The processing of freshwater crustaceans for ecophysiologicalstudies can be simplified considerably if species determination,selection and measurements of the specimens are performed afterfreezing and lyophilization. As the zooplankters do not changeshape during this process, reliable taxonomic determinationcan be performed with dried animals. Bean (Phaseolus vulgaris) landraces in northern Malawi were found to be heterogeneous for many qualitative and quantitative traits. An earlier 21- character evaluation of 25 lines in 15 landraces found significant differences in all characters among lines and among landraces. The present study has concentrated on two factors that are likely to be involved in the generation and maintenance of this heterogeneity: seed- handling practices and natural outcrossing. Seed- handling practices during harvest, storage, and marketing at the village, local market, and national levels were found to result in physical and usually nonselective mixing of many seed types. Experiments determined that outcrossing does occur at a low level among beans in Malawi and can result in the generation of many new seed phenotypes. A scenario was developed integrating seed- handling practices and outcrossing that lays a foundation for understanding the generation and maintenance of variability in these bean landraces. The process whereby landrace heterogeneity arises and the ways it is maintained, if understood, should assist plant breeders in the development of improved germplasm for subsistence farmers.  相似文献

The numbers of adult trout in Loch Leven were estimated in April each year 1968–71 by tagging fish caught by seine net, and estimating the proportion of the total stock tagged from examination of the angling catch during June-August. The vulnerability of tagged fish before June and after August was higher than that of untagged fish. Tag losses, estimated by double marking, were 2.15 % over a whole angling season. The reporting rate of tag recaptures varied within and between years from 0.43 to 0.71. Differential mortality of tagged and untagged fish was unimportant, except in two subsidiary experiments when fish were tagged in June and August, when handling losses reached 2.7%. With adjustment for these measured errors, the stock of trout beginning their third or more years in the loch in April, fell from 126 665 in 1968 to 52 337 in 1971.  相似文献

Over the last few decades several enzymatic processes to improve properties of wool fabrics like felting tendency, shrink resistance, dyeing ability and handling characteristics have been described. Previous investigations into the use of proteases to hydrolyse the cuticles at the surface of wool fibres, resulted in high strength and weight losses. Therefore restriction of the enzyme activity to the wool surface or control of enzyme diffusion to the cortex cells is required.

To change the diffusion behaviour of proteases in wool fibres, the soluble polymer PEG was covalently attached to a protease from Bacillus lentus. Modified enzymes with different molecular weights were compared. These modified enzymes retained up to 80% of their activity in the standard assay while hydrolysis of wool fibres was successfully restricted to cuticles, resulting in a 90% decrease in weight losses compared to non-modified enzymes.  相似文献

Flue-cured tobacco inoculated in the field with A. amstelodami, A. flavus, A. ochraceus, A. repens, A. ruber, and a species of Penicillium was rarely invaded by these fungi. Regardless of inoculum, the predominant fungi reisolated from green tissue were species of Alternaria and Cladosporium. After curing, A. repens, A. niger, and species of Alternaria and a species of Penicillium were the most commonly isolated fungi. The fungus used as inoculum was not the predominant fungus reisolated from green or cured tissue. Conditions during handling and storage prior to marketing probably determine when storage fungi become associated with the leaf and which species becomes predominant.  相似文献

The popularity of date palm (Phoenix dactylifera) fruit is increasing, therefore the demand for high-quality date palm fruit with less or no chemical treatment is the topic of interest for date producers and consumers. The quality of date palm fruit is much dependent on its postharvest handling and processing. For preventing the degradation and maintenance of the high quality of dates during the storage an appropriate harvest and post-harvest processes are required. The process should control the biotic and abiotic factors like insects, fungus, temperature, as well as handling and processing of dates. Therefore, in this work, we reviewed the literature related to the protection of date fruits during their post-harvest life. The commercially viable advance and updated techniques that can be used to avoid storage losses and problems while keeping fruit quality (nutritional, color, flavor, and texture) and microbial safety under optimal conditions are discussed.  相似文献

