Seventy-two lactating Chios ewes were used in two experiments to determine effects of supplemental dietary biotin on productivity and milk composition. The first experiment started after weaning on day 42 postpartum, and lasted 20 weeks, while the second started on week 24 postpartum and lasted 12 weeks. In both experiments, ewes were allocated, after equal distribution relative to milk yield, body weight, time of lambing, and lactation number (i.e., two or three), into three groups of 24 ewes each, and were accommodated in three floor pens/groups of eight ewes/treatment. Ewes were fed one of three diets varying in supplemental biotin (BIOTIN0, no supplemental biotin; BIOTIN3, 3 mg supplemental biotin/ewe/day; BIOTIN5, 5 mg supplemental biotin/ewe/day) in each experiment. Milk, fat, protein, lactose and ash yield, and milk fat content increased linearly (P<0.012) with increased dietary biotin in both studies. Dietary biotin supplementation improved the productive performance of these lactating ewes at both an early and late stage of lactation.  相似文献

The correlations between some meteorological parameters and fertility data were evaluated in Barki x Rahmani crossbred ewes using the records of five consecutive years (2003-2007). Additionally, estrus detection and ultrasonic evaluation were applied on eighteen mature dry ewes during breeding and non-breeding seasons. The effect of lactation was evaluated by monitoring estrus behavior in ninety four lactating ewes from 40 to 120 d after parturition. Moreover, ultrasonography was used to identify ovarian activity in six cyclic and six acyclic non-lactating ewes. Results revealed that relative estrus occurrence and fertile mating were positively correlated (P <0.05) with high temperature and long photoperiod (conditions of summer season), and were negatively correlated (P < 0.01) with rainfall (condition of winter season). During breeding season, estrus rate, serum progesterone concentration, and diameter of largest follicle were significantly (P < 0.05) higher than those observed during the non-breeding season. Furthermore, month of parturition had a significant effect (P < 0.05) on estrus rate of lactating ewes where ewes that lambed in August, September, and October recorded higher estrus rate than those lambed in November and December. However, the lactational strength did not exert any deleterious effect on the reproductive performance of lactating ewes.In conclusion, in Egypt under subtropical conditions, Barki x Rahmani crossbred ewes exerted optimum estrus behavior and fertile mating during summer season. The reduction in estrus activity during lactation was due to the seasonal effect rather than lactational stress.  相似文献

The influence of (1) lactation and (2) three different hormone treatments were measured in fall lambing ewes. By day 67 postpartum, considering lactation effect, 89% of nonlactating and 33 % of lactating ewes showed estrous behavior (P<0.05). Nonlactating control ewes ovulated significantly sooner postpartum than ewes in other treatment groups. Both nonlactating and lactating ewes ovulated prior to demonstrating estrus. In the hormone treatments those containing estrogen tended to have earlier estrus which is assumed to reflect a direct effect. Uterine involution in all animals was generally complete by day 24. No apparent treatment differences in uterine epithelial cells, gland number and size, edema, or interstitial hemorrhage were noted during the 67-day postpartum period studied. Data from this study indicate that the reproductive tract appears acceptable for rebreeding several weeks before the ewes show behavioral estrus.  相似文献

An experiment was performed to study the glucose kinetics of crossbred Holstein cattle feeding on either hay or 5% urea-treated rice straw during early lactation (30 days post partum), mid-lactation (120 days post partum) and late lactation (210 days post partum). Two breeds: Holstein FriesianxRed Sindhi (50:50 = 50% HF) and Holstein FriesianxRed Sindhi (87.5:12.5 = 87.5% HF) were used. In early lactating 87.5% HF animals feeding on either hay or urea-treated rice straw, the high milk yields and lactose secretion were related to glucose uptake by the udder and udder blood flow as compared with those of 50% HF animals. Marked decreases in udder blood flow, glucose uptake, lactose secretion and milk yield were apparent in mid- and late lactation of both groups of 87.5% HF animals. In contrast, both groups of 50% HF animals showed no significant changes in udder blood flow, udder glucose uptake, lactose secretion and milk yields throughout the course of lactation. Total glucose entry rate using 3-[3H] glucose infusion, recycling of glucose carbon and plasma glucose clearance significantly increased during late lactation for 50 and 87.5% HF animals feeding on urea-treated rice straw. The utilization rates of glucose using [U-(14)C] glucose infusion were not significantly different among groups of animals and periods of lactation. It can be concluded that 87.5% HF animals have the genetic potential for a higher milk yield, but a shorter peak yield and poorer persistence in comparison with 50% HF animals. Changes in the utilization of glucose by the mammary gland for milk production in both groups of crossbred animals during feeding on either hay or urea-treated rice straw would be dependent on intramammary changes.  相似文献

