不同碳源和氮源对金针菇降解木质纤维素酶活性的影响   总被引:1,自引:0,他引:1

以3株栽培的金针菇Flammulina velutipes为材料,研究它们在玉米芯和棉子壳以及不同碳源、氮源培养条件下纤维素、半纤维素和木质素降解酶活性的规律。结果表明,不同金针菇菌株的羧甲基纤维素酶、木聚糖酶和漆酶活力显著不同(P<0.001),同时,培养条件对羧甲基纤维素酶、木聚糖酶和漆酶的活力都有显著影响(P<0.001)。在简单碳源存在的条件下,金针菇的羧甲基纤维素酶和木聚糖酶活力远远低于复杂碳源培养基(P<0.05)。全营养培养基上生长的金针菇的羧甲基纤维素酶和木聚糖酶活力低于缺乏碳源和氮源的培养基(P<0.05)。漆酶活力在无简单氮源培养基上低于全培养基(P<0.05)和无葡萄糖培养基(P<0.05),即复杂碳源和氮源培养基上的漆酶活力低于简单碳源和氮源培养基(P<0.05)。  相似文献

对采自青海玉树、青海果洛和云南迪庆的冬虫夏草鲜品进行虫生真菌的分离、纯化,获得3种生长形态不同的真菌QH 2019、GL 2019和YN 2019。通过形态学及分子鉴定,菌株QH 2019为蝙蝠蛾拟青霉Samsoniella hepiali,菌株GL 2019为粉棒束孢Isaria farinosa,菌株YN 2019为玫烟色棒束孢Isaria fumosorosea。对这3株菌的培养条件初步研究,结果表明:菌株QH 2019在1/4 SDAY培养基上菌丝日生长速率最快,为(1.94±0.55)mm/d,且菌丝致密、粗壮,含水率高达(91.90±1.22)%,虫草素和虫草酸均以PDA培养的含量最高,分别为(0.47±0.022)mg/g和(3.24±0.021)mg/g;菌株GL 2019在PDA培养基上菌丝日生长速率、菌丝含水率、虫草素含量和虫草酸含量都最高,分别为(2.37±0.20)mm/d、(88.34±2.00)%、(0.23±0.013)mg/g和(6.92±0.019)mg/g,但其菌丝在1/4 SDAY培养基上生长最为致密、粗壮;菌株YN 2019在PDA培养基上菌丝日生长速率、虫草素和虫草酸的含量最高,分别为:(2.27±0.27)mm/d、(0.50±0.012)mg/g和(11.32±0.16)mg/g,但其菌丝在1/4 SDAY培养基上含水率最高(95.23±1.65)%,且生长最为致密、粗壮。综合评价表明3株真菌中YN 2019菌丝中虫草素和虫草酸的含量高、生长速率快,有较好的药用研究价值。  相似文献

苦马豆素(swainsonine,SW)是由小花棘豆等疯草植物中内生真菌通过酵母氨酸还原酶催化产生的一种吲哚里西啶生物碱,牲畜过量采食含SW的疯草植物会中毒,但是SW具有抗肿瘤和免疫调节活性等医学作用。文中以小花棘豆内生真菌野生株OW7.8及其酵母氨酸还原酶基因缺失突变株M1为研究对象,在固体培养基中添加酵母氨酸、α-氨基己二酸、赖氨酸和哌啶酸培养内生真菌野生株和突变株,利用高效液相色谱-质谱联用(HPLC-MS)技术测定SW,比较不同培养时间下内生真菌野生株和突变株中SW合成动态变化,并对所得数据用统计学软件进行方差分析。研究结果显示:对照组OW7.8中SW含量高于M1;添加前体物可提高菌体内SW的含量;不同前体对SW合成的影响不同,其中添加哌啶酸对SW影响最大。统计学分析结果表明:菌株(基因型)、添加前体化合物对真菌SW合成均有显著影响。  相似文献

