Changes in formation and localizations of phenolic compounds, including flavans, were investigated in the tissues of European and Canadian yew (Taxus baccata L. and T. canadensis Marsh.) during dedifferentiation in vitro. Annual shoots of European yew had the highest capacity for synthesizing these compounds. During the summer growth period, the content of total soluble phenolic compounds and flavans in these shoots was 30–40% higher than in the winter. Cell dedifferentiation and growth in vitro was accompanied by enhanced synthesis of phenolic compounds, including flavans, the change in tissue localization of these compounds, and an increase in the number of cells containing phenolics. Significant accumulation of phenolic compounds in callus cells resulted in necroses following two subcultures in the European and Canadian yew cultures initiated from summer explants, and following seven subcultures of the European yew calli initiated from winter explants. These data allow us to suggest that a high level of phenolic compounds in yew calli could be the reason for their necrosis.  相似文献

A new optically active flavan aglucone, 7-hydroxy-3′,4′-methylenedioxyflavan, and its 7-glucoside have been isolated from the bulbs of Zephyranthes flava, collected at flowering. Additionally, two known flavans, 7,4′-dihydroxy-3′-methoxyflavan and 7-methoxy-2′-hydroxy-4′,5′-methylenedioxyflavan, have been isolated for the first time from this species. The structures of these flavans have been established by comprehensive analyses (UV, IR, 1H NMR, 13C NMR, mass spectrometry, [α]D) of the compounds and their acetates, and also by chemical correlation.  相似文献

One of the goals of chemical ecology is to assess costs of plant defenses. Intraspecific trade-offs between growth and defense are traditionally viewed in the context of the carbon-nutrient balance hypothesis (CNBH) and the growth-differentiation balance hypothesis (GDBH). Broadly, these hypotheses suggest that growth is limited by deficiencies in carbon or nitrogen while rates of photosynthesis remain unchanged, and the subsequent reduced growth results in the more abundant resource being invested in increased defense (mass-balance based allocation). The GDBH further predicts trade-offs in growth and defense should only be observed when resources are abundant. Most support for these hypotheses comes from work with phenolics. We examined trade-offs related to production of two classes of defenses, saponins (triterpenoids) and flavans (phenolics), in Pentaclethra macroloba (Fabaceae), an abundant tree in Costa Rican wet forests. We quantified physiological costs of plant defenses by measuring photosynthetic parameters (which are often assumed to be stable) in addition to biomass. Pentaclethra macroloba were grown in full sunlight or shade under three levels of nitrogen alone or with conspecific neighbors that could potentially alter nutrient availability via competition or facilitation. Biomass and photosynthesis were not affected by nitrogen or competition for seedlings in full sunlight, but they responded positively to nitrogen in shade-grown plants. The trade-off predicted by the GDBH between growth and metabolite production was only present between flavans and biomass in sun-grown plants (abundant resource conditions). Support was also only partial for the CNBH as flavans declined with nitrogen but saponins increased. This suggests saponin production should be considered in terms of detailed biosynthetic pathway models while phenolic production fits mass-balance based allocation models (such as the CNBH). Contrary to expectations based on the two defense hypotheses, trade-offs were found between defenses and photosynthesis, indicating that studies of plant defenses should include direct measures of physiological responses.  相似文献

The effects of cadmium (6.3 × 10?5 M or 10.6 × 10?5 M) on the growth of tea plant (Camellia sinensis L.) callus cultures derived from leaves, stems, and roots and on the formation, in these cultures, of phenolic compounds, including flavans and lignin, which are characteristic of the tea plant, were investigated. In the calli derived from leaves and stems, cadmium treatment decreased the biomass increment, while in the calli derived from roots, growth characteristics remained at the control level. Under the effect of cadmium, the content of phenolic compounds, including flavans, in the leaf calli decreased, while in the stem and root calli, it either increased (at the cadmium concentration of 6.3 × 10?5 M), or was close to a control one (at the cadmium concentration of 10.6 × 10?5 M). The lignin content in the root and stem calli increased, but it did not change in the leaf calli. All this data demonstrate that the cadmium-induced changes in phenolic metabolism of the tea plant callus culture depended both on the cadmium concentration in the medium and on the origin of calli.  相似文献

