The determination of benzodiazepines (carbamazepine and clobazam) in rabbit carcass tissues and larvae of three Calliphorid flies is described. After feeding the rabbits with lethal and toxic doses, samples of larvae and carcass tissues were studied. Residual drugs were determined using Ultra-high-performance liquid chromatography – quadrupole time-of-flight mass spectrometry (UHPLC/QTOF-MS). Benzodiazepines and its main active metabolites have been detected in the rabbit tissues at different retention times depending on the dosage used (lethal or toxic). A total of 1150 insects were collected and 800 larvae of the flies Chrysomya albiceps, Lucilia sericata and L. silvarum were used in the analysis. The presence of benzodiazepines in the rabbit tissues has been shown to typically affect the larval development cycle of the three necrophagous flies. Chrysomya albiceps larvae feed on drugs developed faster, while the development of L. sericata and L. silvarum larvae slowed. These results indicate that drugs have an impact on the life cycles of insects, which suggests that the presence of these substances is a factor that needs to be taken into account when estimating the post-mortem interval (PMI).  相似文献

死后不同时间,在尸体上出现不同种类食尸性蝇类的演替规律,可用于准确推断死亡时间。传统上仅依据蝇类形态学特征来判断种属,但由于蝇类的形态结构复杂和种间形态差异微小等特点,对蝇类尤其是对蝇类幼虫的种属鉴别很难。因此应用分子生物学方法对食尸性蝇类及其幼虫进行种属鉴定非常重要。本研究主要是利用此方法对我国西部部分地区常见双翅目食尸性蝇类包括:开普黑蝇、大头金蝇、丝光绿蝇及部分卵,铜绿蝇、棕尾别麻蝇及部分幼虫和蛹的线粒体DNA(mtDNA)上细胞色素氧化酶辅酶Ⅰ(COⅠ)中278 bp的基因序列进行鉴别。除个别蝇类如丝光绿蝇与铜绿蝇外,该方法均能有效地将上述食尸性蝇类鉴定到种属水平。在我国,它将成为法医鉴别食尸性蝇类种属的可靠依据。  相似文献

Cadaver insects cause dead bodies to break down in ecological processes. Still, the knowledge has been accumulated slowly about the structure and role of the insect assemblages attending to death scenes. This study analysed the successional patterns of insects on pig cadavers in the spring, summer, and autumn from 2016 to 2019 at Yeongdo Island, Busan, South Korea. A sum of 71 insect species belonging to four orders and 21 families were sampled. Predominant insects were largely Diptera and Coleoptera. The majority of the flies were the calliphorids as Chrysomya pinguis (Walker) and Lucilia porphyrina (Walker). The most common beetles were Creophilus maxillosus (L.), Omosita japonica (Reitter), and Staphylinidae sp. regardless of seasons. Occurrence matrices were made from the successional patterns of insect species during 42 sampling intervals in the spring, 38 sampling intervals in the summer, and 26 sampling intervals in the autumn. Permutation analyses of the occurrence matrices showed that the patterns of succession were similar between spring 2016 and 2017 (P = 0.019), between summer 2016 and 2017 (P = 0.003), and between autumn 2016 and 2019 (P = 0.002). The result shows that there are clear patterns in succession between seasons, and provides the reference data to presume the death time, at least in the southeast part of South Korea.  相似文献

