Aphelenchoides saprophilus nematodes fed on sclerotia, mycelium, and alginate-formulated pellets of Sclerotinia sclerotiorum, mycelium of Trichoderma harzianum, and mixed fungal cultures. As many as 500 nematodes were found inside individual sclerotia. Results suggest potential impacts of fungivory on S. sclerotiorum and its ecological interactions with plant hosts and biocontrol fungi.  相似文献

The purpose of this study was to investigate the effects of isolates T22, T9 and T6 of Trichoderma harzianum on isolate F42 of Fusarium oxysporum f. sp. radicis-cucumerinum. The effect of T. harzianum isolates on controlling disease was examined under greenhouse conditions. Results showed that these three isolates, respectively, had the high effect on inhibition of pathogen growth. In examining the severity of disease in the greenhouse, it was found that isolate T22 had the greatest effect on controlling the pathogen. The results of volatile compounds showed that these isolates, respectively, were effective in reducing mycelial growth of isolate F42. The highest peroxidase activity was observed on the fourth day in isolate T6 and the highest phenylalanine ammonia lyase enzyme activity was observed on the fifth day in isolate T22. Based on the results, isolate of T22 showed the greatest effect on the induction of resistance against F42 and may be a successful agent for biological control of root and stem rot of cucumber.  相似文献

Studies were conducted to determine the effects of soil moisture (9, 16 or 24% w/w) and temperature (5, 15, 20 or 25°C) on the control of sclerotia of Sclerotinia sclerotiorum by five fungal agents in sterile and natural field soil. All five biocontrol agents were effective in reducing the survival of sclerotia of S. sclerotiorum in sterile soil under dry (9% moisture) or wet (24% moisture) conditions at 20°C, but only Coniothyrium minitans was effective in natural soil. Coniothyrium minitans was the most effective in reducing sclerotial viability at the temperature range of 15–25°C. Trichoderma virens was effective against sclerotia of S. sclerotiorum to a lesser extent than C. minitans , and in non-autoclaved soil, it performed best at 25°C. Although Epicoccum purpurascens , Talaromyces flavus and Trichothecium roseum were effective against sclerotia of S. sclerotiorum in some instances, they were less effective than C. minitans and T. virens . Sclerotia of S. sclerotiorum conditioned for myceliogenic germination were more vulnerable to attack by the biocontrol agents than dormant sclerotia. The implications are discussed with respect to enhancement of biological control of crop diseases caused by S. sclerotiorum in different geographic regions.  相似文献

A novel 36-kDa endochitinase named chit36 has been isolated and characterized from Trichoderma harzianum Rifai TM. Partial amino acid sequences from the purified protein were used to clone the fungal cDNA, based on polymerase chain reaction with degenerate primers. The complete open reading frame encodes a 344-amino acid protein which shows 84% similarity to a putative chitinase from Streptomyces coelicolor. Chit36 was overexpressed under the pki1 constitutive promoter from Trichoderma reesei via biolistic transformation of T. harzianum TM. Stable transformants showed expression and endochitinase activity of chit36 in glucose-rich medium. Culture filtrates containing secreted CHIT36 as the sole chitinolytic enzyme completely inhibited the germination of Botrytis cinerea conidia. Growth of Fusarium oxysporum f. sp. melonis and Sclerotium rolfsii were significantly inhibited on agar plates on which the Trichoderma transformants had previously been grown.  相似文献

