With the successful development of methods for the isolation and purification of ethanol-insoluble cinnamic acid esters in mint it became possible to initiate kinetic, isotopic studies on purified, ‘insoluble’ derivatives of caffeic acid, ferulic acid and p-coumaric acid. Pulse-feeding experiments were conducted with 14CO2, phenylalanine-U-14C and cinnamic acid-3-14C. The ferulic acid derivative exhibited a significant turnover as compared to the other insoluble derivatives which were relatively stable. Time-course tracer studies were performed to compare the turnover of soluble caffeic acid derivatives with ‘insoluble’ forms of caffeic acid. Caffeic acid associated with a macromolecular fraction consistently showed a higher specific activity than either soluble caffeic acid or the caffeic acid associated with a second insoluble derivative.  相似文献

Rat anterior hemipituitaries incubated in vitro rapidly take up and incorporate into protein D-[6-3H]-glucosamine · HCl, D-[1-14C]mannose and L-[G-3H]fucose. The newly labeled protein was only slowly released into a Krebs-Ringer bicarbonate incubation medium. Glucosamine- or mannose-labeled protein was barely detectable in the medium after a 30–60 min incubation whereas about 4% of all fucose-labeled protein had already been released into the incubation medium by 30 min. Puromycin · 2HCl (1 mM) inhibited incorporation of glucosamine or mannose into protein to 40% or less of control values within 30 min; fucose incorporation was not significantly inhibited before 45 min. Acid hydrolysis followed by amino acid analysis of glucosamine-labeled protein yielded significant amounts of label in glucosamine, galactosamine and apparent glucosamine-degradation products but no significant amount of label in any amino acid.  相似文献

Blood from ducks parasitized with Plasmodium lophurae and normal duck blood were incubated with sodium 14C-1-acetate. After release of the parasites from infected red blood cells (RBC) and concurrent treatment of normal blood, lipids were extracted from cellular material and plasma and lipid classes separated by thin-layer chromatography. Specific activity (dpm/mg lipid) of lipid classes was measured quantitatively by liquid scintillation radioassay and gravimetric analysis. The data indicated that the parasite within the RBC incorporated 14C-labeled lipid precursors.Experiments employing sodium 14C-1-acetate in two concentrations, 50 μCi 14C in 0.91 μmole sodium acetate/50 ml blood and 500 μCi 14C in 9.1 μmole sodium acetate/50 ml blood (1.82 × 10?5M and 1.82 × 10?4M), showed higher 14C incorporation into parasitized blood than normal blood preparations at the higher substrate concentration at 5 hr of incubation. At 1.82 × 10?5 M 14C-1-acetate, the highest specific activity in P. lophurae was associated with lipid alcohols. Monoglycerides and diglycerides were significantly labeled. At the higher acetate concentration (1.82 × 10?4M), monoglyceride and diglyceride lipid classes had the highest specific activity in preparations of partially purified P. lophurae.Lipids of plasma from parasitized blood incubated for 5 hr with both concentrations of labeled acetate exhibited the highest specific activity in the free fatty acid class and sterols.At 24 hr of incubation, the lipids of partially purified P. lophurae had increased specific activity in free fatty acids, diglycerides, monoglycerides, phospholipids, and triglycerides.In plasma from parasitized blood incubated 24 hr with 14C-1-acetate, the highest specific activity and greatest percent of 14C incorporation was found in free fatty acids.  相似文献

Developing ovaries from pharate adults of the silkworm, Bombyx mori, were incubated in a medium containing 14C-trehalose or 14C-glucose, and the effects of diapause hormone on the incorporation of these isotopes into ovary glycogen were studied. The rates of incorporation of 14C-trehalose remained unchanged in ovaries incubated for 36 hr when the medium was renewed at intervals of 12 hr, and showed saturation kinetics against the concentration of the sugar in the medium, giving apparent Km values of 6·0 mM for trehalose.There was no difference in 14C-glucose incorporation by ovaries grown in the presence (+SG) and absence (?SG) of the suboesophageal ganglion (SG). However, when 14C-trehalose was used as a substrate for glycogen synthesis, there was a marked difference in the incorporation between them, i.e. the incorporation was more than 60 per cent higher in +SG ovaries than that in ?SG ovaries. Increased incorporation of 14C-trehalose was also observed in ovaries from SG-removed pharate adults which received an injection of diapause hormone preparations. Maximum stimulation rates (about twofold) appeared 36 hr after the injection. Further, comparable effects on 14C-trehalose incorporation were observed in ovaries which were incubated with diapause hormone preparations added in vitro.These data are discussed in relation to the hormonal regulation of trehalase activity in ovaries.  相似文献

