The existing treatments for nonalcoholic steatohepatitis (NASH) are not completely effective. The need for new alternatives without adverse effects and low cost, such as the flavonoid (−)-epicatechin (EC), which has beneficial effects on lipid metabolism and cardiovascular diseases, arises. The objective of this work was to analyze EC effects in the NASH induced by a Paigen-type diet (PD). Mice were administered with (1) normal chow and water, (2) PD + fructose 30% and (3) PD + fructose 30% + EC (1 mg/kg) per gavage during 9 weeks. At the end of each treatment, serum was collected for analysis of the biochemical profile and liver enzymes. The liver was collected for microscopic analysis and for the evaluation of the relative expression of Plin2, Plin3, CD36, adiponectin and UCP2. Results showed that EC reduced weight gain and decreased triglyceride (TG), low-density lipoprotein cholesterol, TG/high-density lipoprotein and the activity of liver enzymes (alanine aminotransferase and alkaline phosphatase), suggesting lower liver damage. The microscopic analysis showed less “balloonization” of the hepatocyte, small drops of lipids, less accumulation of collagen and infiltration of inflammatory cells as compared to nontreated group. Finally, a decrease in the expression of Plin 2 was observed. While CD36 decreased, adiponectin and UCP2 increased. In conclusion, EC improves the biochemical profile, the microscopic characteristics and protein expression. Therefore, it may be a possible therapeutic approach for NASH since it prevents the progression of the hepatic and metabolic damage induced by high-fat diets.  相似文献

The distribution of acetylcholinesterase(AChE)-positive structures in the developing rat spinal cord was studied with AChE-histochemistry.AChE-positive perikarya were first seen on embryonic day 14(E14) in the ventrolateral portion of the spinal cord.From that time onward.AChE=containing cells appeared gradually in the intermediate gray,dorsal horn and lateral spinal nucleus of the spinal cord in a ventral-to-dorsal,and lateral-to-medial order.No obvious rostral-to-caudal sequence was found.At birth,the distribution pattern of AChE-positive perikarya was basically similar to that in adults.After birth a dramatic increase in the AChE staining intensity extended from postnatal day 5(P5) to postnatal day 21(P21),In addition,two phases of transient AChE staining were observed in the external surface of the dorsal horn from embryonic day 15(E15) to embryonic day 21(E21) and in the marginal layer from embryonic day 21(E21) to postnatal day 14(P14),respectively.  相似文献

Summary Recent investigations into the role of peroxisomes in mammalian lipid metabolism have employed double isotope methodologies to examine the influence of peroxisomal agents on lipid turnover in the liver and extra hepatic tissues of the living animal.The action of these agents, all of which caused extensive changes in the flux of lipid metabolism in the treated animals, may best be viewed in relation to their effects on the common pathway of fatty acid oxidation in peroxisomes.Clofibrate, for example, acts through induction of peroxisomal oxidases and catalase; glycolate and ethanol through activation of this pathway; and aminotriazole and allylisopropylacetamide through inhibition of the catalase step in the sequence.The data from these studies provide support for the concept of an important contributory and regulatory role of peroxisomes in relation to the overall balance of lipid metabolism, and emphasize that these organelles play a significant role in the oxidation of common fatty acids, as well as a potential for the elimination of fatty acids that are poorly oxidized by mitochondria.Additionally, the data raise intriguing questions on the extension of peroxisomal influence to include phospholipid metabolism and the substantial degree of inter-tissue communication which is involved in the balance of lipid metabolism in the whole animal.  相似文献

A temperate peatland located in the St. Lawrence lowlands (Southern Québec) was studied in order to specify the past influence of natural and anthropogenic disturbances on its postglacial development. Seven profiles were analysed for pollen, macroremains, microfossils, macro- and microcharcoal, and loss-on-ignition. Fire incidence was important between 9300 and 8200 cal b.p. and during the last 300 years. The Early Holocene fires were contemporaneous with local waterlogged conditions, which prevented the destruction of peat. In contrast, the effects of the recent fires were enhanced by surface dryness resulting from the creation of at least six successive ditch networks over the entire peatland. The modification of the local hydrology and the regular occurrence of fire induced profound vegetation changes. The bryophytic carpet notably regressed in favour of a shrubby coverage, dominated by Chamaedaphne calyculata, Aronia melanocarpa and Betula populifolia. The results obtained suggest that rehabilitation of such an ecosystem is realistic, although relatively urgent in order to preserve the relict populations of endangered species, such as Woodwardia virginica and Dendroica palmarum. Aims

