Cell cultures in the biocompatibility study of synthetic materials   总被引:1,自引:0,他引:1  
In vitro cytotoxicity (Neutral Red uptake, Kenacid Blue and MTT) and cytocompatibility (cell adhesion and proliferation) tests were applied to the biocompatibility study of a series of poly(ester-ether-ester) block copolymers of potential interest as biomaterials. Our results indicate that the copolymer extracts after 72 hours incubation with a 3T3 mouse fibroblast cell line do not induce significant toxic effects. Furthermore, human umbilical vein endothelial cells seeded on thin copolymer films show a normal pattern of growth. We conclude that thein vitro tests used are a valid instrument to evaluate the potential toxic action of synthetic materials on different cell compartments and that the tested materials seem to be promising for future applications in the field of biomedical devices.  相似文献

A colorimetric assay utilising Neutral Red (C.I. 50040), a nuclear stain, was developed to determine the cellular viability of hybridoma cells in microtitre plates. A linear correlation (r=0.99) was found to exist between the uptake of Neutral Red by viable cells and the viable cell count determined by Trypan blue exclusion test. The linearity stretched over the range of cell concentrations normal in batch cultures (2–30×104/0.2 ml) with as little as ±6% intra-plate well-to-well variation and ±10.2% inter-assay variation.Microscopical examinations of viable hybridoma cells stained with Neutral Red showed that it was located in the nucleus. The possble bifunctional activity of the Neutral Red assay as a test for cellular viability and estimating the DNA content of hybridoma cells is discussed along with its application in a drug screening programme.  相似文献

SUMMARY: Dyes which have been used to detect lipolysis (Nile Blue, Aniline Blue and Neutral Red) and others (Methylene Blue, Toluidine Blue and Thionin) were critically examined for their inhibitory effects in both liquid and solid media. It was confirmed that Nile Blue would inhibit the Gram-positive bacteria tested, even at 2 × 10−5. Gram-negative bacteria were not inhibited at 1 × 10−4 and the colour change in fat media was good. Aniline Blue and Neutral Red did not inhibit Gram-positive organisms at 1 × 10−4 and the colour change was moderately good. The other three dyes were not toxic at 2·5 × 10−5, but there was no colour change. When butter fat saturated with the precipitated bases of Nile Blue, Basic Fuchsin, Crystal Violet or Malachite Green was incorporated in peptone-Yeastrel agar, the resulting media were toxic to Gram-positive bacteria. Fat saturated with the bases of Aniline Blue or Neutral Red permitted growth, but the colour change with lipolytic species was not striking, being one of intensity only. Satisfactory growth and colour changes were obtained using butter fat stained with the inert dye Waxolene Green in medium containing Neutral Red in the aqueous phase, and with butter fat stained with the oxazine base of Nile Blue in agar containing Aniline Blue.  相似文献

The in vitro cytotoxicity of the antimicrobial peptide P34 was evaluated in different eukaryotic cells. The food‐grade bacteriocin nisin was also analysed for comparison. Vero cells were treated with different concentrations (0.02–2.5 μg·ml?1) of antimicrobial peptide P34 and nisin. Cell viability and plasma membrane integrity were checked by MTT [3‐(4,5‐dimethylthiazole‐2‐yl)‐2,5‐diphenyltetrazolium bromide], NRU (Neutral Red dye uptake) and LDH (lactate dehydrogenase) assays. The EC50 values of the peptide P34 in MTT and NRU assays were 0.60 and 1.25 μg·ml?1 respectively, while values of nisin found were 0.50 and 1.04 μg·ml?1. In the LDH assay, the EC50 values were 0.65 and 0.62 μg·ml?1 for P34 and nisin, respectively. The peptide P34 revealed similar haemolytic activity on human erythrocytes (5.8%) when compared with nisin (4.9%). The effects on viability, motility and acrosomal exocytosis of human sperm were also evaluated. Nisin and P34 showed similar effects on sperm parameters. The evaluation of cytotoxicity of antimicrobial peptides is a critical step to guarantee their safe use.  相似文献

Marine heatwaves can lead to rapid changes in entire communities, including in the case of shallow coral reefs the potential overgrowth of algae. Here we tested experimentally the differential thermal tolerance between algae and coral species from the Red Sea through the measurement of thermal performance curves and the assessment of thermal limits. Differences across functional groups (algae vs. corals) were apparent for two key thermal performance metrics. First, two reef‐associated algae species (Halimeda tuna and Turbinaria ornata) had higher lethal thermal limits than two coral species (Pocillopora verrucosa and Stylophora pistillata) conferring those species of algae with a clear advantage during heatwaves by surpassing the thermal threshold of coral survival. Second, the coral species had generally greater deactivation energies for net and gross primary production rates compared to the algae species, indicating greater thermal sensitivity in corals once the optimum temperature is exceeded. Our field surveys in the Red Sea reefs before and after the marine heatwave of 2015 show a change in benthic cover mainly in the southern reefs, where there was a decrease in coral cover and a concomitant increase in algae abundance, mainly turf algae. Our laboratory and field observations indicate that a proliferation of algae might be expected on Red Sea coral reefs with future ocean warming.  相似文献

