The effects of stimulating the midbrain central gray matter (CGM) on neuronal response in the ventroposteromedial (VPM) nucleus produced by stimulating tooth pulp, A-alpha and A-delta fibers of the intraorbital nerve and the caudal nucleus of the spinal trigeminal tract (CN STT) were investigated during experiments on cats under thiopental-chloralose anesthesia. It was found that applying trains of stimuli to the CGM produced excitatory responses in a proportion of the test neurons with latencies of up to 30 msec. Application of conditioning stimulus to the CGM led to suppression of response of efferent stimulation in neurons belonging to low-threshold, convergent, and high-threshold groups. Responses produced in 40% of neurons by stimulating tooth pulp and A-delta fibers of the suborbital nerve, as well as those evoked in 26.4% of thalamic VPM cells by stimulating A-alpha fibers of the suborbital nerve were completely suppressed. The inhibitory effect found when stimulating CGM on response in certain neurons, produced by stimulating both the peripheral nerve and the CN STT, would indicate that the CGM could exert an influence on the activity of thalamic VPM neurons directly.A. A comparative study was conducted on the effects of stimulating different central gray matter (CGM) areas on neuronal activity of the trigeminal nucleus during nociceptive stimulation before and after complete section of medial brainstem structures. The possible role of opiate and non-opiate systems in inhibition of nocieptive afferents is discussed. The CGM was found to be a functionally diverse structure: pain-suppressing and pain-promoting systems exist side by side at this site. The inhibitory influence of the ventromedial CGM is exerted by activation of raphe nucleus neurons; that of the dorsomedial area could well be mediated by involvement of the lateral bulbar area and/or activation of rostral brain structures.I. Stimulating the midbrain central gray matter (CGM) with trains of 10–20 stimuli was found to inhibit response to electrical stimulation of infraorbital nerve and tooth pulp A-alpha and A-delta afferents at 100 msec intervals in 65% of the caudal trigeminal nucleus in neurons tested during experiments on cats under chloralose-Nembutal anesthesia. Response was inhibited most effectively in convergent neurons (activated by stimulating infraorbital nerve and tooth pulp A-alpha and A-delta afferents) to stimulating tooth pulp (0.76) and, to a somewhat lesser degree, to stimulation of A-alpha afferents (0.6). For high-threshold neurons (activated by stimulating infraorbital nerve and tooth pulp A-delta afferents), success rate of inhibiting response under the effects of CGM stimulating measured 0.71 and 0.48 for low-threshold cells (activated by stimulating infraorbital nerve A-alpha afferents). Stimulating CGM produced an excitatory response in 10 caudal trigeminal nucleus neurons within 7.5–20 msec; after this neurons showed no reaction to peripheral nerve stimulation for a 200–450 msec period. The possible involvement of these neurons in pressing the mouth-opening reflex produced by CGM stimulation is discussed in this article.A. A. The effects of stimulating the midbrain central gray matter (CGM) on motoneuronal response in trigeminal nerves were investigated in anesthetized cat. It was found that stimulating the CGM did not induce postsynaptic response in these motoneurones. Conditioning stimulation of the CGM brought about suppression of motoneuronal postsynaptic response to stimulation of tooth pulp and high threshold infraorbital nerve afferents without affecting motoneuronal antidromic response and jaw-opening reflex as induced by stimulating the caudal nucleus of the spinal trigeminal tract. It was thus concluded that stimulating the CGM exerts no direct effect on motoneurons but does have an influence on postsynaptic response — a result of modulation of the afferent spike flow at interneuronal level.A. The effects of severing the spinal trigeminal tract and its caudal nucleus on high-threshold jaw-opening reflex elicited by tooth pulp stimulation were investigated during experiments on cats under chloralose-Nembutal anesthesia. Low-threshold jaw-opening reflex produced by stimulating the A--infraorbital nerve at an intensity 2–3 thresholds in relation to the most excitable fibers on this nerve was also observed, as well as suppression of these reflexes induced by central gray matter stimulation. It was found that spinal trigeminal tract section produces a 8–52% increase in high-threshold reflex. The amplitude of low-threshold reflex either remained unchanged or showed a