There is unanimous consensus that insects are important vectors of foodborne pathogens. However, linking insects as vectors of the pathogen causing a particular foodborne illness outbreak has been challenging. This is because insects are not being aseptically collected as part of an environmental sampling program during foodborne outbreak investigations and because there is not a standardized method to detect foodborne bacteria from individual insects. To take a step towards solving this problem, we adapted a protocol from a commercially available PCR-based system that detects foodborne pathogens from food and environmental samples, to detect foodborne pathogens from individual flies.Using this standardized protocol, we surveyed 100 wild-caught flies for the presence of Cronobacter spp., Salmonella enterica, and Listeria monocytogenes and demonstrated that it was possible to detect and further isolate these pathogens from the body surface and the alimentary canal of a single fly. Twenty-two percent of the alimentary canals and 8% of the body surfaces from collected wild flies were positive for at least one of the three foodborne pathogens. The prevalence of Cronobacter spp. on either body part of the flies was statistically higher (19%) than the prevalence of S. enterica (7%) and L.monocytogenes (4%). No false positives were observed when detecting S. enterica and L. monocytogenes using this PCR-based system because pure bacterial cultures were obtained from all PCR-positive results. However, pure Cronobacter colonies were not obtained from about 50% of PCR-positive samples, suggesting that the PCR-based detection system for this pathogen cross-reacts with other Enterobacteriaceae present among the highly complex microbiota carried by wild flies. The standardized protocol presented here will allow laboratories to detect bacterial foodborne pathogens from aseptically collected insects, thereby giving public health officials another line of evidence to find out how the food was contaminated when performing foodborne outbreak investigations.  相似文献

Sarcophaga dux (Diptera: Sarcophagidae) is a necrophagous flesh fly species with potential forensic value for estimating minimum postmortem interval (PMImin). The basic developmental data and precise age estimates of the pupae are significant for PMImin estimation in forensic investigations. In the present study, we investigated the development data of that species at seven constant temperatures varying from 16 °C to 34 °C, including body length changes of the larve, developmental duration and accumulated degree hours of the preadults. Several reference genes for relative quantification of the differentially expressed genes (DEGs) were firstly selected and evaluated in the pupae of different ages under different temperatures. The DEGs of the insects during the pupal period at different constant temperatures (34, 25 and 16 °C) were further analyzed for more precise age estimation. The results showed that the developmental durations of the preadults at 16, 19, 22, 25, 28, 31 and 34 °C were 1478.6 ± 18.3 h, 726.1 ± 15.8 h, 538.5 ± 0.9 h, 394.1 ± 9.5 h, 375.6 ± 10.8 h, 284.1 ± 7.3 h, and 252.5 ± 6.1 h, respectively. The developmental threshold temperature the flies was 12.27 ± 0.35 °C, and the thermal summation constant was 5341.71 ± 249.29° hours. The most reliable reference genes during the pupal period at different temperatures were found: GST1 and 18S rRNA for the 34 °C group, GST1 and RPL49 for 25 °C, and 18S rRNA and 28S rRNA for 16 °C. The four differential expression genes (Hsp60, A-alpha, ARP, and RPL8) have the potential to be used for more precise age estimation of pupal S. dux. This work provides important basic developmental data and a more precise age estimation method for pupal S. dux, and improves the value of this species for PMImin estimation in forensic investigations.  相似文献

The stability of drugs in biological evidence during collection and storage is of particular concern in forensic investigations. Microbes in the samples and other elements are an essential component of these investigations. In the current study, the HPLC method was used to examine the stability of (1R, 2S)-(-)-Ephedrine hydrochloride in plasma and urine samples inoculated with C. albicans after storage at 37 °C for 48 h and −20 °C for six months. In the stability experiment, MIC50% of (1R, 2S)-(-)-Ephedrine hydrochloride was applied according to MIC and MFC that were determined in this work. This drug had MIC and MFC of 50 and 100 ppm, respectively. In HPLC analysis, the standard (1R, 2S)-(-)-Ephedrine hydrochloride had a retention time of 1.63 and was used to identify this drug in samples that had or had not been exposed to C. albicans. The findings demonstrated that within 48 h at 37 °C, C. albicans had an impact on the drug concentration (10% and more than 15%, respectively, were lost in plasma and urine samples inoculated with C. albicans). In plasma samples, the drug content remained stable at −20 °C for three months, although it degraded in urine samples after one month. In plasma and urine samples, the concentration reduction had surpassed 70% and 50% by the sixth month, respectively. The results of this investigation show that C. albicans can change the stability of (1R, 2S)-(-)-Ephedrine hydrochloride in plasma and urine samples that contain MIC50% of Ephedrine hydrochloride.  相似文献

