We used quantitative histochemistry to investigate the tissue-specific compartmentation of potassium in the laminar pulvinus of Phaseolus coccineus L. at day and night positions of diurnal leaf movement. The assay was based on the potassium-dependent activation of pyruvate kinase. Total potassium levels of pulvini were higher in the light than in the dark [0.88 and 0.57mol (kg dry weight)?1, respectively]. Transverse compartmentation of potassium was studied on three tissue slices, representing the middle part, petiolar and laminar sides of individual pulvini. These were dissected further into 10 distinct subsamples (bundle; motor tissues: extensor, flexor; flanks). In the day position the amount of potassium in the extensor was higher than in the night position [1.92 and 1.50 mol (kg dry weight)?1, respectively]. Flexor changes were opposite [1.13 and 1.65 mol (kg dry weight)?1, respectively]. In the day position there was a steep and consistent increase in potassium content from the innermost to the outermost zones of the extensor. In the night position this was much more variable. Comparable gradients were not detected in flexor samples. Here highest amounts of potassium were recovered from the middle of the motor tissue. The data specify distinct tissue regions involved in osmotic adjustment during leaf movement in Phaseolus coccineus.  相似文献

The circadian movement of the lamina of primary leaves ofPhaseolus coccineus L. depends on circadian changes of the K+, Cl- and (depending on the Cl- availability) malate content in the swelling and shrinking motor cells of the laminar pulvinus. After sowing in soil, the laminar pulvinus develops within about 26 days. When the leaves emerge from the soil (about 6 days after sowing) and the pulvinus starts with the diurnal movement (about 9 days after sowing) the pulvinar dimensions are about half of those of the mature pulvinus. The anatomical structure, however, is basically the same as in the developed pulvinus. In soil-grown plants, the K+, Cl- and malate content as well as the period length of the circadian leaf movement rhythm change in the developing pulvinus. In the embryo of the dry seed, the Cl- content is low (0.03 mmol g-1 DW), the K+ content, however, 22-fold higher than the Cl- content. When the leaves emerge from the soil, the pulvinar K+ and Cl- content is the same as in the whole embryo of the dry seed. In the developing pulvinus the K+ content increases by a factor of 2 and the Cl- content by a factor of 41 in the mature pulvinus. The pulvinar malate content increases between the 6th and 10th day from about 40 to 180Μmol g-1 DW, then decreases until the 17th day and remains thereafter on a low level (around 80 Μmol g-1 DW). These results indicate that the Cl- availability increases in the developing pulvinus with age. It explains furthermore why in young leaves malate was found as counterion to K+ in the osmotic leaf movement motor, in older ones, however, Cl-. The circadian leaf movement starts 9 days after sowing. The period length decreases during the development of the pulvinus from 31.3 to 28.6 h in leaves of intact soil-grown plants. In leaves which were cut from the plants and immersed with their petioles in distilled water, the age dependent decrease of the period length is also found. However, the period lengths are shorter by more than 1 h than in the leaves of intact plants. The increasing Cl- availability in the developing pulvinus does not seem to be the cause for the age dependent shortening of the period length, because the period length in 22 days old Cl- deprived pulvini is the same as in 22 days old pulvini with a high Cl- content.  相似文献

The location and some morphological, anatomical, and functional aspects of the gravity-sensitive pulvini of a selected number of grass shoots are examined. There are two distinct gravity-sensitive regions near the nodal regions of Gramineae. One, the leaf sheath pulvinus, is located at the base of the sheathing leaf bases, and is characteristic of the subfamily Festucoideae. The other, the internodal pulvinus, is located at the base of the internode, a little above the nodal joint. Most members of the Panicoideae possess internodal pulvini, in addition to more or less developed leaf sheath pulvini. Three members of the Oryzoideae examined possess leaf sheath pulvini only, while Phragmites australis (Arundinoideae) possesses both leaf sheath and internodal pulvini. Leaf sheath pulvini of some grasses develop hairs, cork-silica cell pairs or stomatal apparatuses over the epidermis while many others are devoid of any such idioblasts. Both the leaf sheath and internodal pulvini of all grasses examined preferentially exclude, or accumulate very little silica, whereas the regions of the shoot immediately above and below the pulvini in these same grasses accumulate large quantities of silica. Pulvini remain unsilicified or poorly silicified throughout their life and even after several days following geotropic bending. Pulvini are also distinguished from the regions above and below them by the lack of lignin in the bundle cap cells. Lignin is found only in the xylem vascular tissue, and this consists of annular and helical vessel elements only. The bundle cap cells are rich in pectin and are described as collenchymatous. All pulvini possess specialized zones of cells containing starch statoliths. In response to horizontal displacement of the shoots, the lower side of the pulvini grows by cell elongation only. The collenchymatous cells stretch in a manner that results in alternately thin and thick regions of cell wall.  相似文献