The photodynamic activity of Neutral Red and the new monobrominated Neutral Red was studied in suspensions of Staphylococcus aureus. The effect of mannitol and sodium azide in the presence of 25 μm photosensitizer on lethal photosensitization were investigated. The results of the mechanistic evaluation of Neutral Red showed that both mannitol and sodium azide produced a completed protective effect after irradiation without significant differences between them. The evaluation of monobrominated Neutral Red also showed a protective effect of microorganisms with the addition of mannitol. Although sodium azide produced a protective effect of the photoinactivation, it was incomplete and less than that exhibited by mannitol. The results indicate that the starting reagent, Neutral Red, is a producer of radical species, acting through a type I mechanism, whereas the halogenated derivative of Neutral Red produced reactive oxygen species and a contribution of singlet molecular oxygen cannot be discarded in the photoinactivation of Staphylococcus aureus cells. These results, analyzed together with the previously evaluated properties of the dyes, allow us to explain the differences observed in the photoinactivation of Staphylococcus aureus mediated by both azine photosensitizers.  相似文献

Decolorization of synthetic dyes was performed using cultures of white-rot fungi producing ligninolytic enzymes and radical-generating reactions that could be involved in the mechanism of fungal decolorization. Among the white-rot fungi tested, Pleurotus ostreatus exhibited the highest decolorization rates, and also the highest production of laccase and Mn-peroxidase. P. ostreatus strain f6 gave 69% decolorization of Eosin Yellowish, 96% of Evans Blue, 75% of Phenol Red (all at 1 mM) and 88% of Poly B-411 (20 ppm) during a 14-day treatment. Treatment with Cu/succinic acid/H2O2 resulted in 96% decolorization of Evans Blue and Poly B-411 within 24 h. However, only 48% and 2% decolorization was achieved with Phenol Red and Eosin Yellowish, respectively. Similar decolorization rates were also obtained when Cu was replaced with Co. The results show that treatment of dye-containing solutions with both fungal cultures and biomimetic catalytic reactions results in decolorization.  相似文献

The Karner blue butterfly (KBB) (Lycaeides melissa samuelis), a federally listed species, has historically been a component of barren and savannah ecosystems in the northeastern and midwestern United States. In New York, it now occurs primarily on managed, early‐successional sites such as power line corridors (utility rights‐of‐way) from Albany to Glens Falls. Blue lupine (Lupinus perennis), the exclusive larval food source of the KBB, has been suggested to be the most limiting factor for the butterfly within the eastern portion of its range. Power line corridor management maintains early‐successional habitats by suppressing the regrowth of woody species, creating potentially important habitat for the conservation and restoration of the KBB complex. This research compared the effects of several vegetation‐clearing methods on Blue lupine populations and associated communities of nectar species for KBB over an 8‐year period. Methods evaluated differed in intensity (annual, 4‐, or 8‐year intervals) and type (herbicide or mechanical). Blue lupine and plant community responses did not significantly differ among the treatment types applied to the power line corridors. However, Blue lupine cover, clump size, and density of stems per clump increased following the application of treatments in general. The number and cover of nectar species, total herbaceous cover, and species richness also responded positively to treatment overall. The percentage of non‐native species temporarily increased following treatment but declined to near pre‐treatment levels again as woody cover increased. Temporal changes in plant community composition were not related to management methods. The management tools assessed, mowing and herbicide application, will accomplish the goal of halting or reversing succession, maintaining critical habitat for the KBB in a landscape that provides little other suitable habitat.  相似文献