土壤和植物对冬虫夏草寄主昆虫规模化饲养的影响   总被引:2,自引:0,他引:2  
冬虫夏草大量人工培植的关键之一是寄主昆虫的规模化饲养,本文对西藏林芝地区冬虫夏草生境中不同土壤基质在室内对蝙蝠蛾幼虫生长的影响以及寄主幼虫对5种植物的取食行为、嗅觉反应进行了系统研究。结果表明蝙蝠蛾幼虫偏好土质疏松、渗透性好的土壤,土壤因子中湿度和有机质含量与蝙蝠蛾幼虫存活率成正相关。蝙蝠蛾3龄幼虫对适生地植物的取食选择性与嗅觉反应趋性顺序为鹅绒委陵菜>珠芽蓼>小大黄>圆穗蓼>羊角天麻;进一步Spearman相关分析显示,幼虫对各植物的选择系数与可溶性糖(R=0.850,P<0.05)和粗蛋白(R=0.898,P<0.05)都存在显著的正相关关系,而与粗纤维(R=-0.952,P<0.05)有着显著的负相关关系,与粗灰分(R=-0.391,P=0.516)之间没有显著的相关关系;取食鹅绒委陵菜、珠芽蓼后的幼虫单头虫重显著大于取食其他植物的幼虫。这一研究结果为冬虫夏草寄主昆虫规模化饲养提供了理论依据。  相似文献

本文分别采用浓氨水引咳法、酚红排秘法、常压耐缺氧法、对抗亚硝酸钠中毒性缺氧法,心肌缺血性缺氧法,探讨香栓菌子实体不同极性提取物的止咳化痰及其耐缺氧药理活性。将雄性小鼠随机分组(每组10只),分别为阴性对照组(NCG)、阳性对照组、香栓菌石油醚提取物低TSPEE(L) 0.025g/kg、中TSPEE(M) 0.05g/kg、高TSPEE(H) 0.1g/kg剂量组、香栓菌水提物低TSWE(L) 0.5g/kg、中TSWE(M) 1.0g/kg、高TSWE(H) 2.0g/kg剂量组。小鼠的检测指标包括咳嗽潜伏期、咳嗽次数、咳嗽抑制率、止咳活性(%)、酚红排泌量、化痰率(%)、常压条件下耐缺氧存活时间、亚硝酸钠中毒存活时间、急性脑缺血性存活时间及血清中SOD、MDA、IL-2、IL-4、IgG含量。结果表明,TSPEE(L)无明显效果;TSPEE(M)具有显著化痰作用(P<0.05);TSPEE(H)可延长急性脑缺氧存活时间(P<0.05),并提高SOD、IL-2、IL-4、IgG含量,降低MDA含量(P<0.05);TSWE(L)可增强咳嗽抑制率并增强止咳活性,同时延长常压条件下缺氧存活时间(P<0.05);TSWE(M)除中毒性脑缺氧试验外均具有显著性作用(P<0.05);TSWE(H)可增强咳嗽抑制率、提高止咳活性、增加亚硝酸钠中毒存活时间(P<0.05),同时增加常压条件和急性脑缺血条件下缺氧存活时间(P<0.01)并且提高SOD、IL-2、IL-4、IgG含量,降低MDA含量(P<0.01)。综上所述,香栓菌子实体石油醚提取物和水提取物具有止咳化痰及耐缺氧作用。  相似文献

优化子叶节转化法培育大豆MtDREB2A转基因植株   总被引:4,自引:0,他引:4  
将正交因素试验与GUS基因组织化学染色等技术相结合, 优化大豆(Glycine max)品种东农50遗传转化体系, 导入抗旱关键基因MtDREB2A。结果表明, 大豆种子表面消毒, NaClO溶液法与Cl2气熏蒸法的去污染率分别达到98.67%和93.33%。子叶节法转GUS基因组织化学染色率(68.33%)显著高于下胚轴法(14.00%)和胚尖法(0.67%) (P<0.05)。种子萌发5天, 农杆菌(Agrobacterium tumefaciens)培养温度25°C, OD600=0.9, 共培养5天的转GUS基因子叶节最高达72.00%; 恢复培养5天, 草丁膦(3 mg·L-1)、头孢噻肟钠(200 mg·L-1)和羧苄青霉素(300 mg·L-1)筛选诱导分化的转GUS基因不定芽最多为3.33%; 优化的大豆遗传转化体系转化效率为1.11%。转MtDREB2A基因大豆东农50植株根系更加密集, 主根长度和侧根数量均显著高于对照(P<0.05), 证实MtDREB2A基因具有促进大豆根系生长的作用, 为利用该基因进行大豆抗旱育种奠定了坚实的基础并提供了理论依据。  相似文献

