Abscisic acid (ABA) regulates plant adaptive responses to various environmental stresses. 9-cis-epoxycarotenoid dioxygenase (NCED) is the key enzyme of ABA biosynthesis in higher plants. A NCED gene, SgNCED1, was overexpressed in transgenic tobacco plants which resulted in 51–77% more accumulation of ABA in leaves. Transgenic tobacco plants decreased stomatal conductance, transpiration rate, and photosynthetic rate but induced activities of superoxide dismutase (SOD), catalase (CAT), and ascorbate-peroxidase (APX). Hydrogen peroxide (H2O2) and nitric oxide (NO) in leaves were also induced in the transgenic plants. Compared to the wild-type control, the transgenic plants improved growth under 0.1 M mannitol-induced drought stress and 0.1 M NaCl-induced salinity stress. It is suggested that the ABA-induced H2O2 and NO generation upregulates the stomatal closure and antioxidant enzymes, and therefore increases drought and salinity tolerance in the transgenic plants.  相似文献

With the changing climate, plants will be facing increasingly harsh environmental conditions marked by elevated salinity in the soils and elevated concentrations of CO2 in the atmosphere. These two factors have opposite effects on water status in plants. Therefore, our objective was to determine the interaction between these two factors and to determine whether elevated [CO2] might alleviate the adverse effects of salt stress on water status in two barley cultivars, Alpha and Iranis, by studying their relative water content and their water potential and its components, transpiration rate, hydraulic conductance, and water use efficiency. Both cultivars maintained their water status under salt stress, increasing water use efficiency and conserving a high relative water content by (1) reducing water potential via passive dehydration and active osmotic adjustment and (2) decreasing transpiration through stomatal closure and reducing hydraulic conductance. Iranis showed a greater capacity to achieve osmotic adjustment than Alpha. Under the combined conditions of salt-stress and elevated [CO2], both cultivars (1) achieved osmotic adjustment to a greater extent than at ambient [CO2], likely due to elevated rates of photosynthesis, and (2) decreased passive dehydration by stomatal closure, thereby maintaining a greater turgor potential, relative water content, and water use efficiency. Therefore, we found an interaction between salt stress and elevated [CO2] with regard to water status in plants and found that elevated [CO2] is associated with improved water status of salt-stressed barley plants.  相似文献

Bean plants, Phaseolus vulgaris L. cv. Contender, were grown in the spring and summer seasons to study the relationship between xylem Na+/Cl-, transpiration rate, and salt tolerance. Eight-day-old seedlings were transplanted to 50% modified Hoagland solution with 1 mM NaCl. Four days after transfer, one of two treatments was applied: a control of 1 mM NaCl or a treatment of 25 mM NaCl every two days to reach a final treatment concentration of 75 mM NaCl. Plants were sampled on the fourth day after the final salt concentration was reached, eight days after the salinisation treatment began. Relative growth rate was 2.6-fold greater in summer than in spring. However, while no differences were found between treatments in spring, summer salt-treated plants had growth rates that were 31% lower than those of controls. In summer, CO2 assimilation, stomatal conductance, and transpiration rate of salinised plants declined with respect to controls. Leaf Na+ and trifoliolate leaf Cl- were higher in salt-treated plants in summer, although root Na+ was significantly higher in spring. Moreover, in summer salinity inhibited Ca2+ and K+ uptake and changed its distribution. Summer salt-treated plants had an average of 17-fold higher xylem Na+ during the daily cycle, while xylem Cl-, only in the afternoon, showed higher values (1.5-fold) compared to spring-grown plants. Our results suggest that the faster growth response to salt in summer-grown bean was at least partly due to an increase in xylem Na+ independent of the transpiration rate and possibly related to an increase in xylem Na+ influx or/and Na+ recirculation.  相似文献

