Abstract. Experiments were carried out to determine the origin of dialkyl disulphides found in the larval frass of Acrolepiopsis assectella Zell. which enable the specialist parasitoid Diadromus pulchellus Wsm. to find its host. These kairomones are not specific: they are also found in the frass of the generalist Cacoecimorpha pronubana Hb. fed a leek-based diet. They are emitted only after the frass emerges from the gut. Bacteria are present in the gut of larvae and in their frass. The addition of antibiotics to the larval diet prevents bacterial development and suppresses disulphide emission. When grown in the presence of sulphur amino acids, precursors of sulphur volatiles, some bacteria metabolize kairomones. The production of volatile kairomones, responsible for host– parasitoid relationships, depends on the presence of additional organisms, i.e. bacteria, and not on enzymes arising from the plant or the phytophage.  相似文献

The hemocytic changes that attend parasite encapsulation and melanization in larvae of Drosophila, Musca domestica, and Orthellia caesarion are very similar to those changes occurring in noninfected individuals at metamporhosis. Some of the changes include the precocious differentiation and migration of hemocytes. Comparative and quantitative hemocytological data suggest that some stimulus in infected larvae, acting directly on the hemocytes or on the mechanism(s) controlling their activity, causes the cells to leave areas of the body where they are normally found and to encapsulate parasites. Questions concerning the origin and mode of action of the initial stimulus are raised, and as a basis for future experimentation it is proposed that the cellular immune reactions of these insects against internal metazoan parasites result from a hormonal imbalance.  相似文献

Contact kairomones are essential for host-acceptance behaviour by female parasitoids. In the solitary endoparasitoid wasp, Diadromus pulchellus, this behaviour depends mainly on compound(s) in the cocoon of their host, Acrolepiopsis assectella pupae. Extracts of empty cocoons and polypeptides extracted from cocoons were tested in acceptance behaviour assays using cotton fibre lures bearing extracts. Extractions with solvents of increasing polarity indicated that the active compounds were polar, while SDS-PAGE showed that four glycopolypeptides contained enough information to trigger host-acceptance behaviour in female wasps. This kairomonal activity was found to be due to the protein moieties, and was independent of any glycosylation. These four glycopolypeptides might be two variants of two soluble sericin-like polypeptides differing in their degree of glycosylation.  相似文献

In insects, melanotic encapsulation is an important innate immune response against large pathogens or parasites, and phenoloxidase (PO) is a key enzyme in this process. Activation of prophenoloxidase (proPO) to PO is mediated by a serine proteinase cascade. PO has a tendency to adhere to foreign surfaces including hemocyte surfaces. In this study, we showed that in the naïve larvae of the tobacco hornworm Manduca sexta, hemolymph proPO bound to the surface of granulocytes and spherule cells but not to oenocytoids, and about 10% hemocytes had proPO on their surfaces. When larvae were injected with water (injury) or microsphere beads (immune-challenge), hemolymph proPO was activated, and the number of hemocytes with surface proPO/PO increased at 12 h post-injection, but dropped to the normal level at 24 h. Hemocyte surface proPO can be activated in vitro, leading to melanization of these hemocytes. The number of melanized hemocytes from the larvae injected with water or microsphere beads significantly increased. We also showed that neither hemocytes nor cell-free plasma alone triggered melanization of immulectin-2-coated agarose beads in vitro. However, agarose beads were effectively melanized by isolated hemocytes in the presence of cell-free plasma. Our results suggest that activation of hemocyte surface proPO may initiate melanization, leading to the systemic melanization of hemocyte capsules.  相似文献

When eggs from the lateral oviduct of the gregarious parasitoid Apanteles glomeratus were injected with calyx fluid and venom apparatus material into host larvae, Pieris rapae crucivora, most of the eggs were not encapsulated. Apanteles eggs deposited by the parasitoid from which the venom apparatus was removed were usually encapsulated by the host. These results indicate that the parasitoid venom apparatus material is an important factor in suppressing the encapsulation of 1- or 2-day-old eggs in the host. In order to clearly demonstrate that the venom suppresses egg encapsulation but not the encapsulation of other foreign objects, DEAE-Sephadex A-50 ion-exchange particles stained with 0.001% (w/v) Congo Red solution were injected into hosts together with venom apparatus material. The Sephadex particles were encapsulated by host hemocytes. The results suggest that the venom does not inhibit the encapsulation ability of the host.  相似文献

