Peroxidases (POXs) are actively synthesised under stress conditions and induced by biotic/abiotic elicitors. Their age-regulated expression in plants has been a matter of investigation during recent times. The present study focuses on studying the expression of POX at different stages of growth and variation in its inducibility by aqueous extract of Azadirachta indica (neem) fruits. Tomato plants at 6, 8, 10 and 12 weeks were chosen for the study. The third nodal leaf of each plant was treated with neem fruit extract alone or in combination with Pseudomonas syringae pv. tomato. Sampling was performed at an interval of 24 h up to five days and after two weeks of treatment, from both the treated and distal untreated nodes of the plants. POX activity and its isoforms were analysed. The results demonstrated that POX expression and its inducibility by the biotic inducer (neem fruit extract) varies with age of the plant.  相似文献

This study deals with the use of peroxidases (POXs) from Allium sativum, Ipomoea batatas, Raphanus sativus and Sorghum bicolor to catalyze the degradation of free phenolic compounds as well as phenolic compounds contained in wastewater from leather industry. Secretory plant POXs were able to catalyze the oxidation of gallic acid, ferulic acid, 4-hydroxybenzoic acid, pyrogallol and 1,4-tyrosol prepared in ethanol 2% (v:v). Efficiency of peroxidase catalysis depends strongly on the chemical nature of phenolic substrates and on the botanical source of the enzymes. It appeared that POX from Raphanus sativus had the highest efficiency. Results show that POXs can also remove phenolic compounds present in industrial wastewater such as leather industry. Removal of phenolic compounds in wastewater from leather industry by POX was significantly enhanced by polyethylene glycol.  相似文献

During the mid-1980s, Sitobion avenae became recognised as an important vector of barley yellow dwarf virus (BYDV) in the Vale of York. A field trial at the University of Leeds Farm, North Yorkshire, was carried out during the autumn/winter of 1984-85 to evaluate different control procedures against S. avenae-transmitted BYDV and to investigate its epidemiology. Winter barley was sown on three dates in September, and plots were sprayed with either deltamethrin, demeton-S-methyl or pirimicarb on one of three dates between mid-October and mid-November, making a factorial design. Rhopalosiphum padi, the main vector of BYDV in southern England, were rarely found during the experiment, but the numbers of S. avenae were much higher, reaching a peak of 21% of plants infested in the unsprayed plots of the first sowing date. Single applications of each insecticide reduced populations of S. avenae to zero. Some treatments, particularly in the early sown plots and those treated with pirimicarb, however, did allow some recolonisation, and thus led to increased virus incidence and decreased yields. Sprays applied before the end of the migration of S. avenae were more efficient at controlling BYDV if the insecticide was persistent, otherwise a spray after this period, in November, was more effective. Virus incidence, although reduced by sprays, was generally low in plots sown on 18 and 27 September. In contrast, about 11% of plants were infected in unsprayed plots sown on 6 September and a small yield benefit was obtained with insecticidal treatments. Enzyme-linked immunosorbent assay (ELISA) of plants taken from the plots indicated that MAV- and PAV-like strains were present, and were most likely to have been transmitted by S. avenae.  相似文献

Settling and take-off behaviour of Sitobion avenae was studied in the laboratory using infested and uninfested wheat at a range of developmental stages. Of the later developmental stages of wheat alate S. avenae preferred to settle on and gave birth to relatively more offspring on flowering and watery-ripe plants. The time to take-off of alatae which matured on wheat was influenced by both the developmental stage of their host plant and the degree of crowding they experienced. All alatae flew but those isolated on flowering plants stayed longer than those crowded or isolated on any of the other developmental stages tested. Similarly the number of offspring produced before take-off was inversely related to the degree of crowding and positively to the time spent on a plant before take-off.  相似文献

1 The effect of reducing the growth of winter wheat on population size and development of the grain aphid Sitobion avenae was studied. 2 Automatic, mobile, crop shading devices, which reduced radiation comparable to levels found on a cloudy day, whilst minimizing other climatic changes, were used to reduce crop growth. Shading was applied between either GS 31 and 39 (1st node detectable → flag leaf ligule just visible) or GS 39 and 55 (flag leaf just visible → 50% of inflorescence emerged). 3 Sitobion avenae populations were initiated at GS 55 and highest populations were subsequently observed on plants shaded between GS 31 and 39. 4 Individual aphids confined within clip cages were observed to measure development from birth to adult moult, adult lifespan and fecundity. Results indicated that adult lifespan was shortest on plants shaded between GS31 and 39, whilst there were no differences in development time or fecundity. 5 Possible explanations for these observations are discussed.  相似文献

