Variability of taurine (2-aminoethane sulfonic acid) was studied as a function of size in the mussel Mytilus galloprovincialis and tissue specificity. Isometric and/or allometric relationships were established with regard to total soft mass of the mussels between 20 and 60 mm shell length. Relative amounts of taurine dropped significantly with increasing mass of whole soft tissues with an allometric coefficient value of -0.15. The inverse relationship between taurine and increasing size of mussels was confirmed for gill epithelium and labial palp (allometric coefficient values of -0.16 and -0.10, respectively), tissues that, in turn, represented isometric functions with regard to total soft mass. Although relative amounts of taurine were significantly different in digestive gland, mantle and foot, relationships with increasing size of mussels remained unchanged in these tissues. Gill area of mussels was related to soft mass with an allometric coefficient of 0.70 by 2D Image Analysis, but increased to 0.85 when introducing a third dimension, i.e. gill thickness. Results are discussed according to gill structure analysis and taurine functionality.  相似文献

Seasonal variations of six mussel (Mytilus galloprovincialis) biomarkers at two sites in the Mediterranean Sea were compared with physiological indices (condition, growth and gonad maturation), environmental parameters (temperature, salinity and turbidity), and chemical contamination levels. The basal levels of acetylcholinesterase (AChE), DNA adducts, benzo[a]pyrene hydroxylase (BPH), heat-shock proteins (HSP70), metallothioneins (MT) and P-glycoprotein (P-gp)-mediated multixenobiotic resistance (MXR) were estimated as early warning signals in caged mussels sampled at Carteau (native site) and La Fourcade (transplantation site) over a 2-year period. The Carteau and La Fourcade mussels have specific chemical contamination profiles but a similar range of values. For example, both are highly contaminated by heavy metals (201 and 258.4 mg kg(-1) dw, respectively) and considered as moderately impacted for polychlorinated biphenyls (PCBs) and polycyclic aromatic hydrocarbons (PAHs). However, contamination levels at Carteau are twice as high for PAHs (101.5 mg g(-1) dw) and PCBs (90.2 mg g(-1) dw) than La Fourcade. The seasonal contamination trend at Carteau showed six-fold higher levels of pyrolytic pollutants in winter. Although few tissue lesions were detected in individuals studied at either site, greater parasitic infestation was observed at Carteau. The results of findings from the two Mediterranean pilot studies support the adaptability of transplanted mussels to be used as biomarkers and to establish physiological endpoints for chemical contaminant exposure.  相似文献

The relationship between hemic neoplasia, a blood cell disorder in bivalve molluscs, and chemical contaminants was evaluated in the common mussel (Mytilus edulis complex). Hemic neoplasia (HN) is endemic to mussel populations in Puget Sound. The prevalence of hemic neoplasia ranged from 0 to 30% in mussels from nine sites in Puget Sound, Washington. Organic chemical contamination in sediment from these sites range from 0.1 to 64.0 ppm of polycyclic aromatic hydrocarbons (PAHs) and 0.07 to 0.50 ppm chlorinated hydrocarbons. No relationship between the body burden of environmental contaminants and the prevalence of HN in mussels was identified. To evaluate the short-term ability of chemical contaminants to induce HN in mussels, mussels, from a site where mussels were previously determined to be HN free, were fed microencapsulated PAHs (composed of a mixture of phenanthrene, flouranthene, and benzo[a]pyrene) or PCBs (Aroclor 1254) and the prevalence of HN was assessed after 30 days of exposure. Although an apparent increase in HN prevalence (20 to 30%) was observed in all treatments groups except the untreated controls, no significant difference in the prevalence of HN was observed between the control group of mussels fed corn oil (vehicle) and mussels fed either PAHs or PCBs in corn oil. A long-term (180-day) exposure study was conducted to evaluate the influence of PAHs or PCBs in modulating the prevalence of HN in a mussel population already exhibiting a moderate HN prevalence. Mussels, from a site where mussels were previously determined to exhibit a background prevalence of HN, fed microencapsulated PAHs, PCBs, and corn oil (vehicle) over a long time period (180 days), revealed an apparent increased prevalence of HN (30 to 40%) above the low levels (20%) initially present. However, no significant difference in the prevalence of HN was observed between the control group of mussels fed corn oil (vehicle) and mussels fed either PAHs or PCBs in corn oil. Although chemical contaminants have been proposed as a modulating factor in the development and promotion of HN in bivalve molluscs from environmentally stressed and degraded habitats, we find no evidence that chemical contaminants induce or promote the development of HN in the mussel M. edulis complex.  相似文献

