Experiments on the isolated ganglionic ring of the freshwater molluskPlanorbis corneus showed that more neurons (30%) in the ipsilateral mesocerebrum respond to stimulation of the left cerebral nerves than to stimulation of the opposite nerves (13%). A similar picture is observed for neurons of the right mesocerebrum, except that 11% of neurons respond to activation of the left cerebral nerves compared with 39% to stimulation of the ipsilateral right cerebral nerves. Ipsilateral connections of nerve of the visceral complex of ganglia are more clearly defined, as is exemplified by the left pallial nerve, during stimulation of which 68% of neurons in the ipsilateral mesocerebrum were activated, compared with 8% in the contralateral right mesocerebrum. Afferent fibers running in the visceral nerves cross at the level of the abdominal ganglion and not of the cerebral commissure, as might be expected from the structure of the ganglionic ring. The mesocerebral neurons themselves do not form synaptic connections between the mesocerebra, and excitation reaching one of them does not therefore induce any effects in the other.A. A. Summary The caudo-dorsal cells (CDC) in the cerebral ganglia of the pond snail Lymnaea stagnalis synthesize the 36-amino acid ovulation hormone (CDCH). We have used immuno-cytochemistry and in situ hybridization to reveal the localization of neurons and axons containing CDCH-like material.A monoclonal antibody to a fragment of CDCH and a cDNA probe encoding CDCH reacted with the CDC-system, with specific cell groups in the cerebral and pleural ganglia, and with individually occurring neurons throughout the central nervous system. The cells in the pleural ganglia, which were found in about 50% of the preparations studied, are considered as ectopic CDC. They are morphologically similar to CDC in their somal dimensions and axonal organization. By means of immuno-electron microscopy it was shown that these neurons contain secretory vesicles that are similar to those of the CDC. The neurons of the bilateral groups occurring in the cerebral ganglia in addition to the CDC are smaller and more intensely stained than the CDC. Axons of these small neurons probably have varicosities located on the CDC axons in the neuropil of the cerebral ganglion, indicating synaptic contacts. Two major axon tracts could be followed from (or toward) the neuropil of the cerebral ganglion. One tract runs from the cerebral gangion via the pleural and parietal ganglia to the visceral ganglion, giving off branches to most nerves emanating from these ganglia. The other tract could be traced through the cerebro-pedal connective to the pedal ganglia. Only in the right pedal ganglion was extensive axonal branching observed. The nerves emanating from this ganglion contained many more immunoreactive axons than those from the left pedal ganglion. A polyclonal antibody raised against the synthetic fragment of CDCH stained, in addition to the neurons and axons revealed with the monoclonal antibody and the cDNA probe, three other major groups of neurons. Two are located in the cerebral ganglion, the other in the left pedal ganglion.The present findings suggest the presence of a system of neurons that contain CDCH or CDCH-like peptides. The role this system may play in the control of egg-laying and egg-laying behaviour is discussed.  相似文献

R1 and L1 spikes are initiated in at least three different loci: (1) the peripheral axon in the foot, (2) the neuropil of the pleural and/or pedal ganglion, and (3) the neuropil of the abdominal ganglion. Furthermore, R1 and L1 probably have two different mechanisms for spike initiation: (1) sensory (foot), and (2) synaptic (abdominal and/or head ganglia). The different loci for spike initiation account for the bidirectional conduction of R1 and L1 spikes. As sensory (mechanoreceptor) neurons, R1 and L1 have peripheral axons in the ipsilateral posterior pedal nerve, show low threshold responses to stimulation of the ipsilateral posterior foot, they are rapidly adapting their responses do not decrease with repetition, and they are not blocked by high Mg++/low Ca++ solutions. As synaptically-driven neurons, R1 and L1 have widespread bilateral responsiveness, their responses decrease with repetition and their inputs are blocked with high Mg++/low Ca++ solutions. 运用一氧化氮合酶(NOS)组织化学方法研究了软体动物门双壳纲种类中国蛤蜊和腹足纲种类嫁Qi神经系统中NOS阳性细胞以及阳性纤维的分布。结果表明:在蛤蜊脑神经节腹内侧,每侧约有10-15个细胞呈强NOS阳性反应,其突起也呈强阳性反应,并经脑足神经节进入足神经节的中央纤维网中;足神经节内只有2个细胞呈弱阳性反应,其突起较短,进入足神经节中央纤维网中,但足神经节中,来自脑神经节阳性细胞和外周神经系统的纤维大多呈NOS阳性反应;脏神经节的前内侧部和后外侧部各有一个阳性细胞团,其突起分别进入后闭壳肌水管后外套膜神经和脑脏神经索。脏神经节背侧小细胞层以及联系两侧小细胞层的纤维也呈NOS阳性反应。嫁Qi中枢神经系统各神经节中没有发现NOS阳性胞体存在;脑神经节、足神经节、侧神经节以及脑—侧、脑—足、侧—脏连索中均有反应程度不同的NOS阳性纤维,这些纤维均源于外周神经。与已研究的软体动物比较,嫁Qi和前鳃亚纲其它种类一样,神经系统中NO作为信息分子可能主要存在于感觉神经。而中国蛤蜊的神经系统中一氧化氮作为信息分子则可能参与更广泛的神经调节过程。  相似文献

