Three fluorescent pseudomonads isolated from rhizosphere/rhizoplane of crop plants showed in vitro antibiosis against seven fungal and two bacterial plant pathogens on iron-deficient KB medium. Seed bacterization of chick- pea (Cicer arientinum L.), egg plant (Solanum melongena L.), soybean (Glycine max Merr.) and tomato (Lycopersicon esculentum Mill.) with these organisms showed an increased seed germination, shoot height, root length, fresh weight, dry weight and yield. Seed bacterization with one of these strains, RB 8, reduced the number of chick-pea wilted plants in wilt-sick (Fusarium oxysporum f.sp. ciceris) soil. Addition of iron into the soil eliminated the disease suppression. The disease suppression and/or growth enhancement along with the positive root colonization by these organisms indicate their possible use as plant growth-promoting rhizobacteria (PGPR)/biocontrol agents against chick-pea wilt.  相似文献

Pseudomonas fluorescens strains PRS9 and GRS1 (wild type) were made mercury resistant PRS9Hg(r) (147 microM HgCl2) and GRS1Hg(r) (55 microM HgCl2), respectively, in King's medium by enrichment selection and their in situ root colonization studies were carried out. Mercury resistant mutant of PRS9 was stable and resulted in significant increase in root and shoot fresh weight (P < 0.05). Both the mutants are positive for indoleacetic acid (IAA), 'P' solubilization and siderophore production. PRS9, potent 'P' solubilizer, exhibited higher 'P' solubilization as compared to GRS1. After 2 weeks of inoculation, the population level of wild type PRS9 and its mercury resistant mutants has increased (50 fold). Mercury resistance has no adverse effect on the growth promoting properties of mutants besides being comparable in its morphological and physiological properties with their wild type counterpart. Furthermore, mercury resistant character facilitates rhizospheric competition and thus helpful for establishment of growth promoting strains where metal ions are either limiting and/or present at toxic level.  相似文献

AIMS: To identify and compare the relative diversity and distribution of genotypes of culturable fluorescent pseudomonads from soils. METHODS AND RESULTS: Analysis of 160 isolates from seven soil samples using randomly amplified polymorphism DNA methods revealed 53 genotypes, which were subsequently identified by their 16S ribosomal DNA sequences. Phylogenetic analyses of the 53 genotypes along with 43 fluorescent pseudomonad type strains separated the genotypes into 10 distinct clusters that included two phylogenetic groups that were not represented by previously described type strains. CONCLUSIONS: The diversity of genotypes that was obtained from the soil samples was highly variable among the different soils and appeared to be associated with different soil management practices that also influence plant yields. SIGNIFICANCE AND IMPACT OF THE STUDY: The identification and phylogenetic analysis of these genotypes offers opportunities for study of phenotypic traits that may be associated within taxonomically related groups of fluorescent pseudomonad species and how these groups vary in relation to soil management practices.  相似文献

Fluorescent pseudomonads that produce antibiotic 2,4-diacetylphloroglocinol (2,4-DAPG) are important group of PGRP that inhibit a broad spectrum of plant pathogenic fungi. Studying on genetic diversity of 2,4-diacetylphloroglucinol-producing fluorescent pseudomonads has been shown with special importance. The first step to investigate the genetic diversity of these bacteria is detecting of the genes required for the biosynthesis of this antibiotic. The objectives of the current study were detection of phlD gene within fluorescent pseudomonads by a PCR-based assay, and comparison of phenotypic and genotypic characteristics of fluorescent pseudomonads with proven biocontrol potential against some soil-borne phytopathogenic fungi. We used a collection of 47 fluorescent Pseudomonas spp. some with known biological control activity against Macrophomina phaseolina, Rhizoctonia solani, Phytophthora nicotianae var. parasitica, Pythium sp. and Fusarium sp. in vitro and the potential to produce known secondary metabolites such as, siderophore, HCN and protease. The results indicated that 66, 40.42, 63.82,48.94 and 27.65% of strains revealed antagonistic activity against R. solani, M. phaseolina, Pythium sp., P. nicotianae and Fusarium sp., respectively. Rhizoctonia solani recognized as the most vulnerable fungus. Among 47 strains, 76.59, 97.87 and 17% of strains produced protease, siderophore and HCN, respectively. We could detect phlD gene in strains P-5, P-32, P-47. Strain CHA0 was used as positive control for the detection this gene. Overall, there was no obvious link between the existence of phlD gene and inhibition of fungal growth or production of the antifungal metabolites in vitro. But in some strains such as CHA0 and P-5, we saw a link between the existence of phlD and antifungal activities. Studying on detection and diversity of phlD provides a fundamental knowledge for developing a rapid genetic screening system to identify a potential biocontrol strains.  相似文献