Two separate experiments were conducted to examine the effects of exogenous oestrogen on selectivetransfer of IgG1 into mammary secretion of ewes. In one experiment, non-pregnant ewes were induced to lactate artificially by first developing mammary glands with injections of progesterone plus low doses of oestrogen then triggering milk secretion with either glucocorticoid or high doses of oestrogen. In the other experiment, lactating ewes were injected with oestrogen each day for 6 days. The results of the experiments suggest that oestrogen affects selective transfer of IgG1 into mammary secretion of the ewes. Moreover, the results show that, in the absence of high levels of oestrogen in blood, the magnitude of the selective transfer of IgG1 into mammary secretion is related inversely to the synthetic activity of the glandular epithelium.  相似文献

Responses to daily injections of bovine growth hormone (GH, 0.15 mg kg-1 liveweight), beginning on day 10 of lactation, were measured in lactating ewes. Milk yields of GH-treated ewes increased soon after commencement of injections and continued to increase for some 25 days before reaching plateau levels. By comparison, yields of ewes injected with excipient (controls) decreased over the experiment. There was a tendency for contents of milk fat to be higher and milk protein to be lower for GH-treated than for control ewes during the first 15-20 days after injections were started. At the beginning and over the first 15-20 days of the experiment feed intakes of both groups of ewes were similar, but thereafter intakes of GH-treated ewes gradually increased to reach plateau levels some 200-300 g day-1 higher than for control ewes by about day 35. Liveweights of both groups of ewes decreased during the first 2 weeks of treatment then increased, with GH-treated ewes losing, then gaining, more weight than control ewes. The efficiency of food utilization for milk production was higher for GH-treated than control ewes throughout the experiment but digestibility of food organic matter was not different during the eighth week of the experiment. At the end of the experiment, body composition, assessed by dilution of tritiated water, was similar for both groups of ewes. Differences in milk production were not sustained after withdrawal of GH injections. Measurements of tissue uptake of key metabolites were made on days 3 and 45 of GH treatment. On day 3, GH lowered uptake of glucose and non-esterified fatty acids by leg muscle tissue and increased mammary uptake of non-esterified fatty acids. By day 45 there were no apparent differences of tissue uptake of key metabolites. The results indicate that there is a biphasic response to exogenous GH in the lactating ruminant. It appears that initially GH affects nutrient partition thereby increasing supplies to the mammary gland of key nutrients for milk synthesis. In the longer term, GH increases feed intake, which provides sufficient nutrients to sustain increased milk production and also liveweight gain.  相似文献

