The absence of an effective vaccine and the debilitating chemotherapy for Leishmaniasis demonstrate the need for developing alternative treatments. Several studies conducted with Morinda citrifolia have shown various biological activities, including antileishmanial activity, however its mechanisms of action are unknown. This study aimed to analyze the in vivo activity of M. citrifolia fruit juice (Noni) against Leishmania (Leishmania) amazonensis in C57BL/6 mice. M. citrifolia fruit juice from the Brazilian Amazon has shown the same constitution of other juices produced around the world and liquid chromatography–mass spectrometry analysis identified five compounds: deacetylasperulosidic acid, asperulosidic acid, rutin, nonioside B and nonioside C. Daily intragastric treatment with Noni was carried out after 55 days of L. (L.) amazonensis infection in C57BL/6 mice. Parasitic loads, cytokine and extracellular protein matrix expressions of the lesion site were analyzed by qPCR. Histopathology of the lesion site, lymph nodes and liver were performed to evaluate the inflammatory processes. Cytokines and biochemical parameters of toxicity from sera were also evaluated. The Noni treatment at 500 mg.kg-1.day-1 for 60 days decreased the lesion size and parasitic load in the footpad infected with L. (L.) amazonensis. The site of infection also showed decreased inflammatory infiltrates and decreased cytokine expressions for IL-12, TNF-α, TGF-β and IL-10. On the other hand, Noni treatment enhanced the extracellular matrix protein expressions of collagen IV, fibronectin and laminin in the infected footpad as well collagen I and II, fibronectin and laminin in the mock-infected footpads. No toxicity was observed at the end of treatment. These data show the efficacy of Noni treatment.  相似文献

Hippophae salicifolia, Elaeagnus pyriformis, Myrica esculenta and M. nagi are actinorhizal plants growing in the sacred forests of Northeast India with multipurpose uses. The present investigation was undertaken to determine the phenol, flavonoid and flavonol contents of the fresh fruit juice of these plant species including the antioxidant potential by means of DPPH, H2O2 and NO scavenging activity and FRP. The total phenolic, flavonoid and flavonol contents of fruit juice ranged from 321.68±0.06 to 76.67±0.01 mg/g GAE, 272.92±0.07 to 20.12±0.02 mg/g QE and 258.92±0.08 to 18.72±0.02 mg/g QE, respectively. At 2.0 mg/mL concentration, DPPH scavenging activity was found to be the highest in M. esculenta (89.62%) and the lowest in E. pyriformis (17.58%). The reducing power activity was found significantly higher in H. salicifolia juice, which increased with increase in concentration. The H2O2 scavenging activity of H. salicifolia juice was found to be as high as 98.78%, while Elaeagnus juice was found to be less effective with just 48.90%. Juice of H. salicifolia showed the greatest NO scavenging effect of 75.24% as compared to juice of E. pyriformis, where only 37.54% scavenging was observed at the same concentration. Taking into account all the experimental data, it can be said that the fruits of H. salicifolia and both M. nagi and M. esculenta have good antioxidant activity compared to fruits of E. pyriformis.  相似文献

Background

Berry fruit is known for its high contents of various bioactive compounds. The latter constitute of anthocyanins, flavonols and flavanols and posses high antioxidative activity. The highly dynamic antioxidant system can be evaluated in vitro and in vivo in several model organisms. These measurements represent a good approximation of the real potential of bioactive compounds in the cells of higher eucarions. The aim of the study was thus to determine in vitro and in vivo antioxidant activity of different berry juices, which reportedly contain high amounts of phenolics.

Methodology/Principal Findings

Five different berry species were collected from several locations in central Slovenia and juice was extracted from each species separately. Juice was assessed for their in vitro and in vivo antioxidant activity. Phenolic profiles of berries were determined with the use of a HPLC/MS system, in vitro antioxidant activity with the DPPH radical scavenging method and in vivo antioxidative activity using Saccharomyces cerevisiae. The highest diversity of individual phenols was detected for bilberry juice. The highest in vitro antioxidant capacity was determined for blackcurrant juice. A decrease in intracellular oxidation compared to control was observed in the following order: blackcurrant < chokeberry = blueberry < bilberry. The results indicate important differences in antioxidant activity of berry juices between in vitro and in vivo studies.

