用透射电镜观察了箭舌豌豆根瘤侵染细胞。结果表明,有一些小泡出现在细胞质膜和侵入线附近,它们不断向中央液泡运动,并在运动中相互靠近形成小泡团。当其到达中央液泡时,附近的液泡膜产生内吞,形成突起。最后,这些突起脱离液泡膜,在中央液泡中形成一种由管状结构和小泡组成、表面具有一层被膜的特殊内含物。本文还讨论了此种内含物的起源及其与根瘤抗旱和细菌周膜扩增的关系。  相似文献

Abstract: In order to study the effect of mechanical stimulation on the membrane potential (Em) of roots of Lepidium sativum L., the roots were subjected to pulses of perfusion medium expelled from a micropipette positioned 1 mm from the root surface at the level of the elongation zone. On the opposite side of the root, a microelectrode was inserted into rhizodermal or cortical cells of the elongation zone to record the membrane potential during such a pulse. A medium pulse elicited a fast transient hyperpolarisation of up to ?38 mV, reaching a peak approx. 1 min after the start of a pulse. Both degassing the medium and increasing the buffer strength had no effect on the magnitude of the pulse-induced hyperpolarisation. Covering the roots with Parafilm, and thereby preventing an exchange of medium in the vicinity of the root during a pulse, while still providing a mechanical stimulus, also did not abolish the response. We conclude that Lepidium roots are capable of perceiving extremely weak mechanical stimuli (less than 1 mN) which induce rapid transient hyperpolarisations of the plasma membrane.  相似文献

用透射电镜研究了红豆草(Onobrychis viciifolia)根瘤侵染细胞中液泡内含物的超微结构特征。结果表明,早期发育侵染细胞的液泡中只含有少量的纤维状物质。随着细胞的发育,液泡不断变大,液泡中的纤维状物质和膜状物质越来越多。在中央液泡形成后,液泡中的纤维状物质逐渐减少,类细胞质、泡状和膜状物质明显增多,它们常由一层来自液泡膜的膜包围,其形状一般近似圆形或椭圆形。液泡内含物的大量出现可能与红豆草及其根瘤具有高度的抗旱件有关。  相似文献

Using a highly sensitive vibrating electrode, the pattern of naturally occurring electric currents around 1-day-old primary roots of Lepidium sativum L. growing vertically downward and the current pattern following gravistimulation of the root has been examined. A more or less symmetrical pattern of current was found around vertically oriented, downward growing roots. Current entered the root at the root cap, the meristem, and the beginning of the elongation zone and left the root along most of the elongation zone and in the root hair zone. After the root was tilted to a horizontal position, we observed current flowing acropetally at the upper side of the root cap and basipetally at the lower side within about 30 seconds in most cases. After a delay of several minutes, acropetally oriented current was also found flowing along the upper side of the meristematic zone. The apparent density of the acropetal current in the root cap region increased and then decreased with time. Gravitropic curvature was first visible approximately 10 minutes after tilting of the root to the horizontal position. Since the change in the pattern of current in the root cap region precedes bending of the root and is different for the upper and lower side, a close connection is suggested between the current and the transduction of information from the root cap to the elongation zone following graviperception in the cap.  相似文献

The process of gonad development in the frog Rana nigromaculata was observed using the electron microscope. The gonadal medulla was formed by the proliferation and displacement of the epithelial cells within the primordial gonad, and a distinct continuity was observed between the cortical and medullary cells. Sex differentiation of the gonad occurred directly from the sexually indifferent primordial gonads. In the rudimentary testes, the continuity between the cortical and medullary regions increased closer, and the intermingling of cortical and medullary cells was evident. The inner region of the cortex developed into a cord-like structure and subsequently differentiated into rudimentary seminiferous tubules. The medulla differentiated into the testicular rete and efferent duct. In the rudimentary ovaries, the cortex and medulla were separated and the ovarian cavity was formed in the medullary region. In the cortex, the cortical cells surrounding oocytes which had reached the diplotene stage, differentiated into follicular cells. The intrusion of mesenchymal or blastemal cells derived from extragonadal regions into the cortex or medulla was never observed. These findings do not support Witschi's cortico-medullary antagonistic theory of sex differentiation.  相似文献

