The differentiation and obliteration of protophloem sieve elementsin leaves of the grass Aegilops comosa var. thessalica havebeen studied by electron microscopy. These elements differentiatesimilarly to metaphloem sieve elements of the same plant andother monocotyledons. Plasmalemma, smooth endoplasmic reticulum(ER), mitochondria, P-type plastids and sometimes nuclear remnantsconstitute the protoplasmic components at maturity, all areperipherally distributed. The differentiation of end walls intosieve plates and the presence of sieve areas on the lateralwalls indicate that protophloem sieve elements are componentsof sieve-tube. They may be functional for a brief period butsoon after their maturation they are compressed and finallyobliterated by the stretching of actively-growing surroundingcells. The protoplasmic components of mature elements degenerateand are destroyed during obliteration of the sieve elements. Junction complexes of unusual structure form between neighbouringsieve tubes in the secondary phloem of Eucalyptus species. Thick-walledribs support thin-walled ‘sieve areas’. In longitudinalsections the structures have a ‘concertina’- likeappearance. They are relatively large, up to 0.2 mm in length.Electron micrographs confirmed that the structures consistedof thin-walled areas perforated with pores, supported by muchthicker ribs. The structures provide a vast surface area fortransfer of metabolites between sieve tubes compared with thatof lateral wall sieve areas of other plants. Hydrolysis of parenchymacell walls occurs during the development of the junction complexes.The structures are only found when sieve tubes are in closeproximity and it is the redifferentiation and partitioning ofintervening parenchyma cells which result in junction complexformation. A survey for the presence of the structures in thephloem of other genera in the family Myrtaceae was made andthey were found in Tristania and Angophora but were not observedin Acmena and Metrosideros. The sieve elements of Pinus silvestris L., Sorbus aucupariaL., Vitis vinifera L., and Cucurbita pepo L. have been examinedelectron microscopically in ultra-thin section, and the structuresof the corresponding sieve areas or sieve plates have been describedand compared. In Pinus the sieve areas contain groups of connectingstrands which enter the wall from the lumen side as individualsbut coalesce within it in the median cavity. This cavity hasdeveloped by wall breakdown in the middle lamella and primarywall region and corresponds to the median nodule visible undera light microscope. Neither in this nor in the other speciesobserved is there any visible closing membrane. Structural differences between the four species are shown tosuggest that the major evolutionary trend in the evolution ofspecialized conducting strands has been the enlargement of theconnecting strands from groups of small separate strands toa smaller number of larger strands as the median cavity becomesenlarged to form a canal through the wall. The connecting strands appear invariably to be dense, highlyosmiophilic and continuous with the cytoplasmic surface of thecell. No signs of micropores or of other tubular structure inthe strands have been found. The structures thus revealed aremore nearly compatible with active transport of materials acrossthe sieve plate than they are with any purely physical mechanism.It is suggested that they are incompatible with any mass flowhypothesis.  相似文献

Light microscopic studies of the petioles of Lagenaria sicerariareveal that the external phloem of each bicollateral vascularbundle develops earlier than the internal phloem, and that thesieve elements of the external phloem are arranged in the outerand inner zones. Each sieve element of L. siceraria and Momordicacharantia is vertically associated with a maximum of six andtwo companion cells respectively. Discrete granular bodies seenin the cytoplasm of young sieve elements develop into globular,oval, or elongated slime bodies. Enlargement and fusion of slimebodies, and the subsequent dispersal of slime occur in the parietalcytoplasm. The dispersal of slime coincides with degradationof the nucleus and perforation of the pore sites. Before nucleardisorganization, the sieve-element nucleolus is extruded. Slimeafter its immediate dispersal appears amorphous and uniformlydistributed in the sieve elements. Plugs exhibit varying degreesof condensation of slime near the sieve plates. Certain maturesieve elements in the external phloem of L. siceraria have ovalbodies which we consider reaggregated or undispersed slime.Evidence has been obtained that a central cavity occurs in afew, almost mature, sieve elements wherein the cytoplasm includingthe slime is peripheral.  相似文献