Sclerotinia sclerotiorum causes serious yield losses to many crops worldwide. Aspergillus sp. Asp-4, previously shown to inhibit germination of sclerotia of S. sclerotiorum in vitro and in the field, was evaluated in field trials for suppression of this pathogen on oilseed rape. Spray application of Asp-4 to the soil prior to sowing rice in a rice–oilseed rape rotation resulted in a significant reduction in incidence of Sclerotinia stem rot on oilseed rape compared with the non-treated control in two field trials. This application of Asp-4 also resulted in a significant reduction in germination of sclerotia relative to the non-treated control in these field trials, suggesting that this reduction in sclerotial germination led to disease control. Microscopic examination demonstrated that Asp-4 could effectively colonise external and internal portions of sclerotia of S. sclerotiorum in vitro. Incubation of Asp-4 with sterile sclerotial material induced production of β-glucanase and chitinase activities by this isolate; β-glucanase and chitinase being potentially capable of degrading the glucan and chitin polymeric components of sclerotia. Incubation of Asp-4 with sterile sclerotial material also resulted in a significant reduction in dry weight of this sclerotial material relative to the non-treated control in 96?h in vitro experiments. Experiments reported here indicate that Aspergillus sp. Asp-4 has promise as a biological control agent for S. sclerotiorum on oilseed rape. Experiments reported here suggest that disease control results from inhibition of germination of sclerotial resting structures due to mycoparasitic colonisation by Asp-4.  相似文献

Three Trichoderma harzianum isolates viz., Th-Sks, Th-Ke and Th-Ar collected from respective states of India viz., Rajasthan, Kerala and Andhra Pradesh were evaluated for the management of six fungal diseases namely damping off, Fusarium wilt, Rhizoctonia wilt, early leaf spot, late blight and Septoria leaf spot in tomato. During in vitro analysis, T. harzianum isolates inhibited the pathogens’ growth. Isolate Th-Sks was the most virulent antagonist against all the test pathogens and exhibited maximum of 79.47% growth inhibition of Phytophthora infestans. Isolate Th-Sks proved most effective at suppression efficacy in the range of 95–100% and 91–100% against all diseases under glasshouse and in the field conditions, respectively. Tomato seeds treatment with isolate Th-Sks also promoted plant height (78.23 cm) and fruits yield (290 g/plant) during field trial and data were found to be not-significantly different from other isolates. Thus, it is concluded that isolate Th-Sks can be utilised as a biocontrol agent for management of fungal diseases in tomato.  相似文献

The effect of six isolates of Trichoderma harzianum and one isolate of T. koningii on the incidence and severity of tan spot (Pyrenophora tritici-repentis) and leaf blotch of wheat (Mycosphaerella graminicola) was evaluated under field conditions. Significant differences between wheat cultivars, inoculum types and growth stages were found. Three of the isolates tested (T2 for M. graminicola, T7 for P. tritici-repentis and T5 for both of them) showed the best performance in controlling leaf blotch and tan spot when coated onto seed or sprayed onto wheat leaves at different growth stages, with significant severity reduction up to 56%. At tillering, six of the isolates reduced the severity of P. tritici-repentis and M. graminicola compared to the control by up to 39% and 12-53%, respectively. In some experiments, the biocontrol preparation (T2 and T5) gave a level of disease control similar to that obtained with Tebuconazole (70 and 48%, respectively). The effect of Trichoderma against P. tritici-repentis was also observed at the heading stage, when six of the treatments reduced disease severity by 16-35%. This is the first report on the efficacy of Trichoderma spp. against wheat necrotrophic pathogens under field conditions in Argentina.  相似文献

A method is described for the culture of larvae of Scolytus multistriatus and Scolytus scolytus on an artificial medium following exposure to cultures of microorganisms. In control cultures, a natural mortality rate of 21.2% was found for S. multistriatus and 17.6% for S. scolytus. The effects of Trichoderma harzianum, T. polysporum and Scytalidium lignicola on fifth instar larvae of S. scolytus and S. multistriatus reared on the artificial medium were studied. The fungi were larvicidal and larval mortality was increased to more than 80% by inoculation of the larvae with the fungi. Another fungus, Phomopsis oblonga, had little effect on larvae of S. scolytus. The results are discussed in relation to mechanism of pathogenicity of the fungi and their potential use in the control of Dutch elm disease. It is proposed that with modifications, the method is applicable to other bark beetle pests.  相似文献

