Microorganisms strongly influence and are required to generate the selective substrate that provides nutrients and support for fungal growth, and ultimately to induce mushroom fructification under controlled environmental conditions. In this work, the fungal and bacterial microbiota living in the different substrates employed in a commercial crop (compost phase I, II and III, flush 1 and 2, and casing material on day 1, 6 and 8 after compost casing and during flush 1 and 2) have been characterized along the different stages of cultivation by metataxonomic analysis (16S rRNA and ITS2), analysis of phospholipid fatty acid content (PLFAs) and RT-qPCR. Additionally, laccase activity and the content of lignin and complex carbohydrates in compost and casing have been quantified. The bacterial diversity in compost and casing increased throughout the crop cycle boosted by the connection of both substrates. As reflected by the PLFAs, the total living bacterial biomass appears to be negatively correlated with the mycelium of the crop. Agaricus bisporus was the dominant fungal species in colonized substrates, displacing the pre-eminent Ascomycota, accompanied by a sustained increase in laccase activity, which is considered to be a major product of protein synthesis during the mycelial growth of champignon. From phase II onwards, the metabolic machinery of the fungal crop degrades lignin and carbohydrates in compost, while these components are hardly degraded in casing, which reflects the minor role of the casing for nourishing the crop. The techniques employed in this study provide a holistic and detailed characterization of the changing microbial composition in commercial champignon substrates. The knowledge generated will contribute to improve compost formulations (selection of base materials) and accelerate compost production, for instance, through biotechnological interventions in the form of tailored biostimulants and to design environmentally sustainable bio-based casing materials.  相似文献

In the present paper an account is given of observations and experiments on the influence of the casing soil on the cropping of beds of the cultivated mushroom, Psalliota campestris. It is shown from the results of a survey of cropping at thirty-nine mushroom farms that when the compost and cultural conditions are suitable for cropping, the production of mushrooms may be largely determined by the soil used for casing. Casing soils are classified into four grades depending on the numbers of sporophores formed, their rate of growth and the type of mushroom produced. A laboratory method of assessing the cropping potentialities of soils is described, and examples are given of its use for investigating field factors affecting the selection of soils for casing and the nature of the soil factors affecting cropping. It is suggested that chemical substances or specific surfaces in soils may promote cropping, but that close contact with air is necessary for them to be effective. It is concluded that direct trial is the only reliable method of assessing the value of a material for casing and that considerable fundamental research is needed on casing soils.  相似文献

Three insect growth regulator insecticides and an entomopathogenic strain of Bacillus thuringiensis (GC327), products effective against the mushroom sciarid, Lycoriella auripila, were compared for their effect on mushroom cropping. Cyromazine and diflubenzuron were applied as a surface drench to mushroom compost before or after pasteurisation (at filling or spawning, respectively); admixed into casing material (at casing); or at a combination of these times. Hexaflumuron and GC327 were applied only at filling and casing, respectively. The presence of the target pest, L. auripila, had no effect on treatment trends, although it was accounted for in the analysis by use of a yield model. The trial was notable for the disparate effects that cyromazine and diflubenzuron casing treatments had on mushroom cropping. Cyromazine treatments that included application at casing resulted in increases in yield, compared to the untreated control whereas, with diflubenzuron, the opposite was true, with treatment at casing alone causing the greatest reduction overall (10%). GC327 applied at casing was also conspicuous for giving a 13% increase in yield. Treating the crop at casing with either cyromazine or GC327, therefore, resulted in a 15% or 24% increase in yield, respectively, compared to a similar treatment with diflubenzuron. Hexaflumuron applied at filling caused increases in yield compared to application of cyromazine at filling and cyromazine or diflubenzuron at spawning. There were also effects on crop timing. The addition of a cyromazine casing treatment normally caused the distinct flushes of mushrooms to be produced significantly earlier than the untreated control (up to 2.5 days), as did GC327. With diflubenzuron, the earlier flushes were only produced by those treatments that did not include a casing application. The combinations that included a casing treatment with diflubenzuron initially produced mushroom flushes earlier than the untreated control. They became either synchronous with the control or they were delayed. From the crop tolerance perspective, therefore, cyromazine and GC327 would be the sciarid control products of choice for a commercial mushroom grower.  相似文献

