This study focused on the adaptation of Aspergillus niger tolerating high concentration of heavy metals for bioleaching of fly ash. The Plackett-Burman design indicated that Al and Fe inhibited the growth of A. niger (AS 3.879 and AS 3.40) significantly. The single metal (Al and Fe) and multi-metals adapted AS 3.879 strain tolerated up to 3500 mg/L Al, 700 mg/L Fe, and 3208.1mg/L multi-metals, respectively. The order of metal extraction yield in two-step bioleaching of 60 and 70 g/L fly ash using Al adapted, multi-metals adapted and un-adapted AS 3.879 strains was as follows: multi-metals adapted>Al adapted>un-adapted. The multi-metals adapted strain grew with up to 70 g/L fly ash and secreted 256 mmol/L organic acids after 288 h, where 87.4% Cd, 64.8% Mn, 49.4% Zn and 45.9% Pb were dissolved. The extracted metals in TCLP test of the bioleached fly ash by multi-metals adapted strain were under the regulated levels in China.  相似文献

The use of the fungus Aspergillus niger for the bioleaching of heavy metals from spent catalyst was investigated, with fluid catalytic cracking (FCC) catalyst as a model. Bioleaching was examined in batch cultures with the spent catalysts at various pulp densities (1-12%). Chemical leaching was also performed using mineral acids (sulphuric and nitric acids) and organic acids (citric, oxalic and gluconic acids), as well as a mixture of organic acids at the same concentrations as that biogenically produced. It was shown that bioleaching realised higher metal extraction than chemical leaching, with A. niger mobilizing Ni (9%), Fe (23%), Al (30%), V (36%) and Sb (64%) at 1% pulp density. Extraction efficiency generally decreased with increased pulp density. Compared with abiotic controls, bioleaching gave rise to higher metal extractions than leaching using fresh medium and cell-free spent medium. pH decreased during bioleaching, but remained relatively constant in both leaching using fresh medium and cell-free spent medium, thus indicating that the fungus played a role in effecting metal extraction from the spent catalyst.  相似文献

A spent refinery processing catalyst was physically and chemically characterized, and subjected to one-step and two-step bioleaching processes using Aspergillus niger. During bioleaching of the spent catalysts of various particle sizes ("as received", 100-150 microm, <37 microm, and x =2.97 (average) microm) and pulp densities, the biomass dry weight and pH were determined. The corresponding leach liquor was analysed for excreted organic acids along with heavy metal values extracted from the catalyst. Chemical characterization of the spent catalyst confirmed the presence of heavy metal including Al (33.3%), Ni (6.09%) and Mo (13.72%). In general, the presence of the spent catalyst caused a decrease in the biomass yield and an increase in oxalic acid secretion by A. niger. The increase in oxalic acid secretion with a decrease in the catalyst particle size (up to <37 microm) led to corresponding increase in the extraction of metal values. The highest extraction of metal values from the spent catalyst (at 1% w/v pulp density and particle size <37 microm) were found to be 54.5% Al, 58.2% Ni and 82.3% Mo in 60 days of bioleaching. Oxalic acid secretion by A. niger in the presence of the spent catalyst was stimulated using 2-[N-Morpholino]ethanesulfonic acid (MES) buffer (pH 6), which resulted in comparable metal extraction (58% Al, 62.8% Ni and 78.9% Mo) in half the time required by the fungus in the absence of the buffer. Spent medium of A. niger grown in the absence and in the presence of MES buffer were found to leach almost similar amounts of Al and Ni, except Mo for which the spent medium of buffered culture was significantly more effective than the non-buffered culture. Overall, this study shows the possible use of bioleaching for the extraction of metal resources from spent catalysts. It also demonstrated the advantages of buffer-stimulated excretion of organic acids by A. niger in bioleaching of the spent catalyst.  相似文献

