Fusarium populations were investigated in maize grains and their husks about six weeks before harvest in three maize fields in the Manawatu region of New Zealand. The role of litter and soil as reservoirs for these fungi was also examined. Two techniques were used to examine populations, dilution plating and direct plating. Using the dilution plating technique the highest overall populations were found in husks (mean 2.2 x 10(5)/g) and litter (mean 1.4 x 10(5)/g), while similar lower numbers of viable propagules were obtained from grain (mean 2.1 x 10(3)/g) and soil (2.8 x 10(3)/g). With this technique five Fusarium spp. were commonly isolated; F. graminearum (Gibberella zeae), F. culmorum, F. subglutinans, F. oxysporum and F. acuminatum, of which F. graminearum was the most abundant. With the direct plating technique 87% of grains were infected with Fusarium spp., with some grains being infected with more than one species. Segments from husks and litter, 70% and 43% respectively, were colonised by Fusariumr spp. F. graminearum was the most frequent species isolated from maize grain and husk segments (48.3 and 37.7% colonisation respectively). Other species, particularly F. culmorum and F. acuminatum, were also found to be common contaminants. A total of 15 Fusarium spp. was recovered from all material examined by both techniques. Cultures with characteristics resembling those of F. moniliforme were rarely observed.  相似文献

BACKGROUND: The gorgeous frescoes organized by the master Renaissance painter Raphael Sanzio (1483-1520) and illustrating the heavenly adventures of Cupid and Psyche were painted between 1515 and 1518 to decorate the Roman villa (now known as the Villa Farnesina) of the wealthy Sienese banker Agostino Chigi (1466-1520). Surrounding these paintings are festoons of fruits, vegetables and flowers painted by Giovanni Martini da Udine (1487-1564), which include over 170 species of plants. A deconstruction and collation of the cucurbit images in the festoons makes it possible to evaluate the genetic diversity of cucurbits in Renaissance Italy 500 years ago. FINDINGS: The festoons contain six species of Old World cucurbits, Citrullus lanatus (watermelon), Cucumis melo (melon), Cucumis sativus (cucumber), Ecballium elaterium (squirting cucumber), Lagenaria siceraria (bottle gourd) and Momordica balsamina (balsam apple), and two or three species of New World cucurbits, Cucurbita maxima, C. pepo and, perhaps, C. moschata (pumpkin, squash, gourd). The images of C. maxima are the first illustrations of this species in Europe.  相似文献

The complexity of cyclic GMP-binding activity in the 48,000 X g supernatant of three species of the cellular slime molds (Dictyostelium discoideum, Dictyostelium rosarium, and Polysphondylium violaceum) was studied by gel filtration chromatography on AcA 34 Ultrogel. All these species have in common a cyclic GMP-binding protein of molecular weight of about 2.5 X 10(5) which specifically binds this nucleotide. In addition, Scatchard plots of assays carried out with the 48,000 X g supernatant of these species exhibit cyclic GMP-binding activity with an apparent dissociation constant of about 1 nM. None of the cyclic GMP-binding proteins separated by chromatography on AcA 34 Ultrogel was associated to protein kinase activity stimulation. In view of the cyclic GMP function during chemotactic transduction in the cellular slime molds, the possible molecular function for this 2.5 X 10(5)-dalton cyclic GMP-binding protein is discussed.  相似文献

Fusarium head blight is an important disease of cereal crops caused by Fusarium species. It causes not only a reduction in yield, but most Fusarium species (F. graminearum. F. culmorum, F. avenaceum. F. poae) produce also a range of toxic metabolites such as deoxynivalenol (DON) and zearalenone (ZEA). The evaluation of Fusarium species was followed up under natural infection conditions during the growing seasons 2001--2002 and 2002--2003 in two varietal winter wheat experiments on the experimental farm of the Hogeschool Gent at Bottelare. Disease pressure, DON and ZEA content, different Fusarium species as well as growth and yield parameters were determined. In both years there were significant differences between the varieties concerning the susceptibility to Fusarium and the DON content. ZEA was not found in the kernels. The mean deoxynivalenol (DON) content was in 2002 (1,126 mg/kg) higher than in 2003 (0.879 mg/kg) although the mean disease severity was bigger in 2003 than in 2002 what means that the DON content was not always correlated with the disease severity. The Fusarium species most frequently identified in our two field trials (Bottelare) were F. graminearum and F. culmorum Varietal differences in susceptibility to Fusarium species and DON contamination could be detected.  相似文献

