An ethanol-insoluble, high molecular weight fraction was collected from the juice of Morinda citrifolia fruit grown in Viet Nam. The fraction is composed primarily of carbohydrate (67% (w/w)). The polysaccharide fraction consists predominantly of GalAp (53.6mol%), Araf (13.6mol%), Galp (17.9mol%) and Rhap (9.5mol%). Glycosyl linkage analysis suggests the polysaccharide fraction contains mostly the pectic polysaccharides, homogalacturonan (4-GalAp), rhamnogalacturonan I (4-GalAp, 2-Rhap, 2,4-Rhap), arabinan (5-Araf, 3,5-Araf, t-Araf), type I arabinogalactan (4-Galp, 3,4-Galp, t-Araf) and beta-glucosyl Yariv-binding type II arabinogalactan (3,6-Galp, t-Araf). Low levels of xyloglucan (4-Glcp, 4,6-Glcp, t-Xylp, t-Fucp), heteroxylan (4-Xylp) and heteromannan (4-Manp) are also present. 相似文献

In the present study the phytochemical composition and biological activities of the aerial part extracts of Asteriscus graveolens against pathogenic bacteria and leishmania parasite were evaluated. Phytochemical analysis revealed the presence of polyphenols, flavonoids and tannins. The ethyl acetate fraction exhibited high antioxidant potential of 359.72 ± 32.03 μg and 326.76 ± 15.86 μg of ascorbic acid equivalents and 13.32 ± 0.19 μg/mL by PM, FRAP and DPPH assays respectively. This fraction also displayed a moderate antibacterial activity against Listeria monocytogenes ATCC 19115 (MIC = 0.312 mg/mL) and strong antileishmanial activity against both promastigote and amastigote forms with IC50 ranging from 22.93 ± 0.39 μg/mL to 35.23 ± 0.62 μg/mL. Furthermore the ethyl acetate fraction exhibited low cytotoxicity and good selectivity index towards the macrophage cell line Raw 264.7. HPLC analysis of the active fraction revealed the presence of hydroxycinnamic acid as major compound (30.56%). Asteriscus graveolens is a promising source of bioactive compounds that could be potentially used in cosmetic and pharmaceutical industry. 相似文献

Ten flavone compounds, including three new flavonoid glycosides, were isolated from defatted rapeseed, and their protective antioxidant effect on H2O2-induced oxidative damage in human umbilical vein endothelial cells (ECV-304) was investigated. Three new flavonoid glycosides were identified as kaempferol-3-O-[(6-O-sinapoyl)-β-d-glucopyranosyl-(1 → 2)-β-d-glucopyranoside]-7-O-β-d-glucopyranoside (8), kaempferol-3,7-di-O-β-d-glucopyranoside-4'-O-(6-O-sinapoyl)-β-d-glucopyranoside (9), and kaempferol-3-O-[(3-O-sinapoyl)-β-d-glucopyranosyl-(1 → 2)-β-d-glucopyranoside]-7-O-β-d-glucopyranoside (10). The protective effects of all of the isolated compounds on H2O2-induced oxidative damage were assessed, and the activities of superoxide dismutase (SOD) and lactate dehydrogenase (LDH) were measured. All of compounds had a protective effect on H2O2-induced oxidative damage in ECV-304 cells and the presence of a substituted sinapoyl group and its position in the structures were used to elucidate the activity differences. 相似文献