This study was designed to assess if radiofrequency (RF) radiation induces oxidative stress in cultured mammalian cells when given alone or in combination with ferrous ions (FeSO4). For this purpose the production of reactive oxygen species (ROS) was measured by flow cytometry in human lymphoblastoid cells exposed to 1950 MHz signal used by the third generation wireless technology of the Universal Mobile Telecommunication System (UMTS) at Specific Absorption Rate of 0.5 and 2.0 W/kg. Short (5–60 min) or long (24 h) duration exposures were carried out in a waveguide system under strictly controlled conditions of both dosimetry and environment. Cell viability was also measured after 24 h RF exposure using the Resazurin and Neutral Red assays. Several co‐exposure protocols were applied to test if RF radiation is able to alter ROS formation induced by FeSO4 (RF given before or concurrently to FeSO4). The results obtained indicate that non‐thermal RF exposures do not increase spontaneous ROS formation in any of the experimental conditions investigated. Consistent with the lack of ROS production, no change in cell viability was observed in Jurkat cells exposed to RF radiation for 24 h. Similar results were obtained when co‐exposures were considered: combined exposures to RF radiation and FeSO4 did not increase ROS formation induced by the chemical treatment alone. In contrast, in cultures treated with FeSO4 as positive control, a dose‐dependent increase in ROS formation was recorded, validating the sensitivity of the method employed. Seven phenolic compounds, 1 – 7 , including a new organic acid gallate, mucic acid 1‐ethyl 6‐methyl ester 2‐O‐gallate ( 7 ), were isolated from the MeOH extract of the fruits of Phyllanthus emblica L. (Euphorbiaceae). The structures were elucidated on the basis of extensive spectroscopic analysis and comparison with literature data. Upon evaluated for their antioxidant abilities by 1,1‐diphenyl‐2‐picrylhydrazyl (DPPH), 2,2′‐azinobis(3‐ethylbenzthiazoline‐6‐sulfonic acid) (ABTS), and ferric reducing antioxidant power (FRAP) assays. The inhibitory activities against melanogenesis in B16 melanoma cells induced by α‐MSH, as well as cytotoxic activities against four human cancer cell lines were also evaluated. All phenolic compounds, 1 – 7 , exhibited potent antioxidant abilities (DPPH: IC50 5.6 – 12.9 μm ; ABTS: 0.87 – 8.43 μm Trolox/μm ; FRAP: 1.01 – 5.79 μm Fe2+/μm , respectively). Besides, 5 – 7 , also exhibited moderate inhibitory activities against melanogenesis (80.7 – 86.8% melanin content), even with no or low toxicity to the cells (93.5 – 101.6% cell viability) at a high concentration of 100 μm . Compounds 1 – 3 exhibited cytotoxic activity against one or more cell lines (IC50 13.9 – 68.4%), and compound 1 with high tumor selectivity for A549 (SI 3.2).  相似文献

Complexes of the positively charged, nuclear staining, quinone-imine dyes Nile Blue and Neutral Red with negatively charged tetrachloroplatinum (II) have been prepared in an effort to form neutral drugs which could gain ready access to the cellular nucleus and deliver significant quantities of the reactive tetrachloroplatinum anion to the vicinity of the DNA. Elemental analysis showed that both the Nile Blue and Neutral Red complexes with tetrachloroplatinum (II) comprised 2 mol of dye and 1 mol of tetrachloroplatinum, forming Pt(Nile Blue)2 and Pt(Neutral Red)2. Exposure of superhelical pBR322 DNA to the complexes or the dyes for 24 h followed by agarose gel electrophoresis showed that Neutral Red and Pt(Neutral Red)2 had little effect on DNA conformation, but that both Nile Blue and Pt(Nile Blue)2 could produce single-strand DNA breaks in a dose-dependent fashion. Studies in exponentially growing asynchronous, hypoxic, and normally oxygenated EMT6 cells at normal pH (7.40) and pH 6.45 demonstrated that neither dye was highly toxic, but that both complexes were capable of producing significant cytotoxicity. Both complexes killed normally oxygenated cells more efficiently than hypoxic cells, but Pt(Neutral Red)2 was more cytotoxic at pH 6.45, while Pt(Nile Blue)2 killed significantly more cells at normal pH. Both complexes decreased the survival of hypoxic EMT6 cells as indicated by the slope of the radiation survival curve [dose modifying factor (DMF) 2.90 for Pt(Nile Blue)2 and 1.45 for Pt(Neutral Red)2]. Studies with the FSaIIC murine tumor showed that both complexes were active radiosensitizing agents in vivo [DMF 1.76 for Pt(Nile Blue)2 and 1.25 for Pt(Neutral Red)2]. These results indicate that these new platinum complexes have characteristics which may make them and similar complexes effective radiosensitizing agents in humans.  相似文献