Conclusion/Significance

In addition to the total content of phenolic compounds entering the cells, a key factor determining antioxidative activity of berry juices is also the ratio between the compounds. Where high content levels of anthocyanins and very low content levels of flavonols and hydroxycinnamic acids were measured a lower intracellular oxidation has been detected. Specifically, intracellular oxidation increased with higher consumption of hydroxycinnamic acids and lower consumption of anthocyanins in the cells. Antioxidative activity also increased when the consumption of analyzed phenols was rather low.  相似文献

This study aimed at investigating the regulatory system of bacteriocin synthesis by Lactobacillus plantarum strains in vegetables and fruits in a model system. Sterile and neutralized cell-free supernatant (CFS) from L. plantarum strains grown in MRS broth showed in vitro antimicrobial activities toward various indicator strains. The highest activity was that of L. plantarum C2. The antimicrobial activity was further assayed on vegetable and fruit agar plates (solid conditions) and in juices (liquid conditions). A regulatory mechanism of bacteriocin synthesis via quorum sensing was hypothesized. The synthesis of antimicrobial compounds seemed to be constitutive under solid conditions of growth on vegetable and fruit agar plates. In contrast, it depended on the size of the inoculum when L. plantarum C2 was grown in carrot juice. Only the inoculum of ca. 9.0 log CFU ml−1 produced detectable activity. The genes plnA, plnEF, plnG, and plnH were found in all L. plantarum strains. The genes plnJK and plnN were detected in only three or four strains. Reverse-phase high-performance liquid chromatography purification and mass spectrometry analysis revealed the presence of a mixture of eight peptides in the most active fraction of the CFS from L. plantarum C2. Active peptides were encrypted into bacteriocin precursors, such as plantaricins PlnJ/K and PlnH and PlnG, which are involved in the ABC transport system. A real-time PCR assay showed an increase in the expression of plnJK and plnG during growth of L. plantarum C2 in carrot juice.  相似文献

Carotenoids, found in many fruits and vegetables, are antioxidants that protect human skin from UV radiation. In humans, fruit and vegetable intake increases carotenoid contents in skin, which are conventionally assessed by invasive blood tests. In this study, 47 healthy Korean subjects (volunteers) consumed fruit juice containing tomato, apple, strawberry, or grape three times per week for 6 weeks. Skin antioxidant levels were measured by non-invasive resonance Raman spectroscopy. The correlation between skin carotenoid (SC) score with demographic data (age, height, weight) and juice supplementation and changes in SC scores among groups were analyzed. Variations in skin antioxidant levels increased with juice supplementation (p < 0.05). Fruit juice intake was significantly correlated with SC score, indicating increased skin antioxidant levels. Grape and tomato increased skin antioxidant levels and showed higher antioxidant activity than other fruits. Fruit juices containing high levels of carotenoids and antioxidants may provide modest benefits to human health.  相似文献

Many insects feed on only one or a few types of host. These host specialists often evolve a preference for chemical cues emanating from their host and develop mechanisms for circumventing their host’s defenses. Adaptations like these are central to evolutionary biology, yet our understanding of their genetics remains incomplete. Drosophila sechellia, an emerging model for the genetics of host specialization, is an island endemic that has adapted to chemical toxins present in the fruit of its host plant, Morinda citrifolia. Its sibling species, D. simulans, and many other Drosophila species do not tolerate these toxins and avoid the fruit. Earlier work found a region with a strong effect on tolerance to the major toxin, octanoic acid, on chromosome arm 3R. Using a novel assay, we narrowed this region to a small span near the centromere containing 18 genes, including three odorant binding proteins. It has been hypothesized that the evolution of host specialization is facilitated by genetic linkage between alleles contributing to host preference and alleles contributing to host usage, such as tolerance to secondary compounds. We tested this hypothesis by measuring the effect of this tolerance locus on host preference behavior. Our data were inconsistent with the linkage hypothesis, as flies bearing this tolerance region showed no increase in preference for media containing M. citrifolia toxins, which D. sechellia prefers. Thus, in contrast to some models for host preference, preference and tolerance are not tightly linked at this locus nor is increased tolerance per se sufficient to change preference. Our data are consistent with the previously proposed model that the evolution of D. sechellia as a M. citrifolia specialist occurred through a stepwise loss of aversion and gain of tolerance to M. citrifolia’s toxins.  相似文献

