热激处理对冷藏蚕豆种子褐变和有关酶活性的影响   总被引:7,自引:0,他引:7  
研究了45℃30、60和120s短期热激处理对蚕豆种子在1℃贮藏期间褐变和有关酶活性的影响。结果表明,采用45℃ 60s热激处理可显著降低蚕豆在冷藏期间的呼吸强度、PPO和PAL活性,抑制MDA和总酚含量及褐变度的上升,延缓叶绿素和Vc含量的下降。从而起到延缓衰老和保持品质的作用。45℃热处理120s使2周后MDA含量和褐变度上升及Vc含量下降较快,这可能是由于组织受到了热伤害造成的。  相似文献

柑橘果皮褐变严重影响果实的商品价值和耐贮性.以奉节脐橙(Citrus sinensis Osbcck)果实为材料,通过采后常温、涂蜡、低温、机械损伤等处理研究了果实果皮褐变率、苯丙氨酸解氨酶(PAL)活性及PAL上基因在果皮褐变过程中表达水平的变化。结果表明,涂蜡、损伤处理均极显著地提高果实的果皮褐变率,而低温贮藏可显著降低其发生率;贮藏期问各处理的PAL活性均呈上升趋势,损伤处理PAL活性显著高于对照。果实发生褐变或受到机械损伤后,PAL2、PAL6基因的表达均比对照明显增强。结果首次表明脐橙果实PAL活性变化以及PAL2基因的表达与果皮褐变具有非常密切的关系.  相似文献

低温胁迫对不同西葫芦品种幼苗生长与生理特性的影响   总被引:7,自引:0,他引:7  
在昼/夜温度为10℃/5℃的低温胁迫下,对‘特早’、‘早青’、‘冬宝’3个西葫芦(Cucurbit pepoL.)品种的幼苗生长和生理生化指标的变化规律进行研究。结果表明,随着低温处理时间的延长,幼苗单株叶面积、单株干重均有一定程度下降,叶绿素总含量也呈下降趋势;各品种的根系活力显著下降,电导伤害率均明显升高;丙二醛含量在处理9 d时有所下降但整体呈现缓慢上升的趋势,脯氨酸含量呈现逐渐上升的趋势,且处理6 d是脯氨酸和丙二醛两指标测定的理想时期;处理幼苗的POD活性呈"V"字形变化趋势,抗寒性强的品种酶活性也较高,CAT和SOD活性则呈现出先上升后下降的趋势,三者活性均显著高于相应对照,且以处理6 d时酶活性受影响最大。各测定指标间表现出一定的交叉适应和协调性。研究发现,西葫芦耐冷性强弱可根据幼苗电解质渗透率结合根系活力或干物质积累来综合评判,品种‘冬宝’对低温具较强耐受性。  相似文献

O2和CO2配比对气调贮藏梨采后褐变及相关理化因子的影响   总被引:1,自引:0,他引:1  
以采后'丰水'梨果实为材料,在乐扣气调试验箱中研究了O2和CO2配比对果实褐变率、多酚氧化酶(PPO)和过氧化物酶(POD)活性、丙二醛(MDA)和总酚含量的影响,以探讨适宜减轻梨气藏褐变的气体成份.结果表明:在整个贮藏过程(150 d)中,'丰水'梨果肉未发生褐变.从贮藏60 d开始,气调处理和冷藏对照果实的果皮均出现褐变,气调处理在贮藏120 d之前对果皮褐变的影响不显著,而在贮藏120~150 d内可显著减轻果皮的褐变、抑制果皮PPO和POD活性及降低总酚含量.与冷藏对照相比,气调处理可推迟果心褐变的时间,且(8%~10%)O2+3% CO2处理可完全抑制果心的褐变;气调处理亦可降低果心PPO活性、减少总酚及MDA含量;(8%~10%)O2+1% CO2处理能够显著提高果心的POD活性,而(8%~10%)O2+3% CO2处理对果实POD活性的影响不显著.可见,气调贮藏主要是通过降低'丰水'梨果皮PPO、POD活性及总酚含量来减轻组织的褐变,并以(8%~10%)O2+3% CO2处理对果实褐变因子的控制效果较理想.  相似文献

