Effects of different iron concentrations (final concentrations of iron in Artari's medium: 3.7, 37.0, 74.0, and 185.0mmol·L-1) on growth rate and contents of protein, triacylglycerides, and β-carotene in Dunaliella viridis cells at cultivation in different months were investigated. It was shown that the dose-dependent effects of iron were notable for season variability. In the 1stexperimental series (October, 2007), iron at researched concentrations did not affect growth rate of culture and protein, triacylglyceride, and [3-carotene contents in cells. In experimental series conducted respectively in Novem-ber 2007, December 2007, and February 2008, the dose-dependent stimulation of microalgae growth was observed. For each of these experimental series, there were particular dose dependences of protein, triacylglyceride, and β-carotene contents in microalgae cells at cultivation on media with iron at different concentrations. Meanwhile, for all of the four experimental series conducted in different months, variability of growth rate and analyzed parameters of microalgae Dunaliella viridis as control (cultivation without iron) was shown. It is suggested that these functional differences of control cultures of microalgae in different months caused variability in the dose-dependent effects of iron in a Dunaliella viridis culture. The possibility of iron usage for increasing microalgae biomass and for enriching it by β-carotene in Dunaliella viridis culture with initial low productivity and low β-carotene content is considered.  相似文献

Cell carbon and nitrogen in D. viridis are strongly dependent on the culturing conditions. Both elements increase with increasing salinity. At 31°C cell carbon is maximum and cell nitrogen minimum. This temperature was described previously (Jiménez, C., Niell, F. X. & Fernandez, J. A. (1990). Hydrobiologia, 197, 165-72) as the optimal one for achieving the maximum oxygen evolution. These results point out a possible competence for the reducing power during carbon and nitrogen assimilation processes, and under conditions of high photosynthesis (carbon assimilation) there is a partial inhibition of nitrate reduction, making C:N ratio maximum under conditions of maximum net photosynthesis.The study of cell glycerol, nitrate, structural proteins and free amino acids indicates that all of these solutes accumulate in the cells as a result of the high salinity adaptation.  相似文献

Betatene Ltd has established an extensive system for cultivation of D. salina for β-carotene production at Whyalla in South Australia.The operation, believed to be the largest area under man-made algal cultivation in the world, covers over 300 hectares in area, divided into three major cultivation lakes. A necessary requirement of the extensive system is to have a cost-effective harvesting system to handle the relatively large culture volumes. The extraction plant at the lakeside is designed to handle up to one million litre per hour in four parallel modules using a proprietary purely physical harvesting system.Current products are a crystalline β-carotene preparation for food colour applications, a high solubility preparation dispersed in soya bean oil for dietary supplementation and a dried powdered algal extract as an animal feed ingredient.  相似文献

The study evaluated the effect of dietary doses of Euglena viridis on the immune response and disease resistance of Labeo rohita fingerlings against infection with the bacterial pathogen Aeromonas hydrophila. L. rohita fingerlings were fed with diet containing 0 (Control), 0.1 g, 0.5 g, 1.0 g Euglena powder kg−1 dry diet for 90 days. Biochemical (serum total protein, albumin, globulin, albumin:globulin ratio), haematological (WBC, RBC, haemoglobin content) and immunological (superoxide anion production, lysozyme, serum bactericidal activity) parameters of fish were examined after 30, 60 and 90 days of feeding. Fish were challenged with A. hydrophila 90 days post-feeding and mortalities were recorded over 10 days post-infection. The results demonstrate that fish fed with Euglena showed increased levels of superoxide anion production, lysozyme, serum bactericidal activity, serum protein and albumin (P < 0.05) compared with the control group. Following challenge with A. hydrophila less survivability was observed in the control group (56.65%) than the group fed the experimental diets. The group fed 0.5 g Euglena kg−1 dry diet showed the highest percentage survival (75%). These results indicate that Euglena stimulates the immunity and makes L. rohita more resistant to A. hydrophila infection.  相似文献

Preillumination of intact cells of the eukaryotic, halotolerant, cell-wall-less green alga Dunaliella salina induces a dark ATPase activity the magnitude of which is about 3–5-fold higher than the ATPase activity observed in dark-adapted cells. The light-induced activity arises from the activation and stabilization in vivo of chloroplast coupling factor 1 (CF1). This activity, 150–300 μmol ATP hydrolyzed/mg Chl per h, rapidly decays (with a half-time of about 6 min at room temperature) in intact cells but only slowly decays (with a half-time of about 45 min at room temperature) if the cells are lysed by osmotic shock immediately after illumination. The activated form of the ATPase in lysed cells is inhibited if the membranes are treated with ferri- but not ferrocyanide, suggesting that the stabilization of the activated form of CF1 is due to the reduction of the enzyme in vivo in the light.  相似文献