After decades of experience and strongly improved technology, service time of pacemaker generators is expected to increase. To test this hypothesis, we conducted a retrospective review of a large cohort of patients with a pacemaker.

Methods

We reviewed data collected between 1984 and 2006 in the first national Dutch pacemaker registry. This registry covered 96% of all generators implanted. We analysed the time of and reason for explantation of pacemaker generators. A 7-year follow-up interval after first implantation and following replacements was used to analyse changes over time.

Results

During 22 years of data collection, nearly 97,000 first pacemaker generators were implanted. A total of 27,937 (22.4%) generators were explanted within a mean of 6.3 (standard deviation 3.3) years. Reasons for approximately 60% of these explantations were ‘end of life’ of the pacemaker generator or elective system change. Complications or failures such as infections and recalls accounted for approximately 20% of the explantations. For the remaining 20%, the reasons for explantation had not been registered.

Conclusion

Despite progress in technology, a substantial proportion of pacemaker generators is explanted before its expected service time, with one in five generators being replaced due to technical failures, infections or other complications. Furthermore, the time interval between pacemaker implantation and explantation due to normal ‘end of life’ (battery EOL) decreased. Infections continue to rank highly as a cause for pacing system replacement, despite all current preventive measures.

Measurement error (ME) can lead to bias in the analysis of epidemiologic studies. Here a simulation study is described that is based on data from the French Uranium Miners’ Cohort and that was conducted to assess the effect of ME on the estimated excess relative risk (ERR) of lung cancer death associated with radon exposure. Starting from a scenario without any ME, data were generated containing successively Berkson or classical ME depending on time periods, to reflect changes in the measurement of exposure to radon (222Rn) and its decay products over time in this cohort. Results indicate that ME attenuated the level of association with radon exposure, with a negative bias percentage on the order of 60% on the ERR estimate. Sensitivity analyses showed the consequences of specific ME characteristics (type, size, structure, and distribution) on the ERR estimates. In the future, it appears important to correct for ME upon analyzing cohorts such as this one to decrease bias in estimates of the ERR of adverse events associated with exposure to ionizing radiation.  相似文献

The Loess Plateau is a special natural–cultural unit in northern China. Intensive land use in the past has had, and forestation and grass planting at present will have inevitable impacts on plant biodiversity in the Loess Plateau. Based on the analysis of floristic features within three sampling sites with different land use practices and analysis of species richness among different land use types, we discuss impacts of land use on species richness and floristic features in the Northern Loess Plateau. The results drawn from this case study are as follows: (1) It appears that forestation and grass planting have had a positive influence on the local species diversity, but they have contributed little to the native vegetation in terms of conserving its floristic features. (2) Caragana intermedia shrubland, Pinus tabulaeformis forestland, and natural grassland have made important contributions to supporting indigenous species and maintaining local plant biodiversity. (3) There is a significant positive correlation between land use diversity and species richness. These results imply that practicing biodiversity conservation in situ is feasible and the suitable choice for the Loess Plateau. Concrete measures for biodiversity conservation in the area can include setting up small nature reserves and diversifying land use patterns to maintain as much habitat as possible for native vegetation. The artificial Hippophae rhamnoides shrubland should not be further promoted, considering its negative influence on biodiversity conservation.  相似文献