A study based on 12 pregnant and six dry Barki ewes was carried out to examine the changes in blood constituents during pregnancy and lactation periods. The blood parameters were blood hemoglobin, packed cell volume percent (PCV%), mean corpuscular hemoglobin concentration (MCHC), glucose, aspartate aminotransaminase (AST or GOT), alanine aminotransaminase (ALT or GPT), total plasma protein, albumin, globulin, albumin to globulin ratio (A/G), urea and creatinine. During pregnancy all these parameters started to increase significantly, but in different stages, reaching maximum values at parturition. In contrast, dry ewes showed almost stable values during the experimental period. From 10th week to parturition, PCV% and MCHC increased (P<0.01) in pregnant ewes, which resulted in increased (P<0.01) blood hemoglobin. Blood glucose increased from the 4th week of pregnancy to reach its maximum at parturition (60.15–90.08 mg/dl). The two transaminases increased significantly from the 2nd week (52.23–65.02 IU for AST and 8.02–15.12 IU for ALT). Plasma protein with its two components, albumin and globulin, increased significantly at the 6th week, but dropped throughout the 16–18th week of pregnancy. Urea and creatinine began to increase significantly after 10–12 weeks of pregnancy (from 54.73 to 72.11 mg/dl for urea and from 0.882 to 2.475 mg/dl for creatinine). During the first month of lactation, PCV decreased sharply in lactating ewes and was significantly lower than in dry ewes at the 3rd week of lactation (24.25 versus 27.17%), which resulted in a drop in blood hemoglobin at the 4th week (68.42 versus 74.00 g/l). However, lactating ewes maintained significantly higher values of MCHC (30.01–31.19% for lactating versus 29.87–27.48% for dry). In lactating ewes, levels of glucose, ALT, urea and creatinine returned to levels comparable to those in dry ewes. The same occurred with total plasma proteins, mainly due to a sharp decrease in globulin, while albumin remained higher than in dry ewes with a slow decline, which resulted in higher values of A/G ratio during lactation. Aspartate aminotransferase remained higher than in dry ewes.  相似文献

Supplementation of pregnant ewes with long-chain n-3 polyunsaturated fatty acids (PUFA) demonstrably improves indicators of neonatal lamb vigour, potentially improving the number of lambs reared per ewe. The present study investigated the effect of supplementing ewes with fish oil and vitamin E (α-tocopherol acetate) throughout both pregnancy and lactation on the performance of lactating ewes and sucking lambs. Forty-eight ewes were supplemented with one of four concentrates containing either Megalac or fish oil plus a basal (50 mg/kg) or supranutritional (500 mg/kg) concentration of vitamin E from 6 weeks pre-partum until 4 weeks post partum in a two-by-two factorial randomised-block design. All concentrates were formulated to contain approximately 60 g/kg supplemental fatty acids. Ewes were housed, penned on sawdust and offered straw ad libitum. Blood samples were taken from ewes and lambs at intervals throughout the experiment and milk samples were obtained at 21 days into lactation. There was no notable effect of dietary vitamin E concentration upon ewe or lamb performance. Ewe dry-matter (DM) intake and yield were unaffected by dietary treatment, although ewes fed fish oil lost less weight during lactation (-1.88 kg compared with -3.97 kg for Megalac-supplemented ewes; P < 0.01). Milk fat concentrations (67.3 g/kg compared with 91.8 g/kg; P < 0.01) and yields (6.65 g/h v. 9.26 g/h; P < 0.01) were reduced in ewes fed fish oil and these decreases were associated with lower litter-growth rates (0.49 g/day compared with 0.54 g/day; P < 0.05). Milk protein yield was increased by fish oil supplementation (3.82 g/h) compared with Megalac supplementation (3.28 g/h; P < 0.05); moreover, there was an interaction between fat source and vitamin E concentration in that both protein concentration and yield were significantly lower in milk from ewes fed treatment with Megalac + basal vitamin E (MB) compared with the other three treatments. Fish oil supplementation increased the concentrations of C18:1 trans-, cis-9, trans-11 conjugated linoleic acid (CLA), C20:5 (n-3) and C22:6 (n-3) within ewe plasma, milk and lamb plasma. The mechanisms by which fish oil supplementation affects milk composition warrants further investigation.  相似文献

