Contrasting effects on the stomatal index (SI), stomatal density,epidermal cell size and number were observed in four chalk grasslandherbs (Sanguisorba minor Scop., Lotus corniculatus L., Anthyllisvulneraria L. and Plantago media L.) following exposure to elevatedcarbon dioxide concentrations (CO2) in controlled environmentgrowth cabinets. SI of S. minor increased for both leaf surfaces,whilst in A. vulneraria and P. media SI decreased on one surfaceonly. In L. corniculatus , no differences in SI were observedas epidermal cell density changed in parallel with stomataldensity. In L. corniculatus and S. minor stomatal density increasedon both surfaces, whereas in P. media it decreased; in A. vulnerariastomatal density decreased on the abaxial leaf surface alonefollowing exposure to elevated CO2. In the latter three species,SI changed because stomatal density did not change in parallelwith epidermal cell density. The results suggest elevated CO2is either directly or indirectly affecting cell differentiationand thus stomatal initiation in the meristem. In S. minor and P. media leaf growth increased in elevated CO2,because of increased cell expansion of epidermal cells, whereasin L. corniculatus, epidermal cell size decreased and greaterleaf growth was because of an increase in epidermal cell divisions.In A. vulneraria, leaf size did not change, but increased cellexpansion on the adaxial surface suggests CO2 affects leaf surfacesdifferently, either directly or indirectly at the cell differentiationstage or as the leaf grows. These results suggest component species of a plant communitymay differ in their response to elevated CO2. A biophysical analysis of root expansion was conducted in fourchalk downland herbs (Sanguisorba minor Scop., Lotus corniculatusL., Anthyllis vul-neraria L. and Plantago media L.) exposedto either ambient or elevated CO2in controlled environment cabinets.Measurements of fine (F) and extra-fine (EF) root extensionrate (RER), water relations, and cell wall tensiometric extensibilityrevealed differences in the diurnal pattern of root growth betweenspecies. Hydraulic properties of entire root systems and isolated rootsof three contrasting sugarcane clones were evaluated using transpiration-induceddifferences in hydrostatic pressure across intact root systems,root pressure-generated xylem sap exudation, and pressure-fluxrelationships. Regardless of the measurement technique employed,the clones were ranked in the same order on the basis of theirleaf area–specific total root system hydraulic conductance(Croot). All methods employed detected large developmental changesin Grootroot with maximum values occurring in plants with approximately02 m2 total leaf area. Genotypic ranking according to Groot,was reflected as a similar ranking according to root length-specifichydraulic conductance (L) of individual excised roots. Genotypicdifferences in Groot and L were consistent with anatomical characteristicsobserved in individual roots. Patterns of Groot, during soildrying and following re-irrigation suggested that the declinein Groot, observed during soil drying occurred within the rootsrather than at the soil–root interface and may have beencaused in part by xylem cavitation in the roots. Hydraulic and osmotic properties of roots of 2-year-old Norwayspruce seedlings (Plcea abiea (L.) Karst) were investigatedusing different techniques (steady flow, pressure probe, andstop flow technique). Root pressures were measured using theroot pressure probe. Compared to roots of herbaceous plantsor deciduous trees, excised root systems of spruce did not developappreciable root pressure (-0.001 to 0.004 MPa or -10 to 40cm of water column). When hydrostatic pressure gradients wereused to drive water flows across the roots, hydraulic conductivities(Lpr) were determined in two types of experiments: (i) rootpressure relaxations (using the root pressure probe) and (ii)steady flow experiments (pneumatic pressures applied to theroot system or xylem or partial vacuum applied to the xylem).Root Lpr ranged between 0.2 and 810–8m s–1 MPa–1(on average) depending on the conditions. In steady flow experiments,Lpr depended on the pressure applied (or on