deeper water habitats whilst B. sudanica was found along the shoreline in shallower water. B. stanleyi was more widely distributed among the sampling locations (19 sites) than B. sudanica (10 sites). Of the four villages included in the study area, snails from sites near Piida and Bugoigo villages had the highest schistosome infection rates, presumably attributable to the closer proximity of people with intestinal schistosomiasis. After inspection of cross-correlation plots which identified most suitable time lags, snail density dynamics could be associated with seasonal variations inclusive of: air temperature, rainfall, lake level, water temperature, water conductivity and water pH. These temporal observations better reveal the relationship between snail populations and environmental factors, providing important information concerning the relative roles of B. stanleyi and B. sudanica in transmission of S. mansoni and development of integrated strategies for disease control around Lake Albert.  相似文献

Dry and rainy season investigations of diverse freshwater habitats in south-western Nigeria revealed fourteen species of snail comprised of nine pulmonates: Biomphalaria pfeifferi Krauss, Bulinus globosus Morelet, Bulinus rohlfsi Clessin, Lymnaea natalensis Krauss, Physa ( Aplexa) waterloti Germain, Bulinus forskali Ehrenberg, Gyraulus costulatus Krauss, Ferrissia sp, Segmentorbis sp. and five prosobranchs namely, Lanistes libycus Morelet, Lanistes ovum Peters, Pila wernei Philippi, Potadoma moerchi Reeve and Melanoides tuberculata Müller.The influence of diverse chemical and physical properties of water on their occurrence as well as interspecific and snail-plant relationships are discussed and maps showing the distribution of the species encountered are presented. Among the intermediate hosts of Schistosoma, B. globosus was widely distributed and common, B. pfeieri was also widely distributed but infrequent and B. rohlfsi was rare.  相似文献

A new anisakid nematode, Hysterothylacium patagonense n. sp., is described from the intestine of the temperate bass Percichthys trucha (Cuvier & Valenciennes), a freshwater fish from Lake Aluminé, Patagonia, Argentina. It is characterised mainly by the absence of lateral alae, the length ratio of the caecum and ventricular appendix (1:0.9–1.8), the length of the spicules (0.952–1.292 mm) and the presence of minute spines on the tip of the tail. In the same locality and in Lake Huechulafquen, advanced third-stage and fourth-stage larvae of this parasite were also recorded from the digestive tract of the introduced salmonids, Salvelinus fontinalis (Mitchell), Oncorhynchus mykiss (Walbaum) and Salmo trutta L. This is the second Hysterothylacium species described from freshwater fishes in South America. A key to Hysterothylacium species parasitising American freshwater fishes is provided.  相似文献

Land snails of the genus Euhadra (Gastropoda: Bradybaenidae) are indigenous to the Japanese Archipelago. The larvae of an unknown species, tentatively named as Brachylaima sp. B (Trematoda: Brachylaimidae), have been found from Euhadra brandtii sapporo in Hokkaido, the northernmost island of Japan. In this study, a large-scale snail survey covering a wide area of Japan was conducted to confirm the larval parasite from members of Euhadra and related genera. Sporocysts with cercariae were found only from Eu. brandtii sapporo in Hokkaido and Euhadra callizona in central Honshu at low prevalence (1.0–9.6%). The metacercariae were detected widely from 6 species of Euhadra and the related genera at high prevalence (7.1–100%). A molecular identification by DNA barcoding demonstrated almost all of the larvae to be Brachylaima sp. B. Adult worms experimentally raised from the metacercariae were morphologically most similar to Brachylaima ezohelicis in Hokkaido, but could be differentiated by the microstructure of the tegumental surface. We propose Brachylaima lignieuhadrae n. sp. for the unknown species, based on the morphology, DNA profile, host specificity, and geographic distribution. Phylogeography of the new species suggests a possibility that migratory birds serve as the definitive hosts.  相似文献

 Fluorogenic substrate analogues (MUF substrates) are very sensitive in detecting hydrolytic enzymes. This method was adapted for the quantitative analysis of extracellular enzymes in snails and other animals. It was then used to determine cellobiase, chitobiase, protease, esterase, phosphatase and lipase in the digestive tract of Radix peregra and Bithynia tentaculata. The method was sensitive enough to determine the complete enzyme kinetics (K m, V max values) in the esophagus, stomach, digestive gland and intestine of single animals adapted to various natural foods. With the exception of lipase and phosphatase, K m values were between 15 and 30 mmol l–1, independent of prefeeding. Enzyme activities, in contrast, changed with food conditions: cellobiase-activities ranged from 80 to 8000 picokatal, esterase from 500 to 3000 pKat, chitobiase from 150 to 750 pKat and protease from 100 to 1100 pKat per animal. R. peregra had cellobiase activities 10 times higher than B. tentaculata. For chitobiase, esterase and protease the enzyme activities were in the same order of magnitude in both species. All enzymes differed significantly in activity between prefeeding regimes. This variability with prefeeding was much higher in Bithynia than in Radix. Control of soil-transmitted helminth infection and elimination of lymphatic filariasis by periodic chemotherapy increase drug pressure for possible occurrence of resistance against single dose anthelminthics. In veterinary practice, frequent treatment of closed populations has led to a serious problem of anthelminthic drug resistance which is now largely irreversible. Reduced efficacy of single dose drugs against nematodes of humans should be taken as early warnings to tackle the issue in due time. Research and development of sensitive tools for monitoring and early detection of drug resistance is urgently needed to sustain the benefits of helminth control programs gained so far. A concerted action with international partners and the creation of a network of scientists to address this issue is the next pressing public health issue for helminth control.  相似文献