The purpose of this study was to assess chromium handling in non-insulin dependent diabetic patients (NIDDM) compared to healthy volunteers. Chromium handling was evaluated using fasting blood and second morning void urine samples from 93 NIDDM patients and 33 healthy volunteers. Significant differences in chromium homeostasis were seen between patients and controls. NIDDM patients had mean levels of plasma chromium around 33% lower and urine values almost 100% higher than those found in health. Healthy volunteers showed a significant negative correlation between fasting levels of plasma chromium and insulin. This was not evident in NIDDM patients. In the early years of onset of NIDDM, plasma chromium values were inversely correlated with plasma glucose. This was lost in patients with diabetes of more than 2 years duration. We suggest large losses of chromium over many years may exacerbate an already compromised chromium status in NIDDM patients and might contribute to the developing insulin resistance seen in patients with type 2 diabetes.  相似文献

Horticulture is a branch of Agricultural science where it is defined as the science and art of cultivating and handling fruits, vegetables, ornamental plants and several plants having unique medicinal and aromatic values. Horticultural crops provide farmers with high income and have good export quality, but they have a concern about postharvest losses. Hence, increasing productivity and decreasing post-harvest losses by using scientific studies and techniques like biotechnology and nanotechnology could be the simplest possible solution to the above-mentioned problems. Using nanotechnology which is having the characteristics of nanoparticles is proven to be very useful in science and technological applications. Nanotechnology-based formulations increase the product quality and the shelf life of horticultural products and provide multiple ways of inhibiting the growth and development of microorganisms. It is precisely a new edible packaging coverage (film) that controls the exchange of gases and prevents damage from harmful rays such as ultraviolet radiation to a greater extent. Increasing strength by using nano biosensors for labeling products is considered a fundamental process to automated control of storage products. Postharvest rotting of vegetables is recognized to be an oxidative reaction and microbial deterioration as well. This review will address all such nanotechnology-based advancements for minimizing post-harvest losses of horticultural crops and enhancing the socio-economical progress of growers in particular.  相似文献

To obtain correction factors for estimating root dry weight from washed samples and to test the efficiency of various procedures for storing root samples, dry matter losses were determined by simulating root washing methods with roots obtained from a nutrient culture. For sugar beet dry matter losses were higher than values previously found for wheat and ryegrass: about 30% for the procedure normally used and about 40% for samples pretreated with sodium pyrophosphate. The largest share of water-soluble sugars was lost from root samples within one day of storing roots. The N content of roots expressed on the basis of remaining dry matter rose first during handling of the root samples and decreased in samples stored for a longer period. In most cases no cell wall material (cellulose and lignin) is lost from the root samples; expressed on the basis of remaining dry weight the contents consequently rose.Communication no. 2 of the Dutch Programme on Soil Ecology of Arable Farming Systems  相似文献

Ripening of fleshy fruit: Molecular insight and the role of ethylene   总被引:1,自引:0,他引:1  
Development and ripening in fruit is a unique phase in the life cycle of higher plants which encompasses several stages progressively such as fruit development, its maturation, ripening and finally senescence. During ripening phase, several physiological and biochemical changes take place through differential expression of various genes that are developmentally regulated. Expression and/or suppression of these genes contribute to various changes in the fruit that make it visually attractive and edible. However, in fleshy fruit massive losses accrue during post harvest handling of the fruit which may run into billions of dollars worldwide. This encouraged scientists to look for various ways to save these losses. Genetic engineering appears to be the most promising and cost effective means to prevent these losses. Most fleshy fruit ripen in the presence of ethylene and once ripening has been initiated proceeds uncontrollably. Ethylene evokes several responses during ripening through a signaling cascade and thousands of genes participate which not only sets in ripening but also responsible for its spoilage. Slowing down post ripening process in fleshy fruit has been the major focus of ripening-related research. In this review article, various developments that have taken place in the last decade with respect to identifying and altering the function of ripening-related genes have been described. Role of ethylene and ethylene-responsive genes in ripening of fleshy fruit is also included. Taking clues from the studies in tomato as a model fruit, few case studies are reviewed.  相似文献

The ornamental fish trade is estimated to handle up to 1·5 billion fishes. Transportation and handling of fishes imposes a range of stressors that can result in mortality at rates of up to 73%. These rates vary hugely, however, and can be as low as 2%, because they are generally estimated rather than based on experimental work. Given the numbers of ornamental fishes traded, any of the estimated mortality rates potentially incur significant financial losses and serious welfare issues. Industry bodies, such as the Ornamental Aquatic Trade Association (OATA), have established standards and codes of best practice for handling fishes, but little scientific research has been conducted to understand the links between stress, health and welfare in ornamental species. In aquaculture, many of the same stressors occur as those in the ornamental trade, including poor water quality, handling, transportation, confinement, poor social and physical environment and disease and in this sector directed research and some resulting interventions have resulted in improved welfare standards. This review considers the concept of welfare in fishes and evaluates reported rates of mortality in the ornamental trade. It assesses how the stress response can be quantified and used as a welfare indicator in fishes. It then analyses whether lessons from aquaculture can be usefully applied to the ornamental fish industry to improve welfare. Finally, this analysis is used to suggest how future research might be directed to help improve welfare in the ornamental trade.  相似文献