Diel patterns in mobility and feeding behaviour of the larvae of the stream-dwelling trichopteran Sericostoma personatum larvae were investigated. Larvae fed at night on coarse particulate organic matter (CPOM) at the sediment surface. In the daytime they rested a few cm below the sediment surface, during which time their defaecation activity effected a release of fine particulate organic matter (FPOM) into the sediment. The amount of faeces (mean particle size = 0.1 ± 0.044 mm, x ± SD, n = 500) introduced into the sediment by the larvae, evaluated in two experiments, was 0.4–0.56 mg day–1. This amount did not differ significantly from the organic input resulting from bacterial activity (0.36–0.64 mg day–1). The presence of S. personatum larvae increased the sediment organic content by 42.9 mg (75.8 %) and 59.8 mg (185.6%) AFDW per 16 cm3 sediment over a 90-day period, as compared with control systems containing no larvae.  相似文献

The aim of this study is to analyse the impact of maternal age at first birth on low birth weight, preterm birth and low Apgar scores at one minute and at five minutes among live births delivered to primiparous Brazilian women in the city of S?o Paulo. Analyses were based on 73,820 birth records from the 1998 birth cohort. Logistic regression was used to assess the association between maternal age and each outcome variable, controlling for the following risk factors: delivery mode, plurality, sex, maternal education, number of prior losses, prenatal care, race, parity and community development. Maternal ages below 20 and above 30 years were significantly associated with the risks of low birth weight and preterm birth, but no association was found between maternal ages and Apgar score, with the exception that ages 15-19 reduced the odds of a low one-minute score. Even though this result seems to be inconsistent, low birth weight, preterm birth and low Apgar scores measure different dimensions of newborn well-being, and the association of each measure with maternal age is expected to diverge.  相似文献

Acetylcholinesterase (AChE) is one of the fastest enzymes known, even though the active site is buried inside the protein at the end of a 20-A deep narrow gorge. Among the great variety of crystal structures of this enzyme, both in the absence and presence of various ligands and proteins, the structure of a complex of AChE with the pseudo-irreversible inhibitor Mf268 is of particular interest, as it assists in the proposal of a back door for product clearance from the active site. Binding of Mf268 to AChE results in the carbamoylation of Ser200 and liberation of an eseroline-fragment as the leaving group. The crystal structure of the AChE-Mf268 complex, however, proves that eseroline has escaped from the enzyme, despite the fact that the Ser-bound inhibitor fragment blocks the gorge entrance. The existence of alternative routes other than through the gorge for product clearance has been postulated but is still controversially discussed in the literature, as an experimental proof for such a back door is still missing. We have used Monte Carlo-based molecular docking methods in order to examine possible alternative pathways that could allow eseroline to be released from the protein after being cleaved from the substrate by Ser200. Based on our results, a short channel at the bottom of the gorge seems to be the most probable back-door site, which begins at amino acid Trp84 and ends at the enzyme surface in a cavity close to amino acid Glu445. In this paper we present a mathematical model for estimating external mycelium growth of arbuscular mycorrhizal fungi and its effect on root uptake of phosphate (P). The model describes P transport in soil and P uptake by both root and fungi on the single root scale. We investigate differences in soil P depletion and overall P influx into a mycorrhizal root by assuming that different spatial regions of mycelia are active in P uptake. When all external hyphae contribute to P uptake, overall uptake is dominated by the fungus and the most effective growth pattern appears to be the one using a high level of anastomosis. The same is true when only the proportion of external hyphae assumed to be active contributes to uptake. When uptake is restricted to the tips, hyphal contribution to overall P uptake is less dominant; the most effective growth pattern appears to be the one characterised by nonlinear branching where branching stops at a given maximal hyphal tip density. Comparison to measured P depletion in the literature suggests that the scenario where active hyphae are contributing to P uptake is likely to fit the data best. These quantitative predictions promote our understanding of the mycorrhizal symbiosis and its role in plant P nutrition.  相似文献