The failure of Benlate (50% benomyl) to control Mycogone perniciosa on mushroom farms where isolates of the organism were benomyl sensitive was investigated. A comparison was made of carbendazim levels, the major fungitoxic breakdown product of benomyl, in unsterilised casing, autoclaved casing and autoclaved casing mixed with a small proportion (1.7%) of casing previously treated with benomyl but from which all detectable levels of carbendazim had disappeared. Added benomyl was effective in controlling M. perniciosa in autoclaved casing, less effective in unamended casing and ineffective in the amended casing mixture. The recovery of carbendazim from these treatments was directly related to disease occurrence. Studies on farms where benomyl or other carbendazim generating fungicides had been used indicated that a lack of control of M. perniciosa was associated with the disappearance of fungicide from the casing before cropping began. This was not so on a farm where carbendazim generating fungicides had not been used before these investigations. Thiabendazole, a fungicide closely related to the carbendazim generators showed only a slight decline in concentration in casing capable of degrading benomyl. Bacteria isolated from the casing were examined in vitro for their ability to degrade benomyl. Five groups of bacteria were compared and three bacteria, Pseudomonas putida, Pseudomonas aeruginosa and an unidentified member of the Entero-bacteriaceae were most able to degrade benomyl.  相似文献

In this study, the possibility of using tea production waste as a new casing material in mushroom (Agaricus bisporus) cultivation was investigated. Some physical and chemical characteristics of tea waste, fermented tea waste and a mixture of tea waste with peat were compared with that of peat casing, as were their effects on yield. The highest yield was obtained from peat casing. Using tea production waste alone as a casing was not acceptable for assured yield when it was compared with peat. But, a mixture of tea production waste with peat in 1:1 (v:v) ratio increased the yield. There was no significant difference between the mushroom yields of tea production waste+peat and peat casing materials at the end of 30 and 40 days. High salt content, organic and inorganic compounds in casing materials caused reduction of yields. However, a high iron content in casing material gave a significant positive correlation with total yield at 40 days.  相似文献

Experiments were conducted to examine the location of oviposition by the phorid fly Megaselia halterata (Wood) (Diptera: Phoridae) in uncased and cased compost. Clearly, a majority of the gravid females choose oviposition sites directly after entering the top layer of the compost. In uncased compost, 60% of all adults emerged from the top of four compost layers of equal thickness. When the compost was covered by a casing layer which was still uncolonized by Agaricus bisporus, oviposition was further concentrated in the top compost layer. In this situation, 91% of all adults emerged from the top compost layer whereas only 1.5% emerged from the casing. When the casing layer was colonized by mushroom mycelium, 45% of all adults emerged from the casing layer and 53% emerged from the top compost layer. Further concentration in the top compost layer and the casing layer occurred as a result of upward migration of larvae. When compost was cased after oviposition, up to 43% of all adults emerged from the casing layer. We concluded that in the control of phorid infestations with insect pathogenic nematodes, applications in uncased compost can be restricted to the upper compost layer. When compost and casing are filled simultaneously, nematode applications in the casing layer only could be considered.  相似文献

In laboratory tests the distribution of insecticide in peat–chalk mushroom casing greatly affected the number of adult sciarids which emerged from larvae placed in 50 g samples. Only thionazin markedly reduced emergences when granules were clumped in the centre of the sample of casing. The slope of dosage mortality data for clumped granule treatments was less steep than for mixtures and gave estimated LD95 values of 16300 and 26.8 ppm respectively. Thus, the number of adults which emerged when granules amounting to 6.4 mg of chlorfenvinphos were clumped was 65% of those emerging when only 0.4 mg a.i. was mixed into 50 g casing. However, adults did not emerge when 3.2 mg of the same insecticide was mixed throughout the sample. Increased doses did not, therefore, compensate for inadequate mixing. With 0.2–0.4 mg a.i. chlorfenvinphos mixed through the casing, about three times as many sciarids emerged with the intact granules than when pulverized granules were used. This showed the lack of mobility of this chemical in peat-chalk mixtures and the need for it to be homogeneously incorporated in commercial casing. Pirimiphos-ethyl, diazinon and thionazin also gave improved kills when mixed into the casing rather than clumped. A χ2 test of the mortality data suggested a non-linear dosage-mortality relationship for treatments amounting to half the dish. However, LD95 estimates of 37.5 and 62.2 ppm were obtained for mixed and quadrant treatments. Thus, when 3.2 mg chlorfenvinphos was mixed into only half of the volume of casing, the number of adults emerging was only 6% of that from untreated casing. However, larval movement was negligible in untreated casing and this appeared to limit the effectiveness of chlorfenvinphos and pirimiphos-ethyl when they were not evenly distributed throughout the casing. Staphylococcal food poisoning associated with fermented sausages has been a recurring problem. By testing for thermonuclease by direct application of sausage casing disks on the surface of thermonuclease assay agar plates, possible Staphylococcus aureus growth in fermented sausages could be detected simply and rapidly. Koupal-Deibel deoxyribonucleic acid agar was somewhat superior to toluidine blue deoxyribonucleic acid agar for thermonuclease assay of fermented sausage casings. The sensitivity of the thermonuclease casing test was comparable to that of the extraction procedure, and the thermonuclease casing test results were in complete agreement with the thermonuclease assay results by the extraction procedure. The thermonuclease casing test offers government and industry laboratories a useful screening tool which could significantly reduce the problem of staphylococcal enterotoxins in fermented sausages.  相似文献