This article reports the role of indigenous iron-oxidizing microorganisms in bioleaching of heavy metals from anaerobically digested sewage sludge in presence of toxic dyes namely, methylene blue (MB), Remazol black B (RBB) and mixture of both the dyes (DM). To achieve this goal, different concentrations of dyes (0, 500, 10,000, 15,000 and 25,000 mg/L) were added to the sewage sludge (initial pH ~ 7) and subsequently, the solubilization of heavy metals (Cu, Ni, Zn and Cr) was measured at time intervals of 48 h for 16 days. The results showed that an increase in dye concentration adversely affected the population of indigenous iron-oxidizing microorganisms, thereby decreased the bioleaching of metals. The metal solubilization from sludge is more adversely affected in presence of RBB and DM than MB. In majority of cases, the maximum metal solubilization was recorded at dye concentration of 5,000 mg/L. Two factor analyses (ANOVA) suggests the impact of both factors: dye concentration and dye type on metal leaching rate in sludge system.  相似文献

Bacillus megaterium strain MNSH1-9K-1, isolated from a high-metal content site in Guanajuato, Mexico, has the intrinsic capacity to remove vanadium (V) and nickel (Ni) from a petrochemical spent catalyst, and counteract the toxic effects produced in the cell due to the presence of oxidative stress. Since knowledge of the molecular components involved in the microbial resistance to spent catalysts is scarce, this study aimed to identify the proteins potentially involved in the enhanced resistance of a B. megaterium strain, during the removal of metals contained in a spent catalyst. Thus, the current research uses a proteomic approach to investigate and evidence the differences in the molecular resistance mechanisms of two B. megaterium strains, one isolated from a mining site and a wild type strain, when both are exposed to a spent catalyst. In addition, we studied their ability to eliminate nickel (Ni), vanadium (V), aluminum (Al) and molybdenum (Mo). The data presented here may contribute to the knowledge of the molecular mechanisms involved in the resistance of B. megaterium to high metal content wastes, as well as its potential utilization for the recovery of valuable industrial metals.  相似文献

Aluminium toxicity may be an important factor in the decline in vitality of many forest trees and the associated ectomycorrhizal fungal flora. In this study, comparative in vivo 31P NMR investigations on Al-adapted and non-Aladapted fungus of Suillus bovinus in pure culture have produced interesting new data. With respect to intracellular compartments, 31P NMR spectroscopy showed the spectra to differ in a peak-6 ppm appearing in the spectra of the A1-adapted fungus indicating terminal phosphate groups of mobile polyphosphate. Thus, in the Al-adapted fungus the average chain length of mobile polyphosphate is considerably shorter than in the non-Al-adapted fungus. A special method of cyclic phosphate supply followed by block averaging of the NMR spectra was used to determine the kinetic behaviour of phosphate uptake, storage and incorporation into polyphosphate at a constant external pH 3.5. While the Al-adapted fungus showed resistance to Al, an irreversible break-down in phosphate metabolism of the non-Al-adapted fungus by exposure to Al was caused. In comparison with the non-Al-adapted fungus supplied by nutrient solutions omitting Al, the Al-adapted fungus showed higher levels in both phosphate uptake and mobile polyphosphate concentration. As a consequence of these results a de-toxification of freely mobile Al-ions into a stable and insoluble complex in the Al-adapted fungus is considered to be due to a capture of intracellular Al by mobile polyphosphate of shorter chain length.  相似文献

Present work describes the bioleaching potential of metals from low-grade mining ore containing smithsonite, sphaerocobaltite, azurite and talc as main gangue minerals with adapted consortium of Sulfobacillus thermosulfidooxidans strain-RDB and Thermoplasma acidophilum. Bioleaching potential improved markedly by added energy source, acid preleaching and adaptation of microbial consortium with mixed metal ions. During whole leaching period including acid preleaching stage of 960 h and bioleaching stage of 212 days about 76% Co, 70% Zn, 84% Cu, 72% Ni and 63% Fe leached out.  相似文献

During iron oxidation,Thiobacillus ferrooxidans (Ferrobacillus ferrooxidans) was able to tolerate high concentrations of Zn, Ni, Cu, Co, Mn and Al (more than 10 g/litre). Silver and anions of tellurium, arsenic and selenium were toxic in concentrations of 50–100 mg/litre. Molybdenum (as molybdate), at concentrations above 5 mg/litre, was lethal toT. ferrooxidans. During thiosulphate oxidation, the tolerance to Zn, Ni and Co was greatly reduced, cobalt now being at least 2000 times more toxic, and the inhibitory levels of Zn and Ni being 600 mg Zn/litre and 150 mg Ni/litre. During sulphur oxidation, the tolerance to heavy metals extended to concentrations above 5 g/litre. Adaptation to Zn, Ni or Cu during iron oxidation was found to result in increased tolerance to some of the other metals also.  相似文献