Tectona grandis L.f is a timber plant that is commonly referred to as teak. Its wide use as a medicine in the various indigenous systems makes it a plant of importance. A wide gamut of phytoconstituents like alkaloids, phenolic glycosides, steroids, etc. has been reported. A renewed interest in this plant has resulted in scientific investigations by various researchers towards the isolation and identification of active constituents along with scientific proof of its biological activities. The different parts of the plant have been scientifically evaluated for their antioxidant, antipyretic, analgesic, hypoglycemic, wound healing, cytotoxic, and many more biological activities. Documentation of this scientific knowledge is of importance to have consolidated precise information encompassing the various aspects of this plant, which could provide a base for future studies. This review is a compilation of the salient reports on these investigations concerning phytochemistry, the methods used to identify and quantify the constituents, the evaluation methods of the biological activity, toxicological studies, allergies and the patent/patent applications. The phytochemical, antioxidant and mineral composition of hydroalcoholic extract of leaves of Cichorium intybus L., was determined. The leaves were found to possess comparatively higher values of total flavonoids, total phenolic acids. The phytochemical screening confirmed the presence of tannins, saponins, flavonoids, in the leaves of the plant. The leaf extract was found to show comparatively low value of IC50 for 2,2-diphenyl-1-picrylhydrazyl (DPPH) inhibition. The IC50 value of chicory leaves extract was found to be 67.2 ± 2.6 μg/ml. The extracts were found to contain high amount of mineral elements especially Mg and Zn. Due to good phytochemical and antioxidant composition, C. intybus L., leaves would be an important candidate in pharmaceutical formulations and play an important role in improving the human health by participating in the antioxidant defense system against free radical generation. 相似文献

The effects of juice from Morinda citrifolia (noni) on gastric emptying, gastrointestinal transit, and plasma level of cholecystokinin (CCK) in rats were studied. Male rats were given noni by gavage at levels of 0.25, 1, or 4 ml/kg once per day for one or 7 days. The rats in the control group were given water, while the rats in the experimental group were fasted overnight before measurement of gastrointestinal motility. Gastrointestinal motility was assessed in rats 15 min after intragastric instillation of a test meal containing charcoal (10%) and Na251CrO4 (0.5 microCi/ml). Gastric emptying was determined by measuring the amount of radiolabeled chromium contained in the small intestine as a percentage of the initial amount received. Then, gastrointestinal transit was evaluated by calculating the geometric center of distribution of the radiolabeled marker. Finally, blood samples were collected for measurement of CCK by radioimmunoassay. The administration of noni at 0.25 ml/kg, but not at 1 ml/kg and 4 ml/kg, for 1 day significantly inhibited gastric emptying. In contrast, gastric emptying was significantly inhibited by oral noni (0.25, 1, or 4 ml/kg) for 7 days. Intraperitoneal injection of lorglumide (5 or 10 mg/kg), a selective CCK1 receptor antagonist, effectively attenuated the noni-induced inhibition of gastric emptying. The intestinal transit and body weight, food intake, water intake, urine volume as well as feces weight were not altered by the administration of noni either acutely or chronically, but the administration of oral noni (1 ml/kg) for 7 days increased the level of plasma CCK in male rats. These results suggest that oral noni inhibits gastric emptying in male rats via a mechanism involving stimulation of CCK secretion and CCK1 receptor activation. 相似文献

Morinda citrifolia L. (noni) is a tropical plant that has been used for centuries in traditional medicine for numerous purposes and, nowadays, is largely commercialised on the worldwide market as a dietary supplement. Long-term in vitro cultures of Helianthus tuberosus dormant parenchyma explants constitute a classical growth model that can be used to evaluate the proliferative or antiproliferative and/or cytotoxic effects of different mixtures of chemicals present in food or with putative pharmacological applications. Explants of dormant tubers were cultured in vitro with 2%, 10% and 20% (v/v) of noni fruit juice. After four weeks of culture, explant cell growth was reduced 40.5%, 72.5% and 73.9%, respectively, by 2%, 10% and 20% (v/v) of noni juice in comparison to untreated controls. Our results demonstrated that noni fruit juice possesses strong antiproliferative capacity, low cytotoxicity and moderate antioxidant activity. 相似文献

Search for medicinal plants to treat kidney disorders is an important topic on phytotherapeutical research. Plumbago zeylanica L. is an important medicinal plant with hepatoprotective, anti-inflammatory, anti-diabetic, anti-cancer and anti-hyperlipidemic activities. In the present study, the protective effect of hydroalcoholic extract of P. zeylanica (HAPZ) in cisplatin induced nephrotoxicity was analyzed in Swiss albino mice. Treatment with higher dose (400 mg/kg) of HAPZ significantly reversed the adverse effect of cisplatin on kidney weight, serum urea and creatinine, indicating their renoprotective effect. The antioxidant effect of the drug is evident from its significant effect on Catalase, Glutathione peroxidase and lipid peroxidation activities. 相似文献