The mycolytic bacterial strain Bacillus sp. 739 produces extracellular enzymes which degrade in vitro the cell walls of a number of phytopathogenic and saprophytic fungi. When Bacillus sp. 739 was cultivated with Bipolaris sorokiniana, a cereal root-rot pathogen, the fungus degradation process correlated with the levels of the β-1,3-glucanase and protease activity. The comparative characteristic of Bacillus sp. 739 enzymatic preparations showed that efficient hydrolysis of the fungus cell walls was the result of the action of the complex of enzymes produced by the strain when grown on chitin-containing media. Among the enzymes of this complex, chitinases and β-1,3-glucanases hydrolyzed most actively the disintegrated cell walls of B. sorokiniana. However, only β-1,3-glucanases were able to degrade the cell walls of native fungal mycelium in the absence of other hydrolases, which is indicative of their key role in the mycolytic activity of Bacillus sp. 739.  相似文献

The objective of this study was to determine the effectiveness of a 42-kDa endochitinase coded by the ThEn42 gene from Trichoderma harzianum as a potential source of transgenic resistance to Rhizoctonia root rot of barley caused by Rhizoctonia solani AG8 and/or R. oryzae. The gene cThEn42 was codon optimized (GC content increased from 53.3 to 65.1%) and then synthesized to produce the modified cThEn42GC in Pichia pastoris for in vitro tests. Two expression vectors were constructed: one with the fungal signal peptide and the fungal activation peptide [FSP-FAP-cThEn(GC)] and the other with barley chitinase 26 signal peptide followed by the fungal signal and activation peptides [SP(HVChi26)-FSP-FAP-cThEn(GC)]. N-terminal sequencing showed that, of two proteins secreted into liquid medium, FSP was cleaved off faithfully in one protein and both FSP and FAP were cleaved from the other protein. Purified endochitinase provided strong in vitro inhibition of both R. solani AG8 and R. oryzae. The enzyme had an intermediate inhibitory activity against Gaeumannomyces graminis var. tritici, and no inhibitory activity against Fusarium graminearum, F. pseudograminearum, and F. culmorum.  相似文献

Genes from the biocontrol fungus Trichoderma atroviride encoding the antifungal proteins endochitinase or exochitinase (N-acetyl--D-hexosaminidase) were inserted into Marshall McIntosh apple singly and in combination. The genes were driven by a modified CaMV35S promoter. The resulting plants were screened for resistance to Venturia inaequalis, the causal agent of apple scab, and for effects of enzyme expression on growth. Disease resistance was correlated with the level of expression of either enzyme when expressed alone but exochitinase was less effective than endochitinase. The level of expression of endochitinase was negatively correlated with plant growth while exochitinase had no consistent effect on this character. Plants expressing both enzymes simultaneously were more resistant than plants expressing either single enzyme at the same level; analyses indicated that the two enzymes acted synergistically to reduce disease. Selected lines, especially one expressing low levels of endochitinase activity and moderate levels of exochitinase activity, were highly resistant in growth chamber trials and had negligible reduction in vigor relative to control plants. We believe that this is the first report of resistance in plants induced by expression of an N-acetylhexosaminidase and is the first report of in planta synergy between an exochitinase and an endochitinase.  相似文献

From 1985 to 1989 five large scale surveys were made to document the spread of the cassava mealybug (CM) Phenacoccus manihoti Matile‐Ferrero (Hom., Pseudococcidae) and the releases and successful establishment of its exotic parasitoid Epidinocarsis lopezi (De Santis) (Hym., Encyrtidae) through most cassava‐growing areas of Malawi. In a multiple regression analysis involving 29 meteorological, agronomic and plant variables from 476 fields, the duration of E. lopezi's presence was the major factor influencing CM population densities. In the first year the CM was recorded in a particular place, 25% of all tips had more than 100 CM. Wherever E. lopezi had been present for two years or more, CM populations were reduced on average seven times and tips infested with more than 100 CM became rare (1%). In parallel, damage on cassava tips stabilized at a low level. Similarly, along Lake Malawi, 84.9% of all tips had more than 10 CM in 1986; this value stabilized at 3.3%‐4.0% in 1988–1989. Indigenous coccinellids were often abundant in the first year of the CM infestation, but their populations collapsed later, while the frequency of fields with E. lopezi increased. In 1987, farmers abandoned 28.4% of all fields where E. lopezi was just introduced as compared with 2.7% where the parasitoid had been present for two years or more. Overall, satisfactory control was achieved by E. lopezi in all but a few fields concentrated on extremely poor soils characterized by sand dune vegetation. The socioeconomic implications of this ongoing and apparently successful biological control program are discussed.  相似文献

