Hydrolysates of methanolic leaf extracts of Circaea yield the glycoflavones vitexin, isovitexin, vicinin-1, and vicinin-2. Additional flavonoids are found in the unhydrolyzed extracts. Previous studies of Onagraceae report only flavonols and one chalcone. Circaea is assigned to a monogeneric tribe of Onagraceae and has no clear relationship to any other group within the family. The presence of glycoflavones in Circaea serves to underscore the distinctiveness of the genus.  相似文献

Observations of meiotic chromosomes are reported for all 21 species and 3 additional sub species ofGaura (Onagraceae), based upon a study of 647 individuals from 509 naturally occurring populations throughout the range of the genus. The basic chromosome number for the genus isx = 7, and 18 species are diploid withn = 7. Among these, the self-incompatible ones are often highly chromosomally heterozygous, with no homozygous individuals having been found in nature in the perenrennialsGaura lindheimeri andG. villosa, and two-thirds or more of the individuals apparently heterozygous in the following well-sampled species:G. calcicola, G. longiflora, andG. suffulta subsp.suffulta. In contrast, the autogamous species are entirely chromosomally homozygous or nearly so. Two species ofGaura are reported as chromosomal structural heterozygotes, with about 50% pollen abortion:G. biennis andG. triangulata; the translocation systems originated independently of one another. Two of the three polyploid species,G. sinuata andG. drummondii (G. odorata of many authors), are consistently tetraploid (n = 14) and, despite their cytological autotetraploidy, are thought to have originated following interspecific hybridization. They are the only rhizomatous species in the genus and may have had one ancestor in common. The remaining polyploid,G. coccinea, includes populations withn = 7, 14, 21, and 28, as well as evident interploid hybrids and, frequently, supernumerary chromosomes. The relationship among these populations is close and is maintained by frequent hybridization and exchange of genetic material. No other species seems to have participated in their origin, and the association of their chromosomes is consistently that characteristic of autopolyploidy in plants with tetraploid and higher chromosome numbers.  相似文献

The leaves of Saussurea triangulata (Compositae) have been eaten with rice as a wrapping vegetable for preventing neuro-aging. However, the components responsible for the neuroprotective effects of S. triangulata still remain unidentified. In the process of investigating the neuroprotective activity of S. triangulata, we found that a methanol extract of S. triangulata exhibited significant protection against glutamate-induced toxicity in primary cultured rat cortical cells. Three quinic acid derivatives were isolated from the n-BuOH fraction of S. triangulata. Among these three quinic acid derivatives, methyl 5-caffeoylquinic acid (3) exhibited significant neuroprotective activities against glutamate-induced toxicity exhibiting cell viability of about 50%, at concentrations ranging from 0.1 μM to 10 μM. Therefore, the neuroprotective effect of S. triangulata might be due to the inhibition of glutamate-induced toxicity by the quinic acid derivatives from S. triangulata.  相似文献

A survey of the flavonoids of some 92 species of Australian Cyperus, mainly of subtropical or tropical origin, has confirmed a correlation previously reported in this family between flavonoid pattern and plant geography. The pattern found was similar to that of African and South American Cyperaceae, particularly in the occurrence of the rare marker substance, luteolin 5-methyl ether. Tricin and luteolin are relatively common, in glycosidic form, in the leaves while the flavonol quercetin is infrequent. When present, quercetin occurs either in glycosidic form or free as a methyl ether. The 3-monomethyl and 3, 7-dimethyl ethers of kaempferol and quercetin and the 3, 7, ?-trimethyl ether of quercetin are reported for the first time from the Cyperaceae. The flavonoid pattern of inflorescences is distinct from that of the leaves in that tricin is not detectable and that luteolin 5-methyl ether appears to be replaced by 7, 3′, 4′-trihydroxyflavone. In addition, the aurone aureusidin is more commonly present than in the leaves and is occasionally accompanied by two further aurones. The glycoxanthones mangiferin and isomangiferin occur rarely in all three species examined in the section Haspani, i.e. in C. haspan, C. prolifer and C. tenuispica. In general, however, the flavonoid data do not offer any markers which separate off different sections within the genus; there are, however, some significant correlations between the frequency of the flavonoid classes and subgeneric groupings.  相似文献