Résumé Chez Diadromus pulchellus WSM et D. collaris Gr., Hyménoptères Ichneumonidés parasitoïdes spécialistes des nymphes d'A. assectella lépidoptère inféodé aux Allium, des EAG sont enregistrés en réponse à l'odeur du poireau sain ou attaqué par les larves du phytophage, odeurs actives par ailleurs sur leur comportement locomoteur (cinèses). Le thiosulfinate de dipropyle (Ti-P2) très spécifique et abondant dans les Allium, et instable, est le mieux perçu par les deux parasitoïdes mais n'a aucun effet locomoteur chez D. pulchellus contrairement aux disulfures et thiosulfonates, plus stables et moins spécifiques. Les propriétés de l'acétate d'éthyle, actif sur l'antenne des deux parasitoïdes, et sur la locomotion de D. pulchellus expliqueraient en partie les activités biologiques du poireau attaqué sur ces parasitoïdes. Des EAG sont aussi obtenus chez les deux espèces avec l'odeur du chou et de composés soufrés plus ou moins spécifiques des Crucifères. Le thiocyanate de méthyle (TCN-M), le plus actif sur l'antenne et seul efficace sur le comportement locomoteur (de D. pulchellus), expliquerait en partie l'installation reconnue de D. collaris sur Plutella maculipennis spécialiste des Crucifères et rend celle de D. pulchelles vraisemblable.L'odeur des larves de dernier stade d'A. assectella est seule efficace sur l'antenne et la locomotion des femelles de D. pulchellus. Les mâles de ce dernier réagissent (EAG, locomotion) à des odeurs de cocons de l'hôte et particulièrement à ceux occupés par une femelle de leur espèce.  相似文献

The genomic organization of two parasitic wasps was analyzed by DNA reassociation. Cot curves revealed a pattern with three types of components. A highly repetitive DNA, accounting for 15 to 25% of the genome, was identified as satellite DNA. The moderately repetitive DNA corresponds to 26 to 42% of the genome in both species, and shows large variations in complexity, repetitive frequency and a number of sub-components between males and females. These variations are seen as resulting from DNA amplification during somatic and sexual differentiation. Dot blot analyses show that such DNA amplifications concern several types of structural and regulatory genes. The presence of repeated mobile elements was studied by the Roninson method to compare the repeated sequence patterns of Diadromus pulchellus and Eupelmus vuilleti with those of Drosophila melanogaster. The occurrence and organization of mobile elements in these Hymenoptera differ from those of the neighboring order of Diptera. The repetitive and unique components define very large genomes (1 to 3 × 109 base pairs). The genomic organization in Parasitica appears to be an extreme drosophilan type. We propose that the germinal genome of these parasitic wasps is primarily composed of satellite DNA blocks and very long stretches of unique sequences, separated by a few repeated and/or variously deleted, interspersed elements of each mobile element family.  相似文献

Biological control strategies capitalise on natural mechanisms such as predation and parasitism to reduce the need for chemical applications to control insect pests. In Canada, the parasitic wasp Diadromus pulchellus Wesmael (Hymenoptera: Ichneumonidae) is being investigated for its use in the biological control of an invasive crop pest, the leek moth, Acrolepiopsis assectella (Zeller) (Lepidoptera: Acrolepiidae). Large numbers of insects will be needed for releases to ensure that populations of D. pulchellus establish quickly and impact leek moth populations. Since the current culture is not producing the number of insects required for large-scale releases, the accumulation and storage of D. pulchellus might be a viable option to obtain ideal numbers. Currently, little is known about the optimal conditions for the long-term storage and release of D. pulchellus, which overwinter in nature as adults. Using insects from the active culture, the effect of intermittent, short-term warming on cold-stored adults was evaluated for survivorship and fecundity. In accordance with previous findings, females survived cold storage more readily than males. However, the warming regimes employed had no significant impacts on overall survivorship. Cold-stored females had reduced fecundity compared to females maintained in the culture, though no significant differences were noted between the treatments. In addition, the offspring sex-ratio for all treatments was male skewed. Thus, the warming procedures utilised provided no advantages over current techniques for the long-term storage of D. pulchellus intended for release.  相似文献