the flow acrossthe roots) and equalled (0.190.12) to (1.21.7)10–8m s–1 MPa–1 at pressures between 0.2 and 0.4 MPaand (1.51.3)10–8 m s–1 MPa–1 at appliedpressures between 0.8 and 1.0 MPa. When pressures or vacuumwere applied to the xylem, Lpr values were similar. The hydraulicconductivity measured during pressure relaxations (transientwater flows) was similar to that obtained at high pressures(and water flows). Although there was a considerable scatterin the data, there was a tendency of the hydraulic conductivityof the roots to decrease with increasing size of the root system.When osmotic gradients were used to drive water flows, Lpr valuesobtained with the root pressure probe were much smaller thanthose measured in the presence of hydrostatic gradients. Onaverage, a root Lpr=0.01710–8 was found for osmotic andLpr=6.410–8 m s–1 MPa–1 in correspondinghydrostatic experiments, i.e. the two values differed by a factorwhich was as large as 380. The same hydraulic conductivity asthat obtained in osmotic experiments using the pressure probewas obtained by the 'stop flow techniquel. In this technique,the suction created by an osmoticum applied to the root wasbalanced by a vacuum applied to the xylem. Lpr values of rootsystems did not change significantly when measured for up to5 d. In osmotic experiments with different solutes (Na2S04,K2S04, Ca(NO3)2, mannitol), no passive uptake of solutes couldbe detected, i.e. the solute permeability was very low whichwas different from earlier findings on roots of herbs. The effects of NO-3 and NH+4 nutrition on hydroponically grownwheat (Triticum aestivum L.) and maize (Zea mays L.) were assessedfrom measurements of growth, gas exchange and xylem sap nitrogencontents. Biomass accumulation and shoot moisture contents ofwheat and maize were lower with NH+4 than with NO-3 nutrition.The shoot:root ratios of wheat plants were increased with NH+4compared to NO-3 nutrition, while those of maize were unaffectedby the nitrogen source. Differences between NO-3 and NH+4-fedplant biomasses were apparent soon after introduction of thenitrogen into the root medium of both wheat and maize, and thesedifferences were compounded during growth. Photosynthetic rates of 4 mM N-fed wheat were unaffected bythe form of nitrogen supplied whereas those of 12 mM NH+4-fedwheat plants were reduced to 85% of those 12 mM NO-3-fed wheatplants. In maize supplied with 4 and 12 mM NH+4 the photosyntheticrates were 87 and 82% respectively of those of NO-3-fed plants.Reduced photosynthetic rates of NH+4 compared to NO-3-fed wheatand maize plants may thus partially explain reduced biomassaccumulation in plants supplied with NH+4 compared to NO-3 nutrition.Differences in the partitioning of biomass between the shootsand roots of NO-3-and NH+4-fed plants may also, however, arisefrom xylem translocation of carbon from the root to the shootin the form of amino compounds. The organic nitrogen contentof xylem sap was found to be considerably higher in NH+4- thanin NO-3-fed plants. This may result in depletion of root carbohydrateresources through translocation of amino compounds to the shootin NH+4-fed wheat plants. The concentration of carbon associatedwith organic nitrogen in the xylem sap of maize was considerablyhigher than that in wheat. Arabidopsis halleri is a Cd hyperaccumulator; however, the mechanismsinvolved in the root to shoot translocation of Cd are not wellunderstood. In this study, we characterized Cd transfer fromthe root medium to xylem in this species. Arabidopsis halleriaccumulated 1,500 mg kg–1 Cd in the shoot without growthinhibition. A time-course experiment showed that the releaseof Cd into the xylem was very rapid; by 2 h exposure to Cd,Cd concentration in the xylem sap was 5-fold higher than thatin the external solution. The concentration of Cd in the xylemsap increased linearly with increasing Cd concentration in theexternal solution. Cd transfer to the xylem was completely inhibitedby the metabolic inhibitor carbonyl cyanide 3-chlorophenylhydrazone(CCCP). Cd concentration in the xylem sap was decreased by increasingthe concentration of external Zn, but enhanced by Fe deficiencytreatment. Analysis with 113Cd-nuclear