In our investigation, auxins (IAA, IBA and NAA) effectively promoted rooting in soybean hypocotyls. The activity of anionic peroxidase (POX) (pI 3.7) and cationic POX (pI 8.5) was significantly suppressed by exogenous auxins on day 2 (the inductive phase). Some particular anionic POXs (pI 4.0 and pI 5.3) significantly increased in IBA-treated tissues as compared with the control when the incubation time was prolonged to day 3 and day 4 (the initiation phase). We sequenced 5′-flanking region of pI 8.5 and pI 5.3 POX genes using the PLACE and PlantCARE databases to identify several potential cis-regulatory elements. The pI 8.5 POX gene promoter contained two sites that were homologous to sequences commonly found in auxin response elements; motifs ARF/AuxRE and CATATGGMSAUR. During the inductive phase, the activity of pI 8.5 POX was significantly suppressed by the exogenously applied auxins. The pI 8.5 POX gene promoter contained both ARF/AuxRE and CATATGGMSAUR motifs that responded to auxins earlier than the pI 5.3 POX gene. Hence, the pI 8.5 POX gene might belong to primary auxin response genes. The pI 5.3 POX gene, which responded to auxins a day or two later, contained only ARF/AuxRE motif. Moreover, unlike pI 8.5 and pI 3.7 POXs that were suppressed by auxins, the pI 5.3 POX was induced or enhanced by the applied auxins, especially IBA. The pI 5.3 POX might generate H2O2 which caused the auxin-induced growth at the initiation phase during the formation of adventitious root in soybean hypocotyls.  相似文献

DNA barcode (mitochondrial COI) sequences have allowed for species identification of aphids. In this study, we newly found a DNA barcoding problem in a part of the DNA sequences for Sitobion avenae. Five S. avenae individuals showed differences of, on average, 32.60% in the DNA sequences from other conspecific individuals, and a BLAST search revealed that the five sequences are similar to those of aphid parasitoids such as Aphidius, Ephedrus and Praon spp. (Hymenoptera: Braconidae). Based on these results, we concluded that the universal primers used in aphid DNA barcodes can amplify barcode sequences from parasitoid species within host aphids.  相似文献

The grain aphid, Sitobion avenae, is an economically important cereal pest worldwide. Aphid saliva plays an essential role in the interaction between aphids and their host plants. However, limited information is available regarding the proteins found in the saliva of S. avenae. Here, the watery saliva proteins from S. avenae were collected in an artificial diet and identified using a liquid chromatography–mass spectrometry/mass spectrometry analysis. A total of 114 proteins were identified in S. avenae saliva, including several enzymes, binding proteins, and putative effectors, as well as other proteins with unknown functions. In comparison with salivary proteins from nine other aphid species, the most striking feature of the salivary protein from S. avenae was the different patterns of protein functions. Several orthologous proteins secreted by other aphid species such as glucose dehydrogenase, elongation factors, and effector C002 were also detected in S. avenae saliva and speculated to play a significant role in aphid–plant interactions. These results provide further insight into the molecular basis between aphids and cereal plant interactions.  相似文献

为了评估紫外连续处理对第20代以后麦长管蚜Sitobion avenae (Fabricius)种群动态的影响,采用特定年龄生命表方法,设置不同强度(0,0.50,0.70 m W/cm2) UV-B连续多代处理麦长管蚜成虫,统计经紫外处理后的麦长管蚜第22代,第27代,第32代,第37代麦长管蚜生命表种群参数的变化并总结了1—37代麦长管蚜内禀增长率的变化规律。结果表明:(1)处理组供试麦长管蚜平均世代周期(T)呈先显著升高后下降的趋势。(2)低强度处理组内禀增长率(rm)、净增殖率(R0)、周限增长率(λ)均呈先升高后下降又升高的趋势,高强度处理组供试麦长管蚜内禀增长率(rm)、净增殖率(R0)、周限增长率(λ)呈先下降后显著升高的的趋势,且均在G37达到最大值。(3) 1—37代处理组麦长管蚜rm呈现先显著增高后下降又升高又下降又显著升高的变化规律,且均在G37达到最大值。由此可见:麦长管蚜经过多代的UV-B胁迫后,对其生命表参数的影响依然显著,在G37麦长管蚜对紫外的适应能力最强。本研究的创新点在于解析了第22代以后紫外胁迫对麦长管蚜种群动态的影响,从而为全面深入的阐述紫外胁迫对麦长管蚜生长繁殖的影响规律,掌握不同紫外胁迫下的剂量效应关系及麦长管蚜的适应性提供实验基础和理论依据。  相似文献

