The effects of prostaglandin F (PGF) on propulsive activity in segments of isolated colon and on isolated strips of guinea-pig colon were investigated.Using experimental conditions under which spontaneous propulsive activity was negligible, PGF (5×10−8×1×10−6M), added to the bathing medium, increased propulsive activity in a concentration dependent manner. This increase of propulsive activity was abolished in the presence of atropine or tetrodotoxin (1×10−7g/ml).The contractions produced by PGF(5×10−7 − 1×10−5M) in isolated longitudinal and circular smooth muscle strips of guinea-pig colon were unaffected in the presence of atropine or tetrodotoxin (1×10−7g/ml).From these results it is concluded that under the conditions employed in this study propulsive activity stimulated by PGF may depend on the contractions of both muscle layers and stimulation of the peristalic reflex.  相似文献

Prostacyclin (PGI2) produced a biphasic response in canine isolated basilar arteries. In low doses (1 X 10(-8)M-1 X 10(-7)M) PGI2 caused a slight but consistent relaxation of resting muscle tone. In low concentrations (1 X 10(-8)M-1 X 10(-6)M) PGI2 antagonized muscle contractions caused by serotonin or prostaglandin (PG) F2 alpha. This relaxant effect with low doses of PGI2 on the isolated cerebral artery contrasts with findings obtained with other PGs and supports the hypothesis that PGI2 is a mediator of vasodilatation. However, in 1 X 10(-5)M concentrations PGI2 contracted the arterial muscle and did not antagonize contractions induced by serotonin or PGF2 alpha.  相似文献

The cell membranes isolated from bovine corpora lutea bound 3H-prostaglandin (PG) F2α with high affinity and specificity. The specific binding of 3H-PGF2α was detectable at 10?10M added 3H-PGF2α and reached saturation at 10?7M to 10?6M. Unlabeled PGF2α, as low as 10?9M, inhibited the binding of 3H-PGF2α with complete inhibition occurring at 10?6M. The Scatchard analysis of equilibrium binding data revealed that the PGF2α receptors are heterogeneous: Kd1?5.1 × 10?9M, n?289 fmoles/mg protein; Kd2?1.8 × 10?8M, n?780 fmoles/mg protein. The relative affinities of various other PGs for binding to PGF2α receptors were (PGF2α?100%): PGF1α?17.5; PGE1?0.8; PGE2?22.4; PGA1?0.007; PGB1?0.01. The specificity and affinity of 3H-PGF2α binding is consistent with the possibility that this receptor interaction may reflect an initial event in the action of PGF2α as a luteolytic agent.  相似文献

Both intact cortical tissue and isolated cortical cells from the adrenal gland of the rat were analyzed for 6-keto-PGF, the hydrolysis metabolite of PGI2, using high-performance liquid chromatography and gas chromatography-mass spectrometry. 6-Keto-PGF was present in both incubations of intact tissue and isolated cells of the adrenal cortex, at higher concentrations than either PGF or PGE2. Thus, the cortex does not depend upon vascular components for the synthesis of the PGI2 metabolite. Studies in vitro, using isolated cortical cells exposed to 6-keto-PGF (10?6-10?4M), show that this PG does not alter cAMP levels or steroidogenesis. Cells exposed to PGI2 (10?6-10?4M), however, show a concentration-dependent increase of up to 4-fold in the levels of cAMP without altering corticosterone production. ACTH (5–200 μU/ml) increased cAMP levels up to 14-fold, and corticosterone levels up to 6-fold, in isolated cells. ACTH plus PGI2 produced an additive increase in levels of cAMP, however, the steroidogenic response was equal to that elicited by ACTH alone. Adrenal glands of the rat perfused in situ with PGI2 showed a small decrease in corticosterone production, whereas ACTH greatly stimulated steroid release. Thus, while 6-keto-PGF is present in the rat adrenal cortex, its precursor, PGI2, is not a steroidogenic agent in this tissue although it does stimulate the accumulation of cAMP.  相似文献