A total synthesis of four flavans with a furan ring in different positions (14) was achieved successfully. The flavan was synthesized from flavanone by reduction and followed by deoxygenation steps. Hippeastrum x hortorum bulbs produce red pigments upon wounding. Analysis by TLC, HPLC, MS and NMR revealed that this pigment, which is absent in unwounded tissue, is a mixture composed of an orange-coloured chalcone (3,2′4′-trihydroxy-4-methoxychalcone) and 3 flavans (7,4′-dihydroxy-8-methylflavan, 7,3′-dihydroxy-4′-methoxyflavan and 7-hydroxy-3′-4′-methylenedioxyflavan). The colourless flavans can be oxidised to red-coloured dimers or polymers. The induction of these “phytoalexins” is achieved by wounding and can be further amplified by biotic and abiotic elicitors; e.g. cell walls of yeast appeared to be a potent elicitor. Immediately after wounding or elicitation the apparent activities of phenylalanine ammonium lyase (PAL) and peroxidase, which are probably involved in the biosynthesis of the red pigments increase substantially (both enzymes are hardly measurable in unwounded tissue) and reach a transient maximum after 2 ? 3d whereas pigment formation becomes visible after 2d and reaches a maximum after 9 to 12 d. Since cycloheximide inhibits the formation of the phytoalexins, a de novo synthesis of the corresponding enzymes is likely. Total phytoalexin extracts showed an inhibition of bacterial growth (e.g. of Bacillus subtilis, B. megaterium) and of feeding by polyphagous larvae of the moth Syntomis mogadorensis. These data imply that the wounding- and elicitor-induced phytoalexins appear to function as defence compounds against microorganisms and herbivores.  相似文献

It was shown that, in pea (Pisum sativum L.) roots, flavans are a dominating component of the soluble phenol fraction. In plants grown at low temperature (8°C), flavan content during the early growth phase was lower than in plants grown at 22°C, but later it increased and was by 40% higher than in plants grown at 22°C. Total phenol content in the two treatments differed insignificantly. Low growth temperature decreased the content of some phenolic compounds in pea seedling roots.  相似文献

Grapes of the Gamay Beaujolais and Siebel varieties have been sampled during the period from just before the onset of colouration to harvest. Extraction has yielded data on the composition and quantities of lower molecular weight and polymeric flavanoids present. The structure of the major flavans of these grapes has been established and the variation in their relative amounts monitored during the growth season. The basic structural unit of the grape proanthocyanidin polymer is a monomer with (?)-epicatechin stereochemistry.  相似文献

An investigation of Marshallia obovata afforded, in addition to known thymol derivatives, one new ester. Furthermore six new chromone derivatives, all prenylated, were present together with the known spathelia chromene. The main constituent was a flavan already isolated from M. grandiflora but five new flavans were also isolated. Besides other widespread compounds a tetraester of inositol was isolated in minute amounts. All the structures were elucidated by spectroscopic methods or chemical transformations. Surprisingly, even the main 4-methoxyflavan could be split by periodic acid.  相似文献

Principal rotenoids (deguelin, tephrosin, rotenone, and 12a-hydroxyrotenone) (3-30microM) isolated from the stems of Erycibe expansa significantly inhibited invasion of human fibrosarcoma HT1080 cells through Matrigel-coated filters and release of proMMPs-2 and 9. In addition, deguelin and tephrosin showed differentiation-inducing activity in human promyelocytic leukemia HL-60 cells. Furthermore, effects of various constituents isolated from the ethyl acetate-soluble fraction on proliferation of human leukemia U937 cells were examined. As a result, most of isoflavones and several flavans as well as rotenoids showed moderate or substantial anti-proliferative activities.  相似文献

A new flavone xyloside, 1, and two new flavan-3-ol glucosides, 3 and 4, were isolated together with three known flavones, 2, 11, and 12, five known flavans, 5-9, and a known dihydrochalcone, 10, from the stems and leaves of Juniperus communis var. depressa (Cupressaceae) collected in Oregon, U.S.A., and their structures were determined on the basis of spectral evidence. A novel flavone nucleus such as that in 1 is seldom found in nature today, and new methylcatechin glucosides 3 and 4 are also rare in nature. In addition, we investigated the inhibitory activity of individual components, i.e., 8-11, and others, that were abundantly isolated from the same plant material for the Maillard reaction.  相似文献