The host-searching ability of Neoaplectana feltiae Filipjev (= S. bibionis Bovien) (Nematoda: Steinernematidae) in response to larvae of a mushroom fly, Lycoriella solani Winn. was examined in a mushroom substrate. Individuals of L. solani were less attractive for the parasite than larvae of Galleria mellonella L. The nematode juveniles penetrated a 22 cm layer of casing mixture within 2–4 days. In the casing alone nematode effectiveness was better than in mushroom compost or in compost and casing together. In the casing mixture parasite dosages of 20 and 100 juveniles per cm2 led to 22% and 45% parasitization of L. solani respectively, while all G. mellonella larvae were parasitized at both dosages. The prevalence of nematode infection depended on the content of water in the mushroom substrate. The highest N. feltiae infectivity was observed, when the ratio of the dry casing weight to the weight of water content was 1: 2.5. The practical aspects of the observed phenomena, essential for the use of N. feltiae in the protection of commercial mushroom cultivation are discussed.  相似文献

The efficacy of different species of entomopathogenic nematodes was tested against larvae of the mushroom phorid Megaselia halterata (Diptera: Phoridae) and the mushroom sciarid Lycoriella auripila (Diptera: Sciaridae). Sciarid larvae originating from infestations in casing soil during colonization by Agaricus bisporus were almost completely controlled by applications of Steinernema feltiae to the casing soil. When larvae originated from infestations in freshly spawned compost, they could be controlled by compost applications halfway through spawnrunning and by very early casing treatments. The control of phorids in compost was maximally 31% when nematodes were mixed within the infested compost at a concentration of 3 106 nematodes/m2. Only slightly higher reduction rates were obtained at higher concentrations. The control of phorids was more promising in the infested casing layer, in which S. carpocapsae was most successful. At concentrations of 6 and 15 106 nematodes/m2 this species obtained reduction rates of 65 and 73% respectively when it was applied 3 days after the end of the infestation period. Different combinations of peat and chalk or lime sources with differing moisture contents were used to determine how specific physical and chemical properties of the casing soil relate to the growth and water relations of the mushroom. The peat types varied in terms of decomposition and extraction method; the lime addition varied in terms of rate and type (chalk or sugar beet lime). During the colonisation of the casing soil before fruiting, the extension growth rate of mushroom mycelium was most closely correlated (negatively) with the volumetric moisture content of the casing soil. Scanning electron microscopy showed that mycelium growing at a lower casing soil matric potential (Ψm) had a much finer and branched structure than mycelium growing at a higher Ψm. Across all the peat and lime source treatments, a relationship was found between the mean Ψm of the casing soil and mushroom yield, with an optimum Ψm of -7.9 to -9.4 kPa. Mushrooms are produced in ‘flushes’ at about 8-day intervals and during the development of each flush of mushrooms, there was a significant decrease in casing soil Ψm . This decrease (to below -40 kPa) was greatest in the second flush, which was the highest yielding. There were no relationships between mushroom yield and casing soil osmotic potential Ψπ within the range -93 to -154 kPa or any of the other chemical properties and water and air holding characteristics of the casing soils which were determined. Across different casing soil treatments, mushroom dry matter content was negatively correlated with mushroom yield and positively correlated with mushroom tissue osmotic potential. 卵孢小奥德蘑Oudemansiella raphanipes是一类依赖覆土栽培的食用菌。本文采用Illumina MiSeq高通量测序技术结合生态学及统计学分析方法研究卵孢小奥德蘑覆土真菌群落特征和影响因素。结果表明,与原始覆土内真菌群落相比,种植卵孢小奥德蘑后的覆土内真菌群落多样性显著下降;在组成方面,虽然优势菌门仍为子囊菌门Ascomycota,但其中的CladosporiumAcremoniumGibberellaEmericellopsis等属的相对丰度有所降低,而隶属于被孢霉门Mortierellomycota的StilbellaNeocosmosporaPseudogymnoascusPreussia等属的相对丰度呈增加趋势。卵孢小奥德蘑覆土真菌以腐生营养型为主,与原始覆土相比,种植卵孢小奥德蘑后的粪生营养型真菌和木腐菌呈增加的趋势;进一步研究发现pH、速效钾、速效磷和总氮等覆土土壤理化性质显著影响真菌群落的多样性和结构,pH和速效钾含量与真菌群落的丰富度和多样性呈显著负相关关系。研究结果为深入解析卵孢小奥德蘑覆土微生态机制奠定基础。  相似文献