Necrophagous insects, mainly Diptera and Coleoptera, are attracted to specific stages of carcass decomposition, in a process of faunistic succession. They are very important in estimating the postmortem interval, the time interval between the death and the discovery of the body. In studies done with pig carcasses exposed to natural conditions in an urban forest (Santa Genebra Reservation), located in Campinas, State of S?o Paulo, southeastern Brazil, 4 out of 36 families of insects collected - Calliphoridae, Sarcophagidae, Muscidae (Diptera) and Dermestidae (Coleoptera) - were considered of forensic importance, because several species were collected in large numbers both visiting and breeding in pig carcasses. Several species were also observed and collected on human corpses at the Institute of Legal Medicine. The species belonged to 17 different families, 6 being of forensic importance because they were reared from human corpses or pig carcasses: Calliphoridae, Sarcophagidae, Muscidae, Piophilidae (Diptera), Dermestidae, Silphidae and Cleridae (Coleoptera). The most important species were: Diptera - Chrysomya albiceps, Chrysomya putoria, Hemilucilia segmentaria, Hemilucilia semidiaphana (Calliphoridae), Pattonella intermutans (Sarcophagidae), Ophyra chalcogaster (Muscidae), Piophila casei (Piophilidae); Coleoptera - Dermestes maculatus (Dermestidae), Oxyletrum disciolle (Silphidae) and Necrobia rufipes (Cleridae).  相似文献

Although the pupation behavior of blowflies has been widely studied, this preliminary study was done on the vertical dispersal behavior (both ascending and descending) and fly emergence rate of third instar larvae of Chrysomya megacephala (Fabricius) to evaluate weather immature stages of blowflies survive burial and emerge out as adult. Third instar larvae of Chrysomya megacephala were placed at three different depths (5 cm, 25 cm and 45 cm) of soil under laboratory conditions to determine the impact of soil depth on the ascending and descending vertical dispersal behavior and the subsequent emergence of adults. The results of this study, although preliminary, but valuable to the field of forensic entomology because they provide new information about both ascending and descending vertical dispersal behavior of Chrysomya megacephala forensically important species of blowfly. In all the cases, maximum number of pupae recovered at the depth of 0 to 5 cm are 35.5 ± 4.5, 34 ± 1, 25 ± 5, when food was located at 5 cm, 25 cm and 45 cm depth respectively. This means that maximum no of larvae reached to the depth of 0 to 5 cm by ascending dispersal irrespective of at which depth they are placed. Paramount pupae were recovered from shallow burial depth of 0–5 cm in ascending dispersal and showed highest eclosion success i.e. 90.1% followed by 25 cm and 45 cm i.e. 71.7% and 55% respectively. While the number of pupae recovered as well as eclosion success was less in descending dispersal with an average of 62.8%, 39.25% and 33.9% at depths of 5, 25 and 45 cm respectively. This manifest if larvae disperse ascendingly, it increases their chance of survival.  相似文献

In Riyadh, Saudi Arabia, we are reporting two cases of natural death. The two bodies showed different types of habitat, insect colonization and decomposition stage. The first case was about the body of a 65-years-old male, with mummification of the clothed body was found in an outdoor habitat. Different life stages of Dermestes maculatus DeGeer (Coleoptera: Dermestidae were gathered from the cadaver, and due to the advanced degree of decomposition, the PMImin was estimated to be 3 months. The second body belonging to a 40-years-old male, was found in a semi-closed apartment (indoor habitat), and the body was at the end of the bloated decomposition stage. In this case, Musca domestica L. (Diptera: Muscidae) larvae were collected, and the PMImin was estimated to be 4 days. The limited insect activity for the two bodies caused by the advanced decomposition stage in the first case and indoor environment in the second. Average temperatures of (23.3 ± 1.6) °C for the first body and (27.5 ± 1.7) °C2 for the second body. Habitat was the key factor to attract insects to the two cadavers. This study stressed that, for accurate estimates of time since death, knowledge of the impact of different variables on insects found over and around the carrion is essential.  相似文献