The conifer genus Phyllocladus is shown by comparative flavonoid chemistry to be remarkably homogeneous and quite distinct from other studied genera in the Podocarpaceae. It is characterized by the accumulation (in the foliage) of a predominance of flavone O-glycosides, and in particular, luteolin 7- and 3′-O-glycosides. Lower levels of flavonol O-glycosides are also evident. Two flavone glycosides are reported for the first time, luteolin 3′-O-α-L-rhamnopyranoside and luteolin 7-O-α-L-rhamnoside.  相似文献

During a chemosystematic survey of 38 representative species of the Juncaceae for leaf and stem flavonoids, the 5-methyl ether of luteolin was discovered for the first time in plants. It occurs both free and as the 7-glucoside; its identity was confirmed by synthesis. Flavone sulphates were also found in the family and the 7-glucosidesulphates of luteolin and chrysoeriol were characterised for the first time. 7,3′,4′-Trihydroxyflavone and its 7-glucoside, not previously reported in the monocotyledons, were found in three species. The presence of luteolin 5-methyl ether or its glucoside in 70% of the species surveyed serves to distinguish the Juncaceae from the morphologically related Centrolepidaceae, Restionaceae and Thurniaceae. Flavone C-glycosides, common in grasses and sedges, were found only in Prionium, a genus which on anatomical grounds is anomalous in the Juncaceae. Among other phenolics detected during the survey, the uncommon 7,8-dihydroxycoumarin, daphnetin, was identified in Juncus effusus and its 8-methyl ether in four Luzula species. Taken together, these chemical findings show that the Juncaceae are very distinctive in their phenolic pattern and confirm the correctness of assigning them an isolated position in a separate order, the Juncales. The results indicate that the Juncaceae are chemically specialized, in spite of the facts that the family has been regarded as ancestral to the Cyperaceae and Gramineae and that they have been assigned a low advancement index by Sporne.  相似文献

Forty-four chromosome counts from the North American genusCallirhoe are reported, including first-recorded counts ofC. alcaeoides, C. digitata, C. bushii, C. triangulata, andC. involucrata var.tenuissima. Information concerning chromosome numbers from present and previous reports is summarized and evaluated. Observations are presented on meiosis for the genus in general, for male sterile individuals of gynodioecious species, and for selected F1 hybrids. Pollen fertility data are recorded for the latter. Chromosomally three species groups are recognized: an exinvolucellate, diploid species group withn=14; an involucellate, tetraploid and octoploid species pair,n=28 and 56; and an involucellate, diploid and tetraploid species group withn=15 and 30. Intrageneric relationships and those within the tribe Malveae subtribe Malvinae are discussed.  相似文献

Lagerstroemia (crape myrtle) is an important plant genus used in ornamental horticulture in temperate regions worldwide. As such, numerous hybrids have been developed. However, DNA sequence resources and genome information for Lagerstroemia are limited, hindering evolutionary inferences regarding interspecific relationships. We report the complete plastid genome of Lagerstroemia fauriei. To our knowledge, this is the first reported whole plastid genome within Lythraceae. This genome is 152,440 bp in length with 38% GC content and consists of two single-copy regions separated by a pair of 25,793 bp inverted repeats. The large single copy and the small single copy regions span 83,921 bp and 16,933 bp, respectively. The genome contains 129 genes, including 17 located in each inverted repeat. Phylogenetic analysis of genera sampled from Geraniaceae, Myrtaceae, and Onagraceae corroborated the sister relationship between Lythraceae and Onagraceae. The plastid genomes of L. fauriei and several other Lythraceae species lack the rpl2 intron, which indicating an early loss of this intron within the Lythraceae lineage. The plastid genome of L. fauriei provides a much needed genetic resource for further phylogenetic research in Lagerstroemia and Lythraceae. Highly variable markers were identified for application in phylogenetic, barcoding and conservation genetic applications.  相似文献

The subfamily Pomoideae has been surveyed for leaf phenolics and it has been shown that flavone glycosides are present in the genera Sorbus, Aronia, Chaenomeles and Hesperomeles in addition to the previously reported occurrences in Crataegus, Malus and Pyrus. The dihydrochalcone phloridzin, a typical constituent of Malus, has also been found in Docynia. Arbutin and phenolic acid-calleryanin esters are apparently restricted to Pyrus. Naringenin and eriodictyol glucosides have been detected in Pyracantha, Sorbus, Photinia, Chaenomeles and Hesperomeles. A number of Pomoideae phenolics have been found in two Spiraeoideae genera; luteolin 7-glucoside,] luteolin 7-diglucoside, luteolin 7-rhamnosylglucoside and apigenin 7-glucoside in Exochorda and the dihydrochalcone trilobatin in Sorbaria. The chemotaxonomic evidence is consistent with the hypothesis that the Pomoideae evolved through a process of allopolyploidy from primitive members of the Spiracoideae and Prunoideae.  相似文献