Thirty ninhydrin-positive compounds were determined in the sera, amniotic fluid, stomach content and allantoic fluid from 29 bovine fetuses of various gestational ages. Fetal serum was found to contain about 3-fold higher concentrations of free amino acids (FAA) than maternal serum, and allantoic fluid contained about 3-fold higher concentration of FAA than fetal serum. Decreasing concentrations of FAA were found in serum as a function of the crown-rump length for the amino acids taurine, aspartic acid, threonine, serine, ornithine and lysine. Decreasing concentrations of FAA in allantoic fluid were found for threonine, alanine and ornithine, whereas increasing concentrations were found for phosphoserine and methionine as a function of the crown-rump length. Correlations were found between the concentrations of most amino acids in amniotic fluid and stomach content, but fewer correlations were found between the other fluid compartments. The feeding ecology of the green-lipped mussel, Perna canaliculus, was investigated within three intertidal mussel beds along Ninety Mile Beach, northern New Zealand, between August 2000 and March 2001. Adult mussels of different sizes (45-105 mm in shell length) were collected from the intertidal sites about 30 min after being submerged by the incoming tide for gut content analyses. Results of these analyses indicate that mussels consume a variety of phytoplankton, micro- and mesozooplankton, including mussel larvae and post-larvae. Cannibalism of juveniles of up to 620 μm was recorded for intertidal mussels, and conspecifics of up to 2.4 mm were found within the stomachs of additional mussels collected in August 2000 from a nearby subtidal site. For all three intertidal populations, mussel larvae and juveniles contribute about 70% of the food particle consumption during the spawning peak in August, while phytoplankton and other zooplankton constitute the majority of the food source (about 99%) in March, during gametogenesis. Larger intertidal mussels tended to have more food particles in their stomachs than smaller mussels within all three populations. Distinctive differences in food consumption among intertidal populations directly coincide with variations in total particulate matter (TPM), particulate organic matter (POM) and percent organic matter (OM) in the adjacent seawater.Separate experiments designed to test the feeding behavior of mussels feeding at different times during the incoming tide were conducted at one of the intertidal sites during August 2000 and March 2001. Results from these experiments indicate a marked shift in food consumption from bivalves to other mesozooplankton in August, and from phytoplankton to mesozooplankton in March. The observed combination of mussel predatory and grazing behavior over the incoming tide and through the year provides evidence for a strong food-web link between the benthic and pelagic life stages of this species. Furthermore, the high rate of cannibalism during some months of the year suggests that this source of food may significantly contribute to the energy budget of wild populations, with potential implications for evolutionary adaptive success.  相似文献