Context/objective: The mechanisms of immunomodulatory effects of Morinda citrifolia (Noni) were examined through intracellular signaling pathways in the splenocytes and their modulation by phytochemicals using bioinformatics tools. Materials and methods: Noni fruit juices without seeds (NSL) and with seeds (NWS) were co-incubated in vitro with splenocytes from young, middle-aged and old F344 male rats and proliferation of lymphocytes, cytokine production, antioxidant enzyme activities and intracellular signaling markers were measured. Results: NSL decreased lymphoproliferation in early middle-aged rats, and IL-2 and IFN-γ production in old rats. In contrast, NWS enhanced lymphoproliferation in young and old rats, IL-2 and IFN-γ production in middle-aged and old rats. The activities of antioxidant enzymes were augmented by NWS and NSL in old rats. NWS reversed age-related increase in lipid peroxidation in all age-groups, while NSL increased lipid peroxidation in old rats. NSL increased p-ERK in old rats and decreased p-CREB in young and middle-aged rats. In contrast, NWS decreased p-ERK in all age groups and increased p-CREB in old rats. Both NSL and NWS increased p-Akt expression in middle-aged and old rats. Both NSL and NWS suppressed p-NF-κB expression in middle-aged and old rats. Docking studies demonstrated that Noni phytochemicals, damnacanthal, myricetin and ursolic acid, are potent inhibitors of ERK with binding sites in the catalytic and phosphorylation sites of the molecule. Conclusions: These results suggest that Noni fruit juices with or without seeds modulate cell-mediated immunity and antioxidant enzyme activities based on the phytochemicals that may differentially influence cell signaling and therefore, age-associated immunity. 相似文献

Cell suspensions of Morinda citrifolia were cultivated in a B5-medium containing 4% sucrose as the sole carbon source and 1 mg l-1 naphthyl acetic acid (NAA) or 1 mg l-1 2,4-dichloro-phenoxyacetic acid (2,4-D). Both auxins were able to support growth but only in the presence of NAA anthraquinone production was observed. 2,4-D inhibited the production in NAA cultures. Anthraquinone synthesis took place in the growth and the stationary phase and amounts of 0.2–0.4 mmol (about 100–200 mg) g-1 dry weight could be reached.Under both growth conditions sucrose was hydrolyzed extracellularly by invertase. From the resulting monosaccharides, glucose was taken up preferentially and an appreciable uptake of fructose only took place when medium glucose was exhausted. Sugar uptake rates were similar when cells were grown in NAA and in 2,4-D medium but the intracellular sugar contents (expressed on a dry weight basis) differed considerably. The presence of sucrose, glucose and fructose was demonstrated under both growth conditions. The amounts of sucrose and glucose were much lower in the 2,4-D cells than in the NAA-cells especially during the growth phase. Fructose contents were low and comparable, while in NAA cells an unknown sugar (possibly the sugar moiety of the glycosylated anthraquinones) was observed especially at the end of the growth phase and in the stationary phase. The differences in sugar concentrations were even larger due to the lower water contents of the NAA cells.Respiration of 2,4-D cells was much higher than that of NAA cells during the growth phase. A sharp increase in sugar contents (mainly sucrose) occurred in the 2,4-D cells at the end of the growth phase and corresponded with the fall in respiratory activity.A possible correlation between the lack of production of anthraquinones in 2,4-D cells and a less efficient growth metabolism in these cells is discussed.Abbreviations AQ

