Single and joint action bioassays using spent lubrication oil, laundry detergent and a mixture of both (based on a 1:1 ratio of their 96h LC50 values) against the guppy Poecilia reticulata were carried out. On the basis of 96h mortality data, laundry detergent (LC50 value - 0.033g l?1) was found to be approximately 1 147 times more toxic than spent lubrication oil (96hrs LC50 — 37.72g l?1) when acting singly against P. reticulata. Joint action bioassays revealed a synergistic effect, with the mixture of the two test compounds being about 12 000 and 10 times more toxic, respectively, than either spent lubrication oil or laundry detergent acting singly. The significance of these results should affect both local and national waste disposal management policies.  相似文献

This study assessed the histological changes in the epidermis of guppies Poecilia reticulata induced by waterborne zinc (Zn). Laboratory‐reared P. reticulata fry were maintained individually in separate vessels containing artificial water (8 µg l?1 Zn) to which 0, 15, 30, 60 or 120 µg l?1 Zn was added. Their epidermal response to Zn was monitored regularly over 4 weeks. Compared with controls, mucus was rapidly released and mucous cell numbers decreased at all concentrations. Thereafter mucous release, epidermal thickness, numbers and size of mucous cells fluctuated at a rate that varied with Zn concentration, but fluctuations declined after day 18. Results clearly highlight the dynamic nature of the epidermal response to sublethal concentrations of waterborne Zn. In general, low concentrations of Zn induced a rapid response with reduced numbers and size of mucous cells and shift in mucin composition, and a subsequent thickening of the epidermis. Epidermal thickness and mucous cell area fluctuated over time but were normal after a month of exposure to low Zn concentrations. The number of mucous cells, however, remained low. Virtually all mucous cells from fish maintained in 15 and 60 µg l?1 Zn contained acidic mucins throughout the month, whereas fish maintained at 30 µg l?1 Zn responded by production of neutral mucins during the first 12 days followed by a mixture of neutral and acidic mucins. At 120 µg l?1 Zn, the most dramatic effects were the gradual but sustained decrease in numbers and area of mucous cells, and the shift to acidic mucins in these cells. Thus, as concentration of Zn increased, the epidermal responses indicated a disturbed host response (dramatic decline in mucous cell numbers, with mixed composition of mucins), which may have been less effective in preventing Zn uptake across the epithelium.  相似文献

Norepinephrine (NE) and phenylephrine (Phe) were employed to study the ionic requirements for alpha adrenoceptor activation in the teleost Poecilia reticulata melanophores. As expected the beta adrenoceptor blocker, propranolol, increased the sensitivity of the preparation to NE (5.8 times), and was therefore employed in all the experimental procedures. Neither cocaine (a neuronal uptake blocker) nor dexamethasone (an extraneuronal uptake blocker) enhanced the sensitivity of the preparation to NE, suggesting that these inactivating mechanisms would not play a role in P. reticulata pigmentary system. However, in the absence of calcium, the dose-response curve (DRC) to NE was displaced to the left about 3.5 times, whereas the DRC to Phe was not affected. These results indicate that a neuronal uptake is active, but was not demonstrated by the classical pharmacological tools, probably due to an assymmetric display of the nervous endings. The DRC to NE was rightward displaced (14.1 times) in the presence of the calcium channel blocker Verapamil, whereas the DRC to Phe was not affected. These data suggest that P. reticulata melanophores possess a mixed population of alpha1 and alpha2 adrenoceptors, the activation of the latter eliciting an extracellular calcium influx. In sodium-free saline, the DRC to NE was rightward shifted (6.6 times) and the response to Phe was impaired in such a way that the maximal response was not achieved. The DRC to both NE and Phe were rightward displaced (7.9 and 2.7 times respectively) in the presence of the sodium channel blocker tetrodotoxin (TTX) 10?7M. In potassium-free saline, the melanophore sensitivity to Phe was increased, whereas the responses to NE were not affected, suggesting a differential sensitivity of the two alpha adrenoceptor subtypes to the resulting membrane hyperpolarization. Based on the literature and on our data we propose that P. reticulata melanophores possess a mixed population of alpha1 and alpha2 receptors. The activation of both subtypes of alpha adrenoceptors elicits a Na+ ion influx through TTX-sensitive sodium channels. The stimulation of alpha2 adrenoceptors also requires an extracellular calcium influx, through the opening of slow calcium channels.  相似文献