Photosynthetic characteristics of Dunaliella salina with high (red form) and low β-carotene (green form) concentrations were studied. D. salina growing in brine saltworks exhibited a high level of β-carotene (15 pg cell−1). The rate of oxygen evolution as a function of irradiance was higher in the red than in the green form (on chlorophyll basis). Photosynthetic inhibition of the green form was observed above 500 μmol m−2 s−1. The red form appeared more resistant to high irradiance and no inhibition in O2 evolution was observed up 2000 μmol m−2 s−1. However, when these results are expressed on a cell number basis the rate of oxygen evolution was significantly higher in the green form. Carbonic anhydrase (CA) activity (total, soluble, membrane bound) was found in red and green forms. CA was higher in the red form on a chlorophyll basis, but lower if expressed on a protein basis. The light dependent rate of oxygen evolution and photoinhibition depends on the concentration of β-carotene in D. salina cells.  相似文献

β-Mannanase catalyzes endo-wise hydrolysis of the backbone of mannan and heteromannan, which are abundant in the cell wall structure of ungerminated leguminous seeds. The mature β-mannanase originated from Bacillus subtilis was expressed in Pichia pastoris, a methylotrophic yeast, using the leader peptide sequence of Saccharomyces cerevisiae α-factor. The cultivation of β-mannanase expressing Pichia pastoris yields up to 1.8 g/L protein. In the supernatant the activity of the 40 kDa—total mannanase attained a level of 1102.0 IU/mL. The properties of the β-mannanase were characterized. Optimum pH and temperature for the recombinant enzyme were 5.5 and 50°C respectively. Screening of Dunaliella was performed in four provinces along the coast of the Gulf of Thailand (Chanthaburi, Chon Buri, Chachoengsao and Samut Songkhram) and in Nakhorn Ratchasima province in the north-east. Six clones of D. salina were isolated from salt fields in Samut-Songkhram province. Clone DS91008 produced the highest carotenoid content of 80.3 pg cell-1, in modified J/1 medium at 30% NaCl under continuous illumination at 270 μmol m-2 s-1.  相似文献

Variations in the β-carotene, vitamin C and vitamin E content of D. tertiolecta have been shown to result from the nitrogen source used in the culture medium. Differences of 101%, 38% and 69% have been found in β-carotene, ascorbic acid and tocopherol content in mg/g of dry matter, respectively, and differences of 147%, 63% and 37% occurred in β-carotene, vitamin C and E concentrations in mg/litre of culture, respectively. Considering the β-carotene, vitamin C and vitamin E content in mg/g of chlorophyll a, maximum variations occurred in β-carotene content, with differences of 145% among the different nitrogen sources. Maximum β-carotene and vitamin C values were found in urea cultures, whereas urea cultures showed the minimum values for vitamin E.  相似文献

The wild type (Wt) and the polyamine-deficient strain (PAvs) of the halotolerant Dunaliella salina were subjected to stress caused by 3.5 mol/L NaCl concentration. The chloroplasts were isolated and the molecular aspects of their reaction to salt stress were studied together with their recovery response to these hyper-saline conditions.In the Wt, the photosynthetic complexes were found to be severely affected by salt stress under light conditions. Transglutaminases, which are present in chloroplasts as two units of 25 and 50 kDa, were immunorecognized by antibodies raised against rat prostatic gland transglutaminase. The amount, in particular that of the 50 kDa unit, underwent an immediate change following hyper-saline stress. These concentration changes were found to coincide with variations in enzymic activity, which is also affected by the presence or absence of light.The PAvs has a concentration of proteins and chlorophylls which is much lower than that of the Wt. In addition, the PAvs appeared to be more severely affected by both salt and subculture stresses. Its recovery time was also longer. Its TGase activity increased after salt stress and was always higher in the light than in the dark, except soon after subculture, showing an additive stress effect of salt and light. In the PAvs acclimated to high salinity, or immediately after stress application, the chloroplast content of chlorophyll a and b was considerably enhanced, like the TGase activity (by two-fold or more), and these changes exhibited almost coincident behaviours.Some transglutaminase substrates (proteins of 68, 55, 29 and 27 kDa) were found to be similar to those present in higher plants (thylakoid photosynthetic complexes and Rubisco). They were more markedly labelled by [1,4-14C] polyamines when the transglutaminase assay was performed in the light than in the dark, and much more in algae already acclimated to hyper-saline conditions than in those cultured in the optimal saline medium, or subjected to stress. The amount of 68 and 55 kDa polypeptides was particularly high in the 3.5 mol/L NaCl acclimated cells. The possible role of polyamine conjugation in the assembly of chloroplast proteins in cells affected by salt stress is discussed.  相似文献