Abiotic factors, such as lunar phases and tides, have a significant effect on insect development. Reproduction and immature development are usually interlinked to these abiotic factors. The tide is at its highest levels at full moon or new moon, hindering the feeding of the immature or causing their drowning. The oviposition by adult females is also compromised on these days because much of the available food is submerged. Another important abiotic factor is the wind, which displaces odoriferous particles in the air. Wind speed and direction are important elements to indicate potential sources of food for insects. I report on the effects of lunar phases, tides, and wind speed on the Calliphoridae fauna in mangrove swamps. The different species collected were identified, and the predominant species in the area were quantified. A total of 1,710 flies were collected over a 1-year period. Six Calliphoridae flies, Chloroprocta idioidea (Robineau-Desvoidy), Chrysomya megacephala (Fabricius), Chrysomya albiceps (Wiedemann), Chrysomya putoria (Wiedemann), Cochliomyia macellaria (Fabricius), and Lucilia eximia (Wiedemann) were collected. Data indicated that lunar phases have a significant effect on the abundance of C. albiceps (r?=?0.39, p?<?0.01), and that the variation of the tides also affected the abundance of C. putoria (r?=?0.40, p?<?0.00), C. macellaria (r?=?0.41, p?<?0.00), and C. idioidea (r?=?0.31, p?<?0.04). Mites can be found in all imaginable terrestrial habitats, in freshwater, and in salt water. Mites can be found in our houses and furnishings, on our clothes, and even in the pores of our skin—almost every single person carries mites. Most of the time, we are unaware of them because they are small and easily overlooked, and—most of the time—they do not cause trouble. In fact, they may even proof useful, for instance in forensics. The first arthropod scavengers colonising a dead body will be flies with phoretic mites. The flies will complete their life cycle in and around the corpse, while the mites may feed on the immature stages of the flies. The mites will reproduce much faster than their carriers, offering themselves as valuable timeline markers. There are environments where insects are absent or rare or the environmental conditions impede their access to the corpse. Here, mites that are already present and mites that arrive walking, through air currents or material transfer become important. At the end of the ninetieth century, the work of Jean Pierre Mégnin became the starting point of forensic acarology. Mégnin documented his observations in ‘La Faune des Cadavres’ [The Fauna of Carcasses]. He was the first to list eight distinct waves of arthropods colonising human carcasses. The first wave included flies and mites, the sixth wave was composed of mites exclusively. The scope of forensic acarology goes further than mites as indicators of time of death. Mites are micro-habitat specific and might provide evidential data on movement or relocation of bodies, or locating a suspect at the scene of a crime. Because of their high diversity, wide occurrence, and abundance, mites may be of great value in the analysis of trace evidence.  相似文献

ABSTRACT   Taking the conflict over the remains of Ned Kelly as a starting point, in this article I trace the various conceptions of the, body as evidence within the intertwined histories of anthropology, criminology, and medicine to explore how anthropological practice brings the dead into being through exhumation and analysis. I outline the popular rhetorical tropes within which evidentiary claims are situated, exploring how the agency of people after death is understood within the framework of present-day forensic anthropological practice and how this is underwritten by a particular heritage of anatomical analysis. Increases in mortality of trembling aspen (Populus tremuloides Michx.) have been recorded across large areas of western North America following recent periods of exceptionally severe drought. The resultant increase in standing, dead tree biomass represents a significant potential source of carbon emissions to the atmosphere, but the timing of emissions is partially driven by dead‐wood dynamics which include the fall down and breakage of dead aspen stems. The rate at which dead trees fall to the ground also strongly influences the period over which forest dieback episodes can be detected by aerial surveys or satellite remote sensing observations. Over a 12‐year period (2000–2012), we monitored the annual status of 1010 aspen trees that died during and following a severe regional drought within 25 study areas across west‐central Canada. Observations of stem fall down and breakage (snapping) were used to estimate woody biomass transfer from standing to downed dead wood as a function of years since tree death. For the region as a whole, we estimated that >80% of standing dead aspen biomass had fallen after 10 years. Overall, the rate of fall down was minimal during the year following stem death, but thereafter fall rates followed a negative exponential equation with = 0.20 per year. However, there was high between‐site variation in the rate of fall down (= 0.08–0.37 per year). The analysis showed that fall down rates were positively correlated with stand age, site windiness, and the incidence of decay fungi (Phellinus tremulae (Bond.) Bond. and Boris.) and wood‐boring insects. These factors are thus likely to influence the rate of carbon emissions from dead trees following periods of climate‐related forest die‐off episodes.  相似文献