A total of 665 fluorescent pseudomonads, isolated from surface water and from different types of tap water, were classified into 22 groups defined by the results of the following 5 tests: hydrolysis of casein or gelatin, production of N2 from NO inf3 sup- , and growth on sucrose, ethanol and d-sorbitol, respectively. Differences in colonial morphology and in the degree of proteolysis revealed that these groups were inhomogeneous. A more detailed subdivision was achieved by adding the following characters: growth on l-arabinose, d-mannitol, meso-inositol, and adonitol, respectively, and hydrolysis of Tween-80. Repeating the tests for hydrolyzing enzymes, denitrification, and growth on the carbohydrates and alcohols with strains stored in the laboratory for 1 to 3 years revealed that most characters, except hydrolysis of Tween-80, denitrification, and growth on sucrose, were very stable.Forty-five biotypes of the fluorescent pseudomonads were defined, based on the results of the repeated tests and the results of tests on nine aromatic compounds. The observed changes of some characters in a number of isolates did not diminish the value of this classification, but indicated that close relationships exist between many biotypes. About half of the biotypes described in this paper are similar to those defined by Stanier, Palleroni and Doudoroff (1966) and by Doudoroff and Palleroni (1974a), confirming the wide-spread occurrence of well-definable biotypes of fluorescent pseudomonads.Representatives of some biotypes were most frequently isolated from surface water and from tap water prepared from surface water. Tap water prepared from anaerobic or aerobic ground water contained representatives of biotypes which were typical of these water types. Some pseudomonads were found to grow especially in filters. The observed relationships between origin of the fluorescent pseudomonads and their classification into biotypes as defined in this paper supported the presented classification.  相似文献

Three strains of Streptomyces (CAI-21, CAI-26 and MMA-32) were earlier reported by us as having potential for biocontrol of charcoal rot of sorghum, caused by Macrophomina phaseolina (Tassi) Goid., and plant growth promotion (PGP) of the plant. In the present investigation, the three Streptomyces were characterised for their physiological traits (tolerance of salinity, temperature, pH and resistance to antibiotics) and further evaluated in the field for their PGP of rice, grown by a system of rice intensification methods. All three Streptomyces were able to grow in NaCl concentrations of up to 12% (except MMA-32), at pH values between 5 and 13 and temperatures between 20 and 40°C. They were highly resistant to ampicillin and trimethoprim (>800 ppm), sensitive to chloramphenicol, kanamycin and nalidixic acid (50–100 ppm) and highly sensitive to streptomycin and tetracycline (5–25 ppm). When evaluated for their PGP activity on seedlings of rice, % germination and shoot and root lengths were significantly enhanced over the control. In the field, the Streptomyces strains significantly enhanced the panicle length, filled grain numbers and weight, panicle weight, 1000 seed weight, tiller numbers, total dry matter, root length (39–65%), root volume (13–30%), root dry weight (16–24%), grain yield (9–11%) and stover yield (11–22%) over the control. In the rhizosphere soil (0–15 cm from root) at harvest, the population of actinomycetes was significantly enhanced as was microbial biomass carbon (27–83%) and nitrogen (24–43%), dehydrogenase activity (34–152%), available P (13–34%) and N (30–53%) and % organic carbon (26–28%). This study further confirms that the selected Streptomyces have PGP properties.  相似文献

A total of 59 Pseudomonas isolates was obtained from 11 samples of diseased fleece taken from live sheep. All but four of the isolates could be assigned to one of nine Pseudomonas species, of which P. aeruginosa, P. alcaligenes, P. mendocina , and P. putida were the most common. P. aeruginosa was found in four of the fleece samples and, when present, appeared to predominate. Although several of the isolates of P. aeruginosa lacked the ability to produce pyocyanin (and some produced neither pyocyanin nor fluorescein), nearly all produced several virulence factors. Of the other pseudomonads, many produced proteinase, esterase, and catalase, several were able to grow at 42 degrees C and reduce nitrate, and some also produced lipase and hemolysin and, like P. aeruginosa, might serve to initiate (or sustain) the dermatitis frequently associated with fleece rot in sheep.  相似文献