The activities of enzymes which catalyze one step in each of the five major carbon pathways in green plants were measured in secondary pulvini and other tissues of Phaseolus coccineus L. leaves. We were able to detect activities of fumarase (EC 4.2.1.2; tricarboxylic-acid pathway), NAD-glyceraldehyde-phosphate dehydrogenase (NAD-GAPDH, EC 1.2.1.12; glycolysis), 6-phosphogluconate dehydrogenase (6-PGDH, EC 1.1.1.44; oxidative pentose-phosphate pathway), ribulose-1, 5-bisphosphate carboxylase (Rubisco, EC 4.1.1.39; photosynthetic carbon-reduction pathway), and of hydroxypyruvate reductase (HP-R, EC 1.1.1.81; photosynthetic carbon-oxidation pathway). On a protein basis the activities of Rubisco and HP-R in pulvinar regions were very low (below 1 and 2 mol · (kg protein) –-1 · h–-1, respectively), but the activities of fumarase and NAD-GAPDH were between 10- and 5-fold higher compared with the laminar tissue (up to 7 and 50 mol · (kg protein)–-1 · h–-1, respectively). Similarly, the protein specific activities of 6-PGDH were increased in the pulvinus (3–4 compared with approx. 1 mol · (kg protein)–-1 · h–-1 in the leaf blade). No differences in specific activities were detected between day and night positions of the leaves. By applying quantitative histochemical techniques we determined the longitudinal and transversal compartmentation of the activities of fumarase, NAD-GAPDH, and 6-PGDH in pulvinar tissues. Levels of activity of all three enzymes increased towards the middle part of the pulvinus. Here, expressed on a dry-weight (DW) basis, the analysis of cross sections showed highest activities in the outer parts of the extensor in the order given, approx. 0.6, 5, and 0.25 mol · (kg DW)–-1 · h–-1 for fumarase, NAD-GAPDH and 6-PGDH. When related to protein, levels of activity were comparably high within the inner parts of extensor and flexor, and partly also in the abaxial part of the bundle (fumarase, 6-PGDH). The tissue-specific compartmentation of the respective activities is discussed in relation to leaf movement and shows parallels with guard-cell function.Abbreviations Chl chlorophyll - DW dry weight - GAPDH glyceraldehyde-phosphate dehydrogenase - HP-R hydroxypyruvate reductase - Rubisco ribulose-1,5-bisphosphate carboxylase - 6-PGDH 6-phosphogluconate dehydrogenase This investigation was supported by a grant from the Deutsche Forschungsgemeinschaft.  相似文献

The incorporation of 14C into sucrose and hexose phosphates during steady-state photosynthesis was examined in intact leaves of Zea mays L. plants. The compartmentation of sucrose synthesis between the bundle sheath and mesophyll cells was determined by the rapid fractionation of the mesophyll and comparison of the labelled sucrose in this compartment with that in a complete leaf after homogenisation. From these experiments it was concluded that the majority of sucrose synthesis occurred in the mesophyll cell type (almost 100% when the time-course of sucrose synthesis was extrapolated to the time of 14C-pulsing). The distribution of enzymes involved in sucrose synthesis between the two cell types indicated that sucrose-phosphate synthetase was predominantly located in the mesophyll, as was cytosolic (neutral) fructose-1,6-bisphosphatase activity. Stromal (alkaline) fructose-1,6-bisphosphatase activity was found almost exclusively in the bundle-sheath cells. No starch was found in the mesophyll tissue. These data indicate that in Zea mays starch and sucrose synthesis are spatially, separated with sucrose synthesis occurring in the mesophyll compartment and starch synthesis in the bundle sheath.  相似文献

