The goal of this research is to enhance our knowledge of thecontributions of doliolids to the planktonic community as consumersand secondary producers. The objectives are to quantify feedingand growth rates of Dolioletta gegenbauri gonozooids at fourfood concentrations and four temperatures in order to determinetheir impact as grazers throughout the water column. Althoughdoliolids are abundant in numerous regions of the coastal ocean,and are considered to be major planktonic grazers, data on ratesof feeding and growth are scarce. Laboratory experiments wereconducted at 16.5, 20, 23.5 and 26.5°C to quantify removalof a 50:50 volumetric concentration of Thalassiosira weissflogiiand Rhodomonas sp. at four different food concentrations of20, 60, 160 and 390 µg C l–1. Results from theseexperiments suggest that clearance rates are similar at concentrationsfrom 20 to 60 µg C l –1, and decrease as the foodconcentrations increase to 160 and 390 µg C l –1.The ingestion rates increase over a range of phytoplankton concentrationsfrom 20 to 160 µg C l –1, then decrease when abnormallyhigh concentrations of 390 µg C l –1 are offered.Clearance and ingestion rates increase as temperature increasesfrom 16.5 to 26.5°C. The exponential growth rates rangefrom k = 0.2–0.7, with the lowest rates occurring at thehighest food concentration. Growth rates increase with increasingtemperature from K = 0.1–0.3 at 16.5°C to 0.45–0.7at 26.5°C. In each case, the small- and medium-sized zooidshad higher growth rates than the larger gonozooids. These resultssuggest that doliolid feeding and growth rates are a functionof environmental food concentrations and temperatures, and implythat they can be important consumers in a changing neritic environment.  相似文献

Mannitol-1-phosphate dehydrogenase (EC 1.1.1.17 [EC] ) and mannitol-1-phosphatase(EC number yet unassigned) were detected in the brown algae,Spatoglossum pacificum and Dictyota dichotoma. The enzymes wereextracted from algal fronds and their properties were investigatedusing partially purified preparations. Mannitol-1-phosphatase shows maximum activity at pH 7. The enzymehad a narrow substrate specificity. The Km value for mannitol-1-phosphateis 8.3x10–4 M (30°C, pH 7.0). The enzyme is activatedby Mg++ and Mn++and is strongly inhibited by PCMB, Hg++and NaF. Mannitol-1-phosphate dehydrogenase showed maximum activitiesat pH values 6.5 and 10.2 in reductive and oxidative reactions,respectively. The dehydrogenase also showed narrow substratespecificity; mannitol-1-phosphate and NAD or fructose-6-phosphateand NADH2 are utilized, respectively, in oxidative and reductivereactions by the enzyme. Km values for these substrates andthe coenzymes are 2.5x10–4 M and 7.1x10–5 M forthe first pair and 2.8x10–4 M and 1.3x10–5 M forthe latter pair. This enzyme was strongly inhibited by PCMBand Hg++, but was only slightly affected by adenosine phosphates. The biosynthetic pathway from D-glucose to L-(+)-tartaric acid(TA) in detached leaves of the bean, Phaseolus vulgaris L.,was studied in three cultivars, two of which were known to containTA and one of which lacked TA, with the aid of several putativeradiolabeled intermediates, namely D-[l-14C]glucose, D-[6-14C]glucose,D-[U-14C]glucose, D-[U-14C]gluconate, L-[U-14C]-ascorbic acid,L-[l-l4C]idonate, D-xylo-5-[U-14C]hexulosonate, D-xylo-5-[l-14C]hexulosonate,D-xylo-5-[6-l4C]hexulosonate and L-[U-l4C]threonate. D-[U-14C]Glucoseand D-[U-l4C]gluconate were converted to TA with low isotopicyield but this yield was further reduced when leaf tissues weresupplied with unlabeled D-gluconate or D-xylo-5-hexulosonate.D-xylo-5-[U-14C]Hexulosonate and D-xylo-5-[l-14C]hexulosonatewere good precursors of TA. D-xylo-5-[6-14C]Hexulosonate didnot furnish 14C to TA. Addition of a metabolic product of D-xylo-5-hexulosonate(which was labeled by D-xylo-5-[l-14C]hexulosonate but not byD-xylo-5-[6-14C]hexulosonate) to leaves labeled with D-xylo-5-[l-14C]hexulosonatedoubled the incorporation of 14C into TA. L-[U-14C]Ascorbicacid, L-[l-14C]idonate and L-[U-14C]threonate failed to producelabeled TA. A metabolic scheme to accommodate these observationsis presented. Using permeabilized characean cells in which the ionic conditionsat the cytoplasmic side of the tonoplast are easily controlled,effects of Ca2+ ion on tonoplast potential were examined. Whenthe cell was treated with 1 µM Ca2+, the tonoplast potential(EM became positive in a complicated manner in Chara corallinawhile it simply became negative in Nitella axilliformis. Whenthe cell was treated with 9-antracenecarboxylic acid, a Cl-channelinhibitor, Em became more negative and the response of Em toCa2+ was significantly suppressed. In a small temperate lake of the southern Andes, Bosmina longirostrisand Ceriodaphnia dubia coexist with the predaceous water miteLimnesia patagonica. Sampling of natural populations and laboratoryexperiments were carried out. The field population of Limnesiadid not show a numerical response to the density or biomassof its prey. Laboratory experiments showed that the water miterejected C.dubia adults and juveniles as prey, but consumedBosmina. The maximum predation rate was 40 prey predator–1day–1 and a linear relationship between predation rateand prey density was obtained (R2 = 66%). The contribution ofmortality due to predation and the predation risk are too lowto provoke a prey suppression. By means of computer modelling,the densities of predator necessary to explain half of the totalprey mortality were calculated. These densities were one ortwo orders of magnitude higher than those in the field. It isconcluded that Limnesia could really be a suppressor, but thepotential depends greatly on its density.  相似文献