The effects of temperature and host species on the development of Nasonia vitripennis Walker (Hymenoptera: Pteromalidae), a forensically important parasitoid of carrion flies, were studied under laboratory conditions. Development time of N. vitripennis on five species of Calliphoridae (Diptera), Calliphora albifrontalis Malloch, Calliphora dubia Macquart, Lucilia sericata Meigen, Chrysomya rufifacies Macquart, and Chrysomya megacephala Fabricius, were determined under eight constant temperatures (15, 18, 21, 24, 27, 30, 33, and 36 °C). Thermal requirements for development (developmental thresholds, thermal constant, and optimum temperature) of N. vitripennis in each host species were estimated using linear and nonlinear models. Upper and lower developmental thresholds ranged between 36.6–38.4 and 9.6–11.1 °C, respectively. The optimum temperature for development was estimated at between 30.6 and 31.8 °C. Statistical differences in the development time of N. vitripennis on the various calliphorid host species were evident within all temperature treatments, particularly at the upper and lower temperature range investigated. As such, it is recommended that insect‐based estimates of time since death in forensic investigations relying on parasitoid evidence should use host‐specific development data where available.  相似文献

The composition of syninclusions was analyzed in three representative samples of the Late Eocene Rovno amber. Statistical analysis revealed a stable connection between three pairs of taxa in all three samples, viz., the midges Chironomidae and Ceratopogonidae; the flies Dolichopodidae and aphids Germaraphis; and the mite Glaesacarus rhombeus and Sciara zone dipterans (Sciaroidea, Tipuloidea, etc.). The first pair is interpreted as representing the assemblage of aeroplankton insects; the second, the assemblage of dwellers of the trunks of amber trees; and the third pair, the assemblage of dead wood consumers. The results, although provisional, support the idea that the study of syninclusions is a useful instrument for understanding the structure of past communities.  相似文献

Recent investigations have proved the crucial role of nutritional antioxidants to prevent the damage caused by toxic compounds. In this study, the antioxidant effect of date palm fruit extract on dimethoate-induced oxidative stress and nephrotoxicity in rat is investigated and compared with the effect of the well-known antioxidant vitamin C. Male Wistar rats were randomly divided into six groups of ten each: a control group (C), a group that received dimethoate (20 mg/kg body weight) (D), a group given Deglet Nour extract (DNE), a group treated with DNE 30 min before the administration of dimethoate (DNE + D), a group which received VitC (100 mg/kg body weight) plus dimethoate (Vit C + D), and a group given dimethoate for the first month and DNE 30 min after administration of dimethoate, during the second month (D + DNE). These components were daily administered by gavage for 2 months. After completing the treatment period, blood samples from rats were collected under inhaled diethyl ether anesthesia for serum urea, uric acid, and creatinine levels, while the rat kidneys were obtained for enzyme assays and histology. Oral administration of dimethoate in rats induced a marked renal failure characterized by a significant increase in serum creatinine and urea levels (p < 0.01) in addition to a significant decrease in serum uric acid (p < 0.05). Interestingly, these drastic modifications were accompanied by a marked enhancement of lipid peroxidation in kidney, indicating a significant induction of oxidative damage (p < 0.01) and dysfunctions of enzymatic antioxidant defenses. These biochemical alterations were also accompanied by histological changes in kidney revealed by a narrowed Bowman’s space, tubular degeneration, tubular cell desquamation, and tubular dilatation of proximal tubules. Treatment with date palm fruit extract (Deglet Nour) and also with vitamin C significantly (p < 0.05) reversed the serum renal markers to their near-normal levels when compared with dimethoate-treated rats. In addition, Deglet Nour extract and vitamin C significantly reduced lipid peroxidation, restored the antioxidant defense enzymes in the kidney, and improved the histopathology changes. The present findings indicate that in vivo date palm fruit may be useful for the prevention of oxidative stress-induced nephrotoxicity.  相似文献