The effect of acute ozone exposure on the stomatal conductance and leaf water content during rapid desiccation was examined in leaves of two tobacco cultivars, ozone sensitive cv. BelW3 and ozone tolerant cv. Samsun. The relative rate of stomatal closure was constant during leaf desiccation in cv. Samsun but decreased in cv. BelW3 in both ozonated and control plants. Ozone exposure increased the relative rate of stomatal closure and transpiration rate (measured on the following day) in cv. Samsun, but reduced the respective parameters in cv. BelW3. As a result, the plants of ozone-sensitive cultivar, treated with ozone, lost more water during desiccation than control plants. Citrus rootstocks as well as lemon scions differ in their ability to restrict sodium and chloride ions and in their sensitivity to saline stress. To determine the behaviour of different rootstock-scion combinations, 3 lemon cultivars on 3 different rootstocks were grown in containers in a greenhouse and irrigated with 5, 25 and 50 m M NaCl. Growth of the plants and foliar contents of sodium and chloride as well as physiological parameters including transpiration rate, gas exchange, stomatal conductance and chlorophyll content were evaluated. Shoot length of the plants on sour orange and on C. volkameriana showed a greater reduction with salinity than those on C. macrophylla . Accumulation of salt in the leaves was also scion dependent, cv. 'Eureka' having higher concentrations of sodium and chloride than the others. Assimilation rate of CO2 and stomatal conductance were greatly reduced by salinity in the leaves of Verna and Eureka on sour orange. Gas exchange in the leaves was highly correlated with chloride and sodium contents in all lemon-rootstock combinations. C. macrophylla showed a higher resistance to salinity than C. volkameriana and sour orange. Inferences on the mechanisms of action of salt on lemon trees are discussed.  相似文献

In the present study, we Investigated the effects of Increaslng sallnlty on growth, gas exchange, absclslc acld (ABA), calmodulln (CAM), and the relevance to salt tolerance In seedllngs of Populus euphratlca Ollv. and cutUngs of P. "pupularls 35-44" (P. popularls) and P. x euramerlcana cv. 1-214 (P. cv. Itallca). The relatlve growth rates of shoot helght (RGR,) for P. cv. Itallca and P. popularls were severely reduced by Increaslng salt stress, whereas the growth reductlon was relatlvely less in P. euphratica. Slmllarly, P. euphratlca malntalned hlgher net photosynthetlc rates (Pn) and unlt transplration rotes (TRN) than P. cv. Itallca and P. popularls under condltlons of hlgher sallnlty. Sallnity caused a slgnlficant increase In leaf ABA and CaM In the three genotypes after the onset of stress, but NaCl-induced ABA and CaM accumulatlon was more pronounced In P. euphratlca, suggeeUng that P. euphratlca plants are more sensitlve in sensing soil salinlty than the other two poplars. Furthermore, P. euphratica maintained relatively higher ABA and CaM concentrations under conditions of high salinity. The higher capacity to synthesize stress signals, namely ABA and CaM, In P. euphratica and the contrlbuUon of this to the salt resistance of P. euphratica are discussed.  相似文献

The effect of potassium starvation on stomatal conductance was studied in olive trees and sunflower plants, two major crops with greatly differing botanical characteristics. In both species, K(+) starvation inhibited water-stress-induced stomatal closure. In olive trees, potassium starvation favoured stomatal conductance and transpiration, as well as inhibiting shoot growth, in the three cultivars studied: 'Lechín de Granada', 'Arbequina' and 'Chetoui'. However, 'Lechín de Granada' - generally considered more drought-tolerant than 'Arbequina' and 'Chetoui' - proved less susceptible to potassium starvation. Results for olive trees also suggest genetic variability in olive cultivars in relation to potassium requirements for stem growth and the regulation of water transpiration. The results obtained suggest that inhibition of the stomatal closure mechanism produced by moderate potassium starvation is a widespread plant physiological disorder, and may be the cause of tissue dehydration in many water-stressed crops.  相似文献

Two tree medics contrasting in salt tolerance, Medicago arborea and Medicago citrina, were compared to evaluate the relative importance of abscisic acid on leaf growth and stomatal responses to salt stress. Plants were grown for 30 d in solution culture with 1, 50, 100 or 200 mM NaCl. Salinized plants of M. citrina had lower Na+ and Cl- uptake and maintained better leaf growth than M. arborea. In M. citrina, stomatal conductance was only slightly affected by salt and, in consequence, the salt treatment had no significant influence, neither on the CO2 fixation rate nor the transpiration rate in these plants. Moreover, leaf photosynthetic pigments and soluble protein in M. citrina were increased by the presence of NaCl, while a decrease of both parameters with salt was found in M. arborea. However, leaf and xylem ABA increased only in salt-treated M. citrina, while no differences were found among treatments in M. arborea. The role of ion compartmentation, gas exchange parameters and ABA concentrations in relation to salt tolerance in M. arborea and M. citrina is discussed.  相似文献