Laboratory experiments were carried out on the holoplanktonicscyphomedusa Pelagia noctiluca, which exhibits population explosionssome years in the Mediterranean Sea. Feeding experiments performedon small, laboratory-reared medusae showed specific daily rationsof 13 and 35% at 5 and 20 Anemia nauplii prey 1–1 respectively,while basal NH+4 excretion reached 3.9% (specific elementalmetabolic rate) for adults at 22°C. These values appearto be consistent with those previously observed by several authorsfor other planktonic coelenterates. Assuming that such laboratoryphysiological rates are indicative of food requirements in naturalenvironments, and given the high abundance of P. noctiluca observedregionally, we conclude that P. noctiluca can have a strongpredatory impact in the Ligurian Sea (north-west Mediterranean),particularly during the summer and early fall. 3Present address: ORSTOM B.P. 2528, Bamako, Mali  相似文献

The growth rates of four saline-lake diatom taxa were measuredunder varying conditions of salinity (5, 8 and 11), brine type(sulfate- versus bicarbonate-dominated) and nitrogen form (NH4+versus NO3), using a full factorial design. With NO3as the nitrogen source, Cyclotella quillensis, Cymbella pusillaand Anomoeoneis costata exhibited lower growth rates in thesulfate versus bicarbonate media. The strain of Chaetoceroselmorei used in these experiments, isolated from a sulfate-dominatedlake, was unable to grow on NO3 alone. In the NH4+ treatments,neither salinity nor brine type affected the growth rates ofC.quillensis or C.elmorei. When supplied with NH4+, C.pusillaand A.costata had higher growth rates in the bicarbonate versussulfate media, although for C.pusilla the difference on NH4+was not as great as on NO3. The impact of brine typeon NO3 use is consistent with the theory that sulfateinhibits molybdate uptake, as molybdenum is required for NO3use but not NH4+. Cymbella pusilla was the only taxon affectedby changes in salinity. The four taxa used in these experimentsare frequently found in saline lakes and saline-lake sediments,hence they are used in paleoclimate reconstructions; the resultspresented here provide additional information that may enhancethese diatom-based reconstructions.  相似文献