以海南尖峰岭、霸王岭、五指山、吊罗山、鹦哥岭5个热带原始森林土壤为研究对象, 分层采集0-100 cm的土壤样品并分析有机碳含量, 用纵向拟合法和分层估算法分别估算其土壤有机碳密度。结果显示: (1)纵向拟合法计算的5个热带原始森林土壤有机碳密度分别为14.98、18.46、16.48、18.81、16.66 kg·m-2, 分层估算法分别为14.73、16.24、15.50、16.91、15.03 kg·m-2, 前者显著高于后者(p < 0.05); 未经扰动的原始森林土壤, 宜采用纵向拟合法计算土壤有机碳密度。(2) 5个热带原始森林0-30 cm表层土壤有机碳含量分别占0-100 cm的50.50%、48.56%、43.49%、47.37、42.88%。(3)土壤有机碳密度与森林群落Shannon-Wiener指数(p < 0.01)、Simpson指数(p < 0.05)、物种丰富度(p < 0.01)、土壤容重(p < 0.001)存在极显著或显著的负相关关系; 与海拔(p < 0.05)、土壤孔隙度(p < 0.001)、土壤全氮含量(p < 0.001)存在极显著或显著的正相关关系; 与坡向、林分生物量、平均胸径、平均树高无显著相关关系(p > 0.05)。(4)由于海南地处低纬度地区, 其丰富的降水和持续高温条件加速了有机质的分解和物质的再循环, 导致海南森林土壤碳密度远低于全国平均水平。  相似文献

碳、水循环是沙质草地生态系统物质和能量循环的两个关键生态过程, 认识碳、水循环的变化对了解沙质草地生态系统结构与功能对区域气候变化和人类活动的响应具有重要作用。2013年利用箱式法对科尔沁围封和放牧的沙质草地进行了一个生长季的观测研究, 结果表明: (1)在观测周期内, 沙质草地生态系统生产力(GEP)、生态系统呼吸(ER)、蒸散量(ET)在围封和放牧样地之间存在显著差异(p < 0.05)。围封17年样地的GEPERET均最大, 其次为围封22样地的, 放牧样地的最小, 且最大值分别为最小值的2.23倍、1.65倍、1.94倍。(2)碳水(GEPET)之间存在显著的线性正相关关系(p < 0.01), ET可解释GEP 58%-60%的变异, 水分利用效率(WUE)从大到小依次为: 围封22年(2.85 μmol·nmol-1) >围封17年(2.75 μmol·nmol-1) >放牧(2.10 μmol·nmol-1)。(3) GEPER和土壤含水率之间有显著的线性正相关关系(p < 0.01、p < 0.05), 指数模型能够较好地模拟ER对土壤温度变化的响应, ER的温度敏感系数(Q10值)从大到小依次为: 围封17年(1.878) >围封22年(1.733) >放牧(1.477)。因此, 围封能够使退化沙质草地生态系统的碳水循环速率提高, 但围封时间不宜过久。  相似文献

通过研究肿瘤抑制率、脏器指数、肿瘤切片的观察以及血清中影响因子干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)、血管内皮细胞生长因子(VEGF)、过氧化氢酶(CAT)、谷胱甘肽-过氧化物酶(GSH-PX)和超氧化物歧化酶(SOD)等指标的变化对比分析灰树花孔菌 Grifola frondosa子实体粗粉及灰树花孔菌子实体超微粉对Hep-A-22荷瘤小鼠体内抗肿瘤和免疫调节作用。结果表明,各剂量组分均具有抑制肿瘤生长作用,其中灰树花孔菌超微粉高剂量组(1 500mg/kg)抑瘤效果最佳,与模型组比较有极显著性差异( P<0.01)。通过HE染色后的肿瘤细胞可观察到坏死面积增大,同时该组血清中IFN-γ、IL-2、VEGF、GSH-PX和SOD含量显著增加( P<0.01或 P<0.05),并与抑制肿瘤具有一定相关性。  相似文献