The effects of prostacyclin (PGI2) on transepithelial potential difference (PD) and sodium transport were examined in rabbit cortical collecting tubules (CCT) perfused in vitro. Addition of PGI2 (10?6M) to the bathing medium, which was bubbled with 95% O2 – 5% CO2, caused a reversible decrease in PD averaging 49±9.4 (SE)%. Maximal effect was evident between 5–10 min. After addition of PGI2 and PD returned spontaneously towards control values within 30 min., corresponding to the rapid degradation of PGI2. In a more alkaline bathing solution achieved by bubbling with 100% O2, in which the degradation of PGI2 is known to be delayed markedly, the decrease in PD by PGI2 was continuous and dose-dependent, with half-maximal and maximal effects achieved at 10?7 M and 10?5 M, respectively. Neither 10?8 M PGI2 nor vehicle alone exerted significant effects on PD. 6-keto-PGF (10?5M), believed to be the major metabolite of PGI2, had no effect on PD. Lumen-to-bath flux of Na decreased with PGI2 from 9.0 to 5.6 pEq/cm/sec (n=4, p<0.005), although bath-to-lumen flux did not change significantly. In summary, PGI2 caused a dose-dependent decrease in PD of rabbit CCT and inhibited Na absorption in this segment in vitro. These results suggest that PGI2 may play an important role in regulating Na transport in CCT.  相似文献

Inhibitory effects of 3-hydroperoxy-3-methyl-2-phenyl-3H-indole(HPI) on prostaglandin endoperoxide synthase(EC 1.14.99.1) and prostaglandin I2(PGI2) synthetase were compared with those of 15-hydroperoxy-5,8,11,13-eicosatetraenoic acid, namely, 15-hydroperoxyarachidonic acid(15-HPAA) and tranylcypromine (TCP). Sheep seminal vesicle microsomes were used as a source of prostaglandin endoperoxide synthase and bovine aortic microsomes as that of PGI2 synthetase. 15-HPAA and HPI inhibited PGI2 synthetase with IC50s of 5 × 10?7 and 3.5 × 10?6 M, respectively, whereas neither compound had effect on prostaglandin endoperoxide synthase at the concentration inhibiting PGI2 synthetase by 90%. TCP was a weak(IC50 = 5 × 10?4M) PGI2 synthetase inhibitor with low specificity.  相似文献

The contracting and relaxing potencies of anf interactions between a number of prostaglandins (PGs) were studied in vitro on spiral strips of small canine mesenteric arteries (outside diameter < mm). PGF2α and PGE2, the most potent contracting PGs, were nearly equal in potency (EC50 4 × 10?7M) and did not cause relaxation under our experimental conditions. PGI2 and PGE1 were equal and the most potent relaxing PGs (EC50 3 × 10?9M). PGE1 also caused contraction, but this effect was not consistent. PGI2 did not cause contraction in concentrations up to 3 × 10?6M. In higher concentrations, however, it caused abrupt and near maximal contraction. PGD2 was weak in both respect, causing incomplete relaxation and contraction or biphasic effects. Interaction studies showed that PGE1 and PGI2 mutually excluded the relaxing effects of each other. PGE1 also reversed the relaxing effect of isoproterenol. However, pre-exposure to PGD2 did not attenuate the relaxing effect of PGE1 or PGI2 nor was the relaxing effect of PGD2 changed by pre-exposure to PGE1. Two different orders of potency of PGs suggest two PG receptors subserving contraction and relaxation, respectively. Further, it appears that several PGs can act upon both receptors which may explain unusual interactions between the PGs and some of their atypical effects. Finally, the data also suggest that there may be subtypes of the PG receptors subserving contraction and relaxation.  相似文献