Microorganisms have been geologically active in mineral formation, mineral diagenesis and sedimentation via direct action of their enzymes or indirectly through chemical action of their metabolic products. This property of microorganisms is being harnessed during the recent years for extraction of metals from their ores, especially from low-grade ores. In the present study bioleaching of copper from its low-grade chalcopyrite ore using 26 isolates of acidophilic fungi is reported. Most of these fungal strains belonged to the genera Aspergillus, Penicillium and Rhizopus. The leaching experiments were conducted in Czepek Dox minimal medium containing 1% (100 mesh) ore with shaking at room temperature for 20 days. Out of these, 4 isolates exhibited significant bioleaching activities. Maximum leaching of copper (78 mg/L) was observed with Aspergillus flavus (DSF-8) and Aspergillus niger (DOF-1). Nutritional and environmental conditions for optimum bioleaching were standardized. Present study indicates the usefulness of acidophilic fungi in bioleaching of copper from its low-grade ores.  相似文献

In order to explore the bioleaching mechanism and improve the bioleaching efficiency,the micro-bial community in the bioleaching solution was compared with that on the surface of minerals based on the microarray analysis.Meanwhile,the elements composition in the bioleaching solution was analyzed using the ICP-AES method.Results showed that there was a high concentration of S and Cu in the leaching solution which up to 2 380 mg/L and 1 378 mg/L,respectively,after continuously bioleaching of copper-ore concen-trate for 30 days by a mixed culture associated with 12 species of bioleaching microorganisms.Based on the data of microarray,the total of cell number in the surface of minerals was far higher than that in the bi-oleaching solution.Furthermore,the dominant communities on the surface of minerals,such as Acidithiobacillus ferrooxidans,Acidithiobacillus thiooxidans and Acidithiobacillus caldus,were similar to that in the bioleaching solution.However,the relative level of some bacteria,such as Sulfobacillus aci-dophilus and Sulfobacillus thermosulfidooxidans,showed great discrepancy with lower presence in the bi-oleaching solution with respect to the mineral surface.  相似文献

To facilitate the application of anaerobic ammonium oxidation (anammox) to a nitrogen removal process, the effects of heavy metals (Ni, Cu, Co, Zn, and Mo) on anammox bacteria entrapped in gel carriers were examined by conducting continuous feeding tests for each metal. The results show that all anammox activities decreased by more than 10 % when influent concentrations of Ni, Cu, Co, Zn, and Mo were 5, 5, 5, 10, and 0.2 mg/L, respectively. It was observed that the effects of Ni, Cu, Co, and Zn on anammox activity were reversible and that of Mo on anammox activity was irreversible. Anammox activity was not affected when influent containing mixed Ni, Cu, Co, and Zn (0.5 mg/L) was fed into the reactor.  相似文献

Accumulation of different metals and metalloids was assessed in two vegetables radish (Raphanus sativus L.) and spinach (Spinacea oleracea L.) irrigated with domestic wastewater in the peri-urban areas of Khushab City, Pakistan. In general, the metal and metalloid concentrations in radish and spinach were higher at site-II treated with sewage water than those found at site-I treated with canal water. In case of radish at both sites the levels of metals (Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Se, Mo, Cd, and Pb) were below the permissible level except those of Mn, Ni, Mo, Cd, and Pb. At both sites, the transfer factor ranged from 0.047–228.3 mg kg?1 with Cr having the highest transfer factor. The metal pollution index in soil was in the following order: As > Fe > Ni > Zn > Cd > Mo > Se > Co > Pb > Mn > Cr > Cu, respectively. While in case of spinach at both sites, the concentrations of metals and metalloids in vegetable samples irrigated with canal and sewage water were observed below the permissible level except Mn, Ni, Zn, Mo, and Pb. At both sites, the transfer factor ranged from 0.038–245.4 mg kg?1 with Cr having the highest transfer factor. The metal pollution index in soil was in the following order: Cd > Ni > Co > Se > Mn > Zn > Mo > Pb > Fe > Cr > As > Cu, respectively.  相似文献