Chill susceptible insects like Drosophila lose the ability to regulate water and ion homeostasis at low temperatures. This loss of hemolymph ion and water balance drives a hyperkalemic state that depolarizes cells, causing cellular injury and death. The ability to maintain ion homeostasis at low temperatures and/or recover ion homeostasis upon rewarming is closely related to insect cold tolerance. We thus hypothesized that changes to organismal ion balance, which can be achieved in Drosophila through dietary salt loading, could alter whole animal cold tolerance phenotypes. We put Drosophila melanogaster in the presence of diets highly enriched in NaCl, KCl, xylitol (an osmotic control) or sucrose (a dietary supplement known to impact cold tolerance) for 24 h and confirmed that they consumed the novel food. Independently of their osmotic effects, NaCl, KCl, and sucrose supplementation all improved the ability of flies to maintain K+ balance in the cold, which allowed for faster recovery from chill coma after 6 h at 0 °C. These supplements, however, also slightly increased the CTmin and had little impact on survival rates following chronic cold stress (24 h at 0 °C), suggesting that the effect of diet on cold tolerance depends on the measure of cold tolerance assessed. In contrast to prolonged salt stress, brief feeding (1.5 h) on diets high in salt slowed coma recovery, suggesting that the long-term effects of NaCl and KCl on chilling tolerance result from phenotypic plasticity, induced in response to a salty diet, rather than simply the presence of the diet in the gut lumen.  相似文献

Agrochemicals are a major cause of concern for the aquatic environment because of their toxicity, persistence, and tendency to accumulate in the organisms. The impact of these chemicals on aquatic organisms is due to their movement from various diffuse or point sources, which poses a great threat to aquatic fauna especially fishes, which constitute one of the major sources of protein-rich food for mankind. The present study is a first of its kind, where the toxic potential of two sublethal concentration (LC1/10th and LC1/25th) of four different classes of agrochemicals have been tested (Insecticide- Imidacloprid-0.074 ppm, 0.02 ppm, Fungicide-Curzate- 4.9 ppm, 1.96 ppm, Herbicide- Pyrazosulphuron ethyl-50 ppm, 20 ppm and Fertilizer-Micronutrient mixture 500 ppm, 200 ppm) on candidate markers of hypothalamus pituitary-thyroid axis (TSH, T3, T4, TSHβr) in Oreochromis mossambicus (tilapia) by validating hormonal level and mRNA expression. The results reveal that exposure to agrochemicals resulted in a broad range of alterations with maximum damage being caused by insecticide followed by herbicide and fungicide in that order on the thyroid axis. The results of the present study highlight the need for more detailed studies on the effects of agrochemicals that accumulate in organisms and propose that there should be a check on the rampant use of agrochemicals.  相似文献

Drosophila suzukii (Matsumura) (Diptera: Drosophilidae) is a widely distributed pest species of soft-skinned fruits. Recent studies suggest the use of sterile insect technique (SIT) as a control method for this species; however, many factors can impact effectiveness of a SIT programme, including the environmental conditions. Environmental condition is critical at the time of the release and in the days afterwards, since it may impact sterile insects’ survival and ability to mate. Thus, we verified the influence of temperature and relative humidity on mating and survival of fertile and sterile D. suzukii, when insects were food provided or deprived. Highest mating rates occurred when sterile or fertile flies provided with food were exposed to 25ºC or 81%–100% relative humidity, while temperatures of 10 and 35ºC and humidity below 60% impaired mating. Overall, mating rate among food-deprived flies was low in all temperatures and humidity levels tested, but fertile insects were more prone to mate when compared to sterile flies. Survival was negatively influenced by high temperatures, low relative humidity and food deprivation. The information present in this study is useful to be considered for release of sterile D. suzukii.  相似文献