Young shoot tips of species and hybrids from the Prunus section eucerasus contain catechin, epicatechin and flavolans (oligomeric flavans including biflavans). Prunus avium shoot segments, 2 mm in length, were excised from the elongating zone of growing shoots and cultured in vitro on a liquid Murashige and Skoog medium containing indolyl-3-acetic acid and benzyl adenine. Supplementation of the same medium with (+)-catechin and epicatechin resulted in apparent promotion of callus growth. This effect was greater under long day conditions than under continuous dark.  相似文献

Phytochemical investigation of the aerial parts of Potentilla astracanica Jacq. led to the isolation of two isoflavonoid glucosides (1, 2) five flavonoid glycosides (37) including one flavone-C-glycoside (7) as well as two flavans (8, 9). Circular dichroism is a powerful tool for establishing the absolute configuration of flavonoids and proanthocyanidin analogues. It has been utilized to study the configuration of flavanones, dihydroflavonols (3-hydroxyflavanones), flavan-3-ols, flavan-4-ols, flavan-3,4-diols, flavans, isoflavans, isoflavanones, pterocarpans, 6a-hydroxypterocarpans, rotenoids, 12a-hydroxyrotenoids, neoflavonoids, 3,4-dihydro-4-arylcoumarins, 4-arylflavan-3-ols, auronols, homoisoflavanones, proanthocyanidins, and various classes of biflavonoids. Results relevant to the correlation of circular dichroic data and the absolute configuration of the diastereoisomers of some of the above classes of compounds will be discussed.  相似文献

Investigation of the initial stages of infection of pea plants (Pisum sativum L., cv. Marat) with bacteria Rhizobium leguminosarum has demonstrated that interactions with the microsymbiont considerably influence the content and composition of phenolic compounds (PC) in the host plant. The changes were observed both in the total root content of PC and in the content of flavans. Concentrations of PC in various root zones were also different. The root regions featuring high content of PC in a day after inoculation had the lowest number of attached and penetrated bacteria. At the same time, in noninoculated plants, the distribution of PC in the root zones with various sensitivities to R. leguminosarum showed the opposite pattern.  相似文献

A new phenolic compound, broussonone A (1) were isolated from the stem barks of Broussonetia kanzinoki (Moraceae), together with two diphenylpropanes, broussonin A (2), broussonin B (3), two flavans, 7,4'-dihydroxyflavan (4), 3',7-dihydroxy-4'-methoxyflavan (5), and two flavones, 3,7-dihydroxy-4'-methoxyflavone (6), 3,7,3'-trihydroxy-4'-methoxyflavone (7). Compound 1 showed noncompetitive inhibitory activity on pancreatic lipase with an IC(50) of 28.4 μM. In addition, compounds 1-5 significantly inhibited adipocyte differentiation in 3T3-L1 cells as measured fat accumulation using Oil Red O assay.  相似文献

The effect of hormone-like compounds at different concentrations: 2,4-D (2 × 10?6; 2 × 10?5; and 2 × 10?4M) and 1-NAA (2 × 10?7; 2 × 10?6; 2 × 10?5; 4 × 10?5, and 6 × 10?5 M) on the growth and production of phenolic compounds, including flavans and lignin, was investigated in callus culture of tea plant (Camellia sinensis L., a highly productive strain IFR ChS-2). The growth of the culture was vigorous, and production of phenolic compounds therein was efficient in the medium containing 2 × 10?5 M 2,4-D. Substitution of 1-NAA for 2,4-D in all the cases decelerated the growth of the culture. These changes were more pronounced when 2 × 10?7 and 2 × 10?6 M 1-NAA was used; in this case, biomass accumulation decreased by 1.5–2.0 times as compared with control material growing on the medium with 2 × 10?5 M 2,4-D. In the presence of 1-NAA, the content of total soluble phenolic compounds and flavans in the calli rose by 30% on the average as compared with control material. Accumulation of lignin remained essentially the same. Therefore, the replacement of 2,4-D with 1-NAA in the nutrient medium used for the growing of highly productive strain of tea plant callus did not induce considerable changes in its ability to produce phenolic compounds.  相似文献
