Isolated embryos are more suitable for in vivo study of protein synthesis than non-isolated embryos because, after isolation, the uptake of labeled amino acids is about 1000 times higher. However, isolation also stimulates protein synthesis: Up to 4 h after isolation, the capacity to incorporate labeled amino acids increased 7 times. Therefore, data on incorporation obtained with isolated embryos cannot be extended to non-isolated embryos. The increase of protein synthesis was not due to synthesis of specific proteins, but was a general increase. Furthermore, ripening, dormant, and afterripened embryos showed the same degree of increase. Isolation therefore stimulates protein synthesis nonspecifically. When embryos were kept under anaerobic conditions after isolation, protein synthesis did not increase. Therefore, higher oxygen consumption after removal of the seedcoat is probably the cause of the higher incorporation capacity. Furthermore, the activation of protein synthesis lagged several hours behind the increase of oxygen consumption.Abbreviations A

The transition from the growth to the maturation phase in developing seeds of Agrostemma githago L. was found to coincide with major changes in the rate of protein synthesis, the kinds of proteins synthesized, and the composition of the non-proteinbound amino acid pool. Coincident changes were observed in viability and the ability to withstand desiccation. Desiccated mature Agrostemma seeds are dormant and need at least three months of after-ripening. In imbibed dormant and after-ripened seeds no synthesis of storage proteins was observed with the exception of one particular set of storage proteins. Dormant and after-ripened seeds synthesized the same kinds of proteins during early imbibition, indicating an almost identical metabolic state which differs considerably from that of developing seeds. 相似文献

In dark-grown, isolated embryos of Agrostemma githago, a transient period of nitrate-reductase (NR) (NADH: nitrate oxidoreductase, EC 1.6.6.1) activity occurred from 6 to 36 h after the start of imbibition. During this period, NR activity was enhanced by nitrate, 6-benzylamino-purine and ethylene. Ethylene and 6-benzylamino-purine acted synergistically, whereas ethylene had no effect on nitrate induction. Aminoethoxyvinyl-glycine, an inhibitor of ethylene biosynthesis, inhibited the cytokinin-induced increase of NR activity, but had no effect on the nitrate-induced increase. The inhibition by aminoethoxyvinylglycine was overcome completely by ethylene. The ethylene precursor 1-aminocyclopropane-1-carboxylic acid had the same effect on NR activity as ethylene. Our data indicate that NR induction by cytokinins only occurs in the presence of ethylene, and that nitrate enhances NR activity through a mechanism which is distinct from the induction by hormones.Abbreviations ACC

The injection of adrenocorticotropic hormone (ACTH) of prolonged effect at the doses of 4 and 10 M. U. to the intact rats from the 11th till the 15th day of pregnancy resulted in the twofold increase of protein content in the brain and its decrease in the liver of 15 days old embryos, as compared with the control ones. The content of DNA, RNA and proteins in the placenta of experimental animals increased as well. The rate of incorporation of 3H-thymidine in the liver DNA and 14C-leucine in the liver and brain acid-soluble protein decreased within small intervals of time following the treatment. The total radioactivity of proteins in the liver, brain and placenta calculated per DNA unit was similar to the control one whereas the specific radioactivity of total protein in the liver of experimental embryos was higher than in the control. 相似文献

Quiescent maize embryos were found to contain significant amounts of poly-A-rich pre-formed RNA. 14C-amino acid incorporation into trichloroacetic acid precipitable material was detected at slow rate at the begining of imbibition and fastly increased near 18 to 24 h. Polysomal formation was measured during this period. Addition of - amanitin to the incubation system at two 6h-pulse periods showed significant inhibition of the 14C-amino acid incorporation for the 18–24 h-period, but not for the 0–6 h-period. 相似文献

In an inbred population, selection may reduce the frequency of deleterious recessive alleles through a process known as purging. Empirical studies suggest, however, that the efficacy of purging in natural populations is highly variable. This variation may be due, in part, to variation in the expression of inbreeding depression available for selection to act on. This experiment investigates the roles of life stage and early‐life environment in determining the expression of inbreeding depression in Agrostemma githago. Four population‐level crosses (‘self’, ‘within’, ‘near’ and ‘far’) were conducted on 20 maternal plants from a focal population. Siblings were planted into one of three early environmental treatments with varying stress levels. Within the focal population, evidence for purging of deleterious recessive alleles, as well as for variation in the expression of inbreeding depression across the life cycle was examined. In addition, the effect of early environment on the expression of inbreeding depression and the interaction with cross‐type was measured. We find that deleterious recessive alleles have not been effectively purged from our focal population, the expression of inbreeding depression decreases over the course of the life cycle, and a stressful early environment reduces the variance in inbreeding depression expressed later in life, but does not consistently influence the relative fitness of inbred versus outcrossed individuals. 相似文献

Primary leaves of intact bean plants ( Phaseolus vulgaris L.) were treated with benzyladenine (BA) at different stages of ageing, BA promoted the synthesis of RNA, and soluble and insoluble proteins. The effects of BA stimulation differed depending on the age at which the leaf received the hormone treatment. In leaves attached to the plant, BA appeared to stimulate the rate of synthesis more than the rate of decomposition of RNA and protein, resulting in a net increase in RNA and protein. Both chloroplast and cytoplasmic ribosomes were still observed in intact yellowish green leaves. Polysomes in the cytoplasm increased remarkably when BA treatment was begun at late stages. 相似文献

