A field assessment of 26 accessions of Vicia narbonensis and three of V. johannis confirmed previous laboratory studies demonstrating higher levels of resistance to Aphis fabae in these two wild species compared to the closely related crop, Vicia faba. Accessions of V. johannis were significantly more resistant than most accessions of V. narbonensis for all resistance indices measured except survival of aphid nymphs. Plant growth stage significantly affected levels of resistance in both Vicia species, resistance being moderate at pre-bud stage, decreasing on flowering and rising again at pod fill and onset of leaf senescence. Significant intraspecific variability in aphid resistance was found only within the 26 accessions of V. narbonensis, var. serratifolia being more resistant than var. narbonensis. Possible resistance factors and the agronomic potential of these two wild relatives of Faba bean are considered.  相似文献

Investigations were made on the effect of species of fungi isolated from therhizosphere of broad bean (Vicia faba Linn.) variety Giza I, and cotton (Gossypium barbadense Linn.) variety Giza 47, on plant growth. Broad bean rhizosphere fungi differently affected plant growth.Fusarium moniliforme, Penicillium martensii, Rhizopus stolonifer, andCladosporium sphaerospermum stimulated both root and shoot growth.Aspergillus niger andAlternaria tenuis have an inhibitory effect on plant growth. On the other hand, the rhizosphere fungi of cotton namely,Penicillium cyclopium, Aspergillus terreus, Cephalosporium sp.,Aspergillus niger, Cladosporium sphaerospermum, andFusarium oxysporum stimulated plant growth.  相似文献

Minicircular DNAs found in mitochondria of 6-d-old etiolated seedlings of Vicia faba L. were also present in mitochondria isolated from cell suspension cultures and from green leaves of this plant. These results support the suggestion that the plasmid-like molecules found in mitochondria of V. faba are an essential component of the mitochondrial genome. The minicircular DNAs were, apparently, peculiar for the species V. faba since they were found in all three cultivars of this species which were studied. The distribution pattern of plasmid-like DNAs in Vicia villosa L. was completely different and mitochondria from Medicago sativa L. also contained specific minicircular DNAs. Thus, minicircular DNAs are typical for the mitochondrial genomes of several legumes and different plant species have their specific mitochondrial plasmid-like DNAs  相似文献

The effects of salt stress on nitrogenase (N2-ase) activity, growth and nitrogen content ofVicia faba (L.),Medicago sativa (L.) Merrill,Glycine max andVigna sinensis (L.) were investigated. Four levels of salinity were applied and salt treatments were imposed on inoculated and N-fertilized plants.M. sativa tolerated mild levels of salinity but higher salt concentrations depressed N2-ase activity of this species. The other three legumes were considerably affected by salt treatments, and N2-ase activity was significantly reduced by salinity. Vicia faba, carrying elongate nodules, could restore a partial N2-ase activity upon recovery from salt stress whereasG. max andV. sinesis, both with spherical nodules, could not regain significant activity when salinity was removed. Salt stress retarded growth of both inoculated and N-fertilized plants. The nitrogen content of both treatments was also affected by salinity and the effect was more severe for inoculated than N-fertilized plants.  相似文献

The dominating rhizosphere fungi of broad bean (Vicia faba Linn.) variety “Giza 1”, and cotton (Gossypium barbadense Linn.) variety “Giza 47”, were grown in liquid medium. After 10 days, filtrates were obtained and sterilized by filtration through sintered-glass filter. Plants were grown in sterile sand which was supplemented with nutrient solution. Every plant was irrigated with fungal filtrate, unconsumed medium, and water. The filtrates of the rhizosphere fungi of both broad bean and cotton stimulated plant growth.  相似文献

The speciesVicia bithynica andVicia narbonensis, from the same subgeneric section ofVicia faba, show variations in nuclear DNA content Nuclear DNAs, extracted from root tips of the twoVicia species, were characterized by thermal denaturation, analytical ultracentrifugation and reassociation kinetics. The thermal denaturations of DNA, the number of DNA components reassociating with second order kinetics, the proportion of repeated DNA sequences, the frequency of the repeated DNA classes are reported and compared to previous data onVicia faba DNA. Feulgen absorptions at different thresholds of optical density+ of interphase nuclei in cytological preparations of the root meristems ofV. bithynica andV. narbonensis are determined and compared withV. faba analogous determinations. The results, confirming that plant genome is highly flexible, are discussed in relation to other data on the interspecific variations of the nuclear DNA content.  相似文献