Frequency of occurrence of fish species was monitored on a fortnightly basis in four fykes and a weir in the Oosterschelde estuary from 1979 through 1988. This was done in order to record changes in the fish fauna that may have occurred as a response to the construction of a storm-surge barrier in the mouth of the Oosterschelde (1984–1986) and the concomitant building of compartmentalization dams in the landward part. These compartmentalization dams reduced the freshwater inflow into the system. Principal component analysis using the annual averages in frequency of occurrence suggests a slight shift occurred in the fish community separating a cluster of years 1979–1984 from the cluster 1985–1988. Many of the changes in individual species could be attributed to fluctuations in yearclass strength or were part of changes occurring on a wider geographical scale. The only impact of the construction works seems to be the decrease in a number of anadromous fish. Fish traps seem to be useful as a monitoring tool for a number of species. The value of the data collected could be improved if catch size and length-frequency data are recorded.  相似文献

Purpose

Improving the environmental performances of viticulture practices is particularly complicated during unfavorable climatic conditions because of the need for fungal and other pest pressure and the variability of phytosanitary control methods available for winegrowers. However, winegrowers wish to improve the environmental performances of their practices regardless of climatic conditions. The objective of the study was to quantify the variability in environmental impacts due to climatic variations, and changes in viticulture practices induced by climatic variability.

Methods

Life cycle assessment was used to evaluate the environmental impacts of viticulture technical management routes (TMR) implemented on five plots in the Loire Valley for the Chenin Blanc grape variety. These TMRs are representative of the range of practices used in the region, including intensive conventional practices to moderate organic practices. The study covered two different years of production (2011 and 2013) under contrasting climatic conditions. The first year (2011) was hot and dry, which are favorable conditions for viticulture, while the second year (2013) was cold and humid, which are unfavorable conditions.

Results and discussion

During the unfavorable year, the number of phytosanitary treatments and soil maintenance interventions increased for most of the studied TMRs. This meant that the inter-annual variability in environmental impact was significant, depending on the type of TMR, with differences in impacts ranging from 19% to more than 40% between 2011 and 2013. Managing environmental impacts was found to be more difficult for the organic systems when climatic conditions are unfavorable (in 2013). The non-organic TMRs showed less variation in environmental impacts than the organic TMRs between the 2 years studied.

Conclusions

This study shows the importance of taking into account inter-annual variability in environmental assessments of viticulture systems. Indeed, winegrowers do not respond the same way to climate variability mainly with regard to plant protection and soil maintenance. Viticulture is very sensitive to inter-annual climate variations due to the parasitic pressure variability. The different production systems do not give to winegrowers the same possibilities for adapting practices to limit their impact on the environment.

Lineweaver-Burk plots revealed the presence of high (uptake 1) and low (uptake 2) affinity reuptake components for serotonin, dopamine and norepinephrine in the cerebral and in the visceral ganglia of M. edulis.2. The Km, affinity value for norepinephrine was 5 times greater in the visceral ganglia than in the cerebral ganglia, a finding which demonstrates ganglionic variations in norepinephrine uptake.3. The rank order of monoamine affinity for the uptake 1 system is serotonin > norepinephrine > dopamine in the cerebral ganglia, whereas in the visceral it is norepinephrine > serotonin > dopamine.4. Background

Recent studies have shown that the acetabular component frequently becomes deformed during press-fit insertion. The aim of this study was to explore the deformation of the Durom cup after implantation and to clarify the impact of deformation on wear and ion release of the Durom large head metal-on-metal (MOM) total hips in simulators.

Methods

Six Durom cups impacted into reamed acetabula of fresh cadavers were used as the experimental group and another 6 size-paired intact Durom cups constituted the control group. All 12 Durom MOM total hips were put through a 3 million cycle (MC) wear test in simulators.

Results

The 6 cups in the experimental group were all deformed, with a mean deformation of 41.78±8.86 µm. The average volumetric wear rate in the experimental group and in the control group in the first million cycle was 6.65±0.29 mm3/MC and 0.89±0.04 mm3/MC (t = 48.43, p = 0.000). The ion levels of Cr and Co in the experimental group were also higher than those in the control group before 2.0 MC. However there was no difference in the ion levels between 2.0 and 3.0 MC.

Conclusions

This finding implies that the non-modular acetabular component of Durom total hip prosthesis is likely to become deformed during press-fit insertion, and that the deformation will result in increased volumetric wear and increased ion release.