Ascoviruses (AVs) are pathogenic to lepidopteran larvae, and most commonly attack species in the Noctuidae. The unique pathology includes cleavage of host cells into virion-containing vesicles which leads to the milky white colouration of the hemolymph as opposed to the clear hemolymph of healthy larvae. Recently, we showed that a Heliothis virescens AV (HvAV-3e) isolate is able to induce disease in Crocidolomia pavonana F. (Lepidoptera: Crambidae), affecting feeding, growth and survival of infected larvae. In this study, we investigated the effect of different variants of HvAV-3e on diamondback moth, Plutella xylostella (Lepidoptera: Plutellidae) larvae, another non-noctuid host. In hemolymph inoculation bioassays fourth instar larvae showed a significant dose response to each of the HvAV-3e variants and significant differences between the virulence of the three variants were detected. Both second and fourth instars were readily infected with the virus and infected individuals demonstrated significant reductions in food consumption and growth. The majority of infected individuals died at the larval or pupal stage and individuals which developed into adults were usually deformed, less fecund than non-infected controls and died shortly after emergence. In transmission studies, Diadegmasemiclausum (Hymenoptera: Ichneumonidae), a key parasitoid of diamondback moth, infected healthy host larvae during oviposition following previous attack of HvAV-3e infected hosts. In choice tests D. semiclausum did not discriminate between infected individuals but host infection had no detectable impact on the development of immature D. semiclausum or on subsequent adults.  相似文献

Immunosuppression is the main pathological symptom of the diamondback moth, Plutella xylostella (Lepidoptera: Yponomeutidae), parasitized by an endoparasitoid wasp, Cotesia plutellae (vestalis, Hymenoptera: Braconidae). C. plutellae bracovirus (CpBV), which is a symbiotic virus of C. plutellae, has been known to be the main parasitic factor in the host-parasitoid interaction. CpBV-lectin, encoded in the viral genome and expressed in P. xylostella during early parasitization stage, was suspected to play a role in immunoevasion of defense response. Here we expressed CpBV-lectin in Sf9 cells using a recombinant baculovirus for subsequent functional assays. The recombinant CpBV-lectin exhibited hemagglutination against vertebrate erythrocytes. Its hemagglutinating activity increased with calcium, but inhibited by adding EDTA, indicating its C-type lectin property. CpBV-lectin showed specific carbohydrate-binding affinity against N-acetyl glucosamine and N-acetyl neuraminic acid. The role of this CpBV-lectin in immunosuppression was analyzed by exposing hemocytes of nonparasitized P. xylostella to rat erythrocytes or FITC-labeled bacteria pretreated with recombinant CpBV-lectin, which resulted in significant reduction in adhesion or phagocytosis, respectively. The immunosuppressive activity of CpBV-lectin was further analyzed under in vitro encapsulation response of hemocytes against parasitoid eggs collected at 1- or 24-h post-parasitization. Hemocytic encapsulation was observed against 1-h eggs but not against 24-h eggs. When the 1-h eggs were pretreated with the recombinant CpBV-lectin, encapsulation response was completely inhibited, where CpBV-lectin bound to the parasitoid eggs, but not to hemocytes. These results suggest that CpBV-lectin interferes with hemocyte recognition by masking hemocyte-binding sites on the parasitoid eggs.  相似文献

Females of the parasitic phorid Neodohrniphora sp. were collected in the field and released singly inside an observation chamber placed between a laboratory colony of Atta sexdens (L.) and its foraging arena. The number and speed of loaded and unloaded ants returning to the nest, the weight of foragers and their loads, the number of leaf fragments abandoned by ants, and the number of small workers 'hitchhiking' on leaf fragments were measured before phorids were released, while they were in the observation chamber, and after they were removed. Relatively few ants were attacked by Neodohrniphora sp., but the presence of flies prompted outbound ants to return to the nest and caused a significant reduction on the number and mass of foragers. Additionally, the weight of leaf fragments transported by ants was reduced and the number of abandoned fragments increased in response to Neodohrniphora sp. Presence of the parasitoid caused no significant changes in the number of hitchhiking ants. The regular ants' traffic was resumed after phorids were removed, but foraging activity remained below normal for up to three hours. In the field A. sexdens forages mostly at night, but colonies undergo periods of diurnal foraging during which ants are subject to parasitism from several species of phorid flies. Considering that daytime foraging may be necessary for nutritional or metabolical needs, phorids may have a significant impact on their hosts by altering their foraging behavior regardless of the numerical values of parasitism.  相似文献

