Background

The cotton-melon aphid, Aphis gossypii Glover (Hemiptera: Aphididae), is a polyphagous species with a worldwide distribution and a variety of biotypes. North China is a traditional agricultural area with abundant winter and summer hosts of A. gossypii. While the life cycles of A. gossypii on different plants have been well studied, those of the biotypes of North China are still unclear.

Results

Host transfer experiments showed that A. gossypii from North China has two host-specialized biotypes: cotton and cucumber. Based on complete mitochondrial sequences, we identified a molecular marker with five single-nucleotide polymorphisms to distinguish the biotypes. Using this marker, a large-scale study of biotypes on primary winter and summer hosts was conducted. All A. gossypii collected from three primary hosts—hibiscus, pomegranate, and Chinese prickly ash—were cotton biotypes, with more cotton-melon aphids found on hibiscus than the other two species. In May, alate cotton and cucumber biotypes coexisted on cotton and cucumber seedlings, but each preferred its natal host. Both biotypes existed on zucchini, although the cucumber biotype was more numerous. Aphids on muskmelon were all cucumber biotypes, whereas most aphids on kidney bean were cotton biotypes. Aphids on seedlings of potato and cowpea belong to other species. In August, aphids on cotton and cucumber were the respective biotypes, with zucchini still hosting both biotypes as before. Thus, the biotypes had different fitnesses on different host plants.

Conclusions

Two host-specialized biotypes (cotton and cucumber) are present in North China. Hibiscus, pomegranate, and Chinese prickly ash can serve as winter hosts for the cotton biotype but not the cucumber biotype in North China. The fitnesses of the two host-specialized biotypes differ on various summer hosts. When alate aphids migrate to summer hosts, they cannot accurately land on the corresponding plant.  相似文献

The labial gland secretions from males of the bumble bee Bombus (Pyrobombus) perplexus Cresson were analysed by gas chromatography/mass spectrometry (GC/MS) in the electron impact and positive ion chemical ionization mode. The major compound of the complex mixture of alkenols, alkenals, fatty acids, hydrocarbons, wax type esters and steroids is 3,7,11,15-tetramethyl-2,6,10-hexadecatrien-1-ol (geranylcitronellol), considerable amounts of hexadecan-1-ol and Z-9-hexadecen-1-ol were also found. All alcohols were present as esters of the detected acids. In older samples both the acids and the alcohols sometimes could not be detected in the GC; therefore, the possibility to check the detected acid-alcohol pattern by interpreting the wax type ester peaks is very instructive. Moreover, the labial gland contains a rich mixture of mono- and di-unsaturated straight chain hydrocarbons. The similarity in composition of the labial glands of the North American B. (Pyrobombus) perplexus with the Eurasian species B. (Pyrobombus) hypnorum corroborates the assumption that the two species are conspecific. The likely supposition that the hydrocarbons could play an essential role in the chemical communication in bumble bees is discussed.  相似文献

 Scleroderris canker is a very damaging disease of conifers caused by a fungal pathogen, Gremmeniella abietina var ‘abietina’. This fungal pathogen is now known to comprise a number of distinct races and biotypes. In North America, two races, an indigenous North American race and an introduced European race, are present. In Europe, three distinct biotypes have been reported within the European race: one in the Alps, another in Fennoscandia, and a third that overlaps with the first two. We used random amplified microsatellites (RAMS) and DNA sequencing with arbitrary primer pairs (SWAPP) to design five PCR primer pairs flanking polymorphic regions of the genome of the European race of G. abietina. Length polymorphisms produced by repeats of basic units in microsatellites were distinguished by electrophoresis of PCR products in agarose gels, and point mutations were identified by low-ionic-strength single-strand conformation polymorphisms (LIS-SSCP). Some primers generated private alleles in the European biotype and the psychrophilic Alpine and Fennoscandian biotypes, i.e., alleles that were fixed within the two groups but polymorphic between them. Conversely, one pair of primers amplified at least 3, 4, and 7 alleles in the Fennoscandian, Alpine, and European biotypes, respectively. The Alpine and Fennoscandian biotypes, although geographically separated, were genetically more closely related to one another than to European biotype, which has an overlapping distribution. However, both Alpine and Fennoscandian biotypes have similar ecotypic adaptation. The orange blossom wheat midge, Sitodiplosis mosellana (Géhin) (Diptera: Cecidomyiidae), is a significant pest of wheat (Triticum spp.) grown in the Northern Hemisphere. It was accidently introduced to North America over 200 years ago and has subsequently spread throughout the northern Great Plains. Since 2010, several Canadian spring wheat varieties containing the resistance gene Sm1 have been released. Due to the potential of wheat midge populations to evolve virulent biotypes to Sm1, cultivars containing Sm1 are grown with a susceptible cultivar in an interspersed refuge. An understanding of the genetic diversity of wheat midge populations could provide important information on the potential development of resistance to Sm1. In the current study, we used two mitochondrial genes (CO1 and ND4) from wheat midge collected across the northern Great Plains and Québec in North America to assess population structure and genetic diversity. We found limited genetic diversity and population structure across the sampled North American populations. We also assessed North American haplotype similarity to wheat midge collected from Europe and China and found high similarity between North American and European populations, although sampling in Europe was limited. This supports the hypothesis that North American populations originated from Europe.  相似文献