The composition of an artificial medium and environmental conditions are described for the in vitro rearing of the egg parasite Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae). The medium was composed of defined amounts of protein, carbohydrates, lipid, salts, and vitamins, but also contained up to 40% insect hemolymph. The hemolymph was necessary to induce pupation. T. pretiosum eggs were obtained by dissection of Heliothis virescens (F.) (Lepidoptera: Noctuidae) eggs. In vitro reared T. pretiosum were similar in size to H. virescens reared T. pretiosum, and females were fecund.
Résumé Les oeufs de Trichogramma pretiosum ont été obtenus par dissection d'oeufs d'Heliothis virescens. T. pretiosum Riley (Hymenoptère, Trichogrammatidae) a été élevé avec succès sur un substrat synthétique. Outre des quantités définies de protéines, glucides, lipides, éléments minéraux et vitamines, la ration contenait aussi jusqu'à 40% d'hémolymphe de Manduca sexta. L'hémolymphe était nécessaire pour induire la nymphose. En plus de la nourriture, les conditions d'environnement sont apparues extrêmement importantes pour élever T. pretiosum dans des conditions satisfaisantes. Le contrôle de l'humidité relative, en particulier, était le facteur le plus important. Les adultes produits au cours de cette étude étaient d'apparence normale; ils se sont accouplés sans problèmes, les femelles étaient fécondes et leur taille ne différait pas de celle d'individus élevés sur H. virescens.

An efficient yeast gene expression system with GAL10 promoter that does not require galactose as an inducer was developed using Δgal80 mutant strain of Saccharomyces cerevisiae. We constructed several combinations of gal mutations (Δgal1, Δgal80, Δmig1, Δmig2, and Δgal6) of S. cerevisiae and tested for their effect on efficiency of recombinant protein production by GAL10 promoter using a lipase, Candida antarctica lipase B (CalB), as a reporter. While the use of Δgal1 mutant strain required the addition of a certain amount of galactose to the medium, Δgal80 mutant strain did not require galactose. Furthermore, it was found that the recombinant CalB could be produced more efficiently (1.6-fold at 5 L-scale fermentation) in Δgal80 mutant strain than in the Δgal1 mutant. The Δgal80 mutant strain showed glucose repressible mode of expression of GAL10 promoter. Using Δgal80 mutant strain of S. cerevisiae, CalB was efficiently produced in a glucose-only fermentation at volumes up to 500 L.  相似文献

We examined the assimilation of Cd, Cr, and Zn by the green mussel Perna viridis under complicated food conditions, including combinations of different compositions and concentrations of food (diatom and sediment), and variable food quantity and quality during particle digestion. At different combinations of food composition and quantity (5 mg l−1 and 20 mg−1, below and above the pseudofeces production), the Cd assimilation was significantly dependent on the food composition. The Cd assimilation efficiency (AE) decreased with increasing proportions of sediments in the diets, but its assimilation was not significantly affected by food concentration. In contrast, the assimilation of Cr and Zn decreased significantly with increasing food concentrations, whereas food composition did not significantly affect their AEs. Variations in metal gut passage time accounted partially for the difference in AEs among different combinations of food composition and quantity. By changing the type of particles during metal digestion, their AEs were maintained comparably at a low particle load (1 mg l−1), suggesting that variation of food quality during digestion did not affect metal assimilation. At a higher particle load (5 mg l−1), variation of food type during digestion affected the AEs of Cr and Zn. An increase in food concentrations from 1 to 15 mg l−1 during digestion resulted in a significant decrease in the AEs of Cr and Zn bound with either sediments or diatoms. Conversely, decreasing the food concentrations from 15 to 1 mg l−1 did not affect the AEs of metals, except for Zn bound with diatoms. Overall, our results highlighted the metal-specificity in their assimilation as influenced by complicated food environments, probably caused by different metal geochemical and biological behavior in the mussels. Feeding selectivity may have a greater control on the influx rate into the mussels than metal assimilation.  相似文献

The reaction of plasma membrane ATPase from yeast with Mg2+ and Mg · ATP was studied in a temperature range of 10 – 30°C. The random mechanism of activation by Mg2+ and the pseudocompetitive inhibition at higher concentrations was not altered when the temperature was varied, nor were the kinetic constants representing substrate binding. However, at low temperature, the affinity of the enzyme for Mg2+ is greatly reduced. The Arrhenius plot of log V vs. 1/T shows straight lines with an inflection point at 24°C, which disappears in the presence of detergent. Calorimetric studies of the plasma membranes show a transition point at the same temperature. From these findings we suppose that Mg2+ is bound at a regulatory site of the ATPase, which is influenced by the surrounding phospholipids.  相似文献