In the HymenopteraDiadromus pulchellus diploid males have been observed in samples collected in the wild and bred in the laboratory. Using a yellow body color mutant strain, a protocol of crosses, involving inbred individuals, allows the routine production of such diploid males. These males result from the development of fertilized ova and emerge normally. Their preimaginal viability is similar to those of other individuals, and their imaginal viability does not differ from that of haploid males. Diploid males present normal external morphology and neither mosaicism nor intersexuality was observed. However, they are bigger than haploid males and have head size and wing length similar to those of females. The significance of diploid male viability in hymenopteran populations is discussed.  相似文献

Abstract.  1. Life-history traits such as immunity are often characterised by the presence of large phenotypic variation, but it often remains unclear how and why this variation is maintained by selection.
2. Here an annual social insect, the bumblebee Bombus terrestris , was used to study variation in encapsulation response of males and workers. Bumblebees are a suitable system to study offspring immunity because they are host to a broad variety of different parasites. Bumblebee males, in particular, have a long lifespan compared with other social insect males and their immunity should therefore be an important element for colony reproductive success.
3. Encapsulation response, which was used here as a measurement for the generalised immune defence capacity of an individual, was found to be a highly variable trait. High levels of worker response correlated with low levels of colony parasitism rates.
4. Encapsulation response was found to be (a) lower in males compared with sister workers, and (b) lower in late-produced cohorts compared with early ones.
5. In colonies with delayed sexual reproduction, males had a lower encapsulation response. Thus, investments into immunity seemed reduced in later male cohorts and those eclosing later in the season, perhaps because males had a shorter expected remaining time to acquire matings. The results presented add further evidence that immune defence is a key variable defining colony fitness in social insects.  相似文献

The potential of trap cropping as a pest control strategy was explored for two specialist moth species, where oviposition behaviour determines larval distribution. Oviposition behaviour was studied at three different scales: Individual plants in the laboratory, small scale field experiments with and without cages, and medium scale field trials. The study insects were the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), a specialist on Brassica species and the leek moth, Acrolepiopsis assectella (Z.) (Lepidoptera: Acrolepiidae), a specialist on Allium species. The diamondback moth preferred Indian mustard, Brassica juncea (L.) over white cabbage, B. oleracea var alba (L.). The leek moth did not show preference when offered leek, Allium porrum (L.) and chives, A. schoenoprasum (L.). However, the leek moth did prefer to oviposit on plants of a larger biomass over smaller plants. In both the small scale and the medium scale field studies the trap crops reduced the number of eggs laid on the primary crop. The total number of eggs laid in the monoculture and the trap crop treatments were the same in the small scale field cage study, but the females distributed their eggs differently. More eggs were laid on trap crops compared with primary crops and this pattern was also validated in the medium scale field trials. In this study only the direct effects of trap cropping were considered. Indirect effects of vegetation diversity such as enhancement of natural enemies could potentially increase the efficacy of trap cropping.  相似文献

Chaperonin GroEL assists protein folding in the presence of ATP and magnesium. Recent studies have shown that several divalent cations other than magnesium induce conformational changes of GroEL, thereby influencing chaperonin-assisted protein folding, but little is known about the detailed mechanism for such actions. Thus, the effects of divalent cations on protein encapsulation by GroEL/ES complexes were investigated. Of the divalent cations, not only magnesium, but also manganese ions enabled the functional refolding and release of 5,10-methylenetetrahydroforate reductase (METF) by GroEL. Neither ATP hydrolysis nor METF refolding was observed in the presence of zinc ion, whereas only ATP hydrolysis was induced by cobalt and nickel ions. SDS-PAGE and gel filtration analyses revealed that cobalt, nickel and zinc ions permit the formation of stable substrate-GroEL-GroES cis-ternary complexes, but prevent the release of METF from GroEL.  相似文献

