Summary Pistils ofTrifolium repens L. andT. ambiguum Bieb. were cultured on an agar-based modified Murashige-Skoog medium. Pistils with and without accessory floral parts were removed from flowers of selected clones ofT. repens, hand-pollinated under aseptic conditions, and planted on the medium. Pistils cultured without accessory floral parts showed no evidence of fertilization after 2 weeks. However, 52% of thoseT. repens pistils cultured with calyx lobes and pedicels contained ovules with maturing embryos 12 days after in vitro cross-pollination. Pistils fromT. ambiguum intraspecific cross-pollinations could not be cultured successfully under the same conditions; however, addition of various combinations of auxin, cytokinin, and gibberellic acid enhanced embryo growth. Fertilization and partial embryo development occurred in interspecific crosses betweenT. ambiguum andT. repens orT. hybridum only whenT. ambiguum was used as the pistillate parent. These results indicate that embryological development under in vitro conditions closely parallels in situ development although growth regulator requirements may vary among species. Summary An analysis of phenotypic and genetic variability was performed on Phleum ambiguum populations obtained by vegetative propagation and grown in different environments. The investigation on a single character and the canonical analysis on several plant traits indicate that, while genetic variability (h2) is essentially constant, phenotypic variance and covariance are affected by different environments and successive clonations. The observed changes seem not to be reversible when plants, transferred to a different environment, are returned to the original one. Within-clones variability seems to be affected by environmental conditions without being directly related to them. The results reported seem to be in good agreement with the phenotypic divergence hypothesis.  相似文献

Trifolium ambiguum M. Bieb and T. repens L. are taxonomically related but very difficult to cross. The rare hybrids so far reported between these two species were obtained only by embryo culture. This difficulty has been overcome in the present research by the creation of a “fertile bridge” between T. ambiguum and T. repens. Characters of interest can now be transferred from T. ambiguum to T. repens by using this “fertile bridge” without the use of sophisticated techniques. An array of backcross progenies was generated from crosses between a T. ambiguum×T. repens F1 hybrid (8x H-435) and its parental species. The 8x hybrid was cross-fertile only with T. repens and resulted in 145 seeds from 1578 reciprocal crosses. Eleven of nineteen initially grown BC1F1 plants were all hexaploid with an average pollen stainability of 41.6%. A high frequency of multivalents at metaphase-I indicated that both autosyndetic and allosyndetic pairing occurred. Backcrosses of 6x BC1F1 plants to T. repens resulted in 5x BC2F1 plants with an average pollen stainability of 59.3%. On the other hand, 6x BC1F1×6x T. ambiguum crosses did not produce any seed and only two pentaploid plants were obtained from 6x BC1F1×4x T. ambiguum crosses. The difficulty encountered in generating 6x backcross progeny with 6x T. ambiguum was overcome by intercrossing the 6x BC1F1 plants and producing 6x BC1F2 plants with an average pollen stainability of 65.8%. One of these 6x BC1F2 plants was cross-compatible as a female with 6x T. ambiguum and resulted in CBC2 plants that were all cross-compatible with 6x T. ambiguum. The 6x BC1F2 plants are likely to be superior to 6x BC1F1 progeny, as they have exhibited better expression of the combined rhizomatous and stoloniferous growth habit, improved fertility, more frequent nodal rooting and heavier nodulation. Consequently, the 6x BC1F2 plants can either be used directly in the selection programme or as a “fertile bridge” between the two parental species. The present work has resulted in the development of a series of fertile hybrids by the manipulation of chromosome numbers, combining the agronomic characteristics of the parent species in varying genome balances and at a range of ploidy levels. Several species of ciliates produce and accumulate low molecular weight toxic compounds in specialised membrane-bound ejectable organelles: extrusomes. These compounds can be used in predator–prey interaction for killing prey as well as for chemical defence. Here, we describe the isolation and characterisation of 2-(3-methylbut-2-enyl)benzene-1,4-diol(mono-prenyl hydroquinone), the extrusomal defensive toxin of the freshwater heterotrich ciliate Spirostomum ambiguum. The toxin was purified at homogeneity by RP-HPLC, and its structural characterisation was carried out through NMR and MS measurements. In vivo experiments involving S. ambiguum and Climacostomum virens in predator–prey interaction, and the analysis of cytotoxic activity of mono-prenyl hydroquinone on a panel of free-living freshwater ciliates, indicated that the toxin is very effective in S. ambiguum’s chemical defence.  相似文献

