Nitrogen-fixing activity associated with different wetland rice varieties was measured at various growth stages by an in situ acetylene reduction method after the activities of blue-green algae (cyanobacteria) in the flood water and on the lower portion of the rice stem were eliminated. Nitrogen-fixing activities associated with rice varieties differed with plant growth stages. The activities increased with plant age, and the maximum was about at heading stage. The nitrogen fixed during the whole cropping period was estimated at 5.9 kg of N per ha for variety IR26 (7 days) and 4.8 kg of N per ha for variety IR36 (95 days). The population of aerobic heterotrophic N2-fixing bacteria associated with rice roots and stems was determined by the most-probable-number method, using semisolid glucose-yeast extract and semisolid malate-yeast extract media. The addition of yeast extract to the glucose medium increased the number and activity of aerobic heterotrophic N2-fixing bacteria. The glucose-yeast extract medium gave higher counts of aerobic N2-fixing bacteria associated with rice roots than did the malate-yeast extract medium, on which Spirillum-like bacteria were usually observed. The lower portion of the rice stem was also inhabited by N2-fixing bacteria and was an active site of N2 fixation.  相似文献

Many industrial activities produce H2S, which is toxic at high levels and odorous at even very low levels. Chemolithotrophic sulfur-oxidizing bacteria are often used in its remediation. Recently, we have reported that many heterotrophic bacteria can use sulfide:quinone oxidoreductase and persulfide dioxygenase to oxidize H2S to thiosulfate and sulfite. These bacteria may also potentially be used in H2S biotreatment. Here we report how various heterotrophic bacteria with these enzymes were cultured with organic compounds and the cells were able to rapidly oxidize H2S to zero-valence sulfur and thiosulfate, causing no apparent acidification. Some also converted the produced thiosulfate to tetrathionate. The rates of sulfide oxidation by some of the tested bacteria in suspension, ranging from 8 to 50 µmol min?1 g?1 of cell dry weight at pH 7.4, sufficient for H2S biotreatment. The immobilized bacteria removed H2S as efficiently as the bacteria in suspension, and the inclusion of Fe3O4 nanoparticles during immobilization resulted in increased efficiency for sulfide removal, in part due to chemical oxidation H2S by Fe3O4. Thus, heterotrophic bacteria may be used for H2S biotreatment under aerobic conditions.  相似文献

Precipitation is a major driver of biological processes in arid and semiarid ecosystems. Soil biogeochemical processes in these water‐limited systems are closely linked to episodic rainfall events, and the relationship between microbial activity and the amount and timing of rainfall has implications for the whole‐system carbon (C) balance. Here, the influences of storm size and time between events on pulses of soil respiration were explored in an upper Sonoran Desert ecosystem. Immediately following experimental rewetting in the field, CO2 efflux increased up to 30‐fold, but generally returned to background levels within 48 h. CO2 production integrated over 48 h ranged from 2.5 to 19.3 g C m−2 and was greater beneath shrubs than in interplant spaces. When water was applied on sequential days, postwetting losses of CO2 were only half a large as initial fluxes, and the size of the second pulse increased with time between consecutive events. Soil respiration was more closely linked to the organic matter content of surface soils than to rainfall amount. Beneath shrubs, rates increased nonlinearly with storm size, reaching an asymptote at approximately 0.5 cm simulated storms. This nonlinear relationship stems from (1) resource limitation of microbial activity that is manifest at small time scales, and (2) greatly reduced process rates in deeper soil strata. Thus, beyond some threshold in storm size, increasing the duration or depth of soil moisture has little consequence for short‐term losses of CO2. In addition, laboratory rewetting across a broad range in soil water content suggest that microbial activity and CO2 efflux following rainfall may be further modified by the routing and redistribution of water along hillslopes. Finally, analysis of long‐term precipitation data suggests that half the monsoon storms in this system are sufficient to induce soil heterotrophic activity and C losses, but are not large enough to elicit autotrophic activity and C accrual by desert shrubs.  相似文献