Effects of temperature on egg, larval and pupal developmental times of the leek moth were examined at five constant temperatures ranging from 12 to 20°C. Using this data, a linear degree-day model for the leek moth was developed. The model gave the threshold temperature of T h =6°C, and 630 degree-days were required above the threshold temperature, to complete the development of the leek moth. To validate the model in the field, flight activity during the summers of 1998 and 1999 was studied using pheromone traps. If the summer is warm the leek moth can have two generations per year in Sweden, but the probability of completing two generations within leek plantations is very limited. The likelihood of the leek moth becoming a pest is highly dependent on access to alternative host plants for the first generation of leek moths.  相似文献

Summary Catecholamines and related o-diphenols extracted from the cuticle and hemolymph of adult cockroaches during sclerotization and pigmentation of the cuticle were analyzed by reverse phase HPLC with electrochemical detection. At ecdysis, dopamine (DA) oconjugates predominated in the hemolymph of Periplaneta americana, P. australasiae, P. fuliginosa, P. brunnea, and Blatta orientalis (Blattidae); Blattella germanica (Blattellidae); and Gromphadorhina portentosa and Blaberus craniifer (Blaberidae). n-Acetyldopamine (NADA) conjugates were second in abundance in these species, but were major in the hemolymph of the other blaberoid species, Leucophaea maderae and Nauphoeta cinerea. After ecdysis NADA became the major hemolymph catecholamine in all species as DA decreased rapidly. n--Alanyldopamine (NBAD) concentrations in the hemolymph remained low in all species, although NBAD and its metabolite, n--alanylnorepinephrine (NBANE), were generally the major catecholamines in tanning cuticle. Catechol (1,2-dihydroxybenzene) occurred mainly as a conjugate(s) at high levels in the hemolymph of nymphs and adults of all blattid species. Only trace amounts were detected in B. germanica and Cryptocercus punctulatus (Cryptocercidae), and none was found in any of the blaberoid species. High concentrations of NBANE and NBAD accumulated in tanning cuticle of B. germanica, G. portentosa, and all blattid species, whereas NADA and DA predominated in cuticle from the other blaberoid species, particularly L. maderae and N. cinerea. However, cockroaches as a group appear to utilize both the n-acetyl and n--alanyl catecholamines for stabilization of the exoskeleton. We tested the ability of saturated n-monocarboxylic acids ranging from eight to 12 carbons in length to self-assemble into vesicles, and determined the minimal concentrations and chain lengths necessary to form stable bilayer membranes. Under defined conditions of pH and concentrations exceeding 150 mM, an unbranched monocarboxylic acid as short as eight carbons in length (n-octanoic acid) assembled into vesicular structures. Nonanoic acid (85 mM) formed stable vesicles at pH 7.0, the pK of the acid in bilayers, and was chosen for further testing. At pH 6 and below, the vesicles were unstable and the acid was present as droplets. At pH ranges of 8 and above clear solutions of micelles formed. However, addition of small amounts of an alcohol (nonanol) markedly stabilized the bilayers, and vesicles were present at significantly lower concentrations (∼20 mM) at pH ranges up to 11. The formation of vesicles near the pKa of the acids can be explained by the formation of stable RCOO…HOOCR hydrogen bond networks in the presence of both ionized and neutral acid functions. Similarly, the effects of alcohols at high pH suggests the formation of stable RCOO…HOR hydrogen bond networks when neutral RCOOH groups are absent. The vesicles provided a selective permeability barrier, as indicated by osmotic activity and ionic dye capture, and could encapsulate macromolecules such as DNA and a protein. When catalase was encapsulated in vesicles of decanoic acid and decanol, the enzyme was protected from degradation by protease, and could act as a catalyst for its substrate, hydrogen peroxide, which readily diffused across the membrane. We conclude that membranous vesicles produced by mixed short chain monocarboxylic acids and alcohols are useful models for testing the limits of stabilizing hydrophobic effects in membranes and for prebiotic membrane formation.  相似文献