为了筛选小花棘豆Alternaria oxytropis内生真菌最稳定的内参基因,以小花棘豆内生真菌OW7.8菌株为研究对象,ACTIN、TUB、GAPDH、APRT、18S rRNA为候选内参基因,利用qRT-PCR技术,GeNorm、NormFinder和BestKeeper软件共同评价候选内参基因的表达稳定性;以培养基中分别添加苦马豆素合成前体化合物赖氨酸(L-Lysine acid,L-Lys)、α-氨基己二酸(L-2-Aminoadidpic acid,L-A)和哌啶酸(L-Pipecolic acid,L-P)的不同生长时间的菌株为实验组,不添加为对照,利用qRT-PCR对真菌酵母氨酸还原酶基因(saccharopine reductase gene,Sac)表达分析。研究表明,Actin为表达最稳定的内参基因,实验组Sac表达量均高于对照组;3种前体化合物均促进Sac的表达,其中添加L-P的真菌Sac表达量最高,生长32 d的真菌的表达量达到最大值;添加L-A和L-Lys的真菌Sac表达量分别在生长第26天和第23天达到最大值。本研究结果为探索Sac对苦马豆素合成的影响提供了基础理论数据。  相似文献

通过蛇足石杉(Huperzia serrata)离体培养研究不同培养基对培养物细胞特征、总DNA特征、生物增长量和石杉碱甲含量的影响。结果表明,Sh、W和Shx三种培养基中,培养物分化成叶状体、根状体和苔藓状体3种不同的形态,三者的叶细胞拟由单细胞出芽成串丛生发育成组织,苔藓状体和根状体比叶状体的细胞表面有更多的根和假根生长;总DNA也存在差异,35、45和55天的苔藓状体均出现DNA梯带,表明形态变异可能与细胞凋亡相关;叶状体、根状体和苔藓状体的生物量增长分别为(1 788±31)%、(833±27)%和(1 963±52)%,石杉碱甲的含量分别为71.7±1.54、20.1±0.82和0μg·L–1(P0.01)。上述结果表明,培养基成分变化可以使相同的基因型材料生成不同的形态,且生长速度和代谢产生石杉碱甲的量等都发生明显变化,叶状体是产生石杉碱甲的最好形态。  相似文献

The effect of season on yield and physical properties of agars extracted from Gracia gracilis and G. bursa-pastoris were determined. The agar yield from G. gracilis was maximum during spring (30%) and minimum during autumn (19%). In G. bursa-pastoris, the agar yield was greatest in summer (36%) and lowest in winter (23%). Agar yield from G. bursa-pastoris was positively correlated with temperature (r=0.94; P<0.01) and salinity (r=0.97; P<0.01) and negatively with nitrogen content (r=−0.93; P<0.01). Agar gel strengths fluctuated from 229 to 828 g cm−2 and 23 to 168 g cm−2 for G. gracilis and G. bursa-pastoris, respectively. The gelling temperature showed significant seasonal variation for both species. Chemical analysis of agar from the two seaweeds indicated variation in 3,6-anhydrogalactose and sulfate content (P<0.01). Furthermore, there was an inverse correlation between the two chemical variables. In general, agar extracted from G. gracilis possessed better qualities than agar extracted from G. bursa-pastoris and can be considered a candidate for industrial use.  相似文献

本文介绍从药用植物丹参内生真菌中筛选新获得的一株对丹参生长和丹酚酸含量具有显著促进作用的菌株,Cladosporium sp. SM58。运用菌根生物技术,于离体、盆栽和田栽条件下,分别将丹参组培苗或幼苗接种SM58固体菌种后,观察测定菌株SM58对丹参组培苗生长的影响,以及丹参幼苗接菌培植6个月后菌株SM58对丹参丹酚酸有效成分含量和生物量的影响;同时进行显微观察,对菌株SM58侵入田栽丹参根组织的状况进行分析。结果表明,SM58菌株不仅对丹参组培苗的株高、根长和生物量均有显著促生长作用(P<0.01),而且对盆栽和田栽丹参的丹酚酸有效成分含量和根干重的提高也均有显著效果(P<0.01)。与对照相比较,接菌组田栽丹参的根干重、总酚酸含量、丹酚酸A的含量分别提高68%、47%、11%。显微观察显示SM58菌丝主要通过侵染丹参的根表皮细胞和皮层细胞而对丹参产生影响。本研究表明,SM58菌株能对丹参品质和产量产生有益影响,是一株具有很好研究价值和开发应用的活性菌。  相似文献