Many measures have been proposed to mitigate gaseous emissions and other nutrient losses from agroecosystems, which can have large detrimental effects for the quality of soils, water and air, and contribute to eutrophication and global warming. Due to complexities in farm management, biological interactions and emission measurements, most experiments focus on analysis of short-term effects of isolated mitigation practices. Here we present a model that allows simulating long-term effects at the whole-farm level of combined measures related to grassland management, animal housing and manure handling after excretion, during storage and after field application. The model describes the dynamics of pools of organic carbon and nitrogen (N), and of inorganic N, as affected by farm management in grassland-based dairy systems. We assessed the long-term effects of delayed grass mowing, housing type (cubicle and sloping floor barns, resulting in production of slurry and solid cattle manure, respectively), manure additives, contrasting manure storage methods and irrigation after application of covered manure. Simulations demonstrated that individually applied practices often result in compensatory loss pathways. For instance, methods to reduce ammonia emissions during storage like roofing or covering of manure led to larger losses through ammonia volatilization, nitrate leaching or denitrification after application, unless extra measures like irrigation were used. A strategy of combined management practices of delayed mowing and fertilization with solid cattle manure that is treated with zeolite, stored under an impermeable sheet and irrigated after application was effective to increase soil carbon stocks, increase feed self-sufficiency and reduce losses by ammonia volatilization and soil N losses. Although long-term datasets (>25 years) of farm nutrient dynamics and loss flows are not available to validate the model, the model is firmly based on knowledge of processes and measured effects of individual practices, and allows the integrated exploration of effective emission mitigation strategies.  相似文献

A modified litter bag design and handling procedure were tested to establish whether these reduced the exaggerated fragmentation losses that occur with standard litter bags. The modified design was compared with standard coarse (5 mm) and fine (0.25 mm) mesh litter bags using Phragmitesleaf litter. All were positioned in a section of a reedbed subject to water level management but negligible water flow. Breakdown rates were significantly reduced with the modified design but these were still significantly greater than those in the fine mesh bags. Owing to the extended period, results were influenced by invertebrates colonising the fine mesh bags. The significance of bag design and invertebrate colonisation are discussed.  相似文献

Favourable economics of handling and application of manure are of fundamental importance to encourage the implementation of emission-reducing application techniques. The economics of manure application depend on the costs of the equipment and the time to carry out the field operation. In this study the costs of application techniques designed to reduce ammonia losses were assessed and compared with the costs of conventional broadcast spreading across a range of farm characteristics. A model was developed to calculate the costs and time requirements of manure application. Data on factors affecting the costs were used from different countries in Europe. The calculations showed that for a range of farm characteristics with a manure production of 1000-3000 m(3)y(-1), the costs of manure application by trailing hose, trailing foot, shallow injector and arable land injector were approximately 2 m(-3) higher than for broadcast spreading. The cost difference between broadcast spreading and the other application techniques decreased with farm size. The average additional costs of manure application by a trailing foot or a shallow injector decreased by 15% on small extensive farms to more than 50% on intensive farms, when the fertiliser value of the nitrogen was taken into account. The field application itself took less than 50% of the operating time in the process of the manure handling and application. With an increasing application rate, the relative contribution of the time for field application decreased.  相似文献

Nine recombinant FixL heme domains from Bradyrhizobium japonicum previously were shown to exhibit mass instability independent of many environmental factors (J.D. Satterlee, C. Suquet, A. Bidwai, J. Erman, L. Schwall, R. Jimenez, Biochemistry 47 (2008) 1540-1553). Two of those recombinant proteins were produced in remote laboratories. Mass losses begin appearing at completion of isolation and comprise a substantial proportion of samples within 1-3 days of storage and handling. Thus, degradation occurs during the time frame of experiments and crystallization. Detailed understanding of this instability is desired in order to formulate stable heme-PAS sensor domains for experimentation and for a mechanistic interpretation. However, mass spectra of the full length heme-PAS domain, BjFixLH140-270, are complex by 1-3 days following isolation due to broad features and a high density of overlapping peaks, so that individual peak assignments are at present ambiguous. This stymies direct, quantitative interpretation of the source of the observed mass losses. To solve this dilemma amino-terminal primary sequencing and MALDI-TOF (Matrix Assisted Laser Desorption Ionization-Time of Flight) mass spectrometry monitoring of three terminal variants of BjFixLH140-270 have been achieved. The working hypothesis, that the experimentally observed mass losses originate in the PAS protein sequence termini, has been substantiated. This establishes a basis for interpreting the more complex results from aging full length BjFixLH140-270.  相似文献