1. When one pedal ganglion is removed, snails first crawl using the unoperated side of the foot, but in 4-8 weeks the operated side exhibits an anterior-to-posterior gradient of recovery. 2. A ganglion bud bridges the site of the missing ganglion and axons project from intact central ganglia into the foot. 3. Rhythmic activity in right and left pedal nerve pairs is correlated during locomotion in the regenerated snails. 4. The oscillator in the remaining pedal ganglion drives bilaterally coordinated activity. It was shown earlier that some neurons in Helix CNS express the mRNA of the precursor of neuropeptide GFAD. Using the data obtained with the help of the whole-mount in situ hybridization, we tried to identify a group of such neurons, namely, the pedal caudo-ventral group and to determine their possible functions. The local extracellular stimulation of the pedal caudo-ventral group resulted in movements of reproductive organs in the semi-intact preparation and suppressed the activity of the modulatory neurons controlling feeding and defensive behavior. Application of synthetic peptide GFAD (10(-8) mol/l) also activated movements of the reproductive organs and suppressed the activity of the modulatory neuron controlling feeding behavior. Stimulation of the labial nerves resulted in suppression of caudo-ventral neurons with simultaneous activation of the modulatory neuron controlling feeding behavior. The obtained evidence suggests that the caudo-ventral neurons can regulate movements of the reproductive organs and also coordinate different functions in realization of the integral sexual behavior. This group of neurons inhibits the modulatory neurons controlling the forms of behavior incompatible with courtship, i.e., feeding and defensive forms.  相似文献

The distribution of substances related to the tetrapeptide APGWa was investigated in the central nervous system (CNS) and the reproductive apparatus of Helix aspersa by immunocytochemistry. In the CNS, APGWa immunoreactive neurons were detected in all ganglia except the pedal ganglia. Concerning the mesocerebrum of the cerebral ganglia, only neurons of the right mesocerebral lobe reacted positively to the antiserum. In the genital apparatus, positive neurons fibres were seen in the muscular layer of the penis and, in the gonad, an immunoreactive material occurred on the heads of some spermatozoa. On the basis of these observations and of previous electrophysiological studies, an implication of AGPWa-like peptides in the control of mating behaviour is proposed. The significance of the positive reaction of the spermatozoa remains unclear.  相似文献

Cobalt axonal iontophoresis and intracellular recordings were used to identify a cluster of several motor neurons innervating the penis-retractor muscle of Aplysia. Intracellularly recorded motor neuron action potentials elicited direct, one-for-one, constant latency excitatory junctional potentials (ejps) in individual muscle fibers. The axons of motor neurons could be recorded extracellularly in the penis-retractor nerve and stimulation of the nerve backfired the motor neurons. Perfusion of the ganglion, the muscle, or both with solutions of either increased Mg++/decreased Ca++ or increased Ca++ sea water indicated that the presumed motor neuron impaled was not a sensory cell and that interneurons were not intercalated in the pathway. Innervation of muscle fibers was found to be functionally polyneuronal and diffuse. The ejps were found to undergo marked facilitation with repetitive motor-neuron stimulation. The motor neurons were isolated in a distinct cluster in the right pedal ganglion. Their electrical activity was characterized by spontaneous irregular action potentials and a moderate input of postsynaptic potentials.  相似文献

