Plant elongation in Allium cepa L. cv. Dorata di Parma was stimulated by end-of-day far-red radiation, while the same treatment was ineffective with respect to bulbing response. It was concluded that the Pfr-dependent reactions which control bulbing are completed during the long daily light period (18 h). Day breaks of 3 h fluorescent white light, in the middle of the inductive photoperiod were inhibitory to bulbing. Repeated brief far-red irradiations could substitute for continuous far-red irradiations lasting 3 h in the middle of the photoperiod. Red light alone or applied immediately after each far-red irradiation inhibits bulb formation.  相似文献

Bulb development was followed in four onion cultivars growingin controlled environments under 17 h days in factorial combinationsof photon-flux densities (PFD) of 111 and 333 µmol m–2s–1 with red: far-red spectral ratios (R : FR) of 2.88and 1.41. A PFD of 111with R : FR of 0.86 was used in a secondexperiment. The lower the R : FR or the higher the PFD the higherthe mean bulbing ratio and the ratio of bulb plus sheath toleaf blade dry weight and the earlier the bulb swelling. LowerR : FR accelerated scale initiation as did the higher PFD underR : FR 2.88 but not under R : FR 1.41. In high PFD bulb swellingoccurred before scale initiation because many sheaths of bladedleaves thickened, but in low PFD and low R : FR bulb swellingand scale initiation were concurrent. In the low PFD a low R: FR increased soluble carbohydrate concentrations in sheathsand scales compared to high R : FR. In high PFD, soluble carbohydrateconcentrations did not vary with R : FR and were higher thanunder low PFD. Summary Significant accumulation of photoconvertible protochlorophyll(ide) in the cotyledons of the mustard seedling takes place from 24 h after sowing onwards (25° C). The rate of accumulation in darkness is greatly increased by a pretreatment with red or far-red light. The strong effect of continuous red light, given from the time of sowing, remains fully reversible by a 756 nm-light pulse up to about 18 h after sowing. On the other hand, the effect of continuous far-red light which can be detected at 15 h after sowing is not influenced by a subsequent application of 756 nm-light pulses. An interpretation of the data requires the concept that continuous red light and continuous far-red light act from different sites. Chlorophyll a (Chl a) accumulation in the cotyledons of Scots pine seedlings (Pinus sylvestris L.) is much higher in the light than in darkness where it ceases 6 days after germination. When these darkgrown seedlings are treated with continuous white light (3,500 lx) a 3 h lag phase appears before Chl a accumulation is resumed. The lag phase can be eliminated by pretreating the seedlings with 7 h of weak red light (0.14 Wm-2) or with 14 red light pulses separated by relatively short dark periods (<100 min). The effect of 15s red light pulses can be fully reversed by 1 min far-red light pulses. This reversibility is lost within 2 min. In addition, the amount of Chl a formed within 27 h of continuous red light is considerably reduced by the simultaneous application of far-red (RG 9) light. It is concluded that phytochrome (Pfr) is required not only for the elimination of the lagphase but also to maintain a high rate of Chl a accumulation in continuous light. Since accumulation of 5-aminolevulinate (ALA) responds in the same manner as Chl a accumulation to a red light pretreatment it is further concluded that ALA formation is the point where phytochrome regulates Chl biosynthesis in continuous light. No correlation has been found between ALA and Chl a formation in darkness. Mustard (Sinapis alba L.) seedlings were irradiated with continuous far-red light either with or without a pretreatment with 3 or 6 h of the same far-red light, separated by a 15 h dark period. The pretreatment increases the initial rate of anthocyanin accumulation — as caused by the 2nd light treatment — at least 6-fold but leads to an earlier cessation of anthocyanin accumulation. Moreover, the pretreatment seems to shorten the apparent lag-phase of anthocyanin accumulation considerably but it does not eliminate the lag. If the pretreatment with far-red light is terminated before the seedling reaches competence (with regard to phytochrome and anthocyanin synthesis) the pretreatment has no effect on the apparent lag-phase even though the future capacity of anthocyanin biogenesis is considerably stimulated by the pretreatment. The effect of light on the accumulation of phytochrome in germinatingAvena embryos was determined. A quantitative ELISA using monoclonalantibody AFRC MAC 56 was used to measure specifically type 1(or dark) phytochrome. A pulse of red light given after 14 himbibition but prior to the onset of type 1 phytochrome synthesis,strongly inhibited subsequent type 1 phytochrome accumulation.This effect of red light at 14 h was reversible by far-red lightindicating the involvement of phytochrome. Red light also inhibitedphytochrome synthesis after 18 h and 24 h imbibition but after24 h, far-red light did not reverse the effect. The effect ofred light treatment at 18 h was reversed by giving a pulse offar-red light at any time up to 30 h. Seed germination was notinfluenced by light under the conditions of these experiments.It is proposed that type 2 (or light) phytochrome may be responsiblefor photoregulation of type 1 phytochrome synthesis in germinatingAvena embryos. The effects of red and far-red light on the enhancement of in vitro nitrate reductase activity and on nitrate accumulation in etiolated excised maize leaves were examined. Illumination for 5 min with red light followed by a 4-h dark period caused a marked increase in nitrate reductase activity, whereas a 5-min illumination with far-red light had no effect on the enzyme activity. The effect of red light was completely reversed by a subsequent illumination with the same period of far-red light. Continuous far-red light also enhanced nitrate reductase activity. Both photoreversibility by red and far-red light and the operation of high intensity reaction under continuous far-red light indicated that the induction of nitrate reductase was mediated by phytochrome. Though nitrate accumulation was slightly enhanced by red and continuous far-red light treatments by 17% and 26% respectively, this is unlikely to account for the entire increase of nitrate reductase activity. The far-red light treatments given in water, to leaves preincubated in nitrate, enhanced nitrate reductase activity considerably over the dark control. The presence of a lag phase and inhibition of increase in enzyme activity under continuous far-red light-by tungstate and inhibitors of RNA synthesis and protein synthesis-rules out the possibility of activation of nitrate reductase and suggests de novo synthesis of the enzyme affected by phytochrome.  相似文献