以美国‘紫李’为试材,测定经间歇升温和热处理后果实的褐变度、抗氧化酶活性、抗氧化剂含量、膜质过氧化水平、总酚和可滴定酸含量的变化。结果表明:间歇升温和热处理可适当恢复因冷害而降低的细胞抗氧化活性,清除活性氧自由基,减少膜脂过氧化产物丙二醛的积累,抑制多酚氧化酶和过氧化物酶活性升高,强化抗低温防御系统,阻止多酚逆境代谢发生,使冷害和褐变症状得以延缓和减轻;同时,还可抑制可滴定酸含量的减少和固酸比的上升,延缓后熟衰老。间歇升温处理,李贮藏两个月果实品质良好。初步认为,-0.5~0℃贮藏,每15 d加温至18~20℃并保持l d,是贮藏美国‘紫李’适宜的变温模式。  相似文献

以‘翠冠’梨为材料,研究了套双层遮光纸袋梨果实贮藏过程中蔗糖、果糖、葡萄糖、山梨醇及糖代谢中酶活性的变化规律。结果表明,贮藏套袋梨果实中果糖、葡萄糖、山梨醇和蔗糖含量都低于未套袋对照;套袋梨果实中山梨醇脱氢酶活性在贮藏的前5d都低于对照,贮藏10d后活性均高于对照,且与山梨醇含量呈现极显著正相关;贮藏套袋梨果实中蔗糖磷酸合酶(SPS)及蔗糖合酶(SS)分解和合成方向活性都是前期低于对照,贮藏后期都高于对照,且蔗糖含量与蔗糖磷酸合酶和蔗糖合酶(分解方向)活性都呈显著正相关;贮藏的套袋梨果实和对照中的山梨醇含量与果糖含量均呈极显著负相关,蔗糖含量与葡萄糖含量呈极显著负相关,即在贮藏过程中山梨醇可能转化为果糖,而蔗糖则转化成葡萄糖。  相似文献

以紫斑牡丹种子为试验材料,研究不同浓度的赤霉素(GA_3)处理对种子生根以及生根过程中营养物质、酶活性和内源激素水平变化的影响,为探讨紫斑牡丹种子萌发机制提供依据。结果表明:(1)GA_3处理能够促进种子生根,并以300 mg/L GA_3处理对种子生根效果最好,与对照相比可提前14.67 d生根,生根率可达71.00%。(2)与对照相比,GA_3处理可以在0~15 d时促进种子淀粉水解和可溶性糖的积累,并加速可溶性蛋白的消耗,在0~30 d促进过氧化物酶(POD)活性的提高,从而促进种子萌发生根。(3)在种子沙藏生根过程中,种子脱落酸(ABA)含量呈下降趋势,赤霉素(GA)、玉米素核苷(ZR)和吲哚乙酸(IAA)含量均表现出先上升后下降的趋势,与对照相比,GA_3处理可使种子GA、ZR和IAA的含量在沙藏前期明显上升,以解除种子休眠。研究发现,外源GA_3处理可以调控紫斑牡丹种子内源激素含量和POD活性的变化,促进营养物质转化,从而提前解除种子休眠使其萌发。  相似文献

研究了蚜虫危害胁迫后不同抗蚜性苜蓿品种叶片内丙二醛含量及防御性酶活性的动态变化。结果表明:在蚜虫刺吸诱导的过程中,高感品种的MDA含量始终高于高抗品种,并且高感和高抗品种均保持上升的趋势;高感品种的SOD、POD和PAL活性始终低于高抗品种,其中高感和高抗品种的SOD和POD活性均表现先上升后下降的趋势,而PAL活性上升到高峰后均趋于稳定;CAT活性在高感和高抗品种间表现为交替的上升下降;高抗品种的PPO活性前期低于高感品种,而后期高于高感品种。由此可见,在蚜虫危害胁迫下,高感和高抗品种间MDA、SOD、POD、PAL和PPO活性的变化与苜蓿的抗蚜性密切相关,均可作为苜蓿抗蚜性鉴定的生理指标,而CAT活性变化与苜蓿抗蚜性的联系有待进一步研究。  相似文献