The total surface lipids, including the wax particles, of the adult whiteflies of Bemisia tabaci and Trialeurodes vaporariorum were characterized. At eclosion, there were similar amounts of long-chain hydrocarbons, aldehydes, alcohols and wax esters. Within a few hours post-eclosion, long-chain aldehydes and long-chain alcohols were the dominant surface lipid components, C34 on B. tabaci and C32 on T. vaporariorum. Hydrocarbons, mainly n-alkanes, were minor components of the surface lipids. The major wax esters were C46 on B. tabaci and C42 on T. vaporariorum. The major acid and alcohol moieties in the wax esters of B. tabaci were C20 and C26, respectively, and of T. vaporariorum were C20 and C22, respectively. Both B. tabaci and T. vaporariorum had a minor wax ester composed of the fatty acid C18:1 esterified to the major alcohols, C34 and C32, respectively. Bemisia were readily distinguished from Trialeurodes based on the composition of their wax particles and/or their wax esters; however, no differentiating surface lipid components were detected between biotypes A and B of B. tabaci.  相似文献

Intraspecific variation in four New Zealand species of Chionochloa, C. flavescens, C. pallens, C. rigida; and C. rubra, was investigated by examining the major carbon chain lengths of fatty acids, alcohols, aldehydes, wax esters and alkanes of the epicuticular waxes. The major even-carbon chain lengths ranged generally from C24 to C32 in the acids, alcohols and aldehydes; C29 to C33 in the alkanes; and even-carbon chains between C36 and C52 in the wax esters. A computer program was used to calculate the degree of similarity between samples in terms of chain length distribution. In C. rigida eastern and western South Island localities were identified; in C. flavescens Canterbury and Nelson, western South Island and southern North Island regions were recognized; and C. pallens and C. rubra were divisible into four regions; Canterbury, Nelson, western South Island and southern North Island. The possible elongation-decarboxylation pathways and the specificity of the enzymes in the biosynthetic pathways of epicuticular wax synthesis suggest the possibility that the northwest Nelson region could be a biogenetic centre from which wax synthesis has diversified along three routes, one to the western South Island, another to eastern South Island and the third to southern North Island. Identification of each of the four species based on the distribution of the carbon chain lengths in the individual lipid fractions is impossible unless the locality of collection is known. Intraspecific variation in lipid composition is not coincident with patterns of variation already reported.  相似文献

Pulsed field gel electrophoresis using OFAGE, TAFE, and CHEF systems has been used to more fully characterize karyotypic variation within the two closely related fungal species of Ophiostoma ulmi sensu lato. Twelve wild-type and laboratory strains, representing the less agressive species O. ulmi and both of the biotypes of the more aggressive species O. novo-ulmi were studied and their karyotypes determined. Depending on the strain, a minimum of four to a minimum of eight chromosomal DNA bands were present that fall into three distinct size classes, with one exception. Strain CESSI6K (O. novo-ulmi, North American aggressive subgroup) contains a unique chromosomal DNA band which comigrated near a Saccharomyces cerevisiae chromosome of 0.95 Mb. This unique band was the smallest O. ulmi s. l. chromosomal DNA observed. Seven of the twelve strains shared a common chromosomal DNA banding pattern, whereas each of the other five had a unique karyotype. There was no correlation between chromosome profile and species, as some O. novo-ulmi and O. ulmi strains shared common electrophoretic karyotypes.  相似文献