Nuclear factor erythroid 2-related factor 2 (Nrf2) is over-expressed in many types of tumor, promotes tumor growth, and confers resistance to anticancer therapy. Hence, Nrf2 is regarded as a novel therapeutic target in cancer. Previously, we reported that luteolin is a strong inhibitor of Nrf2 in vitro. Here, we showed that luteolin reduced the constitutive expression of NAD(P)H quinone oxidoreductase 1 in mouse liver in a time- and dose-dependent manner. Further, luteolin inhibited the expression of antioxidant enzymes and glutathione transferases, decreasing the reduced glutathione in the liver of wild-type mice under both constitutive and butylated hydroxyanisole-induced conditions. In contrast, such distinct responses were not detected in Nrf2−/− mice. In addition, oral administration of luteolin, either alone or combined with intraperitoneal injection of the cytotoxic drug cisplatin, greatly inhibited the growth of xenograft tumors from non-small-cell lung cancer (NSCLC) cell line A549 cells grown subcutaneously in athymic nude mice. Cell proliferation, the expression of Nrf2, and antioxidant enzymes were all reduced in tumor xenograft tissues. Furthermore, luteolin enhanced the anti-cancer effect of cisplatin. Together, our findings demonstrated that luteolin inhibits the Nrf2 pathway in vivo and can serve as an adjuvant in the chemotherapy of NSCLC.  相似文献

Ártioposthia triangulata is a terrestrial planarian that feeds on earthworms. Up to 76% of field collected A. triangulata fed on Eisenia fetida earthworms at 10°C in the laboratory. In one experiment, the feeding rate declined from 63% of earthworms attacked to 36% and 34% in the first, second and third feeding opportunities over a week. The average number of E. fetida eaten was 1.4 per planarian per week. The gain in weight of individual A. triangulata was significantly related to the amount of earthworm tissue lost during the feeding process. Over a one week period, the amount of earthworm tissue lost was related to the total amount presented to the planarians but not to their size. Frequency of attack was, however, related to planarian size. The results are discussed in relation to an hypothetical population of earthworms in pasture. It is concluded that the results support the contention that A. triangulata could seriously deplete earthworm populations.  相似文献

The major flavonoids of Marchantia polymorpha var. polymorpha and aquatica are the 7-O-β-d-glucuronides of apigenin and luteolin, luteolin 3′-O-β-d-glucuronide, luteolin 7,3′-di-O-β-d-glucuronide, and the 7,4′-di-O-β-d-glucuronides of apigenin and luteolin. These are accompanied by minor amounts of apigenin, luteolin, luteolin 3′,4′-di-O-β-d-glucuronide and luteolin 7,3′,4′-tri-O-β-d-glucuronide. All the luteolin di- and triglucuronides except the 3′,4′-di- substituted compound are new natural products.  相似文献

The flavonoids of 2 samples of Conocephalum conicum gametophyte tissue have been studied, one from U.S.A. and the other from Germany. Common to both samples were vicenin-2, lucenin-2, the 7-O-glucuronides of apigenin, chrysoeriol and luteolin and the previously unknown 7-O-glucuronide 4′-O-rhamnosides of apigenin, chrysoeriol and luteolin. Additionally the German sample contained the 7,4′-di-O-glucuronides of apigenin and luteolin and a new compound, apigenin 7-O-diglucuronide 4′-O-glucuronide. The North American sample contained, additionally, luteolin 7,3′-di-O-glucuronide, luteolin 7-O-glucuronide 3′,4′-di-O-rhamnoside (a new triglycoside) and 2 further derivatives of luteolin 7-O-glucuronide. Evidence is presented for the existence of geographic faces of C. conicum and for the qualitative invariability of the flavonoid patterns with changing season or environment.  相似文献

