随着整合生物学思想的发展,生态学与免疫学的相互渗透与交叉,产生了生态免疫学这一崭新的学科,自从其诞生虽然只有短短的十几年时间,但发展迅速。生态免疫学主要从免疫代价的视角来解释生活史权衡、性选择和种群动态变化等生态学问题。动物的免疫功能对其抵抗疾病和最终的生存起至关重要的作用,影响动物免疫的因素具有多样性和复杂性的特点,而研究动物免疫功能变化的原因和结果一直是生态免疫学研究的重要内容。免疫防御是否具有能量或资源代价,这种代价是否昂贵是生态免疫学需要回答的基本问题之一,大量的实验已表明免疫防御的代价是昂贵的。由于能量或资源不是无限的,有限的能量或资源必须在多种经常相互竞争的生理功能间进行分配,这导致了免疫功能与动物的生长、繁殖等生活史组分之间的权衡,很多的研究表明增加一个过程的投资会降低对另一过程的投资。免疫同样在性选择特征进化以及维持雌性偏爱性修饰的雄性中发挥至关重要的作用,免疫功能障碍假说认为睾丸激素负责第二性征的产生并同时具有免疫抑制作用,表达性征的代价是降低了免疫功能,这使得宿主对病原体或寄生物攻击的易感性增加,因此只有高质量的雄性个体才能充分表达性征同时又不遭受大量寄生负荷。综述了生态免疫学的概念、研究内容以及未来研究需要关注的领域。  相似文献

There has been considerable recent interest in the effects oflife-history decisions on immunocompetence in birds. If immunocompetenceis limited by available resources, then trade-offs between investmentin life-history components and investment in immunocompetencecould be important in determining optimal life-history traits.For this to be true: (1) immunocompetence must be limited byresources, (2) investment in life-history components must benegatively correlated with immunocompetence, and (3) immunocompetencemust be positively correlated with fitness. To gather such empiricaldata, ecologists need to be able to measure immunocompetence.We review techniques used to measure immunocompetence and howthey are applied by ecologists. We also consider the componentsof the immune system that constitute immunocompetence and evaluatethe possible consequences of measuring immunocompetence in differentways. We then review the empirical evidence for life-historytrade-offs involving immune defense. We conclude that thereis some evidence suggesting that immunocompetence is limitedby resources and that investment in certain life-history componentsreduces immunocompetence. However, the evidence that immunocompetenceis related to fitness is circumstantial at present, althoughconsistent with the hypothesis that immunocompetence and fitnessare positively correlated. We argue that future work needs toexamine the fitness effects of variation in immunocompetenceand suggest that artificial selection experiments offer a potentiallyimportant tool for addressing this issue.  相似文献

Multidirectional interactions among the immune, endocrine, and nervous systems have been demonstrated in humans and non-human animal models for many decades by the biomedical community, but ecological and evolutionary perspectives are lacking. Neuroendocrine-immune interactions can be conceptualized using a series of feedback loops, which culminate into distinct neuroendocrine-immune phenotypes. Behavior can exert profound influences on these phenotypes, which can in turn reciprocally modulate behavior. For example, the behavioral aspects of reproduction, including courtship, aggression, mate selection and parental behaviors can impinge upon neuroendocrine-immune interactions. One classic example is the immunocompetence handicap hypothesis (ICHH), which proposes that steroid hormones act as mediators of traits important for female choice while suppressing the immune system. Reciprocally, neuroendocrine-immune pathways can promote the development of altered behavioral states, such as sickness behavior. Understanding the energetic signals that mediate neuroendocrine-immune crosstalk is an active area of research. Although the field of psychoneuroimmunology (PNI) has begun to explore this crosstalk from a biomedical standpoint, the neuroendocrine-immune-behavior nexus has been relatively underappreciated in comparative species. The field of ecoimmunology, while traditionally emphasizing the study of non-model systems from an ecological evolutionary perspective, often under natural conditions, has focused less on the physiological mechanisms underlying behavioral responses. This review summarizes neuroendocrine-immune interactions using a comparative framework to understand the ecological and evolutionary forces that shape these complex physiological interactions.  相似文献