From cell suspension cultures of Morinda citrifolia five known anthraquinones, rubiadin, lucidin, morindone, lucidin-3-β-primeveroside and morindone-6-β-primeveroside, and seven new anthraquinones were isolated. Cell cultures of Morinda citrifolia L. are capable of accumulating substantial amounts of anthraquinones. Chorismate formed by the shikimate pathway is an important precursor of these secondary metabolites. Isochorismate synthase (EC 5.4.99.6), the enzyme that channels chorismate into the direction of the anthraquinones, is involved in the regulation of anthraquinone biosynthesis. Other enzymes of the shikimate pathway such as deoxy-D-arabino-heptulosonate 7-phosphate synthase (EC 4.1.2.15) and chorismate mutase (EC 5.4.99.5) do not play a regulatory role in the process. The accumulation of anthraquinones is correlated with isochorismate synthase activity under a variety of conditions, which indicates that under most circumstances the concentration of the branchpoint metabolite chorismate is not a rate-limiting factor. Anthraquinone biosynthesis in Morinda is strongly inhibited by 2,4-D, but much less by NAA. Both auxins inhibit the activity of isochorismate synthase proportionally to the concomitant reduction in the amount of anthraquinone accumulated. However, the correlation between enzyme activity and rate of biosynthesis is less clear when the activity of the enzyme is very high. In this case, a limiting concentration of precursor may determine the extent of anthraquinone accumulation. Partial inhibition of chorismate biosynthesis by glyphosate leads to less anthraquinone accumulation, but also to a reduction in ICS activity. The complexity of the interference of glyphosate with anthraquinone biosynthesis is illustrated by the effect of the inhibitor in cell cultures of the related species Rubia tinctorum L. in these cells, glyphosate leads to an increase in anthraquinone content and a concomitant rise in ICS activity. All data indicate that the main point of regulation in anthraquinone biosynthesis is located at the entrance of the specific secondary route. 相似文献

In the current study, antioxidant, antibacterial activities, and the phenolic compositions of extracts from Helianthemum canum L. Baumg . (Apiaceae) aerial parts were investigated for the first time. The H . canum was extracted with 70% methanol (HCM eOH ) and water (HCW ). Both extracts were determined by total phenolic contents (3 mg/ml), flavonoids (1.5 mg/ml), flavonols (1.5 mg/ml), qualitative–quantitative compositions, iron (II ) chelation activities (0.1 – 5 mg/ml), free radical scavenging activities (DPPH ?: 0.01 – 0.6 mg/ml and ABTS +?: 0.125 – 0.5 mg/ml) and the effect upon inhibition of β ‐carotene/linoleic acid co‐oxidation (1 mg/ml). The peroxidation level was also determined using the thiobarbituric acid method (0.01 – 1.5 mg/ml). The results of the activity tests given as IC 50 values were estimated from non‐linear algorithm and compared with standards. Antibacterial activities of extracts and standards were evaluated against Gram‐ negative and ‐positive ten standard strains using disc diffusion and broth microdilution methods. The MIC results (312.5 – 2500 μg/ml) against tested microorganisms varied from 625 to 2500 μg/ml. In HPLC analysis, 3,5‐dihydroxybenzoic acid was found as the main substance in both extracts. These results showed that HCM eOH was richer in phenolic compounds (284.13 ± 0.30 mg GAE /g extract) from HCW (244.55 ± 0.35 mg GAE /g extract). In conclusion, H . canum extracts showed in vitro antibacterial and antioxidant activities. 相似文献

Various neurodegenerative diseases e.g., Alzheimer's affect many elderly people that eventually leads to disability and premature death. No effective treatment for these neurodegenerative diseases has been developed yet. Oxidative stress-mediated neurodegeneration is one of the key pathophysiological factors involved in these diseases. Since numerous botanic components are well-known potent antioxidants, in this study, we aim to examine the neuroprotective effects of phytochemical compositions from the water, methanolic, and 95% ethanolic extracts of the Crataegus pinnatifida fruit against hydrogen peroxide (H2O2)-induced cytotoxicity in PC12 neuronal cells. Our results revealed that the methanolic extract showed the most favorable antioxidant activities in scavenging various sources of free radicals. All three extracts concentration (0.5–5 μg/ml)-dependently protected PC12 against H2O2-induced cell death with the methanolic extract being the most potent one. Component elucidation revealed that the methanolic extract contained the greatest amounts of total phenolic compounds, flavonoids and tannin content. Conversely, the water soluble and 95% ethanolic extracts yielded a comparable amount of total triterpenoid. Our results indicated that methanolic and 95% ethanolic extracts of C. pinnatifida fruit show the potential to be novel neuroprotective agents to be considered in nutraceutical products for preventing oxidative-related disorders. Further investigation is necessary to verify the neuroprotective efficacy and mechanisms in vivo. 相似文献