This is the first report of the presence of 15-acetyl-deoxynivalenol on corn ears in the field prior to harvest and in combination with deoxynivalenol and zearalenone. This study was designed to determine testes masses, total number of spermatozoa ejaculated per copulatory episode, and the pattern of sperm numbers in successive ejaculates in prairie voles (Microtus ochrogaster), montane voles (M. montanus), pine voles (M. pinetorum), and meadow voles (M. pennsylvanicus). Prairie voles displayed mean totals of 2.7 ejaculations and 30.5 X 10(6) spermatozoa before reaching a satiety criterion; montane voles 3.4 ejaculations and 19.0 X 10(6) spermatozoa, pine voles 2.4 ejaculations and 3.3 X 10(6) spermatozoa, and meadow voles 2.5 ejaculations and 25.5 X 10(6) spermatozoa. In all species the number of spermatozoa decreased in successive ejaculates. Significant species differences were noted for the total number of spermatozoa ejaculated and number of spermatozoa ejaculated in each of the first 3 ejaculates. Species differences also were noted for testes mass, with meadow voles having the largest testes and pine voles having the smallest. These data can be compared to similar data on laboratory rats and deer mice and related to recent theory regarding sperm numbers, testes sizes, and mating systems. In general, the species with large testes appear to ejaculate more spermatozoa. The significance of species differences in testes mass and total sperm numbers remains unclear, but may relate to the occurrence of multiple mating by females during a single receptive period.  相似文献

In 1998–2000 a monitoring of the spectrum of Fusarium species on winter wheat was carried out in the Rhineland. The epidemic spread ofFusarium spp. on wheat plants during growing season was investigated as well as the grain contamination after harvest.F avenaceum was the Fusarium species isolated most frequently followed byF culmorum, F poae andF graminearum. Microdochium nivale occurred considerably only in 1998. Both, susceptibility and plant height of the cultivars were correlated to the incidence of Fusarium species /M nivale on harvested kernels; interactions with cropping intensities were detected. The incidence ofF poae seemed to be independent of the cultivar-specific Fusarium susceptibility. Despite the lack of disease symptoms, between growth stages 45–85 mycelium ofFusarium spp. was detectable in the leaves as well as conidia on the leaf surfaces.  相似文献

A mycological survey was carried out for the first time, on red flint corn samples from the northwestern Andinian region of Argentina in the 1999 and 2000 harvest seasons. Species of the genus Fusarium were the most prevalent component of the flint corn mycoflora present in all provinces. F. verticillioides was the predominant Fusarium isolated in the 1999 harvest season in the the region, and was found at higher incidence level than those observed on commercial semident corn hybrids harvested in the main corn production area in Argentina (Pampean region). During the 2000 harvest season, Fusarium graminearum was most commonly isolated species in Salta province.  相似文献