magnetic resonance (NMR)showed that the chemical shift of 113Cd in the xylem sap wasthe same as that of Cd(NO3)2. Metal speciation with Geochem-PCalso showed that Cd occurred mainly in the free ionic form inthe xylem sap. These results suggest that Cd transfer from theroot medium to the xylem in A. halleri is an energy-dependentprocess that is partly shared with Zn and/or Fe transport. Hydrodictyon africanum can photosynthesize at high pH underconditions in which HCO3 rather than CO2 is the carbonspecies entering the cell. A passive entry of HCO3 seemsunlikely; a metabolic HCO3 pump is proposed. It is possiblethat such a pump is related to a light-dependent reaction specificto the use of HCO3. This reaction is dependent on photosystem2, but appears to be independent of ATP. These characteristicsare similar to those of active lightdependent Cl influx in H.africanum, and suggest a similar energy source for the two pumps.The HCO3 pump may be electrogenic.  相似文献

A study was made of ultrastructural aspects and ion distributionin roots of Phaseolus coccineus as affected by NaCl and Na2SO4salinity. In the proximal region of the root, xylem parenchymacells are differentiated as transfer cells with well developedwall protuberances adjacent to the half-bordered pits of thevessels. The cytoplasm of these transfer cells contains cisternaeof rough endoplasmic reticulum, the number of which was increasedgreatly when the plants were grown in the presence of NaCl orNa2SO4. The cisternae of the endoplasmic reticulum are oftenassociated closely with the plasmalemma and interconnected withit by fibrillar bridges. Wall protuberances occur also in the exodermis and epidermisof the more apical region of the root. Their function is stillunknown. P. coccineus excludes Na, but not Cl, from the leaves by retainingit particularly in the proximal region of the root. X-ray microanalysisof unfixed, frozen, hydrated specimens revealed that the transfercell-type xylem parenchyma cells in salt-treated roots accumulatedNa relative to both the adjoining xylem vessels and the corticalcells and showed very high Na/K and Na/Cl ratios. It is suggestedthat the xylem parenchyma cells can reabsorb Na from the vessels,probably in exchange for K, and that Na exclusion from the shootis at least partly mediated by this process. Previous papers have shown that abscisic acid can inhibit transportof ions across the root to the xylem vessels, resulting in reducedexudation from excised roots or inhibiting guttation from intactplants. However, it has not been established whether the inhibitionwas due to a reduction in salt transport (Js) or in permeabilityof the roots to water (Lp). This paper investigates the effectof ABA on Lp and Js separately. It is shown that Lp increasedin ABA and then fell, but was about the same as in control rootswhen transport was inhibited. The effect of ABA on exudationtherefore appeared to be mainly due to reduction in Js. Inhibitionof Js was also present in intact, transpiring plants and sowas not due to reduced water flow. The inhibition of ion releaseto the xylem affected Na+, Mg2+, Ca2+, and phosphate as wellas the major ion in the exudate, K+. It is concluded that ABAinhibits salt transport to the shoot by acting on ion transportinto the xylem, and not by reducing water flow coupled withsalt transport.  相似文献

Myrothamnus flabellifoliusWelw. is a desiccation-tolerant (‘resurrection’)plant with a woody stem. Xylem vessels are narrow (14 µmmean diameter) and perforation plates are reticulate. This leadsto specific and leaf specific hydraulic conductivities thatare amongst the lowest recorded for angiosperms (ks0.87 kg m-1MPa-1s-1;kl3.28x10-5kg m-1MPa-1s-1, stem diameter 3 mm). Hydraulic conductivitiesdecrease with increasing pressure gradient. Transpiration ratesin well watered plants were moderate to low, generating xylemwater potentials of -1 to -2 MPa. Acoustic emissions indicatedextensive cavitation events that were initiated at xylem waterpotentials of -2 to -3 MPa. The desiccation-tolerant natureof the tissue permits this species to survive this interruptionof the water supply. On rewatering the roots pressures thatwere developed were low (2.4 kPa). However capillary forceswere demonstrated to be adequate to account for the refillingof xylem vessels and re-establishment of hydraulic continuityeven when water was under a tension of -8 kPa. During dehydrationand rehydration cycles stems showed considerable shrinking andswelling. Unusual knob-like structures of unknown chemical compositionwere observed on the outer surface of xylem vessels. The aim of this work was to discover if the respiratory metabolismof the roots of Plantago lanceolata L. differed from that ofthe roots of Plantago major L. Measurements of oxygen uptakeand dry weight of excised root systems during growth of seedlingsprovided evidence that the two species differed in the amountof respiration needed to support a given increase in dry weight.Excised root systems were given a 6-h pulse in [U-14C]sucrosefollowed by a 16.5-h chase in sucrose. The detailed distributionof 14C amongst the major components of the roots at the endof the pulse and the chase revealed no significant differencebetween the two species. Patterns of 14CO2 production from [1-14C],[2-14C], [3,4-14C], and [6-14C]glucose of excised root systemsfrom plants of three ages were similar for the two species.It is suggested that there is no conclusive evidence for anysignificant inherent difference in the respiratory metabolismof the roots of the two species. Elongation growth of abraded hypocotyl sections of Vigna unguiculataunder xylem perfusion was markedly promoted a few minutes afterthe application of an acid aerosol generated from a solutionof HCl. At the beginning of the acid-induced growth, intracellularpressure (Pi) began to decrease and the membrane potential betweenthe symplast and the xylem apoplast (Vpx) began to depolarize.Subsequently, Pi and Vpx remained at a reduced level and a depolarizedlevel, respectively, while the promotion of elongation growthcontinued for more than 4 hours. The electrogenic componentof the xylem membrane potential (Vpxact) gradually increasedto about twice that before acid treatment. There was a closecorrelation between the enhanced growth and the decrease inintracellular pressure within 30 min after application of acidbut little correltion after 60 min. By contrast, there was littlecorrelation between the promotion of growth and the activityof the xylem pump after 30 min while a close correlation wasobserved after 60 min. It is inferred that the acid-induced activation of water uptakeconsists of two major processes, in series, that are drivenby different forces: the rapid uptake of water for more than30 min, driven by hydrostatic force generated by loosening ofcell walls; and a long-lasting enhancement of water uptake forat least 4 h, which is driven by osmotic force that is generatedby the canal system within the xylem. Ultrastructural features and the distribution of soluble ionshave been examined in mature roots of Zea mays plants grownin both NaCl and Na2SO4 salinities. When the plants were grown in either salt, the Na concentrationincreased proximally along the root with a concomitant declinein the K concentration. Both trends were reversed in the shoot. X-ray microanalysis of deep-frozen, fully hydrated specimensshowed that in salt-treated roots Na, and Cl, or S were distributedabout stoichiometrically in the cortex and endodermis. Na wasusually less concentrated than the anion in the lumens of thevessels, but was concentrated markedly relative to either Clor S in the adjoining xylem parenchyma cells. In the older, proximal parts of seminal roots of plants grownboth without salt (controls) and in the presence of either NaClor Na2SO4, wall developments occurred in xylem parenchyma cellsat the half-bordered pits in which the cell wall became markedlythicker and possessed a loosely packed fibrillar structure.These structures were not comparable with the transfer-celltype of protuberances reported in the roots of other species. In the xylem parenchyma of plants grown in the presence of Na2SO4there were dramatic increases in the quantities of rough endoplasmicreticulum, ribosomes, and mitochondria relative both to controlsand NaCl treatments. The results are discussed in relation to the possible functionof the xylem parenchyma of the mature root in the reabsorptionof Na from the xylem sap, which may mitigate adverse effectsof salinity in salt-sensitive glycophytes.  相似文献