A phenol-sodium dodecyl sulfate procedure has been used to extract DNA, RNA, and protein from 3-, 4.5-, and 6.5-day-old preimplantation rabbit embryos that were incubated for different lengths of time in the presence of labeled precursors. The results suggest that the newly-synthesized RNA and protein are more stable in the growing blastocyst than in the trophoblast at the time of implantation.This conclusion was derived from the results of embryo incorporation kinetics. The results also suggest that the increase in protein and RNA content in preimplantation rabbit blastocysts was preceded by an increase in protein synthesis accompanied by low catabolism. 相似文献

Effects of Cultivation on the Germination Character of the Corn Cockle (Agrostemma githago L.) 总被引:1,自引:0,他引:1
Germination tests were done on 19 populations of Agrostemmagithago to examine their responses in relation to the culturalhistory of the plant and to try to determine what changes mayhave accompanied cultivation and the part these may have playedin the establishment and subsequent decline of the species asa weed. Differences found from one population to another were foundto be relatively minor, and the responses of all the collectionsresembled those of other species of Silenoideae distributednaturally around the Mediterranean basin. Differences were establishedbetween adventive populations of A. githago and species naturallydistributed in deciduous woodland and steppeland zones of Europe,even when growth patterns and habitat preferences were relativelysimilar. It was concluded that A. githago had originated in an area witha Mediterranean climate and that there had been few changesin the germination responses of the species during its periodof association with man as a weed of cultivation. The significanceof this result is discussed in relation to the decline of thespecies in Europe during the last hundred years, and with respectto the effects of cultivation on the germination of species,including crop plants, of Mediterranean origin which possessgermination responses similar to those of Agrostemma. 相似文献

In vitro RNA synthesis by tobacco mosaic virus and cowpea chlorotic mottle virus replicase were inhibited by cordycepin triphosphate. Inhibition could be overcome with higher concentrations of ATP in assay mixtures but not with UTP. Products synthesized in vitro by tobacco mosaic virus RNA replicase in the presence of inhibitor revealed replicative form but not replicative intermediate RNAs. These results suggest that cordycepin triphosphate competes specifically with ATP and results in premature termination of viral RNA synthesis in vitro. 相似文献

Dormant and after-ripened seeds of Agrostemma githago (corn-cockle)were pretreated in polyethylene glycol 400 (PEG) solutions attemperatures which would have allowed germination if the seedshad been imbibed in water, viz. 4?C or 20?C for after-ripenedseeds, and 4?C for dormant seeds. Pretreated seeds germinatedfaster than untreated seeds. The maximum decrease of the T50(time to 50% germination) was 66%. Furthermore, pretreated seedswere capable of germination at supra-optimal temperatures whichotherwise had inhibited germination completely (20?C for dormantseeds and 30?C for after-ripened seeds). The percentage germinationat a supra-optimal temperature was considerably higher whenthe seeds had been primed at a temperature at which they developedmore extension power. The advantageous effects of the osmotic pretreatment were lessthan might be expected when the osmoticum had inhibited onlycell elongation. This was largely, if not fully, due to a generaldetrimental effect of osmotic stress and not to a selectiveinhibition of the processes which occur during the pregerminativephase in preparation for growth. Thus, during priming seedscomplete all or almost all processes which occur in water-imbibedseeds prior to radicle emergence. Gibberellins and the photoperiodic control of stem elongation in the long-day plant Agrostemma githago L. 总被引:1,自引:0,他引:1
Agrostemma githago is a long-day rosette plant in which transfer from short days (SD) to long days (LD) results in rapid stem elongation, following a lag phase of 7–8 d. Application of gibberellin A20 (GA20) stimulated stem elongation in plants under SD, while 2-isopropyl-4-dimethylamino-5-methylphenyl-1-piperidine-carboxylate methyl chloride (AMO-1618, an inhibitor of GA biosynthesis) inhibited stem elongation in plants exposed to LD. This inhibition of stem elongation by AMO-1618 was overcome by simultaneous application of GA20, indicating that GAs play a role in the photoperiodic control of stem elongation in this species. Endogenous GA-like substances were analyzed using reverse-phase high-performance liquid chromatography and the d-5 corn (Zea mays L.) assay. Three zones with GA-like activity were detected and designated, in order of decreasing polarity, as A, B, and C. A transient, 10-fold increase in the activity of zone B occurred after 8–10 LD, coincident with the transition from lag phase to the phase of rapid stem elongation. After 16 LD the activity in this zone had returned to a level similar to that under SD, even though the plants were elongating rapidly by this time. However, when AMO-1618 was applied to plants after 11 LD, there was a rapid reduction in the rate of stem elongation, indicating that continued GA biosynthesis was necessary following the transient increase in activity of zone B, if stem elongation was to continue under LD. It was concluded that control of stem elongation in A. githago involves more than a simple qualitative or quantitative change in the levels of endogenous GAs, and that photoperiodic induction alters both the sensitivity to GAs and the rate of turnover of endogenous GAs.Abbreviations AMO-1618