From an analysis of the Brownian motion of particles in sieveelements of Heracleum mantegazzianum and Heracleum sphondylium,Barclay and Johnson have suggested that the in situ viscosityof sieve tube sap is four to six times higher than has previouslybeen assumed. In particular, they obtained a value for the sapviscosity of about 10–2 Pa s, which compares with a valueof 2 x 10–3 Pa s for a 20 per cent (w/v) sucrose solution.The present paper describes a reanalysis of their data. It isargued that Barclay & Johnson underestimated the Brownianmotion of sieve element particles and so overestimated the sapviscosity. An exact correction was not possible, but it is concludedthat the in situ viscosity of Heracleum sieve tube sap mustbe less than 3 x 10–3 Pa s, which corresponds to a sucroseconcentration of less than 29 per cent. Hence it may not beunreasonable to suppose that the viscosity of sieve tube sapis determined primarily by the concentration of sucrose, ashas been assumed in theoretical analyses of the Munch hypothesis.It is also concluded that the sieve tubes studied by Barclayand Johnson were not functional, in the sense that they didnot exhibit an axial bulk flow of sap. The study of isolated phloem in Heracleum has been extendedto intact functioning sieve tubes. Techniques of phloem dissectioncombined with Nomarski interference optics have been developedto permit useful visual observations, photomicrography, andciné photographs of sieve tubes which are apparentlynormal. In these preparations, plastids and organelles calledby us ‘marker particles’ are visible in rapid bouncingmotion, and the state of dispersal of these particles is relatedto the amount of damage done to the preparations. The movement of the marker particles and their subsequent fixationshows that they are apparently attached to or restrained byan invisible network in situ. The network is very sensitiveto disturbance and readily collapses around the sieve platesto form slime plugs upon damage to the sieve element. The markerparticles do not move through the cell nor across sieve platesin mature Heracleum. In young cells cyclosis is observable alongthe periphery and this suggests that a vacuole may then be present.In undamaged mature cells there was evidence neither of a vacuolenor of trans-cellular tubules of any size optically detectable. The motion of the particles was greater than Brownian movementand appeared to be under some physiological control. Their movementprobably indicated the presence of an operating transport phenomenon,either because solution was moving past them in the sieve tubeor because they were themselves attached to a contractile networkactively in pulsatory motion. Nearby companion and parenchymacells showed normal cytoplasmic streaming. Proposed mechanisms of translocation involving cytoplasmic streamingdo not seem to be applicable to the phloem of Heracleum. Themovement of the marker particles seemed to agree best with amechanism of ‘activated’ mass flow.  相似文献

In differentiating sieve elements of Aegilops comosa var. thessalicadictyosomes are abundant and they produce numerous smooth vesicles.Coated vesicles seem to bud from smooth ones. Since both kindsof vesicles appear both in the cytoplasm and in associationwith the plasmalemma, it is proposed that they move to and fusewith the plasmalemma transferring products for cell wall synthesis.During differentiation sub-plasmalemmal microtubules are initiallyscarce and randomly oriented but soon afterwards they becomenumerous and transversely oriented to the long axis. Cellulosemicrofibrils in the cell wall appear to run parallel to themicrotubules and the latter may regulate microfibril orientation. Root protophloem sieve elements develop wave-like wall thickenings,which are, during development, overlaid by microtubules perpendicularto the long axis. Just after maturation these thickenings progressivelybecome smooth and finally the walls appear uniform in thickness.The wave-like wall thickenings may function as stored wall material,utilized in later stages of development when wall material willbe needed and its synthesis will be impossible because of theabsence of a synthesizing mechanism in the highly degraded protoplastsof mature sieve elements. It is suggested that in this way thethickenings may enable root protophloem sieve elements to growand keep pace with the active clongation of the surroundingcells. The phloem of very young petioles of Nelumbo nucifera Gaertn.