Summary The leaching of six Eastern coals was investigated using experimental coal columns subjected to simulated leaching events. Measurements of CO2 assimilation and specific enrichment cultures indicated that the microbial communities of all leachates were dominated by iron- and sulfur-oxidizing chemoautotrophic bacteria. Comparison of CO2 assimilation rates in leachates and core samples of leached coal indicated that most chemoautotrophs remained within coal columns during leaching. Mean numbers of chemoautotrophic bacteria in leachate samples were correlated with concentrations of dissolved iron and sulfate. Leachates from unwashed, run-of-mine coals contained more chemoautotrophs and more iron and sulfate than did leachates from washed, final product coals. After several leachings, the ratio of sulfur oxidizers to iron oxidizers tended to increase. These data suggest that the chemoautotrophic community of final product coals may be pyritelimited. Aerobic heterotrophs constituted a minor component of the microbial community in leachates from the six coals and their abundance and metabolic activity were apparently not influenced by the beneficiation history of the coal. Changes in rates of acetate metabolism may have been related to microbial succession within the heterotrophic community of coal columns. In all leachates, rates of tritiated methylthymidine assimilation were correlated with rates of acetate incorporation but not with CO2 assimilation, even though autotrophs dominated the microflora. Thus, thymidine assimilation rates appear to reflect activities or growth of mainly heterotrophic microorganisms in leachate.  相似文献

Storm events resulted in a marked reduction in the benthic nitrifying activity of a stream receiving geothermal inputs of ammonium. Subsequently, nitrifying activity demonstrated a logarithmic increase until a stable activity was reached 10 to 12 days after the storm event. The rate of increase of poststorm nitrifying activity was used to calculate the nitrifier growth rate in situ (0.0075 to 0.0116 h−1) which was lower than the growth rates observed for laboratory cultures of nitrifying bacteria isolated from the sediments (0.0233 to 0.0990 h−1).  相似文献

The enhanced mineralization of organic nitrogen by bacteriophagous protozoa is thought to favor the nitrification process in soils, in which nitrifying bacteria have to compete with heterotrophic bacteria for the available ammonium. To obtain more insight into this process, the influence of grazing by the bacteriovorous flagellate Adriamonas peritocrescens on the competition for limiting amounts of ammonium between the ammonium-oxidizing species Nitrosomonas europaea and the heterotrophic species Arthrobacter globiformis was studied in the presence of Nitrobacter winogradskyi in continuous cultures at dilution rates of 0.004 and 0.01 h-1. The ammonium concentration in the reservoir was maintained at 2 mM, whereas the glucose concentration was increased stepwise from 0 to 7 mM. A. globiformis won the competition for limiting amounts of ammonium when the glucose concentration in the reservoirs increased, in agreement with previously described experiments in which the flagellates were not included. The numbers of nitrifying bacteria decreased as the numbers of heterotrophic bacteria rose with increasing glucose concentrations. Critical C/N ratios, i.e., ratios between glucose and ammonium in the reservoirs at which no nitrate was found in the culture vessels, of 12.5 and 10.5 were determined at dilution rates of 0.004 and 0.01 h-1, respectively. Below these critical values, coexistence of the competing species was found. The numbers of nitrifying bacteria decreased more in the presence of flagellates than in their absence, presumably by selective predation on the nitrifying bacteria, either in the liquid culture or on the glass wall of the culture vessels. Despite this, the rate of nitrate production did not decrease more in the presence of flagellates than in their absence. This demonstrates that no correlation has to be expected between numbers of nitrifying bacteria and their activity and that a constant nitrification rate per cell cannot be assumed for nitrifying bacteria. Above the critical C/N ratios, low numbers of nitrifying bacteria were still found in the culture vessels, probably because of attachment of the nitrifying bacteria to the glass wall of the culture vessels. Like the numbers of heterotrophic bacteria, the numbers of flagellates increased when the glucose concentrations in the reservoirs increased. Numbers of 2 × 105 and 12 × 105 flagellates ml-1 were found at 7 mM glucose at dilution rates of 0.004 and 0.01 h-1, respectively. It was concluded that the critical C/N ratios were practically unaffected by the presence of protozoa. Although nitrate production rates were equal in the presence and absence of flagellates, the numbers of nitrifying bacteria decreased more strongly in their presence. This indicates a higher activity per nitrifying cell in the presence of flagellates.  相似文献