Artesunate (ART) is an anti-malaria drug that has been shown to exhibit anti-tumor activity, and functional lysosomes are reported to be required for ART-induced cancer cell death, whereas the underlying molecular mechanisms remain largely elusive. In this study, we aimed to elucidate the molecular mechanisms underlying ART-induced cell death. We first confirmed that ART induces apoptotic cell death in cancer cells. Interestingly, we found that ART preferably accumulates in the lysosomes and is able to activate lysosomal function via promotion of lysosomal V-ATPase assembly. Furthermore, we found that lysosomes function upstream of mitochondria in reactive oxygen species production. Importantly, we provided evidence showing that lysosomal iron is required for the lysosomal activation and mitochondrial reactive oxygen species production induced by ART. Finally, we showed that ART-induced cell death is mediated by the release of iron in the lysosomes, which results from the lysosomal degradation of ferritin, an iron storage protein. Meanwhile, overexpression of ferritin heavy chain significantly protected cells from ART-induced cell death. In addition, knockdown of nuclear receptor coactivator 4, the adaptor protein for ferritin degradation, was able to block ART-mediated ferritin degradation and rescue the ART-induced cell death. In summary, our study demonstrates that ART treatment activates lysosomal function and then promotes ferritin degradation, subsequently leading to the increase of lysosomal iron that is utilized by ART for its cytotoxic effect on cancer cells. Thus, our data reveal a new mechanistic action underlying ART-induced cell death in cancer cells.  相似文献

用透射电镜观察了沙冬青叶肉细胞液泡中泡状内含物的形成。在早期,这种泡状内含物位于细胞质中,它由大小不等、形态各异的小泡组成,后经液泡膜内吞进入液泡。液泡中的泡状内含物主要位于两个正常叶绿体之间,附近的细胞质较多,内有丰富的内质网、高尔基体、质膜管状突起和由它们产生的小泡。也有一些液泡泡状内含物出现在解体叶绿体附近。前者主要来自内质网、高尔基体和质膜,后者则主要起源于解体的叶绿体。这种泡状内含物的形成可能与增强植物的抗冻性有关。  相似文献

Lepidium sativum (garden cress) seed oil was examined for its antimicrobial, antioxidant, and anti-inflammatory activities. The oil was obtained by hydrodistillation, where gas chromatography coupled with mass spectrometry that utilized to study its chemical composition. Microdilution method was used to test the antimicrobial effect of oil against Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Pseudomonas aeruginosa, Salmonella enterica, Klebsiella pneumoniae, and Candida albicans. The antioxidant activity was assessed by radical scavenging activity assay using 2,2-diphenyl-1-picrylhydrazyl radical. The major constituents found in the oil were 7,10-hexadecadienoic acid, 11-octadecenoic acid, 7,10,13-hexadecatrienoic acid, and behenic acid. The minimum inhibitory concentration (MIC) against all pathogens was 47.5 mg/ml, except for Salmonella enterica, which showed MIC of 90 mg/ml. The oil demonstrated antioxidant activity in a dose dependent pattern, with a half maximal inhibitory concentration (IC50) value of 40 mg/ml, and exerted anti-inflammatory activity, wherein 21% protection was shown at a concentration of 300 μg/ml. Thus, L. sativum seed oil shows antimicrobial, antioxidant, and anti-inflammatory properties.  相似文献