1. Growth rates of seven species of planktonic algae were determined in culture over a range of temperature from 2 to 35 °C. Additional observations on growth and viability were made for 13 species in the temperature range 20–35 °C. 2. There was a wide range of growth rates between species at their optimal temperatures, from 1.7 divisions day?1 (Asterionella formosa) to 0.3 divisions day?1 (Ceratium furcoides). 3. There were considerable differences between species for growth at low and high temperature. Certain algae, including the diatom A. formosa and the flagellates Cryptomonas marssonii, Dinobryon divergens and Eudorina unicocca var. unicocca, had growth rates of 0.4 divisions day?1 or more at 5 °C. The cyanophyte Tychonema (formerly Oscillatoria) bourrellyi, the xanthophyte Tribonema sp., the desmid Staurastrum cingulum and the large dinoflagellate C. furcoides grew poorly or not at all at this temperature. All 21 species tested could grow at 25 °C, but many – including most of the diatoms, some cyanophytes, and all the flagellates – failed to grow persistently at 30 °C. Only Aphanizomenon flosaquae survived with moderate increase at 35 °C, a lethal temperature for the other species. 4. The applicability was considered of proposed quantitative formulations of the rate‐temperature relationship. Simple exponential relationships applied only to very limited lower ranges of temperature. The relationship proposed by B?lehrádek was a better fit over a wider temperature range, but still excluded rate‐decline at high temperature. 5. The interspecific differences found are of potential significance for restrictions in natural distributions associated with season, altitude (especially above 500 m) and latitude.  相似文献

  • The development of different pollen morphs by one specimen – pollen heteromorphism – occurs in ca. one‐third species of the genus Viola. Melanium section species (pansies) stand out in producing the widest range of apertures among Viola species. Aperture number decreases with elevation increase, and faster germination of five‐aperturate pollen as compared with three‐aperturate has previously been postulated.
  • We re‐examined pollen heteromorphism in the context of its viability, and made correlations with elevation (>1500 m a.s.l. versus <1500 m a.s.l.), soil type (metalliferous versus non‐metalliferous; MET versus NMET) and chromosome number based on selected study criteria of ca. 20% karyologically and morphologically strongly differentiated but genetically closely related pansies.
  • A total of 79% of analysed species were heteromorphic, forming three‐ to six‐colp or ate pollen per individual flower. Mean aperture number and pollen viability were not affected by soil type (MET versus NMET). Mean aperture number was also not influenced by elevation or species chromosome number. Positive correlations were established between aperture number and pollen viability, negative between pollen viability and elevation (increasing altitude of 100 m decreased pollen viability by 0.4%) and lack of correlation between chromosome number and pollen viability.
  • The varied frequencies of different pollen morphs among species are not under the general pressure of ecological conditions, as previously postulated for the species of Melanium section. Rather, this trait in pansies, similar to other floral characters (e.g. long, curved nectar spur, ‘landing platform’, posterior petals with nectar guides), is adaptive but dependent on the breeding system (inbreeding versus outbreeding) of the individual species.

A species recovery project for the endangered fish Red‐finned Blue‐eye (Scaturiginichthys vermeilipinnis) commenced in 2009 at Edgbaston Reserve in central western Queensland. To date, the project has included relocations and control of alien Eastern Gambusia (henceforth Gambusia, Gambusia holbrooki), using the piscicide rotenone. Preventing Gambusia colonising springs where Red‐finned Blue‐eye occur or have been relocated is crucial to the success of the project. As such, this study tested the use of porous silt fences that were installed around five springs from 2012. Results indicate that silt fencing is a suitable barrier material and that provided installed silt fences are serviced regularly, the material is an effective method of quarantining populations of endangered species in isolated aquatic environments. Four of the five installed fences were effective, and maintenance consisted of small repairs at regular intervals (quarterly) followed by replacement after 2 years. As such, silt fencing may have wider application in the conservation of endangered fish and other organisms, particularly in areas where isolated waterbodies occur and where their size renders exclusion fencing a practical recovery action.  相似文献

