Sterol compositions of the cold water ophiuroids Ophioplocus januarii and Ophionotus victoriae and of the tropical ophiuroids Ophiocoma echinata and Ophiocoma wendtii are reported. The four sterol mixtures contain Δ5 mono- and di-unsaturated common 3β-hydroxy-sterols. Ophioplocus januarii and O. victoriae contain 24-methylcholesta-5,24(28)-dien-3β-ol and 24ζ-ethylcholesta-5,24(28)-dien-3β-ol in higher abundance than in O. echinata. These sterols were not found in O. wendtii. An interesting finding is the presence of Δ5,24(28)-24-n-propylidenecholesterol in 7.6% in Ophionotus victoriae.  相似文献

Using living and herbarium material, a survey of polyhydroxyalkaloids in species of 52 genera of Araceae revealed the presence of 2,5-dihydroxymethyl-3,4-dihydroxypyrrolidine (DMDP) and -homonojirimycin (HNJ) in leaves of Nephthytis Schott, Anchomanes Schott, Pseudohydrosme Engl. (tribe Nephthytideae), Aglaonema Schott and Aglaodorum Engl. (tribe Aglaonemateae). Levels were high in living plants, ranging from 0.1 to 1% dry weight DMDP and 0.04 to 0.6% HNJ. Isomers of HNJ, such as -3,4-di-epi-homonojirimycin, were also present in the five genera. Seven of the eight Nephthytis species examined also contained deoxymannojirimycin at levels, in fresh material, of 0.1–0.2% dry weight. Lower levels of DMDP (mean 0.009%) and HNJ (mean 0.002%) were detected in species of Amorphophallus Blume & Decne and Pseudodracontium N.E. Br. (tribe Thomsonieae). The similarity in polyhydroxyalkaloid chemistry between Nephthytideae and Aglaonemateae concurs with recently published chloroplast restriction site data in suggesting a relationship between these tribes.  相似文献

Relationships between two plant communities in the south-western Alps, high-altitude manured meadows (Triseto-Polygonion) and lower tall grass prairies (Adenostylion) have been examined by means of the flavonoid analysis of specimens belonging to six species common to those two associations. The species, collected at eight stations of the Ligurian Alps and the Ubaye valley, are: Trollius europaeus (Ranunculaceae), Chaerophyllum aureum (Umbellifereae), Silene cucubalus (Caryophyllaceae), Polygonum bistorta) (Polygonaceae), Alchemilla vulgaris (Rosaceae) and Centaurea montana (Compositae). In every species, factorial analysis of correspondence was applied to the distribution of flavonoid glycosides. This led to the partition of the populations into two groups according to the two sectors (Ligurian Alphs Ubaye valley) of the collection area. Secondly, populations of two species, Alchemilla vulgaris and Centaurea montana, exhibited further correlation of their flavonoid pattern with their phytosociological origin.  相似文献

It is of interest to understand what kinds of physiological and biochemical changes occur in insects if the homeostasis of trehalose in the hemolymph is disrupted by the infection with a recombinant baculovirus containing a secretory-trehalase gene. For this purpose, two recombinant non-occluded Autographa california multicapsid nucleopolyhedroviruses (AcMNPVs), vTREVL and vERTVL, containing a trehalase cDNA of the mealworm beetle, Tenebrio molitor, were constructed. The trehalase cDNA was inserted in the sense orientation downstream of the polyhedrin promoter for vTREVL, and in the anitsense orientation for vERTVL. The active trehelase of T. molitor was found outside of cells when SF-9 cells or larvae of the cabbage armyworm, Mamestra brassicae, were infected with vTREVL. In the hemolymph of vTREVL-infected larvae, expression of the active trehelase was followed by disappearance of trehalose and appearance of glucose. However, the mortality time of virus-infected 5th instar larvae increased in the following order: AcMNPV C6 (wild-type virus) ≤ vERTVL < vTREVL. The symptoms (the browning and liquefying of the host body) of NPV infection were moderated considerably in vTREVL-infected larvae.  相似文献

The major pentasaccharides Fuc(1-2)[GalNAc(1-3)]Gal(1-4)[Fuc(1-3)]Glc and Fuc(1-2) [Gal(1-3)]Gal(1-4)[Fuc(1-3)]Glc, which are normally present in the urine of bloodgroup A Leb and B Leb healthy subjects, were each found to be contaminated by a minor component when analysed by1H-NMR. The chemical structure of lipid A, from the marine -proteobacterium Pseudoalteromonas haloplanktis 14393, a main product of lipopolysaccharide hydrolysis (1% AcOH), was determined using chemical methods and NMR spectroscopy. The lipid A was shown to be -1,6-glucosaminobiose 1,4-diphosphate acylated with two (R)-3-hydroxyalkanoic acid residues at C3 and C3 and amidated with one (R)-3-hydroxydodecanoyl and one (R)-3-dodecanoyloxydodecanoyl residue at N2 and N2, respectively.  相似文献