Fusarium is one of the important phytopathogenic genera of microfungi causing serious losses on cucurbit plants in Kermanshah province, the largest area of cucurbits plantation in Iran. Therefore, the objectives in this study were to isolate and identify disease-causing Fusarium spp. from infected cucurbit plants, to ascertain their pathogenicity, and to determine their phylogenetic relationships. A total of 100 Fusarium isolates were obtained from diseased cucurbit plants collected from fields in different geographic regions in Kermanshah province, Iran. According to morphological characters, all isolates were identified as Fusarium oxysporum, Fusarium proliferatum, Fusarium equiseti, Fusarium semitectum and Fusarium solani. All isolates of the five Fusarium spp. were evaluated for their pathogenicity on healthy cucumber (Cucumis sativus) and honeydew melon (Cucumis melo) seedlings in the glasshouse. F. oxysporum caused damping-off in 20–35 days on both cucurbit seedlings tested. Typical stem rot symptoms were observed within 15 days after inoculation with F. solani on both seedlings. Based on the internal transcribed spacer (ITS) regions of ribosomal DNA (rDNA) restriction fragment length polymorphism (RFLP) analysis, the five Fusarium species were divided into two major groups. In particular, isolates belonging to the F. solani species complex (FSSC) were separated into two RFLP types. Grouping among Fusarium strains derived from restriction analysis was in agreement with criteria used in morphological classification. Therefore, the PCR-ITS-RFLP method provides a simple and rapid procedure for the differentiation of Fusarium strains at species level. This is the first report on identification and pathogenicity of major plant pathogenic Fusarium spp. causing root and stem rot on cucurbits in Iran.  相似文献

Klebsiella was found to multiply and colonize growing radish root bulb surfaces following the inoculation of seeds with 101–104 cells. All 29 cultures ofKlebsiella originally isolated from 5 different sources were capable of growth to 106–107 colony-forming units/g of root bulb within 1 week after seed germination. Linear regression analysis illustrated differences inKlebsiella survival rates over 4 weeks of radish plant development. Analysis of covariance showed the survival ability wasKlebsiella from vegetables>mastitis>human, water, and pulp mill isolates. It was also shown thatKlebsiella species 2 had a significantly higher survival rate than the otherKlebsiella species. This finding correlates well with the observation thatKlebsiella species 2 is the most commonKlebsiella species isolated from vegetables. Average densities for allKlebsiella groups at plant harvest (5 weeks) ranged from 103–105 colony-forming units/g of radish plant. The possible health significance of these densities ofKlebsiella on vegetables consumed raw by humans is discussed.  相似文献

Blood-derived hemopoietic stem cells were collected using a continuous (10 patients) or a semi-continuous flow separator (2 patients) in some patients with acute non-lymphocytic leukemia. For ten out of those patients, five to seven leukaphereses were performed during a short period (median = 11 days) of marrow recovery following severe aplasia induced by an intensive chemotherapy. The mean number of CFU-GM cells collected per leukapheresis and per patient respectively was 6.7 X 10(4)/kg and 38.5 X 10(4)/kg. This latter number was similar to that obtained during a marrow harvest performed for a bone marrow transplantation, suggesting that high numbers of hemopoietic stem cells can be collected from the peripheral blood in leukemic patients and used for autologous transplantation.  相似文献

Ten wet grassland species were fumigated with four concentrations of ozone (charcoal-filtered air, non-filtered air and non-filtered air plus 25 or 50 nl l(-1) ozone) in open-top chambers during one growing season to investigate the long-term effect of this air pollutant on various growth variables. Only Eupatorium cannabinum showed ozone-related foliar injury, while five species reacted with significantly ozone-enhanced senescence. Premature senescence was paralleled by a significant ozone-induced reduction of green leaf area in Achillea ptarmica, E. cannabinum and Plantago lanceolata. At the intermediate harvest performed after 28 days shoot weights were significantly decreased by ozone in A. ptarmica and increased in Molinia caerulea. At the final harvest performed at the end of the growing season two other species, Cirsium dissectum and E. cannabinum had a significantly reduced shoot weight due to ozone. Root biomass was determined only at the intermediate harvest. The root:shoot ratio (RSR) was significantly reduced in C. dissectum, while it increased in M. caerulea. Seven of the species developed flowers during the experiment. While no significant ozone effects on flowering date and flower numbers were detected, flower weights were significantly reduced in E. cannabinum and P. lanceolata.  相似文献