Plant-microbe interactions in the rhizosphere are the determinants of plant health, productivity and soil fertility. Plant growth-promoting bacteria (PGPB) are bacteria that can enhance plant growth and protect plants from disease and abiotic stresses through a wide variety of mechanisms; those that establish close associations with plants, such as the endophytes, could be more successful in plant growth promotion. Several important bacterial characteristics, such as biological nitrogen fixation, phosphate solubilization, ACC deaminase activity, and production of siderophores and phytohormones, can be assessed as plant growth promotion (PGP) traits. Bacterial inoculants can contribute to increase agronomic efficiency by reducing production costs and environmental pollution, once the use of chemical fertilizers can be reduced or eliminated if the inoculants are efficient. For bacterial inoculants to obtain success in improving plant growth and productivity, several processes involved can influence the efficiency of inoculation, as for example the exudation by plant roots, the bacterial colonization in the roots, and soil health. This review presents an overview of the importance of soil-plant-microbe interactions to the development of efficient inoculants, once PGPB are extensively studied microorganisms, representing a very diverse group of easily accessible beneficial bacteria.  相似文献

Endophytic bacteria associated with the roots of coastal sand dune plants were isolated, taxonomically characterized, and tested for their plant growth-promoting activities. Ninety-one endophytic bacterial isolates were collected and assigned to 17 different genera of 6 major bacterial phyla based on partial 16S rDNA sequence analyses. Gammaproteobacteria represented the majority of the isolates (65.9%), and members of Pseudomonas constituted 49.5% of the total isolates. When testing for antagonism towards plant pathogenic fungi, 25 strains were antagonistic towards Rhizoctonia solani, 57 strains were antagonistic towards Pythium ultimum, 53 strains were antagonistic towards Fusarium oxysporum, and 41 strains were antagonistic towards Botrytis cinerea. Seven strains were shown to produce indole acetic acid (IAA), 33 to produce siderophores, 23 to produce protease, 37 to produce pectinase, and 38 to produce chitinase. The broadest spectra of activities were observed among the Pseudomonas strains, indicating outstanding plant growth-promoting potential. The isolates from C. kobomugi and M. sibirica also exhibited good plant growth-promoting potential. The correlations among individual plant growth-promoting activities were examined using phi coefficients, and the resulting data indicated that the production of protease, pectinase, chitinase, and siderophores was highly related.  相似文献

Many species of pseudomonads produce fluorescent siderophores involved in iron uptake. We have investigated the DNA homology between the siderophore synthesis genes of an opportunist animal pathogen, Pseudomonas aeruginosa, and three plant-associated species Pseudomonas syringae, Pseudomonas putida and Pseudomonas sp. B10. There is extensive homology between the DNA from the different species, consistent with the suggestion that the different siderophore synthesis genes have evolved from the same ancestral set of genes. The existence of DNA homology allowed us to clone some of the siderophore synthesis genes from P. aeruginosa, and genetic mapping indicates that the cloned DNA lies in a locus previously identified as being involved in siderophore production.  相似文献

The genetic diversity of plant growth-promoting rhizobacterial (PGPR) fluorescent pseudomonads associated with the sugarcane (Saccharum officinarum L.) rhizosphere was analyzed. Selected isolates were screened for plant growthpromoting properties including production of indole acetic acid, phosphate solubilization, denitrification ability, and production of antifungal metabolites. Furthermore, 16S rDNA sequence analysis was performed to identify and differentiate these isolates. Based on 16S rDNA sequence similarity, the isolates were designated as Pseudomonas plecoglossicida, P. fluorescens, P. libaniensis, and P. aeruginosa. Differentiation of isolates belonging to the same group was achieved through different genomic DNA fingerprinting techniques, including randomly amplified polymorphic DNA (RAPD), amplified ribosomal DNA restriction analysis (ARDRA), repetitive extragenic palindromic (REP), enterobacterial repetitive intergenic consensus (ERIC), and bacterial repetitive BOX elements (BOX) analyses. The genetic diversity observed among the isolates and rep-PCR-generated fingerprinting patterns revealed that PGPR fluorescent pseudomonads are associated with the rhizosphere of sugarcane and that P. plecoglossicida is a dominant species. The knowledge obtained herein regarding the genetic and functional diversity of fluorescent pseudomonads associated with the sugarcane rhizosphere is useful for understanding their ecological role and potential utilization in sustainable agriculture.  相似文献