A strain of Thiobacillus ferrooxidans was adapted to grow at higher concentrations of copper by single step culturing in the presence of 20 g/L (0.314 mol/L) cupric ions added to 9K medium. Exposure to copper results in change in the surface chemistry of the microorganism. The isoelectric point of the adapted strain (pI=4.7) was observed to be at a higher pH than that of the wild unadapted strain(pI=2.0). Compared to the wild strain, the copper adapted strain was found to be more hydrophobic and showed enhanced attachment efficiency to the pyrite mineral. The copper adsorption ability of the adapted strain was also found to be higher than that of the wild strain. Fourier transform infrared spectroscopy of adapted cells suggested that a proteinaceous new cell surface component is synthesized by the adapted strain. Treatment of adapted cells with proteinase-K, resulted in complete loss of tolerance to copper, reduction in copper adsorption and hydrophobicity of the adapted cells. These observations strongly suggest a role played by cell surface modifications of Thiobacillus ferrooxidans in imparting the copper tolerance to the cells and bioleaching of sulphide minerals.  相似文献

Abstract

The bioavailability and leachability of heavy metals play a major role in the toxicity of heavy metals in the compost applied for soil conditioning. A rotary drum composter was used for the study of heavy metal bioavailability and leachability during water hyacinth composting with a mixture of cattle manure, sawdust and lime. Lime was added in 1, 2 and 3% to the mixture of water hyacinth, cattle manure and sawdust at a ratio of 6:3:1 respectively. Influences of physico-chemical parameters on heavy metal bioavailability and leachability were studied during the process. The bioavailability of heavy metals solubility and diethylene triamine penta-acetic acid extractability was examined. The toxicity characteristics leaching procedure (TCLP) test was performed for assessing the hazardous properties of compost. The nutrients and the total concentration of heavy metals were increased during the composting process. The lime was very effective in reducing water solubility, plant availability and leachability of the selected heavy metals (Zn, Cu, Mn, Fe, Ni, Pb, Cd and Cr) during the process. The addition of lime provided a buffer against the decrease in pH and a sufficient amount of Ca that would improve the metabolic activity during composting. The addition of an excess amount of lime slowed the metabolic activity of the microbes due to its high alkalinity. The TCLP test confirms that the heavy metals concentrations in the control and in the lime-amended compost were below the threshold limits.  相似文献

A cost-effective way to reduce lead released into the environment is through immobilization with readily available soil amendments. Leaching of Pb from four shooting range soils treated with two phosphate sources was evaluated in soil columns. Phosphate was applied at a P/Pb molar ratio of 4:1 with two-thirds of the P supplied from phosphate rock (PR) and one-third from phosphoric acid (PA). The soils were incubated for 18 hr and leached with toxicity characterization leaching procedure (TCLP) and synthetic precipitation leaching procedure (SPLP) fluids, respectively. PR/PA treatment reduced Pb in TCLP leachate to below regulatory limit of 5 mg/L after 18 hr of incubation. Reduction of Pb leached was more dependent on clay content than total Fe. PR/PA treatment had no effect on pH of TCLP leached soils, while it led to a drastic lowering of pH that mobilized Pb in SPLP leached soils. However, leaching of Pb reduced with time in SPLP leached soils because pH increased gradually. PR/PA treatment was more effective in SPLP leached soils with low total soil Pb because they received lower amounts of PA. PR/PA treatment may be more efficient when applied as split applications in soils with a low to medium Pb loading and a high buffering capacity.  相似文献

Environmental laws concerning spent catalysts disposal have become increasingly more severe in recent years. Due to the toxic nature of spent catalysts, their disposal can pollute the environment. The recovery of heavy metals decreases the environmental impact of the waste catalysts and the recycled product can be further used for industrial purposes. Bio-hydrometallurgical approaches, such as bioleaching, appear to offer good prospects for recovering valuable metals from spent refinery catalysts. Currently, identifying and modifying the parameters that influenced the efficiency of bioleaching is important for industrial sector. The biological system can be further improved through optimizing the bioleaching parameters, such as the nutrient culture media, amount of oxygen and carbon dioxide, pH, temperature, inoculum, metal resistance of microorganisms, chemistry of solid waste, particle size of solid waste, solid liquid ratio, bioleaching period, size of substrate, shaking speed, and also the development of more effective bioleaching microorganisms. In our previous review (Asghari et al. in J Ind Eng Chem 19:1069–1081, 2013), information available in the literature on the bioleaching fundamentals of spent catalysts with a focus on recent developments was reviewed in detail. In this study, the effects of most important factors that influence an efficient bioleaching process of spent refinery catalysts with the hope that these valuable and useful data can help determine the most efficient process will be discussed. The details of metals recovery with a focus on the effects of different variables in the bioleaching such as reaction time, pulp density, initial pH, particle size, nutrient concentration, temperature and buffer will also be presented.  相似文献