1. Methods for measuring concentrations of total and individual free amino acids (TFAA, FAA), other amino compounds and humic substances (HS) in media containing the following three treatments and the control are described: (i) the non-axenic aquatic macrophyte Ceratophyllum demersum alone; (ii) the pulmonate snail, Biomphalaria glabrata alone; (iii) the snail, plant and the epiphytic bacteria and algae together as a four-component modular system; (iv) control without organisms. 2. TFAA accumulated to give asymptotic values of 2.5 μm , 10 μm and 15 μm in treatments (i), (ii) and (iii), respectively, by the end of the 30-day incubation period. The mass-specific accumulation rate in the treatment with the snail alone [89.4 nmol g–1 (wet weight minus shell) day–1] was approximately ten times that with the plant alone [8.4 nmol g–1 (wet mass) day–1]. No FAA or HS could be detected at any time in (iv). 3. On the second day of incubation the concentrations of TFAA and of some individual amino acids were significantly higher in the treatment containing the snail and plant together than the sum of the concentrations in the treatments with the plant and snail alone, presumably due to an increase in the snails’ metabolic rate and ‘sloppy feeding’. 4. The differences in the relative abundance of amino compounds accumulating in media conditioned by snails alone (e.g. much larger proportions of ammonia, ethanolamine and phosphoserine than in plant-conditioned media) and plants alone (e.g. larger proportions of asparagine and glutamine than in snail-conditioned media) suggest that snails and plants may derive mutual benefits by exchanging amino compounds. 5. The accumulation patterns of the more recalcitrant HS differ from those of the amino acids in two respects. First, the HS concentrations continued to increase throughout the 30-day incubation period in all three treatments. Second, most of the HS in the module originates from the plant as both the concentrations and mass-specific accumulation rates were significantly higher in the treatment with the plant alone [2.6 mg l–1, 9.09 μg g–1 (wet mass) day–1, respectively] than in the treatment with the snail alone [0.75 mg l–1 and 7.7 μg g–1 (wet mass) day–1, respectively]. 6. The possible reasons for the differences in the accumulation patterns of FAA and HS in the three treatments are discussed. Evidence is also given in support of the testable hypothesis that the four components of the module derive several mutual benefits, including those arising from the release of dissolved organic matter, such as FAA and HS, by living organisms.  相似文献

1. Young chum salmon were fed on the basal and taurine-supplemented diets for 30 days in freshwater (FW) and for 25 days in seawater (SW). Levels of taurine, major free amino acids (FAA) and non-protein nitrogen (NPN) in various tissues were determined. 2. Tissue taurine levels were higher when fish were fed on the taurine-supplemented diets. All tissues of the SW fish did not contain higher taurine levels than those of the FW. 3. Levels of major FAA in the tissues differed little between fish fed on the basal and taurine-supplemented diets and also between the FW and SW fish. 4. No difference was observed in tissue NPN levels between fish ingesting the basal and taurine-supplemented diets; the levels were slightly higher in the SW fish.  相似文献

The biochemical composition (proteins, carbohydrates, glycogen, total lipids and lipid classes) of the mussel Mytilus galloprovincialis was investigated during an experimental culture using mussel seed from areas with different degree of exposure to the Prestige oil spill. The aim of the study was to identify alterations in the biochemical composition of mussel seed from natural populations commonly used in Galicia for mussel raft culture that might be linked to previous oil exposure. We have selected three mussel seed populations from Pindo, Miranda and Redes, that were characterised in a previous study according to the oil exposure three months after the spill. These populations were transplanted to a raft culture system in the Ría de Ares-Betanzos where our experimental culture followed standard commercial techniques from March 2003 to February 2004. Mussels from Pindo (characterised as the most affected area by the oil spill) showed marked differences in lipid composition with regard to other populations in the content of triacylglycerols, (P<0.001), free fatty acids (P<0.001) and phospholipids (P<0.05) at the onset of the culture. Although these differences in lipid composition might reflect their previous exposition to hydrocarbons, this pattern did not last longer most likely due to depuration of hydrocarbons stored in the tissues or by the development of certain tolerance to PAHs. These significant differences were not detected between Miranda (designed as hardly affected area) and Redes (designed as reference area) which may reflect that Miranda mussels were not affected or only hardly affected by the spill. With the exception of the onset of the culture, biochemical composition showed similar patterns in all mussel populations. Then, the fact of being cultured in a common environment seemed to be more responsible for the long-term variability in the energetic reserve than the origin of the populations or their previous biochemical status.  相似文献

The seasonal variability of the intracellular free amino acid (FAA) concentration was studied in 5 Macoma balthica populations and 7 Mytilus spp. populations along their European distribution. Because of the well known physiological role of FAA as organic osmolytes for salinity induced cell volume regulation in marine osmoconformers, FAA variations were compared in bivalve populations that were exposed to high vs. low intraannual salinity fluctuations. In general, seasonal FAA variations were more pronounced in M. balthica than in Mytilus spp. In both bivalve taxa from different locations in the Baltic Sea, highest FAA concentrations were found in autumn and winter and low FAA concentrations were measured in summer. Seasonal patterns were less pronounced in both taxa at locations with constant salinity conditions. In contrast to Baltic Sea populations, Atlantic and Mediterranean bivalves showed high FAA concentrations in summer and low values in winter, regardless of seasonal salinity fluctuations. Significant seasonal FAA variations at locations with constant salinity conditions showed that salinity appeared not to be the main factor in determining FAA concentrations. The seasonal patterns of the main FAA pool components, i.e. alanine, glycine and taurine, are discussed in the context of seasonal variations in environmental factors (salinity, temperature) and physiological state (glycogen content, reproductive stage).  相似文献