Droschia stebbingii (Green) belonging to the family Monophlebidae (Hemiptera: Coccoidea) is a serious, phytophagous and dimorphic pest. D. stebbingii and D. mangiferae (Green) with sixty-two common hosts and minor morphological variations pose problems towards their taxonomic identification. Light and scanning electron microscopy were used to outline the ultrastructural details of cuticular wax producing structures of D. stebbingii. Three types of wax-pores along with tubular duct were observed on the head, thorax and abdomen. Quinquelocular and multilocular disc-pores exhibited variations in the central loculus with single and double opening. Short curly wax strands were secreted from the outer loculi whereas central loculus secreted long tubular strands. Additionally, bilocular pores secreting curly wax were observed having two large loculi separated by a septum. GC–MS analysis of cuticular wax indicated the presence of hydrocarbons and wax esters forming majority of the covering along with long chain alcohols, ketones and acids. Variations in the percentage of hydrocarbon and wax esters content were observed for two populations of D. stebbingii collected from different hosts. Hydrocarbons were abundant in populations collected on Pongamia pinnata (L.) Pierre whereas long chain esters dominated the wax profile for the population collected from Tectona grandis L.f. Compositional variations in the cuticular wax of D. stebbingii on different host along with knowledge on the ultrastructural wax pores and waxy secretion would provide an insight to the existing knowledge of this scale insect.  相似文献

The cuticular waxes of leaves of Coffea arabica cv. ‘Catuaí Vermelho’, C. arabica cv. ‘Obatã’, Coffea canephora cv. ‘Apoatã’, Coffea racemosa and two hybrids between C. arabica and C. racemosa were extracted by rapid washing of the surface with chloroform. The waxes were fractionated by thin layer chromatography over silicagel. The fractions of the constituent classes were characterized by infrared spectroscopy and the distribution of the homologs of the n-alkanes and n-primary alcohols was determined by GC/MS and GC/FID. Among the samples analyzed, leaves of C. racemosa have the highest content of foliar wax (22.9 μg cm−2). Most samples contain either n-alkanes (C. canephora and C. racemosa) or n-primary alcohols (C. arabica) as predominant wax constituents. The distribution of n-alkanes allowed the distinction of C. racemosa from the other samples; the distribution of alcohols allowed the distinction of the three species. The two hybrids have waxes similar to the wax of C. arabica.  相似文献

We used randomly amplified polymorphic DNA (RAPD)-PCR to estimate genetic variation among isolates of Trichoderma associated with green mold on the cultivated mushroom Agaricus bisporus. Of 83 isolates examined, 66 were sampled during the recent green mold epidemic, while the remaining 17 isolates were collected just prior to the epidemic and date back to the 1950s. Trichoderma harzianum biotype 4 was identified by RAPD analysis as the cause of almost 90% of the epidemic-related episodes of green mold occurring in the major commercial mushroom-growing region in North America. Biotype 4 was more closely allied to T. harzianum biotype 2, the predominant pathogenic genotype in Europe, than to the less pathogenic biotype 1 and Trichoderma atroviride (formerly T. harzianum biotype 3). No variation in the RAPD patterns was observed among the isolates within biotype 2 or 4, suggesting that the two pathogenic biotypes were populations containing single clones. Considerable genetic variation, however, was noted among isolates of biotype 1 and T. atroviride from Europe. Biotype 4 was not represented by the preepidemic isolates of Trichoderma as determined by RAPD markers and PCR amplification of an arbitrary DNA sequence unique to the genomes of biotypes 2 and 4. Our findings suggest that the onset of the green mold epidemic in North America resulted from the recent introduction of a highly virulent genotype of the pathogen into cultivated mushrooms.  相似文献