(Nelumbium speciosum Willd.) was studied with the light microscope.The elongated, mature sieve elements contain slime, plugs, strands,and numerous plastids. Some sieve elements remain nucleatedfor a brief period even after the sieve plates are well developed.The companion cells numbering 8–14 undergo disintegrationbefore the elongation of the ontogenetically related sieve elementis completed. They are uninucleate to begin with but later becomebinucleate and finally degenerate and obliterate. The variousstages in their ontogeny and disintegration are described. Ofthe very few specialized phloem parenchyma cells present, someare associated with sieve elements. They have slime body-likestructures, and plastid-like bodies which group together andeventually disintegrate.  相似文献

The willow aphid Tuberolachnus salignus feeds by inserting itsstylets into a single sieve element. If the insect is severedfrom its proboscis whilst feeding, sap from the sieve tube exudesfrom the cut end of the stylet canal and may continue for manyhours. By attaching a capillary tube the rate of exudation canbe measured and the sap analysed. Normally the rate of exudationwas about 1 mm.3 per h. This means a movement of water and sucroseof 100 cm./h. or 100 sieve elements per min. The sap was foundto contain between 5 and 15 per cent, sucrose, up to 0.4 percent, raffinose, no reducing sugars, and about 0.5 per cent,amino-acids. Girdles some distance above and below exuding stylets failedto stop exudation (Tables I and II). This indicates a rapidsealing of the cut ends of the sieve tube and a switch-overin source of supply from the leaves to storage cells in thestem. The use of isolated stem segments and even irrigated stripsof bark followed. Using these, the following points have beenestablished, (a) There is no polarity of movement (Table IV);(b) a minimal length of about 16 cm. of stem or 800–1,000sieve elements are needed to give the full exudation rate (Fig.5); (c) when the D.P.D. on the inside of the bark is raised(Figs. 6, 7, and 8), the rate of exudation declines, but theconcentration of the exudate rises and exudation continues,indicating a maintenance of turgor, even in face of a D.P.D.of 20 atm. or more. Secretion of sugar into the sieve tube continuesalbeit slowly even against a sieve tube concentration of 50per cent. A study has been made of the structure of the sieve tubes inthe phloem of seedlings of Cucurbita maxima kept in total darknessfor 2 or 3 days. All cytoplasmic components were found to beparietal in their distribution. The parietal system was closelyapplied to the cell membrane and appeared to be supported bya continuous framework of endoplasmic reticulum (ER) with whichP-protein was intimately associated. The ER-P-protein complexwas highly compact in some sieve elements and loosened to variousdegrees in others. The pores in the sieve plates were eitherunobstructed or occluded by components of the parietal complexin various ways, occlusion not always being accompanied by noticeabledisruption of the parietal system. In visibly undisturbed sievetubes, in which the ER-P-protein complex was in a highly compactstate, occlusion appeared accidental, arbitrary and withoutany alignment of the components present in the pores. It issuggested that the distribution of the cytoplasmic componentsin the parietal position represents a true-to-life conditionof the sieve tube, preserved due to control of the ‘surge’artefact to which transporting sieve tubes are susceptible.However, the organization of sieve tube probably changes withthe state of transport and the highly compact condition of theER-P-protein complex as well as unobstructed or arbitrarilyobstructed sieve plate pores represent a state of ‘rest’or low transport. The changes in structure of the endoplasmic reticulum (ER) andits associations with other cell components have been studiedin differentiating protophloem sieve elements of root tips ofTriticum aestivum. In the young sieve elements single ER cisternaebearing ribosomes are dispersed in the cytoplasm. As differentiationprogresses ER increases in amount while a small proportion ofit aggregates into stacks or becomes associated with the nuclearenvelope and the mitochondria. These modifications occur inthe last two sieve elements containing ribosomes and coincidewith most dramatic changes in the degenerating nucleus. Stacksconsist of relatively few ER cisternae and may be encounteredfree in the cytoplasm or applied to the nuclear envelope. Electron-densematerial accumulates