SYNOPSIS. The modes of pattern regulation found in the ciliates Dileptus and Paraurostyla are compared. These forms are systematically distant but both possess very extensive regenerative capacities. They are characterized by 2 types of ciliary patterns: the ciliature of Dileptus has largely a simple pattern composed of single kinetosomes while that of Paraurostyla has a complex pattern composed of aggregates of kinetosomes interconnected by amorphic matter. In both ciliates a change in cell size evokes pattern regulation which differs substantially in the extent of pattern replacement, timing, and localization of morphogenetic activities. It is concluded that these differences result from the pattern constituents.  相似文献

The fine structure of the dorsal bristle complex and pellicle of non-developing Euplotes eurystomus is described in detail by scanning and transmission electron microscopy. The bristle-pit unit is a highly differentiated complex of organelles. The bristle complex is composed of a pair of kinetosomes (basal bodies) joined by a connective. The anterior kinetosome bears the bristle cilium, which contains a polarized network of particles (“lasiosomes”). The posterior kinetosome bears a very short, knob-like “condylocilium,” and has an associated striated fiber. Accessory ribbons of microtubules are also associated with the kinetosome couplets. Parasomal sacs, a septum connecting the bristle cilium to the anterior wall of the pit, core granules of the kinetosomes, and large membranous ampules are described. The organization of the bristle complex bears many similarities to the somatic ciliature of other ciliates. The pellicle of Euplotes is composed of a continucus outer cell membrane subtended by membranous alveoli, which contain a “fibrous mat.” Two sheets of subpellicular microtubules (longitudinal and transverse) are located just beneath the alveoli. The “epiplasm” seen in some other ciliates is apparently absent in Euplotes. The texture of the cell surface is a pattern of folds or rugae composed of the outer cell membrane and the upper membrane of the alveolus. The pattern of rugae probably defines the “silverline-system” of light microscopy.  相似文献

Two glycoproteins bands isolated from the cyst wall protein pattern of two colpodid ciliates, Colpoda inflata (gp46CI) and Colpoda cucullus (gp46CC) were analysed for their amino acid composition. Both glycoproteins are very rich in glycine and have a relatively high hydrophobicity, containing additionally many leucine and alanine residues. Their high degree of similarity is both quantitative and qualitative. Compared with just two previously published reports, their amino acid compositions are similar to those found in the hydrolysed cyst wall total proteins from the ciliates C. steinii and Paraurostyla spp. The amino acid composition corroborates that they are indeed glycoproteins, because asparagine, an amino acid residue suitable for the attachment to N-acetylglucosamine by its amide group (N-glycan), is abundant in both proteins. We discuss our data in relation to other glycine-rich proteins and a comparison with amino acid composition protein databases is carried out.  相似文献

SYNOPSIS. Morphogenesis, and the cortical structures of Brooklynella hostilis, a cyrtophorine gymnostome ciliate ectoparasitic on marine fishes, were studied from protargol silver-impregnated preparations and with the aid of electron microscopy. The pattern of morphogenesis of Brooklynella is close to that found in less differentiated species of the families Chlamydodontidae (e.g., in the genus Trithigmostoma) and Dysteriidae (e.g., in the genus Hartmanula). The full number of kineties in the opisthe is restored after division from a segment of the left one of the 3 kinetics producing the oral rows. The oral rows consist of a double row of kinetosomes arranged in a zig-zag pattern; only the outer row is ciliated, the inner one being barren. However, the positions of the postciliary and transverse fibers indicate that the oral rows are not homologs of an undulating membrane but are akin to a membranelle. In association with the ventral somatic kinetosomes there are 4 postciliary fibers; a rather aberrant, transversally oriented kinetodesma; 2 microtubular, transverse fibers plus a transverse fibrousspur; and one to several ribbons of subkinetal microtubular fibers. Not directly associated with the kinetosomes are fibrous strands running subpellicularly between the kinetosomes and also deep into the cytoplasm. The cortical structures of Brooklynella are compared with those of some other groups of ciliates of about the same phylogenetic level in which the subkinetal microtubules can also be found– rhynchodine, suctorian, and chonotrich ciliates. The nasse consists of 6–8 nematodesmata not closely associated with the microtubular cytopharyngeal tube. The former have a distinctly developed densely fibrous capitulum containing barren kinetosomes which originally produced the nematodesma during stomatogenesis; the capitulum is connected by a fibrous link to the microtubular shaft. Extending from the oral rows to the capitula are fibrous structures strongly reminiscent of filamentous reticulum in hymenostome and peritrich ciliates. The structure of the posterio-ventral glandular organelle is also described and discussed.  相似文献