Trifolium ambiguum M.B. is a potentially important clover species,but it has not been used widely as a forage legume because itnodulates sparsely or not at all and the nodules formed areusually ineffective. In some compatible grafts of T. hybridumon T. ambiguum effective nodules developed on the roots of thestock. In addition, effective nodulation was obtained on thesexual hybrid between these two species. Summary Previous experiments have revealed that the maize transposable element Activator (Ac) may become active during tissue culture. The objective of the present study was to determine whether a second transposable element, Suppressor-mutator (Spm), could also be activated in tissue culture and detected in regenerated maize plants. Approximately 500 R1 progeny of 143 regenerated plants (derived from 49 embryo cell lines) were crossed as males onto an Spm-responsive tester stock. Spm activity was observed in two R1 progeny of a single regenerated plant. This plant had been regenerated from Type II (friable embryogenic) callus of an A188 × B73 genetic background after 8 months in culture; the absence of Spm activity in four other plants regenerated from this same callus demonstrates that Spm activity was not present before culturing. Approximately 20 Spm-homologous DNA sequences were detected in each of the inbreds used to initiate the tissue cultures; it is presumed that one of these became active to give rise to Spm activity.  相似文献

对根霉(Rhizopus)属的十个种或变种共二十四株菌的菌体可溶性蛋白和酯酶同工酶进行了电泳的研究得到对这个属分类上更多的依据.在严格控制培养,提取和电泳条件的情况下,同一株菌不同批次所得菌体蛋白电泳图谱有较好的重复性.在相同的条件下,每个种的根霉有各自特征性蛋白图谱,种内不同菌株的蛋白图谱和酯酶酶谱基本相同.特别是形态特征明显、分类地位明确的种,种内各株的图谱也较一致,如R. stolonifer;与R.circinans.在确定新变种R. delemar var. latoapicalis时,电泳图谱与R.delemar var.delema:有明显不同,起到了佐证作用.因此认为,蛋白图谱与酯酶酶谱相辅相成,在根霉种的分类中是一有效的辅助手段.  相似文献

The influence of various growth regulators, in different quantities, and the physiological and biological stage of the plant, on the in vitro propagation of myrtle (Myrtus communis L.) were evaluated. It was found that the proliferation was dependent on both the medium and the period in which the sampling was performed. The highest rate of in vitro shoot proliferation was obtained by using 6-benzyladenine and α-naphthyleneacetic acid as growth regulators, starting from nodes sampled at the beginning of May. Rooting was achieved by either transplanting the shoots directly into soil or by culturing on a medium containing 1 mg 1−1 of indole-3-acetic acid. Selected strains of Chondrus crispus were acclimated to relativelywarm Pacific waters off the Baja California peninsula, Mexico, and werecultured in semi-exposed oceanic conditions. Specific techniques weredeveloped for culturing this species in semi-exposed oceanic environment,in an effort to establish the basis for its commercial farming. This is the firsttime that Chondrus crispus has been cultured in the ocean and outsideits natural Atlantic environment. The information generated in this studyis being used to feed an economic model to test the feasibility of thecommercial culturing of Chondrus crispus.  相似文献

Summary In vitro rooted grape (Vitis vinifera L.) plantlets (4 or 8 wk from culturing microcuttings) were plantedex vitro in polythene sachets (24×12 cm) filled to one-third their height with planting mixture. The sachets were misted, closed, and incubated at ambient temperature (25–30°C) under 16 h photoperiod (40–50 μ E·m−2.·sec−1) for 1,2, or 3 wk before opening. Maximum establishment with no or minimum damage toin vitro formed leaves was obtained with 3 wk of closed sachet incubation in both age groups. Opening the sachet at 2 wk from planting resulted in marginal scorching of lower leaves and some reduction in establishment and vigor. Opening at 1 wk led to severe leaf scorching and significant reduction in establisment particularly in 4-wk-oldin vitro plantlets while growth was more affected in 8-wk-old ones. Relative humidity ofin vitro culture vessel was 68–75% while closed sachets had RH values of 45–77% depending on length of incubation and ambient RH. Diurnal variation in RH of sachet in relation to ambient RH was the major factor that facilitated acclimatization rather than the overall fall in RH during the period of closed incubation. Satisfactory acclimatization of plantlets to withstand the open sachet RH (50–55%) by 3 wk and the ambient RH (30–40%) by 4 wk was achieved. Monitoring water loss from detached leaves of plantlets showed a significant reduction between the date of planting and Week 3, and again between Weeks 3 and 4. Comparison of growthin vitro andex vitro suggested that shifting toex vitro earlier was more beneficial. This observation was confirmed by transferring 3-, 4-, and 5-wk-old plantlets fromin vitro rooting medium toex vitro and recording the growth at 8 wk fromin vitro culturing when 3 wkin vitro plus 5 wkex vitro combination showed maximum vigor. The leftover stumps after subculturing of 1–4-mo.-old stock cultures could also be effectively used forex vitro establishment.  相似文献