Controlled atmosphere storage of winter white cabbage   总被引:1,自引:0,他引:1  
In four laboratory experiments and five commercial scale trials carried out between 1977 and 1979, five cvs of winter white cabbage were stored at 0–2 °C for 29–43 wk in controlled atmospheres (C A) consisting of c. 5–6% CO2, 3% O2 and the remainder N., or in air. The incidence of rots caused by Botrytis cinerea was reduced and the incidence of ‘pepper-spotting’ (a non-microbial leaf necrosis disorder) was reduced or eliminated by storage in CA. Storage and trimming weight losses were lower and the recoveries of trimmed cabbage suitable for marketing or processing up to 22% higher in cabbage stored in CA than in those stored in air. Sensory assessments of four cvs tested indicated that CA-stored cabbages retained their green colour, crisp texture and fresh flavour better than the corresponding air-stored samples.  相似文献

Background

Iron deficiency anemia remains a major global health problem. Higher iron demands provide the potential for a targeted preventative approach before anemia develops. The primary study objective was to develop and validate a metric that stratifies recommended dietary iron intake to compensate for patient-specific non-menstrual hemorrhagic losses. The secondary objective was to examine whether iron deficiency can be attributed to under-replacement of epistaxis (nosebleed) hemorrhagic iron losses in hereditary hemorrhagic telangiectasia (HHT).

Methodology/Principal Findings

The hemorrhage adjusted iron requirement (HAIR) sums the recommended dietary allowance, and iron required to replace additional quantified hemorrhagic losses, based on the pre-menopausal increment to compensate for menstrual losses (formula provided). In a study population of 50 HHT patients completing concurrent dietary and nosebleed questionnaires, 43/50 (86%) met their recommended dietary allowance, but only 10/50 (20%) met their HAIR. Higher HAIR was a powerful predictor of lower hemoglobin (p = 0.009), lower mean corpuscular hemoglobin content (p<0.001), lower log-transformed serum iron (p = 0.009), and higher log-transformed red cell distribution width (p<0.001). There was no evidence of generalised abnormalities in iron handling Ferritin and ferritin2 explained 60% of the hepcidin variance (p<0.001), and the mean hepcidinferritin ratio was similar to reported controls. Iron supplement use increased the proportion of individuals meeting their HAIR, and blunted associations between HAIR and hematinic indices. Once adjusted for supplement use however, reciprocal relationships between HAIR and hemoglobin/serum iron persisted. Of 568 individuals using iron tablets, most reported problems completing the course. For patients with hereditary hemorrhagic telangiectasia, persistent anemia was reported three-times more frequently if iron tablets caused diarrhea or needed to be stopped.

Conclusions/significance

HAIR values, providing an indication of individuals’ iron requirements, may be a useful tool in prevention, assessment and management of iron deficiency. Iron deficiency in HHT can be explained by under-replacement of nosebleed hemorrhagic iron losses.  相似文献

Mullerian inhibiting substance (MIS), a large glycoprotein secreted by the fetal and neonatal testis, is responsible for regression of the Mullerian ducts in the male embryo. This fetal growth regulator has been purified more than 2000-fold from crude testicular incubation medium following fractionation on a triazinyl dye affinity support. A high yield of 60% recovered activity was achieved in the absence of exogenous carrier protein by stabilizing MIS with 2-mercaptoethanol, EDTA, and Nonidet-P40 and eliminating losses in the handling and concentration of MIS fractions. Although affinity elution with nucleotides has proved successful in other systems, MIS could not be eluted with ATP, GTP, or AMP, with or without divalent metal ions. Nucleotide elution, however, does remove contaminating proteins prior to MIS recovery with high ionic strength. The 2000-fold-purified MIS fraction, although not homogeneous, shows a reduction-sensitive band after SDS-gel electrophoresis that has been proposed to be the MIS dimer.  相似文献