This study was focused on the analysis of the phenolic content, antioxidant, antibacterial, anti-cholinesterase, anti-tyrosinase, anti-amylase and anti-glucosidase activity of three solvent extracts from Cotoneaster nummularia. Moreover, water extract was tested in terms of mutagenic/anti-mutagenic effects. The antioxidant activities of these extracts were evaluated by DPPH, ABTS, O2, metal chelating, phosphomolybdenum, β-carotene/linoleic acid, ferric and cupric reducing power assays. Enzyme inhibitory activities were also examined with colorimetric methods. Generally, methanol and water extracts exhibited excellent biological activities. These extracts were rich in phenolic and flavonoid content. Furthermore, Cotoneaster extracts indicated appreciable antibacterial properties against human pathogen strains. HPLC analysis showed that ferulic acid, chlorogenic acid, (-) – epicatechin and (+)-catechin were the major phenolics in extracts tested. These data offer that these extracts from C. nummularia may be considered as a potential source of biological agents for developing functional foods or drug formulations.  相似文献

Microorganisms with biocontrol capabilities against plant pathogens are considered as one of the most promising approaches for healthy crop management. In this study, ethyl acetate extracts of 25 Bacillus strains were investigated for their antagonistic effect on Xanthomonas citri subsp. citri (Xcc), which causes the citrus bacterial canker (CBC) disease. Among them, 21 strains exerted antibacterial activity against wild-type Xcc strains. Based on the strength of the antibacterial activity, nine Bacillus strains were selected for 16S rRNA analysis. 16S rRNA sequence homology revealed that several strains were closely related to B. velezensis, where strains with no antibacterial activity grouped as the soil-associated community of B. amyloliquefaciens. B. velezensis Bv-21 exhibited the highest antibacterial activity against wild type and streptomycin resistant Xcc with inhibition zones of 22.91 ± 0.45 and 20.28 ± 0.53, respectively. Furthermore, B. velezensis Bv-21 strain was tested for biocontrol activity against a streptomycin-resistant XccM4 in detached susceptible citrus leaves. The strain reduced the incidence of CBC by 26.30% and pathogen density of XccM4 by 81.68% over control. The results of the study strongly suggest that B. velezensis can be used as an effective and eco-friendly biocontrol agent either by itself or as an active compound, against both, the wild-type and streptomycin-resistant Xcc.  相似文献

The citrus processing industry aims to maintain turbidity and attractive colour of the juice obtained from Sicilian blood oranges. Nevertheless, the presence of pectinesterase (PE, E.C. 3.1.1.11) causes the loss of these peculiar characteristics, due to precipitation of colloids and very fine pieces of pulp in suspension, with negative effects on colour and clarification of the juices. This study compares total PE activity of Sicilian blood oranges (Sanguinello, Moro, Tarocco) with the blonde cultivar Navel, checking enzyme stability with various pasteurisation times and temperatures conditions. Decimal reduction time and temperature (D and z) as well as the kinetic constant (k) were established to optimise and increase the shelf-life of the pasteurised juice. Finally, a heat treatment (85 °C × 3 min) of both microbiological and enzymatic efficacy has been developed that does not compromise anthocyanin stability; this treatment could be used by the citrus fruit processing industry as a valid alternative in the production of blood orange juices.  相似文献