The fat body of Periplaneta americana incorporates labelled leucine into the protein during the period of oöcyte formation. The protein isolated from the ovary shows radioactivity only in the 20 hr treated animals; in the 1 hr treated animals the 14C activity is below background levels. The protein contents of the fat body and ovary were measured during the reproductive stages. The measurements indicate that the fat body may not store the protein which it synthesizes during reproduction.  相似文献

Divinyl-132,173-cyclopheophorbide-a enol was in vivo produced as a metabolite of divinyl-chlorophyll-a by protists and in vitro prepared by the intramolecular cyclization of methyl divinyl-pyropheophorbide-a, one of the divinyl-chlorophyll-a derivatives. The 1H NMR spectra in CDCl3 showed that the obtained product took exclusively its enol form in the solution. The intramolecular cyclization of chlorin π-system at the C132 and C173 positions affected the optical properties of such chlorophyll derivatives including the non-fluorescent emission of the enol.  相似文献

Quipazine (2-(1-piperazinyl)quinoline maleate), an agent with anti-tremorine and serotonin-like activity, was found to inhibit the uptake of 3H-dopamine and 3H-serotonin into rat striatal tissue in vitro. Quipazine was shown to be three times more effective as an inhibitor of serotonin uptake than dopamine uptake, the IC50's being 2.98 × 10?5M and 1.00 × 10?4M, respectively. These data suggest that quipazine exerts serotonergic and dopaminergic effects in the central nervous system.  相似文献

The relative rates of biosynthesis of cardenolide and sapogenin steroids of Digitalis purpurea were estimated by their uptake of 14CO2. The incorporation of label into both groups, although initially slow, indicated that biosynthesis occurs even at the end of the growing season. The sapogenins were produced more rapidly than the cardenolides at this stage of plant development. Within the group of sapogenins, digitogenin, the trihydroxy compound, was produced at a greater rate than the dihydroxy steroid gitogenin. In the case of the cardenolides, the trihydroxy gitoxigenin was produced at a slower rate than the dihydroxy digitoxigenin.  相似文献

Incorporation of leucine and changes in different protein fractions have been studied during Sorghum grain development. Most of the label from the injected leucine-[14C] was found in glutelin and residue fraction towards later stages of maturity. The label in albumin, globulin and prolamin decreased with a concomitant increase in label in glutelin and residue proteins. The concentration of lysine, aspartic acid and glycine decreased while that of leucine, proline, alanine, tyrosine, phenylalanine, and cystine increased during grain development. Increase in serine, methionine, valine and isoleucine was only marginal. The proportion of glutamic acid was high at all stages of grain development. Glutelin fraction resolved into two peaks on gel chromatography, only one of which with higher MW was labelled, while in albumin both the peaks were found to be labelled. After U14C leucine or U14C valine injections into haemolymph of adult female Glossina morsitans during late pregnancy, radioactivity was detected in the post-parturient female and its larval offspring in the injected material, lipids, and a range of non-essential amino acids. The level of radioactivity recorded from the third instar larva was higher than that remaining in the injected adult, and the activity was higher in amino acids than in the lipid fraction. Radiometric analysis of oöcyte and intra-uterine progeny 24 hr after haemocoelic administration to females of labelled leucine or valine revealed a pattern of radioactivity coincident with growth characteristics of these young stages. Rate of leucine uptake by the in utero third instar larva was slightly higher than that of valine, and this instar continues feeding even only a few hours before parturition. For both labelled materials, expired carbon dioxide and excreta from remales in early pregnancy showed significantly higher radioactivity than those in late pregnancy. Uric acid is the main nigrogenous waste of leucine and valine metabolism, though small amounts of these amino acids are also lost during excretion, with valine elimination being higher than leucine.  相似文献

Incubation in vitro of adult Brugia pahangi in an apparatus which permitted the separate exposure of the anterior, middle, or posterior region of the worms to medium-containing radioactively labeled d-glucose, l-leucine, and adenosine has provided evidence that these materials are taken up in physiologically significant amounts by a transcuticular route. No evidence for an oral ingestion of materials has been obtained from worms in vitro, but in vivo an oral uptake of Trypan blue has been demonstrated. The ultrastructure and cytochemical staining reactions for nonspecific esterase, acid phosphatase (EC-3.1.3.2), and leucine naphthylamidase of the gut and body wall are described.  相似文献