1 Defoliation by larvae of the pine processionary moth, Thaumetopoea pityocampa, is negligible in stands of Pinus pinaster colonized by the Argentine ant Linepithema humile which preys fiercely on the young moth larvae. In contrast, such damage is widespread where pine plantations are colonized by native ants, predominantly Tapinoma nigerrimum and Lasius niger, which seemingly disregard the larvae. 2 Where L. humile- and native ant-occupied sectors adjoin, there is a 20–50 m overlap in the transition area between L. humile- and native ant-occupied pines. This was most evident in a > 500 ha plantation where there was severe or very severe T. pityocampa attack in native ant sectors contrasting with none in adjoining L. humile sectors. 3 Predation by L. humile is no doubt enhanced by its existence as super-colonies over very large areas, by its foraging activity and recruitment on trees throughout the time when T. pityocampa and other prey are present, and by honeydew- producing Homoptera which help retain foraging L. humile workers in pine tree crowns. 4 The role of L. humile could be enhanced by cultivations that disturb the soil and restrain ground vegetation.  相似文献

The Madagascan endemic, Bryophyllum delagoense (Crassulaceae), is a major weed in Queensland, Australia. Despite having first been recorded in Australia in the 1940s, it is far more invasive there than on the African mainland where it was introduced more than 170 years ago. This may be due to a number of factors, one of which could be the occurrence of new natural enemy associations in southern Africa. Among the insects of crassulaceous plants that have extended their host ranges, a stem-boring weevil, Alcidodes sedi, was studied to elucidate its status as a natural enemy of B. delagoense in southern Africa and as a candidate biological control agent for introduction to Australia. Laboratory studies indicated that damage inflicted by adult and larval feeding caused significant reductions in stem length and number of leaves. Preliminary host-range trials revealed that A. sedi can complete its development on other species in the Crassulaceae, including most of the introduced Bryophyllum species and some Kalanchoe species native to South Africa. Despite the oligophagous nature of A. sedi and the fact that it can complete its development on a number of ornamental species in the Crassulaceae, it should be considered a potential biological control agent in Australia. All of the native Crassulaceae in Australia are in the genus Crassula, most of which are very small and therefore unlikely to support the development of a large weevil like A. sedi. However, additional host-range trials will have to be undertaken in Australia to determine whether the weevil can be considered safe for release.  相似文献

St. Helena is a small South Atlantic island with a highly degraded, but internationally significant, terrestrial flora. The 2500 remaining gumwoods, Commidendrum robustum, are an important part of this remnant flora. In 1991, a South American scale insect, Orthezia insignis, was identified attacking the gumwood trees. By 1993, severe infestations had killed over 100 trees. If the number of dead trees had continued to increase exponentially, all 2500 trees would have died by 1995. This outcome was likely given abundant alternative host plants, and lack of natural enemies of O. insignis. Host range tests showed that the other members of the endemic genus Commidendrum were also at risk. The coccinellid predator, Hyperaspis pantherina, was released on St. Helena in 1993. H. pantherina numbers increased from 1994, coinciding with a 30× decrease in scale numbers. Scale outbreaks have not been reported on St Helena since 1995, and culturing of H. pantherina was discontinued because insufficient O. insignis prey could be found in the field. Extensive blackening from sooty molds on the surviving trees in 1995 suggested that the predator was effective just in time to prevent death of most trees. H. pantherina appears to have saved the field population of a rare endemic plant from extinction.  相似文献