The aim of the current study is to analyze the phytochemical, antioxidant and antimicrobial activities of 34 extracts prepared from Pistacia atlantica Desf. subsp. atlantica, according to gender, organ type (roots, buds and fruits), geographical location and stage of ripening. Bud extracts exhibited the highest phenolic content (565.74 ± 9.84 mg GAE/g DM), followed by fruit and root extracts. TFC and TTC ranged from 0.38 ± 0.03 to 1.92 ± 0.11 mg CE/g DM and from 0.37 ± 0.03 to 16.54 ± 0.94 mg CE/g DM, respectively. For DPPH and TAC assays, the values varied from 0.038 ± 0.000 to 1.331 ± 0.114 mg/mL and 1.58 ± 0.06 to 43.64 ± 2.58 mg AAE/g DM, respectively. Besides, bud extracts showed the highest bioactivity against pathogenic bacteria and a slight antifungal effect. Additionally, HPLC-DAD analysis revealed that the caffeic acid and the dimethyl-allyl caffeic acid characterized the bud extract, while the rutin and the hydroxytyrosol were abundant in the red fruit extract. The present evidence suggests that P. atlantica may be considered as a potential source of new additives for therapeutic, food and cosmetic products. 相似文献

Five series of dihydrotriazine derivatives containing chalcone (13a–i), phenoxy acetophenone (14a–b), benzyl benzene (15a–c), naphthoxyl acetophenone (16a–b) and benzyl naphthalene (17a–h) moieties were designed and synthesized. The antibacterial and antifungal activities of these compounds were evaluated against several strains of Gram-positive and Gram-negative bacteria, as well as a single fungus. Compound 17h was found to be the most potent of all of the compounds tested, with an MIC value of 0.5?μg/mL against several Gram-positive (Staphylococcus aureus 4220 and QRSA CCARM 3505) and Gram-negative (Escherichia coli 1924) strains of bacteria. However, this compound was inactive against Pseudomonas aeruginosa 2742 and Salmonella typhimurium 2421, indicating that its antibacterial spectrum is similar to those of the positive controls gatifloxacin and moxifloxacin. A new series of complexes of a ligand 4′, 7, 8-trihydroxy-isoflavone with transition metal (zinc, copper, manganese, nickel, cobalt) and selenium have been synthesized and characterized with the aid of elemental analysis, IR, electron ionization mass spectrum (EI-MS) and 1H NMR spectrometric techniques. The compounds were evaluated for their in vitro antibacterial activities and antitumor properties. The metal complexes were found to be more active than the free ligand. Investigation on the interaction between the complexes and calf-thymus DNA (CT DNA) showed that the absorbance of CT DNA increased and the maximum peak (λmax = 260 nm) red-shifted, while the intensity of fluorescence spectra of Epstein-Bart DNA (EB-DNA) gradually weakened, which indicated that all of these metal complexes tightly combined with CT DNA. 相似文献

The increasing resistance of infectious microbial organisms to the existing arsenal of antibiotic drugs is on the rise. There is a growing demand for the new antibiotics that are cost effective and easily available to the common people. In search of new antimicrobial entities, this report deals with the in vitro antimicrobial activities of the crude extracts of leaves of Ehretia Serrata. The methanolic extract and its sub-fractions namely n-hexane, chloroform, ethyl acetate, n-butanol and residual water fraction were screened against a range of 30 different bacterial strains and their zones of inhibition (ZI) and minimum inhibitory concentration (MIC) were subsequently evaluated. Methanolic extract has shown activity against all the tested microorganisms such as Azospirillum lipoferum, Escherichia coli, Pseudomonas aeruginosa, Stenotrophomonas maltophilia and Enterococcus sp. with ZOI ranged from 10.3 to 29.0 mm. Moreover, the MIC values of methanolic extract and its sub-fractions against the tested bacteria ranged from 0.8 to 5.1 mg/mL. GC–MS analysis of sub-fractions revealed the presence of mono (2-ethylexyl) 1,2-benzenedicarboxylate, diisooctyl-1,2-benzenedicarboxylate, 3,5-dehydro-6-methoxypivalate-cholest-22-ene-21-ol, and 3,5-bis (1,1-dimethylethyl)-4-hydroxybenzene-propanoic acid.