We report the isolation of a cDNA clone encoding a neuropeptide precursor named preproGFAD from the central nervous system (CNS) of the snail Helix lucorum. Analysis of the expression of this gene shows that it is neurospecific and expressed in several groups of CNS neurons. Most notable is the expression of preproGFAD gene in the right mesocerebrum, where the neurons controlling mating behavior are located. The expression in this particular region is observed in adult animals but not in juvenile ones. The preprohormone is 108 amino acids long and contains a hydrophobic leader peptide and eight Lys-Arg recognition sites for endoproteolysis. The post-translational processing of the prohormone may lead to the generation of seven tetrapeptides, Gly-Phe-Ala-Asp-COOH (GFAD). This peptide has the same sequence as two previously isolated peptides from a related snail, Achatina fulica. The first of them (achatin-I) contains D-Phe; the second (achatin-II) is its L-Phe-containing stereoisomer. Injection of synthetic D-GFAD in nanomolar concentrations into intact animals caused an increase of the heartbeat rate and opening of the genital atrium. In preparations containing CNS with intact innervation of reproductive organs, bath application of D-GFAD caused extensive movements of the penis but not of other reproductive organs. Intracellular activation of individual neurons expressing the preproGFAD gene also elicited penis movements. D-GFAD also suppressed activity of neurons modulating feeding behavior. Our data therefore indicate that the preproGFAD gene encodes the precursor of a neuropeptide that participates in the regulation of male mating behavior. © 1998 John Wiley & Sons, Inc. Stimulation of the tail nerve (pedal nerve 9, p9) of the mollusk, Aplysia californica, causes release of serotonin (5-HT), which mediates sensitization of withdrawal responses. There are about 35 serotonin-immunoreactive (5-HT-ir) axons in p9, yet the cell bodies of these axons have not been located. Backfills of p9 were combined with 5-HT immunohistochemistry to locate the cell bodies of 5-HT-ir neurons with axons in p9. About 100 neurons had axons in p9. Only about ten neurons, however, were both backfilled and 5-HT-ir. These double-labeled neurons were all located in the pedal ganglion associated with p9, which had a total of approximately 42 5-HT-ir somata. The discrepancy between the number of 5-HT-ir axons and double-labeled cell bodies is not likely due to neurons having multiple axons in the nerve; intracellular fills suggest that these neurons do not branch before entering p9. Additionally, no evidence was found for peripheral 5-HT-ir cell bodies that project axons centrally through p9. Thus, approximately 70% of the neurons that give rise to the 5-HT-ir axons in tail nerve are unaccounted for, but likely to reside in the pedal ganglion.  相似文献

In the simultaneous hermaphrodite snail Lymnaea stagnalis, copulation as a male is controlled by neurons that send axons to the male copulatory organs via a single penis nerve. Using direct mass spectrometry of a penis nerve sample, we show that one of the molecular ions has a mass corresponding to GAPRFVamide, previously identified from the buccal ganglia, and named Lymnaea inhibitory peptide (LIP). The identity of this peptide is confirmed by partial peptide purification from the penis nerve, followed by post source decay mass spectrometry. We cloned the LIP-encoding cDNA, which predicts a prohormone that gives rise to five copies of LIP (now re-named LIP A), two other -FVamide peptides (LIPs B and C), and five structurally unrelated peptides. The LIP gene is expressed in neurons of the right cerebral ventral lobe that send their axons into the penis nerve. We show that the LIP A peptide is present in these neurons and in the penis nerve, and confirmed the presence of LIP B and C in the penis nerve by post source decay mass spectrometry. Finally, we demonstrate that LIP A, B and C inhibit the contractions of the penis retractor muscle, thereby implicating their role in male copulation behavior.  相似文献