Cluster root formation in white lupin (Lupinus albus L.) isinduced mainly by phosphorus (P) starvation, and seems to beregulated by the endogenous P status of the plant. Increasedformation of cluster roots, when indole acetic acid is suppliedto the growth medium of P sufficient plants, and inhibitoryeffects of kinetin application suggest the involvement of endogenousphytohormones (auxins and cytokinins), which may act in an antagonisticmanner in the P-starvation response. Phosphorus deficiency-inducedadaptations of white lupin, involved in P acquisition and mobilizationof sparingly available P sources, are predominantly confinedto the cluster roots, and moreover to distinct stages duringtheir development. Increased accumulation and exudation of citrateand a concomitant release of protons were found to be mainlyrestricted to mature root clusters after prolonged culture (3–4weeks) under P-deficient conditions. Inhibition of citrate exudationby exogenous application of anion channel antagonists such asethacrynic- and anthracene-9-carboxylic acids may indicate involvementof an anion channel. Phosphorus deficiency-induced accumulationand subsequent exudation of citric acid seems to be a consequenceof both enhanced biosynthesis and reduced turnover of citricacid in the cluster root tissue, indicated by enhanced expressionof sucrose synthase, fructokinase, phosphoglucomutase, phosphoenol-pyruvatecarboxylase, but reduced activity of aconitase and slower rootrespiration. The release of acid phosphatase and of phenoliccompounds (isoflavonoids) as well as the induction of a putativehigh-affinity P uptake system was more highly expressed in juvenile,mature and even senescent cluster regions than in apical zonesof non-proteoid roots. The lichen compound usnic acid (UA) is a lipophilic weak acid that acts as a proton shuttle and causes loss of mitochondrial inner membrane potential. In the current study we show that UA treatment induced the formation of autophagosomes in human cancer cells, but had minimal effects on normal human fibroblasts. However, autophagic flux was incomplete, degradation of autophagosomal content did not occur and acidification was defective. UA-treated cells showed reduced ATP levels and activation of AMP kinase as well as signs of cellular stress. UA is thus likely to trigger autophagosome formation both by energy depletion and stress conditions. Our findings indicate that the H+-shuttling effect of UA operates not only in mitochondria as previously shown, but also in lysosomes, and have implications for therapeutic manipulation of autophagy and pH-determined drug distribution.  相似文献

Abstract. The phototropic and the geotropic sensitivities of cress hypocotyls differed in etiolated and in green seedlings. In etiolated seedlings, phototropism was poorly developed and geotropism seemed to be the dominant orientation mechanism. In green seedlings, although geotropic sensitivity was slightly greater, phototropic responses were markedly enhanced, such that photo-signals could over-ride, or even reverse, geocurvature. The 'de-etiolation' light pretreatments required to bring about these changes in the photo-and geo-systems were different. The slight enhancement of geocurvature could be achieved by short-term exposure to red light and was reversed by far-red irradiation. The marked stimulation of photocurvature required extensive irradiation pretreatments.  相似文献

The glycolytic enzyme aldolase is concentrated in a domain around stress fibers in living Swiss 3T3 cells, but the mechanism by which aldolase is localized has not been revealed. We have recently identified a molecular binding site for F-actin on aldolase, and we hypothesized that this specific binding interaction, rather than a nonspecific mechanism, is responsible for localizing aldolasein vivo.In this report, we have used fluorescent analog cytochemistry of a site-directed mutant of aldolase to demonstrate that actin-binding activity localizes this molecule along stress fibers in quiescent cells and behind active ruffles in the leading edge of motile cells. The specific cytoskeletal association of aldolase could play a structural role in cytoplasm, and it may contribute to metabolic regulation, metabolic compartmentation, and/or cell motility. Functional duality may be a widespread feature among cytosolic enzymes.  相似文献