The mesophyll protoplasts were isolated from the Solanum tuberosum (S. tbr) clones of different ploidy level (4x Bzura cv., 2x H-8105, and 2x ZEL-1136) as well as from the wild species: S. bulbocastanum (S. blb, 2x) and two accessions of S. nigrum (S. ngr, 6x). Additionally, the protoplasts were isolated from the cell suspensions of Bzura cv. and H-8105 clone. The conditions of protoplast isolation as well as the media for their culturing and regeneration, were selected and optimized for the studied genotypes. For mesophyll protoplasts, the shooting calli were produced by all the cultured protoclones except that of S. bulbocastanum. The shoots excised from the protoplast-derived calli developed into whole plants in all the studied potato clones but only in one accession of S. nigrum, i.e. S. ngr var. gigantea. As for suspension-cell-derived protoplasts, only H-8105 clone produced the regenerative type of calli, though normal shoots could not be obtained. The regenerative capacity of the protoplasts isolated from leaves and cell suspensions is compared and discussed. We regret to report the death of M. Sc. A study was made of environmental factors affecting the growth of Rhizoclonium riparium in order to evaluate its suitability for large-scale culturing. The results indicate that under the natural conditions prevailing at Taishi, Taiwan, this species can grow year-round, with a monthly biomass production (oven-dried) of 945–1540 kg ha−1 pond surface (assuming a pond depth of 1 m). The specific growth rate ranged from –2.1 to 10.4% per day. Salinity and temperature, both influenced the rate significantly, with optimal values at 20% and 25 °C, respectively. Short (2-mm) lengths of filaments had a higher specific growth rate than longer (20 mm) filaments. Under rotational culturing conditions, the specific growth rate was reduced when flow was increased.  相似文献

Homografting of Arabidopsis thaliana scions on stocks of A. thaliana and heterografting on other species were used to study the compatibility and the ontogeny of graft union formation. Highly compatible homografting with scions of young leafy inflorescence stems was obtained on stocks of inflorescence stems growing from large 3-month-old A. thaliana plants. Histologic analysis revealed four developmental stages of graft union formation in Arabidopsis homografting: (1) development of a necrotic layer, (2) callus proliferation in the grafted scion, (3) differentiation of new vascular tissues within the scion, and (4) a full vascular graft union formation between the scion and the stock. Vascular connections were formed within the callus bridge between rootstocks and scions 15 days after grafting. Heterografts of Arabidopsis on two members of Brassicaceae, cabbage (Brassica) and radish (Raphanus), showed partial incompatible interaction with a lower level of vascular differentiation. Arabidopsis grafting on tomato (Solanaceae) rootstock showed complete incompatibility and limited noncontinuous differentiation of new vascular tissues that did not cross the scion/stock boundary. Although lacking scion/stock vascular connections, Arabidopsis scions grafted onto tomato rootstock flowered and produced seeds. This may indicate some nonvascular functional connections between the two plants, probably of parenchyma cells, further emphasizing the usefulness of Arabidopsis as a model plant for studying various levels of the complicated scion/stock relationships expressed in grafting biology. Experiments with dye transport in the xylem showed that although in general there was an agreement between the histologic study and dye transport, in Arabidopsis homografts water transport frequency was lower than functional and histologic compatability. We conclude that homografting and heterografting of Arabidopsis inflorescence stems is a convenient and reproducible method for studying the fundamental cellular genetic and molecular aspects of grafting biology.  相似文献

Stark spectroscopy is a powerful technique to investigate the electrostatic interactions between pigments as well as between the pigments and the proteins in photosynthetic pigment–protein complexes. In this study, Stark spectroscopy has been used to determine two nonlinear optical parameters (polarizability change Tr(Δα) and static dipole-moment change |Δμ| upon photoexcitation) of isolated and of reconstituted LH1 complexes from the purple photosynthetic bacterium, Rhodospirillum (Rs.) rubrum. The integral LH1 complex was prepared from Rs. rubrum S1, while the reconstituted complex was assembled by addition of purified carotenoid (all-trans-spirilloxanthin) to the monomeric subunit of LH1 from Rs. rubrum S1. The reconstituted LH1 complex has its Qy absorption maximum at 878 nm. This is shifted to the blue by 3 nm in comparison to the isolated LH1 complex. The energy transfer efficiency from carotenoid to bacteriochlorophyll a (BChl a), which was determined by fluorescence excitation spectroscopy of the reconstituted LH1 complex, is increased to 40%, while the efficiency in the isolated LH1 complex is only 28%. Based on the differences in the values of Tr(Δα) and |Δμ|, between these two preparations, we can calculate the change in the electric field around the BChl a molecules in the two situations to be E Δ ≈ 3.4 × 105 [V/cm]. This change can explain the 3 nm wavelength shift of the Qy absorption band in the reconstituted LH1 complex.  相似文献

