A recently proposed hypothesis [Nick et al. (1990) Planta 181:162] suggests that, in maize coleoptiles, tropistic curvaturemight be caused by a stimulus-induced trans-organ gradient overthe orientation of cortical microtubules adjacent to the outercell wall of the outer epidermis. This gradient, in turn, iscontrolled by a light-induced redistribution of auxin. The hypothesiswas tested by following the behaviour of microtubules for variouslight stimuli using indirect immuno-fiuorescence in epidermalstrips as assay. Analysis of gravitropic straightening, nasticcurvature on the horizontal clinostat, effects of tonic irradiationwith red and/or blue light, and experiments involving opposinglight pulses demonstrate that bending direction and microtubuleorientation gradients are not as closely linked as predicted:Considerable bending can be produced without detectable gradientsof microtubule orientation, and conspicuous gradients of microtubuleorientation are not necessarily expressed as corresponding curvature.Thus, a monocausal relationship between microtubules and tropismis excluded. Furthermore, a comparison of tonic light effectson microtubules to earlier studies into the impact of lightupon auxin content indicate that the relationship between auxinand microtubules might be more complex than hitherto assumed.It is concluded that, at least in maize coleoptiles, growthcan be regulated by various mechanisms, and that microtubules,although somehow related to tropism, are probably not the causeof the fast tropistic responses. The tropical maize race Tuxpeño is a well-known race of Mexican dent germplasm which has greatly contributed to the development of tropical and subtropical maize gene pools. In order to investigate how it could be exploited in future maize improvement, a panel of maize germplasm accessions was assembled and characterized using genome-wide Single Nucleotide Polymorphism (SNP) markers. This panel included 321 core accessions of Tuxpeño race from the International Maize and Wheat Improvement Center (CIMMYT) germplasm bank collection, 94 CIMMYT maize lines (CMLs) and 54 U.S. Germplasm Enhancement of Maize (GEM) lines. The panel also included other diverse sources of reference germplasm: 14 U.S. maize landrace accessions, 4 temperate inbred lines from the U.S. and China, and 11 CIMMYT populations (a total of 498 entries with 795 plants). Clustering analyses (CA) based on Modified Rogers Distance (MRD) clearly partitioned all 498 entries into their corresponding groups. No sub clusters were observed within the Tuxpeño core set. Various breeding strategies for using the Tuxpeño core set, based on grouping of the studied germplasm and genetic distance among them, were discussed. In order to facilitate sampling diversity within the Tuxpeño core, a minicore subset of 64 Tuxpeño accessions (20% of its usual size) representing the diversity of the core set was developed, using an approach combining phenotypic and molecular data. Untapped diversity represents further use of the Tuxpeño landrace for maize improvement through the core and/or minicore subset available to the maize community.  相似文献

Redistribution of Surface Antigens– a General Property of Animal Cells?   总被引:7,自引:0,他引:7  
MEMBRANE antigens of animal cells are reported to be localized either in discontinuous discrete areas (H-21,2, HL-A3, organ4 and immunoglobulin (Ig)5–9 or continuously (species4 and blood group9,10). These results have been obtained by immunofluorescence (IFL) and electron microscopy performed by double layer principles except in the case of Ig where direct IFL technique has been used. The influence on the localization pattern of marker molecules by variables such as Ig concentration of the reagents used, effects of interactions between Ig molecules in the double layer and temperature dependence of the reaction steps have not been investigated. Redistribution of lymphocyte surface Ig molecules induced by anti-Ig antibody has been described and this was observed to be temperature dependent occurring at 20° C but not at 0° C11,12. Here I discuss the distribution and localization of different surface antigens (blood group substance A and B, species, H-2 and organ) on several cell types (monkey kidney, human lymphoid cells, HeLa, human thyroid cells, RK13 and L-cells of C3H/K) as recorded by direct and indirect IFL technique. I show that the temperature and Ig concentration used exert an influence on the distribution of these antigens. The cells used in IFL tests were cultured and harvested as described before10. IFL staining was performed according to Möller13. The stained cells were examined with a Leitz ‘Orthoplan’ ultraviolet microscope. Direct IFL technique was applied to two test systems. A FITC-conjugated rabbit antiserum to blood group substance A was used on monkey (Macaca fascicularis) kidney cells known to be A-positive10 and a FITC-conjugated horse anti-human thymocyte globulin were tested on a human lymphoid cell line Robinson (obtained from Dr Moore, Buffalo). In both test systems the staining pattern was completely confluent at all temperatures used: 4° C, 20° C and 37° C. The staining pattern was not related to antibody concentration.  相似文献

Background

In prokaryotes and some eukaryotes, genetic material can be transferred laterally among unrelated lineages and recombined into new host genomes, providing metabolic and physiological novelty. Although the process is usually framed in terms of gene sharing (e.g. lateral gene transfer, LGT), there is little reason to imagine that the units of transfer and recombination correspond to entire, intact genes. Proteins often consist of one or more spatially compact structural regions (domains) which may fold autonomously and which, singly or in combination, confer the protein''s specific functions. As LGT is frequent in strongly selective environments and natural selection is based on function, we hypothesized that domains might also serve as modules of genetic transfer, i.e. that regions of DNA that are transferred and recombined between lineages might encode intact structural domains of proteins.