Aims Plants can enhance their photosynthetic efficiency and competitiveness by adjusting canopy structure and radiation interception. The objective of this paper was to quantify the relationship between canopy structure (crown depth and crown area) and light interception (LI) in a Salix matsudana stand under three different stand densities in a flood plain of Zhangye.Methods Our study site is located at the Heihe flood plain of Xichengyi in Ganzhou district, Zhangye City, Gansu Province in the middle Heihe River, where S. matsudana is the dominant species. Based on stand density (10 m × 10 m), the S. matsudana community is divided into three types: low density (I, 25-36 Ind.·plot-1), medium density (II, 37-48 Ind.·plot-1), and high density (III, 49-60 Ind.·plot-1). Community characteristics, soil physical and chemical properties of each type were measured. At each plot, we measured photosynthetically active radiation (PAR), LI, net photosynthetic rate (Pn), transpiration rate (Tr), crown depth, crown area, leaf area index (LAI), twig numbers, twig length, and bifurcation angle. The standardized major axis (SMA) estimation method was used to determine the relationships between LI and canopy structure.Important findings With increasing in stand density, we found that soil moisture increased, and soil electric conductivity decreased, while twig length and crown depth increased, and PAR, twig numbers, bifurcation angle and crown area decreased. LAI and LI, Pn and Tr reached their maximum at the stand of medium density. There was a significant, positive correlation and negative correlation (p < 0.01), respectively, between the LI, crown depth and crown area at low density (I), whereas low significant (p < 0.05) at high density (III), and high significantly positive correlation (p < 0.01) at the medium density (II). S. matsudana has more horizontal branches that reduce LI. Canopy thickness and increased crown area at low density. More vertical distribution of branches at high density, and a more balanced spacial distribution were found at medium density.  相似文献

为了比较糙皮侧耳栽培种在不同常规栽培基质上的漆酶活性,分析更适合糙皮侧耳生长的栽培基质,以1株糙皮侧耳Pleurotus ostreatus栽培菌株为研究材料,研究在固态和液态发酵条件下添加木屑、玉米芯和棉籽壳这3种栽培基质后,对其产漆酶活性的影响。结果表明,不同栽培基质对糙皮侧耳漆酶活性具有极显著的影响(P<0.001);不同发酵方法对糙皮侧耳漆酶活性也具有极显著的影响(P<0.001),仅第2天差异不显著。固体发酵与液体发酵条件下,糙皮侧耳在棉籽壳培养基上所检测到的漆酶活性均高于在木屑或者玉米芯培养基上,表明棉籽壳对提高糙皮侧耳漆酶活性的诱导能力更强。此外,糙皮侧耳在棉籽壳培养基上能够快速分泌漆酶,表明棉籽壳对缩短糙皮侧耳漆酶分泌时间的诱导能力更强。  相似文献

Aims The storage of soil and plant samples has important significance for ecological studies, but has not been widely used. This study aims to compare total carbon/nitrogen content of soil and plant samples before and after long term storage, and further to investigate the feasibility of archiving samples for time series ecological studies at large temporal scales.Methods Soil and plant samples were collected in the growing season in 2011. Carbon/nitrogen mass fraction were analyzed after four years of storage, and were compared with the data obtained before storage using pairwise t-test and linear regression.Important findings Nitrogen mass fractions of stored samples were linearly correlated to the data before storage along the 1:1 line under different storage conditions, and the correlation coefficient r was greater than 0.98 (except for soil samples stored at temperature lower than 20 °C and with particle size <2 mm, r = 0.91). The carbon mass fraction after storage was changed by the storage conditions. Carbon mass fractions of stored samples with particle size <0.15 mm were linearly correlated to the data before storage along the 1:1 line (r > 0.98). Carbon mass fractions of samples with particle size <2 mm increased after storage, and the slope of the linear relationship was 1.26 and 1.04 for soil and plant samples respectively. These results indicated that, nitrogen content of stored samples was stable under different storage conditions, while the stability of carbon content was affected by sample particle size but by storage temperature. Archived samples used for carbon/nitrogen analysis were suggested to be ground to particle size <0.15 mm under fully dry and completely sealed conditions.  相似文献