Secretory class III plant peroxidases (POXs) catalyze the oxidation of various reductants, and are encoded by a large multigene family. In rice, 42 independent expressed sequence tags for POXs have been identified. By RNA gel blot analysis using specific probes, we show here that 21 rice POX genes are unique in their developmental, organ specific and external stimuli-responsive expression. This would suggest that encoded POX isoenzymes are involved in a broad range of physiological processes in rice plants, individually.  相似文献

为了探究高压静电场对麦长管蚜Sitobion avenae Fabriciusde的影响。采用特定年龄生命表的方法,设置2、4、6 k V/cm辐射强度处理小麦种子和出生24h内的1龄若蚜。统计连续20代电场胁迫后麦长管蚜生命表种群参数、繁殖参数以及存活率的变化。结果表明:(1)经高压静电场处理后麦长管蚜的内禀增长率(r)在2 k V/cm和4 k V/cm强度时受影响显著(P0.05),但两者表现的世代不同,2 k V/cm和4 k V/cm的最低值分别出现在第20代和第10代。(2)麦长管蚜的净增值率(R0)、平均世代周期(T)、繁殖力(F)、成虫寿命和产仔天数等参数均呈现前期受高压静电场影响各处理组均显著低于对照组(P0.05),随着世代的增加高压静电处理组与对照组无显著差异。(3)麦长管蚜的特定年龄阶段存活率曲线受世代和高压静电场强度的影响显著。研究结论:麦长管蚜的生长发育和繁殖受世代和高压静电场强度的双重影响,且两者具有交互效应。研究的创新点在于:模拟自然条件下(同时对植物和昆虫进行高压静电场辐射处理)高压静电场对麦长管蚜的影响,为蚜虫生态遗传进化和生态控蚜提供理论依据。  相似文献

Differences were observed in the total fecundity and patterns of reproduction of four morphs of Sitobion avenae (F.). The reproductive rates of apterous and alate exules declined gradually with time but decreased rapidly in gynoparae and oviparae. Under short daylengths apterous exules gave birth in three phases; the first consisting mainly of gynoparae, the second of males and the third of non-gynoparous viviparae. The strategic importance of the sequence of morphs and the reproductive behaviour of each is discussed in relation to the maximization of overwintering population size.

Résumé Des différences ont été observées entre les fécondités totales et les modes de reproduction de quatre types de Sitobion avenae. Les taux de reproduction diminuent graduellement dans le temps chez les aptères et ailés virginopares et rapidement chez les gynopares et ovipares. En photophases courtes, les virginopares aptères donnent naissance à trois types: le premier composé essentiellement de gynopares, le second de mâles et le troisième de vivipares. L'importance stratégique de la succession des types et du comportement reproducteur de chacun est discutée en relation avec l'accroissement maximum de la taille de la population hivernante.

ELISA-based surveys during 1985–87 in three major cereal-growing areas of Spain confirmed the presence of barley yellow dwarf virus (BYDV). Samples of small grain cereals and grasses with and without BYDV-like symptoms were collected in the central, southwestern, and northeastern Spain. Infections were found in all cereal species sampled and in some grasses. About 37 % of the samples collected in 1985 were infacted with isolates of the PAV serotype. Isolates of the RPV serotype were less common, and were detected only in samples from the central region at El Encin, Madrid. Only a single sample, collected from El Encin in 1987, was unequivocally diagnosed as containing an isolate of the MAV serotype. Aphid vector population dynamics was monitored during fall and winter of 1984–87 in the central region. Rhopalosiphum padi L. appeared to be the most abundant species during fall and winter months, infesting grasses and volunteer wheat. Other species present were Sitobion avenae (F.), Metopolophium dirhodum (Walker) and Rhopalosiphum maidis (Fitch). Both R. padi and S. avenae seem to be anholocyclic in the central region of Spain, and are able to remain and reproduce on wheat volunteers and grasses until the beginning of spring. S, avenae populations increase quickly on wheat volunteers in April, while populations of R. padi remain low. Therefore, spread of S. avenae-transmitted BYDV types to neighbouring cereal fields seem more likely to occur than spread of other types. Other possible virus reservoirs, such as maize, also need investigation for a better understanding of BYDV epidemiology in the central and other cercal-growing areas of Spain.  相似文献