between the contiguous cisternae of thestacks. ER-attached ribosomes persist even in nearly maturesieve elements, but their pattern of arrangement becomes changed.The structural evidence indicates that only a few highly degradedER elements are retained in fully mature sieve elements. Histochemical localization of ATPase was carried out on phloemtissues from vegetative and reproductive sinks of Ricinus communis,using lead precipitation procedures. Reaction products werelocalized mainly at the plasma membrane of the sieve elements,companion cells and phloem parenchyma cells. Activity was alsopresent in plasmodesmata, the tonoplast of companion cells anddispersed P-protein within the sieve element lumen. The resultsare discussed in relation to the possible involvement of a plasmamembrane ATPase in apoplastic and symplastic unloading fromthe phloem conducting tissues. Sieve tube elements occur in the rhizomes and petioles of Marsileaquadrifolia. These are either thick walled with compound sieveplates in oblique end walls or thin walled with simple sieveplates in transverse end walls. Vessels are restricted to themetaxylem in the roots where the phloem contains sieve cellsonly. The sieve pores are invariably callose lined and as inother pteridophytes, excepting the Lycopsida, refractive spherulesare ubiquitous in the sieve elements of Marsilea. The luminaof the protoxylem tracheary elements in the rhizomes and petiolesare occluded by tyloses but probably remain functional in theroots. Pericycle cells backing on to the root protoxylem armspossess wall ingrowths. Transfer cells are however absent fromthe vascular tissue of the rhizomes and leaves. It is suggestedthat their presence in the root pericycle is related to theretrieval of ions from the xylem sap which may be particularlycritical in water plants. The incidence of transfer cells incryptogams appears to be far more sporadic than in angiosperms.The root endodermis of Marsilea possesses a casparian stripand abundant vacuolar tannin deposits. The aerial shoots of Equisetum hyemale L. var. affine (Engelm.)A. A. Eat. were examined with the electron microscope as partof a continuing study of sieveelement development in the lowervascular plants. Young E. hyemale sieve elements are distinguishablefrom all other cell types within the vascular system by thepresence of refractive spherules, proteinaceous bodies whichdevelop within dilated portions of the endoplasmic reticulum(ER). Details of cell wall thickening differ between protophloemand metaphloem sieve elements. Following cell wall thickeningthe ER increases in quantity and aggregates into stacks. Shortlythereafter, nuclear degeneration is initiated. During the periodof nuclear degeneration some cytoplasmic components-dictyosomes,microtubules and ribosomes-degenerate and disappear, while organellessuch as mitochondria and plastids persist. The latter undergostructural modifications and become parietal in distribution.Eventually the massive quantities of ER are reduced, leavingthe lumen of the cell clear in appearance. At maturity the plasmalemma-linedsieve element contains a parietal network of tubular ER, aswell as mitochondria, plastids, and refractive sphemh At thistime many of the spherules are discharged into the region ofthe wall. Sieveelement pores occur in both lateral and end walls.At maturity many pores are traversed by large numbers of ERmembranes. The metaphloem sieve elements of the mid-internodalregions apparently are sieve-tube members. End walls of sieve elements of minor veins of the leaves ofBeta vulgaris L. do not contain the multi-perforate sieve plateswhich typically occur on the end walls of sieve-tube membersof major veins. Instead, both end and side walls of the sieveelements of minor veins contain scattered pores which may occursingly or in small numbers. These pores are similar to thosewhich are grouped in sieve plates of major veins in size, possessionof callose and plugs of filaments. In addition to these pores,there are tubular connections 0.1 µ in diameter throughcharacteristically thickened parts of the cell wall betweensieve cells and companion cells. Young sieve elements from petioles and rachises of Davalliafijiensis Hook were examined with an electron microscope. Evidencewas obtained that implicated both the endoplasmic reticulum(ER) and the Golgi apparatus in the formation of refractivespherules. Numerous connections were observed between smooth,tubular ER and peripheral tubules of the dictyosomes, indicatingthat these two cytoplasmic components are parts of a singleendomembrane system.
