Despite the tremendous progress mode in the understanding o f the molecular biology of Leishmania and the clinical possibilities presented by some experimental chemotherapeutic agents, no new drugs have been developed for the treatment of leishmaniasis since the introduction of the pentovalent antimoniols more than 50 years ago. As reviewed here by Maurice M. Iwu, Joan E. Jackson and Brion G. Schuster, recognition of the current extensive use of herbal therapy in Leishmania-endemic regions has renewed interest in evaluation of plant remedies used in traditional medicine as sources of potential antileishmanials.  相似文献

Many types of action can be taken in favour of the conservation and sustainable use of medicinal plants. Some of these are undertaken directly at the places where the plants are found, while others are less direct, such as some of those relating to commercial systems, ex situ conservation and bioprospecting. In the latter cases, actions taken will not lead to in situ conservation unless they feedback to improvements in the field. Probably the single most important role for medicinal plants in biological conservation is their use to achieve conservation of natural habitats more generally. This stems from the special meanings that medicinal plants have to people, related to the major contributions that they make to many people's lives in terms of health support, financial income, cultural identity and livelihood security. Problems associated with biopiracy or (the other side of the coin) excessive restrictions on research have come to assume policy prominence in the general thematic area of medicinal plant conservation and use. The fair and equitable sharing of benefits from bioprospecting is required under the Convention on Biological Diversity, but it is not always easy to achieve these ideals in practice. While experience is accumulated in how this may practically be achieved, it is important, at the present time, that controls imposed on scientific research to prevent biopiracy or theft of local and indigenous intellectual property do not unduly restrict research that has little or nothing to do with these matters.  相似文献

Metabolomics uses high-resolution mass spectrometry to provide a chemical fingerprint of thousands of metabolites present in cells, tissues or body fluids. Such metabolic phenotyping has been successfully used to study various biologic processes and disease states. High-resolution metabolomics can shed new light on the intricacies of host-parasite interactions in each stage of the Plasmodium life cycle and the downstream ramifications on the host’s metabolism, pathogenesis and disease. Such data can become integrated with other large datasets generated using top-down systems biology approaches and be utilised by computational biologists to develop and enhance models of malaria pathogenesis relevant for identifying new drug targets or intervention strategies. Here, we focus on the promise of metabolomics to complement systems biology approaches in the quest for novel interventions in the fight against malaria. We introduce the Malaria Host-Pathogen Interaction Center (MaHPIC), a new systems biology research coalition. A primary goal of the MaHPIC is to generate systems biology datasets relating to human and non-human primate (NHP) malaria parasites and their hosts making these openly available from an online relational database. Metabolomic data from NHP infections and clinical malaria infections from around the world will comprise a unique global resource.  相似文献

We have compared the species richness of medicinal plants and the differential patterns of use amongst settlements in the Andean communities of Northwest Argentina which have differing levels of isolation. About 259 ethnoespecies, belonging to 74 plant families, were included, representing between 70 and 80% of the total estimate. The results indicate that Coronopus didymus is the most relevant and important species. The method of use of medicinal plants and the ailments treated by rural doctors compared to those of the layperson is different. Native and exotic plants are used differently according to the body system treated. There are some relationships between internal and external use and body systems and recipes. The greater medicinal species richness found in the less isolated locations is due to external enriching cultural influences.  相似文献

Trypanosoma evansi is a hazardous pathogenic parasite infecting a broad variety of livestock and affects wildlife worldwide. Trypanosoma evansi has gained resistance to most drugs used; therefore, it requires alternative medicines. The objective of this research was to investigate the impact of Indigofera oblongifolia leaf extract (IE) on T. evansi-induced hepatic injury.Mice were once infected with 1000 T. evansi. The treated group was gavaged with 100 mg/Kg IE after infection. Histological and biochemical changes in mice hepatic tissue were studied. Also, the oxidative damage in the liver was evaluated through determining the level of glutathione (GSH), Malondialdehyde (MDA), nitric oxide (NO) and catalase (CAT) markers. IE was able to suppress the induced parasitemia due to infection. Also, IE improved the histological liver architecture. Furthermore, the liver enzymes, alanine aminotransferase (ALT), aspartate aminotransferase (AST), and alkaline phosphatase (ALP) activity were improved after IE mice were treated. IE protects against hepatic damage caused by trypanosomiasis in mice. Further studies are needed to isolate the active compounds in IE and to monitor these compunds’ ameliorative function.  相似文献

