Following ovariectomy, adult female rats did not increase their food-reinforced bar pressing during 30-min tests, but they responded significantly more frequently for food than did control animals during 2- and 24-hr bar-press tests. Meal pattern data obtained during the 24-hr test demonstrate that ovariectomy increases meal size and decreases meal frequency, although the reduction in the number of meals did not fully compensate for the alteration in meal size. These findings suggest that ovariectomy does not increase the motivation to initiate a meal, but does result in the taking of a larger meal. The implication of these findings to body-weight set-point interpretations of ovarian obesity are briefly discussed.  相似文献

Pneumocyte type II produces a phospholipid, dipalmitoyl lecithin, which is stored in and secreted from the cell's inclusion bodies and is indispensable for alveolar stability. Cloned rat lung type II cells were harvested at monolayer confluence and homogenized in swelling buffer. After sequential differential centrifugations, the crude membrane fraction was subjected to discontinuous sucrose density gradient centrifugation at 65,000 × g. Quality of the relevant fractions was monitored by enzyme activities and phase contrast and electron microscopy of two major bands at densities 1.16 and 1.18, respectively. The less dense band contained only small quantities of organelles, little cytochrome c oxidase, and some glucose 6-phosphatase, but had a significant (Na+, K+)-ATPase activity; this and ultrastructural evidence certified the product as a suitable plasma membrane preparation. Upon sodium dodecyl sulfate-poly acrylamide gel electrophoresis, the protein pattern consisted of 11 major protein bands between 13,000 and 68,000 Mr ranges, and several minor ones. The lipid pattern was studied by two-dimensional thin layer chromatograpy, followed by various group reactions (e.g., amine, unsaturation, phosphorus, sugars). In the two major phospholipids, phosphatidyl choline and phosphatidyl ethanolamine, palmitic acid was the least abundant of four major fatty acids, accounting for 14.20% in phosphatidyl choline and 5.70% in phosphatidyl ethanolamine, whereas the most abundant were stearic and palmitoleic with about 28% each in phosphatidyl choline, and palmitoleic (29.90%) and oleic (23.05%) in the ethanolamine phosphatide. Apparently, the palmitic acid containing phosphatidyl choline must be in the lamellar inclusion bodies of type II cells and not in their plasma membranes.  相似文献

It is generally held that the decline in courtship which is seen at the beginning of incubation during the reproductive cycle of the male ring dove (Streptopelia risoria) is the result of changes in endocrine secretion rather than changes in response to the external situation. To test this hypothesis, male ring doves proceeding through a cycle with their mates were tested at selected intervals to determine whether there was a decline in courtship behavior in the course of a reproductive cycle. Four conditions were examined. Inexperienced and experienced males were tested with a stimulus female in two situations; once after the mate and nest were removed from the home cage, and once after the mate only was removed. In the former situation the male courted a stimulus female irrespective of the stage of the cycle he had reached with his mate. In the latter situation, as incubation with the mate progressed, males continued to incubate when the stimulus female was introduced. Thus, the male's display of courtship during the reproductive cycle is influenced by the external stimulus situation presented by the female and the nest.  相似文献

An improved method for the isolation and quantitation of bile acids from rat feces was developed. This method employs an initial Soxhlet extraction of the solid fecal material, esterification of the bile acid fraction with dry methanol/HCl and quantitation using a combination of tlc and glc techniques. In addition, identification of the individual components of the fecal bile acid fraction is accomplished by tlc and glc-ms. This method has proven useful for the quantitation and identification of the fecal bile acids during sterol metabolism measurements.  相似文献

Cultured L1210 lymphocytic leukemic cells resistant to cytosine arabinoside or Cytoxan were frozen under different conditions for up to 5 months and transplanted into recipient mice. Biochemical determinations including DNA, total RNA and drug resistance suggested that the more rapid, less expensive method of freezing mouse leukemic cells enables good retention of each parameter. Examination of cellular and subcellular RNA fingerprints indicated several alterations in the localizations f certain subspecies of RNA. Nonetheless, all cells retained their overall viability and the capacity to induce leukemia in CDF1 mice.  相似文献

N-alkylporphyrins are formed when certain agents such as 3,5-diethoxycarbonyl-2,4,6-trimethyl-1,4-dihydropyridine or ethylene interact with cytochrome P-450 in rats. It is likely that the iron protoporphyrin complex in cytochrome P-450 is first alkylated and then demetallated to form the free base N-alkylprotoporphyrins that are observed. An iron complex of N-methylprotoporphyrin IX dimethyl ester, chloro-N-methylprotoporphyrin IX dimethyl ester iron(II), shows the following properties: a double Soret band (λmax = 435 nm, with a shoulder at 390 nm) relatively facile reduction (E12 for Fe(III)/Fe(II) of 0.385 V vs Ag/AgCl in acetonitrile) and facile demetallation by acid or good nucleophiles such as thiophenol. A knowledge of such properties should be useful in determining the mechanism of formation of N-alkylprotoporphyrins in vivo.  相似文献

A superfusion model was used to study in vitro gastric inhibitory polypeptide (GIP) release from hamster small intestinal mucosa. A 10% glucose solution, in both fed and fasted hamsters, produced a prompt, sustained, three-fold rise in mean GIP release. In contrast, superfusion of a solution of 10% mannitol did not alter release of the peptide. This model provides potential for elucidation of the mechanisms through which glucose and other agents release GIP and other gastrointestinal peptides.  相似文献

Trypsin-activated protease activity was observed in the supernatants of conA-treated unfractionated or monocyte-depleted cultures at 1 h of incubation and trypsin-inhibitory activity was detected at 24 h. In contrast, supernatants from wheat germ agglutinin (WGA)-treated cultures or untreated cultures exhibited trypsin-inhibitory activity at 1 h and trypsinactivated protease activity after 24 h. The expression of proteases and protease inhibitors may be early events that occur during the activation of quiescent lymphocytes to a proliferative state.  相似文献

A rabbit antiserum has been prepared against the B95-8 transforming strain of EBV. The antiserum has a high virus neutralizing titer (approximately 1:1000) against both the marmoset B95-8 EBV and the human P3HR-1 EBV. The neutralizing antibodies may be absorbed completely with EBV producer cell lines, but not with nonproducer cell lines or producer cell lines treated with phosphonoacetic acid (PAA) so as to be nonproducer. After repeated absorption with PAA-treated B95-8, the serum remains reactive with the membranes of producer cell lines as judged by immunofluorescence or the 125I--Staphylococcal protein A radioimmunoassay. Thus the neutralizing antigens are expressed on the membranes of producer cell lines and may be purified from this source using the serum and 125I--Staph A binding as an assay. The ability of the serum to differentiate between producer and nonproducer cells by means of cell surface determinants has been exploited to achieve a separation of these two populations from the same culture. Immunoprecipitation by the protein A technique shows that the serum recognizes two polypeptides from producer cells of approximate molecular weights 150,000 and 75,000.  相似文献

Azo coupling reactions of N-α-acetylhistidine, N-α-acetyltyrosine, and N-α-acetyllysine with p-methylbenzenediazonium ion were investigated as model reactions to obtain information on the relative reactivity of the histidine, tyrosine, and lysine moieties of protein, separated from structural effects. The azo coupling yields of the amino acids increased as the pH of the reaction medium was increased, indicating that the ractive species are the imidazole anion of histidine, the phenolate anion of tyrosine, and the neutral ε-amino group of lysine. It was calculated, based on percentage yields of the azo products, that the imidazole anion is more reactive than the phenolate anion and the ε-amino group, respectively.  相似文献
