Tyrocidine, a peptide antibiotic produced by Bacillus brevis (ATCC 8185), relaxes superhelical DNA in a biphasic manner and induces 'packaging' of the DNA at higher concentrations. This was concluded from studies using the sensitive 4,5',8-trimethylpsoralen photobinding technique [Sinden, R. R., Carlson, J. O. & Pettijohn, D.-E. (1980) Cell 21, 773-783]. Relaxed DNA is not affected by tyrocidine whereas linearized molecules become packaged. The linear gramicidin synthesized by the same strain reverses the tyrocidine-induced relaxation as well as the packaging, an observation which might be of biological relevance.  相似文献

Effects of the two peptide antibiotics tyrocidine and the linear gramicidin on exponential growing cultures of Bacillus brevis (ATCC 8185) were studied. Both antibiotics are produced by this bacterial strain. Our results demonstrate that the addition of tyrocidine leads to inhibition of RNA synthesis followed by a cessation of growth. In contrast, gramicidin does not affect net RNA synthesis and the cells continue to grow. However, sporulation is inhibited by gramicidin. The addition of a mixture of tyrocidine and gramicidin prevents partially the inhibitory effect of tyrocidine on RNA synthesis. The results presented are essentially in agreement with in vitro results described previously (8,9).  相似文献

This paper presents evidence that the two peptide antibiotics tyrocidine and linear gramicidin, produced by Bacillus brevis ATCC 8185, are required for the induction of sporulation in the producer organism. When tyrocidine synthesis was specifically blocked with 2-amino-3-hydroxy-3-phenylpropanoic acid [Mach, B., Reich, E., and Tatum, E. L. (1963) Proc. Natl Acad. Sci. USA, 50, 175-181], sporulation and gramicidin synthesis were inhibited, but both processes could be restored by the addition of tyrocidine. Certain other amino acids such as L-tyrosine inhibited both sporulation and peptide antibiotic synthesis in nitrogen-limited cultures. When either tyrocidine or linear gramicidin was added together with L-tyrosine, neither sporulation nor peptide antibiotic synthesis was restored. On the other hand, the addition of both tyrocidine and linear gramicidin effectively reversed the inhibition of sporulation by L-tyrosine. These experiments demonstrate that sporulation of B. brevis depends on either the endogenous synthesis or the addition of both tyrocidine and linear gramicidin. The fact that endogenous as well as exogenous peptides could effect sporulation argues against the involvement of artifacts, such as the depletion of intracellular nucleotide pools caused by the surfactant properties of added peptide antibiotics.  相似文献

The interactions of three polypeptide antibiotics (polymyxin B, gramicidin S, and valinomycin) with artificial lecithin membranes were studied by nuclear magnetic resonance (NMR). Combination of 31P and 2H NMR allowed observation of perturbations of the bilayer membrane structure induced by each of the antibiotics in the regions of the polar headgroups and acyl side chains of the phospholipids. The comparative study of the effects of these membrane-active antibiotics and the lipid bilayer structure demonstrated distinct types of antibiotic-membrane interactions in each case. Thus, the results showed the absence of interaction of polymyxin B with the dimyristoyllecithin membranes. In contrast, gramicidin S exhibited strong interaction with the lipid above the gel to liquid-crystalline phase transition temperature: disordering of the acyl side chains was evident. Increasing the concentration of gramicidin S led to disintegration of the bilayer membrane structure. At a molar ratio of 1:16 of gramicidin S to lecithin, the results are consistent with coexistence of gel and liquid-crystalline phases of the phospholipids near the phase transition temperature. Valinomycin decreased the phase transition temperature of the lipids and increased the order parameters of the lipid side chains. Such behavior is consistent with penetration of the valinomycin molecule into the interior of the lipid bilayers.  相似文献

Activity of polymyxin B nonapeptide alone and in combination with other antibiotics against clinical strains of Pseudomonas and enteric bacteria was studied. It was shown that nonapeptide was highly active against Pseudomonas and moderately active against enteric bacteria. In combination with rifampicin, fusidic acid or erythromycin the nonapeptide had a potentiating effect on the tested strains.  相似文献

A treatment of Bacillus thuringiensis cells with gramicidine D in the medium containing the yeast and polysaccharides increases the lag-phase up to 12 h without a change of the rate during the logarithmic phase of the culture growth. The exoprotease activity of cells treated with gramicidine reaches a maximum value 4 h earlier in comparison with the control culture, and the activity level is increased 2-fold. At a concentration increasing the exoprotease activity, gramicidine was found to induce the permeability of Bac. thuringiensis membranes for potassium ions. An additional introduction of 250 mM KCl or NaCl into the medium inhibits only the exoprotease activity of cells treated with gramicidine. It is assumed that the ability of gramicidine to increase the exoprotease activity of Bac. thuringiensis may be due to a change in the intrabacterial ionic composition.  相似文献

Wood waste powder from Tectona grandis containing melanin was less susceptible to enzymatic hydrolysis than powder without melanin. About a 53% increase in saccharification was noted when melanin was removed. Melanin caused inhibition to all cellulolytic enzymes, but in different degrees. Endo-beta-1,4-glucanase and beta-glucosidase were markedly inhibited when melanin was preincubated with enzyme, while exo-beta-1,4-glucanase was severely inhibited when melanin was preincubated with substrate. The latter was found to be dependent on the contact time. The activities of endo-beta-1,4-glucanase and beta-glucosidase were noncompetitively inhibited by melanin.  相似文献