The response of endothelial adenylate cyclase (AC) to prostaglandins (PGE1, PGE2, PGF, PGF, PGD2 and PGI2) and the relationship of PGE2 to adrenergic systems were investigated in cerebrovascular endothelial cultures. E-type prostaglandins and PGI2 were more effective in stimulating endothelial AC (EC50 = 3 × 10?7M, and 3 × 10?7M, respectively) than prostaglandins of the F-series and PGD2 which activated AC at high doses only. A modulation of endothelial AC response to either PGE2 or norepinephrine (NE) was observed in the presence of both agents in the system. It was manifested by a dose-dependent NE inhibition of the PGE2-stimulated formation of cAMP, which was partially restored by phentolamine. Alpha and β-adrenergic agonists (α, clonidine and 6-fluoronorepinephrine; β, isoproterenol) also partly blocked while forskolin and PGE2 synergistically stimulated the production of cAMP in the endothelial cultures. These findings strongly suggest that the interaction of prostaglandins and α- and β-adrenergic agonists with the AC system in cerebrovascular endothelium may play a role in the regulation of the cerebral microcirculation and/or blood pressure.  相似文献

Prostacyclin when added to medium perfusing rat and rabbit hearts caused an increase in perfusion pressure at concentrations from 1 pg/ml ? 1 ng/ml (2.8 × 10?12 ? 2.8 × 10?9M) and a decrease at higher concentrations. Rhythm disturbances were observed with low prostacyclin concentrations in 6 of 10 rat hearts and 2 of 5 rabbit hearts studied. Increased heart rates were seen in the isolated rat hearts but not in the rabbit hearts. Force of contraction of isolated rat hearts was increased with increasing prostacyclin concentrations up to 100 pg/ml. Higher concentrations decreased contractile force. No inotropic effects were seen with rabbit hearts.  相似文献

Human venous endothelial cells synthesize prostacyclin (PGI2) in response to treatment with histamine. The amount of PGI2 produced is proportional to the histamine concentration over the range of 10?7 to 10?5 M, with a maximal response at 2–5 × 10?6 M. PGI2 synthesis occurs as a burst lasting less than 3 minutes after histamine addition. The H1 histamine receptor antagonist pyrilamine causes an 87% inhibition of PGI2 synthesis, whereas the H2 antagonist cimetidine gives no significant inhibition, suggesting that PGI2 synthesis in response to histamine is mediated by an H1 receptor.  相似文献

The musculature of the oviduct consists of an outer, irregular layer of longitudinal muscle and an inner layer of circular muscle. The four basic modes of activity—compression, segmentation, peristalsis, and reverse peristalsis—were evident in the isolated oviduct. These spontaneous events often occurred in an organized sequence. In fact eggs could be transported down the lateral oviducts by this myogenic activity once the sphincter between the common oviduct and vagina was severed. Myographic recordings were made of only the contractions of the longitudinal muscles. L-glutamate caused a distinct phasic contraction at 2.2 × 10?5 M. The response became larger and more complex as the concentration of the amino acid was increased. Acetylcholine (1.6 × 10?5 M) caused either a phasic or tonic response, or a combination thereof. By contrast, 5HT and tyramine simply increased the frequency of small phasic contractions, although in some preparations both monoamines caused an inhibition. The ecdysones, a juvenile hormone analogue (1 × 10?6 M), and prostaglandin E2 had no effect on oviduct activity. Initially high KCI solutions (162 mM) without Ca++ induced a strong contraction but subsequent additions failed to do so. However, when a high KCI solution (158 mM) with 2 mM Ca++ was added to the preparation the response was partially restored. Also the potent calcium antagonist Mn++ (2mM) can suppress spontaneous activity.  相似文献

Results from this study have indicated serotonin-sensitive adenylate cyclase activity in adult rat brain. The enzyme is localized in the synaptosomal plasma membrane and apparently has multiple activation sites for serotonin with specific activity maxima occuring at serotonin concentrations of 5 × 10?10, 5 × 10?9, 1 × 10?8, and 5 × 10?8 moles/liter. The production of cyclic AMP at these sites appears to be unaffected by 1 × 10?5M fluphenazine, while 1 × 10?5M tryptamine, methysergide, and ergonovine decreased the stimulatory effect of 1 × 10?8M 5-HT by 30 percent, 80 percent, and 57 percent respectively.  相似文献