覆土是影响双孢蘑菇产量、质量和出菇整齐度的重要因子,利用现代分子生态学的方法快速、准确地对不同覆土基质微生物结构特征进行检测,以进一步了解微生物群落与双孢蘑菇相互作用关系。测定了不同覆土的理化特性,应用PCR技术对不同覆土材料提取总DNA,扩增细菌16S rDNA和真菌28S rDNA,运用变性梯度凝胶电泳技术对PCR产物进行分析,研究双孢蘑菇不同覆土基质微生物结构特征。结果表明:不同处理的覆土材料微生物群落的基因具有多样性,其中细菌群落基因多样性存在差异,使用纯泥炭与粉碎稻草处理差异最大,相似性仅为62%;通过真菌28S rDNA变性梯度凝胶电泳结果显示,粉碎稻草处理多样性指数最高,达3.576,但随着泥炭比例的提高,覆土处理中真菌群落的多样性相对减少;栽培试验发现,双孢蘑菇子实体形成量、总产量可能与覆土中的真菌群落多样性呈负相关。  相似文献

The aim of this research was to show the mathematical data obtained through the correlations found between the physical and chemical characteristics of casing layers and the final mushrooms’ properties. For this purpose, 8 casing layers were used: soil, soil + peat moss, soil + black peat, soil + composted pine bark, soil + coconut fibre pith, soil + wood fibre, soil + composted vine shoots and, finally, the casing of La Rioja subjected to the ruffling practice. The conclusion that interplays in the fructification process with only the physical and chemical characteristics of casing are complicated was drawn. The mathematical data obtained in earliness could be explained in non-ruffled cultivation. The variability observed for the mushroom weight and the mushroom diameter variables could be explained in both ruffled and non-ruffled cultivations. Finally, the properties of the final quality of mushrooms were established by regression analysis.  相似文献

The internal pressure strength of tubing and casing often cannot satisfy the design requirements in high pressure, high temperature and high H2S gas wells. Also, the practical safety coefficient of some wells is lower than the design standard according to the current API 5C3 standard, which brings some perplexity to the design. The ISO 10400: 2007 provides the model which can calculate the burst strength of tubing and casing better than API 5C3 standard, but the calculation accuracy is not desirable because about 50 percent predictive values are remarkably higher than real burst values. So, for the sake of improving strength design of tubing and casing, this paper deduces the plastic limit pressure of tubing and casing under internal pressure by applying the twin shear unified strength theory. According to the research of the influence rule of yield-to-tensile strength ratio and mechanical properties on the burst strength of tubing and casing, the more precise calculation model of tubing-casing''s burst strength has been established with material hardening and intermediate principal stress. Numerical and experimental comparisons show that the new burst strength model is much closer to the real burst values than that of other models. The research results provide an important reference to optimize the tubing and casing design of deep and ultra-deep wells.  相似文献

Improved yield and biological efficiency (BE) of Pleurotus eryngii var. eryngii were achieved by supplementation of substrate with a commercial delayed-release nutrient and use of a casing overlay. Yield increases of 14% were achieved from cased substrates that were supplemented at time of casing with delayed-release nutrient (Remo’s). Use of a casing layer enhanced yield by 141% over non-cased substrates. When casing and substrate supplementation were combined, yield increased 179% over non-cased/non-supplemented substrates. Mushrooms harvested from cased substrates were darker in color and solids contents were lower compared to non-cased substrates. An additional break of mushrooms was harvested from non-cased “spent” substrate by fragmenting and re-supplementing the substrate prior to the application of a casing overlay. Three production methods were compared for their effect on mushroom yield: “standard”, “casing” and “casing after first break”. Casing of the substrate before first break (“casing” production method) resulted in the highest yield and biological efficiency.  相似文献