(−)-Epigallocatechin, an antibiotic found in Elaeagnus glabra, and 28 other related plant flavonoids were tested for their antibacterial activity against Proteus vulgaris and Staphylococcus aureus. A free 3′,4′,5′-trihydroxy B-ring and a free 3-OH were necessary for antibacterial activity. Metal ions such as iron can induce DNA damage by inducing reactive oxygen species (ROS) and oxidative stress. Vitamin C is one of the most widely consumed antioxidants worldwide, present in many fruits and vegetables, especially inMalpighia glabra L., popularly known as acerola, native to Brazil. Acerola is considered a functional fruit due to its high antioxidant properties and phenolic contents, and therefore is consumed to prevent diseases or as adjuvant in treatment strategies. Here, the influence of ripe and unripe acerola juices on iron genotoxicity was analyzed in vivo using the comet assay and micronucleus test. The comet assay results showed that acerola juice exerted no genotoxic or antigenotoxic activity. Neither ripe nor unripe acerola juices were mutagenic to animals treated with juices, in micronucleus test. However, when compared to iron group, the pre-treatment with acerola juices exerted antimutagenic activity, decreasing significantly micronucleus mean values in bone marrow. Stage of ripeness did not influence the interaction of acerola compounds with DNA, and both ripe and unripe acerola juices exerted protective effect over DNA damage generated by iron.  相似文献

To reveal the genetic mechanism of host selection in a monophagous fruit flyDrosophila sechellia, olfactory responses and oviposition preferences of this species were compared with those of closely related polyphagous species,D. simulans andD. melanogaster. Adult flies ofD. sechellia were strongly attracted to the ripe fruit ofMorinda citrifolia which is known to be the sole breeding site of this species. They were also attracted to the odor ofn-caproic acid which is contained in the ripe fruit ofM. citrifolia and is presumably responsible for the characteristic odor of the fruit. In contrast,D. simulans andD. melanogaster showed a strong repulsion ton-caproic acid. In parallel with the olfactory responses,D. sechellia females laid eggs preferentially on a medium containingn-caproic acid, to which the other two species showed an aversion. Genetic analyses using the hybrid progeny betweenD. sechellia andD. simulans suggested that the species differences in these behaviors are controlled by gene(s) located on the second chromosome.  相似文献

Alicyclobacillus acidoterrestris is one of the most spoilage-causing bacteria in fruit juices. Control of A. acidoterrestris in fruit juices by bificin C6165 (Pei et al. in J Appl Microbiol 114(5):1273–1284, 2013), a bacteriocin produced by Bifidobacterium animalis subsp. animalis CICC 6165, was described in this study. Activity spectrum of bificin C6165 was investigated and sixteen strains of A. acidoterrestris were sensitive to bificin C6165 in diluted Apple Juices. In the commercial fruit juices, vegetative cells of A. acidoterrestris were inactivated by bificin C6165 at 40 μg/ml. The inhibitory effect of bificin C6165 was better at lower pH (pH 3.5) and at a higher temperature of 45 °C. Furthermore, electron microscopy examination of the vegetative cells treated with bacteriocin revealed substantial cell damage and bacterial lysis. The result suggested that primary mode of action of bificin C6165 was most probably due to pore formation. Although no significantly activity of bificin C6165 was observed against the endospores of A. acidoterrestris in commercial apple juice, the addition of bacteriocin contributed to the reduction of the thermal resistance of A. acidoterrestris spores. Additionally, encapsulation of bificin C6165 with Ca-alginate gel was investigated. Encapsulation of bificin C6165 provided a promising method to control A. acidoterrestris in food juice industry.  相似文献