Photoinhibition was examined in paraquat-resistant and paraquat-susceptible biotypes of Hordeum glaucum Steud., Hordeum leporinum Link., Arctotheca calendula (L.) Levyns., and Conyza bonariensis (L.) Cronq. Plants were photoinhibited at low temperature, and the extent of photoinhibition determined by O2 evolution and 77 K fluorescence. No difference in the degree of photoinhibition was detected between paraquat-resistant and paraquat-susceptible biotypes for any of the species examined. C. bonariensis plants were also photoinhibited by treatment without CO2 at either 21% (volume/volume) O2 or 4% (volume/volume) O2, and again no difference was observed between the paraquat-resistant and paraquat-susceptible biotypes in reduction of the ratio of variable fluorescence to maximal fluorescence. This is in contrast to a recent report (MAK Jansen, Y Shaaltiel, D Kazzes, O Canaani, S Malkin, J Gressel, [1989] Plant Physiol 91: 1174-1178 in which it was claimed that a paraquat-resistant biotype of C. bonariensis was more tolerant of photoinhibition than a paraquat-susceptible biotype. We conclude that paraquat-resistant biotypes of these plant species are not more tolerant of photoinhibition when compared with the paraquat-susceptible biotypes.  相似文献

Invasive events by Bemisia tabaci (Gennadius) biotypes in various parts of the world are of continuing interest. The most famous is biotype B that has caused great economic losses globally. In addition, biotype Q has also recently been reported to be a new invasive pest. These two biotypes have been monitored for some time in the Western North Pacific region, but the invasive events and population genetic structures of these two biotypes are still not clear in this region. In this study, the mitochondrial cytochrome oxidase I (COI) gene was used to reconstruct a phylogenetic tree for identifying biotypes B and Q and to study the relationships between invasive events and ornamental plants. Population genetic analyses of mtCOI sequences were also used to study the genetic relationships within and between populations. A combination of a phylogenetic tree and haplotype analysis suggested the recent invasion of biotype Q in this region is related to the international ornamental trade from the Mediterranean region. Low levels of haplotype diversity and nucleotide diversity indicate that the presence of biotypes B and Q in the Western North Pacific region are caused by multiple invasions. Hierarchical analysis of molecular variance supports the hypothesis of multiple invasions. In addition, high sequence identities and low genetic distances within and between populations of the two biotypes revealed that these invasive events occurred recently. The low levels of genetic differentiation revealed by pairwise F (ST) values between populations also suggests the invasions were recent. Therefore, results of this study suggested that biotypes B and Q entered this region through multiple recent invasions. A quarantine of agricultural crops may be necessary to prevent further invasions.  相似文献

The possibility of increased invasiveness in cultivated varieties of native perennial species is a question of interest in biofuel risk assessment. Competitive success is a key factor in the fitness and invasive potential of perennial plants, and thus the large-scale release of high-yielding biomass cultivars warrants empirical comparisons with local conspecifics in the presence of competitors. We evaluated the performance of non-local cultivars and local wild biotypes of the tallgrass species Panicum virgatum L. (switchgrass) in competition experiments during two growing seasons in Ohio and Iowa. At each location, we measured growth and reproductive traits (plant height, tiller number, flowering time, aboveground biomass, and seed production) of four non-locally sourced cultivars and two locally collected wild biotypes. Plants were grown in common garden experiments under three types of competition, referred to as none, moderate (with Schizachyrium scoparium), and high (with Bromus inermis). In both states, the two “lowland” cultivars grew taller, flowered later, and produced between 2x and 7.5x more biomass and between 3x and 34x more seeds per plant than local wild biotypes, while the other two cultivars were comparable to wild biotypes in these traits. Competition did not affect relative differences among biotypes, with the exception of shoot number, which was more similar among biotypes under high competition. Insights into functional differences between cultivars and wild biotypes are crucial for developing biomass crops while mitigating the potential for invasiveness. Here, two of the four cultivars generally performed better than wild biotypes, indicating that these biotypes may pose more of a risk in terms of their ability to establish vigorous feral populations in new regions outside of their area of origin. Our results support an ongoing assessment of switchgrass cultivars developed for large-scale planting for biofuels.  相似文献

