The electrical properties (variation of capacitance and conductance with frequency) of a plant stem can be conveniently measured in vivo by time domain dielectric spectroscopy. In this technique a voltage step is applied to a stem. The resulting polarization current is sampled by a microprocessor and Fourier-transformed to yield these properties. Spectra were obtained for seven electrode separations along a Poinsettia stem. The inverse capacitance and conductance were plotted vs separation for 50 frequencies from .35 to 350 Hz. Least-square fits yielded the effective dielectric constant and conductivity of the stem over this frequency range. In this way electrode effects were eliminated. A similar procedure was carried out for Coleus. A log-log plot of dielectric constant vs frequency shows a two-stage linear decrease for both plants. The conductivity is primarily DC. The dielectric loss decreases smoothly with frequency for Coleus. These results are compared to those for bone and the inorganic material hollandite. The dielectric properties seem best described by a cooperative, many-body approach.  相似文献

The dielectric properties of the Tobacco Mosaic Virus (TMV) have been measured using time domain dielectric spectroscopy (TDDS) in the temperature range from 1 to 40 degrees C. A single dielectric dispersion is observed in the MHz range. The activation energy of the process is found to be in the range 1-2 kcal/mol. The experimental data could not be completely accounted for by current theoretical models, but evidence indicates that the dielectric loss arises from polarisation of charge on and around the virus.  相似文献

In the biopharmaceutical industry, mammalian and insect cells as well as plant cell cultures are gaining worldwide importance to produce biopharmaceuticals and as products themselves, for example in stem cell therapy. These highly sophisticated cell-based production processes need to be monitored and controlled to guarantee product quality and to satisfy GMP requirements. With the process analytical technology (PAT) initiative, requirements regarding process monitoring and control have changed and real-time in-line monitoring tools are now recommended. Dielectric spectroscopy (DS) can serve as a tool to satisfy some PAT requirements. DS has been used in the medical field for quite some time and it may allow real-time process monitoring of biological cell culture parameters. DS has the potential to enable process optimization, automation, cost reduction, and a more consistent product quality. Dielectric spectroscopy is reviewed here as a tool to monitor biochemical processes. Commercially available dielectric sensing systems are discussed. The potential of this technology is demonstrated through examples of current and potential future applications in research and industry for mammalian and insect cell culture.  相似文献

Results are presented from experimental studies of the structure of an ac surface discharge excited by a metal needle over a plane dielectric surface. A barrier corona discharge was ignited in atmospheric-pressure argon at frequencies of the applied sinusoidal voltage from 50 Hz to 30 kHz. In experiments, the area of a dielectric covered with the discharge plasma increased with applied voltage. The discharge structure in diffuse and streamer modes was recorded using a digital camera and a high-speed image tube operating in a frame mode. It is found that, in the positive and negative half-periods of the applied voltage, the structure of the surface discharge is substantially different. The statistical characteristics of the branching surface streamers in the positive and negative half-periods are determined as functions of the voltage frequency. The most intense lines in the emission spectrum of the barrier corona are determined for both half-periods. The correlation between the dynamics of the emission intensity and the dynamics of the discharge current and voltage is investigated.  相似文献

Summary and Conclusions  The present work aimed to characterize transdermal drug delivery systems of pinacidil monohydrate in vivo by monitoring the effect of the TDDS on blood pressure of methyl prednisolone acetate induced hypertensive rats. The blood pressure of rats was measured using a noninvasive rat BP instrument based on cuff tail technique. A significant fall in rat BP (P<.01) was observed in treatment of hypertensive rats with all the formulations, which was maintained for 48 hours. Interformulation comparison revealed that formulation B-4 was the most effective with 37.96% reduction in BP (160.33±4.96 vs 99.44±4.46 mmHg). It was concluded that a single patch application of pinacidil TDDS (B-4) can effectively control hypertension in rats for 2 days. The system holds promise for clinical studies. The classical cable equation, in which membrane conductance is considered constant, is modified by including the linearized effect of membrane potential on sodium and potassium ionic currents, as formulated in the Hodgkin-Huxley equations for the squid giant axon. The resulting partial differential equation is solved by numerical inversion of the Laplace transform of the voltage response to current and voltage inputs. The voltage response is computed for voltage step, current step, and current pulse inputs, and the effect of temperature on the response to a current step input is also calculated.The validity of the linearized approximation is examined by comparing the linearized response to a current step input with the solution of the nonlinear partial differential cable equation for various subthreshold current step inputs.All the computed responses for the squid giant axon show oscillatory behavior and depart significantly from what is predicted on the basis of the classical cable equation. The linearization procedure, coupled with numerical inversion of the Laplace transform, proves to be a convenient approach which predicts at least qualitatively the subthreshold behavior of the nonlinear system.  相似文献