The NLRP3 inflammasome is a caspase-1 containing multi-protein complex that controls the release of IL-1β and plays important roles in the innate immune response. Since NLRP3 inflammasome is implicated in the pathogenesis of a variety of diseases, it has become an increasingly interested target in developing therapies for multiple diseases. We reported the current study to determine how luteolin, a natural phenolic compound found in many vegetables and medicinal herbs, would modulate NLRP3 inflammasome in both the in vivo and in vitro settings. First, we found that a high-fat diet upregulated mRNA expression of NLRP3 inflammasome components Asc and Casp1 in adipose tissue of ovariectomized mice, which were greatly reduced by dietary supplementation with luteolin. Of note, Asc and Casp1 expression in adipose tissue correlated with mRNA levels of Adgre1 encoding F4/80, an established marker for mature macrophages. We also demonstrated that luteolin inhibited NLRP3 inflammasome-derived caspase-1 activation and IL-1β secretion in J774A.1 macrophages upon diverse stimuli including ATP, nigericin, or silica crystals. Luteolin inhibited the activation step of NLRP3 inflammasome by interfering with ASC oligomerization. Taken together, these findings suggest that luteolin supplementation may suppress NLRP3 induction and activation process and thus potentially would be protective against NLRP3-mediated inflammatory diseases.  相似文献

Traces of luteolin, an important rhizobial nod gene inducer in Rhizobium meliloti, are released by alfalfa (Medicago sativa L.) seeds, but most luteolin in the seed exudate is conjugated as luteolin-7-O-glucoside (L7G). Processes affecting the production of luteolin from L7G in seed exudate are poorly understood. Results from this study establish that (a) seed coats are the primary source of flavonoids, including L7G, in seed exudate; (b) these flavonoids exist in seeds before imbibition; and (c) both the host plant and the symbiotic R. meliloti probably can hydrolyze L7G to luteolin. Glycolytic cleavage of L7G is promoted by glucosidase activity released from sterile seeds during the first 4 hours of imbibition. Thus, L7G from imbibing alfalfa seeds may serve as a source of the nod-gene-inducing luteolin and thereby facilitate root nodulation by R. meliloti.  相似文献

Thirty-three Sonchus, one Embergeria, one Babcockia and five Taeckholmia species were surveyed for their phenolic constituents. The coumarins scopoletin and aesculetin were found as major constituents of Embergeria, Babcockia and Taeckholmia species, and in lesser amount in some Sonchus species. Six flavone glycosides were identified: apigenin 7-glucuronide, apigenin 7-rutinoside, luteolin 7-glucoside, luteolin 7-glucuronide, luteolin 7-rutinoside and luteolin 7-glucosylglucuronide and the systematic significance of their distribution is discussed.  相似文献

Emergence of the bi-subunit topoisomerase I in the kinetoplastid family (Trypanosoma and Leishmania) has brought a new twist in topoisomerase research related to evolution, functional conservation and preferential sensitivities to the specific inhibitors of type IB topoisomerase family. In the present study, we describe that naturally occurring flavones baicalein, luteolin and quercetin are potent inhibitors of the recombinant Leishmania donovani topoisomerase I. These compounds bind to the free enzyme and also intercalate into the DNA at a very high concentration (300 µM) without binding to the minor grove. Here, we show that inhibition of topoisomerase I by these flavones is due to stabilization of topoisomerase I–DNA cleavage complexes, which subsequently inhibit the religation step. Their ability to stabilize the covalent topoisomerase I–DNA complex in vitro and in living cells is similar to that of the known topoisomerase I inhibitor camptothecin (CPT). However, in contrast to CPT, baicalein and luteolin failed to inhibit the religation step when the drugs were added to pre-formed enzyme substrate binary complex. This differential mechanism to induce the stabilization of cleavable complex with topoisomerase I and DNA by these selected flavones and CPT led us to investigate the effect of baicalein and luteolin on CPT-resistant mutant enzyme LdTOP1Δ39LS lacking 1–39 amino acids of the large subunit [B. B. Das, N. Sen, S. B. Dasgupta, A. Ganguly and H. K. Majumder (2005) J. Biol. Chem. 280, 16335–16344]. Baicalein and luteolin stabilize duplex oligonucleotide cleavage with LdTOP1Δ39LS. This observation was further supported by the stabilization of in vivo cleavable complex by baicalein and luteolin with highly CPT-resistant L.donovani strain. Taken together, our data suggest that the interacting amino acid residues of topoisomerase I may be partially overlapping or different for flavones and CPT. This study illuminates new properties of the flavones and provide additional insights into the ligand binding properties of L.donovani topoisomerase I.  相似文献