Stop codon reassignments have occurred very frequently in ciliates. In some ciliate species, the universal stop codons UAA and UAG are translated into glutamine, while in some other species, the universal stop codon UGA appears to be translated into cysteine or tryptophan. The class Litostomatea has been hypothesized to be the only group of ciliates using the universal genetic code. However, the hypothesis was based on a statistical analysis of quite small sequence dataset which was insufficient to elucidate the codon usage of the class among such highly deviated phylum. In this study, together with the updated database sequence analysis for the class, we approached the problem of stop codon usage by examining the capacity of the translation termination factor eRF1 for recognizing stop codons. Using in vivo assay systems in budding yeast, we estimated the activity of eRF1 from two litostome species Didinium nasutum and Dileptus margaritifer. The results clearly showed that Didinium and Dileptus eRF1s efficiently recognize all three stop codons. This is the first experimental evidence that strongly supports the hypothesis that litostome ciliates use universal genetic code.  相似文献

The morphogenesis of ciliary zones has been studied in the ciliate Eudiplodinium maggi which has a vestigial somatic infraciliature consisting of subcortical barren kinetosomes. These replicate and migrate into the subequatorial ciliary anlagen of the dividing cells. The kinetid pattern of the barren kinetosomes changes sequentially to that of the ciliary zones. Postciliary and transverse microtubules are derived from basal and cortically directed microtubules while a dense spur coincides with the origin of the kinetodesma. All kinetics produce potential cytopharyngeal components. Nematodesmata originate in granular subkinetosomal dense plates, while two of the three types of cytopharyngeal ribbon are formed from tansverse microtubules. A spatial selectivity ensures that these components extend only from specific kinetids. It is concluded that 91) morphogenesis of the intricate cytoskeletal network of infraciliature involves a “pattern generator” in the vestigial somatic kinetids; and 92) entodiniomorph stomatogenesis in not truly apokinetal but is more like the telokinetal process of related ciliates.  相似文献

This study examines previously undescribed general and cytopharyngeal features of the genus Entodinium. The cytopharynx contains three types of microtubular ribbons underlying the cytostomal membrane as well as a loose palisade of nematodesmata. A protoesophagus composed of microtubular bundles associated with a fibrous wall lies internally to one side of an extrusible peristome on which the adoral zone of syncilia (AZS) is mounted. Macronuclear structures are very similar to those of other ophryoscolecids. The micronucleus has chromatin bodies forming a compact mass but lacks the thick wall found in other species. A tubular spongiome surrounds the contractile vacuole and the cytoproct is relatively undifferentiated. Cortical structure follows the usual five-layered ophryoscolecid pattern with subcortical barren kinetosomes arranged into indistinct kineties. The infraciliature of the AZS has kinetosomes set upon a subkinetal rod and with associated bifurcated kinetodesmata and transverse microtubules, some of which extend into the cytopharynx. Components newly described for Entodinium are the one to three postciliary microtubules and the interkinetosomal centro-lateral strand, all of which are present in other species of ophryoscolecid ciliates. The infraciliature of the paralabial ciliary tuft shows similar components to that of the main AZS, but lacks the subkinetal rod. The microtubular components of the cytopharynx are discussed in relation to the “alimentary” structures in other ophryoscolecids, and a relationship of these structures to dietary differences is suggested.  相似文献

The somatic infraciliature is composed of a relatively small number of discontinuous kinetics, formed by groups of few kinetosomes (pallets). The buccal ciliature is composed of an adoral zone of membranelles and a paroral formation otherwise unknown in ciliates, with many short kineties, which lie on a rigid stem. We propose that P. metchnikoffi is a primitive hypotrich and, consequently, we present a new classification system for hypotrichs.  相似文献