Present study was intended with the aim to document the pre-existence traditional knowledge and ethnomedicinal uses of plant species in the Palas valley. Data were collected during 2015–2016 to explore plants resource, their utilization and documentation of the indigenous knowledge. The current study reported a total of 65 medicinal plant species of 57 genera belonging to 40 families. Among 65 species, the leading parts were leaves (15) followed by fruits (12), stem (6) and berries (1), medicinally significant while, 13 plant species are medicinally important for rhizome, 4 for root, 4 for seed, 4 for bark and 1 each for resin. Similarly, thirteen species were used as a whole while twelve species as partial for medicinal purpose. Further, it is concluded that every part of plants such as bulb, rhizome, roots, barks, leaves, flowers, fruit and seed were used for various ailments. Moreover, among 65 plants species, 09 species are threatened and placed into Endangered (EN) and Least Concern (LC) categories of IUCN. The recorded data are very useful and reflects the significance of the Palas valley as medicinal plants resource area.  相似文献

It has been proposed that the basis of severe malarial anaemia, a major cause of morbidity and mortality in endemic areas, is multifactorial. Inappropriately low reticulocytosis is observed in malaria patients suggesting that insufficient erythropoiesis is a major factor. Clinical studies provide conflicting data concerning the production of adequate levels of erythropoietin (EPO) during malaria. Plasmodium chabaudi AS causes non-lethal infection in resistant C57BL/6 mice, and lethal infection in susceptible A/J mice. In P. chabaudi AS infected C57BL/6 and A/J mice, which experience varying degrees of severity of anaemia, kidney EPO production is appropriate to the severity of anaemia and is regulated by haematocrit level. Neutralisation of endogenous EPO during infection leads to lethal anaemia while timely administration of exogenous EPO rescues mice although reticulocytosis is suppressed in proportion to the parasitemia level. Characterisation of alterations in splenic erythroid compartments in naive and P. chabaudi AS infected A/J mice revealed that infection, with or without EPO treatment, leads to sub-optimal increases in TER119+ erythroblasts compared to EPO-treated naive mice. A lower percentage of TER119+ erythroblasts in infected mice undergo terminal differentiation to become mature haemoglobin-producing cells. Furthermore, there is a shift in transferrin receptor (CD71) expression from TER119+ cells to a non-erythroid population. Deficiencies in the number and maturation of TER119+ erythroblasts during infection coincide with blunted proliferation to EPO stimulation in vitro by splenocytes, although a high frequency express EPO receptor (EPOR). Together, these data suggest that during malaria, EPO-induced proliferation of early EPOR+ erythroid progenitors is suppressed, leading to sub-optimal generation of TER119+ erythroblasts. Moreover, a shift in CD71 expression may result in impaired terminal maturation of erythroblasts. Thus, suppressed proliferation, differentiation, and maturation of erythroid precursors in association with inadequate reticulocytosis may be the basis of insufficient erythropoiesis during malaria.  相似文献

A possible protective role of IL-18 in host defense against blood-stage murine malarial infection was studied in BALB/c mice using a nonlethal strain, Plasmodium yoelii 265, and a lethal strain, Plasmodium berghei ANKA. Infection induced an increase in mRNA expression of IL-18, IL-12p40, IFN-gamma, and TNF-alpha in the case of P. yoelii 265 and an increase of IL-18, IL-12p40, and IFN-gamma in the case of P. berghei ANKA. The timing of mRNA expression of IL-18 in both cases was consistent with a role in the induction of IFN-gamma protein expression. Histological examination of spleen and liver tissues from infected controls treated with PBS showed poor cellular inflammatory reaction, massive necrosis, a large number of infected parasitized RBCs, and severe deposition of hemozoin pigment. In contrast, IL-18-treated infected mice showed massive infiltration of inflammatory cells consisting of mononuclear cells and Kupffer cells, decreased necrosis, and decreased deposition of the pigment hemozoin. Treatment with rIL-18 increased serum IFN-gamma levels in mice infected with both parasites, delayed onset of parasitemia, conferred a protective effect, and thus increased survival rate of infected mice. Administration of neutralizing anti-IL-18 Ab exacerbated infection, impaired host resistance and shortened the mean survival of mice infected with P. berghei ANKA. Furthermore, IL-18 knockout mice were more susceptible to P. berghei ANKA than were wild-type C57BL/6 mice. These data suggest that IL-18 plays a protective role in host defense by enhancing IFN-gamma production during blood-stage infection by murine malaria.  相似文献