Abstract: Pridefine (AHR-1118) is a pyrrolidine derivative with clinically established antidepressant efficacy. Previous work from this laboratory indicates that pridefine is a reuptake blocker of catecholamines and serotonin with weak releasing activity. This study characterized the mode of amine uptake inhibition by pridefine as noncompetitive. The uptake experiments were performed utilizing ouabain instead of zero-degree controls to differentiate between the passive and active components of uptake. Furthermore, the passive component was resolved into diffusion and binding of substrate. Correction was made for the effects of ouabain on binding. Kinetic constants determined from Lineweaver-Burk plots were: Km= 3 × 10?7 M for NE, Km= 9 × 10?8 M for DA, and Km= 3 × 10?8 M for 5-HT. Dixon analyses of uptake at various pridefine concentrations indicated noncompetitive inhibition with Ki= 2.5 × 10?6 M for NE uptake, Ki= 2.0 × 10?6 M for DA uptake, and Ki= 1 × 10?5 M for 5-HT uptake. These constants compare well with IC50 values for the same transmitters: NE, IC50= 2.4 × 10?6 M; DA, IC50= 2.8 × 10?6 M; 5-HT, IC50= 1.0 × 10?5 M. The in vitro results indicate that pridefine is relatively specific as a catecholamine uptake blocker. It differs from tricyclic antidepressants which are reportedly competitive inhibitors of monoamine uptake. The possible mechanisms by which pridefine acts as a noncompetitive inhibitor are discussed.  相似文献

Sperm collection methods and the effect of osmolality, ions, sugar, temperature, pH and dilution ratio on sperm motility were investigated in guppies Poecilia reticulata. The present study revealed that the sperm was motile in a wide range of osmolalities (200–470 mOsm kg?1) either in Hanks balanced‐salt solution (HBSS) or in non‐electrolyte solutions such as glucose or sucrose. Sperm collected from crushing testes yielded lower motility and shorter motility duration than samples collected without crushing but gentle disruption. Dilution ratios within the range of 1:50 to 1:500 of sperm to HBSS had minimal effect on sperm motility during extended refrigerated storage. Examination of storage temperature showed that refrigerated storage at 4° C was superior to room temperature (25° C). Sperm was found to tolerate a wide range of pH from 5·6 to 7·8, but motility was affected negatively by pH values >7·8.  相似文献

In this study the uptake by guppies (Poecilia reticulata) of 1,2,3-trichlorobenzene (TCB) and hexachlorobenzene (HCB) from sediment-free water was compared with uptake in the presence of suspended sediment. The results show that the influence of suspended sediment on the uptake of chlorobenzenes varies with test compound. For TCB uptake was not influenced by the presence of suspended sediment. This is probably due to the large amount of the chemical which is dissolved relative to the amount which is present in the sorbed state. For the more hydrophobic HCB, the concentration found in the fish from the system with suspended sediment was significantly higher than in fish from the control experiment.  相似文献