Three ancient varieties of wheat and two modern spring-sown cultivars were investigated in the field to assess their resistance to the cereal aphids Sitobion avenae and Metopolophium dirhodum; the monitoring of natural populations and detailed observations using clip-cages were both undertaken. The ancient variety Einkorn showed both antixenotic and antibiotic resistance to S. avenae, and antibiotic resistance to M. dirhodum. The resistance to S. avenae was greater than that to M. dirhodum. The value of Einkorn as a component of a resistance-breeding programme is discussed with reference to its agronomic and genetic characteristics and with reference to the pest status of cereal aphids.  相似文献

The study of aphid host selection and feeding behavior is difficult because aphids have to penetrate the plant to reach their feeding site, phloem tissue. The activity of the stylets, salivation or food intake, can not be observed externally and requires an indirect visualization technique such as the Electric Penetration Graph (EPG). The plant selection behavior of Sitobion avenae on potato varied depending on whether an ethological or EPG method was used to study it. A similar variation did not occur with Myzus persicae or Rhopalosiphum padi. The application of water-based silver conductive paint onto the thorax, as normally used for EPG, or onto the abdomen of Sitobion avenae alates resulted in increased duration and frequency of probing compared to results from ethological observations. Our results indicated that EPG manipulations might have different effects on different species of aphids and that a comparison of EPG and ethological data is required to confirm that the EPG method does not bias aphid feeding behavior.  相似文献

Three ancient varieties of wheat and two spring-sown modern varieties were screened in the laboratory to assess their resistance to the cereal aphids Sitobion avenae and Metopolophium dirhodum. Resistance was measured in terms of non-preference and antibiosis on plants at three growth stages. The ancient variety Einkorn was the most resistant in terms of both non-preference and antibiosis to both species of aphids at all growth stages examined. The ancient variety Emmer and the modern Sicco exhibited some resistant properties, whereas the ancient Spelt and modern Timmo were relatively susceptible to aphid attack.  相似文献

Insect pests can reduce wheat yield by direct feeding and transmission of plant viruses. Here we report results from laboratory and field phenotyping studies on a wide range of wheat, including landraces from the Watkins collection deriving from before the green revolution, more modern cultivars from the Gediflux collection (north‐western Europe) and modern UK Elite varieties, for resistance to the bird cherry‐oat aphid, Rhopalosiphum padi (Homoptera: Aphididae) and the English grain aphid, Sitobion avenae (Homoptera: Aphididae). A total of 338 lines were screened for R. padi and 340 lines for S. avenae. Field trials were also conducted on 122 Watkins lines to identify wheat bulb fly, Delia coarctata, preference on these landraces. Considerable variation was shown in insect performance among and within different wheat collections, with reduced susceptibility in a number of varieties, but phenotyping did not identify strong resistance to aphids or wheat bulb fly. Field trials showed within collection differences in aphid performance, with fewer aphids populating lines from the Watkins collection. This differs from development data in laboratory bioassays and suggests that there is a pre‐alighting cue deterring aphid settlement and demonstrates differences in aphid preference and performance on older plants in the field compared with seedlings in the laboratory, highlighting the need for phenotyping for aphid resistance at different plant growth stages. No association was identified between performance of the different insect species on individual varieties, potentially suggesting different nutritional requirements or resistance mechanisms.  相似文献

Heritable microbial symbionts can have important effects on many aspects of their hosts’ biology. Acquisition of a novel symbiont strain can provide fitness benefits to the host, with significant ecological and evolutionary consequences. We measured barriers to horizontal transmission by artificially transferring facultative symbionts from the grain aphid, Sitobion avenae, and five other aphid species into two clonal genotypes of S. avenae. We found the symbiont Hamiltonella defensa establishes infections more easily following a transfer from the same host species and that such infections are more stable. Infection success was also higher when the introduced symbiont strain was more closely related to the strain that was originally present in the host (but which had previously been removed). There were no differences among successfully established symbiont strains in their effect on aphid fecundity. Hamiltonella defensa did not confer protection against parasitoids in our S. avenae clones, although it often does in other aphid hosts. However, strains of the symbiont Regiella insecticola originating from two host species protected grain aphids against the pathogenic fungus Pandora neoaphidis. This study helps describe the extent to which facultative symbionts can act as a pool of adaptations that can be sampled by their eukaryote hosts.  相似文献