Methodology/Principal Findings

We selected 1,462 orthologous gene sets representing 144 prokaryotic genomes, and applied a rigorous two-stage approach to identify recombination breakpoints within these sequences. Recombination breakpoints are very significantly over-represented in gene sets within which protein domain-encoding regions have been annotated. Within these gene sets, breakpoints significantly avoid the domain-encoding regions (domons), except where these regions constitute most of the sequence length. Recombination breakpoints that fall within longer domons are distributed uniformly at random, but those that fall within shorter domons may show a slight tendency to avoid the domon midpoint. As we find no evidence for differential selection against nucleotide substitutions following the recombination event, any bias against disruption of domains must be a consequence of the recombination event per se.

Conclusions/Significance

This is the first systematic study relating the units of LGT to structural features at the protein level. Many genes have been interrupted by recombination following inter-lineage genetic transfer, during which the regions within these genes that encode protein domains have not been preferentially preserved intact. Protein domains are units of function, but domons are not modules of transfer and recombination. Our results demonstrate that LGT can remodel even the most functionally conservative modules within genomes.  相似文献

Plant hormones are important biotic factors to regulate root growth. Among the seven kinds of plant hormones, auxin and gibberellin (GA) are strong accelerators of shoot growth, but these are not always accelerators for root growth. The classical views of root-growth regulation by auxin and gibberellin are summarized and current theory of the regulation mechanism is described in this review. The concentration-dependent deceleration of root growth is a key to understanding the auxin action on roots, since the endogenous concentration of indole-3-acetic acid (IAA) is inversely proportional to the growth rate. As massive IAA is transported from shoots to roots by polar transport, the influx speed of IAA mainly controls IAA levels in root cells. The classical view of IAA transport in roots has been supported by recent discoveries of IAA-carrier proteins such as AUX1, PINs and MDRs. The role of plasma membrane-located H+-ATPase and its regulation by IAA has also been described for the acid growth phenomenon caused by the acidification of root cell walls.

Compared to auxins, GA functions in roots are less remarkable. Nevertheless, GA also plays an indispensable role in the normal development of roots, since artificial GA-depletion causes abnormal expansion and suppression of root elongation. The GA-requirement for normal root growth was unveiled by the use of chemical inhibitors and mutants of GA biosynthesis. GA function that keeps root morphology long and slender is ascribed to the arrangement of cortical microtubules, cellulose microfibrils and unknown additional factor(s). Cross talks among plant hormones were recently found in the signal transduction pathways mainly in aerial organs. GA and IAA de-repress gene expression by degrading the gene-repressing proteins via the ubiquitin-mediated proteasome system. Another interaction of IAA and GA in growth regulation is the enhancement of GA1 level by IAA. Since the final biochemical steps of growth regulation take place in cell walls, possible cross talks are also conceivable in cell wall formation and modification.  相似文献

A theoretical model based on the water redistribution mechanism is proposed to predict the volumetric strain of motor cells in Mimosa pudica during the seismonastic movement. The model describes the water and ion movements following the opening of ion channels triggered by stimulation. The cellular strain is related to the angular velocity of the plant movement, and both their predictions are in good agreement with experimental data, thus validating the water redistribution mechanism. The results reveal that an increase in ion diffusivity across the cell membrane of <15-fold is sufficient to produce the observed seismonastic movement.  相似文献

The recent activity in designing, validating and implementing short-term tests for carcinogens has been spurred by the fairly convincing correlation between the carcinogenicity and mutagenicity of chemicals and by the assumption that mutations are somehow involved in neoplastic transformation. Moreover, it has been tacitly assumed that the mutagenic capacity alone of compounds would induce regulatory agencies to pass rules for their removal from man's environment, and would lead the public to avoid them. The actual response, however, is quite different. Government departments shy away from making any decisions on the basis of in vitro test systems, the public at large is becoming irritated by daily announcements that many of their cherished habits could adversely affect their health, and industries feel threatened and may reduce their search for new beneficial chemicals. The reluctance to accept wholeheartedly the mutagenicity tests for the detection of carcinogens is partly due to the uncertainty about the involvement of mutations in the formation of benign and malignant tumors. Following the initial rapid advances in the detection of environmental chemicals with carcinogenic and mutagenic properties, we seem to have arrived at the cross roads: we must now set new priorities for future research, and must make an unbiased assessment of the actual hazard of a compound to man and the human population.  相似文献