Vacuoles of immature apple fruit (Malus pumila Mill. var. domesticaSchneid.) were obtained by purification using Ficoll densitygradient centrifugation after lysis of the protoplasts by bothmild osmotic shock and the addition of EDTA and BSA. The recoverywas about 35% of the protoplasts. The isolated vacuoles hada mean diameter of about 100 µm. The distribution of sugars, organic acids, phenolic compoundsand amino acids in the vacuole, the cytoplasm and the free spacewas determined. Almost all of the fructose and glucose, themajor sugars of the tissue, were found in the vacuole. Sorbitolwas mainly located in the free space and the vacuole, and sucrosein the free space and the cytoplasm. More than 90% of the malicacid, the main organic acid, was located in the vacuole. Almostall of the phenolic compounds were also deposited in the vacuole. The volumes of the vacuole, the cytoplasm and the free spacein the whole tissue were calculated from the cell numbers ofthe whole tissue, the volume of the isolated protoplasts, andthe volume of the vacuoles present in the protoplast. The soluteconcentration in each compartment was estimated: vacuoles, 888mM; cytoplasm, 37 mM; free space, 57 mM. All three floating plants have roots bearing laterals derivedfrom both pericycle and endodermis. In Pistia and Eichhornialaterals arise within the meristem of the mother root; in Hydrocharisthey arise from mature tissue. In Pistia and Hydrocharis theepidermis becomes anatomically discrete between cortex and cap:in Pistia it is derived from the endodermis of the mother root,in Hydrocharis from the pericycle. The epidermis is not discretein Eichhornia and is derived from the pericycle of the motherroot with the cortex. Stathmokinetic data were used to construct timetables of developmentwhich show how the differences arise. In Pistia the first periclinaldivision of the endodermis-derived tissue individualizes theepidermis and occurs early, before a quiescent centre forms.In Hydrocharis the epidermis also becomes discrete before thepole of the meristem becomes quiescent, but it does so by apericlinal division of the pericycle-derived tissue. In Eichhorniapericlinal divisions occur in the outermost layer of the pericycle-derivedtissue long after quiescence has set in at the pole and afterthe fourth periclinal division in the endodermis derived cap.Its epidermis therefore never becomes anatomically discretethough it becomes functionally discrete because its polar cellsstop dividing as in the other plants. The involvement of the endodermis of mother roots in the formationof laterals is discussed in relation to the state of differentiationat sites of primordium formation, discreteness of the epidermisand subsequent fate of primordia. 为研究外源硫化氢(H2S)对豌豆(Pisum sativum)胚根生理特性及其边缘细胞的影响,测定了不同浓度外源H2S处理下豌豆根长、根尖组织可溶性蛋白质含量、抗氧化酶(APX、CAT、POD和SOD)活性、根尖边缘细胞存活率及其粘胶层相对面积的变化。结果表明:低浓度(0-40μmol·L-1)H2S可促进豌豆胚根生长,根尖组织可溶性蛋白质含量升高,SOD、APX和POD活性增加,CAT活性降低,根尖边缘细胞存活率上升,粘胶层相对面积变小。高浓度(60-80μmol·L-1)H2S可抑制豌豆胚根生长,可溶性蛋白质含量和边缘细胞存活率明显下降,APX和POD活性降低,CAT活性升高,SOD活性没有明显变化,粘胶层相对面积变大;边缘细胞染色体凝集并边缘化,然后逐渐降解并伴随粉末化,细胞质膜皱缩。因此,推测H2S可能在植物体内发挥着重要的生理作用。  相似文献

The position of the contractile vacuoles in non-dividing Paramecium aurelia, as judged by that of the pores, is relatively constant both laterally and longitudinally, as is the distance between them. Some variability of the distance between the pores persists from the recent fission among the smaller animals of normal shape. At the onset of fission two new vacuoles appear, one anterior to each of the old vacuoles; all the vacuoles must undergo relocation because their mean positions now are somewhat removed from that in the non-dividing animals. The pores of the front daughter are too close together: the anterior pore is too far back, and the posterior is too far forward; likewise the pores of the hind daughter are too close together: the anterior too far forward as well as the posterior. The relocation does not begin until shortly before separation of the daughters and is practically complete by the time the daughters have assumed their normal length/width ratio after separation. A neuroneme connects the pore to one of the basal granules adjacent to its place of origin, now far removed.
Occasionally more than one new pore appears at the usual time and place during very early fission; these are very close together and usually serve a single vacuole. However, most of them fail to survive the relocation following this and the next fission, so they are found in the posterior position very rarely. But, in some individuals they may survive and serve a single or separate vacuoles. In a certain clone of Paramecium aurelia multiple pores and vacuoles are rather frequent; these are, of course, the result of the tendency to produce multiple pores and vacuoles at the time of fission.  相似文献