People use medicinal plants as diet, and for treatment of infectious and noninfectious diseases and they use brief procedures like frying and cooking to do so. Medicinal plants; Moringa oleifera, Azadirachta indica, and Lepidium sativum which is believed to have active components that help to treat and manage various diseases were investigated for their antibacterial activities against Staphylococcus aureus, Salmonella Typhi, Streptococcus agalactiae and Shigella boydii. Solvent methanol and aqueous were used for extraction of crudes by means of maceration. Susceptibility testing was determined by using disc diffusion method and Minimum inhibitory concentration was determined by broth dilution method. Heat treated plant material activity against test pathogen was aimed to identify resistance capacity of plant material at different interval of time and temperature. All plant extracts under study was active against all tested pathogen after exposure to 45 °C for 30 min. The antibacterial activities of the non-heat-treated extracts of Azadirachta indica were relatively low; the results of this study show that extracts of Azadirachta indica have better residual antibacterial activities. Methanol extracts of all plant leaves showed highest activity as compared to the aqueous extracts. This is probably assigning the choice of extraction solvent for extracting desired active phytochemical from plants. Many of the people in the study area were illiterate and they did not have awareness about the ways use of medicinal plants. They use the medicinal plants by cooking and frying for different purposes. In the main, plant material can be affected as the temperature of treatment is increases with respect to various times of exposures.  相似文献

Malaria is the major life threatening parasitic disease and the cause of a global public health problem. The failure of vector eradication programs and the appearance and spread of drug resistant parasites have posed the urgent challenge of developing effective, safe and affordable anti-malarial drugs. The design of such drugs is largely based on the targeting of agents to the parasite-based machinery for host digestion and to the products of hemoglobin catabolism. Iron chelators, by depriving intracellular parasites from essential iron, lead to selective suppression of parasite growth. However, by acting on parasite-impaired macrophages, chelators can also expedite resumption of phagocytosis and elimination of parasites. In order to be clinically effective, chelators need to be maintained in the blood for extensive time periods. Therapeutic doses can be attained with appropriate drug combinations and formulations or delivery devices and these must be presented in a form well tolerated by the host. The early documentation that chelation therapy has activity against human malaria has paved the road for the design of novel and more efficient remedies based on short-term iron deprivation.  相似文献

Antimalarial treatment strategies based on in vitro studies are limited by the paucity of pharmacodynamic information for dosage regimen design. We postulated that a murine model could be used for pre-clinical stages of drug development, especially in dose–response studies and evaluation of combination therapies. Swiss mice infected with Plasmodium berghei parasites (2–5% starting parasitaemia) were given dihydroartemisinin (0–100 mg/kg single dose). Parasite density was regularly determined from thin blood films. A parasite population growth model comprising parasite multiplication, decline in erythrocyte count with increasing parasitaemia and parasite clearance after drug administration was developed. This model described the rise in parasitaemia following inoculation, the nadir following dihydroartemisinin administration, and the subsequent resurgence of parasitaemia (analogous to ‘recrudescence’). At doses of 10, 30 and 100 mg/kg dihydroartemisinin, there was a graded response with 2.5 ± 1, 5 ± 1 and 12 ± 4-fold decreases in parasitaemia, respectively. The nadir parasitaemia (at 21–27 h) was also dose-dependent. This study demonstrates that a murine malaria pharmacodynamic model is a valuable tool for understanding how single drugs and their dosing schedules alter the time course and level of infection.  相似文献