A growing body of literature reports the importance of non-prey food sources in boosting fitness of arthropod natural enemies, thus further contributing to their pest control efficacy. Although resources such as nectar, pollen, or honeydew have received a fair amount of research attention, little is known about the role of fruit juices. Under natural conditions, Tephritid fruit fly parasitoids enjoy ample access to fallen or damaged fruits and their saccharide-rich juices, and wasp fitness can potentially benefit in multiple ways from access to these resources. In this study, we compared the effect of fruit juice with other food resources on multiple fitness parameters in parasitoids that commonly forage on fallen, damaged fruits: the braconid Diachasmimorpha longicaudata and figitid Aganaspis pelleranoi. Parasitoids were subject to simple or combined diets of guava juice (Psidium guajava), honey and pollen, and their effect on wasp longevity, ovarian dynamics and (body) carbohydrate levels was assessed. For both species, adult longevity proved lowest on simple diets of water, guava juice, or pollen, while greatest longevity was attained on honey or combined diets. For D. longicaudata, egg load did not differ between the various diets, while A. pelleranoi egg load was higher for individuals that had access to honey or pollen, but did not differ between newly emerged wasps and those fed guava juice. In both parasitoid species, total sugars, fructose, and glycogen levels were highest in wasps fed with honey or combined diets and lowest under (simple) guava juice, pollen, or water diets. In conclusion, D. longicaudata and A. pelleranoi attained superior longevity and body nutrient levels with access to high-sucrose sugar sources, such as honey, but benefited comparatively little from access to guava juice. Our work hints the role of high-sucrose foods such as (extra-) floral nectar or artifical sugar sprays in boosting fitness of fruit fly parasitoids. We further discuss the relevance of these findings for fruit fly biological control, in crops such as guava.  相似文献

Plants have always been the prime focus in medicine industries due to their enormous ethnobotanical uses and multitude of biological and therapeutic properties. In the current study, preliminary phytochemical composition, Total phenolic content (TPC), and total flavonoid content (TFC) with the antioxidant and antibacterial activity of hydroalcoholic extract and n-hexane, chloroform and n-butanol fractions of five selected medicinal plants [Tephrosia purpurea (L.) Pers., Lavandula stoechas L., Aesculus indica (Wall. ex Cambess.) Hook, Iris ensata Thunb., and Kalanchoe pinnata (Lam.) Pers.] from Pakistan, have been evaluated. TPC and TFC were determined by Folin-Ciocalteu’s and AlCl3 methods respectively. The antioxidant activity was performed by DPPH, ABTS, FRAP, and CUPRAC while the antibacterial potential of these plants was determined by agar well diffusion assay. K. pinnata (Lam.) Pers. exhibited the highest TPC (695 ± 13.2 mg.GA.Eq.g-1DE ± SD) in n-butanol fraction and the highest TFC in its chloroform faction (615 ± 6.31 mg Q.Eq.g−1 DE ± SD). The n-butanol fraction and hydroalcoholic extract of I. ensata Thunb. exhibited strong antioxidant potential by DPPH and CUPRAC assays respectively, whereas K. pinnata (Lam.) Pers. n-butanol fraction exhibited the strongest reducing potential. The hydroalcoholic extract of all tested plants exhibited significant antibacterial activity against tested bacterial strains with ZI (12–18 mm). Conclusively, K. pinnata (Lam.) Pers. (Family: Crassulaceae) and I. ensataThunb. (Family: Iridaceae) exhibited the highest antioxidant and antibacterial potential. They can be explored for the isolation of phytoconstituents responsible for this potential and serve as a lead for the production of new natural antioxidants and antibacterial agents that can be used to cure various diseases.  相似文献

Fruit juices are targets of spoilage moulds, yeasts and acid tolerant bacteria. They might be contaminated with bacteria from raw materials, environment, packaging and during the handling of the product. These contaminations have frequently resulted in the spoilage of fruit juice and consequently commercial losses. The objective of this study was to determine the influence of propolis in apple juice againstEscherichia coli andE. coli O157:H7 strains of the spoilage and pathogenic bacteria. For this purpose, apple juice was obtained from fresh apples and then was pasteurised. The pH value, titrable acidity (as % malic acid) and Brix degree of this apple juice were 3.72±0.10, 0.67±0.05% and 12.1±0.01, respectively. Propolis extract at 1,2 and 5% concentrations were tested to determine ofE. coli andE. coli O157:H7 inhibition using paper disc diffusion method. The control treatment had no propolis extract. The apple juices were contaminated with these bacteria, and the activity of propolis was observed at first, 18th, 24th, 48th and 72nd hours at 4 and 25°C. The number of cells in the tubes was counted using serial dilution method. Results indicated that propolis extract at 2 and 5% concentrations had significant antimicrobial activity againstE. coli andE. coli O157:H7, therefore we can conclude that propolis extract is worthy of further study as a natural preservative for the foods prone to microbial spoilage.  相似文献