The immunocompetence handicap hypothesis postulates that secondary sexual traits are honest signals of mate quality because the hormones (e.g. testosterone) needed to develop secondary sexual traits have immunosuppressive effects. The best support for predictions arising from the immunocompetence handicap hypothesis so far comes from studies of insects, although they lack male-specific hormones such as testosterone. In our previous studies, we found that female mealworm beetles prefer pheromones of immunocompetent males. Here, we tested how juvenile hormone (JH) affects male investment in secondary sexual characteristics and immune functions in the mealworm beetle, Tenebrio molitor. We injected male mealworm beetles with JH (type III) and found that injection increased the attractiveness of male pheromones but simultaneously suppressed immune functions (phenoloxidase activity and encapsulation). Our results suggest that JH, which is involved in the control of reproduction and morphogenesis, also plays a central role in the regulation of a trade-off between the immune system and sexual advertisement in insects. Thus, the results reflect a general mechanism by which the immunocompetence handicap hypothesis may work in insects.  相似文献

Sexual dimorphism in immune function is a common pattern in vertebrates and also in a number of invertebrates. Most often, females are more 'immunocompetent' than males. The underlying causes are explained by either the role of immunosuppressive substances, such as testosterone, or by fundamental differences in male and female life histories. Here, we investigate some of the main predictions of the immunocompetence handicap hypothesis (ICHH) in a comparative framework using mammals. We focus specifically on the prediction that measures of sexual competition across species explain the observed patterns of variation in sex-specific immunocompetence within species. Our results are not consistent with the ICHH, but we do find that female mammals tend to have higher white blood cell counts (WBC), with some further associations between cell counts and longevity in females. We also document positive covariance between sexual dimorphism in immunity, as measured by a subset of WBC, and dimorphism in the duration of effective breeding. This is consistent with the application of 'Bateman's principle' to immunity, with females maximizing fitness by lengthening lifespan through greater investment in immune defences. Moreover, we present a meta-analysis of insect immunity, as the lack of testosterone in insects provides a means to investigate Bateman's principle for immunity independently of the ICHH. Here, we also find a systematic female bias in the expression of one of the two components of insect immune function that we investigated (phenoloxidase). From these analyses, we conclude that the mechanistic explanations of the ICHH lack empirical support. Instead, fitness-related differences between the sexes are potentially sufficient to explain many natural patterns in immunocompetence.  相似文献

Sexual dimorphism in immunocompetence, usually in the directionof inferior male immunocompetence, has historically been explainedas the result of proximate physiological mechanisms such asthe immunosuppressive effects of the male hormone testosterone.More recently, it has been argued that this pattern is bestunderstood as a result of resource-based trade-offs betweenmale mating effort and immune defense, a trade-off that femalesdo not make. The central prediction of this hypothesis is thatas the strength of sexual selection on males increases, themagnitude of the sex differences in immunocompetence will increase.Two implicit assumptions of this argument are that 1) longevityis of more importance for female than for male fitness and 2)that the primary benefit of immunocompetence is increased longevity.However, both of these assumptions may not be as broadly applicableas has been argued. We have modeled the optimal allocation toimmunocompetence for males and females without making theseassumptions. We find that the optimal allocation to immune defensefor males decreases as the strength of sexual selection increases,as predicted. However, males may still invest more, relativeto females, into immunocompetence if the impact of parasiteson condition differs for the sexes and/or if the relationshipbetween condition and reproduction differs for the sexes. Weargue that these previously overlooked assumptions may be criticalfor predicting sex-specific patterns of immunocompetence.  相似文献