Summary Short term changes in the soluble sugar, starch, and cell-wall carbohydrate content of the mustard seedling have been studied in the different organs during phytochrome induced photomorphogenesis in continuous far-red. The program was: imbibition of seeds 36 hrs dark far-red irradiation. Kinetics have been followed up to 12 hrs after the onset of irradiation.There are no substantial changes in carbohydrate content in the cotyledons and the radicle. In the cotyledons in far-red after a lag-phase of 3 hrs, there is a decrease in oligosaccharide content, and after a lag-phase of 6 hrs, an increase in cell-wall synthesis. The reducing sugar and starch content is not altered upon irradiation. In the radicle immediately after the onset of far-red, there is a temporary rise in the reducing sugar and cell-wall carbohydrate content. However, 6 hrs later the values in far-red again parallel those of the dark control.The important phytochrome dependent changes take place in the hypocotyl. In far-red after a lag-phase of 3 hrs the glucose accumulation is markedly retarded, the sucrose and starch content no longer increased, and the fructose content even decreases below the 3 hrs value. The glucose: fructose ratio, which is constant in dark, is shifted in favour of glucose. The lag-phase of phytochrome controlled hypocotyl elongation is about 1 hr, the lag-phase of the inhibition of cell-wall carbohydrate synthesis is in about the same order of magnitude.There seems to be neither any immediate connection between sugar content and cell-wall carbohydrate synthesis, as shown by the difference in lag-phases, nor does there seem to be any direct relationship between hypocotyl inhibition and overall synthesis of cell-wall material. The relative inhibition of cell-wall synthesis is less than one third of that of hypocotyl elongation (Figs. 5,6). Apparently phytochrome controls hypocotyl elongation by influencing the cell-wall structure.In spite of the fact that fat degradation is higher in far-red than in dark and respiration higher in dark than in far-red (Friederich, 1968), 6 hrs after the onset of far-red the increase of total carbohydrate content declines compared with that in dark. This finding leads to the conclusion that the efficiency of the fat-carbohydrate-transformation is higher in dark than in far-red.  相似文献

We investigated the effect of mixed radiation from light-emitting diodes (LEDs) on the growth and flowering of ageratum, marigold, and salvia bedding plants. Blue, red, and far-red lights were applied under controlled environmental conditions for 28 d. Both the combination of blue-plus-red radiation as well as fluorescent lighting treatment (control) caused increases in dry weights, but shoot lengths were shortest when plants were exposed to blue plus red light compared with either red or blue plus far-red treatments. The number of floral buds as well as the occurrence of flower opening for ageratum and salvia plants was also enhanced under the blue plus red mixture. Likewise, carbohydrate accumulation was stimulated by that combination compared with the other radiation treatments.  相似文献

Four days oldAmaranthus seedlings responded to light treatment with an increase of amaranthin accumulation. With increasing irradiation time, red light caused a saturation effect. Blue light induced a high irradiation response. The blue light effect was reversible to a certain extent by far-red irradiation given at the end of the treatment with blue light. Intermittent red light (3 h red light, 3 h dark, …) caused a higher amaranthin accumulation than 24 h continuous red light. Microsomal fractions isolated from coleoptiles of dark or far-red light grown corn show ATP-dependent Ca2+ accumulation. The microsomal transport from dark but not from far-red light grown tissue could be stimulated by calmodulin. Ca2+ accumulation into mitochondria from coleoptiles of far-red light grown corn in also inhibited as compared to the dark controls. Light irradiation of isolated microsomes and mitochondria had no effect on either Ca2+ uptake nor efflux.  相似文献

