The pineal organ of masu salmon Oncorhynchus masou was maintained in a flow-through, whole-organ culture (superfusion) system and melatonin secretory profiles were determined at 15 °C under light-dark cycles of 12:12 h (LD 12:12) or the same in combination with constant darkness (DD) for 72 h. Under LD 12:12, superfused pineal organs showed a rhythmic melatonin secretion with high and low rates during the dark phase and the light phase, respectively. When the pineal organs maintained under LD 12:12 for 24 h were transferred to DD, melatonin secretion was consistently activated and no endogenous component was evident. When the pineal organs maintained under DD for 48 h were transferred to LD 12:12, melatonin secretion was reduced only during the light phase. These results indicate that melatonin secretion from the superfused pineal organ of masu salmon is regulated not by an intra-pineal circadian oscillator but by the environmental LD cycles, via local photoreceptors.  相似文献

The core temperature of the rainbow trout Oncorhynchus mykiss (3·5 kg) dropped to 1·0° C during the first 6 h of chilling at 0·5° C, remained stable until 24 h, and dropped significantly to 0·7° C after 39 h. Blood plasma osmolality increased and muscle moisture content decreased gradually with increasing chilling time. After 39 h of chilling, the rainbow trout experienced 40 mosmol l-1 higher blood plasma osmolality and 2·8% less muscle moisture content compared with initial values. In the Atlantic salmon Salmo salar (5·3 kg), core temperature dropped to 1·3° C and blood plasma osmolality increased significantly during the first 6 h of chilling at 0·5° C, but remained relatively stable throughout the rest of the experimental period. After 39 h of chilling, the salmon experienced 20 mosmol l-1 higher blood plasma osmolality and 0·5% less muscle moisture content compared with initial values. In rainbow trout muscle moisture content was inversely related to blood plasma osmolality indicating reduced seawater adaptation with increasing hours of chilling. No such relationship was observed in the Atlantic salmon. Hence, changes in plasma osmolality and muscle moisture in the Atlantic salmon do not indicate osmoregulatory failure since the new levels, once established, were maintained throughout the chilling time.  相似文献

Emergence pattern and developmental status at emergence of Baltic salmon fry from the Umeälven hatchery stock (63°50'N, 20°25'E) were studied at 6, 10 and 12°C in the laboratory. The number of days and degree days from hatching to 50% emergence decreased exponentially with increasing temperature. Synchronization of emergence increased with temperature. Optimal temperature for incubation of yolk-sac alevins was 10°C, which resulted in the largest fry at emergence and the lowest death rate. Fry kept at 6°C had the lowest mean weight and at 12°C the highest death rate. The fry emerged at an earlier developmental state with more yolk at 12°C than at 6°C. The Baltic salmon had a faster developmental rate during the gravel-phase, as compared to more southern Atlantic salmon populations.  相似文献

Returning adult salmon caught at the mouth of the River Dee, Aberdeenshire, were transferred to tanks in the laboratory. For fish placed in fresh water, sea lice remained attached for up to 6 days, though most lice were lost in the first 48 hours. Few lice were lost from salmon maintained in sea water. The experiments were conducted in water within a temperature range of 12·8 to 16° C, equivalent to summer river temperatures in the Aberdeenshire Dee.  相似文献

In order to compare the effects on reproductive performance of short-term or prolonged exposure to elevated temperatures during vitellogenesis, female Atlantic salmon Salmo salar were held at a water temperature of 22° C for periods of 4 or 6 weeks during the austral summer and autumn. Plasma levels of 17β-oestradiol (E2), testosterone (T) and vitellogenin (Vtg) were monitored and reproductive success was compared to that in groups of fish maintained at 14 or 22° C for 12 weeks from mid-January. Significant endocrine effects were observed within as few as 3 days of the commencement of exposure to 22° C, when plasma levels of E2 ( c. 0·5 ng ml−1) and Vtg ( c. 1·4 mg ml−1) were approximately half those observed in fish maintained at 14° C ( c. 1·0 ng ml−1 and 2·7 mg ml−1 respectively). The fertility and survival to the eyed stage of ova from fish held at 14° C exceeded 85 and 70% respectively, whereas ova from fish held at 22° C for 6 or 12 weeks exhibited significantly reduced fertility (<70 and <45% respectively) and survival ( c. 40 and 13% respectively). In spite of significant endocrine effects at all stages, a 4 week exposure to 22° C only generated significant reductions in egg fertility (<65%) and survival ( c. 30%) when it occurred between mid-February and mid-March. Together, these data confirm that high temperature spikes can affect reproductive success as strongly as more prolonged exposures, and indicate that there is a critical period of reproductive sensitivity to elevated temperature in late February and early March in this stock of Atlantic salmon.  相似文献