Disentangling the sources of variation in developing an effective immune response against pathogens is of major interest to immunoecology and evolutionary biology. To date, the link between immunocompetence and genetic variation at the major histocompatibility complex (MHC) has received little attention in wild animals, despite the key role of MHC genes in activating the adaptive immune system. Although several studies point to a link between MHC and immunocompetence, negative findings have also been reported. Such disparate findings suggest that limited statistical power might be affecting studies on this topic, owing to insufficient sample sizes and/or a generally small effect of MHC on the immunocompetence of wild vertebrates. To clarify this issue, we investigated the link between MHC variation and seven immunocompetence proxies in a large sample of barn owls and estimated the effect sizes and statistical power of this and published studies on this topic. We found that MHC poorly explained variation in immunocompetence of barn owls, with small‐to‐moderate associations between MHC and immunocompetence in owls (effect size: .1 ≥ r ≤ .3) similar to other vertebrates studied to date. Such small‐to‐moderate effects were largely associated with insufficient power, which was only sufficient (>0.8) to detect moderate‐to‐large effect sizes (r ≥ .3). Thus, studies linking MHC variation with immunocompetence in wild populations are underpowered to detect MHC effects, which are likely to be of generally small magnitude. Larger sample sizes (>200) will be required to achieve sufficient power in future studies aiming to robustly test for a link between MHC variation and immunocompetence.  相似文献

The immunocompetence handicap hypothesis suggests that the immune system competes for resources with sexually selected ornaments; variation in ornaments might reflect genetic variation for immunocompetence. We tested this genetic prediction by mating scorpionfly females to males differing in the expression of a condition-dependent ornament trait, saliva secretion, and then comparing offspring immunocompetence. We found several indications of an immunocompetence handicap in our study: females had superior immunocompetence compared with males, the different immune traits were positively correlated, and there were indications of genetic variation in immune traits. However, we found no significant difference in the immunocompetence of offspring derived from males differing in ornament expression, only a tendency for sons of ornamented males to possess slightly better immunocompetence. The estimated effect of fathers on offspring immunocompetence was rather small, but it might be a sufficient benefit of female choice, provided that the costs of choice are small. We conclude that the genetic benefit of female choice is small concerning offspring immunocompetence, but the immunocompetence handicap principle might nevertheless work in scorpionflies.  相似文献

Sex differences in immunity are often observed, with males generally having a weaker immune system than females. However, recent data in a sex-role-reversed species in which females compete to mate with males suggest that sexually competitive females have a weaker immune response. These findings support the hypothesis that sexual dimorphism in immunity has evolved in response to sex-specific fitness returns of investment in traits such as parental investment and longevity, but the scarcity of data in sex-reversed species prevents us from drawing general conclusions. Using an insect species in which males make a large but variable parental investment in their offspring, we use two indicators of immunocompetence to test the hypothesis that sex-biased immunity is determined by differences in parental investment. We found that when the value of paternal investment was experimentally increased, male immune investment became relatively greater than that of females. Thus, in this system, in which the direction of sexual competition is plastic, the direction of sex-biased immunity is also plastic and appears to track relative parental investment.  相似文献

A new era of cancer immunotherapy has brought not only successful cancer vaccines but also immunomodulators, such as those that target checkpoint blockade in order to induce endogenous host immune responses. However, the immune system of cancer patients can be compromised through multiple means, including immune suppression by the tumor and by prior therapies such as chemotherapy and radiation. Therefore, a comprehensive means of assessing patient immunocompetence would seem helpful for determining whether patients are ready to benefit from immunotherapy, and perhaps even which immunotherapy might be most appropriate for them. Unfortunately, there are no standardized tests for immune competence, nor is there agreement on what to measure and what will be predictive of outcome. In this review, we will discuss the technologies and assays that might be most useful for this purpose. We argue for a comprehensive approach that should maximize the chances of developing predictive biomarkers for eventual clinical use.  相似文献