Permeability of Boric Acid Across Lipid Bilayers and Factors Affecting It   总被引:13,自引:0,他引:13  
Boron enters plant roots as undissociated boric acid (H3BO3). Significant differences in B uptake are frequently observed even when plants are grown under identical conditions. It has been theorized that these differences reflect species differences in permeability coefficient of H3BO3 across plasma membrane. The permeability coefficient of boric acid however, has not been experimentally determined across any artificial or plant membrane. In the experiments described here the permeability coefficient of boric acid in liposomes made of phosphatidylcholine was 4.9 × 10−6 cm sec−1, which is in good agreement with the theoretical value. The permeability coefficient varied from 7 × 10−6 to 9.5 × 10−9 cm sec−1 with changes in sterols (cholesterol), the type of phospholipid head group, the length of the fatty acyl chain, and the pH of the medium. In this study we also used Arabidopsis thaliana mutants which differ in lipid composition to study the effect of lipid composition on B uptake. The chs1-1 mutant which has lower proportion of sterols shows 30% higher B uptake compared with the wild type, while the act1-1 mutant which has an increased percentage of longer fatty acids, exhibited 35% lower uptake than the wild type. Lipid composition changes in each of the remaining mutants influenced B uptake to various extents. These data suggest that lipid composition of the plasma membrane can affect total B uptake. Identifying the environmental factors responsible for the formation of a species' distribution limit is challenging because organisms interact in complex ways with their environments. However, the use of statistical niche models in combination with the analysis of phenotypic variation along environmental gradients can help to reduce such complexity and identify a subset of candidate factors. In the present study, we used such approaches to describe and identify factors responsible for the parapatric distribution of two closely‐related livebearer fish species along a salinity gradient in the lowlands of Trinidad, West Indies. The downstream distribution limits of Poecilia reticulata were strongly correlated with the brackish–freshwater interface. We did not observe significant phenotypic variation in life‐history traits for this species when comparing marginal with more central populations, suggesting that abrupt changes in conditions at the brackish–freshwater interface limit its distribution. By contrast, Poecilia picta was present across a wide range of salinities, although it gradually disappeared from upstream freshwater localities. In addition, P. picta populations exhibited an increase in offspring size in localities where they coexist with P. reticulata, suggesting a role for interspecific competition. The parapatric distribution of these two species, suggests that P. reticulata distributions are limited by an abiotic factor (salinity), whereas P. picta is limited by a biotic factor (interspecific competition). Similar parapatric patterns have been previously described in other systems, suggesting they might be a common pattern in nature. © 2013 The Linnean Society of London  相似文献

A method for the measurement of reactive oxygen species (ROS) in human hepatic tissue has been developed. The method is based on the EPR detection of the nitroxide radical produced by reaction of the hydroxylamine spin-probe bis(1-hydroxy-2,2,6,6-tetramethyl-4-piperidinyl)decandioate with ROS generated under pseudo-physiologic conditions in fine needle biopsies of healthy (10 controls) and diseased (22 patients) human liver. Measures of malonaldehyde in 9 liver biopsies (3 controls and 6 patients) have also been obtained by high pressure liquid chromatography and values parallel those obtained by the spin-probe technique. The amount of ROS found in healthy human liver (median = 1.8 × 10-11 mol/mg) was significantly lower than values found in liver affected by hepatitis B (median = 5.8 × 10-10 mol/mg; p < 0.02) or by hepatitis C (median = 2.7 × 10-9 mol/mg; p < 0.003) as well as compared to some other non-viral liver diseases (NVLD): autoimmune hepatitis, primary biliary cirrhosis, primary schlerosing cholangitis (median = 9.8 × 10-9 mol/mg; p < 0.005). NVLD also showed significantly higher ROS levels compared to hepatitis B (p < 0.04) and hepatitis C (p < 0.04).

Short-term (3h) acquisition of iron (16 nmol 59FeCl3 l−1) from oxic, alkaline fresh water was assessed in rainbow trout Oncorhynchus mykiss in the presence or absence of a range of iron chelators, all of which had differing binding affinities for ferric iron [100 μmol l−1 of desferrioxamine (DFO), Log10K1 32·5; citric acid Log10K1 11·9; nitrilotriacetic acid (NTA) Log10K1 15·9, CP20 and CP94 (Log10K1 > 30), as well as humic acid (HA), Log10K1 5·04, 5 mg l−1]. In the absence of chelators (control conditions) O. mykiss acquired iron from the water under laboratory lights (wavelength range of the lights 440–650 nm, peak intensity 548–626 nm) via the gill. In these conditions iron uptake onto the gill had a maximum transport capacity (Jmax) of 11·2 pmol Fe g−1 h−1 (gill organ mass) and a Km of 21·3 nmol Fe l−1 h−1. Furthermore, there were two components to iron accumulation into the carcass of these fish, a slow rate of aqueous iron uptake at low concentrations (6–24 nmol Fe l−1), followed by a faster rate of uptake at higher iron concentrations (48–96 nmol Fe l−1), suggesting that the rate-limiting step of iron uptake at low iron concentrations is the apical entry step. O. mykiss also acquired iron in the presence of HA, although the majority of the other chelators prevented iron uptake. Ultraviolet light (354 nm) treatment of Fe-DFO increased iron bioavailability. Results suggest that rainbow trout are able to access either the predicted very low concentrations (picomolar) of ferrous iron present in fresh water or the ferric oxide complexes present in oxic environments. The iron uptake rate measured (0·75 pmol g−1 h−1) would be sufficient to provide a substantial proportion (c. 85%) of the daily iron requirements of growing salmonid fry.  相似文献