In female ungulates lactation carries a high energetic cost and mothers often devote more care to sons than to daughters. The aims of this study were to determine whether lactating bighorn ewes have higher foraging time, bite rate or selectivity of forage than barren ewes and whether reproductive status affects migration patterns. Ewes with male lambs were predicted to spend more time foraging and to have a higher bite rate than ewes with female lambs. There were no differences in foraging behavior according to ewe reproductive status from April to August. In September-November lactating ewes had a higher bite rate and spent more time foraging than nonlactating ewes but lamb gender did not affect foraging behavior. Lactating ewes gained less weight than nonlactating ones until mid-August; from mid-August to late September mass gain was similar for both groups of ewes. Nonlactating ewes spent more time standing but reproductive status did not affect vigilance behavior or step rate while foraging. Ewes with lambs did not differ from nonlactating ewes in step rate. Pregnant ewes migrated earlier than barren ewes to alpine areas in spring. By so doing they abandoned areas with good quality forage presumably to give birth in areas safer from predation. All ewes spent most of the summer in the alpine range but nonlactating ewes returned to the winter range earlier than lactating ewes, probably to profit from the abundant forage there.  相似文献

Specific changes in milk composition during lactation in the tammar wallaby (Macropus eugenii) were correlated with the ages of the developing pouch young (PY). The present experiment was designed to test the hypothesis that the sucking pattern of the PY determines the course of mammary development in the tammar wallaby. To test this hypothesis, groups of 60-day-old PY were fostered repeatedly onto one group of host mothers so that a constant sucking stimulus on the mammary gland was maintained for 56 days to allow the lactational stage to progress 42 days ahead of the age of the young. Analysis of the milk in fostered and control groups showed the timing of changes in the concentration of protein and carbohydrate were essentially unaffected by altering the sucking regime. The only change in milk protein secretion was a small delay in the timing of down-regulation of the secretion of whey acidic protein and early lactation protein in the host tammars. In addition, the rates of growth and development of the foster PY were significantly increased relative to those of the control PY because of ingesting more milk with a higher energy content and different composition than normal for their age. The present study demonstrates that the lactating tammar wallaby regulates both milk composition and the rate of milk production and that these determine the rates of PY growth and development, irrespective of the age of the PY.  相似文献

Orexins are hypothalamic peptides implicated in the regulation of ingestive and other behaviours. Here we investigated prepro-orexin expression and hypothalamic orexin-A and -B levels in lactating rats, which display marked hyperphagia, with or without food restriction for 2 days or treatment with bromocriptine, which inhibits milk production and thus reduces the energy losses of lactation. Neither prepro-orexin gene expression nor hypothalamic orexin-A peptide levels were changed in any of these lactating groups compared with age-matched virgin controls. However, hypothalamic orexin-B levels were significantly higher in lactating rats that were food-restricted for 2 days (P<0.05) compared with non-lactating controls and with lactating rats that were either freely-fed or bromocriptine-treated. Thus, food restriction superimposed on lactation selectively increases hypothalamic orexin-B levels, suggesting that orexin-A and -B may be differentially released or cleared. Changes in orexin-B availability may influence physiological activities other than energy homeostasis, perhaps inducing arousal.  相似文献

Twenty-two polytocous lactating Chios ewes were used to test the effects of bovine somatotropin (bST) on reproduction, progesterone concentration and LH secretion during estrus. Half of the ewes were injected every second week with 160 mg bST in a prolonged release vehicle, from the fifth day post partum until the end of lactation, while the remaining ones were used as controls. All animals were fed the same amount of ration. Supplementation with bST resulted in an increase of milk production (P<0.05) and an insignificant trend for delayed resumption of normal estrous cycles. Although there were no differences between groups, there was also a tendency for the bST group to display lower progesterone concentrations during the first three fortnights after the onset of normal estrous cycles and higher ones during the last three fortnights of the experiment, compared with the control group. Duration of the first normal luteal phase after delivery of the bST group was found to be shorter compared with the control group (P<0.05). After estrous synchronization the bST group showed a shorter estrus compared with the control group (P<0.05). Average and baseline LH concentrations during synchronized estrous in the bST group was lower (P<0.001) compared with the control group. Additionally, the conception rate did not differ between the two groups. This study supports the concept that the beneficial effects of bST treatment on milk production outweigh the potential deleterious effects on reproduction.  相似文献