Cryptococcus gattii (Cg) is an emerging pathogen of both healthy and immunocompromised patients worldwide. Understanding the molecular genetic basis of virulence and physiology of this pathogen will be critical for defining its pathogenic mechanisms. The purine biosynthetic gene, URA5 encoding orate phosphorybosyltransferase (OPRTase), has been successfully used as a selectable marker for gene disruption by transformation and homologous recombination in Cg. Here, we report the characterization of ura5 auxotrophy and URA5 reversion phenomenon at the molecular, genetic, and structural levels, and use of ura5URA5 reversion as a tool for reconstitution of gene of interest and auxotrophic marker to their native loci. We identified a single mutation of GG128T→GAT with substitution of glycine to aspartic acid at amino acid position 43 resulting in ura5 auxotrophy. The ura5URA5 reversion on CSM lacking uracil (CSM-U) was found to be a rare phenomenon with a reversion frequency of 0.000002%, and sequence analysis of URA5 from all the reverted strains revealed mutation of GA128T→GGT back to its ancestral state. The URA5 allele in the reverted strains was fully functional, as demonstrated by the excellent growth of these strains on medium lacking uracil, as well as by the ability of this allele to efficiently transform ura5 mutant to restore prototrophy. The deduced Cg URA5 protein modeled on the known crystal structures of OPRTase from Salmonella typhimurium (1LH0_A, 1STO) and from Escherichia coli (1ORO_A) indicated that the glycine 43 of Cg URA5 was situated on a conserved loop, and it’s substitution to more globose aspartic acid may have resulted in URA5 inactivation in auxotrophic strain. The advantages of this approach for the generation of a reconstituted strain are (1) that it restores the functionality of the native URA5, (2) that it eliminates an additional biolistic delivery of exogenous URA5, and (3) that it allows easy selection of reconstituted strains with homologous integration. This strategy was successfully used for the generation of Cg can2+CAN2/URA5 homologous reconstituted strains, which grew in ambient air to the wild-type level while can2 mutant exhibited severe growth defect under similar conditions. Monoxenic symbioses between the arbuscular mycorrhizal (AM) fungus Glomus intraradices and two nontransformed tomato root organ cultures (ROCs) were established. Wild-type tomato ROC from cultivar “RioGrande 76R” was employed as a control for mycorrhizal colonization and compared with its mutant line (rmc), which exhibits a highly reduced mycorrhizal colonization (rmc) phenotype. Structural features of the two root lines were similar when grown either in soil or under in vitro conditions, indicating that neither monoxenic culturing nor the rmc mutation affected root development or behavior. Colonization by G. intraradices in monoxenic culture of the wild-type line was low (<10%) but supported extensive development of extraradical mycelium, branched absorbing structures, and spores. The reduced colonization of rmc under monoxenic conditions (0.6%) was similar to that observed previously in soil. Extraradical development of runner hyphae was low and proportional to internal colonization. Few spores were produced. These results might suggest that carbon transfer may be modified in the rmc mutant. Our results support the usefulness of monoxenically obtained mycorrhizas for investigation of AM colonization and intraradical symbiotic functioning.  相似文献

Cell carbon and nitrogen in D. viridis are strongly dependent on the culturing conditions. Both elements increase with increasing salinity. At 31°C cell carbon is maximum and cell nitrogen minimum. This temperature was described previously (Jiménez, C., Niell, F. X. & Fernandez, J. A. (1990). Hydrobiologia, 197, 165-72) as the optimal one for achieving the maximum oxygen evolution. These results point out a possible competence for the reducing power during carbon and nitrogen assimilation processes, and under conditions of high photosynthesis (carbon assimilation) there is a partial inhibition of nitrate reduction, making C:N ratio maximum under conditions of maximum net photosynthesis.The study of cell glycerol, nitrate, structural proteins and free amino acids indicates that all of these solutes accumulate in the cells as a result of the high salinity adaptation.  相似文献

Summary A micropropagation procedure for the adult cherimoya tree (Annona cherimola Mill.) is described. Axillary shoot proliferation was obtained after culturing nodal sections from Annona cherimola cv. ‘Fino de Jete’, on Murashige and Skoog (MS) medium supplemented with 2.28 μM zeatin. Roots were induced after preincubation of shoots for 3d in light on MS basal medium supplemented with lgl−1 activated charcoal, followed by culturing for 10 d (7 d dark and 3 d light) on MS medium with 492 μM indole-3-butyrie acid (IBA), 15 gl−1 sucrose, and 200 mgl−1 citric acid. Sixty-eight percent of induced shoots rooted after transferring to the same medium without auxin and with the macroelements at half strength and the sucrose at 20gl−1. About 65% of rooted shoots survived after acclimatization. The procedures described herein may prove useful for clonal micropropagation of selected genotypes of cherimoya.  相似文献