A comparative localization of Na+,K+-ATPase and ouabain-sensitive H+,K+-ATPase in rat skin was performed using in situ RNA hybridization and immunohistochemistry. Na+,K+-ATPase was predominantly detected in the basal layer of the epithelium, whereas the ouabain-sensitive H+,K+-ATPase, in the granular and prickle cell layers. The genes of these ATPases are thus expressed in epithelial cells at different stages of their development. The hypothesis was advanced that the ouabain-sensitive H+,K+-ATPase is involved in maintaining the skin pH value. The probes specific to the mRNAs of the full-size -subunit of the ouabain-sensitive H+,K+-ATPase and its truncated form were used to establish a similar distribution of both mRNA variants in skin.  相似文献

Smoking or ingestion of cannabis causes cognitive, perceptual and behavioural changes, which are responsible for impaired performance in driving motor vehicles. In this paper a novel liquid chromatographic assay for the selective quantification of Δ9-tetrahydrocannabinol, the major indicator of a present cannabis intoxication in saliva, is described. The method involves a column-switching procedure and requires an extremely simple pre-treatment of the sample. Deproteinized saliva was directly injected into the chromatographic system. The clean-up and enrichment procedure was performed in an immunoaffinity column, followed by the transfer of the antigens to an octylsilica analytical column. The immunoaffinity sorbent was obtained by covalent immobilization of specific antibodies on epoxy-activated silica. The mobile phase consisted of methanol-aqueous 0.15 mol/1 NaCl solution (elution programmed) and the analyte was detected by measuring the UV absorption at 220 nm. Using an injection volume of 4.5 ml (dilution 3:2, v/v) the limit of quantification was 20 ng/ml, at a signal-to-noise ratio of 5. Recoveries were estimated to be in the range of 70%. A method for estimating immobilized enzyme reaction progress curves, using simultaneous non-linear regression analysis of 2–3 substrate concentrations with time, is presented. These facile procedures involve using nested Gauss–Newton curve fitting algorithms on a Microsoft EXCEL spreadsheet. We refer to our technique as "nested" because the analysis consists of two or three mutually parameter-dependent sets of computations associated with bi- or termolecular enzyme-catalyzed reactions, respectively. We have applied the method to immobilized glucose oxidase-catalyzed reactions ([ -glucose] and [O2] with time) and found that the kinetic parameters from initial velocity data were similar to those determined by the nested curve fitting method discussed herein.  相似文献

Functionally active preparations of Na+,K+-ATPase isozymes from calf brain that contain catalytic subunits of three types (1, 2, and 3) were obtained using two approaches: a selective removal of contaminating proteins by the Jorgensen method and a selective solubilization of the enzyme with subsequent reconstitution of their membrane structure by the Esmann method. The ouabain inhibition constants were determined for the isozymes. The real isozyme composition of the Na+ pump from the grey matter containing glial cells and the brain stem containing neurons was determined. The plasma membranes of glial cells were shown to contain mainly Na+,K+-ATPase of the 11 type and minor amounts of isozymes of the 22(1) and the 31(2) type. The axolemma contains 21 and 31 isozymes. A carbohydrate analysis indicated that 11 enzyme preparations from the brain grey matter substantially differ from the renal enzymes of the same composition in the glycosylation of the 1 isoform. An enhanced sensitivity of the 3 catalytic subunit of Na+,K+-ATPase from neurons to endogenous proteolysis was found. A point of specific proteolysis in the amino acid sequence PNDNR492 Y493 was localized (residue numbering is that of the human 3 subunit). This sequence corresponds to one of the regions of the greatest variability in 1-, 2-, 3-, and 4-subunits, but at the same time, it is characteristic of the 3 isoforms of various species. The presence of the 3 isoform of tubulin (cytoskeletal protein) was found for the first time in the high-molecular-mass Na+,K+-ATPase 31 isozyme complex isolated from the axolemma of brain stem neurons, and its binding to the 3 catalytic subunit was shown.  相似文献

The structure of a new nonasaccharide isolated from human milk has been investigated. Leaf resins in the leguminous genera Hymenaea and Copaifera may play a role in restricting infection by the associated leaf fungus, Pestalotia. We tested this hypothesis by assessing growth of the geographically widespread Pestalotia subcuticularis presented with different compositions of resins. Leaf resins of Hymenaea and Copaifera are composed of the same suite of sesquiterpene hydrocarbons, but these vary quantitatively to form discrete compositional patterns among individuals and populations. Leaf resins may also contain sesquiterpene oxides; caryophyllene oxide, which may have been formed from the caryophyllene precursor common in these resins, inhibited fungal growth in vitro.  相似文献

In Alzheimer's disease, abnormal processing of the amyloid precursor protein (APP) is thought to play an important role in amyloid deposition. We investigated the effect of heparin, a highly sulfated glycosaminoglycan related to heparan sulfate, on the secretion of the ß-secretase cleavage product of APP (sAPPß) in a human neuroblastoma cell line. Heparin induced an increase in the secretion of total APP, and an even greater relative increase in the secretion of sAPPß. The effect on sAPPß was specific to heparin. These data support the hypothesis that highly sulfated heparan sulfate proteoglycans may promote amyloidogenic pathways of APP metabolism.  相似文献