The means by which calcium is transported into the milk produced by mammary glands is a poorly understood process. One hypothesis is that it occurs during exocytosis of secretory products via the Golgi pathway, consistent with the observation that the SPCA1 Ca2+-ATPase, which is expressed in the Golgi, is induced in lactating mammary tissue. However, massive up-regulation of the PMCA2bw plasma membrane Ca2+-ATPase also occurs during lactation and is more strongly correlated with increases in milk calcium, suggesting that calcium may be secreted directly via this pump. To examine the physiological role of PMCA2bw in lactation we compared lactating PMCA2-null mice to heterozygous and wild-type mice. Relative expression levels of individual milk proteins were unaffected by genotype. However, milk from PMCA2-null mice had 60% less calcium than milk from heterozygous and wild-type mice, the total milk protein concentration was lower, and an indirect measure of milk production (litter weights) suggested that the PMCA2-null mice produce significantly less milk. In contrast, lactose was higher in milk from PMCA2-null mice during early lactation, but by day 12 of lactation there were no differences in milk lactose between the three genotypes. These data demonstrate that the activity of PMCA2bw is required for secretion of much of the calcium in milk. This major secretory function represents a novel biological role for the plasma membrane Ca2+-ATPases, which are generally regarded as premier regulators of intracellular Ca2+.  相似文献

After intravenous administration of alloxan (50 mg kg-1 liveweight) to lactating ewes, there were triphasic changes in plasma glucose and insulin. Almost immediately, plasma insulin decreased and hyperglycaemia occurred, then, between c. 5-12 h, insulin increased and ewes became hypoglycaemic. Thereafter, insulin decreased and glucose increased from c. 20 h after alloxan and the diabetic state was established. Changes in glucose production and utilization correlated with changes in plasma glucose. Exogenous insulin was administered from 30 h after alloxan, and it took some 2 weeks to stabilize ewes. During this period, when mild hyperglycaemia persisted, milk yields and feed intakes were decreased but milk fat content was elevated. Once ewes were stabilized, plasma glucose, milk yield, feed intake and milk fat content returned to levels prior to alloxan. These observations are consistent with insulin playing a role in the aetiology of the 'low milk fat syndrome' in the ruminant. It appears that the alloxan-treated, insulin-stabilized ewe would be a useful model for studying the role of insulin during lactation, but it is necessary to allow time for animals to overcome effects of administration of alloxan.  相似文献

Early lactation parameters are difficult to estimate from commercial dairy records due to the small number of records available before the peak of production. A biological model of lactation was used with weekly milk records from a single Holstein herd to estimate these early lactation parameters and the secretion rate of milk from the average cell throughout lactation. A genetic analysis of the lactation curve parameters, calculated curve characteristics and secretion rate traits was undertaken. Early lactation traits were found to have little genetic variation and effectively zero heritability. Secretion rate traits for milk, protein, lactose and water were all moderately heritable and highly genetically correlated (>0.87) but fat secretion rate had lower genetic correlations with the other secretion rates. A similar pattern of correlations was seen between total lactation yield traits for fat, protein, lactose and water. The genetic correlations between the lactation curve traits and the secretion rate traits were calculated. Total milk yield, peak yield and maximum secretion potential were all highly correlated with milk, lactose and water secretion rates but less so with fat and protein secretion rates. In particular, fat secretion rate had a moderate to low genetic correlation with these lactation curve traits. Persistency of lactation was highly correlated with fat and protein secretion rates, more persistent lactations being associated with lower rates of secretion of these milk components. Similar levels of heritability were found, where trait genetic parameters were directly equivalent to those derived from the same dataset by random regression methods. However, by using a biological model of lactation to analyse lactation traits new insights into the biology of lactation are possible and ways to select cows on a range of lactation traits may be achieved.  相似文献