The localization was studied of α- and β-galactosidases in frozen sections of Ca-formol fixed root tips using simultaneous azocoupling reaction. In all species studied (Allium cepa,Cucurbita maxima, Lupinus albus, Pisum sativum, Vicia faba, Zea mays) positive results were obtained, the localization being ubiquitous (according to localization typology given here). InVicia faba andZea mays the isoenzymes of α- and β-galactosidases were revealed by means of acrylamide gel electrophoresis, using authors’ modification of Reisfeld method, in whole root tips, particular growth zones and separately in cortex and central cylinder. No differences were observed comparing stele and cortex. Whereas characteristic isoenzyme patterns were found in individual growth zones in maize, no differences appeared in broad bean. A comparison was made of thein situ localization and of the isoenzyme patterns of α- and β-galactosidases with α- and β-glucosidases. In the case of galactosidases, positive results appear with both α- and β-galactoside. The rising of pH to neutrality leads to considerable decrease in the activity of both galactosidases.  相似文献

The culture filtrates of the rhizosphere fungi of broad bean (Vicia faba L.) and cotton (Gossypium barbadense L.) were analysed for the presence of plant growth substances of auxin and gibberellins nature. Bioassay test and chromatographic analysis indicated that these fungi, each synthesized different auxins in their culture medium. These auxins were indole compounds. Similarly the rhizosphere fungi produced in their culture medium some gibberellins and gibberellin-like substances.  相似文献

For the separation of α- and β-glucosidase and α- and β-galactosidase isoenzymes fromZea mays L. andVicia fabaL. root tips the system of disc electrophoresis in polyacrylamide gel developed for basic protein separation proved most suitable. The detection was carried out by a simultaneous azocoupling reaction. In maize α-glucosidase was not detected, β-glucosidase gave 3, α-galactosidase 4, and β-galactosidase 3 zones. In broad bean a- and β-glucosidases were absent, α-galactosidase gave 2 and β-galactosidase 3 zones, α- and β-galactosidase activity zones correspond principially to each other in their position. In maize one zone gives a positive reaction for both β-glucosidase and α- and β-galactosidaso.  相似文献

Strains of fungi isolated from pulse crops: pea (Pisum sativum) and faba beans (Vicia faba) plants with symptoms of Ascochyta blight, footrot and stems lesions have been examined under laboratory conditions for their ability to produce ascochitine and metabolites toxic toArtemia salina. BothAscochyta pisi Lib and Ascochyta fabae LK Jones isolates formed ascochitine in yields of 20–480mg/kg. The highest yield of ascochitine was produced on rice and the lowest on maize grain. Ascochyta pinodes andPhoma medicaginis varpinodella (LK Jones) Boerema (formerlyAscochyta pinodella LK Jones) did not produce ascochitine. Crystalline ascochitine was found to be of moderate toxicity toArtemia salina larvae (LC50 = 85μg/cm3BSM*). Extracts ofPhoma medicaginis var,pinodella cultures were found to be highly toxic toArtemia salina.  相似文献

Electrophoretic analysis of five enzyme systems, LAP, PGI, SKDH, SOD and 6-PGDH, among 102Vicia accessions representingV. bithynica and seven species of theV. narbonensis complex, namelyV. eristalioides, V. kalakhensis, V. johannis, V. galilaea, V. serratifolia, V. narbonensis andV. hyaeniscyamus, has been performed. The recorded variation was tentatively assigned to 41 allelic genes at eight loci; intraspecific variation was observed in all species except forV. eristalioides. The results obtained were compared with the corresponding data reported earlier forV. faba. Hierarchical grouping of the investigated taxa, includingV. faba, was based onNei's genetic identities calculated from the allozyme frequency data.Vicia faba andV. bithynica were shown to be most distantly related to one another and to the remaining species investigated.Vicia serratifolia appeared to be a peripheral member of theV. narbonensis complex. The results are discussed with reference to genetic diversity and taxonomic relationships of the species under study.  相似文献

The insecticide pyrethroid II, representing synthetic pyrethroids of the second generation, was not found to be mutagenic inSalmonella typhimurium strains TA1535, TA100, TA1538, TA98 and TA97. High concentrations of the tested insecticide inhibited the germination and length of roots of germinating seeds, the height of plants cultivatedin vitro, and slightly increased the frequency of aberrant anaphases and telophase in root-tips ofVicia faba andHordeum vulgare.  相似文献