Clinical Relevance

This study was determined to explore the deformation of the Durom cup after implantation and to clarify the impact of deformation on wear and ion release of the prosthesis. Deformation of the cup after implantation increases the wear of MOM bearings and the resulting ion levels. The clinical use of the Durom large head prosthesis should be with great care.  相似文献

Summary The vibrational sensitivity of awake pigeons was tested with the heart rate conditioning method. This method proved to be a very sensitive instrument for the behavioural measurement of the pigeon's sensitivity to mechanical stimuli.Sine wave vibrational stimuli between 50 and 2000 Hz were applied to the 1st, 7th and 16th primary feathers of the wing. The resulting threshold curves were U- or V-shaped with 3 characteristics: (i) The frequency of best response was either 800 or 900 Hz. (ii) Within the broad frequency range of vibrational sensitivity (50 to 2000 Hz) the sensitivity was extremely high at the best frequency: threshold amplitudes lay between 0.5 and 0.09 m. (iii) The threshold curves showed very sharp tuning at best frequency with bandwidths between 0.3 and 0.7 octaves (measured at best frequency 10fold above threshold). Field trials were carried out from June to August in 2004 at Wuyishan (Wuyi Mountains), Fujian province, China, to determine the effects of habitat diversification on arthropod communities. Two Chinese cabbage, Brassica chinensis, field 1 (F1) and field 2 (F2) surrounded by diverse vegetable cultivars were selected, while a monoculture of Chinese cabbage served as the control field (CK). The results showed that: (i) when comparing insect abundance of each order between different habitats, significantly higher numbers of lepidopterous insects (39.76% from the each order) and lower densities of Hymenoptera (19.82%) were found in CK than in F1 and F2; (ii) compared with CK, F1 and F2 had a lower percentage of species richness and an abundance of herbivorous insects, but increased richness, abundance and biodiversity of predatory insects; (iii) no differences were observed in neutral insects' guild between different fields; and (iv) the dominant species for each guild depends on the habitat types and sampling dates. This study suggests that intercropping could conceivably be used in these habitats to increase the population of natural predators, thus achieving desirable and ecologically friendly results in vegetable fields.  相似文献

Purpose

Life cycle assessment (LCA) has been increasingly implemented in analyzing the environmental performance of buildings and construction projects. To assess the life cycle environmental performance, decision-makers may adopt the two life cycle impact assessment (LCIA) approaches, namely the midpoint and endpoint models. Any imprudent usage of the two approaches may affect the assessment results and thus lead to misleading findings. ReCiPe, a well-known work, includes a package of LCIA methods to provide assessments on both midpoint and endpoint levels. This study compares different potential LCIA results using the midpoint and endpoint approaches of ReCiPe based on the assessment of a commercial building in Hong Kong.

Methods

This paper examines 23 materials accounting for over 99 % of the environmental impacts of all the materials consumed in commercial buildings in Hong Kong. The midpoint and endpoint results are compared at the normalization level. A commercial building in Hong Kong is further studied to provide insights as a real case study. The ranking of impact categories and the contributions from various construction materials are examined for the commercial building. Influence due to the weighting factors is discussed.

Results and discussion

Normalization results of individual impact categories of the midpoint and endpoint approaches are consistent for the selected construction materials. The difference in the two approaches can be detected when several impact categories are considered. The ranking of materials is slightly different under the two approaches. The ranking of impact categories demonstrates completely different features. In the case study of a commercial building in Hong Kong, the contributions from subprocesses are different at the midpoint and endpoint. The weighting factors can determine not only the contributions of the damage categories to the total environment, but also the value of a single score.

Conclusions

In this research, the midpoint and endpoint approaches are compared using ReCiPe. Information is whittled down from the inventories to a single score. Midpoint results are comprehensive while endpoint results are concise. The endpoint approach which provides additional information of damage should be used as a supplementary to the midpoint model. When endpoint results are asked for, a LCIA method like ReCiPe that provides both the midpoint and endpoint analysis is recommended. This study can assist LCA designers to interpret the midpoint and endpoint results, in particular, for the assessment of commercial buildings in Hong Kong.  相似文献