We determined the effect of low molecular weight components (LMWC) from healthy juvenile and adult Macrobrachium rosenbergii hemolymph on lectin activity and oxidative burst (OB) in hemocytes. In an attempt to identify the LMWC that affect the lectin's hemagglutinating activity or oxidative burst, we determined the hemolymph carbohydrates and free amino acids (FAA) concentration. The LMWC (<2000 Da) were obtained after dialysis of the hemolymph. Our results showed that LMWC from juveniles exerted a greater inhibition on lectin than LMWC from adult hemolymph. Production of superoxide radicals by hemocytes was lower in the presence of juvenile (p<0.05) as compared to adult LMWC. FAA composition of the hemolymph and of LMWC from adults showed higher proportion of alanine (which corresponded to 25% of total FAA) and proline (>20%); whereas, in juveniles, the main FAA identified were glycine (>40%) and alanine (26%). N-Acetyl-D-glucosamine (GlcNAc) was the main sugar residue in the hemolymph and LMWC from juveniles; its concentration was 2.4 times higher than glucose (Glc), whereas, in adults, Glc was the main free sugar residue. Our results suggest that the proportion of FAA and carbohydrates in the hemolymph of M. rosenbergii seems to be correlated with the maturation process; furthermore, the high proportion of free GlcNAc and glycine regulate, in the juvenile stage, lectin activity and cellular oxidative mechanisms, respectively.  相似文献

The concentrations of total free amino acids (TFAA) and humic substances (HS) accumulating in media conditioned by axenic and non-axenic duckweed fronds (Lemna minor L.) were analyzed at various time intervals over a 21-day incubation period with the aid of a Shimadzu HPLC system. In the non-axenic Lemna cultures, the concentrations of both TFAA and HS continued to increase throughout the incubation period, although the rate of increase was higher in the initial stages. In contrast, the concentrations of both TFAA and HS reached asymptotic values in media conditioned by non-axenic Lemna after 10–12 days. As a result, the concentrations of both FAA and HS became significantly higher in media conditioned by axenic Lemna fronds than in those conditioned by non-axenic Lemna from days 10–12 until the end of the experiment. The possible reasons for the differences in the accumulation patterns of TFAA and HS in media conditioned by axenic and non-axenic Lemna and their ecological significance are discussed.  相似文献

Rabbit lymphocytes exposed to hyposmotic media first swell and then recover their initial volume within 6 min. During volume recovery, free amino acids (FAA) decrease from 451.1 to 208 nmoles/mg protein. Taurine was the dominating FAA, accounting for 70% of the FAA pool. The time course of 3H-taurine release induced by hyposmolarity followed that of volume recovery. Efflux of 3H-taurine in an 8 min period was 17.8% (of total labeled taurine accumulated during loading) in an isosmotic medium. Reducing osmolarity to 0.87, 0.75, 0.62, and 0.5 increased this release to 24.8%, 38.1%, 56.4% and 70.9%, respectively. The volume-sensitive release of 3H-taurine was unaffected by omission of external Na+ or Ca++ and was reduced by 23% in the absence of Cl-. It was unaffected by agents disrupting the cytoskeleton or by tetraethylammonium, barium, quinidine, and gadolinium, but was 26% reduced by DIDS. Taurine release was inhibited at 4 degrees C, but was unchanged at 15 degrees C or 25 degrees C. An involvement of FAA, particularly taurine, in lymphocyte volume regulation is suggested.  相似文献