Legume pulvini have a clearly delimited endodermis, whose variable content has been associated with the velocity and type of leaf movement: pulvini in leaves with fast nastic movement contain starch grains; pulvini in leaves with slow nastic movements have calcium oxalate crystals as well as starch grains in the endodermis. However, the studies carried out to date have involved few legume species. This study therefore purported to examine the consistency of this hypothesis in other legumes. Thus, the structure and content of the pulvinus endodermal cells of nine legumes of the Brazilian cerrado, with different types and velocities of leaf movement, were investigated: slow nyctinastic and heliotropic movements ( BAUHINIA RUFA, COPAIFERA LANGSDORFFII, SENNA RUGOSA - Caesalpinioideae; ANDIRA HUMILIS and DALBERGIA MISCOLOBIUM - Faboideae; STRYPHNODENDRON POLYPHYLLUM - Mimosoideae), slow heliotropic movement ( ZORNIA DIPHYLLA - Faboideae), and fast seismonastic and slow nyctinastic and heliotropic movements ( MIMOSA RIXOSA and MIMOSA FLEXUOSA - Mimosoideae). Samples were prepared following standard plant anatomy and ultrastructure techniques. The endodermis of all the species contains starch grains. In the species displaying only slow movements, calcium oxalate prismatic crystals were observed in addition to starch grains, except in ZORNIA DIPHYLLA. In conclusion, oxalate crystals occur only in endodermal cells of pulvini that display slow movements, while starch grains are always present in pulvinus endodermal cells of plants with any kind of movement.  相似文献