Response of two spring wheat (Triticum aestivum L.) cultivars, salt tolerant SARC-I and salt sensitive Potohar, to different concentrations of NaCl was examined under glasshouse conditions. Eighteen-day-old plants of both the lines grown in sand culture were irrigated with 0 (control), 80, 160 or 240 mM NaCl in full strength Hoagland's nutrient solution. Shoot fresh and dry masses, and leaf area per plant of SARC-I at the vegetative stage, were significantly greater than those of cv. Potohar at higher salt concentrations, however, relative growth rate (RGR) of cv. Potohar was significantly higher than that of SARC-I. SARC-I had higher net photosynthetic rate (PN), stomatal conductance (gs) and transpiration rate (E) than cv. Potohar at the vegetative stage, but the cultivars did not differ significantly in water-use efficiency (PN/E), intrinsic water use efficiency (PN/gs), and intercellular/ambient CO2 concentration ratio. At the grain development stage, SARC-I had significantly higher PN and gs in the flag leaf than cv. Potohar under salinity. SARC-I was superior to cv. Potohar with respect to number of grains per spike, number of grains per spikelet, mean grain mass, and grain yield per plant at all NaCl concentrations.  相似文献

Pepper species and cultivars, Capsicum annuum cv. Bell Boy, C. annuum cv. Kulai and C. frutescens cv. Padi, differing in drought tolerance were investigated for their water relations, stomatal responses and abscisic acid (ABA) content during water stress. C. frutescens cv. Padi exhibited a greater osmotic adjustment than C. annuum cultivars. Stomatal conductance of cv. Bell Boy was more sensitive to water stress than that of cvs. Kulai and Padi. In all pepper genotypes, stomatal closure was triggered in the absence of a large decrease in leaf water status. ABA content in xylem sap and leaf was higher in C. annum cultivars compared to C. frutescens cv. Padi. The regulative role of ABA in the rapid plant stomatal reactions in response to salinity was investigated. The influence of the short-term salinity on the overall ABA accumulation and its distribution within the mature leaf (revealed by immunohystochemical technique) and stomatal conductance of barley (Hordeum vulgare L.) were determined. Rapid bulk leaf ABA accumulation and increase in ABA immunolabeling in the mesophyl and guard cells of stomata were shown. The bulk ABA increasing in mature barley leaves coincided with stomatal closure induced by salt treatment indicating on the ABA contribution to the rapid stomatal closure.  相似文献

以采自甘肃民勤一年生的沙拐枣幼苗为试材,对不同NaCl浓度(0、50、100、200、300mmol·L~(-1))处理下沙拐枣光合生理特性进行分析,并对各生理指标与地上生物量进行灰色关联度分析,以探讨荒漠植物沙拐枣的抗盐机理,为沙拐枣的保护及其恢复荒漠生态系统稳定提供理论依据。结果显示:随着NaCl浓度的升高,沙拐枣同化枝内脯氨酸含量逐渐增大,而其可溶性糖含量逐渐减小;在低浓度NaCl(50mmol·L~(-1) NaCl)处理下,同化枝光合参数均增加,且净光合速率(Pn)、气孔导度(Gs)、蒸腾速率(Tr)均达到最大值,比对照分别显著增加了33.3%、68.0%、60.8%;与50mmol·L~(-1) NaCl相比,处理浓度超过50mmol·L~(-1) NaCl时,Pn、Gs、Tr均降低;同化枝叶绿素b含量随着NaCl浓度的增加而降低,而叶绿素a和总叶绿素含量均呈先增加后降低的趋势。灰色关联度分析发现,同化枝的Tr、Gs、Ci以及叶绿素b与地上生物量的关联度较大。研究表明,低盐浓度NaCl激活了沙拐枣的某些生理机制,有利于植株的光合作用和生长,而植物在高盐浓度胁迫时能通过调节脯氨酸和可溶性糖的含量,减少叶绿素含量、Pn和Tr等维持自身的生长。  相似文献

We report the effects of the root hemiparasite Striga hermonthica (Del.) Benth. on the growth and photosynthesis of two cultivars of sorghum: CSH-1, a susceptible variety, and Ochuti, which shows some tolerance to S. hermonthica in the field. Within 4 d of parasite attachment to the host roots, infected plants of both cultivars were significantly shorter than uninfected controls. At 55 d, infected plants of both cultivars had significantly less shoot and root biomass, and significantly smaller leaf areas than uninfected controls. The dry weight of S. hermonthica attached to host roots was insufficient at this stage to explain the decreased growth in terms of a competing sink for carbon and nitrogen. Leaf chlorophyll and nitrogen per unit area were greater in infected plants of both cultivars compared with control plants. However, whereas photosynthesis and transpiration in young leaves of infected CSH-1 plants declined with time when compared with controls, the rates in infected Ochuti plants were similar to those in uninfected controls throughout the time course of observation. In both cultivars, a strong correlation was observed between the rate of photosynthesis and stomatal conductance during photosynthetic induction, but infection resulted in a much slower induction than in controls. In CSH-1 plants, both steady-state photosynthesis and stomatal conductance were lower than in controls, whereas in leaves of Ochuti steady-state photosynthesis and stomatal conductance eventually reached the same values as in the control leaves. Results from AlCi analysis and also from determination of 13C isotope discrimination were consistent with a stomatal limitation to photosynthesis in the leaves of Striga-infected plants. The concentration of the plant growth regulator abscisic acid (ABA) was measured in the xylem sap of infected CSH-1 plants only, and was found to be twice that of uninfected plants. A possible role of ABA in determining host response to infection by S. hermonthica is discussed.  相似文献