以低品位黄铜矿溶液为原料,浸矿制备Cu2+能有效提高低品位黄铜矿的利用价值。基于浸矿过程中存在多因素影响的现象,通过正交试验与神经网络分析方法,对浸矿条件(接种量、矿石品位、Fe2+添加量及浸矿溶液pH)实行优化。结果表明:在正交试验组中最佳试验结果为浸矿产128.753mg/LCu^2+;BP神经网络优化后的最佳实验组合为微生物接种量12%、矿石品位0.3%、添加Fe^2+24g/L及浸矿溶液pH1.7,该条件下验证试验产Cu^2+ 141.352mg/L,通过正交试验及神经网络优化提高了微生物浸出低品位黄铜矿酸性溶液Cu^2+的产量。  相似文献

In this study, it was considered that the biosorption of heavy metals by biomass might occur during the bioleaching of fly ash. This work is focused on the biosorption behavior of Al, Fe, Pb and Zn by Aspergillus niger during the bioleaching process. The fungal biomass was contacted with heavy metals solution which extracted from fly ash by using gluconic acid as leaching agent. The equilibrium time for biosorption was about 120 min. The biosorption experiment data at initial pH 6.5 was used to fit the biosorption kinetics and isotherm models. The results indicated that the biosorption of Al, Fe and Zn by A. niger biomass were well described by the pseudo-first order kinetic model. The pseudo-second order kinetic model was more suitable for that of Pb. The Langmuir isotherm model could well describe the biosorption of Fe, Pb and Zn while the Freundlich model could well describe the biosorption of Al. Furthermore, the biosorption of metal ions decreased evidently in the presence of fly ash as compared to that in the absence of fly ash. This research showed that although the biomass sorption occurred during the bioleaching process, it did not inhibit the removal of Al, Fe, Pb and Zn evidently from fly ash.  相似文献

Investigation of the spatial distribution of metals was conducted for two constructed wetlands used as tertiary treatment in Chia Nan University of Pharmacy and Science (CNU) and Metal Processing Industries (MPI) located in Tainan, Taiwan. These two distinguished sites were selected to compare the distribution of metals for constructed wetlands treating different types of wastewater. Along the distance, samples of water, sediment, and macrophytes were analyzed for metals including Al, Cd, Cr, Cu, Fe, Mn, Ni, Pb, and Zn. Additionally, measurements of water quality including temperature, pH, EC, ORP, DO, TSS, BOD, COD, and turbidity were performed. Results show that, at CNU, wastewater contained higher organic consititute (BOD 29.3 +/- 11.7 mg/, COD 46.7 +/- 33.6 mg/L) with low metals content. Wastewater at MPI contained low level of organic consititute (BOD 7.1 +/- 3.3 mg/L, and COD 66.0 +/- 56.5 mg/L) and higher metals content. Metals distribution of both sites showed similar results where metals in the sediments in the inlet zone have greater concentrations than other areas. The constructed wetlands can remove Cd, Cu, Ni, Pb, and Zn. However, there was no removal of Al, Cr, Fe, and Mn. A distance along the constructed wetlands had no effect on metal concentrations in macrophyte and water.  相似文献

Bioleaching of uranium was carried out with Turamdih ore sample procured from Uranium Corporation of India Limited, Jaduguda. The bacterial strain that was used in the leaching experiments was isolated from the Jaduguda mine water sample. Efficiency of bioleaching was studied by varying parameters like pulp density and initial ferrous concentration as source of energy. It is observed that the efficiency of bioleaching was 49% at 10% pulp density (w/v) and initial pH 2.0. Addition of external has no effect on efficiency of bioleaching showing domination of direct leaching mechanism over indirect.  相似文献