6-benzylaminopurine (BAP) delays leaf abscission of soybeanGlycine max (L.) Merr. Abscission of the distal pulvinus ofprimary leaves was induced in 12-d-old seedlings or explantsby removal of the leaf blade. BAP applied to the cut end ofthe pulvinus following leaf blade removal delayed abscission.Discoloration of the pulvinus occurred before abscission commencedand the number of grana in chloroplasts within cortical parenchymacells of the pulvinus decreased over time following leaf bladeremoval. BAP prevented discoloration of pulvinus tissues anda decrease in grana number. Starch grains within amyloplastsof cells of the starch sheath in the pulvinus disappeared followingleaf blade removal, whereas starch accumulated within the abscissionzone prior to abscission. BAP prevented this apparent redistributionof starch and instead promoted an increase in starch withinplastids of cortical parenchyma cells of the pulvinus. Duringthe abscission process, cells within the separation layer enlargedand their nuclei and nucleoli became more evident prior to theirseparation from one another. Cell separation resulted from breakdownof middle lamellae and partial degradation of primary cell walls.Cycloheximide applied directly to the external surface of theabscission zone inhibited abscission in a similar way to theBAP treatment. These results suggest that BAP prevents abscissionby altering patterns of starch distribution in the pulvinusand abscission zone and by inhibiting the synthesis of proteinsthat typically appear de novo in induced abscission zone tissues. The energy requirements of the various phases of the circadian clock in the laminar pulvini cells of primary leaves of Phaseolus coccineus L. were investigated using 4-h pulses of NaCN (5 mM) and NaN3 (1 mM). The induced phase shifts were calculated from the timing of the subjective night position during the third cycle after the treatment. Both inhibitors produce advances during phases which are correlated with the upward movement of the leaf (ca. 0–12 h after the maximum of the subjective night position) and during phases which are correlated with the downward movement of the leaf (ca. 20–28 h after the maximum of the subjective night position). Maximal advances are induced during the phase which is correlated with the maximum of the subjective night position (hour 0), whereas during phases which are correlated with the subjective day position (ca. 12–20 h after the maximum of the subjective night position) the inhibitors have no effect or induce only small advances. These results demonstrate that the part of the circadian cycle which, according to Bünning's tension-relaxation model of the circadian clock, is characterized by features of relaxation, represents a sequence of phases with decreasing energy requirement, whereas the tension part of the circadian cycle represents a sequence of phases with increasing energy requirement. The energy requirement for changing and maintaining the leaf positions was investigated by continuously offering NaCN, NaN3, and dinitrophenol (DNP) to leaves with intact and half (flexor cut away) pulvini. The substances inhibit in both pulvini the upward movement or induce a downward movement, depending on the leaf position, when the transfer to the inhibitor solution takes place. These results give evidence that the movement of intact pulvini reflects the turgor (volume) state of the extensor cells and that the increase of turgor (volume) and high turgor (volume) state requires more energy than the decrease of turgor (volume) or low turgor (small volume) state. Therefore, the time course of the energy requirements of the circadian clock and the clock-controlled turgor (volume states or leaf movement) is out of phase during a circadian cycle. Consequently the reaction of the clock-controlled leaf movement to the reduced energy supply can mask the clock behavior in pulse and step experiments. The phase response curves towards CN- and N 3 - reflect the time course of the CN--induced membrane depolarizations (the energy requirement of the electrogenic pump) in extensor cells of the pulvinus (Freudling et al. (1980), Plant Physiol. 65, 966–968), and both are out of phase with the time course of the energy requirement of the turgor. Consequently it is hypothesized that in Phaseolus advances are due to membrane depolarization and that at least in this organism electric properties of the plasmalemma are essentially involved in the mechanism of the circadian clock.Abbreviations LD light-dark cycle - LL continuous light - DNP dinitrophenol This paper is dedicated to Professor Erwin Bünning on the occasion of his 75th birthdayIn this paper zero corresponds to the second maximum of the subjective night position of the leaves after transfer to constant conditions. Zero to twelve hours corresponds approximately to the upward movement of the leaves, 12–20 h to the elevated (subjective day) position, and 20–28 h to the downward movement of the leaves. In other circadian systems Pittendrigh's CT (circadian time) convention is used. CT 00 is the time of dawn after a 12-h light/12-h dark cycle. Since in Phaseolus the plants are raised in a LD cycle different from 12:12 and since the phases at dawn differ considerably from leaf to leaf and are furthermore not precisely determinable (whereas the subjective night position of the leaves is a well-defined and recognizable phase) this convention is not followed in Phaseolus. Phase zero in Phaseolus corresponds to approximately CT 18 in other systems  相似文献

Summary Using energy-dispersive X-ray microanalysis, the concentrations of ions, especially potassium and chlorine, were determined in different tissues of primary and tertiary pulvini of Mimosa pudica. It was shown that stimulating the leaf was followed by ion displacements which were most striking in the outer extensor cells, resulting in turgor loss. Since Ca concentration remains relatively constant in cell walls of collapsed cells, the changes of K concentration are best described by the K:Ca ratio. After stimulation the K:Ca ratio dropped in the outer extensor of the primary pulvinus from 775.3 to 2.37 in the cytoplasm, and from 542.2 to 9.25 in the cell wall. Changes in chlorine content were less striking in the primary pulvinus. The KCl ratios in some cases were lower than 1.0, which indicates that Cl content can increase, while K content is diminished. In the non-stimulated tertiary pulvini the outer extensor cells show high concentrations of Cl, but much lower Cl concentrations were found after stimulation. In contrast to the primary pulvinus the K content of the tertiary pulvini is very low. In the vascular tissues of both primary and tertiary pulvini stimulation is followed by a release of K and Cl out of the sieve element cytoplasm into the apoplast. K then appears accumulated in the cell walls of the collenchymatous tissue. These displacements lead to the assumption that the collenchymatous apoplast temporarily functions as a reservoir for K and to a lesser extent for Cl. With regard to the mechanism of leaf movement after stimulation, the accumulation of ions in the apoplast seems to be initiated by the decrease of water potential triggered by an apoplastic accumulation of unloaded sucrose (Fromm and Eschrich 1988a). The resulting turgor release in the outer extensor is accompanied by an efflux of ions.Supported by the Deutsche Forschungsgemeinschaft  相似文献