In previous experiments, rates of picoplankton uptake into coral communities were controlled by sponge and ascidian biomass. Those experimental communities, however, had relatively few sponges and ascidians. In contrast, turbulent transport of particles into the momentum boundary layers can limit particle removal by layered, dense bivalve populations. In this study, the role of water velocity in controlling particulate nutrient-uptake by rubble communities was evaluated, in which the rubble was more completely covered by sponges and ascidians. Picoplankton uptake was proportional to concentration over a range of cell concentrations from 3.0 × 105 to 9.5 × 105 heterotrophic bacteria ml−1, 4.1 × 104 to 1.2 × 105 Synechococcus sp. ml−1 and 6.3 × 103 to 1.8 × 104 picoeukaryotes ml−1. The first-order uptake rate constants, normalized to sponge and ascidian biomass, were similar to previous experimental communities. Picoplankton uptake increased 1.6-fold over a 7-fold change in water velocity, 0.05–0.35 m s−1. This increase has been interpreted as a result of higher turbulent transport within the rough coral community (canopy), as indicated by a 1.6-fold increase in the bottom friction with increasing water velocity.  相似文献

Aims: To determine the contribution of potential modes of action of a Bacillus cereus aquaculture biological control agent in inhibition of the fish pathogen, Aeromonas hydrophila. Methods and Results: When B. cereus was tested in plate well inhibition studies, no production of antimicrobial compounds was detected. Bacillus cereus had a high growth rate (0·96 h?1), whereas Aer. hydrophila concentration decreased by c. 70% in co‐culture experiments. In nutrient limitation studies, B. cereus had a significantly higher growth rate when cultured under glucose (P < 0·05) and iron (P < 0·01) limitation in comparison with Aer. hydrophila. Bacillus cereus glucose (0·30 g l?1 h?1) and iron (0·60 mg l?1 h?1) uptake rates were also significantly higher (P < 0·01) than the Aer. hydrophila glucose (0·14 g l?1 h?1) and iron (0·43 mg l?1 h?1) uptake rates. Iron uptake was facilitated by siderophore production shown in time profile studies where relative siderophore production was c. 60% through the late exponential and sporulation phases. Conclusions: Competitive exclusion by higher growth rate, competition for organic carbon and iron, facilitated by siderophore production, could be identified as mechanisms of pathogen growth inhibition by B. cereus. Significance and Impact of the Study: This study is the first elucidation of the mechanism of action of our novel B. cereus biological agent in growth attenuation of pathogenic Aer. hydrophila. This study enhances the application knowledge and attractiveness for adoption of B. cereus NRRL 100132 for exploitation in aquaculture.  相似文献

A study was made of the time course and kinetics of [3H]GABA uptake by dispersed cell cultures of postnatal rat cerebellum with and without neuronal cells. The properties of GABA neurons were calculated from the biochemical difference between the two types of cultures. It was found that for any given concentration of [3H]GABA, or any time up to 20 min, GABA neurons in cultures 21 days in vitro had an average velocity of uptake several orders of magnitude greater than that of nonneuronal cells. In addition, the apparent Km values for GABA neurons for high and low affinity uptake were 0.33 × 10−6 M and 41.8 × 10−4 M, respectively. For nonneuronal cells, the apparent Km for high affinity uptake was 0.29 × 10−6 M. The apparent Vmax values for GABA neurons for high and low affinity uptake were 28.7 × 10−6 mol/g DNA/min and 151.5 mmol/g DNA/min, respectively. For nonneuronal cells, the apparent Vmax for high affinity uptake was 0.06 × 10−6 mol/g DNA/min. No low affinity uptake system for nonneuronal cells could be detected after correcting the data for binding and diffusion. By substituting the apparent kinetic constants in the Michaelis-Menten equation, it was determined that for GABA concentrations of 5 × 10−9 M to 1 mM or higher over 99% of the GABA should be accumulated by GABA neurons, given equal access of all cells to the label. In addition, high affinity uptake of [3H]GABA by GABA neurons was completely blocked by treatment with 0.2 mM ouabain, whereas that by nonneuronal cells was only slightly decreased. Most (75–85%) of the [3H]GABA (4.4 × 10−6 M) uptake by both GABA neurons and nonneuronal cells was sodium and temperature dependent.  相似文献