In lentic systems, hydrology can be dramatically altered after storm events, potentially modifying the carbon budget. In particular, rapid increases in the surface water carbon dioxide partial pressure (pCO2) have been observed following such events. Several processes may explain these shifts in lake CO2 dynamics, including vertical mixing, increases in metabolism, and increases in external loading. To evaluate the relative importance of these various processes, we reconstructed the whole-lake daily CO2 budget using concurrent estimates of lake metabolism and daily CO2 mass balance budgets in two lakes with distinct morphometries located in Québec, Canada. We found that storm events caused variable, but significant, changes in whole-lake CO2 mass. Such events influenced CO2 dynamics indirectly by inducing shifts in lake metabolism, and directly by importing CO2 by the inflowing storm waters. Storm intensity (in terms of total amount of precipitation) influences the balance between these two processes, but the final outcome depends on lake morphometry. Our results suggest that when storms are intense enough to drive lake water renewal rate beyond 1% day?1, external CO2 loadings became the dominant process, overwhelming internal CO2 production. Lakes with slower hydrological turnover, however, are more susceptible to internal regulation and may simply re-allocate CO2 from the hypolimnion to the epilimnion following a storm event. Our results thus suggest that this tightening of the watershed-lake-atmosphere linkage by climatic events is strongly modulated by lake morphometry. These features should be considered when predicting the impact of future climate change on regional C budgets and emissions.  相似文献

The absence of nitrification in soils rich in organic matter has often been reported. Therefore, competition for limiting amounts of ammonium between the chemolithotrophic ammonium-oxidizing species Nitrosomonas europaea and the heterotrophic species Arthrobacter globiformis was studied in the presence of Nitrobacter winogradskyi in continuous cultures at dilution rates of 0.004 and 0.01 h−1. Ammonium limitation of A. globiformis was achieved by increasing the glucose concentration in the reservoir stepwise from 0 to 5 mM while maintaining the ammonium concentration at 2 mM. The numbers of N. europaea and N. winogradskyi cells decreased as the numbers of heterotrophic bacteria rose with increasing glucose concentrations for both dilution rates. Critical carbon-to-nitrogen ratios of 11.6 and 9.6 were determined for the dilution rates of 0.004 and 0.01 h−1, respectively. Below these critical values, coexistence of the competing species was found in steady-state situations. Although the numbers were strongly reduced, the nitrifying bacteria were not fully outcompeted by the heterotrophic bacteria above the critical carbon-to-nitrogen ratios. Nitrifying bacteria could probably maintain themselves in the system above the critical carbon-to-nitrogen ratios because they are attached to the glass wall of the culture vessels. The numbers of N. europaea decreased more than did those of N. winogradskyi. This was assumed to be due to heterotrophic growth of the latter species on organic substrates excreted by the heterotrophic bacteria.  相似文献

Mutants ofAnabaena variabilis, unable to fix nitrogen under aerobic conditions, were used to determine whether nitrogenase synthesis is subject to O2 repression, as is the case in some heterotrophic bacteria. Nitrogenase activity in the mutants was induced as heterocysts matured under microaerophilic conditions. However, addition of 5% O2 to the assay system inhibited activity by 95%. Under aerobic conditions, nitrogenase activity in the mutants could not be detected, but an activity-independent, immunological assay showed that the Fe-Mo protein was present at levels similar to those found in wild-type and mutant strains induced microaerophilically. Reducing the O2 tension of an aerobically induced mutant resulted in a rate of nitrogenase activity induction twice the rate under continuous microaerophilic conditions. These results indicate that O2 does not repress Fe-Mo protein synthesis in these mutants.  相似文献