The accurate measurement of fructose 2,6-bisphosphate from plants such as wheat is fraught with difficulty. Extraction and assay methods for fructose 2,6-bisphosphate that give near 100% recovery of the metabolite, and a linear response with volume have therefore been developed for extracts prepared from wheat leaves of different ages. Amounts of fructose 2,6-bisphosphate in different regions of leaves generally showed a positive correlation with chlorophyll content. Measurements of sucrose and starch in third leaves harvested at different times of the diurnal cycle demonstrated that sucrose is the major form in which photosynthate is stored in the leaf, but starch can account for up to about 30% of the stored carbohydrate. Virtually all of the carbohydrate accumulated as starch and sucrose during the day was degraded at night. Amounts of fructose 2,6-bisphosphate were generally lower in extracts prepared from leaves harvested in the light than in the dark. Additionally, there was no change in either the amount of fructose 2, 6-bisphosphate or the ratio of sucrose to starch in samples prepared from leaves harvested at different times of the day. These results are broadly consistent with a role for fructose 2,6-bisphosphate in the regulation of sucrose synthesis and the partitioning of carbohydrate between sucrose and starch in wheat leaves.  相似文献

The compartmentation of solutes in suspension cells of Saccharum sp. during different growth phases in batch culture was determined using CuCl2 to permeabilize the plasma membrane of the cells. The efflux of cytosolic and vacuolar pools of sugars, cations and phosphate was monitored, and the efflux data for phosphate were compared and corrected using data from compartmentation analysis of phosphate as determined by 31P-nuclear magnetic resonance spectroscopy. The results show that sucrose is not accumulated in the vacuoles at any phase of the growth cycle. On the other hand, glucose and fructose are usually accumulated in the vacuole, except at the end of the cell-culture cycle when equal distribution of glucose and fructose between the cytosol and the vacuole is found. Both Na+ and Mg2+ are preferentially located in the vacuoles, but follow the same tendency as glucose and fructose with almost complete location in the vacuole in the early culture phases and increasing cytosolic concentration with increasing age of the cell culture. Potassium ions are always clearly accumulated in the cytosol at a concentration of about 80 mM; only about 20% of the cellular K+ is located inside the vacuole. Cytosolic phosphate is little changed during the cell cycle, whereas the vacuolar phosphate pool changes according to total cellular phosphate. In general there are two different modes of solute compartmentation in sugarcane cells. Some solutes, fructose, glucose, Mg2+ and Na+, show high vacuolar compartmentation when the total cellular content of the respective solute is low, whereas in the case of ample supply the cytosolic pools increase. For other solutes, phosphate and K+, the cytosolic concentration tends to be kept constant, and only excess solute is stored in the vacuole and remobilized under starvation conditions. The behaviour of sucrose is somewhat intermediate and it appears to equilibrate easily between cytosol and vacuole.Abbreviation NMR nuclear magnetic resonance The very cooperative help by Dr. J. Summary After illumination intact leaves of Zea mays contain sucrose and starch. The latter is located mainly in the bundle sheath cells. When 0.5 mm wide leaf strips are incubated with sucrose solution, the starch deposit in the bundle-sheath chloroplasts is greatly increased by light. When isolated bundle sheath cells are suspended in water or solutions of sucrose and various metabolites they are not capable of synthesizing starch. An appreciable production of starch in the chloroplasts of isolated bundle sheath cells can be observed only in the presence of glucose-1-phosphate.  相似文献