Onion thrips is a major threat to onion crop throughout the world. It is a potential vector of Iris yellow spot virus and causes significant economic damage to bulb production. Phenotypic and biochemical traits of onion cultivars were assessed against Thrips tabaci. Onion Gawran LR-241 (OG) cultivar was tolerant against the infestation of T. tabaci whereas Onion White (OW) was susceptible. Number and size of stomata, cuticle thickness, cell wall thickness and surface wax of onion leaves were studied with the help of scanning electron microscope and quantitative and qualitative analysis was carried out to estimate epicuticular wax and other bio-chemical components through GC/MS. Onion Gawran has thick cell wall, sharp and dense wax crystals, wider central angle and small sized stomata compared to other cultivars. Epicuticular wax components of OG cultivar were heptacosane (5.2%), octacosanol-1 (9.2%), 2-methyl octacosane (4.2%), heptadecanol-1 (5.2%), hexacosanol-1 (4.2%), azulene, 1,4-dimethyl-7-(1-methylethyl) (36.9%), hexadecanoic acid (1.95%), heptadecane (4.2%), triacontanol-1 (5.8%) and hentriacontanone-16 (23.40%). Azulene, 1, 4-dimethy-l-7-(1-methy-l-ethyl) was only found 36.9% in OG but absent in other three cultivars. 2-methyl octacosane was absent in Poona Red Desi and OW cultivars. Hentriacontanone-16, 2-methyl octacosane, fatty alcohols (Octacosanol-1 and Triacontanol-1) and azulene, 1, 4-dimethy-l-7-(1-methy-l-ethyl) were effective in the formation of epicuticular wax in onion cultivars. It implies that phenotypic and biochemical characteristics of OG cultivar proved as resisting features to T. tabaci.  相似文献

The germination ecology of Ambrosia artemisiifolia and A. trifida glyphosate susceptible biotypes sampled in marginal areas, was compared with that of the same species but different biotypes suspected of glyphosate resistance, common and giant ragweed, respectively. The suspected resistant biotypes were sampled in Roundup Ready® soybean fields. Within each weed species, the seeds of the biotype sampled in marginal area were significantly bigger and heavier than those of the biotype sampled in the soybean fields. A. artemisiifolia biotypes exhibited a similar dormancy and germination, while differences between A. trifida biotypes were observed. A. artemisiifolia biotypes showed similar threshold temperature for germination, whereas, the threshold temperature of the susceptible A. trifida biotype was half as compared to that of the resistant A. trifida biotype. No significant differences in emergence as a function of sowing depth were observed between susceptible A. artemisiifolia and suspected resistant A. trifida biotype, while at a six-cm seedling depth the emergence of the A. artemisiifolia susceptible biotype was 2.5 times higher than that of the A. trifida suspected resistant biotype. This study identified important differences in seed germination between herbicide resistant and susceptible biotypes and relates this information to the ecology of species adapted to Roundup Ready® fields. Information obtained in this study supports sustainable management strategies, with continued use of glyphosate as a possibility.  相似文献

Host-handling behaviors of Eretmocerus mundus (Mercet) (Hymenoptera: Aphelinidae) toward first to fourth instar nymphs of the Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) B and Q biotypes and of Trialeurodes vaporariorum (Westwood) (Homoptera: Aleyrodidae) were compared under laboratory conditions. The host acceptance rates differed among host stages but not among host species or biotypes. In all host species and biotypes, the parasitoids stopped their host-handling behavior before oviposition more often with older hosts. The total host-handling time was longer for the fourth instars than for younger instars, mainly due to the longer oviposition times and probing times. E. mundus showed marking behavior for the B. tabaci B and Q biotypes, but not for T. vaporariorum. In conclusion, E. mundus could parasitize all whitefly biotypes and species used in this study, but both B. tabaci biotypes were more suitable hosts than T. vaporariorum. The younger instars were good oviposition targets in all hosts.  相似文献