Summary Procedures for the serum-free culture of a density fractionated population of bovine bronchial epithelial cells have been established. Epithelial cells dispersed by protease digestion were fractionated by density equilibrium centrifugation, followed by plating of the small basal-like population on type I collagen-coated culture dishes. Two or three passages of 1:4 split enriched for a population of actively dividing cells, which could be stored in liquid nitrogen for subsequent use. Clonal growth assays revealed optimum proliferation using a 1:1 mixture of medium RPMI 1640 and LHC-9, a medium employed for human bronchial epithelial cells. Cellular growth rate, which was 0.6 to 1.3 doublings per day depending on the cell preparation, was conveniently decreased by supplementing LHC-9 medium with less than 50% RPMI. In contrast to airway epithelial cell cultures from other species, serum stimulated the growth of bovine bronchial epithelial cells in this system. Transforming growth factorβ1, however, inhibited growth and induced differentiation into a squamous phenotype. Also in contrast with other systems, the bovine cells were resistant to growth inhibition by 100 nM tetradecanoyl phorbol acetate or 1μM calcium ionophore A23187. Combination of phorbol ester with ionophore decreased mitotic activity, although induction of squamous morphology was not observed. Therefore, growth inhibition and squamous differentiation were not tightly coupled in this system. Finally, biologically synthesized matrix deposited by these cells stimulated growth rate. This culture system will therefore be useful in assessing the activities of both soluble and matrix-associated factors in the absence of serum.  相似文献

The different acid invertase activity (total, soluble, wall-bound and extracellular) in calli induced on explants (cotyledon, petiole, hypocotyl and leaf) originated from Medicago strasseri seedlings were evaluated. In cultures subjected to 16 h photoperiod, the highest total, soluble and extracellular activities were found in calli from leaves cultured in medium 12 (MS with 0.01 mg·dm−3 (0.045 μM) of TDZ), elevated amounts of total and wall-bound invertase being found in calli induced on petioles in 12G medium (MS with 0.01 mg·dm−3 (0.045 μM) TDZ and 3.104 mg·dm−3 glycerol). In cultures maintained in darkness, the activity detected was lower than that observed in cultures under light conditions. The highest amounts of enzyme was bound in calli cultured on medium 12 (total and extracellular invertase) -leaves- and medium 12D (MS with 0.001 mg·dm−3 (0.0045 μM) TDZ) (soluble invertase) -using hypocotyls. In general, the different forms of invertase activity studied seem to appear in greatest amounts in calli induced under light conditions using leaves as explant and TDZ as growth regulator.  相似文献

Morpho-physiological and biochemical responses of Arabidopsis thaliana (accession N1438) to bicarbonate-induced iron deficiency were investigated. Plants were grown in cabinet under controlled conditions, in a nutrient solution containing 5 μM Fe, added or not with 10 mM NaHCO3. After 30 days, bicarbonate-treated plants displayed significantly lower biomass, leaf number and leaf surface area as compared to control plants, and slight yellowing of their younger leaves was observed. Potassium (K+) content was not modified by bicarbonate treatment in roots, whereas it was significantly diminished in shoots. Their content in ferrous iron (Fe2+) and in leaf total chlorophylls was noticeably lower than in control plants. Root Fe(III)-chelate reductase and phosphoenolpyruvate carboxylase (PEPC) activities were significantly enhanced, but leaf ribulose 1.5-bisphosphate carboxylase (Rubisco) activity was decreased.  相似文献

We determined the effect of several concentrations of cadmium (0, 5, 10, and 20 μg/l) on cellular viability in the microalgae Scenedesmus sp. and Dunaliella viridis, by measuring growth at 0, 24, 48, 72, and 96 h and pigment production at 10 days. Algae were obtained from the Nonvascular Plant Laboratory collection, in the Facultad Experimental de Ciencias, Universidad del Zulia, Venezuela. Growth was measured by cellular counting, while pigment content was evaluated using conventional spectrophotometric techniques. Growth of both species decreased in the exposed cultures comparing with the control, but its behavior was similar, because in both control and exposed cultures, its was observed an adaptive phase in the first hours, as well as a growth phase after 72 h. Cadmium concentrations above 10 μg/l produced an adverse effect on pigment production, depending on the concentration and/or exhibition time. However, even though cadmium inhibited growth and pigment production, levels of both parameters indicated cellular viability, demonstrating the adaptability of the algae cultures when they were exposed to the metal.  相似文献