PRO-LONG涂膜对采后贮藏荔枝果实色泽和酶活性变化的影响   总被引:2,自引:0,他引:2  
本文以荔枝品种“槐枝”为材料,以1.5%和2.5%Pro-long涂膜处理果实,研究Pro-long涂膜对果后贮于4℃的荔枝果皮色泽变化和有关酶活性的变化。对照和处理的HunterL值和b值均随贮藏时间的延长而降低,处理的比对照的下降慢。采后贮藏开始21d内,对照和处理的HuntCra值变化很小,保持相对稳定,之后的贮藏时间内有较显著的下降,说明贮藏初期果实的红色特性变化较少,之后变化很明显,这与果皮花色素苷、类黄酮、总酚的变化是相对应的。在贮藏后期,处理的HuntCra值下降明显较对照慢。Hunter值的变化与在贮藏过程中果实外观逐渐变成暗红和揭变以及在果实的来后根变中起着重要作用的多酚氧化酶和部分地涉及褐变过程的过氧化物酶的变化相对应。处理的过氧化物酶活性增加较对照慢,而且,与对照比较,多酚氧化酶的峰值稍稍延迟了。未发现l‘5%和2.5%处理间有明显差别。因此,Pro-long涂胶对荔枝的应用可以部分地降低多酚氧化酶和过氧化物酶,影响果皮花色素苷的降解、类黄酮和总酚的变化,延迟了褐变过程的发生。  相似文献

用25mW红外线激光辐照铁棍山药芦头10、15和30min后,其芦头芽萌发受抑,块茎含水量、可溶性总糖和可溶性蛋白含量下降幅度均变小,丙二醛(MDA)和O_(2·)~-含量上升幅度也变小,贮藏前期的超氧化物歧化酶(SOD)活性升高幅度增大,贮藏后期的SOD活性下降幅度变小,以辐照30min的效果为最佳。  相似文献

为了开发高效的食用菌绿色保鲜剂,以新鲜的白色金针菇为供试材料,以花椒精油和丁香精油为供试熏蒸剂,分别在常温(25±1)℃和低温(4±1)℃条件下开展了适用于金针菇保鲜的精油种类和浓度的筛选试验,并对金针菇贮藏期内的感官评价、失重率、呼吸强度、褐变度、多酚氧化酶(PPO)活性、苯丙氨酸解氨酶(PAL)活性、丙二醛(MDA)以及总酚含量进行了测定。结果显示,常温(25±1)℃条件下用0.1 mL·kg-1花椒精油和0.5 mL·kg-1丁香精油保鲜效果优于其他处理,且有统计学意义(P<0.05),0.1 mL·kg-1花椒精油和0.5 mL·kg-1丁香精油处理组感官评分分别高于对照组23.4%和27.8%,二者均能够抑制金针菇褐变、减轻腐败变质,且有统计学意义(P<0.05);在低温(4±1)℃贮藏试验中发现,0.1 mL·kg-1花椒精油和0.5 mL·kg-1丁香精油均能有效抑制呼吸强度和PPO活性的升高(P<0.05),其呼吸高峰较对照组分别降低了28.3%和39.6%;贮藏15 d后,精油处理组PPO活性较对照组分别降低了8.2%和16.6%;精油处理有效降低了MDA含量的产生,保持着较高的总酚含量、减轻腐烂褐变的程度。第15天时,对照组MDA含量为1.75 μmol·g-1,而花椒精油和丁香精油处理组MDA含量分别比对照组低0.15、0.40 μmol·g-1,丁香精油处理组显著低于花椒精油处理组和对照组(P<0.05)。研究结果表明,0.1 mL·kg-1花椒精油和0.5 mL·kg-1丁香精油均对金针菇采后贮藏保鲜效果显著,其中,0.5 mL·kg-1丁香精油的保鲜效果最明显,在15d的贮藏期内,金针菇依然保持着良好的品质,而对照组已经轻微褐变,部分开始腐烂。研究结果为花椒精油和丁香精油应用于金针菇采后贮藏保鲜提供了理论依据。  相似文献