Rotifers are a relatively well-studied component of lacustrinesystems but their role is only poorly understood in estuaries.Three species of the genus Synchaeta—S. baltica, S. triophthalmaand S.cecilia—dominate the cold-water assemblage of rotifersin Chesapeake Bay. Laboratory experiments were conducted todetermine the temperature dependence of egg development time(EDT) for each species; EDT varied over an approximate rangeof 90–9 h as temperature (T) varied from 2 to 22°C.The EDT/temperature relationships could be closely fitted bya simple polynomial equation of the form log(EDT) = a + b(logT) + c(log T)2 for each species. Natural populations of thesethree rotifers were sampled during a cruise in the Potomac River(7–11 March 1983). Estimates of specific reproductiverates (b) were calculated based on the previously defined EDT/temperaturerelationship and the observed ratio of eggs/rotifers for eachspecies. The two most abundant species, S.triophthalma and S.cecilia,showed a clear dependence of b on the observed chlorophyll aconcentrations. Maximum reproductive rates ({small tilde}0.015h–1) were attained only at relatively high phytoplanktondensities within a bloom of Heterocapsa triquetra where thechlorophyll a concentrations exceeded 10 µg l–1.Estimates of secondary production suggest that Synchaeta spp.may contribute to the trophic flow of carbon in this systemwith a significance at least similar to that of the planktoniccopepods.  相似文献

Laboratory experiments were conducted to determine whether thetwo cyclopoid copepods. Cyclops vicious and Mesocyclops leuckaru.exploit the same food resources. The food requirements of juvenilesof the two cyclopoid copepods were investigated. Moreover, theimportance of algae for the predaceous adults was studied. Naupliiof both M leuckaru and C.vicinus successfully developed intocopepodites when fed the motile algae Chlamydomonas reinhardtii.Chlamydomonas sphaeroides and Cryptomonas sp. Threshold foodconcentrations for naupliar development varied between offeredalgae and between the two cyclopoid species. The food thresholdfor successful naupliar development, when reared on C.reinhardui,was lower for M.leuckarti (0.3 mg C 1–1) than for C.vicinus(0.5 mg C l–1) whereas a similar food threshold was foundusing Cryptornonas sp (0.3 mg C –1) and C.sphaeroides(<0.2 mg C 1–1), Naupliar development time was inverselyrelated to food concentration. Food required for copepoditedevelopment differed for the two cyclopoid species. Cyclopsvicinus was able to develop to the adult stage on a pure dietof any one of the three algal species. whereas M.leuckarti requireda prey supply of the rotifer Brachionus rubens. Food composition.i.e. algal species, algal concentration and rotifer abundance,influenced copepodite survivorship of both cyclopoids and wasalways higher in the presence of B.rubens. Under similar foodconditions, mortality was higher for M.leuckarti than for Cvicinus. Mesocyclops leuckaru females were very dependent onanimal food. The predation rate of M.leuckaru was not lowerin the presence of algae. Egg production of M.leuckarti waslow on a pure algal diet and significantly higher when B rubenswas present. The results were used to discuss the life cyclestrategy and the possibility of exploitative competition ofthe two cyclopoid copepods.  相似文献