The role of free amino acids (FAA) in oocyte hydration during final maturation has been studied in plaice Pleuronectes platessa and lemon sole Microstomus kitt by in vivo and in vitro measurements. In vitro final maturation was initiated by the administration of human chorionic gonadotropin on large vitellogenic oocytes. The eggs produced in vitro had the same fraction of their total amino acid pool present in the free form as the in vivo hydrated eggs, regardless of whether FAA had been present in the incubation medium or not. The FAA pool in the mature egg was increased 10–15 times that of the oocyte, and the two FAA pool profiles differed strongly. The FAA profiles of the egg groups (intra- as well as interspecific) were almost identical except that the taurine content was lower in eggs in vitro . A major protein band of about 100 kDa was present on SDS electrophoretic gels of oocytes but missing on gels of hydrated eggs. This protein, presumably a lipovitellin, is the most likely origin of the egg FAA pool. We suggest that marine fishes with pelagic eggs share a common mechanism for oocyte hydration whereby partial hydrolysis of specific yolk proteins to FAA creates a major part of the osmotic potential needed for the water influx.  相似文献

Fourteen acidic and neutral free amino acids (FAA) were investigated in soft tissue of Macoma balthica from different depth zones of the Gulf of Gdansk (Baltic Sea) over a full seasonal cycle. The dry weight of the bivalves and physico-chemical parameters of overlying bottom water and surface sediments were measured simultaneously at each site. In the brackish waters of the Baltic, the main pool of FAA is composed of Ala, Gln, Arg, Gly and Orn which represent approximately 80% of the total. Compared to the full saline environments, the composition of FAA in the clams from the Baltic differs substantially. The differences can be attributed to the lower salinity of the Baltic. In the Baltic, Gly appears to play a most important role in regulating intracellular osmolarity in the clams, a function performed primarily by Tau in Atlantic and North Sea populations. Spatio-temporal variations of the FAA are affected by biotic and environmental parameters; their respective influence differs with the amino acids. The concentration of Arg depends on its uptake from the external medium. However, its level might be temporarily modified by stress-induced metabolic transformation (e.g. hydrolysis to Orn) caused by changes in the ambient environment. The concentration of Ala increases with depth, probably because of physiological adaptations of the animal to diminishing oxygen concentration through anaerobic glucose catabolism. Biosynthesis of Ala, similarly to Gln, in the shallower zone is generally related to the physiological state of an organism. The concentration of Gly is most likely regulated by internal mechanisms driven by gonadal development and reproduction.  相似文献

Freshwater mussels are ecologically and economically important members of many aquatic ecosystems, but are globally among the most imperiled taxa. Propagation techniques for mussels have been developed and used to boost declining and restore extirpated populations. Here we use a cohort of propagated mussels to estimate the intrinsic variability in size and growth rate of Lampsilis siliquoidea (a commonly propagated species). Understanding the magnitude and pattern of variation in data is critical to determining whether effects observed in nature or experimental treatments are likely to be important. The coefficient of variation (CV) of L. siliquoidea soft tissues (6.0%) was less than the CV of linear shell dimensions (25.1–66.9%). Size-weight relationships were best when mussel width (the maximum left-right dimension with both valves appressed) was used as a predictor, but 95% credible intervals on these predictions for soft tissues were ∼145 mg wide (about 50% of the mean soft tissue mass). Mussels in this study were treated identically, raised from a single cohort and yet variation in soft tissue mass at a particular size class (as determined by shell dimensions) was still high. High variability in mussel size is often acknowledged, but seldom discussed in the context of mussel conservation. High variability will influence the survival of stocked juvenile cohorts, may affect the ability to experimentally detect sublethal stressors and may lead to incongruities between the effects that mussels have on structure (via hard shells) and biogeochemical cycles (via soft tissue metabolism). Given their imperiled status and longevity, there is often reluctance to destructively sample unionid mussel soft tissues even in metabolic studies (e.g., studies of nutrient cycling). High intrinsic variability suggests that using shell dimensions (particularly shell length) as a response variable in studies of sublethal stressors or metabolic processes will make confident identifications of smaller effect sizes difficult.  相似文献