Despite intensive studies in ecological immunology, few have investigated variation in immune defence among natural populations; in particular, there is a lack of knowledge of the sources of spatial variability in immune defence in the wild. Here we documented variation among twelve populations of the freshwater crustacean Gammarus pulex in the activity of the prophenoloxidase (ProPO) system, which is an important component of invertebrate immunity. We then tested for trade-offs between investment in immune defence and fitness-related traits such as survival and fecundity, as well as for environmental causes of variability (water temperature and conductivity, parasite prevalence). Levels of immune defence differed among populations, with environment partly explaining this population effect, as immune activities were negatively related to water conductivity and acanthocephalan parasite prevalence. There was a strong variation among populations for the maintenance of the ProPO system, while variation in its use was relatively weak. Such a pattern could be partly explained by the relative costs associated with the maintenance and/or the use of the ProPO system. Investment in the ProPO system was negatively correlated to survival, whereas it was positively related to female fecundity and resource storage. However, variation in immunity did not predict resistance to bacterial infection among populations, suggesting that measuring the activity of the ProPO system might not be sufficient to estimate immunocompetence at the population level. These results suggest that investment in immune function is a variable trait, which might be locally optimized as a result of both life history trade-offs and environmental conditions, highlighting the need to combine them in a common framework.  相似文献

Females often select mates on the basis of sexual signals, which can be reliable indicators of male quality when the costliness of these signals prevents cheating. The immunocompetence handicap hypothesis (ICHH) provides a mechanistic explanation of these costs, by proposing a trade-off between immune function and sexual displays. This trade-off arises because testosterone enhances sexual signals, but suppresses immune function. Many studies have investigated the ICHH by administrating testosterone, and a recent meta-analysis found little evidence that testosterone suppressed immune function. However, another component of the ICHH, which has received less empirical interest, suggests that there may also be an interaction in the other direction, with immune activation suppressing testosterone levels. We present a meta-analysis to test for this effect. Overall, there was a strong suppressive effect of experimental immune activation on testosterone levels (r=-0.52), regardless of whether live pathogens or non-pathogenic antigens were used to challenge the immune system. The latter is important because it shows that immune activation per se suppresses testosterone levels. Thus, a trade-off between immunocompetence and sexual displays may primarily be generated by the effect of immune activation on testosterone, rather than the opposite effect that has received most attention.  相似文献

The level of expression of secondary sexual characters has been suggested to signal male ability to resist parasitic infestations. To test this idea, several studies have examined the link between sexual signals and immunocompetence in birds. However, most of them have used only a single aspect of immune response to evaluate immunocompetence. We investigated the relation between bill colour and immunocompetence in captive male European blackbirds, Turdus merula, during the breeding season by assessing both cell-mediated and humoral components of the immune system. The blackbird is a sexually dimorphic species with bill colour varying from yellow to orange in males. Humoral immunity was assessed by measuring both primary and secondary responses to sheep red blood cell inoculation. Cell-mediated immunity was estimated with a delayed cutaneous hypersensitivity response to an injection of a mitogen (phytohaemagglutinin). No relation was found between male bill colour and the primary humoral response. However, males with orange bills showed a lower secondary humoral response but a higher cell-mediated immune response than males with yellow bills. Thus, the relation between immunocompetence and a secondary sexual trait may differ markedly depending on which component of the immune system is under consideration. We discuss our results in relation to mechanisms involved in sexual selection. The endocrine system serves as a mediator by which the body integrates environmental cues to organize physiological alterations, including changes in immunocompetence. Hormones are central mechanisms that contribute to the onset and timing of key life history events, the allocation of time and energy between competing functions, and in general modulate phenotypic development and variation. Here we provide a very brief review of testosterone and immunity, which highlights the physiological costs that elevated testosterone levels can incur as a result of reproductive investments. We focus primarily on nonhuman primates where possible. Although there is substantial evidence that testosterone exerts some influences on immune responses, results from in vivo studies involving human and nonhuman primates have yielded equivocal results regarding such immunomodulatory actions. There may be several reasons for this, including variation in study design, immunological measures used, levels of other hormones present, host energy status, and even social conditions. We therefore review some of these potential methodological issues, concluding that increased care must be taken to analyze seasonal variability in energy budgets, to collect an adequate number of samples from known individuals, to account for status in the dominance hierarchy when applicable, and to use multiple measures of immunity. We must also seek to understand the collaborative effects of multiple hormones (particularly dehydroepiandrosterone and estradiol) with relation to their downstream immunological effects, assessing both individual and multiplicative actions in both males and females. Such efforts would benefit from the development of additional noninvasive immune measures for primates.  相似文献