Schooling chum salmon Oncorhynchus keta were biased towards the water surface (median position <1 m) under isothermal conditions (10° C) in a water column simulator (WCS). Thermal stratification (24/10° C) inhibited upward movement with fish congregating at the thermocline and displaying a clear avoidance of potentially lethal surface waters. A tri-phase model based on piece-wise nonlinear regression was used to describe the distribution shifts of chum salmon during a change from isothermal to thermally stratified conditions. Fish distribution was consistent with thermoregulatory behaviour and exhibited 'attraction', 'preference' and 'avoidance' phases. The thermal preference of 50% of the fish lay between 12·2 and 20·2° C, however, >83·5% of the fish occupied a 'preferred' temperature range of 13·7–17·9° C. The mean temperature at which 50% of chum salmon avoided rising temperature by shifting deeper in the water column and using the cooler thermocline was 20·2° C, and 90% avoidance occurred at 22·9° C. Behavioural responses to thermal stratification were consistent amongst underyearling fish of differing size and age.  相似文献

Hatchery reared Atlantic salmon ( Salmo salar L.) parr were tagged with the standard I.C.E.S./I.C.N.A.F. salmon tag at water temperatures of 15°C and 6°C. They were then subjected to swimming trials in a tunnel respirometer. Following this treatment the 15°C fish developed necrotic infarcts around the tag insertions. Provided they did not become infected these lesions healed, but if the fish were retested 9 days later, the wound enlarged, became extremely inflamed, and the fish died soon after. The effect of sodium lactate and storage temperature on toxigenesis by proteolytic (Pr) and nonproteolytic (Np) Clostridium botulinum spores inoculated in processed 'sous-vide'-type beef, chicken breast and salmon was explored. Three g samples of beef and salmon homogenates with 0, 2.4 and 4.8% (w/w) lactate and of chicken with 0, 1.8 and 3.6% (w/w) lactate were placed in 24-well tissue culture plates. The samples were inoculated with 104 spores of pools of Pr (4A + 2B + 2F strains) or Np (4B + 4E strains), vacuum-packaged in barrier bags, and stored at 16 and 30°C for Pr and at 4, 8, 12 and 30°C for Np for up to 90 d. Lactate at 2.4% in beef and 1.8% in chicken delayed toxigenesis by Np for 40 d at 12°C and by Pr for 28 d at 16°C. Delaying toxigenesis for similar periods of time in salmon required 4.8% lactate and 12°C for Np, and 2.4% lactate and 16°C for Pr. Increasing levels of lactate and decreasing temperature significantly delayed toxigenesis of Cl. botulinum in the 'sous-vide' products.  相似文献

This study investigated behavioural thermoregulation by subyearling fall (autumn) Chinook salmon Oncorhynchus tshawytscha in a reservoir on the Snake River, Washington, U.S.A. During the summer, temperatures in the reservoir varied from 23° C on the surface to 11° C at 14 m depth. Subyearlings implanted with temperature-sensing radio transmitters were released at the surface at temperatures >20° C during three blocks of time in summer 2004. Vertical profiles were taken to measure temperature and depth use as the fish moved downstream over an average of 5·6–7·2 h and 6·0–13·8 km. The majority of the subyearlings maintained average body temperatures that differed from average vertical profile temperatures during most of the time they were tracked. The mean proportion of the time subyearlings tracked within the 16–20° C temperature range was larger than the proportion of time this range was available, which confirmed temperature selection opposed to random use. The subyearlings selected a depth and temperature combination that allowed them to increase their exposure to temperatures of 16–20° C when temperatures <16 and >20° C were available at lower and higher positions in the water column. A portion of the subyearlings that selected a temperature c. 17·0° C during the day, moved into warmer water at night coincident with an increase in downstream movement rate. Though subyearlings used temperatures outside of the 16–20° C range part of the time, behavioural thermoregulation probably reduced the effects of intermittent exposure to suboptimal temperatures. By doing so, it might enhance growth opportunity and life-history diversity in the population of subyearlings studied.  相似文献