Allium cepa L. leaves were subjected to enzymatic (pectolyase) and mechanical manipulation in order to ascertain the contribution made by various leaf tissues to the total sugar uptake by the leaf. In order to develop an understanding of the basic anatomy and ultrastructure of the Allium leaf and assess the integrity of the tissue before and after enzymatic and mechanical manipulation, a light- and transmission-electron-microscopy study was performed. One outcome of this study was the discovery that the chloroplasts of the bundle-sheath cells contain starch. The function of these inclusions in relation to carbohydrate pools and translocation is discussed. Kinetic curves for sucrose and fructose uptake by leaf discs derived from control and modified leaves are presented. In addition, kinetic curves for the tissues removed by the enzymatic treatment (inner parenchyma, bundle sheath and some vascular parenchyma) and the vascular bundles were also obtained. All tissues exhibited the same linear plus saturable profile as the dicotyledon, Beta vulgaris, with the exception of fructose uptake into the inner parenchyma and bundle-sheath cells; in this case the response was linear. The effect of anoxia on uptake of exogenous sucrose was also investigated. Anaerobiosis inhibited both the linear and saturable component of sucrose influx. Adenine-nucleotide levels were obtained using high-performance liquid chromatography for control (air) and anoxia-treated (N2) leaf discs. A general loss of adenine nucleotides was observed. The results presented indicate that all tissues of the leaf retrieve exogenous sugar such that the kinetic curves derived from leaf discs cannot represent phloem loading, per se.Abbreviations Mes 2-(N-morpholino)ethanesulfonic acid - E.C. energy charge  相似文献

The compound, palmate lamina of Lupinus palaestinus reorients photonastically, as well as phototropically in response to non-directional and directional light signals, respectively, by structural deformations of pulvini. When the excitation provided by directional light is maintained constant (fluence rate, angle of incidence and azimuth, with respect to the leaflet laminae), the entire lamina reorients towards it at a constant angular velocity over a considerable time interval and displacement. The laminar pulvinules are considerably longer than the subtending common petiolar pulvinus and therefore contribute most to laminar reorientation. The pulvinar region is characterized by transverse folds around its circumference, and longitudinal rib-like thickenings on the external walls of its epidermis that facilitate axial and transverse deformations. Specialized “joints”, at the distal and proximal ends of each pulvinule, contribute most to its flexing. Anthocyanin is notable by its absence. Specialized “motor” tissues surrounding the central vascular core participate in pulvinar deformation by undergoing directional and differential volume changes. The bundle sheath is characterized by numerous starch grains. The multi-layered cortical parenchyma exhibits an abundance of transversely oriented primary pit fields and associated plasmodesmata. When the leaflet lamina rotates around its midrib, the pulvinus twists along its axis, exhibiting epidermal and cortical deformation. The functional significance of these specializations is discussed.  相似文献

The aim of this work was to investigate the effects on carbohydrate metabolism of a reduction in the capacity to degrade leaf starch in Arabidopsis. The major roles of leaf starch are to provide carbon for sucrose synthesis, respiration and, in developing leaves, for biosynthesis and growth. Wild-type plants were compared with plants of a starch-excess mutant line (sex4) deficient in a chloroplastic isoform of endoamylase. This mutant has a reduced capacity for starch degradation, leading to an imbalance between starch synthesis and degradation and the gradual accretion of starch as the leaves age. During the night the conversion of starch into sucrose in the mutant is impaired; the leaves of the mutant contained less sucrose than those of the wild type and there was less movement of 14C-label from starch to sucrose in radio-labelling experiments. Furthermore, the rate of assimilate export to the roots during the night was reduced in the mutant compared with the wild type. During the day however, photosynthetic partitioning was altered in the mutant, with less photosynthate partitioned into starch and more into sugars. Although the sucrose content of the leaves of the mutant was similar to the wild type during the day, the rate of export of sucrose to the roots was increased more than two-fold. The changes in carbohydrate metabolism in the mutant leaves during the day compensate partly for its reduced capacity to synthesize sucrose from starch during the night.  相似文献

The maize (Zea mays L.) stem pulvinus is a disc of tissue located apical to each node that functions to return a tipped stem to a more upright position via increased cell elongation on its lower side. We investigated the possibility that reactive oxygen species (ROS) and hydrogen peroxide (H2O2), in particular, are involved in the gravitropic response of the pulvinus prior to initiation of the growth response by employing the cytochemical stain 3,3'-diaminobenzidine (DAB). DAB polymers were found in the bundle sheath cells of gravistimulated pulvini in association with amyloplasts after 1 min of gravistimulation, and the signal spread throughout the cytosol of these cells by 30 min. Furthermore, treatment of maize stem explants containing pulvini with 1 mm H2O2 on their upper sides caused reversal of bending polarity. Similar, though less dramatic, results were obtained via application of 1 mm ascorbic acid to the lower side of the explants. In addition, we determined that a maize cytoplasmic aconitase/iron regulatory protein 1 (IRP1) homolog is up-regulated in the pulvinus bundle sheath cells after gravistimulation using suppressive subtractive hybridization PCR (SSH PCR), real-time RT-PCR and in situ hybridization. Although we do not yet know the role of the IRP1 homolog in the pulvinus, the protein is known to be a redox sensor in other systems. Collectively, our results point to an increase in ROS quite early in the gravitropic signalling pathway and its possible role in determining the direction of bending of the pulvini. We speculate that an ROS burst may serve to link the physical phenomenon of amyloplast sedimentation to the changes in cellular biochemistry and gene expression that facilitate directional growth.  相似文献