Bupranolol is a promising candidate for transdermal drug delivery system (TDDS) development. The effect of permeation enhancers on the in vivo delivery and beta-blocking effect of reservoir type TDDS was studied in comparison with intravenous BPL in rabbits. The beta-blocking effect was quantified by measuring the inhibition of isoprenaline induced tachycardia in rabbits after BPL administration via transdermal and intravenous routes. The reservoir type TDDS containing a hydroxypropyl cellulose gel and polyethylene membrane was used as a control device. In comparison, the TDDS containing skin penetration enhancers, either 2-pyrrolidone or partially methylated beta cyclodextrin (PMbetaCD) were evaluated. The control device (no enhancer) produced about 52% inhibition of isoprenaline induced tachycardia at 2 h and the effect continued over 24 h application period, however, the devices with 2-pyrolidone or PMbetaCD produced about 85% inhibition of isoprenaline induced tachycardia at 3 h and the same effect continued over 24 h application period. Likewise, the AUC of these devices were significantly higher than that of control device. The intravenous bupranolol showed rapid decline in the pharmacodynamic effect with time indicating its rapid elimination. The in vivo delivery of bupranolol (as estimated by a mass balance study) from the devices made with pyrolidone or PMbetaCD was 3-fold higher than that of control. The results of this study strongly suggest that the penetration enhancers in the TDDS increased the in vivo delivery of BPL, thereby increased the beta-blocking activity of BPL by 50-60% higher than control, enabling the reduction of the TDDS patch size, accordingly.  相似文献

In previous work on the in vitro frog gastric mucosa it was found that step currents produced a transmucosal voltage response of about 100 seconds duration. The voltage response can be formally represented by five resistors in series, with four of the resistors shunted by dielectric capacitors. It was shown that the two resistance-capacitor subcircuits with the longest time constants demand capacitors of the order of Farads cm−2 and that this portion of the transient can be explained on the basis of mechanisms involving the polarization of e.m.f.'s. In the present paper a conceptual unstirred-layer model is presented in which the transient voltage response to step currents is comparable to the observed response in the gastric mucosa. In both the model and in the gastric mucosa the voltage response for small currents is linear and bilateral and deviates from linear bilateral behavior at higher current densities. The implication of the unstirred-layer model is discussed in relation to other models previously proposed as explanations for the long-time constant transient.  相似文献

Studies were carried out on extraction and retrieval of potassium from water hyacinth (Eichhornia crassipes). The stem and leaf were subjected to 13 treatments. The highest rate of K removal following HCl treatment was 69.7% K. Most effective removal of suspended organic substances, Ca2+ and Mg2+ were achieved at pH approximately 13, when 88.0% of K remained in filtrate. Maximum K in precipitate following this step was achieved with tartaric acid additions at n(C4H6O6)/n(K+) of 1.72 when precipitating at 4 degrees C for 3h, which resulted in 72.3% of K removal from the solution. Over the entire process, 44.3% of K in the dried stem-leaf sample of water hyacinth was retrieved in the form of KC4H5O6. This process demonstrated the potential for use of water hyacinth as a resource of potassium to produce potassium salts and provide a valuable end use for the plant, which could be highly invasive in aquatic ecosystems.  相似文献

Summary We report here a kinetic study of the mechanism by which the weak acid TTFB (4,5,6,7-tetrachloro-2-trifluoromethylbenzimidazole) transports protons across phospholipid bilayer membranes. A previous kinetic study of the homologous dichloro compound, DTFB, revealed that the rate limiting step for proton translocation was the back diffusion of the neutral, HA, form of the weak acid; we conclude here that this is also the rate limiting step for proton translocation with TTFB. At high concentrations of either DTFB or TTFB the charged permeant species is an HA 2 complex. The kinetic analysis and independent measurements reveal that the permeability of the membrane to HA and adsorption coefficients of A and HA are an order of magnitude higher for TTFB than for DTFB. When either DTFB or TTFB was present in a solution where the pH was less than the pK of the weak acid, an unusual relaxation in the current was noted on application of a voltage step. The amplitude of the relaxation decreased as the voltage was increased. This relaxation is possibly due to a reorientation of the benzimidazole molecules at the membrane-solution interface. We also report experiments performed with DTFB on mitochondria. It was possible to reconcile these results with the bilayer data and, therefore, with the chemiosmotic hypothesis by postulating that the dielectric constant of the mitochondrial membrane is greater than that of a bilayer formed with decane as a solvent. To demonstrate the effect of dielectric constant on permeability, we replaced decane by 1-chlorodecane. This increased the capacitance of the artificial bilayer by a factor of two and the permeability of the bilayer to the A form of DTFB by two orders of magnitude.  相似文献