Brown trout ( Salmo trutta ), Atlantic salmon ( Salmo salar ), brook trout ( Salvelinus fontinalis ) and lake trout ( Salvelinus namaycush ) fry entering the free-feeding stage, were tested for overhead cover preferences in stream channels at different temperatures and water velocities. Atlantic salmon showed strong preferences for overhead cover, brown trout moderate preferences, whereas lake trout had preferences only at high temperatures, i.e. 12.4–19.2°C. Brook trout showed no cover preferences. Temperature influenced cover preferences of Atlantic salmon and brown trout considerably. An examination of data on both water quality in the Thames Estuary and on the count of salmon. Salmo salar L . trapped in fresh water above the head of the tide in 1982–1989, was carried out to establish statistical correlations. The annual and monthly return of salmon as 1-sea-winter fish (grilse) in June to September was negatively correlated with water temperature, a nil catch being associated with a maximum value of 24.2° C, coupled with lower values maintained over substantial lengths of the estuary (e.g. 21.5° C over a distance of no more than 50 km). The annual return was negatively related to the extent to which the estuary was predicted to be lethal from the combination of low concentration of dissolved oxygen (DO) and high temperature, a reduction in DO of 1 mg 1 1 being equivalent to an increase in temperature of 4° C. The annual return for the whole year was directly related to the return in July to September. Depending upon the year, the monthly returns were related to both DO and temperature; they were reduced to a tenth at a 95 percentile DO of 2.7 mg 1 1, whilst the weekly catches were reduced to zero at 2.4 mg 1−1. Weekly catches increased with river flow and daily catches increased with both river flow and tidal height. The few mortalities observed in the estuary in July are generally related to the quality of the water as predicted from the combination of high temperature and low DO.  相似文献

Eggs and alevins from 21 families of pink salmon, Oncorhynchus gorbuscha , from five odd-year broodline stocks spawning in southern British Columbia were incubated under controlled water temperatures of 4° C, 8° C and 12° C. There were significant differences in egg survival among stocks and among families within stocks at all incubation temperatures, but the differences were greatest at 4° C. Alevin survival was at least 97% for each stock at each temperature. The most northern spawning stocks had higher egg survival at 4° C than did the others. Hatching time of the alevins and emergence time of the fry were similar for all five stocks. Alevins hatching at 8° C were longer than those hatching at 4°C or 12°C, but there were no stock differences in alevin length or tissue weight. Stocks with larger eggs produced alevins of greater total weight and more yolk. Emergent fry from Vancouver Island stocks had the greatest tissue weight at 12° C, but Fraser River fry were heaviest at 8° C. There were significant differences among families within stocks for alevin and fry size parameters, suggesting that family variation should be accounted for in studies of salmonid developmental biology.  相似文献

The incidence of histamine- or tyramine-producing lactic acid bacteria was examined in several products of vacuum-packed sugar-salted fish (salmon, halibut, mackerel). No histamine-producing isolates were observed, whereas the majority of tyramine-producing isolates were identified as Carnobacterium spp. These organisms were shown to be important members of the microbial flora during storage of vacuum-packed sugar-salted salmon at 5°C. The amount of tyramine produced was reduced by lowering the temperature from 9°C to 4°C for all of five strains of carnobacteria or lactobacilli. The majority of tyramine was produced during the exponential growth phase for Carnobacterium piscicola N 5 and Lactobacillus viridescens N 69. The ability of these bacteria to produce tyramine may be used as an index of microbial quality/acceptability of stored vacuum-packed sugar-salted fish.  相似文献

The capacity of tropical whitespotted bamboo sharks Chiloscyllium plagiosum to metabolically compensate, at both the whole‐animal and biochemical levels, to prolonged exposure to temperatures higher (30° C) and lower (20 and 15° C) than their native temperature (24·5° C) was examined. As expected, whitespotted bamboo shark oxygen consumption increased upon exposure to 30° C and decreased at 20 and 15° C. Initial changes in oxygen consumption were maintained even after months at the experimental temperature, indicating that whitespotted bamboo sharks did not compensate metabolically to the experimental temperatures. Maximal activities and thermal sensitivity of citrate synthase and lactate dehydrogenase from whitespotted bamboo shark white locomotor muscle were similar between control animals maintained at 24·5° C and those maintained at 15° C, indicating that cold‐exposed animals did not compensate at the biochemical level. Similarly, lactate dehydrogenase activity did not change following prolonged exposure to 30° C. White muscle from whitespotted bamboo sharks maintained at 30° C had significantly lower citrate synthase activity than did control animals. This result was surprising given the lack of metabolic compensation at the whole‐animal level. Overall, whole‐animal oxygen consumption measurements supported the hypothesis that animals from thermally stable environments lacked the capacity to metabolically compensate to altered temperatures. Enzymatic results, however, suggested that the metabolic potential of muscle could change following temperature acclimation even in the absence of metabolic compensation at the whole‐animal level.  相似文献