A detailed immunocytochemical and biochemical study of the location and expression of frustulins, a family of proteins associated with the frustules of diatoms, has been performed for Cylindrotheca fusiformis Reimann et Lewin, Navicula pelliculosa (Brébisson et Kützing) Hilse, and Navicula salinarum (Grunow) Husted. Immunocytochemistry revealed that frustulins, which share homologous epitopes but are different in size, were predominantly located in the organic casing. Based on timed immunolocalization experiments and Western blotting analysis of cell extracts obtained sequentially after repleting silicate to Si-synchronized cells, the continuous presence of the frustulins in the mature and parental organic casing of the examined species was observed. The frustulins of N. pelliculosa appeared as proteins similar to those of C. fusiformis, sharing identical epitopes. The extractions, however, yielded a markedly lower abundance of frustulins in N. pelliculosa. Peak concentrations of extracted frustulins appeared to be expressed just ahead of the silicification process in C. fusiformis, whereas the level of expression in N. pelliculosa increased along with maturation of the new valves. For N. salinarum, the presence of the frustulins could not be confirmed properly by Western blotting, most probably because of the small sample volumes, inefficient extraction, and a lower amount of homologous frustulins in the casing of this species. It is concluded that the frustulins of these species are not associated with the silicalemma of the newly formed silica deposition vesicles and therefore do not seem to be involved in the silicification process itself. Overall, the results imply a structural role of the frustulins in the casing of diatoms rather than a regulation of the silicification process.  相似文献

The entomopathogenic nematodes Steinernema feltiae (Biosys strain #27) and Heterorhabditis heliothidis were evaluated for the larval control of a mushroom-infesting sciarid, Lycoriella mali, and for the effects of these nematodes on mushroom (Agaricus bisporus) production. In a series of small-scale mushroom crops, infective-stage H. heliothidis and S. feltiae were applied to the mushroom casing surface in the irrigation water or incorporated into the casing material at densities ranging from 28 to 1120 and 11 to 1120 nematodes cm-2 of casing surface respectively. The mortality of L. mali larvae ranged from 52 to 100% for H. heliothidis and 38 to 100% for S. feltiae. Both nematode species reduced mycelial coverage on the casing surface at primordia initiation. Neither mushroom strain (off-white or white hybrid) or method of application (incorporation into or irrigation onto the casing surface) altered the effect on mycelial coverage. The nematodes's negative effect on mycelial growth confounded the benefit of fly control. At high nematode densities (up to 1120 nematodes cm-2), damage-free mushroom yields for the first week of harvest were less than those from the untreated control. However, at lower nematode densities, at or below 140 cm-2, the nematodes had less effect on mushroom growth, and consequently, damage-free mushroom yields for the first week of harvest were frequently greater than those from the untreated control. In the absence of flies, the first-week mushroom yield generally declined with increasing nematode densities for both white and off-white mushroom hybrids. After 4 weeks of harvest, accumulated mushroom yields had nearly recovered from the earlier decline.  相似文献

The mushroom growing bed consists of an underlying compost which is covered with casing (a peat and chalk or lime mixture) to induce fruit-body formation. The influences of the depths of the compost and casing layers on the mushroom texture, susceptibility to bruising, and ease of detachment from the growing bed surface were investigated. Textural properties were determined on cubes of cap tissue and detachment torque was determined by using an instrumented rotating suction cup which was lowered on to the caps. The proportion of mushrooms which detached at the base from the bed surface was recorded, since this is an important parameter for an automatic (robotic) harvesting system. Shallow casing resulted in mushrooms that were firmer and less likely to deform plastically, but that showed greater discolouration after a standard bruising treatment. Shallower casing also increased the proportion of mushrooms that detached at the base. Increasing the depth of compost produced firmer mushrooms, significantly increased the detachment torque and reduced the proportion of mushrooms detached at the base. The proportion of mushrooms that detached at the base correlated negatively with the plastic deformation of cap tissue. A positive correlation was found between mushroom dry matter content and tissue firmness.  相似文献

Two substrates, a non-composted grain spawn substrate and a traditional composted substrate, each covered with peat-based casing that contained varying amounts of activated carbon (AC) and each receiving different heat-treatment durations, were tested for Agaricus bisporus mushroom production. The amounts of AC were 0, 5, 10, 15, and 20% v/v, and the heat treatments were 0, 60, and 180 min at 121 °C and 103.4 kPa. Overall, the addition of AC up to 10–15% of casing for a grain spawn substrate increased mushroom yield. However, the addition of AC to the casing for compost substrates had no significant effect on yield, whereas heat-treating the casing increased yield. The onset of fruiting was retarded in grain spawn treatments not receiving AC with heat-treatment durations of 60 and 180 min, whereas this effect was not as apparent for the compost substrates. On average, mushroom yield was greater for the grain spawn substrate (366 g) than for compost substrate (287 g). For grain spawn substrate, the results show that the addition of AC ranging from 5% to 10% was adequate for maximum mushroom production.  相似文献