Non-invasive and fully automated conductimetric measurements of electrolyte and bacterial samples were achieved in a closed volume test cell, comprising a magnetic field coil and detector. By monitoring field induced currents in sample electrolytes the magnitude of the sample current was shown to vary as the inverse of the sample impedance. The impedance characteristic was shown to be that of an LCR resonant circuit. This characteristic is primarily a function of the applied frequency and the solution/cell properties being dependent on the solution conductivity and dielectric permittivity at any given concentration. Small changes in sample dielectric permittivity in the presence of a large background conductivity are shown to be significant.The apparatus described can provide fixed or swept frequency conductivity measurements in the range 1 kHz to 2.25 MHz with a lower conductivity sensitivity of 0.9 × 10−3 Scm−1. Bulk impedimetric characteristics of cell suspensions are derived by a two stage measurement.  相似文献

The application of nanotechnology in medicine, known as nanomedicine, has introduced a plethora of nanoparticles of variable chemistry and design considerations for cancer diagnosis and treatment. One of the most important field is the design and development of pharmaceutical drugs, based on targeted drug delivery system (TDDS). Being inspired by physio-chemical properties of nanoparticles, TDDS are designed to safely reach their targets and specifically release their cargo at the site of disease for enhanced therapeutic effects, thereby increasing the drug tissue bioavailability. Nanoparticles have the advantage of targeting cancer by simply being accumulated and entrapped in cancer cells. However, even after rapid growth of nanotechnology in nanomedicine, designing an effective targeted drug delivery system is still a challenging task. In this review, we reveal the recent advances in drug delivery approach with a particular focus on gold nanoparticles. We seek to expound on how these nanomaterials communicate in the complex environment to reach the target site, and how to design the effective TDDS for complex environments and simultaneously monitor the toxicity on the basis of designing such delivery complexes. Hence, this review will shed light on the research, opportunities and challenges for engineering nanomaterials with cancer biology and medicine to develop effective TDDS for treatment of cancer.  相似文献

This paper deals with the development and the parameter identification of an anaerobic digestion process model. A two-step (acidogenesis-methanization) mass-balance model has been considered. The model incorporates electrochemical equilibria in order to include the alkalinity, which has to play a central role in the related monitoring and control strategy of a treatment plant. The identification is based on a set of dynamical experiments designed to cover a wide spectrum of operating conditions that are likely to take place in the practical operation of the plant. A step by step identification procedure to estimate the model parameters is presented. The results of 70 days of experiments in a 1-m(3) fermenter are then used to validate the model.  相似文献

In the biopharmaceutical industry, adherent growing stem cell cultures gain worldwide importance as cell products. The cultivation process of these cells, such as in stirred tank reactors or in fixed bed reactors, is highly sophisticated. Cultivations need to be monitored and controlled to guarantee product quality and to satisfy GMP requirements. With the process analytical technology (PAT) initiative, requirements regarding process monitoring and control have changed and real-time on-line monitoring tools are recommended. A tool meeting the new requirements may be the dielectric spectroscopy for online viable cell mass determination by measurement of the permittivity. To establish these tools, proper offline methods for data correlation are required. The cell number determination of adherent cells on microcarrier is difficult, as it requires cell detachment from the carrier, which highly increases the statistical error. As an offline method, a fluorescence assay based on SYBR®GreenI was developed allowing fast and easy total cell concentration determination without the need to detach the cells from the carrier. The assay is suitable for glass carriers used in stirred tank reactor systems or in fixed bed systems, may be suitable for different cell lines and can be applied to high sample numbers easily. The linear dependency of permittivity to cell concentration of suspended stem cells with the dielectric spectroscopy is shown for even very small cell concentrations. With this offline-method, a correlation of the cell concentration grown on carrier to the permittivity data measured by the dielectric spectroscopy was done successfully.  相似文献