Fusarium oxysporum f.sp. melonis and root-knot nematode (Meloidogyne javanica) are destructive pathogens on cucurbits in Varamin area of Iran. The interaction between two pathogens was studied on local melon cultivars, Garmsar and Sooski. Inoculum of Meloidogyne javanica was prepared on susceptible cultivar, Rutgers using single egg mass method in greenhouse. Inoculum of Fusarium oxysporum f.sp. melonis (race 1) was prepared using Richard solution. A concentration of 2 x 10(5) micro conidia of fungus and 2000, 3000, 4000, 5000 eggs of nematode was used in 1 kg of autoclaved soil. Plants were inoculated with nematode at 2-3 leave stage then with fungus 2 weeks after nematode inoculation. The experiment was conducted in factoriel design based on CRD with 20 treatments, including varieties in 2 levels (Garmsar and Sooski), nematode in 5 levels (0, 2000, 3000, 4000, 5000 eggs) and fungus in 2 levels (presence and absence) and 3 replicates. The index that evaluated were growth index including fresh and dry weight of shoot and root, height, Fusarium wilt index and root gall index. Results of this experiment showed that all of treatments comparison to control were significantly different (p = 0.05) in growth index. Combination of fungus and nematode (5000 eggs) caused the most decrease in growth index on Garmsar and Sooski.  相似文献

Trichothecenes are a large family of sesquiterpenoid secondary metabolites of Fusarium species (e.g., F. graminearum) and other molds. They are major mycotoxins that can cause serious problems when consumed via contaminated cereal grains. In the past 20 years, an outline of the trichothecene biosynthetic pathway has been established based on the results of precursor feeding experiments and blocked mutant analyses. Following the isolation of the pathway gene Tri5 encoding the first committed enzyme trichodiene synthase, 10 biosynthesis genes (Tri genes; two regulatory genes, seven pathway genes, and one transporter gene) were functionally identified in the Tri5 gene cluster. At least three pathway genes, Tri101 (separated alone), and Tri1 and Tri16 (located in the Tri1-Tri16 two-gene cluster), were found outside of the Tri5 gene cluster. In this review, we summarize the current understanding of the pathways of biosynthesis, the functions of cloned Tri genes, and the evolution of Tri genes, focusing on Fusarium species.  相似文献

Microbiological challenge study was carried out to verify the microbial stability of bottled waters against four different mold species isolated from bottled water (Fusarium sp.; Cladosporium sp.; Penicillium chrysogenum and Aspergillus fumigatus) and to follow the growth of the molds in bottled water. Twelve types of bottled water with different mineralization and CO2 level in PET and glass packages were collected from 4 European countries. Three different inoculation levels of spore suspensions were used to contaminate bottled water samples. The surviving colony forming unit (CFU) numbers and visual growth were monitored during the investigation period (26 weeks). The results of surviving CFU showed that the fungal growth is mostly determined by the carbonation level and the type of the mold strain. Neither the inoculation level nor the mineral content had any significant effect on the survival of the different mold strains. Results showed decreased CFU numbers in carbonated waters, while slow decreasing, stagnation or even some growth in still waters. A. fumigatus was the most resistant test species. None of the other tested mold strains survived the first 12-week test period in carbonated water. Visual growth was not detected in carbonated water samples, in contrast to all of the non-carbonated samples.  相似文献

In a previous taxonomic study based on multilocus sequencing of Fusarium from clinical specimens and hospital environments, the most common lineage was Fusarium solani species complex group 1 (FSSC 1) which is conspecific with F. solani f. sp. cucurbitae race 2, a pathogen of cucurbit fruits. The aims of our study were to determine if clinical and environmental isolates of FSSC 1 are plant pathogens and members of the same biological species as cucurbit isolates, and to determine if all isolates can germinate, grow and sporulate at 37 degrees C. Isolates from the different sources did not differ in virulence on zucchini fruits. All FSSC 1 isolates were pathogenic and produced more rot than FSSC isolates from plant hosts other than cucurbits. Both mating types were found among isolates from each of the sources, and all isolates were sexually compatible with cucurbit isolates. All isolates germinated, grew and sporulated at 37 degrees C. This is the first report in which plant pathogenicity has been verified for a collection of human clinical isolates. Our data are consistent with the hypothesis that all FSSC 1 isolates, regardless of source, are a single biological species, equally virulent plant pathogens and tolerant of the human body temperature.  相似文献