The flacca mutant in tomato (Lycopersicon esculentum Mill. cv Rheinlands Ruhm) was employed to examine the effects of a relatively constant diurnal water stress on leaf growth and water relations. As the mutant is deficient in abscisic acid (ABA) and can be phenotypically reverted to the wild type by applications of the growth substance, inferences can be made concerning the involvement of ABA in responses to water stress. Water potential and turgor were lower in leaves of flacca than of Rheinlands Ruhm, and were increased by ABA treatment. ABA decreased transpiration rates by causing stomatal closure and also increased the hydraulic conductance of the sprayed plants. Osmotic adjustment did not occur in flacca plants despite the daily leaf water deficits. Stem elongation was inhibited by ABA, but leaf growth was promoted. It is concluded that, in some cases, ABA may promote leaf growth via its effect on leaf water balance.  相似文献

Abstract Soil waterlogging decreased leaf conductance (interpreted as stomatal closure) of vegetative pea plants (Pisuin sativum L. cv. ‘Sprite’) approximately 24 h after the start of flooding, i.e. from the beginning of the second 16 h-long photo-period. Both adaxial and abaxial surfaces of leaves of various ages and the stipules were affected. Stomatal closure was sustained for at least 3 d with no decrease in foliar hydration measured as water content per unit area, leaf water potential or leaf water saturation deficit. Instead, leaves became increasingly hydrated in association with slower transpiration. These changes in the waterlogged plants over 3 d were accompanied by up to 10-fold increases in the concentration of endogenous abscisic acid (ABA). Waterlogging also increased foliar hydration and ABA concentrations in the dark. Leaves detached from non-waterlogged plants and maintained in vials of water for up to 3 d behaved in a similar way to leaves on flooded plants, i.e. stomata closed in the absence of a water deficit but in association with increased ABA content. Applying ABA through the transpiration stream to freshly detached leaflets partially closed stomata within 15 min. The extractable concentrations of ABA associated with this closure were similar to those found in flooded plants. When an ABA-deficient ‘wilty’ mutant of pea was waterlogged, the extent of stomatal closure was less pronounced than that in ordinary non-mutant plants, and the associated increase in foliar ABA was correspondingly smaller. Similarly, waterlogging closed stomata of tomato plants within 24 h, but no such closure was seen in ‘flacca’, a corresponding ABA-deficient mutant. The results provide an example of stomatal closure brought about by stress in the root environment in the absence of water deficiency. The correlative factor operating between the roots and shoots appeared to be an inhibition of ABA transport out of the shoots of flooded plants, causing the hormone to accumulate in the leaves.  相似文献

* Root respiration, stomatal conductance, leaf transpiration and photosynthetic rates were measured in phytotron and field-grown plants following the application of 5 or 10 nM lumichrome, 10 nM ABA (abscisic acid) and 10 ml of 0.2 OD600 infective rhizobial cells. * Providing soybean and cowpea roots with their respective homologous rhizobia and/or purified lumichrome increased the concentration of this molecule in xylem sap and leaf extracts. Relative to control, rhizobial inoculation and lumichrome application significantly increased root respiration in maize, decreased it in lupin, but had no effect on the other test species. * Applying either lumichrome (10 nM), infective rhizobial cells or ABA to roots of plants for 44 h in growth chambers altered leaf stomatal conductance and transpiration in cowpea, lupin, soybean, Bambara groundnut and maize, but not in pea or sorghum. Where stomatal conductance was increased by lumichrome application or rhizobial inoculation, it resulted in increased leaf transpiration relative to control plants. Treating roots of field plants of cowpea with this metabolite up to 63 d after planting showed decreased stomatal conductance, which affected CO2 intake and reduction by Rubisco. * The effect of rhizobial inoculation closely mirrored that of lumichrome application to roots, indicating that rhizobial effects on these physiological activities were most likely due to lumichrome released into the rhizosphere.  相似文献