ABSTRACT. Dileptids are haptorid ciliates with a conspicuous proboscis belonging to the oral apparatus and carrying a complex, unique ciliary pattern. We studied development of body shape, ciliary pattern, and nuclear apparatus during and after binary fission of Dileptus terrenus using protargol impregnation. Additional data were obtained from a related species, Pseudomonilicaryon brachyproboscis . Division is homothetogenic and occurs in freely motile condition. The macronucleus is homomeric and condenses to a globular mass in mid-dividers. The proboscis appears in late mid-dividers as a small convexity in the opisthe's dorsal brush area and maturates post-divisionally. The oral and dorsal brush structures develop by three rounds of basal body proliferation. The first round generates minute anarchic fields that will become circumoral kinetofragments, while the second round produces the perioral kinety on the right and the preoral kineties on the dorsal opisthe's side. The dorsal brush is formed later by a third round of basal body production. The formation of various Spathidium -like body shapes and ciliary patterns during ontogenesis and conjugation of Dileptus shows a close relationship between spathidiids and dileptids. On the other hand, the peculiarities of the dileptid morphology and ontogenesis indicate a long, independent evolution.  相似文献

SYNOPSIS. The fine structure of the tomite stage of Hyalophysa chattoni was examined with particular attention to its kinetal apparatus. The pellicle, thick and dense compared with that of other ciliates, is formed of three layers. The inner layer is composed of short fibrils oriented perpendicular to the surface. The cytoplasm around the oral passage and beneath falciform field 8 is crowded with dense inclusion bodies of unknown function. Dorsal to the oral passage is the rosette, a disc-shaped organelle subdivided by septa in the form of incomplete radii about a central chamber containing a tuft of cilia. The septa are composed of 3 membranes enclosing a fine layer of cytoplasm. At their inner ends 20 mμ fibers run dorsally and ventrally. Dense clumps of fibrous material line the luminal surface of the septa. Rows of fusiform trichocysts parallel the kineties. The trichocysts are composed of a finely periodic, moderately electron-dense material surrounded by 20 mμ fibrils oriented along the long axis of the trichocyst. Between and below the kinetosomes and the rows of trichocysts are electron-dense vesicles 300 mμ in diameter and bounded by a loose membrane. The large “trichocysts,” the “gros trichocystes” of Chatton and Lwoff, whose appearance heralds the beginnings of trichocystogenesis, prove to be canaliculi opening to the surface. Four separate ciliary membrane systems—the oral ciliature (XYZ), falciform field 8, falciform field 9, and the ogival field—are located on the ventral surface of the tomite. Each differs from the others and from the somatic kineties in the fibrillar organization around its kinetosomes. In the somatic kineties the kinetodesmos is a dense, periodic fiber which is formed of stacks of up to 18 subfibers, each arising from the base of a kinetosome. The kinetosomes are short (300 mμ) and contain dense central granules. In some kineties, alternating between the kinetosomes, are elliptical kinetosome-like structures which do not bear cilia and perhaps provide a reservoir of kinetosomes for future growth of the kinety.  相似文献

Two hypotrichous ciliates, Paraurostyla wuhanensis nov. spec. from Wuhan (China) and a new North American population of the Paraurostyla weissei complex, were studied based on live observations and protargol impregnation. Paraurostyla wuhanensis nov. spec. differs from the congeners by the combined features of having six or seven frontal cirri, 2–4 frontoventral cirri, 5–7 ventral rows, and yellow-greenish cortical granules. Ontogenesis proceeds as in the type species, except that fewer frontoventral cirri are formed in the new species. The morphology of the new population of the Paraurostyla weissei complex corresponds well with other American populations. In the phylogenetic trees based on the 18S rRNA gene, Paraurostyla sequences nest in a large clade together with Apoamphisiella and Notohymena. Monophyly of Paraurostyla is rejected by the results of the approximately unbiased test analyses. Morphological, morphogenetic, and phylogenetic analyses show a close relationship between the North American populations of the Paraurostyla weissei complex and Apoamphisiella, indicating that the taxonomic position of the former needs to be reassigned. A new combination, viz. Apoamphisiella polymicronucleata (Merriman, 1937) comb. nov., a reevaluation of the Paraurostyla weissei complex, and emended diagnoses of Paraurostyla and Apoamphisiella, are provided.  相似文献