As an attempt to understand some aspects of the protein catabolism in the resting yam (Dioscorea rotundata) tuber with respect to the storage problems of the tuber, gamma-glutamyl transpeptidase (gamma-GTP) was isolated from healthy tubers after aerobic storage for varying periods. The molecular weight was determined by SDS-gel electrophoresis and the activity by spectrophotometry using gamma-glutamyl alpha-naphthylamide as substrate. Molecular weight and activity were plotted versus duration of the tuber storage. The molecular weight of the gamma-GTP from newly harvested tuber was 2.2 . 10(5). Both the mol. wt and activity decreased linearly with tuber storage, the two processes having approximately equal first order rate constant, 1.2 . 10(-2)/week, with a half-life of 58 weeks. High molecular weight forms of tuber gamma-GTP when dissociated and gel electrophorized gave in each case a lower molecular weight form that also had enzymic activity. A plot of the mol. wt versus the activity gave a straight line which intersected the mol. wt axis at 200000, which is the mol. wt of the smaller subunit of gamma-GTP. These results show that yam tuber gamma-GTP undergoes progressive disintegration during tuber storage and in this process the components are lost probably from the larger subunit after proteolytic cleavage and subsequent dissociation of the cleaved components.  相似文献

研究了在厚度为0.04 mm的聚乙烯(PE)袋内充气成分为2%O2、5%CO2和93%N2于10℃下贮藏时气调袋装去壳雷笋褐变和木质化进程.与对照相比,低氧气调包装(MAP)抑制了去壳雷笋贮藏前期丙二醛(MDA)的生成,显著抑制了过氧化物酶(POD)(P<0.05)和苯丙氨酸解氨酶(PAL)活性(P<0.01),最终显著抑制了笋肉的褐变(P<0.01),显著抑制了木质素(P<0.05)和纤维素的合成,从而延长了保鲜期.去壳雷笋的褐变可能由POD和PAL活性作用引起.POD和PAL活性的增加也可能诱导了木质素的合成.  相似文献

Dormancy is a mechanism that regulates the timing of sprouting (germination) of affected plant parts as well as ensures that the food quality of edible parts is maintained in storage until the following growing season. In yam, however, little is known about the control of tuber initiation or tuber dormancy. The objective of this study was to determine the effects of selected plant growth regulators (PGRs) on tuber initiation and dormancy, using an in vitro system. In two replicated experiments, 2-chloroethylphosphonic acid (ethephon, an ethylene source), abscisic acid (ABA) and gibberellin (GA3) – and their inhibitors silver nitrate, fluridone and 2-chloroethyl-trimethylammonium chloride, respectively – were added at two concentrations to the culture medium prior to explant culture. Dates of micro-tuber initiation and sprouting (end of dormancy) and tuber number were recorded. In the control (no PGR) in Experiment 1, micro-tubers were initiated at the base of the stem after 176 days and sprouted 235 days later, that is 411 days after culturing. Most PGR treatments had only small effects (±30 days) on the duration of dormancy and the time of micro-tuber initiation. However, in GA3 micro-tuber initiation occurred after 76 days, about 100 days earlier than in the control, whereas fluridone affected the position of micro-tubers and duration of dormancy. With fluridone treatments, tubers were found at the base of the stem (normal position) and on lower and upper nodes. Lower node tubers sprouted within 225 days of culturing compared with about 420 days after culturing at other nodal positions and in other PGR treatments. These data suggest an important role for ABA and gibberellic acid in yam micro-tuber initiation and the induction of dormancy.  相似文献

The control of field tuber dormancy in the yam (Dioscorea cayenensisD. rotundata complex) is poorly understood. Although studies have examined single environmental factors and chemical treatments that might prolong tuber dormancy and storage, only a few were focused on further tuber sprouting. The present study concerns microtubers obtained by in vitro culture. When microtubers were harvested (after 9 months of culture) and directly transferred on a new medium without hormones, the tubers rapidly sprouted in in vitro conditions. No dormancy was observed in this case. Harvested microtubers were also stored dry in jars in sterile conditions during 2 to 18 weeks before in vitro sprouting. In this case, microtubers stored during 18 weeks sprouted more rapidly than those stored 8 weeks. A constant “dormancy-like period” (storage duration + sprouting delay) was observed, between 20 and 28 weeks respectively for the more rapid and the slower microtubers. The size of the tubers used for the storage had great influence on further sprouting. The larger they were, the better they sprouted. Light during storage had no effect on the sprouting delay while a temperature of 25 °C permit a quicker sprouting than 18 °C. The medium used to obtain microtubers could also have an effect on sprouting rate.  相似文献