Mating as a male in the simultaneous hermaphrodite freshwater snail, Lymnaea stagnalis, comprises a series of complex behaviors that are a prelude to copulation. Copulatory behavior itself is assumed to be controlled by various types of peptidergic neurons as well as serotonergic cells. Here we report the primary structure of two peptides that were extracted from a cluster of neurons that innervates the penial complex and that is located in the anterior lobe of the right cerebral ganglion. The sequences of the peptides were determined as: Ala-Pro-Gly-Trp-amide and Ser-Gly-Ser-Asp-Tyr-Cys-Glu-Thr-Leu-Lys-Glu-Val-Ala-Asp-Glu-Tyr-Ile-Leu- Leu- Ser-Tyr-Lys-Ile-Glu-Glu-Gln-Arg-Ala-Ala-Asp-Cys-Gly-Gly-Glu-Pro-Pro-Asn- Ser- Gln(amide), respectively. The longer peptide is a homodimer. Both peptides are processed from the recently identified Ala-Pro-Gly-Trp-amide prohormone, which is expressed in the neurons of the anterior lobe of the right cerebral ganglion. Ala-Pro-Gly-Trp-amide could also be recovered from the penial complex. This peptide, when applied in vitro, inhibits the contractions of the penis retractor muscles evoked by serotonin in a dose-dependent fashion.  相似文献

Abstract: Male copulatory behavior in the snail Lymnaea stagnalis is controlled by several types of peptidergic neurons, including a cluster of neurons in the ventral lobe of the right cerebral ganglion that show immunoreactivity to myomodulin-A of Aplysia and innervate the penis complex. We identified structurally myomodulin-A and three related peptides from Lymnaea and showed that they are present in a characteristic ratio in both the penis nerve and penis complex, suggesting that they are processed from a single precursor and transported from the ventral lobe to the penis complex. All four peptides decreased the relaxation time of electrically evoked contractions of the penis retractor muscle. However, their effects on the amplitude of contraction were different, ranging from no effect to an increase or a decrease in the amplitude. A mixture of the peptides in a ratio as determined by direct mass spectrometry of the penis nerve decreased the contraction time, the relaxation time, and the amplitude. These effects resemble those of one particular peptide in the mixture. The direct mass spectrometry determinations of the peptide profile in the penis nerve suggest that many more, as yet unidentified, neuropeptides are involved in modulation of muscle activities of the penis complex.  相似文献

Neurons having large cell bodies in the anterior dorsal median cluster in the metathoracic ganglion of the locust Schistocerca gregaria and the grasshopper Romalea microptera were studied by direct dye injection and reverse filling combined with elyctrical stimulation and recording. Eight, possibly nine, are of the unpaired type, with a T branch leading into left and right axons that leave the ganglion to terminate in muscles. Another six are probably paired, and may be interneurons. Five of the 8 or 9 unpaired neurons have one axonal branch in both N4 and N5, on both sides: the others have but a single branch. One of the nine, DUMETi, has left and right axons exclusively innervating the jumping muscles, and another, DUMDL, has left and right axons exclusively innervating the dorsal longitudinal flight muscles. Neither the locations, sizes or numbers of somata, nor their locations were as constant as is the case for ordinary ventral motoneurons.  相似文献

1. Central pathways for bag cell activation were identified by examining the frequency of spontaneous egg laying episodes in animals with central connective lesions. Bilateral lesions of the cerebropleural (but not the cerebropedal) connectives abolished spontaneous egg laying. In contrast, bilateral lesions of all cerebral ganglion peripheral nerves did not abolish spontaneous egg laying, suggesting that sensory input to the cerebral ganglion is not necessary for activating the bag cells. 2. Backfilling either pleuroabdominal connective labelled cell bodies in the cerebral ganglia (via the ipsilateral cerebropleural connective) that could project to the bag cells. Focal extracellular stimulation of these stained clusters activated the bag cells in isolated brains. 3. Central pathways for initiating egg laying behaviors were identified by selectively eliciting bag cell discharges in animals with central connective lesions. Bilateral lesions of the cerebropedal (but not the cerebropleural) connectives completely abolished elicited egg laying behaviors. 4. Pathways for motor output during rhythmic head and neck movements were identified by eliciting bag cell discharges in animals with peripheral nerve lesions. Bilateral lesions of the four tegumentary nerves in combination with the anterior pedal nerve completely abolished elicited egg laying behaviors, indicating that these nerves are necessary for normal motor output. A normal pattern of egg laying behaviors occurred when the four tegumentary and the anterior pedal nerves were left intact and all other pedal ganglion nerves were lesioned bilaterally, indicating that these nerves are also sufficient for normal motor output.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