Growth, pigmentation and antibacterial activity of Monascus purpureus Went (starch fungus) were affected by zinc. Zinc at concentrations of 2 × 10-3M and 3 × 10-3M nearly stopped the growth, pigmentation and antibiotic production of both wild type and strain NI IS in liquid medium. Relatively vigorous growth, high pigment production, and strong antibacterial activity of NIIS still occurred on solid medium with the same zinc concentrations. Wild type was hardly affected by zinc concentrations lower than 2 × 10-3M. The growth of strain NI IS was significantly inhibited even by 5 × 10-5M zinc, but pigment production and antibacterial activity were highly promoted by 5 × 10-5M zinc. Increase of glucose concentration also promoted pigment and antibiotic productions of strain NI IS. Glucose effects were intensified as 1 × 10-3M zinc was added. Zinc may act as a growth inhibitor and concomitantly as a stimulant for glucose uptake and for the synthesis of metabolites such as pigments and antibiotics.  相似文献

Abstract: Primary astrocyte cultures from neonatal rat brains show uptake of [3H]norepinephrine ([3H]NE). This uptake has a high-affinity component with an apparent Km of approximately 3 × 10?7 M. At 10?7 M [3H]NE both the initial rate of uptake and steady-state content of [3H]NE is inhibited by up to 95% by omission of external Na+. The Na+-dependent component of this uptake is totally inhibited by the tricyclic antidepressants desipramine (DMI) and amitryptyline with IC50 values of 2 × 10?9 and 4 × 10?8 M, respectively. Inhibition of [3H]NE uptake by DMI shows competitive kinetics. These characteristics are essentially identical to those found for high-affinity uptake of NE in total membrane or synaptosome fractions from rodent brains and suggests that such uptake in neural tissue is not exclusively neuronal.  相似文献

The kinetics and thermodynamics of Ga(III) exchange between gallium mononitrilotriacetate and human serum transferrin as well as those of the interaction between gallium-loaded transferrin and the transferrin receptor 1 were investigated in neutral media. Gallium is exchanged between the chelate and the C-site of human serum apotransferrin in interaction with bicarbonate in about 50 s to yield an intermediate complex with an equilibrium constant K 1 = (3.9 ± 1.2) × 10−2, a direct second-order rate constant k 1 = 425 ± 50 M−1 s−1 and a reverse second-order rate constant k −1 = (1.1 ± 3) × 104 M−1 s−1. The intermediate complex loses a single proton with proton dissociation constant K 1a = 80 ± 40 nM to yield a first kinetic product. This product then undergoes a modification in its conformation which lasts about 500 s to produce a second kinetic intermediate, which in turn undergoes a final extremely slow (several hours) modification in its conformation to yield the gallium-saturated transferrin in its final state. The mechanism of gallium uptake differs from that of iron and does not involve the same transitions in conformation reported during iron uptake. The interaction of gallium-loaded transferrin with the transferrin receptor occurs in a single very fast kinetic step with a dissociation constant K d = 1.10 ± 0.12 μM and a second-order rate constant k d = (1.15 ± 0.3) × 1010 M−1 s−1. This mechanism is different from that observed with the ferric holotransferrin and suggests that the interaction between the receptor and gallium-loaded transferrin probably takes place on the helical domain of the receptor which is specific for the C-site of transferrin and HFE. The relevance of gallium incorporation by the transferrin receptor-mediated iron-acquisition pathway is discussed.  相似文献