Nitrate assimilation was examined in two cultivars (Banner Winterand Herz Freya) of Vicia faba L. supplied with a range of nitrateconcentrations. The distribution between root and shoot wasassessed. The cultivars showed responses to increased applied nitrateconcentration. Total plant dry weight and carbon content remainedconstant while shoot: root dry weight ratio, total plant nitrogen,total plant leaf area and specific leaf area (SLA) all increased.The proportion of total plant nitrate and nitrate reductase(NR) activity found in the shoot of both cultivars increasedwith applied nitrate concentrations as did NO3: Kjeldahl-Nratios of xylem sap. The cultivars differed in that a greaterproportion of total plant NR activity occurred in the shootof cv. Herz Freya at all applied nitrate concentrations, andits xylem sap NO3: Kjeldahl-N ratio and SLA were consistentlygreater. It is concluded that the distribution of nitrate assimilationbetween root and shoot of V. faba varies both with cultivarand with external nitrate concentration. Respiratory gas exchange and incorporation of 14C-leucine intoprotein were studied in proximal root segments from 25-day-oldmaize plants grown for the last ten days in 50 mM Na2SO4. 14C-leucineincorporation, and oxygen uptake in the presence of glucose,were as large in Na2SO4-grown tissues tested under saline conditionsas in tissues exposed to non-saline solutions throughout Thisadaptation was attributed to an increased metabolic capacityof Na2SO4-treated tissues, because these tissues, when returnedto non-saline solutions, evolved oxygen and incorporated 14C-leucinefaster than tissues exposed continuously to non-saline solutions. These changes are interpreted as a ‘compensation’for the inhibitory effects found when non-adapted tissues wereexposed to 50 mM Na2SO4. Moreover, we have related them to ultrastructuralchanges observed previously in xylem parenchyma cells of thesetissues, and to the possible involvement of these xylem parenchymacells in the re-absorption of sodium from the ascending xylemfluid Zea mays L., maize, salt-stress, respiration, protein synthesis  相似文献

This paper reports autecological field-studies in Singaporeon Drymoglossum piloselloides (L.) Presl., an epiphytic fernof the humid tropics which is capable of performing Crassulaceanacid metabolism (CAM). As indicated by the gas exchange patternsand by the occurrence of a diurnal malic acid rhythm, the plantalso features CAM in situ at its natural sites. Both in well-wateredand in naturally droughted plants external CO2 was taken upsolely during the night. Water stress decreased nocturnal CO2uptake,but left the synthesis and storage of malic acid unaffected.This indicates that CO2 recycling of respiratory CO2 by CAMis ecophysiologically important at the high night temperaturestypical of the tropical habitats of the fern. The plants showeda diel fluctuation of cell-sap osmotic pressure which paralleledthat of malic acid, while the fluctuation of the xylem tensionfollowed the curve of transpiration more closely than it followedthat of the malic acid content. CAM in D. piloselloides wasclearly not limited by natural access to mineral ions and nitrogen.It is concluded that the ecophysiological advantage of CAM forD. piloselloides lies in a better water use efficiency as comparedwith C3 ferns and in the salvaging of carbon by CO2 recycling. Merr.) plants:An estimation of the contribution of current photosynthate toureides in the xylem stream.