The fauna associated with hard bottom mussel beds along the exposed Pacific coast of Chile was examined. The abundance of adult (>10 mm body length) purple mussels Perumytilus purpuratus varied between 32 and 75 individuals per 50 cm2, and their biomass between 4.8 and 8.6 g AFDW per 50 cm2 at eight sampling sites between Arica (18°S) and Chiloé (42°S). At all sampling sites, the associated fauna was dominated by suspension-feeding organisms (cirripeds, spionid and sabellid polychaetes, a small bivalve) followed by grazing peracarids and gastropods. Predators and scavengers also reached high abundances while deposit- and detritus-feeding organisms were of minor importance. The majority of organisms associated with these hard bottom mussel beds feed on resources obtained from the water column or growing on the mussels rather than on materials deposited by the mussels. This is in contrast to the fauna associated with mussel beds on soft bottoms, which comprises many species feeding on material accumulated by mussels (faeces and pseudofaeces) and deposited within the mussel bed. Many of the organisms dwelling between mussels both on hard bottoms and on soft bottoms have direct development, but organisms with pelagic development also occur abundantly within mussel beds. We propose that species with direct development are disproportionately favoured by the structurally complex habitat with diverse interstitial spaces between the mussels, which provides ample shelter for small organisms. We conclude that mussels on hard-bottoms primarily provide substratum for associated fauna while mussels on soft bottoms provide both substratum and food resources. Electronic Publication  相似文献

The accumulation of nitrogen in different cellular pools by the macroalgae Ulva fenestrata (Postels and Ruprecht) (Chlorophyta) and Gracilaria pacifica (Abbott) (Rhodophyta) was studied in a laboratory experiment. After 8 or 9 days of nitrogen enrichment, nitrate, ammonium, free amino acid (FAA), protein, chlorophyll (chl), phycoerythrin (PE), and insoluble nitrogen pools were extracted and analyzed, and their relative contribution to total nitrogen (TN) was assessed. In U. fenestrata, the nitrate and ammonium enrichments resulted in a significant increase of TN from 2.41% dry weight (dw) to 4.19% and 4.71% dw, respectively. All the extracted N pools increased significantly. In G. pacifica, TN increased more under ammonium enrichment than under nitrate enrichment. In both macroalgae, proteins and FAA were the most important N storage pools. Protein-N ranged from 700 to 2300 μmol N·g dw−1 (43%–66% of TN) and contributed the most to TN increase (41%–89%). The FAA pool was always larger in G. pacifica than in U. fenestrata. In both species, the FAA pool accounted for 4%–17% of TN (70–600 μmol N·g dw−1). In U. fenestrata, nitrate can represent a temporary storage pool: it accumulated up to 200 μmol N·g dw−1 (7% of TN) and contributed more than FAA to overall increase in cellular nitrogen. In contrast, G. pacifica had a small nitrate pool. The PE pool in G. pacifica increased with TN but was never more than 9% of total protein-N or 6% of TN, and it was less important than FAA as a storage pool. All TN was recovered in the extracted and insoluble N pools at the end of the experiment in U. fenestrata. Summary. We studied in vivo production of taurine, hypotaurine and sulfate following subcutaneous administration of L-cysteinesulfinate (CSA) to rats and mice. When 5.0 mmol/kg of body weight of CSA was injected to rats, increased urinary excretions of taurine, hypotaurine and sulfate in 24 h urine were 617, 52 and 1,767 μmol/kg, respectively. From these results together with our previous data, sulfate production was calculated to be 1.6 times greater than taurine production. Increased contents (μmol/g of wet tissue) over the control of taurine and hypotaurine in mouse tissues at 60 min after the injection of 5.0 mmol/kg body weight of CSA were: liver, 3.5 and 9.9; kidney, 0.3 and 5.2; heart, 3.7 and 0.2; blood plasma, 0.4 and 0.2, respectively. Upon loading of hypotaurine or taurine, tissue contents of these amino acids in liver and kidney increased greatly. Our results indicate that liver is the most active tissue for taurine production, followed by kidney, and that external CSA, hypotaurine and taurine are easily taken up by these tissues.  相似文献