We have used small subunit rRNA gene sequences to determine the phylogenetic relationships of species in three genera of endosymbiotic ciliates. We have confirmed that the astome Anoplophrya marylandensis is related to ciliates in the Class Oligohymenophorea, supporting the view that astomes are derived from hymenostome-like ancestors. We confirmed that Plaglotoma lumbrici, formerly considered to be a heterotrich, is a stichotrich spirotrich ciliate most closely related to Paraurostyla weissei in this analysis. Thus, the somatic polykinetids of Plagiotoma can be concluded to be cirri. We report the details of our isolation of Nyctotheroides deslierresae and Nyctotheroides parvus and confirm previous reports that these clevelandellids are related to the metopid and caenomorphid ciliates, now placed in the Class Armophorea.  相似文献

SYNOPSIS The membranes, epiplasm, and fiber systems are described in the somatic cortex of Glaucoma chattoni strain HZ-1. Kinetodesmal fibers, postciliary and transverse microtubular ribbons, basal microtubules, transverse fibers and transverse accessory material are associated with kinetosomes. Longitudinal microtubular ribbons and mitochondria occur interkinetally. In the buccal cortex, the membranes, epiplasm and fibers of the 3 membranelles, the paroral kinety, the ribbed wall, and the cytostome are described. Comparisons between G. chattoni and other ciliates reveal ultrastructural differences of possible systematic significance. In the somatic cortex of this and other tetrahymenines. Iongitudinal microtubular ribbons and basal microtubules occur concurrently. In the buccal cortex, alveoli are absent in tetrahymenine membranelles. A table is presented of the fiber systems associated with single somatic kinetosomes of various ciliates whose cortical ultrastructure has been studied to date.  相似文献

We studied the morphology of four new dileptid ciliates, using standard methods. Dileptus microstoma, which was discovered in Benin (Africa), is outstanding in having a very small oral opening (~4 μm), an interrupted dorsal row of contractile vacuoles, and ampulliform extrusomes. Dileptus semiarmatus, which was discovered in Austria, possesses extrusomes only in the right posterior half of the proboscis and has very widely spaced circumoral and perioral kinetids. Dileptus longitrichus, which was discovered in Japan, is almost unique in having up to 15 μm long brush bristles and a row of contractile vacuoles each in ventral and dorsal side of body. Pseudomonilicaryon brachyproboscis, which was discovered in Greece, differs from the congeners by the narrowly ellipsoidal micronuclei, the dimorphic dorsal brush, the extrusomes, and the contractile vacuole pattern. Four new features are introduced for distinguishing species in dileptids: shape of micronucleus, monomorphic/dimorphic dorsal brush, shape of oral opening, and spacing of circumoral dikinetids. The terrestrial dileptids share several distinct morphological features that are probably adaptations to the soil environment: (1) the body is comparatively slender and small, what is likely related to the narrowness of the habitat; (2) the proboscis is short, which increases the relative volume of the trunk, what might be related to its fragility and/or to the space available for prey digestion; (3) the long dorsal bristles might foster prey recognition; and (4) the pronounced body flexibility in all dileptids likely fosters their high diversity in the narrow and wrinkled soil habitat.  相似文献

ABSTRACT. The ultrastructure of the oral region and the ultrastructural architecture of the basket of Prorodon aklitolophon and Prorodon teres are described. the oral region of Prorodon consists of: 1) A circumoral kinety at the anterior pole of the cell surrounding the typically slit-shaped cytostomial funnel. This kinety is composed of inversely oriented dikinetids in which both kinetosomes are ciliated and are associated with a very short postciliary microtubular ribbon and a few transverse microtubules; 2) Three brush rows aligned in parallel and extended meridionally in the anterior part of the cell. the individual brush rows consist of dikinetids, but in contrast to the dikinetids around the cytopharynx they are not inverted and only the anterior kinetosomes bear specialized short brush cilia and are associated with a divergent-tangential transverse microtubular ribbon. the posterior kinetosome is non-ciliated and bears a prominent convergent postciliary microtubular ribbon. Schematized dikinetid patterns of both oral regions of Prorodon are provided. In addition, a three-dimensional reconstruction of the basket of the genus Prorodon based on serial thin sections is presented. A phylogenetic tree, mainly based on stomatogenic data, is given to show the phylogenetic relationships of some prostomatid genera as well as the hypothesized sistergroup relationship of colpodid and prostomatid ciliates.  相似文献