Biofilms were cultivated on polycarbonate strips in rotating annular reactors using South Saskatchewan River water during the fall of 1999 and the fall of 2001, supplemented with carbon (glucose), nitrogen (NH4Cl), phosphorus (KH2PO4), or combined nutrients (CNP), with or without hexadecane, a model compound representing aliphatic hydrocarbons used to simulate a pollutant. In fall 1999 and fall 2001, comparable denitrification activities and catabolic potentials were observed in the biofilms, implying that denitrifying populations showed similar activity patterns and catabolic potentials during the fall from year to year in this river ecosystem, when environmental conditions were similar. Both nirS and nirK denitrification genes were detected by PCR amplification, suggesting that both denitrifying bacterial subpopulations can potentially contribute to total denitrification. Between 91.7 and 99.8% of the consumed N was emitted in the form of N2, suggesting that emission of N2O, a major potent greenhouse gas, by South Saskatchewan River biofilms is low. Denitrification was markedly stimulated by the addition of CNP, and nirS and nirK genes were predominant only in the presence of CNP. In contrast, individual nutrients had no impact on denitrification and on the occurrence of nirS and nirK genes detected by PCR amplification. Similarly, only CNP resulted in significant increases in algal and bacterial biomass relative to control biofilms. Biomass measurements indicated a linkage between autotrophic and heterotrophic populations in the fall 1999 biofilms. Correlation analyses demonstrated a significant relationship (P ≤ 0.05) between the denitrification rate and the biomass of algae and heterotrophic bacteria but not cyanobacteria. At the concentration assessed (1 ppb), hexadecane partially inhibited denitrification in both years, slightly more in the fall of 2001. This study suggested that the response of the anaerobic heterotrophic biofilm community may be cyclic and predictable from year to year and that there are interactive effects between nutrients and the contaminant hexadecane.  相似文献

Although the absence of nitrate formation in grassland soils rich in organic matter has often been reported, low numbers of nitrifying bacteria are still found in these soils. To obtain more insight into these observations, we studied the competition for limiting amounts of ammonium between the chemolithotrophic ammonium-oxidizing species Nitrosomonas europaea and the heterotrophic species Arthrobacter globiformis in the presence of Nitrobacter winogradskyi with soil columns containing calcareous sandy soil. The soil columns were percolated continuously at a dilution rate of 0.007 h-1, based on liquid volumes, with medium containing 5 mM ammonium and different amounts of glucose ranging from 0 to 12 mM.A. globiformis was the most competitive organism for limiting amounts of ammonium. The numbers of N. europaea and N. winogradskyi cells were lower at higher glucose concentrations, and the potential ammonium-oxidizing activities in the uppermost 3 cm of the soil columns were nonexistent when at least 10 mM glucose was present in the reservoir, although 107 nitrifying cells per g of dry soil were still present. This result demonstrated that there was no correlation between the numbers of nitrifying bacteria and their activities. The numbers and activities of N. winogradskyi cells decreased less than those of N. europaea cells in all layers of the soil columns, probably because of heterotrophic growth of the nitrite-oxidizing bacteria on organic substrates excreted by the heterotrophic bacteria or because of nitrate reduction at reduced oxygen concentrations by the nitrite-oxidizing bacteria. Our conclusion was that the nitrifying bacteria were less competitive than the heterotrophic bacteria for ammonium in soil columns but that they survived as viable inactive cells. Inactive nitrifying bacteria may also be found in the rhizosphere of grassland plants, which is rich in organic carbon. They are possibly reactivated during periods of net mineralization.  相似文献

Hartbeespoort Dam is a hypertrophic man-made lake which is located in the Transvaal Province of South Africa. This region has recently experienced its most severe drought of the century. However, on three occasions in the summer rainy seasons of 1984 and 1985, major rainfalls (> 50 mm) occurred which caused large inflows to the lake. Inflowing river water entered as a density current causing marked silting of the water. Within the epilimnion (0–10 m) prior to these rainfalls there was usually no variation of bacterial numbers with depth, but heterotrophic bacterial activity (glucose uptake) decreased with depth concomitant with primary production. With the increased river inflow bacterial numbers did not increase but bacterial activity at the bottom of the epilimnion (10 m) increased to as high as 2.7 µg C l–1 h–1 in January 1985, reversing the depth profile of bacterial activity within the epilimnion. This resulted in decreased glucose concentrations (Kt + Sn) and turnover times. Heterotrophic activity per cell increased by between 2.5 and 5 times. These data demonstrate that storm events are important phenomena causing short-term changes in the metabolic activity of planktonic heterotrophic bacteria in lakes.  相似文献

In chlorophyll-containing spores of Onoclea sensibilis, depletion of lipid reserves during germination is correlated with increases in the activity of the glyoxylate cycle enzymes isocitrate lyase and malate synthase. In Onoclea, the heterotrophic activity associated with lipid catabolism occurs at the same time that autotrophic activity is taking place. Increases in chlorophyll content and in the activity of glycolate oxidase were recorded during the earliest stages of spore germination. In this species, there is no temporal separation of heterotrophic and autotrophic reactions. Concurrent increases in glyoxylate and glycolate cycle activities appear to occur naturally.  相似文献