Glycine (1–50 mM) increases the rate of the dark-induced (scotonastic) movements and decreases the amplitude and the rate of the light-induced (photonastic) movements of the secondary pulvini of Mimosa pudica leaves. The uptake of glycine is accompanied by a long-lasting dose-dependent increase in the alkalinity of the bathing medium of the excised pulvini. The data are in agreement with a H+-glycine co-transport mechanism within the pulvinar cells. Fusicoccin (50 M), known to promote H+–K+ exchange, antagonizes the effects of glycine on the movements and the alkalization of the bathing medium of the excised pulvini. The present results argue for the hypothesis that proton fluxes mediate the scotonastic and photonastic pulvinar movements.Abbreviations Gly glycine - FC fusicoccin - P1 primary pulvinus - P2 secondary pulvinus  相似文献

Pulvinus activity of Phaseolus species in response to environmental stimuli plays an essential role in heliotropic leaf movement. The aims of this study were to monitor the continuous daily pulvinus movement and pulvinus temperature, and to evaluate the effects of leaf movements, on a hot day, on instantaneous leaf water-use efficiency (WUEi), leaf gas exchange, and leaf temperature. Potted plants of Phaseolus vulgaris L. var. Provider were grown in Chicot sandy loam soil under well-watered conditions in a greenhouse. When the second trifoliate leaf was completely extended, one plant was selected to measure pulvinus movement using a beta-ray gauging (BRG) meter with a point source of thallium-204 (204Tl). Leaf gas exchange measurements took place on similar leaflets of three plants at an air temperature interval of 33–42°C by a steady-state LI-6200 photosynthesis system. A copper-constantan thermocouple was used to monitor pulvinus temperature. Pulvinus bending followed the daily diurnal rhythm. Significant correlations were found between the leaf-incident angle and the stomatal conductance (R 2 = 0.54; P < 0.01), and photosynthesis rate (R 2 = 0.84; P < 0.01). With a reduction in leaf-incidence angle and increase in air temperature, WUEi was reduced. During the measurements, leaf temperature remained below air temperature and was a significant function of air temperature (r = 0.92; P < 0.01). In conclusion, pulvinus bending followed both light intensity and air temperature and influenced leaf gas exchange.  相似文献

To evaluate daytime and nighttime carbon balance and assimilate export in soybean (Glycine max [L.] Merrill) leaves at different photon flux densities, rates of CO2 exchange, specific leaf weights, and concentrations of sucrose and starch were measured at intervals in leaves of pod-bearing `Amsoy 71' and `Wells II' plants grown in a controlled environment room. Assimilate export was estimated from CO2 exchange and change in specific leaf weight. Total diurnal assimilate export was similar for both cultivars. Large cultivar differences existed, however, in the partitioning of carbon into starch reserves and the relative amounts of assimilate exported during the day and the night. Total amounts of both daytime and nighttime export increased with increasing photon flux density, as did sucrose and starch concentrations, specific leaf weight, and rate of respiratory carbohydrate loss at night. Cultivar differences in nighttime rate of export were more closely related to the differences in amount of assimilate available at the end of the day than to differences in daytime rate of net CO2 assimilation. Daytime rates of export, however, were closely related to daytime rates of net CO2 assimilation within each cultivar. The total amount of starch depleted during the 10-hour night increased as starch concentration at the beginning of the night increased.  相似文献