Studies of the relationship between sexual traits and immune function have been at the forefront of sexual selection during the last decade. Whereas evidence is accumulating that there is a trade-off between sexual ornamentation and immunocompetence, the reasons for this trade-off are still unknown. Importantly, most studies have addressed this issue only at the adult stage, when sexual ornamentation and immune function may be fully developed. We show here that juvenile cell-mediated immune response is negatively correlated with subsequent size of an adult sexual ornament in the Chinese quail, Coturnix chinensis. This suggests that the cost of development of a functional immune system is traded off against secondary sexual traits, and that costs of high immunocompetence in juveniles may not be manifested until sexual maturity. Ontogenetic studies of the development of the immune function and associated costs and trade-offs are likely to provide a more complete picture of the links between sexual selection and immunobiology.  相似文献

Inbreeding resulting from severe population bottlenecks may impair an individual's immune system and render it more susceptible to disease. Although a reduced immune response could threaten the survival of highly endangered species, few studies have assessed the effect of population bottlenecks on immunocompetence. We compared the counts of leucocytes and external, blood and gastrointestinal parasite loads in two populations of the endemic New Zealand robin Petroica australis to assess the immunocompetence of birds in a severely bottlenecked population relative to its more genetically diverse source population. Despite similar parasite loads in both populations, robins in the severely bottlenecked population showed lower counts of both total leucocyte and total lymphocyte numbers. When the immune system was experimentally challenged using the phytohaemagglutinin skin test, robins in the severely bottlenecked population exhibited a significantly lower immune response than the source population, suggesting that birds passing through a severe bottleneck have a compromised immunocompetence. Our results confirm that severe bottlenecks reduce the immune response of birds and highlight the need to avoid severe bottlenecks in the recovery programmes of endangered species.  相似文献

The avian spleen has been frequently used in studies of avian ecology, parasitology, and evolution to infer immune system strength in birds. Traditionally, it has been assumed that a large spleen is representative of a strong immune system and conclusions based on this assumption have led to interesting interpretations of the role of disease and parasitism, for example in predator-prey interactions. This assumption of a positive relationship between spleen size and immune system strength has been made despite an incomplete understanding of the physiology of the avian spleen and little evidence of the validity of such an assumption. In this response, we demonstrate that the assumption of a predictable, positive relationship between spleen size and immunocompetence may be unjustified based on what is known of avian splenology. We also review recent research that may indicate that the inverse of the above assumption is true and we discuss general limitations of the use of the spleen as an indicator of immune system strength in birds. Finally, we make recommendations for future research topics in this field of study.  相似文献

A simple version of the immunocompetence handicap hypothesizesthat through condition-dependence, the size of the sexual traitmay be positively related to immune function at the populationlevel. In the present study, we investigated the relationshipbetween sexual advertisement and immune function in a naturalpopulation of male wolf spiders, Hygrolycosa rubrofasciata (Araneae:Lycosidae). Males of H. rubrofasciata have a costly and condition-dependentacoustic signal, courtship drumming. In the mating season, malesdrum against dry leaves while wandering around the habitat searchingfor receptive females. Males increase their mating success byincreasing their drumming rate and mobility. We used drummingrate and mobility measured without female proximity as estimatesof sexual advertisement. As estimates of male immune function,we used encapsulation rate and lytic activity. Encapsulationrate is a common challenging technique, which measures immuneresponse against multicellular parasites. Lytic activity isa monitoring technique, which measures immune response againstpathogens. Our results show that males with higher drummingrate had higher encapsulation rate. This suggests that femalesmight use drumming rate as a signal for choosing males withgood immunocompetence. Moreover, our results show that maleswith higher mobility had higher lytic activity. As females aremore likely to encounter those males that have higher mobility,this might also select males with better immune function. Ourresults suggest that the immunocompetence handicap might workalso among spiders, although we could not assess the causalityof the relationship between sexual selection and immune functionin this correlational study.  相似文献