It has been previously shown that some amino acids and their derivatives are capable of regulating the activity of adenylyl cyclase (AC) and guanylate cyclase (GC) in free-living ciliates Dileptus anser and Tetrahymena. The aim of this work was to study the molecular mechanisms of action of methionine, tyrosine, alanine and neurohormone serotonin on the activity of enzymes-cyclases and the identification of their specific receptors in D. anser and T. pyriformis. Methionine and serotonin significantly increased the basal AC activity in both ciliates, and the AC effect of serotonin in T. pyriformis was carried out with the participation of Ca2+-dependent form of AC and heterotrimetic G proteins. AC stimulating effect of tyrosine and alanine was expressed weakly and only detected in D. anser. Serotonin is both ciliates and alanine in D. anser stimulated GC activity, whereas methionine and tyrosine had no effect on GC. Methionine and serotonin bind to surface receptors of the ciliates with high affinity. K(D) for [methyl-3H] methionine binding to D. anser and T. pyriformis were 7.5 and 35.6 nM, and for [3H] serotonin binding were 2.7 and 4.7 nM, respectively. Alanine and tyrosine bind to the ciliates with low affinity. Thus, ciliates D. anser and T. pyriformis have chemosignaling systems regulated by amino acids and their derivatives and including the enzymes with cyclase activity. There is an assumption that these systems are similar to hormonal signaling systems of higher eukaryotes and are their predecessors.  相似文献

利用电镜酶细胞化学方法,对大尾柱虫(Urostylagrandis)、冠突伪尾柱虫(Pseudourostylacristata)、魏氏拟尾柱虫(Paraurostylaweissei)三种纤毛虫细胞内的酸性磷酸酶(ACPase)进行了定位观察。发现在它们体内ACPase的阳性反应颗粒除分布在消化腔隙内独立的泡状结构上和被消化的食物组织中外,在细胞质中的小圆泡内也有分布。所得结果证实了消化腔隙和小圆泡都具有消化功能。作者推测消化腔隙主要消化食物,小圆泡则可能与自身物质(如衰老胞器等)的消化有关;并推测这种消化腔隙与小圆泡共存是纤毛虫消化胞器由食物泡演化为消化腔隙系统的中间形式。此外,在大尾柱虫纤毛内部观察到非溶酶体性的ACPase。  相似文献

(1) Ciliated protozoa are viewed as unicellular organisms structured in a hierarchy of organizational levels that include the macromolecular, suborganellar, unit organellar, organellar complex, and organellar system. (2) The ciliate cortex is divided into two major functional regions, the somatic region and the oral region. The fundamental component of the cortex is an organellar complex, the kinetid, whose organizing centre is the kinetosome with which are associated three fibrillar associates diagnostic of ciliates. These three fibrillar associates are the periodically striated kinetodesmal fibril and two microtubular ribbons, the transverse and postciliary ribbons. (3) Somatic and oral kinetids are found to be of three major types: monokinetids are composed of one kinetosome and its fibrillar associates; dikinetids are composed of two kinetosomes and their fibrillar associates; polykinetids are composed of more than two kinetosomes and their fibrillar associptes. (4) The mechanisms underlying kinetid function and development remain largely unexplored. Research into the molecular biology and ultrastructure, especially of mutant forms, should provide basic insights in the near future. (5) The conservation of kinetid structure across major phyla of organisms suggests that this subcellular structure should be useful in phylogenetic analysis despite the concepts of ‘chemical identity’ and ‘organic design’. (6) The evolutionary rate of change of oral features is greater than that of somatic features, probably due to developmental and ecological factors. Nevertheless both cortical regions are constrained by the phenomenon of structural conservatism; that is, the conservation of structure through time is inversely related to the level of biological organization. (7) Eight major groupings of ciliate species are recognized, based on ultrastruc-tural features of the cortex. Several examples of differences between these eight groups and the groups presently recognized are discussed.  相似文献