Rapid Effects of Abscisic Acid on Ion Uptake in Sunflower Roots   总被引:1,自引:0,他引:1  
Short-term effects of ABA, ABA + kinetin and kinetin on ion (86Rb-potassium and phosphate) and water uptake in sunflower plants (Helianthus annuus var. californicus) were examined with a continuous-recording technique. Ion uptake in the roots and transport to the shoots were also investigated by conventional tracer uptake experiments and by sap bleeding experiments with excised roots. After addition of 5 × 10?6-4 × 10?5M ABA to the root medium there was an immediate decrease (30–70%) in the rate of ion uptake which lasted 30–70 min. The rate of water uptake was not significantly affected as measured with this method. Ion transport to the shoots and to the bleeding sap of excised roots was decreased by ABA. ABA-induced inhibition of ion uptake was abolished by the presence of kinetin, and uptake was slightly stimulated by 2 × 10?5M kinetin alone. We suggest that concentration gradients of ABA or rapid changes in the ABA-kinetin balance in the roots affect ion uptake and transport.  相似文献

Noradrenaline, a Transmitter Candidate in the Retina   总被引:5,自引:3,他引:2  
The occurrence, metabolism, uptake, and release of noradrenaline were studied in the bovine retina with the following results. (1) Small amounts of noradrenaline occur in the retina and are restricted to the area corresponding to the inner nuclear and plexiform layers. (2) Retinal tissue can metabolise [14C]dopamine to form quantities of [14C]noradrenaline. (3) [14C]Noradrenaline can also be partly metabolised to form [14C]normetanephrine. (4) When bovine retinas were incubated with 5 × 10-7 M-[3H]noradrenaline for 20 min and processed for autoradiography, most of the label was associated with apparent nerve processes in the inner plexiform layer. Biochemical analysis showed that more than 95% of the label was noradrenaline. (5) [14C]Noradrenaline uptake saturated with increasing noradrenaline concentrations and followed Michaelis-Menten kinetics. This uptake could be accounted for by two processes, a high-affinity system with a Km1 of 5 × 10-8 M and a Vmax1 of 0.193 pmol/mg/10 min and a low-affinity system with a Km2 of 6.3 × 10-5 M and a Vmax2 of 0.109 nmol/mg/10 min. (6) Noradrenaline uptake was strongly dependent on temperature and sodium, less dependent on potassium, and independent of calcium and magnesium ions. (7) Centrally acting drugs, such as desipramine, imipramine, desmethylimipramine, and amitriptyline, inhibited noradrenaline uptake by more than 55% at the concentration of 5 × 10-5 M. These drugs at the same concentration diminished dopamine uptake by less than 30%. (8) Noradrenaline uptake is stereospecific, the (-) isomer having a greater affinity for the uptake sites than the (+) isomer. (9) [14C]Noradrenaline in the retina could be released by increasing the external potassium concentration. This release was calcium-dependent and was blocked by 20 mM-cobalt chloride. The present studies could be interpreted as supporting the idea that noradrenaline acts as a transmitter in the retina.  相似文献

It is now well established that the size distribution of phytoplankton plays an important role in primary production processes and nutrient dynamics of coastal environment. In situ observations showed that nanophytoplankton (3–20 μm) contributed 72.08% and58.18% of phytoplankton biomass and 58.32% and 41.14% of primary productivity to Xiamen Western Waters and the northern Taiwan Strait, respectively; picophytoplankton (0.2–3 μm) dominated the biomass (64.70%) and productivity (66.09%) in the southern Taiwan Strait. Furthermore, nanophytoplankton accounted for 75% of phosphate uptake with the highest rate constant (8.3×10-5 s-1) and uptake rate in unit water volume (5.4×10-5 mmol dm-3s-1); picophytoplankton had the highest uptake rate in unit biomass (5.4×10-5 mmol mg-1s-1) and photosynthetic index (3.8 mgC mgChl a-1h-1). All the results highlighted the remarkable characteristics of small size ranged (0.2–20 μm) phytoplankton in subtropical coastal environments: main contributor to phytoplankton biomass and production, high efficiency on organic carbon production and nutrient recycling. Uptake of ammonium by intact soybean (Glycine max Merr. cv. Amsoy) plants can be represented by 3 phases of a single, multiphasic mechanism in the range 1.78 × 10-5-3.57 × 10-3M. Each phase covers a limited concentration range and obeys Michaelis-Menten kinetics. The multiphasic pattern of NH4+ uptake is remarkably consistent at all stages of soybean growth (20, 40, 60 and 80 days).  相似文献