Nine cyclic diarylheptanoids, 1 – 9 , including two new compounds, i.e., 9‐oxoacerogenin A ( 8 ) and 9‐Oβ‐D ‐glucopyranosylacerogenin K ( 9 ), along with three acyclic diarylheptanoids, 10 – 12 , and four phenolic compounds, 13 – 16 , were isolated from a MeOH extract of the bark of Acer nikoense (Aceraceae). Acid hydrolysis of 9 yielded acerogenin K ( 17 ) and D ‐glucose. Two of the cyclic diarylheptanoids, acerogenin A ( 1 ) and (R)‐acerogenin B ( 5 ), were converted to their ether and ester derivatives, 18 – 24 and 27 – 33 , respectively, and to the dehydrated derivatives, 25, 26, 34 , and 35 . Upon evaluation of compounds 1 – 16 and 18 – 35 for their inhibitory activities against melanogenesis in B16 melanoma cells, induced with α‐melanocyte‐stimulating hormone (α‐MSH), eight natural glycosides, i.e., six diarylheptanoid glycosides, 2 – 4, 6, 9 , and 12 , and two phenolic glycosides, 15 and 16 , exhibited inhibitory activities with 24–61% reduction of melanin content at 100 μM concentration with no or almost no toxicity to the cells (88–106% of cell viability at 100 μM ). In addition, when compounds 1 – 16 and 18 – 35 were evaluated for cytotoxic activity against human cancer cell lines, two natural acyclic diarylheptanoids, 10 and 11 , ten ether and ester derivatives, 18 – 22 and 27 – 31 , and two dehydrated derivatives, 34 and 35 , exhibited potent cytotoxicities against HL60 human leukemia cell line (IC50 8.1–19.3 μM ), and five compounds, 10, 11, 20, 29 , and 30 , against CRL1579 human melanoma cell line (IC50 10.1–18.4 μM ).  相似文献

Wing size and shape, expressed as wing loading and aspect ratio respectively, together with bill morphology are parameters that can reveal differences related to the foraging ecology of seabirds. Six species of booby (Sulidae) that inhabit the Pacific are the focus of this study: four mainly pelagic species, Masked Booby Sula dactylatra, Nazca Booby Sula granti, Red‐footed Booby Sula sula and Brown Booby Sula leucogaster, and two coastal species, Blue‐footed Booby Sula nebouxii and Peruvian Booby Sula variegata. Pelagic boobies showed segregation among species in body mass and relative bill size, and they differed in wing morphology (wing loading and aspect ratio) from the coastal boobies. The coastal Peruvian and Blue‐footed Boobies are largely allopatric but overlap in northern Peru. In their area of sympatry, they showed evidence of character displacement in body size and in wing and bill morphology, which suggests that competition plays an important role in sympatry. This study improves our understanding of ecological interactions among Pacific boobies and of how selective pressures have shaped their ecomorphology and foraging behaviours.  相似文献

A new steroid, 20‐hydroxyisofucosterol (stigmasta‐5,24(28)‐diene‐3β,20β‐diol) ( 7 ), along with six known compounds 1 – 6 were isolated from the MeOH extract of the leaves of Sauropus androgynus L. Merr . (Euphorbiaceae). The structure of new steroid was determined by HR‐APCI‐MS and various NMR techniques in combination with literature data. Subsequently, their anti‐inflammatory, cytotoxic activities against five human cell lines, as well as inhibitory activities against the α‐MSH induced melanogenesis on the B16 cell line were evaluated. As the results, steroid compounds, 6 and 7 exhibited moderate cytotoxic to HL60, AZ521, SKBR3, and A549 tumor cell lines (IC50 26.9 – 45.1 μm ) with high tumor selectivity for A549 relative to WI38 cell lines (SI 2.6 and 3.0, resp.). And, flavonoid compounds, 4 and 5 exhibited superior inhibitory activities against melanogenesis (67.0 – 94.7% melanin content), even with no or low toxicity to the cells (90.1 – 99.6% cell viability) at the concentrations from 10 to 100 μm . Furthermore, Western blot analysis suggested that compound 5 could inhibit melanogenesis by suppressing the protein expressions of MITF, TRP‐1, TRP‐2, and tyrosinase.  相似文献

  • 1 Phytoplankton species composition, numerical abundance, spatial distribution and total biomass measured as chlorophyll a concentration were studied in relation to environmental factors in September 1994 (dry season) and March 1995 (rainy season), respectively, in the Kenyan waters of Lake Victoria; 103 species were recorded.
  • 2 Blue‐green algae (Cyanophyceae) were most diverse, followed by diatoms (Bacillariophyceae), green algae (Chlorophyceae) and dinoflagellates (Dinophyceae).
  • 3 Twinspan separated the phytoplankton communities in the Nyanza Gulf and those in the open lake during both seasons. During the dry season, the Nyanza Gulf was strongly dominated by blue‐greens, while diatoms dominated in the open lake. During the rainy season, blue‐greens remained dominant in the Nyanza Gulf although the number of species found was lower than during the dry season; in the open lake, blue‐greens replaced diatoms as the dominant group and there were more species than in the dry season.
  • 4 Canonical correspondence analysis indicated that the phytoplankton species distribution was significantly correlated with turbidity during the dry season and with SiO2 during the rainy season. Chlorophyll a concentrations ranging from 2.0 to 71.5 mg m‐3 in the dry season and 2.0–17.2 mg m‐3 in the rainy season confirm earlier reports of increasing phytoplankton biomass in Lake Victoria since the 1960s.