The distribution of central axons of receptor cells of the eyes and the locations of neurons sending axons into the optic nerves were studied in the cerebral ganglia of the pulmonate mollusksLymnaea stagnalis andHelix sp. by the method of axonal transport of cobalt chloride injected via the optic nerves. Afferent fibers of these nerves form terminal ramifications (chiefly dorsally) in the middle part of the cerebral ganglion. Some of them pass through the commissure to the symmetrical region of the opposite cerebral ganglion. Neurons innervating the eyes are located in several regions of both cerebral ganglia. InLymnaea they are distributed near the point of entry of the optic nerve, in the region of the commissure, the mesocerebrum, and the posterior part of the ganglion. InHelix these neurons are found in the same regions except in the posterior part of the ganglion. In electrophysiological experiments responses of neurons in these parts of the cerebral ganglion to adequate stimulation of the eye were recorded. Differences in the character of responses and also the presence of neurons indifferent to stimulation of the eye are evidence of the functional heterogeneity of these areas. This suggests that morphologically separate visual centers do not exist in the cerebral ganglion of the Pulmonata. Neurons giving specific responses to stimulation of the eye and evidently belonging to different levels of the visual system (afferent or efferent divisions) are closely connected both with each other and with cells of other functional systems.A. The nervous system is ectodermal in origin. All nerve ganglia arise separately by proliferation and later delamination from the ectoderm, not by invagination. They become secondarily connected to one another by commissures and connectives developing as extensions from the peripheral layer of ganglionic nerve cells. Rudiments of the cerebral, pedal, pleural and intestinal (parietal) ganglia arise almost simultaneously at a relatively early stage (Stage V). The cerebral ganglia develop from the ectoderm of the head plates. Rudiments of the pedal and pleural ganglia are separate at their inception. They later fuse (Stage VI) to form a pleuro-pedal ganglionic mass on each side. The 2 intestinal ganglia are symmetrical at the beginning, but they soon lose their symmetry as a result of torsion. The right ganglion crosses to the left over the gut and persists as the supraintestinal ganglion. The left or subintestinal ganglion shifts to the right and forward, and fuses with the right pleural ganglion (Stage VIII), thus obscuring the chiastoneury. The paired buccal and single visceral (abdominal) ganglia start differentiating in Stage VII. The former develop from the ectodermal wall of the stomodaeum, while the visceral ganglion delaminates from the right wall of the visceral sac, then shifts to the left during torsion. The statocysts develop early (Stage V) from 2 ectodermal invaginations on either side of the rudimentary foot. They later separate from the overlying ectoderm and statoconi appear in their lumina. Contrary to earlier reports on related ampullariids, the osphradium proved to be ontogenetically older than the mantle and mantle cavity. It starts differentiating as a thickened ectodermal plate in the right wall of the visceral sac (Stage V). During torsion, it becomes engulfed in the mantle cavity and shifts to the left side, then is carried forward as the mantlegrow. The eyes develop late (Stage IX) as ectodermal invaginations which rapidly separate from the ectoderm to form closed vesicles. Their cells start differentiating before hatching to form the retina, in which pigment is deposited, and the inner cornea. The lens is secreted in the lumen of the eye and grows by addition of concentric layers of secretion.  相似文献

The immunostaining pattern for the peptide gastrin/cholecystokinin 8 (gastrin/CCK8) in the molluscan central nervous system has been considered. The changes in the distribution of gastrin/CCK8 immunoreactivity were analyzed in the neurons of different areas of the cerebral ganglia (mesocerebrum and metacerebrum) and in the buccal ganglia of the terrestrial snail Helix aspersa, during rest and active phases. During the period of inactivity and after one day of activity, there were several immunoreactive neurons in the mesocerebrum and metacerebrum of the snails and in the buccal ganglia, whereas after 7 days of activity the number of labeled neurons decreased. Data suggested a storage of gastrin/CCK8 in the neurons when behavioral activities in which the peptide is involved (such as feeding-related behavior) are suppressed or reduced. The different percentage of gastrin/CCK8 immunoreactive neurons in the left and right mesocerebrum provides information about the activities controlled by these neurons, which could be related to the adaptive evolution and plasticity of the brain in terrestrial pulmonates.  相似文献