ABSTRACT

Ground current commonly referred to as “stray voltage” has been an issue on dairy farms since electricity was first brought to rural America. Equipment that generates high-frequency voltage transients on electrical wires combined with a multigrounded (electrical distribution) system and inadequate neutral returns all contribute to ground current. Despite decades of problems, we are no closer to resolving this issue, in part, due to three misconceptions that are addressed in this study. Misconception 1. The current standard of 1 V at cow contact is adequate to protect dairy cows; Misconception 2. Frequencies higher than 60 Hz do not need to be considered; and Misconception 3. All sources of ground current originate on the farm that has a ground current problem. This case study of a Wisconsin dairy farm documents, 1. how to establish permanent monitoring of ground current (step potential) on a dairy farm; 2. how to determine and remediate both on-farm and off-farm sources contributing to step potential; 3. which step-potential metrics relate to cow comfort and milk production; and 4. how these metrics relate to established standards. On-farm sources include lighting, variable speed frequency drives on motors, radio frequency identification system and off-farm sources are due to a poor primary neutral return on the utility side of the distribution system. A step-potential threshold of 1 V root mean square (RMS) at 60 Hz is inadequate to protect dairy cows as decreases of a few mV peak–peak at higher frequencies increases milk production, reduces milking time and improves cow comfort.  相似文献

The dielectric properties of human lymphocyte suspensions were studied by time domain dielectric spectroscopy (TDDS). Nine populations of malignant and normal lymphocytes were investigated. Analysis of the dielectric parameters of cell structural parts were performed in the framework of Maxwell-Wagner mixture formula and the double-shell model of cell. The specific capacitance of the cell membranes was estimated by the Hanai-Asami-Koisumi formula. It was shown that the dielectric permittivity, capacitance and conductivity values of cell membranes are higher for normal lymphocytes than for the malignant ones. The difference of the same parameters for normal B- and T-cells is also discussed.  相似文献

骨形态发生蛋白(BMPs)是一类调节骨组织发育的生长因子。BMP-2是BMP家族中诱骨活性最强的。在骨组织工程研究和临床应用中需要大量的BMP-2。因此,研究出一种能够有效地大量生产BMP-2的方法是十分必要的。随着植物分子生物学的进展,转基因植物被用作一种生物反应器来生产目的蛋白。以gus作为报告基因,研究了重组人bmp-2基因在烟草中的表达。通过GUS活性检测、半定量PCR和Western blotting分析了根、茎、叶组织中基因表达的水平,结果显示融合蛋白在根和茎组织中表达量显著高于叶组织。由于根和茎组织中蛋白组成与叶组织相比相对简单,提示其更易于进行目的蛋白的纯化。  相似文献

The stability of bioreductive drug delivery systems (TDDS) was monitored at various pH values and in the presence of glutathione (GSH). Results suggest that steric hindrance due to conformational constraint in TDDS led to an increase in stability of TDDS toward nucleophilic degradation under aqueous conditions. Nanopore analysis is an emerging technique of structural biology which employs nanopores, such as the α‐hemolysin pore, as a biosensor. A voltage applied across a membrane containing a nanopore generates a current, which is partially blocked when a molecule interacts with the pore. The magnitude (I) and the duration (T) of the current blockade provide an event signature for that molecule. Two peptides, CY12(+)T1 and CY12(?)T1 with net charges + 2 and ? 2, respectively, were analysed using different applied voltages and all four possible orientations of the electrodes and pore. The four orientations were vestibule downstream (VD), vestibule upstream (VU), stem downstream (SD) and stem upstream (SU) where vestibule and stem refer to the side of the pore on which the peptide was placed and downstream and upstream refer to the application of a positive or negative electrophoretic force, respectively. For CY12(+)T1, the effect of voltage on the event duration was consistent with translocation in the VD and SD configurations, but only intercalation events were observed in the VU and SU configurations. For CY12(?)T1, translocations were only observed in the VD and VU configurations. The results are interpreted in terms of two energy barriers on either side of the lumen of the pore. The difference in height of the barriers determines the preferred direction of exit. Electroosmotic flow and current rectification due to the pore as well as the dipole moment and charge of the peptide also play significant roles. Thus, factors other than simple electrophoresis are important for determining the interaction of small peptides with the pore.