Increases in international travel and migratory flows have enabled infectious diseases to emerge and spread more rapidly than ever before. Hence, it is increasingly easy for local infectious diseases to become global infectious diseases (GIDs). National governments must be able to react quickly and effectively to GIDs, whether naturally occurring or intentionally instigated by bioterrorism. According to the World Health Organisation, global partnerships are necessary to gather the most up-to-date information and to mobilize resources to tackle GIDs when necessary. Communicable disease control also depends upon national public health laws and policies. The containment of an infectious disease typically involves detection, notification, quarantine and isolation of actual or suspected cases; the protection and monitoring of those not infected; and possibly even treatment. Some measures are clearly contentious and raise conflicts between individual and societal interests. In Europe national policies against infectious diseases are very heterogeneous. Some countries have a more communitarian approach to public health ethics, in which the interests of individual and society are more closely intertwined and interdependent, while others take a more liberal approach and give priority to individual freedoms in communicable disease control. This paper provides an overview of the different policies around communicable disease control that exist across a select number of countries across Europe. It then proposes ethical arguments to be considered in the making of public health laws, mostly concerning their effectiveness for public health protection.  相似文献

【目的】解析Actinoplanes sp.SE50/110(简称SE50/110)中阿卡波糖脱氧氨基糖单元的生物合成机制。【方法】经过BLASTp分析,推测了Acb A、Acb B和Acb V负责阿卡波糖脱氧氨基糖单元的生物合成。首先,本研究在SE50/110中分别构建了acb A、acb B和acb V的同框缺失和回补突变株。然后,利用大肠杆菌BL21(DE3)/p Gro7分别对Acb A、Acb B和Acb V成功实现了可溶性表达。最后,以D-葡萄糖-1-磷酸为起始底物,通过体外催化反应,研究脱氧氨基糖单元的生物合成过程和相关蛋白的酶学性质。【结果】在SE50/110中分别缺失acb A、acb B和acb V基因后,相应突变株均丧失了阿卡波糖的合成能力,将acb A、acb B和acb V基因分别回补后,各菌株又恢复了阿卡波糖的合成能力,证明了它们均为阿卡波糖生物合成的必需基因。在体外酶促反应中,D-葡萄糖-1-磷酸-胸腺嘧啶转移酶Acb A催化D-葡萄糖-1-磷酸和d TTP合成d TDP-D-葡萄糖,对D-葡萄糖-1-磷酸的Km值为(0.185±0.053)mmol/L,Vmax为(2.366±0.217)μmol/(min·mg);对d TTP的Km值为(4.964±1.089)mmol/L,Vmax为(60.310±5.419)μmol/(min·mg)。d TDP-D-葡萄糖-4,6-脱水酶Acb B催化d TDP-D-葡萄糖转化为d TDP-4-酮基-6-脱氧-D-葡萄糖,Km值和Vmax分别为(0.353±0.089)mmol/L和(306.401±28.740)μmol/(min·mg)。氨基转移酶Acb V催化d TDP-4-酮基-6-脱氧-D-葡萄糖生成d TDP-4-氨基-4,6-双脱氧-D-葡萄糖,Km值和Vmax分别为(1.411±0.293)mmol/L和(3.447±0.279)μmol/(min·mg)。【结论】本研究阐明了阿卡波糖脱氧氨基糖单元的生物合成过程,为全面解析阿卡波糖生物合成途径奠定了基础。同时,测定了相关酶的动力学参数,为代谢工程改造SE50/110,提高阿卡波糖产量提供了重要的理论依据。  相似文献

Alligatorweed (Alternanthera philoxeroidesG.) has become a serious weed in different crops in China. A fungal pathogen was found in Chongqing and Sichuan Provinces and was identified as a species in the Fusarium genus. The fungus produced macroconidia and chlamydospores abundantly on potato sucrose agar (PSA) plates. The bestconidial production and germination and colonygrowth of Fusarium sp. were at 23–31°C and pH 6.7–7.0. Light period and flooding did not affect fungal growth and conidium formation. The herbicides, glyphosate and paraquat, inhibited the fungal development in vitro. The fungus did not affect seed germination and seedling growth of paddy rice, wheat, maize, oilseed rape and broad bean inlaboratory or greenhouse trials. Inoculum density and wetness duration influenced the efficiency of Fusarium sp. to control alligatorweed; a concentration of 1.0 × 105 spores–1 ml and 12 h of high humidity duration after inoculation produced goodinfections on the weed at 23°C in the laboratory. When the fungus was applied to alligatorweed grown in greenhouse and in the field, good biocontrol efficiency was obtained: the plants started to wilt after four to five (greenhouse) or six days (field), and were killed 9–10 (greenhouse) or 13–14 (field) days after spraying the fungal inoculum. This was similar to the control efficiency resulting from glyphosate treatment. Therefore, this Fusarium sp. appeared to be a good candidatefor further studies and a promising biocontrol agent to manage alligatorweed in some terrestrial and aquatic crops.  相似文献

When donor snails, Lymnaea stagnalis, infected with Echinoparyphium aconiatum were held at 13 C for at least 24 days, transplantation of newborn rediae from them to uninfected snails was 2.5 times as successful as when donor snails were held at 23 C. The difference does not seem to be do to the fact that rediae transferred from 13 C-snails were slightly larger than those from 23 C. This effect of temperature was not observed in analogous experiments with another echinostome, Isthmiophora melis. The mechanism of this phenomenon found in the experiments with E. aconiatum is still unknown.  相似文献