–J. exp. Bot. 39: 1015–1023. Well-nodulated, water-cultured soybean plants were allowed toassimilate 13CO2 at a constant specific activity for 10 h andthe 13C-labelling of total carbon and ureides in xylem sap wasinvestigated. Labelled carbon appeared very rapidly in the xylem stream. Percentageof labelled carbon (relative specific activity, RSA) in xylemsap was 18% at 2 h after the start of 13CO2 assimilation andreached 53% at the end of the 10 h assimilation. The amountof labelled carbon exported from nodulated roots to the shootsvia the xylem during the 10 h labelling period accounted for33% of total labelled carbon imported into the nodulated roots.Ureides (allantoin and allantoic acid) in xylem sap were stronglydependent on currently assimilated carbon. The RSA of ureidesin xylem sap had reached 83% at the end of the assimilationperiod. Labelled carbon in ureides accounted for 51% of totallabelled carbon returned from nodulated roots to the shootsvia the xylem during the 10 h assimilation period. A treatmentwith 20 mol m–3 nitrate in the culture medium for 2 ddecreased the ureide concentration in the xylem sap slightly,but greatly decreased the RSA of ureides. By comparing the data with the results of analysis of the xylemsap of nodule-detached plants, it was concluded that the majorityof labelled carbon exported to the xylem stream from noduleswas in ureide form. A considerable amount of carbon was alsoreturned from roots to shoots via the xylem stream but it wasmore dependent on (non-labelled) carbon reserved in the roottissues. Nodulated 1-1.5-year-old plants of Acacia littorea grown inminus nitrogen culture were each partnered with a single seedlingof the root hemiparasite Olax phyllanthi. Partitioning of fixedN between plant organs of the host and parasite was studiedfor the period 4–8 months after introducing the parasite.N fluxes through nodules of Acacia and xylem-tapping haustoriaof Olax were compared using measured xylem flows of fixed Nand anatomical information for the two organs. N2 fixation duringthe study interval (635 µg N g FW nodules–1 d–1)corresponded to a xylem loading flux of 0.20 µg N mm–2d–1 across the secretory membranes of the pencycle parenchymaof the nodule vascular strands. A much higher flux of N (4891µg mm–2 d–1) exited through xylem at the junctionof nodule and root. The corresponding flux of N from host xylemacross absorptive membranes of the endophyte parenchyma of Olaxhaustorium was 1.15 µg N mm–1 d–1, six timesthe loading flux in nodules. The exit flux from haustorium toparasite rootlet was 20.0 pg N mm–1 d–1, 200-foldless than that passing through xylem elements of the nodule.Fluxes of individual amino compounds in xylem of nodule andhaustorium were assessed on a molar and N basis. Individuals ofArabidopsis thaliana, collected in different naturalpopulations, were grown in controlled and elevated CO2in a glasshouse.Following germination, root growth of progeny of different linesof these populations was studied in control and elevated atmosphericCO2. No significant direct effect of atmospheric CO2concentrationcould be demonstrated on root growth. An important parentaleffect was apparent, namely that root length and branching weredecreased in seeds collected from a mother plant which had beengrown in elevated CO2. This was correlated with smaller seeds,containing less nitrogen. These parental effects were geneticallyvariable. A technique used for hydroponics was adapted to measure instantaneousroot water uptake from the soil for a leaf succulent CAM species,Agave deserti. Comparisons were made to previously modelledwater fluxes for A. deserti and to Encelia farinosa, a non-succulentC3species. Net CO2uptake and transpiration forA. deserti underwell-watered conditions occurred primarily at night whereasroot water uptake was relatively constant over 24 h. Leaf thicknessdecreased when transpiration commenced and then increased whenrecharge from the stem and soil occurred, consistent with previousmodels. A drought of 90 d eliminated net CO2uptake and transpirationand reduced the water content of leaves by 62%. Rewetting theentire root system for 7 d led to a full recovery of leaf waterstorage but only 56% of maximal net CO2uptake. Root water uptakewas maximal immediately after rewetting, which replenished rootwater content, and decreased to a steady rate by 14 d. Whenonly the distal 50% of the root system was rewetted, the timefor net CO2uptake and leaf water storage to recover increased,but by 30 d gas exchange and leaf water storage were similarto 100% rewetting. Rewetting 10 or 20% of the root system resultedin much less water uptake; these plants did not recover leafwater storage or gas exchange by 30 d after rewetting.