Endothelin-1 (ET-1) is a potent vasoconstrictor peptide that induces characteristically long-lasting contractions. We used rat aortic rings to investigate the role of protein kinase C (PKC) in ET-1-induced contractions and prostacyclin (PGI2) release. ET-1 (10−9 M) produced a gradual and sustained contraction in rat aortic rings. Pretreatment of aortic rings with different doses (10−9 M and 10−6 M) of diltiazem (voltage-sensitive L-type calcium channel blocker) produced significant inhibition of ET-1- and PDBu-induced contractions and PGI2 release. Inhibition was first noted at 10−9 M and was complete at 10−6 M. Conversely, pretreatment of aortic rings with different doses (10−9 M and 10−6 M) of calcium channel blockers (thapsigargin, an intracellular calcium channel blocker, or conotoxin, a voltage-sensitive N-type calcium channel blocker) produced no changes on ET-1- or PDBu-induced contraction or PGI2 release. These results provide further support for the concept that PKC mediates ET-induced contractions and PGI2 release in rat aortic rings via an increase in intracellular calcium and this increase is due to the influx of extracellular calcium and not to the release of calcium from the sarcoplasmic reticulum.  相似文献

Results from this study indicate that adult rat brain posesses guanylate cyclase activity sensitive to serotonin (5-HT) and localized in the synaptic plasma membrane. The enzyme appears to have multiple activation sites for 5-HT with specific activity maxima at the 5-HT concentrations of 5 × 10?10M and 7 × 10?8M respectively. The rates of guanosine-3′:5′-monophosphate (cyclic GMP) formation at these concentrations of 5-HT are, respectively, 170% and 307% above the endogenous or basal production rate of 2.7±0.3picomoles/minute/milligram of synaptosomal membrane protein. We have also been able to identify four distinct types (Type #1, #2, #3, and #4) of high affinity, specific binding sites for 5-HT on isolated synaptosomal membranes from rat brain. Dissociation constants of 2.6 × 10?10M, 2.5 × 10?9M, 7.0 × 10?9M, and 4.6 × 10?8M, characterize the binding of 5-HT to our sites of Type #1 through Type #4 respectively. The specific, high affinity binding was saturated at 5-HT concentrations of 5 × 10?10M for the Type #1 sites, 5 × 10?9M for our Type #2 sites, 1 × 10?8M for our Type #3 sites, and 7 × 10?8M for our Type #4 sites. The 5-HT concentrations producing saturation of our specific binding sites of Type #1 and Type #4 are virtually identical to those that elicit the two maxima of 5-HT stimulated cyclic GMP production, indicating that a membrane-bound guanylase cyclase may be closely associated with certain 5-HT receptors and/or re-uptake sites.  相似文献

14α-Hydroxymethyl-5α-cholest-7-en-3β-ol (I) and 14α-hydroxymethyl-5α-cholest-6-en-3β-ol (II) have been prepared by chemical synthesis from 3β-acetoxy-7α,32-epoxy-14α-methyl-5α-cholestane. Compound I, previously shown to be efficiently convertible to cholesterol upon incubation with rat liver homogenate preparations, has been found to be a potent inhibitor of sterol synthesis in animal cells in culture. Compound I caused a 50% reduction of the levels of HMG-CoA reductase activity in cultures of L cells and fetal liver cells at concentrations of 3 × 10?6 M and 8 × 10?6 M, respectively. Compound II, the Δ6-analogue of I, caused a 50% suppression of the enzyme activity in the two cell types at even lower concentrations, 5 × 10?7 M and 2 × 10?6 M, respectively. Concentrations of I and II required to specifically inhibit sterol synthesis from acetate were similar to those required to suppress the levels of HMG-CoA reductase activity.  相似文献