The photosynthetic production of oxalic acid in Oxalis corniculata   总被引:2,自引:0,他引:2  
The biogenesis of oxalic acid in Oxalis corniculata has beeninvestigated. In O. corniculata the bulk of the oxalic acidis produced by CO2 fixation both in light and in darkness butthe rate of its photosynthetic formation is much higher thanin darkness. Several other plants some of which are known toaccumulate oxalic acid e.g., Biophytum sensitivum, Averrhoacarambola, Impatiens balsamina, Amorphophallus campanulatusand Colocassia antiquorum also fix 14CO2 into oxalic acid photosyntheticallywithin 1 min of exposure to the gas. In O. corniculata 14C canbe detected in oxalic acid within 5 sec and about half of thetotal 14C fixed in the 70% ethanol soluble fraction can be locatedin this compound after 5 min. This is accompanied by a declineof radioactivity in two compounds, the chromatographic behaviourand melting points of one of which and its DNP hydrazone aresimilar to those of an authentic sample of glyoxylic acid. Whenglyoxylate 1, 2-14C is incubated with Oxalis leaf homogenateit is converted to oxalate-14C. Glycolate is also metabolizedto oxalate. The conversion of both glycolate and glyoxylateare favoured by light. The C2 compounds acetate and glycinehowever are utilized rather poorly. Young, A. J., Collins, J. C. and Russell, G. 1987. Ecotypicvariation in the osmotic responses of Enteromorpha intestinalis(L.) Link.—J. exp. Bot. 38: 1309–1324. The physiological basis for salt tolerance has been studiedin the euryhaline marine alga Enteromorpha intestinalis. Adaptationto dilute and concentrated seawaters has been investigated inthree separate populations of this alga: marine, rock pool andestuarine. Internal K+, Na+ and Cl levels have been determined usingtracer efflux analyses. K+ has been shown to be the major osmoticsolute within this alga. Cellular levels of Cl and, inparticular, Na+ are low although levels in the cell wall arehigh. Levels of these ions varied considerably between the separateplants; K+ levels within marine plants of E. intestinalis aretwo to four times those found in the other populations. Thetertiary sulphonium compound ß-dimethylsulphonio-propionateis maintained at relatively high levels, although it remainsfairly insensitive to change in the external salinity. Changes in the tissue water content and cell volume are large,particularly within the estuarine plants. The thin cell wallsof these plants allow large changes in volume in the diluteconditions experienced in an estuary, while low turgor preventscell rupture. Thicker cell walls and small cells of the marineand rock pool plants assist in tolerating high and low externalosmotic potential—the estuarine plants respond poorlyto concentrated seawater. Equipment is described which delivers air with concentrationsof CO2 and water vapour closely controlled in the ranges 0 to2500 ppm and 5 to 15 mb respectively, at flow rates of up to10 1 min-1, to each four leaf chambers. The leaf temperatureis controlled to ±0.5 °C and, with a light intensityof 0.3 cal cm-2 min-1 visible radiation (0.4 to 0.7 µm)leaf temperature can be maintained at 17.5 °C.The apparatusused to measure the concentration differences between the watervapour and CO2 entering and leaving the leaf chamber (used tocalculate transpiration, photosynthetic, and respiration rates)is described in detail.Results of tests, which show the necessityfor mounting a fan within the leaf chamber, are reported.Typicallight- and CO2-response curves are given for kale leaves (Brassicaoleracca var. acephala) and an attempt is made to quantify theerrors in the measurement of photosynthesis and transpiration.  相似文献

Rates of CO2 and HCC3 fixation in cells of various Chlorellaspecies in suspension were compared from the amounts of 14Cfixed during the 5 s after the injection of a solution containingonly 14CO2 or H14CO3. Results indicated that irrespectiveof the CO2 concentration during growth, Chlorella vulgaris 11h and C. miniata mainly utilized CO2, whereas C. vulgaris C-3,C. sp. K. and C. ellipsoidea took up HCO3 in additionto CO2. Cells of C. pyrenoidosa that had been grown with 1.5%CO2 (high-CO2 cells) mainly utilized CO2, whereas those grownwith air (low-CO2 cells) utilized HCO3 in addition toCO2. Cells that utilized HCO3 had carbonic anhydrase(CA) on their surfaces. The effects of Diamox and CA on the rates of CO2 and HCO3fixation are in accord with the inference that HCO3 wasutilized after conversion to CO2 via the CA located on the cellsurface. CA was found in both the soluble and insoluble fractions;the CA on the cell surface was insoluble. Independent of the modes of utilization, the apparent Km (NaHCO3)for photosynthesis was much lower in low-CO2 cells than in high-CO2ones. N-Acetyl-[3H]glucosamine supplied to intact 3 d old castor beanendosperm tissue was incorporated into TCA-insoluble productpresumed to be glycoprotein. After an incubation time of 2 hthe major paniculate location of this product within the cellwas the endoplasmic reticulum. Cell-free preparations containingparticulate enzymes transferred N-acetyl-[14C]glucosamine fromUDP-N-acetyl-[14C]glucosamine into a fraction soluble in chloroform/methanol(2: 1, by vol), a fraction soluble in chloroform/methanol/water(10: 10: 3, by vol.), and an insoluble residue. Mild acid hydrolysisreleased the saccharide moieties from the lipids. Paper chromatographicanalysis of the released saccharides established that the C/M-solubleproducts contained both N-acetyl-[14C]glucosamine and N, N'-diacetyl-[14C]chitobiose.In contrast, N-acetyl-[14C]glucosamine released from the C/M/W-solubleproduct was contained in an oligosaccharide, probably in associationwith unlabelled mannose residues. The stimulatory effect ofdolichol monophosphate and the inhibitory effect of tunicamycinon saccharide-lipid synthesis indicated that N-acetyl-glucosamineis transferred to a glycopolymer by the established reactionsof the dolichol monophosphate pathway. The enzymes catalysingthe constituent reactions of this pathway were exclusively locatedin the ER.  相似文献