为了明确异淀粉酶基因(ISA 3)在山药淀粉代谢中的作用,该研究以‘毕克齐’和‘大和长芋’山药为试验材料,测定了块茎中淀粉及组分含量和异淀粉酶活性等;采用RT-PCR技术克隆了ISA 3,并进行生物学分析及山药块茎不同膨大期和不同组织间ISA 3基因的表达等。结果表明:(1)山药‘大和长芋’的直链淀粉、支链淀粉和总淀粉含量均显著高于‘毕克齐’,且两品种的淀粉含量随生长发育的变化均呈先升高后降低的趋势,并均于种植后120 d时达到最高,但‘毕克齐’的异淀粉酶(ISA)活性在整个膨大期均高于‘大和长芋’。(2)成功克隆获得山药ISA 3开放阅读框长1584 bp,编码527个氨基酸;ISA3为亲水性蛋白。(3)不同品种块茎在膨大时期的ISA 3基因表达趋势不同,‘毕克齐’中呈先显著上调随后下调,而在‘大和长芋’中表达总体下调,且在山药的叶、茎和块茎中均有表达,存在明显的组织特异性。(4)ISA活性与山药淀粉及支链淀粉含量呈显著和极显著正相关关系,但ISA活性与ISA 3的表达量呈负相关关系。研究表明,异淀粉酶参与了山药块茎中淀粉的合成,且主要对支链淀粉的合成起关键作用,ISA 3基因的表达可能对异淀粉酶活性和淀粉的合成起负调控作用。  相似文献

Rabbit kidney cortex tissue slices were made ischemic (37 degrees C) for 60 min and then either reperfused in warm (37 degrees C) oxygenated physiologic buffer for 210 min or placed in UW Na gluconate solution (+/- quinacrine; 100 micromol/L) for 18 h followed by warm aerobic reperfusion. Slices were sampled at intervals and analyzed for malondialdehyde (MDA) content by HPLC. Control (nonischemic) slices had no change in MDA content over the duration of the experiment. Hypothermic storage of nonischemic slices did not result in any increase in MDA during reperfusion. Ischemic slices showed significant increases in MDA content during the first 1.5 h of reperfusion and remained elevated for the remainder of the experiment. Hypothermic storage of warm ischemic kidney slices resulted in a significant decrease in MDA content during the storage period. However, MDA content in these slices increased during warm reperfusion and was significantly higher than that in nonischemic controls. Quinacrine added during hypothermic storage of warm ischemic slices significantly decreased slice MDA content during warm reperfusion, an effect which was lost by increasing the storage solution calcium content. This study shows that aerobic hypothermic storage can aid in reducing oxidative stress in warm ischemic kidney tissue during reperfusion. This study suggests that the effects of quinacrine are at the level of the mitochondrion and not as an antioxidant compound.  相似文献

为探讨蒭雷草(Thuarea involuta)在热带珊瑚岛干旱环境下的适应能力,对干旱胁迫下蒭雷草叶片抗逆生理指标的变化进行了研究。结果表明,干旱胁迫初期,叶片的丙二醛(MDA)含量随胁迫程度增加的差异不显著;随胁迫时间的延长,除重度胁迫下先迅速增加后急速下降外,其余处理的变化较小。超氧化物歧化酶(SOD)、过氧化物酶(POD)和过氧化氢酶(CAT)活性随干旱胁迫程度增加而升高;随时间延长,SOD活性不断上升,POD活性基本稳定,CAT活性则先下降后上升。除轻度胁迫下可溶性蛋白(SP)含量低于对照外,其余处理均随干旱胁迫程度增加而增加;干旱胁迫下的脯氨酸(Pro)含量随时间延长呈先上升后下降的变化趋势,但在处理第18天时不同胁迫程度间均差异不显著。因此,蒭雷草具有较强的抗旱能力,可用于南海诸岛的人工植物群落构建和植被恢复以营造良好生态环境。  相似文献