Studies were conducted to examine interrelationships between the heterotrophic and phototrophic populations within an epilithic community in the outlet stream of a high alpine lake. Levels of nitrates, phosphates, and total organic compounds in the lake were consistently near the lower limits of detectability. Microscopic examination of the community by phase-contrast light microscopy and scanning electron microscopy revealed diatoms, filamentous algae, and bacteria embedded within a dense gelatinous matrix. Chlorophyll a and primary productivity measurements had peak values in early August, with subsequent declines. Bacterial heterotrophic activity, as measured by Vmax, turnover rate, and relative activity, increased significantly as the phototrophic community declined. This trend in heterotrophic activity was not accompanied by an increase in total bacterial numbers as determined by epi-illuminated fluorescence microscopy. These results suggest that the phototrophic community responded to changes in, or interactions among, various chemical and physical factors throughout the study period. The catabolic activity of the sessile bacteria appeared to be positively influenced by changes in the mat environment resulting from the decline of the phototrophic populations.  相似文献

Mixotrophic growth of the facultatively autotrophic acidophile Thiobacillus acidophilus on mixtures of glucose and thiosulfate or tetrathionate was studied in substrate-limited chemostat cultures. Growth yields in mixotrophic cultures were higher than the sum of the heterotrophic and autotrophic growth yields. Pulse experiments with thiosulfate indicated that tetrathionate is an intermediate during thiosulfate oxidation by cell suspensions of T. acidophilus. From mixotrophic growth studies, the energetic value of thiosulfate and tetrathionate redox equivalents was estimated to be 50% of that of redox equivalents derived from glucose oxidation. Ribulose 1,5-bisphosphate carboxylase (RuBPCase) activities in cell extracts and rates of sulfur compound oxidation by cell suspensions increased with increasing thiosulfate/glucose ratios in the influent medium of the mixotrophic cultures. Significant RuBPCase and sulfur compound-oxidizing activities were detected in heterotrophically grown T. acidophilus. Polyhedral inclusion bodies (carboxysomes) could be observed at low frequencies in thin sections of cells grown in heterotrophic, glucose-limited chemostat cultures. Highest RuBPCase activities and carboxysome abundancy were observed in cells from autotrophic, CO2-limited chemostat cultures. The maximum growth rate at which thiosulfate was still completely oxidized was increased when glucose was utilized simultaneously. This, together with the fact that even during heterotrophic growth the organism exhibited significant activities of enzymes involved in autotrophic metabolism, indicates that T. acidophilus is well adapted to a mixotrophic lifestyle. In this respect, T. acidophilus may have a competitive advantage over autotrophic acidophiles with respect to the sulfur compound oxidation in environments in which organic compounds are present.  相似文献

Climate change scenarios predict tropical cyclones will increase in both frequency and intensity, which will escalate the amount of terrestrial run-off and mechanical disruption affecting coastal ecosystems. Bacteria are key contributors to ecosystem functioning, but relatively little is known about how they respond to extreme storm events, particularly in nearshore subtropical regions. In this study, we combine field observations and mesocosm experiments to assess bacterial community dynamics and changes in physicochemical properties during early- and late-season tropical cyclones affecting Okinawa, Japan. Storms caused large and fast influxes of freshwater and terrestrial sediment – locally known as red soil pollution – and caused moderate increases of macronutrients, especially SiO2 and PO43−, with up to 25 and 0.5 μM respectively. We detected shifts in relative abundances of marine and terrestrially derived bacteria, including putative coral and human pathogens, during storm events. Soil input alone did not substantially affect marine bacterial communities in mesocosms, indicating that other components of run-off or other storm effects likely exert a larger influence on bacterial communities. The storm effects were short-lived and bacterial communities quickly recovered following both storm events. The early- and late-season storms caused different physicochemical and bacterial community changes, demonstrating the context-dependency of extreme storm responses in a subtropical coastal ecosystem.  相似文献