8种贵州药用植物内生放线菌的分离及多样性研究   总被引:1,自引:0,他引:1  
【背景】药用植物内生菌能产生多种活性较好的次级代谢产物,是新抗生素发现的重要来源。目前,药用植物内生菌的研究主要集中在内生真菌,内生放线菌研究相对较少,是一个尚未充分开发的新领域。【目的】研究贵州药用植物内生放线菌多样性,以期发现新种放线菌或产新活性物质的放线菌,为微生物药物研究奠定基础。【方法】药用植物样品表面消毒后用粉碎机粉碎,采用9种不同分离培养基分离放线菌;通过提取放线菌基因组DNA,PCR扩增16S rRNA基因序列,测定序列并提交EzBioCloud数据库进行序列比对,构建系统进化树,进行内生放线菌多样性分析。【结果】通过形态特征初步排重,从8种药用植物样品中共分离得到内生菌62株,测定其16S rRNA基因序列,序列比对结果表明其中的57株菌分布于放线菌域的5目8科16属,分别是短小杆菌属(Curtobacterium)、链霉菌属(Streptomyces)、微杆菌属(Microbacterium)、纤维单胞菌属(Cellulomonas)、植物杆菌属(Plantibacter)、原小单孢菌属(Promicromonospora)、动球菌属(Kineococcus)、壤球菌属(Agrococcus)、沼杆菌属(Patulibacter)、气微菌属(Aeromicrobium)、拉贝达氏菌属(Labedella)、叶居菌属(Frondihabitans)、冷杆菌属(Frigoribacterium)、节杆菌属(Arthrobacter)、动孢菌属(Kineosporia)、Amnibacterium属,其中短小杆菌属(Curtobacterium)为优势菌属,菌株M8JJ-5和M2KJ-4的16S rRNA基因序列分别与有效发表菌株AmnibacteriumkyonggienseKSL51201-037T(FJ527819)和Aeromicrobiumfastidiosum DSM10552T (Z78209)的相似性最高,相似性分别为97.29%和98.95%,可能为潜在新物种。【结论】贵州药用植物中蕴藏着多样性丰富的放线菌且具有从中发现新放线菌的潜力,能为新抗生素的发现提供菌种储备,也为今后对该地区放线菌资源的认识和开发利用提供理论依据。  相似文献

本文研究海南兴隆地区12种不同类型药用植物微量元素的生物吸收系数(Ax)及其含量水平变化特性,结果表明不同科、属、种植物对同一元素的吸收量差异显著,以Mg>Al>Fe>Mn>Zn>Cu为序;同属不同品种植物在不同生境中对微量元素的生物吸收系数各有差别,海南槟榔(Ax 0.30×10-6、12.51×10-6)>泰国槟榔(Ax 0.29×10-6、11.59×10-6)>云南槟榔(Ax 0.27×10-6、11.56×10-6),元素间吸收量相比以Mo>Zn>Cu>Mn>Ni为例;同种植物不同器官微量元素的吸收量也是不同的,例如巴戟天的含量以根(218.48×10-6)>叶(115.19×10-6)>茎(113.02×10-6)。植物与土壤中元素含量消长变化关系密切,相比之下,植物中Fe、Mn、Zn含量均高于Cu、Pb、Ni。绞股蓝(189.26)>巴戟天(169.04)>益智(161.70)>槟榔(155.88)。  相似文献

峨眉山素有“天然药园”之美称,据初步统计共有野生药用植物1400余种(含变种、变型)。但由于各种原因,截止到21世纪初,峨眉山有17种药用植物被列入中国生物多样性研究报告濒危植物名录中;有4种原分布较少、药用价值较高的种群已渺无踪迹;另有149种也已难见芳容。为引起多方重视,共同保护峨眉山这一“天然药园”笔者特将初步考察研究结果报道于后,以求得更有效的保护办法,使“天然药园”能更好的造福子孙、造福人类。  相似文献

BackgroundHowever, broad adoption of herbal remedies for giardiasis is at present hampered by uncertain findings of investigation not always sufficiently powered. This study was aimed at systematically reviewing the existing literature in herbal medicines to treat giardiasis.MethodsThis review was carried out 06- PRISMA guideline and registered in the CAMARADES-NC3Rs Preclinical Systematic Review and Meta-Analysis Facility (SyRF) database. The search was performed in five databases which are Scopus, PubMed, Web of Science, EMBASE, and Google Scholar without time limitation for all published articles (in vitro, in vivo, and clinical studies). The searched words and terms were: “Giardia”, “giardiasis”, “extract”, “essential oil”, “herbal medicines”, “anti-Giardia”, “In vitro”, “In vivo”, “clinical trial” etc.ResultsOut of 1585 papers, 40 papers including 28 in vitro (70.0%), 7 in vivo (17.5%), 2 in vitro/ in vivo (5.0%), and 3 clinical trials (7.5%) up to 2020, met the inclusion criteria for discussion in this systematic review. The most widely used medicinal plants against Giardia infection belong to the family Lamiaceae (30.0%) followed by Asteraceae (13.5%), Apiaceae (10.5%). The most common parts used in the studies were aerial parts (45.0%) followed by leaves (27.4%) and seeds (7.5%). The aqueous extract (30.0%), essential oil (25.4%) and hydroalcholic and methanolic (10.5%) were considered as the desired approaches of herbal extraction, respectively.ConclusionThe current review showed that the plant-based anti-Giardia agents are very promising as an alternative and complementary resource for treating giardiasis since had low significant toxicity. However, more studies are required to elucidate this conclusion, especially in clinical systems.  相似文献