In excised wheat leaves, the activity of nitrate reductase was enhanced by a brief pulse of red light and this increase was reversed by far-red light irradiation. Even under continuous far-red light, nitrate reductase activity increased by 258% after 18 h. When leaves were kept in distilled water during exposure to red light and then transferred to potassium nitrate, there was no difference in endogenous nitrate concentration. The nitrate reductase activity was the same whether leaves were floated in potassium nitrate or in distilled water during irradiation. Partial to complete inhibition of enzyme activity was observed when leaves were incubated in actinomycin-D and cycloheximide respectively, following 4 h of red light irradiation.In vitro irradiation of extract had no significant effect on nitrate reductase activity  相似文献

Summary Determinations of cell-doubling times using the technique of colchicine-induced metaphase accumulation showed that after 40 h exposure to red or far-red light the rates of cell division in young rice (Oryza sativa L. cv. Ballila) shoot apices were faster than in dark controls. In red light, the increase was already taking place after 16 h of irradiation but in far-red the rates at this time were slower than in the dark controls. Seedlings became more responsive to far-red light as they continued to grow in darkness between 2 1/2 and 4 1/2 days. Mitotic activity at the apex increased in the leaf primordium and decreased in the sub-summit corpus between the 4th and 5th days of growth in darkness at 30° C.  相似文献

Transgenic tomato [Lycopersicon esculentum (=Solanum lycopersicum)] lines overexpressing tomato PHYA, PHYB1, or PHYB2, under control of the constitutive double-35S promoter from cauliflower mosaic virus (CaMV) have been generated to test the level of saturation in individual phytochrome-signalling pathways in tomato. Western blot analysis confirmed the elevated phytochrome protein levels in dark-grown seedlings of the respective PHY overexpressing (PHYOE) lines. Exposure to 4 h of red light resulted in a decrease in phytochrome A protein level in the PHYAOE lines, indicating that the chromophore availability is not limiting for assembly into holoprotein and that the excess of phytochrome A protein is also targeted for light-regulated destruction. The elongation and anthocyanin accumulation responses of plants grown under white light, red light, far-red light, and end-of-day far-red light were used for characterization of selected PHYOE lines. In addition, the anthocyanin accumulation response to different fluence rates of red light of 4-d-old dark-grown seedlings was studied. The elevated levels of phyA in the PHYAOE lines had little effect on seedling and adult plant phenotype. Both PHYAOE in the phyA mutant background and PHYB2OE in the double-mutant background rescued the mutant phenotype, proving that expression of the transgene results in biologically active phytochrome. The PHYB1OE lines showed mild effects on the inhibition of stem elongation and anthocyanin accumulation and little or no effect on the red light high irradiance response. By contrast, the PHYB2OE lines showed a strong inhibition of elongation, enhancement of anthocyanin accumulation, and a strong amplification of the red light high irradiance response.  相似文献

Mutations in a component of phytochrome A (phyA)-specific light signal transduction, SPA1, result in enhanced responsiveness of Arabidopsis seedlings to red and far-red light. Here, we have examined the effects of spa1 mutations on the two known modes of phyA function, the high-irradiance responses (HIRs) to continuous irradiation with far-red light and the very-low-fluence responses (VLFRs) to inductive pulses of light that establish only a small proportion of active phyA. spa1 mutants exhibited an enhanced VLFR under hourly pulses of far-red light for hypocotyl growth inhibition, cotyledon unfolding, anthocyanin accumulation, block of greening in subsequent white light and negative regulation of phyB signaling. We provide evidence that the phenotype of spa1 mutants in red light is also caused by an increase in the VLFR. Taken together, our results indicate that light-induced hypocotyl growth inhibition in spa1 mutants is primarily due to a VLFR. While wild-type seedlings required hourly pulses of far-red light to induce a VLFR, infrequent irradiation with far-red pulses (every 12 h) was sufficient to induce a strong VLFR of hypocotyl elongation in spa1 mutants. This shows that the effect of the VLFR was more persistent in spa1 mutants than in the wild type. We, therefore, propose that SPA1 has an important function in reducing the persistence of phyA signaling. spa1 mutations also enhanced the HIRs of anthocyanin accumulation and of phyA-mediated responsivity amplification towards phyB. Thus, our results suggest that spa1 mutations amplify both the phyA-mediated VLFR and the HIR.  相似文献

Irradiation of excised stem segments from de-etiolated seedlings of Cuscuta planiflora for 24 h with mixtures of red and far-red light with red to far-red ratios between 0.02 to 1.0 enhanced coiling and formation of prehaustoria. Maximum number of prehaustoria were recorded when red:far-red was near 0.1. Coiling and prehaustoria were observed whenever estimated in vivo Pfr/Ptotal at photoequilibrium was between 0.06 and 0.67. Irradiation of excised stem segments from white light grown seedlings with 12 h blue light also promoted coiling and prehaustoria formation after another 38 h in darkness. Coiling and prehaustoria were not observed in segments pulsed with 10 min red light at the end of 12 h in blue light. Coiling and prehaustoria were observed after photoreversible end-of-day far-red/red/far-red pulses but not after red/far-red/red pulses. A far-red pulse may not reverse inhibition by end-of-day red pulse when far-red is given more than 12 h after the red pulse.  相似文献