The influx and efflux of Na+ across the plasma membrane of Characorallina and Chara longifolia were examined under mild saltstress conditions. Na+ influx was found to be rapid in bothspecies with the freely exchangeable cytoplasmic Na+ cominginto isotopic equilibrium with external 22Na+ within 1 h ofexposure to isotope. Cytoplasmlc Na+ concentration and Na+ influxwere greater in C. corallina than in C. longifolla under thesame conditions. Na+ influx across the tonoplast was much lowerthan the flux across the plasma membrane. Na+ efflux was stimulatedat pH 5 relative to pH 7 by 218% in C. coralllna and 320% inC. longifolia. In both species externally applied Li+ inhibitedNa+ efflux at pH 5 but not at pH 7. Na+ etflux was not significantlyinhibited by amiloride. Four methods of determining the substrate requirements for synthesisof a kiwifruit [Actinidia deliciosa (A. Chev.) C. F. Liang etA. R. Ferguson var. deliciosa cv. Hayward] berry were comparedusing data derived from common kiwifruit berry samples collectedfrom anthesis to fruit maturity. The four methods were basedon fruit proximal analysis, elemental analysis, heats of combustion,or tissue carbon content. All methods gave similar patternsof seasonal costs and values of final cost to the plant (mean1.21 g glucose g–1 season–1) but there was lessagreement for growth respiration (mean 0.147 g glucose g–1season–1). This is the first time that a continuous recordof growth cost over the course of development has been presented,and the trends in seasonal cost reflect the uptake into andsynthesis of the different biochemical constituents in the fruit.The differences between the results of each method reflect theunderlying assumptions used in their development. A cDNA for the phytochrome of the fern Adiantum capillus-venerisL. was cloned and sequenced. The deduced phytochrome is 50{smalltilde}55% identical to phytochromes of seed plants, and 68%identical to Selaginella phytochrome. Short-term studies were undertaken of source and sink relationshipsin Pisum sativum, cultivar Orfac, plants grown in a controlledenvironment. 14C-distribution assays were conducted after 24or 48 h 14Cphotoassimilate translocation from a single leafsubtending either a vegetative or a reproductive node. The primaryallocation of 14C-assimilate was achieved within 24 h: therewas no significant secondary movement of 14C within the subsequent24 h. The pod was strongly but not exclusively dependent onthe subtending leaf. Out of the total 14C fixed by a leaf theproportion that was exported within 24 h was related to the14C-sink capacity of the pods. A rapid non-combustive 14C-assay method is described whereby14C-tissue fragments are rendered translucent prior to directscintillation counting of the sample. In terms of cpm per mgobtained the latter method was comparable to a wet combustionmethod adapted for insoluble 14C-tissue.  相似文献

Two planktonic cyclopoid copepods (Tropocyclops prasinus andMesocyclops longisetus) were raised in the laboratory to obtainlife history information (duration of embryonic and post-embryonicdevelopment, reproductive performance, longevity, and stage-specificlength and weight values). Animals were maintained at 20 and25°C, and fed ad libitum. Development times were temperaturedependent when food was not limiting, with shorter periods ofembryonic and post-embryonic development and decreased longevityat 25°C. Laboratory data on the duration of developmentand biomass, together with population dynamics data obtainedin the field, were used to estimate summer and winter biomassand production of these species in a shallow reservoir, LagoaDourada, Brazil. The maximum production rate of T. prasinus,attained during summer, was 2.8 mg dry weight (DW) m–3day–1 and the highest daily production:biomass (P:B) ratiowas 0.29, whereas for M. longisetus the maximum production ratewas 1.4 mg DW m–3 day–1 and the highest daily P:Bratio was 0.39, in the winter. Over short time intervals (everyother day), there was great variability of the species productionrates. Species production rates were low compared to valuesreported in the literature for the same or other species ofequivalent sized copepods from both tropical and subtropicalregions.  相似文献