Hydrogen ion concentrations (pH) of the digestive tracts of channel catfish were determined for fish of two sizes (892 and 134 g average weight) at two environmental temperatures (28 and 23°C). Acidic conditions (pH 2–4) were present in the stomach contents of all catfish with slightly higher pH values in stomachs of fish at 28°C. The pH increased to slightly alkaline values (pH 7–9) in the duodenum and reached a maximum level (pH 8.6) in the upper intestinal region and then decreased in the lower segments to approach neutrality in the colon. The pH of the bile ranged from 6.1 to 7.5 and was higher in fish maintained at 28°C. Higher environmental temperatures (28°C) resulted in a slightly lower pH throughout the intestinal and colon segments. The larger catfish had lower intestinal length/body weight ratios than smaller catfish. As the result of distension due to increased food consumption, catfish maintained at 28°C had shorter intestinal tracts than catfish maintained at 23°C.  相似文献

We examined Hsc70 in gill, liver and caudal fin from coho salmon ( Oncorhynchus kisutch ) before (ambient, ~12 °C) and after a sublethal heat shock of 25 °C for 1 h. Increased levels were observed for at least 48 h in all three tissues. Attempts to demonstrate isoforms of this heat-shock protein were not successful using four different antibodies. However, one of these antibodies recognized isoforms in brine shrimp and oysters, two organisms in which the heat-shock response has been well characterized. Extracts of those organisms and coho salmon tissues were run on the same gels and evaluated on the same Western blots, at the same time. We believe that our results provide a reliable account of Hsc70 in these fish under these conditions. Limited experiments showed that induced thermotolerance was achieved in these coho salmon, lasting for about 1 week after the sublethal heat shock. These data suggest that increased levels of Hsc70 are correlated with induced thermotolerance in these fish.  相似文献

Male and virgin female Aedes taeniorhynchus were maintained on a sugar solution at constant temperatures, at split-temperatures, and at alternating temperatures from immediately after emergence until death in order to study the effect of temperatures on their longevity. Life spans were found to be temperature dependent at constant temperatures of 22, 27, and 32°C, but they were divided into ‘ageing’ and ‘dying’ phases at split-temperatures. The rate of ageing, which was independent of temperatures, was the same in males whether they were transferred from low to high or high to low temperatures. The rate of ageing was the same in females transferred from 22°C or 27 to 32°C, but much longer than expected when transferred from 22 to 27°C. Also, the rate of ageing was the same for females transferred from 32°C to either 22 or 27°C and from 27 to 22°C. The rate of dying was essentially temperature dependent in both males and females with slight temperature compensation occurring in some cases. Life spans were the same in males when alternated between 22?27°C, 27?32°C, and 22?32°C. In females they were same at 27?32°C and 22?32°C, but were much longer than expected at 22?27°C. It is concluded that the threshold theory is confirmed when mosquitoes are maintained at split-temperatures and at alternating temperatures in their optimum range of temperatures.  相似文献

In this study, effects of stock origin, fish size, water flow and temperature on time of river ascent of adult Atlantic salmon Salmo salar were tested. Brood stocks were collected in eight Norwegian rivers situated between 59 and 69° N. The fish were reared to smolts, individually tagged and released in the River Imsa, south-west Norway (59° N). Adults from all stocks approached the Norwegian coast concurrently, but Atlantic salmon ≥70 cm in natural tip length entered coastal water slightly earlier during summer than smaller fish. Atlantic salmon <70 cm, however, ascended the river significantly earlier and at lower water flow and higher water temperature than larger fish. Although largest in size, the fish from the northern populations (62–69° N) ascended the River Imsa almost 1 month earlier than those from the south (59–60° N). They seemed less restricted by the environmental factors than the fish originating from the more southern rivers. There was no apparent trend among years in time of river ascent. Maximum ascent per day occurred at water discharges between 12·5 and 15 m3 s−1 and at water temperatures between 10 and 12·5° C. There was a significant positive correlation between water flow and river ascent during the first part of the upstream run from July to September with best correlation for September, when multiple regression analysis indicated that water temperature had an additional positive effect. Stock origin, fish size and water discharge were important variables influencing the upstream migration of Atlantic salmon in small rivers.  相似文献