叶大小-叶脉密度的权衡关系是植物叶经济谱理论的基础, 对理解资源竞争条件下植物叶片的物理构建与生理代谢的关系具有重要的意义。该文采用标准化主轴估计(standardized major axis estimation, SMA)的方法, 按芨芨草(Achnatherum splendens)株丛密度设置I (>12丛·m-2)、II (8-12丛·m-2)、III (4-8丛·m-2)和IV (<4丛·m-2) 4个密度梯度, 以叶面积和叶干质量分别表示叶大小, 对张掖洪泛平原湿地不同密度条件下芨芨草种群的叶大小和叶脉密度的关系进行研究。结果表明: 随着芨芨草株丛密度的降低, 湿地群落的土壤含水量逐渐减小、土壤电导率逐渐增加, 芨芨草的净光合速率(Pn)、蒸腾速率(Tr)和分枝数呈先增大后减小的趋势, 叶面积、叶干质量、比叶面积和株高呈逐渐减小趋势、光合有效辐射(PAR)和叶脉密度呈逐渐增加趋势; 芨芨草叶大小和叶脉密度在高密度(I)和低密度(IV)样地均呈极显著负相关关系(p < 0.01), 中密度(II、III)样地二者呈显著负相关关系(p < 0.05); 叶大小和叶脉密度回归方程的SMA斜率在不同密度样地均显著小于-1 (p < 0.05), 即芨芨草叶大小和叶脉密度呈“此消彼长”的权衡关系。在高密度湿地群落芨芨草倾向于大叶片低叶脉密度的叶片构建模式, 在低密度湿地群落选择小叶片高叶脉密度的异速生长模式, 体现了密度制约下湿地植物的生物量分配格局和资源利用对策。  相似文献

Aims Anthropogenic pollutants cause an increase in ground-level ozone concentration, which is a known threat to plant growth and yield and has been extensively observed worldwide. Since ozone is only slightly soluble in water, it is deposited mainly through dry deposition in terrestrial ecosystem. The object of this study was to analyze the characteristics of ozone dry deposition and to estimate the contribution of stomatal and non-stomatal ozone deposition pathways to total ozone deposition in a winter wheat field.Methods The research site was a winter wheat (Triticum aestivum) field located in Yongfeng experimental station of Nanjing University of Information Science & Technology. The data used in this study were collected from March 16, 2016 to May 30, 2016. We observed ozone dry deposition with an eddy-covariance system. This system mainly included a 3D sonic anemometer, an open-path infrared absorption spectrometer, a fast-response ozone chemiluminescent analyzer and a slow-response ozone monitor. We simultaneously measured meteorological data including solar radiation (SR), air temperature (T), air relativity humidity (RH), wind speed, net radiation, and rainfall. All raw data were recorded with data-logger and averaged every 30 min.Important findings Half hourly means of ozone concentrations (CO3), ozone flux (FO3) and ozone dry deposition velocity (Vd) in the winter wheat field were 32.9 nL·L-1, -5.09 nmol·m-2·s-1, 0.39 cm·s-1, and the ranges of them were 16-58 nL·L-1, -2.9- -11.7 nmol·m-2·s-1, 0.17-0.63 cm·s-1, respectively. FO3 and CO3/Vd were found to be mismatched with phase peaks occurring at different time intervals. The ecosystem was more effective on ozone dry deposition, under conditions of moderate to high SR (SR ≥ 400 W·m-2), moderate T and humility (T = 18 °C and RH > 40%). The relationship between Vdmax and SR was this function (y = 1.06 -exp (-0.0094 - x)). Vdmax increased with SR When SR < 400 W·m-2, and Vdmax reached its maximum when SR =400 W·m-2. Vdmax maintained its maximum when SR ≥ 400 W·m-2. The relationship between Vdmax and T was “bell” curve (y = 1.06 - (x - 18)2/169). Vdmax reached its maximum when T = 18 °C. Vdmax decreased with RH when RH < 40 % (y = 0.030x - 0.106). The variation of Vd might uncertainty when RH was high. There was a liner positive relationship between friction velocity (u*) and Vd, but this relationship was not significant. The mean day-to-day and daytime contributions of stomatal and non-stomatal ozone deposition pathway to total ozone deposition were 32%, 68% and 42%, 58%, respectively, during the whole experimental period.  相似文献