The concentration of iron in all mammalian milks falls during lactation while the infant's iron requirement increases. Little is known, however, about the entry of iron into milk. Recently, transferrin receptors have been identified on lactating rat mammary plasma membranes, which may regulate iron entry into mammary tissue, potentiating its availability for subsequent transport into milk. This study was conducted to determine what relationship exists between the declining concentration of milk iron and the transferrin receptor concentration during various stages of lactation. Minimal transferrin receptors were detected in nulliparous rats. Total mammary transferrin receptor content increased during early and mid-lactation while milk iron concentration decreased. The continued appearance of high levels of transferrin receptors throughout lactation, without a concomitant increase in milk iron concentration, suggests a need for iron for functions other than cellular growth or secretion into milk to meet infant needs.  相似文献

The effect of L-DOPA on milk removal and on prolactin release during suckling or milking was studied in lactating ewes. Various doses of L-DOPA (25, 50, 100 and 200 mg per animal) were injected iv 30 min before the suckling or milking period. Control ewes were injected with 0.9% NaCl solution only. Milking induced a significant long-lasting release of prolactin. An inhibition of milk removal was obtained with the dose of 200 mg of L-DOPA. An inhibition of prolactin secretion was observed related to the dose of drug administered. The inhibitory effect of 200 mg of L-DOPA on the secretion of prolactin after milking lasted for about 120 min, and thereafter a significant increase in serum prolactin level occurred. This increase in serum prolactin was not due to a "rebound" effect of L-DOPA, since the milking stimulus had to be present to induce the delayed increase in prolactin. Doses of 25 or 50 mg of L-DOPA prevented the surge of prolactin observed immediately after milking, but a long-lasting release of prolactin was obtained thereafter. The inhibitory effect of L-DOPA on prolactin release could be overridden by the suckling or milking stimuli according to the dose administered. The suckling stimulus was more effective than milking in overriding the inhibitory effect of the low dose of L-DOPA. The results indicate that milk removal and prolactin release induced by milking or suckling in lactating ewes is inhibited by an increase in monoamines at the hypothalamic-hypophyseal level.  相似文献

Ghrelin, a novel acylated peptide and endogenous ligand for growth hormone (GH) secretagogue receptor, was originally isolated from rat and human stomachs. In addition to its GH-releasing activity, ghrelin plays an important role in many physiological functions, including food intake, gastric acid secretion, neonatal development, and so on. In this study, the effect of daily treatment with ghrelin on milk production was investigated in lactating rats and the development of the pups was monitored. Daily subcutaneous injection of ghrelin into nursing dams for 8 days from parturition caused a significant increase in milk yield and litter weight gain. When litters nursed by ghrelin-treated and saline-treated dams were interchanged on day 4 of lactation, the growth curves were reversed. Daily injections of ghrelin also increased plasma GH levels. Northern blot analysis revealed that daily injection of ghrelin significantly increased mammary casein mRNA expression. In addition, RT-PCR analysis showed that a ghrelin receptor was present in the mammary glands of lactating rats. These results suggest that ghrelin may play an important role in milk production in lactating dams.  相似文献

A single injection of ergocryptine (0.5 mg/kg liveweight) given to ewes 0.5-20 days prepartum or two injections (0.5 mg/kg liveweight per injection) given c. 30 and 10 days prepartum reduced concentrations of plasma prolactin to negligible (less than 5 ng/ml) values for 4 weeks after parturition, but did not affect concentrations of growth hormone and placental lactogen. Milking of treated ewes had no effect on concentrations of plasma prolactin during the first 4 weeks of lactation, but concentrations of growth hormone were increased during the 10-20 min period after milking. The half-life of prolactin in plasma was estimated as 21 min. In spite of the dramatic effect of ergocryptine on plasma prolactin all treated ewes secreted copious quantities of milk of normal composition. Mean daily yields of ewes treated with ergocryptine were not significantly different (P greater than 0.05) from those of untreated control ewes, but the mean +/- s.e.m. of total milk production over the first 3 weeks of lactation for ergocryptine-treated ewes was significantly lower (P less than 0.05) than that of control ewes (9.5 +/- 1.11 v. 14.1 +/- 1.20 kg milk). The results suggest that prolactin is not an essential component of the lactogenic and galactopoietic complexes of hormones in the ewe.  相似文献