Fragments of chopped lung from indomethacin treated guinea-pigs had an anti-aggregating effect when added to human platelet rich plasma (PRP), probably due to the production of prostacyclin (PGI2) since the effect was inhibited by 15-hydroperoxy arachidonic acid (15-HPAA, 10 μg ml?1). Both 15-HPAA (1–20 μg ml?1 min?1) and 13-hydroperoxy linoleic acid (13-HPLA, 20 μg ml?1 min?1) caused a marked enhancement of the anaphylactic release of histamine, slow-reacting substance of anaphylaxis (SRS-A) and rabbit aorta contracting substance (RCS) from guinea-pig isolated perfused lungs. This enhancement was not reversed by the concomitant infusion of either PGI2 (5 μg ml?1 min?1) or 6-oxo-prostaglandin F (6-oxo-PGF, 5 μg ml?1 min?1). Anaphylactic release of histamine and SRS-A from guinea-pig perfused lungs was not inhibited by PGI2 (10 ng - 10 μg ml?1 min?1) but was inhibited by PGE2 (5 and 10 μg ml?1 min?1). Antiserum raised to 5,6-dihydro prostacyclin (PGI1) in rabbits, which also binds PGI2, had no effect on the release of anaphylactic mediators. The fatty acid hydroperoxides may enhance mediator release either indirectly by augmenting thromboxane production or by a direct effect on sensitized cells. On isolated rabbit mesenteric arteries pretreated with phenoxybenzamine (10-5M) and contracted with prostaglandin F (PGF) dopamine (10?6M to 3×10?4M) and isoprenaline (10-9M to 10-5M) caused a dose-related relaxation. Pindolol (10?7M) significantly suppressed the effects evoked by isoprenaline, but did not affect those produced by dopamine. The dopamine receptor antagonist metoclopramide (5×10?5 and 10?4M), however, shifted the dose-response curve for dopamine-induced relaxation significantly to the right in a concentration dependent manner without affecting relaxations caused by isoprenaline or papaverine. These results demonstrate for the first time a specific antagonism to dopamine-induced relaxation on rabbit mesenteric arteries in vitro. They support the hypothesis of the existence of specific dopamine receptors in vascular smooth muscles.  相似文献

Corpora lutea explanted from rats on the sixth day of pregnancy responded to luteinizing hormone (LH; 5 μg/ml) in vitro with a two- to five-fold increase in cellular cyclic AMP (cAMP) concentration. The maximal cAMP level was reached within 60 min and maintained to the end of the 2 hr-incubation. On incubation with prostaglandin F (PGF) in addition to LH, this rise in cAMP accumulation was prevented. For significant suppression, 1.4 × 10?5M PGF was required. In the absence of LH, PGF (4.2 × 10?5M) caused no change in cellular cAMP. Addition of PGF (4.2 × 10?5M) to the incubation medium after the maximal response to LH was attained, caused the cAMP concentration to return to its basal level within 15 min. This abrogation of LH-stimulated cAMP accumulation represents the earliest and hence possibly the triggering event in PGF-induced luteolysis.  相似文献

The formaldehyde method was used to examine the interaction of PGE1 with morphine, β-endorphin and Met-enkephalin on rat mast cells by their effects on IgE-mediated 14C-serotonin release. PGE1 (2×10?8?2×10?5 M) caused a dose-related inhibition of the mediator release 1 min after an antigen challenge, and morphine (3×10?7?3×10?5 M) reversed this PGE1 effect dose-dependently and stereospecifically; naloxone (2×10?4 M) antagonized this action of morphine. β-Endorphin (3×10?7?10?5 M) and Met-enkephalin (3×10?6?10?4 M) mimicked this morphine action dose-dependently and were antagonized by naloxone (2×10?4 M). These results suggest that morphine and endorphins modulate immunological mediator release from rat mast cells through opioid receptors.  相似文献