Given that surgical castration is a painful practice performed on millions of pigs every year, a need to identify novel reliable pain assessment tools exists in order to test anaesthetic and analgesic protocols that may reduce related pain. Two treatments were considered: handling (H) and surgical castration (C). Physiological (cortisol, lactate, glycaemia, rectal and eye temperature) and behavioural variables (latency to move after treatment and alterations in posture and walking) were analysed. Cortisol showed the greatest level in C piglets within 20 min after the surgical procedure and a positive correlation with glucose concentration. Eye temperature was higher in C piglets, and the same difference was detected for rectal temperature 3 h after castration. Behavioural parameters revealed that C piglets had longer latency to move and a higher percentage of them showed alterations in posture and walking. Results of this study showed that, in castrated piglets behavioural and physiological alterations occur mainly in the first 3 h from treatment. Latency to move, alterations in posture and walking, and eye temperature appear to give additional and useful information in piglet pain assessment. However, differently from the behavioural parameters considered, eye temperature involves several manipulations of the animals and a long process to acquire the data.  相似文献

Density-dependent habitat selection theory was mainly tested on active foragers and therefore its applicability to trap-building predators is poorly understood. The high sensitivity of trap-building predators to changes in their physical environment, combined with their limited movement capability, can lead to habitat specialization, reducing their utilization of alternative habitats. We studied density-dependent habitat selection in two pit-building antlions, differing in their habitat utilization spectrum. The habitat generalist, Myrmeleon hyalinus, inhabits and performs equally well in both sand- and loess-derived soils, although preferring the former, more productive coarse-grained soils. In contrast, the habitat specialist, Cueta lineosa, only inhabits fine-grained soils such as loess, while showing reduced foraging performance in coarse-grained sandy soils. We allowed larvae to select between these two soils, while manipulating con-specific density and initial stocking position. Irrespective of the initial stocking position, the number of M. hyalinus pits in the sand was positively correlated with the number of con-specific pits in the loess, implying that this species is a density-dependent habitat selector. Furthermore, these patterns of density-dependence were consistent with the expectations of ideal pre-emptive distribution (i.e., strong non-linearity in the distribution of antlions between soils with increased total density), suggesting that interference competition largely dictates habitat selection in this species. In contrast, the habitat specialist showed constant habitat selectivity, as neither con-specific density nor initial stocking positions influenced its habitat preference. Although mainly tested on active foragers, habitat selection theory can be applicable for trap-building predators, demonstrating how mechanisms operating at the individual level influence spatial distribution patterns.  相似文献

Sperm cryopreservation is a tool for the conservation of the genetic material of animals of genetic importance or for species preservation. In the case of domestic cats, this can be used to generate information about seminal harvest, evaluation and preservation, which is especially important due to its applicability to wild felids. This study evaluated seminal samples harvested by urethral catheterisation from 13 adult domestic cats. Samples were cryopreserved with experimental groups of extenders were defined by the penetrating cryoprotectant: 6% glycerol (GLY6%), 3% dimethylacetamide (DMA3%) and 3% dimethylformamide (DMF3%). The samples were thawed and evaluated by conventional microscopy and by computer-assisted sperm analysis (CASA). The structural and functional membrane integrity was assessed by supravital tests (EOS), hypoosmotic swelling tests (HOST) and flow cytometry (FC). There was a correlation (P < 0.05) between total motility and EOS (r = 0.54), HOST and FC (r = −0.62) and total motility and flow cytometry (r = 0.63), indicating that these are complementary parameters that increase the accuracy of the feline sperm quality evaluation post-thaw. The results regarding the structural and functional integrity of the sperm plasma membrane did not differ (P > 0.05) among groups. However, the DMA3% group had a lower (P < 0.05) percentage of morphological changes in the sperm tail compared to samples cryopreserved with GLY6% and DMF3%. Additionally, DMA3% provided lower values of immobile sperm post-thaw when compared to DMF3%. DMA is an interesting alternative to GLY and superior to DMF for the cryopreservation of feline semen at the studied concentrations.  相似文献

Experiments with isolated epidermal strips of maize coleoptiles, pretreated with auxin and further incubated on sucrose agar containing different concentrations of auxin (indole-3-acetic acid, IAA or naphthalene-1-acetic acid, NAA) and/or naphthylphthalamic acid (NPA), are described. Preincubation for 2h with 2 . 10?4M IAA or 10?5M NAA in buffer, followed by 30 min wash in buffer results in measurable cell elongation during a subsequent incubation for 6 h on sucrose agar. Addition of 10?4M NPA inhibited the response to auxin and this inhibition could be reversed by providing IAA in addition to NPA. Inner tissue fragments (without outer epidermis) did not respond to external IAA. These results lead to the conclusion that auxin secretion at the outer epidermis may be an essential step in auxin-regulated coleoptile growth.  相似文献

Models for population dynamics in rivers and streams have highlighted the importance of spatial and temporal variations for population persistence. We present a novel model that considers the longitudinal variation as introduced by the sinuosity of a meandering river where a main channel is laterally extended to point bars in bends. These regions offer different habitat conditions for aquatic populations and therefore may enhance population persistence. Our model is a nonstandard reaction–advection–diffusion model where the domain of definition consists of the real line (representing the main channel) with periodically added intervals (representing the point bars). We give an existence and uniqueness proof for solutions of the equations. We then study population persistence as the (in-) stability of the trivial solution and population spread as the minimal wave speed of traveling periodic waves. We conduct a sensitivity analysis to highlight the importance of each parameter on the model outcome. We find that sinuosity can enhance species persistence.  相似文献