The studies on the occurrence ofFusarium andAlternaria in faba bean seed have been conducted for three years. The fungal flora in seed lots from healthy plant and from plant inoculated withAscochyta fabae were determined. In addition the occurrence ofFusarium andAlternaria in three parts of seed: testa, cotyledones, and embryo axis were investigated. It was found thatFusarium andAlternaria were present in faba bean seed, but the frequency of occurrence varied depending upon cultivar and experimental year.  相似文献

Abstract

Faba bean (Vicia faba L.) is a valuable food and feed crop worldwide, but drought often limits its production, and its genome is large and poorly mapped. No information is available on the effects of genomic regions and genes on drought adaptation characters such as stomatal characteristics in this species, but the synteny between the sequenced model legume, Medicago truncatula, and faba bean can be used to identify candidate genes. A mapping population of 211 F5 recombinant inbred lines (Mélodie/2 × ILB 938/2) were phenotyped to identify quantitative trait loci (QTL) affecting stomatal morphology and function, along with seed weight, under well-watered conditions in a climate-controlled glasshouse in 2013 and 2014. Canopy temperature (CT) was evaluated in 2013 under water-deficit (CTd). In total, 188 polymorphic single nucleotide polymorphisms (SNPs), developed from M. truncatula genome data, were assigned to nine linkage groups that covered ~928 cM of the faba bean genome with an average inter-marker distance of 5.8 cM. 15 putative QTLs were detected, of which eight (affecting stomatal density, length and conductance and CT) co-located on chromosome II, in the vicinity of a possible candidate gene—a receptor-like protein kinase found in the syntenic interval of M. truncatula chromosome IV. A ribose-phosphate pyrophosphokinase from M. truncatula chromosome V, postulated as a possible candidate gene for the QTL for CTd, was found some distance away in the same chromosome. These results demonstrate that genomic information from M. truncatula can successfully be translated to the faba bean genome.  相似文献

The two final products of radiolytic degradation of cytosinei.e. parabanic and oxaluric acids were investigated as regards their cytogenetic effects. Broad bean (Vicia faba L.) root meristem was used as experimental material. While the oxaluric acid was not able to induce chromosomal aberrations to a greater extent, the parabanic acid acts as a clastogenic compound. When applied on a resting broad bean root meristem at concentration from 10-3 to 10-2 M it induces chromosome and chromatid aberrations, especially breakages. Their frequency reaches 9–12% at the highest tested concentration. The same concentrations of the parabanic acid increases mitotic index of the first cell generation of primary root.  相似文献

Pseudothecia of Didymella fabae, the teleomorph of Ascochyta fabae, were first observed on faba bean (Vicia faba) debris in Spain during autumn 1995. Most pseudothecia were mature by December–February. The ascospores gave rise to typical cultures of A. fabae, and conidia from these cultures caused ascochyta blight symptoms on inoculated faba bean plants. Placing straw‐bearing pseudothecia over the plants to allow ascospore discharge also resulted in typical ascochyta blight symptoms. Pseudothecia maturity and discharge of ascospores from the infested faba bean straw overlapped with the vegetative stage of the faba bean crop, which occurs in southern Spain during winter as the crop is sown in autumn and harvested in spring. These observations indicate that ascospores may serve as primary inoculum for the disease.  相似文献

Treatment of two-week-oldBrassica campestris andTrigonella foenum-graecum plants with morphactin andVicia faba, Antirrhinum orontium, andPapaver somniferum with Niagara, induced marked variations in the orientation and ontogeny of stomata and the epiderma cells. Morphactin—chlorflurenol at 12.5, 62.5, 125, 250, and 500 ppm, caused marked damage of the shoot apices and changes in the epidermal tissue, such as divisions of the guard cells, reduction in the size of the stomata, and epidermal cells. Niagara—ethyl-hydrogen-1-propylphosphonate at 100, 500, 1 000 5 000, and 10 000 ppm caused thickenings of the epidermal cell walls and differentiation of new meristemoids from the epidermal cells, contiguous stomata, and incomplete development of the guard cells.  相似文献

A procedure was developed for satisfactory freeze-sectioning of root tips. The use of Ca formol-fixed material kept and frozen in Holt's syrup is recommended. The existence and different localization of 2 fractions of non—specific esterase was verified in root tips ofVicia faba. The same results were revealed in fixed and unfixed material. The dynamics ofin situ reaction was followed with respect to optimal incubation time. The results with substrates of different chain length support the existence of 2 fraction of the studied enzyme, none of which, concerning substrate specificity, is a lipase. It follows from the present studies inVicia faba and other species (Cucurbita pepo, Lupinus albus, Pisum sativum Zea mays), that non-specific esterase localization is not directly given by histogenesis.  相似文献