Fruit juices are an important part of the modern diet in many countries. However, few data are available concerning the microbiological quality of the fruit juices sold in Greece. Using standard microbiological procedures, we conducted a bacteriological survey of commercially sold, pasteurized, shelf-stable fruit juices from retail markets. A total of 120 samples of fruit juices sold in various retail markets were examined for their bacteriological quality. The pH of the tested juices was 2.4–4.8. Bacteria were isolated from 51 samples (42.5%) and fungi from 78 samples (65%). Escherichia coli O157:H7 was detected in four of the analyzed samples (3.34%), and Staphylococcus aureus was detected in four different samples (3.34%). In 11 samples (9.1%), the total number of microorganisms detected was as high as 125 colony-forming units (CFU). Acidophilic microorganisms were isolated from 26 samples (21.7%) and Blastomyces was detected in 46 samples (38.3%). All samples were negative for Lactobacillus, Clostridium perfrigens, Salmonella spp., Bacillus cereus, total coliforms, E. coli, and Listeria monocytogenes.Many of the microorganisms detected may cause disease in humans; thus, a number of the tested samples did not meet the Greek guidelines for the microbiological quality of juices. Use of a Hazard Analysis Critical Control Point (HACCP) system should be generally introduced into the juice industry sector to improve the quality of fruit juices, as well as other manufactured foods.  相似文献

Amoxycillin (α-amino-p-hydroxybenzylpenicillin) is a new semi-synthetic penicillin with a broad spectrum of antibacterial activity similar to that of ampicillin. Penicillin-sensitive strains of staphylococci, streptococci, and pneumococci were sensitive to concentrations of 0·1 μg or less of amoxycillin/ml. Strains of Haemophilus influenzae were inhibited by a level of 0·5 μg/ml, and most strains of Escherichia coli, Proteus mirabilis, Shigella sonnei, Salmonella species, and Streptococcus faecalis were sensitive to a concentration of 5 μg or less of amoxycillin/ml. Penicillinase-producing strains of Staphylococcus aureus and strains of Pseudomonas aeruginosa, indole-positive Proteus, Klebsiella, and Enterobacter were insensitive to amoxycillin. The new penicillin was bactericidal in activity, as with other penicillins, and its antibacterial activity was not reduced in the presence of serum. After oral administration to volunteer subjects amoxycillin produced serum concentrations twice as high as those obtained with similar doses of ampicillin, and the penicillin was recovered unchanged in high concentrations in the urine. The absorption of amoxycillin was not greatly influenced by food, and administration of probenecid resulted in increased and more prolonged concentrations of amoxycillin in serum.  相似文献

Two hundred and nineteen bacterial strains were isolated from cow dung. Among these, 59 isolates displayed nematicidal activity against the model nematode Caenorhabditis elegans. Of the 59 bacterial strains, 17 killed >90 % of the tested nematodes within 1 h. Based on their 16S rRNA sequences, these 17 strains were identified as Alcaligenes faecalis, Bacillus cereus, Proteus penneri, Providencia rettgeri, Pseudomonas aeruginosa, Pseudomonas otitidis, Staphylococcus sciuri, Staphylococcus xylosus, Microbacterium aerolatum, Pseudomonas beteli. Among these 17 strains, 14 produced volatile organic compound(s) that inhibited the mobility of the C. elegans nematodes. These 14 strains also showed nematicidal activity against a plant pathogenic nematode Meloidogyne incognita. This is the first report demonstrating nematicidal activity for strains in genera Proteus, Providencia and Staphylococcus.  相似文献