The type specimen of the gastornithid Zhongyuanus xichuanensis Hou, 1980, from the Early Eocene of Henan (central China) is redescribed and compared with European and North American representatives of Gastornis. Differences between the Chinese specimen (the distal end of tibiotarsus) and its European and North American counterparts are of a minor nature, and it is concluded that it can be referred to as Gastornis xichuanensis (Hou, 1980). Stratigraphic evidence suggests that Gastornis originated in Europe, where it is known as early as the Paleocene. Whether the Asian Gastornis dispersed from North America via the Bering land bridge, or directly from Europe across the Turgai Strait, remains uncertain.  相似文献

The proportion of foragers in ant colonies is a fairly constant species-specific characteristic that could be determined by intrinsic or extrinsic factors. If intrinsic factors are relevant, species with similar life history characteristics (e.g., colony size and foraging strategies) would be expected to have a similar proportion of foragers in their colonies. Within the genus Pogonomyrmex, North American species can vary largely in their colony size, whereas only species with small colonies are known in South America. We studied the characteristics of the foraging subcaste in three sympatric South American species of Pogonomyrmex harvester ants, and compared it with the available information on other species of the same genus. We used two mark-recapture methods and colony excavations to estimate the number and proportion of foragers in the colonies of P. mendozanus, P. inermis, and P. rastratus, and to test the relationship between forager external activity levels and abundance per colony. Forager abundance in the three studied species was lower than in most North American species. The percentage of foragers in their colonies ranged 7–15 %, more similar to North American species with large colonies than to those with small colony size. Foraging activity was positively correlated with forager abundance in all three species, implying that colony allocation to number of foragers allows for higher food acquisition. Further comparative studies involving a wider range of traits in South and North American species would allow to unveil the role of environmental factors in shaping each species’ particular traits.  相似文献

This investigation was aiming at the phytochemical characterisation of Origanum vulgare subsp. vulgare from Austria. To study the qualitative and quantitative composition of essential oil compounds 374 individual plants were analysed via GC. The volatiles of Austrian O. vulgare subsp. vulgare were found to be complex mixtures of 53 mono- and sesquiterpenes. Among the sesquiterpenes β-caryophyllene, germacrene D, E-E-α-farnesene, germacrene D-4-ol and caryophyllene oxide were frequently present in higher amounts. The monoterpenes were mainly made up of sabinyl-compounds [mainly sabinene (up to 48.4%) and cis-sabinene hydrate (up to 57.8%)] and/or cymyl-compounds [mainly p-cymene (up to 49.7%), γ-terpinene (up to 21.5%) and carvacrol (up to 32.9%)] that were accompanied by usually smaller amounts of bornyl-compounds and acyclic compounds. Some exceptional (in O. vulgare rare) chemotypes were detected. The essential oil content of Austrian O. vulgare subsp. vulgare ranged between 0.1 and 1.8%. The content of rosmarinic acid was analysed by HPLC and ranged from 0.6 mg/g dry mass up to 37.2 mg/g dry mass. No arbutin could be detected in the analysed populations.  相似文献

Kloeckera apiculata 34-9 was selected from the rhizosphere soil for its high efficacy in controlling citrus green and blue mold. In this study, the effect of the antagonistic yeast K. apiculata on citrus cuticular wax was investigated in ripening Newhall navel orange (Citrus sinensis L. Osbeck). Our results show that K. apiculata triggers the production of cuticular waxes and surface wax morphology changes in the fruit surface. 15 K. apiculata-responsive differentially expressed genes (DEGs) were identified for wax metabolism by using the Affymetrix citrus genome GeneChip. Using GC–MS, 46 wax compounds were found in the Newhall fruit surface. On one hand, esters including docosanoic acid, 1,2,3-propanetriyl ester and 9-hexadecenoic acid, 9-octadecenyl ester were up-regulated approximately twofold in the treatment condition compared with the control; and on the other hand, the fatty acids and fatty alcohols decreased by 74.4% and 72.3%, respectively, in the treatment condition. The new wax production and increased hydrophobicity of the Newhall surface resulting from the treatment may influence spore adhesion and germination. Furthermore, the response of the fruit waxes to the K. apiculata stimulus is likely to be regulated by intra-cellular H2O2 signaling. This study demonstrated the response fruit waxes to K. apiculata in Newhall navel oranges, thus providing new clues that aid our understanding of the mechanisms of action of antagonistic yeasts in postharvest fruits.  相似文献