Most heterotrophic bacteria assimilate CO2 in various carboxylation reactions during biosynthesis. In this study, assimilation of 14CO2 by heterotrophic bacteria was used for isotope labeling of active microorganisms in pure cultures and environmental samples. Labeled cells were visualized by microautoradiography (MAR) combined with fluorescence in situ hybridization (FISH) to obtain simultaneous information about activity and identity. Cultures of Escherichia coli and Pseudomonas putida assimilated sufficient 14CO2 during growth on various organic substrates to obtain positive MAR signals. The MAR signals were comparable with the traditional MAR approach based on uptake of 14C-labeled organic substrates. Experiments with E. coli showed that 14CO2 was assimilated during both fermentation and aerobic and anaerobic respiration. The new MAR approach, HetCO2-MAR, was evaluated by targeting metabolic active filamentous bacteria, including “Candidatus Microthrix parvicella” in activated sludge. “Ca. Microthrix parvicella” was able to take up oleic acid under anaerobic conditions, as shown by the traditional MAR approach with [14C]oleic acid. However, the new HetCO2-MAR approach indicated that “Ca. Microthrix parvicella,” did not significantly grow on oleic acid under anaerobic conditions with or without addition of NO2, whereas the addition of O2 or NO3 initiated growth, as indicated by detectable 14CO2 assimilation. This is a metabolic feature that has not been described previously for filamentous bacteria. Such information could not have been derived by using the traditional MAR procedure, whereas the new HetCO2-MAR approach differentiates better between substrate uptake and substrate metabolism that result in growth. The HetCO2-MAR results were supported by stable isotope analysis of 13C-labeled phospholipid fatty acids from activated sludge incubated under aerobic and anaerobic conditions in the presence of 13CO2. In conclusion, the novel HetCO2-MAR approach expands the possibility for studies of the ecophysiology of uncultivated microorganisms.  相似文献

Strong inhibitory effects of the anionic surfactant linear alkylbenzene sulfonate (LAS) on four strains of autotrophic ammonia-oxidizing bacteria (AOB) are reported. Two Nitrosospira strains were considerably more sensitive to LAS than two Nitrosomonas strains were. Interestingly, the two Nitrosospira strains showed a weak capacity to remove LAS from the medium. This could not be attributed to adsorption or any other known physical or chemical process, suggesting that biodegradation of LAS took place. In each strain, the metabolic activity (50% effective concentration [EC50], 6 to 38 mg liter−1) was affected much less by LAS than the growth rate and viability (EC50, 3 to 14 mg liter−1) were. However, at LAS levels that inhibited growth, metabolic activity took place only for 1 to 5 days, after which metabolic activity also ceased. The potential for adaptation to LAS exposure was investigated with Nitrosomonas europaea grown at a sublethal LAS level (10 mg liter−1); compared to control cells, preexposed cells showed severely affected cell functions (cessation of growth, loss of viability, and reduced NH4+ oxidation activity), demonstrating that long-term incubation at sublethal LAS levels was also detrimental. Our data strongly suggest that AOB are more sensitive to LAS than most heterotrophic bacteria are, and we hypothesize that thermodynamic constraints make AOB more susceptible to surfactant-induced stress than heterotrophic bacteria are. We further suggest that AOB may comprise a sensitive indicator group which can be used to determine the impact of LAS on microbial communities.  相似文献

This paper aims to investigate the effect of H2O2 and paraquat on the activities of superoxide dismutase (SOD) and catalase (CAT), and membrane lipid peroxidation (LPO) levels in newly isolated Streptomyces sp. M3004. SOD activities of Streptomyces sp. M3004, grown in 10 mM and 30 mM H2O2, were significantly lower than the control cultures. On the other hand, as an antioxidant enzyme, CAT activity in both H2O2 treatment conditions increased significantly compared with the control. These activity values in 10 mM and 30 mM H2O2 treatment on the 48th hour of incubation were 3.8- and 6.6-fold higher than the control, respectively. SOD activity decreased significantly with respect to paraquat concentration, which was added at the start of the incubation. CAT activities increased significantly in 1.0 mM and 3.0 mM paraquat treatments compared to control. As an indicative marker of membrane damage, LPO levels of the novel isolate Streptomyces sp. M3004 treated with H2O2, and paraquat stress conditions were significantly higher than the control. Nevertheless, compared with the 30 mM H2O2 in both treatment conditions, LPO levels in 10 mM H2O2 were significantly higher.