A highly sensitive fluorimetric assay using 3-O-methylfluorescein phosphate as substrate was used in the determination of K+-dependent phosphatase activity in preparations of rat skeletal muscle. The gastrocnemius muscle was chosen because of mixed fibre composition. Crude, detergent treated homogenate was used so as to avoid loss of activity during purification. K+-dependent phosphatase activities in the range 0.19–0.37 μmol · (g wet weight)−1 · min−1 were obtained, the value decreasing with age and K+-deficiency. Complete inhibition of the K+-dependent phosphatase was obtained with 10−3 M ouabain. Using a KSCN-extracted muscle enzyme the intimate relation between K+-dependent phosphatase activity and (Na+ + K+)-activated ATP hydrolysis could be demonstrated. A molecular activity of 620 min−1 was estimated from simultaneous determination of K+-dependent phosphatase activity and [3H]ouabain binding capacity using the partially purified enzyme preparation. The corresponding enzyme concentration in the crude homogenates was calculated and corresponded well with the number of [3H]ouabain binding sites measured in intact muscles or biopsies hereof.  相似文献

The cerebrosides were first isolated by Thudicum in 1874 and the structures were established by Carteret al. in 1950 (for review, see [2]). In 1961 Shapiro and Flowers [3] reported the first total synthesis of a cerebroside1 (Fig. 1) which was identified with the natural sample, only through comparison of their i.r. data. In order to confirm the absolute configuration at C-2 of natural cerebroside1, we describe here an unambiguous synthesis of two stereoisomeric cerebrosides1 and2, and found that the1H-NMR spectra of the synthetic1 (Fig. 2) was completely identical with that of the natural cerebroside reported recently by Dabrowskiet al. The effect of pH and transmembrane pH on the efficiency of the proton pump of the mitochondrialbc 1 complex bothin situ and in the reconstituted state was studied. In both cases the H+/e ratio for vectorial proton translocation by thebc 1 complex respiring at the steady state, under conditions in which the transmembrane pH difference (pH) represents the only component of the proton motive force (p), was significantly lower than that measured under level flow conditions. The latter amounts, at neutral pH, to 1 (2 including the scalar H+ release). In the reconstituted system steady-state pH was modulated by changing the intravesicular buffer as well as the intra/extra-liposomal pH. Under these conditions the H+/e ratio varied inversely with the pH. The data presented show that pH exerts a critical control on the proton pump of thebc 1 complex. Increasing the external pH above neutrality caused a decrease of the level flowH +/e ratio. This effect is explained in terms of proton/electron linkage inb cytochromes.  相似文献

The mannuronan C-5-epimerase AlgE2 is one of a family of Ca2+-dependent epimerases secreted by Azotobacter vinelandii. These enzymes catalyze the conversion of β- -mannuronic acid residues (M) to - -guluronic acid residues (G) in alginate. AlgE2 has been produced by fermentation with a recombinant strain of Escherichia coli, isolated and partially purified. Epimerization with AlgE2 increased the content of G-residues in different alginates from starting values of 0–45% up to approximately 70%. The new G-residues were mainly present in short blocks. Although G-residues may be introduced next to pre-existing G-residues, AlgE2 was not able to epimerize strictly alternating MG-structures. The epimerization with AlgE2 was greatly affected by the concentration of Ca2+. The type of alginate used as substrate affected the reaction rate and the reaction pattern especially at low Ca2+ concentration. AlgE2 appears to act by a preferred attack mechanism where the enzyme associates with different sequences in the alginate depending on the concentration of Ca2+. During epimerization, AlgE2 occasionally causes cleavage of the alginate chain. The observed frequency corresponds to 1–3 breaks per 1,000 M-units epimerized.  相似文献

Summary In male mice homozygous for both p s and hpy, two recessive, pleiotrophic, mutations, gametogenesis is normal through meiosis but no functional spermatozoa are produced. Spermiogenesis is abnormal from the Golgi phase on. The types of abnormalities seen during the early and mid-stages of Spermiogenesis are characteristic of those associated with the presence of the p s mutation whereas those associated with the hpy mutation appear during the later stages of spermatid development. While centriolar ultrastructure was normal, axonemal structures were only rarely encountered and no late spermatids with recognizable flagella were seen. Some late spermatids showed head abnormalities of the type characteristic of the ps mutation while others were recognizable as being of the hpy type. A released gamete usually consisted of a distorted nucleus and associated acrosome enclosed in a tightly fitting plasma membrane. No spermatids exhibiting a novel phenotype were encountered. The findings support the view that, despite their simultaneous presence in the double homozygote, each mutation acts autonomously. These studies also allow a similar inference to be made with respect to the extent of the interrelationship of the other major sub-processes of Spermiogenesis.The author wishes to express his thanks to Mr. Clifford S.