slight tendency to rise or fall. Brief stimulation of the central gray matter produced a 100% decrease in high-threshold reflex in intact animals compared with a 40–60% decrease after section of the trigeminal tract. Protracted stimulation of the central gray brought about an 80% decline in high-threshold reflex in intact animals as against 25–30% after section. The degree to which brief stimulation of the central gray produced depression of low-threshold stimulation remained unchanged by trigeminal tract section. Protracted stimulation of the central gray matter brought about a 25–50% reduction in low-threshold reflex in intact animals and a reduction of 75% in three animals and 15–20% in four animals. This implied that the caudal nucleus of the spinal trigeminal tract exerts a more substantial influence on the process of high- than low-threshold reflex inhibition when the central gray matter is stimulated.A. The effects of stimulating the periaqueductal gray matter (PAG) on two types of startle reflex (spino-bulbo-spinal reflex produced by intensive stimulation of the peripheral nerves and low-threshold tactile spino-reticulo-spinal reflex) as well as high-threshold jaw-opening reflex arising in response to tooth pulp stimulation were investigated in cats anesthetized with chloralose. Simulating most PAG test sites led to pronounced inhibition of jaw-opening reflex, profound depression of spino-bulbo-spinal reflex, and moderate inhibition of tactile reflexes. The facilitatory effect of stimulating a number of PAG sites on the latter reflexes was demonstrated. Effects of PAG stimulation fell into two classes: brief, measurable in hundreds of msec and more prolonged, measured in minutes and seconds. Findings would indicate certain differences between the effects of PAG stimulation low-threshold (non-nociceptive) and high-threshold (nociceptive) startle reflexes, of which the possible mechanisms are discussed.A. Background activity was recorded in 272 neurons of the ventrolateral thalamic nucleus before and after systemic haloperidol and droperidol injection at a cataleptic dose using intracellular techniques during chronic experiments on cats in a drowsy condition. Brief burster discharges lasting 5–50 msec and following on at a high intraburst spike rate (of 200–450 Hz) were characteristic of neuronal activity in intact animals. Regular discharges occurred at the rate of 2–2.5 Hz or occasionally 3–4 Hz in 15% of cells. Numbers of neurons with the latter activity pattern rose to 22 and 30%, respectively, following haloperidol and droperidol injection. Both irregular and prolonged (80–300 msec) regular discharges were recorded in one third of the total. A relatively low intraburst spike rate (of 60–170 Hz) was observed in 37% of cells following 10 days' haloperidol injection. These changes are thought to be produced by intensified inhibitory effects on neurons of the thalamic ventrolateral nucleus from the substantia nigra and reticular thalamic nucleus following blockade of dopaminergic and -adrenergic receptors.A. Projections of the central cerebellar nuclei to the intralaminar thalamic nuclei were studied in cats with the use of light
and electron microscopy. Almost all intralaminar nuclei were shown to obtain cerebello-thalamic projections. The entire complex
of the central cerebellar nuclei serves as a source of such projections; yet, involvement of different nuclei is dissimilar.
Destruction of the central and, especially, caudal regions of the fastigial nucleus evoked in the intralaminar thalamic nuclei
degenerative changes in the nerve fibers (from swelling and development of varicosities up to total fragmentation). Pathological
phenomena could be noticed in the most caudal regions of the above thalamic nuclear group, including the medial dorsal nucleus.
Projections of the cerebellar interpositus nucleus were directed toward nearly the same regions of the intralaminar nuclei;
degeneration was more intensive (covered thecentrum medianum) when posterior regions of the interpositus nucleus were destroyed. Destruction of the lateral cerebellar nucleus evoked
a similar pattern of pathological changes, but degeneration was also observed in some structures of the ventral and anterior
nuclear groups of the thalamus. Electron microscopic examination showed that degeneration of dark and light types developed
in the fiber preterminals and terminals. It can be concluded that the central cerebellar nuclei project not only to the ventral
complex of the thalamic nuclei, but also to the anterior, medial, and intralaminar nuclear groups (rostral and caudal portions). 相似文献

The effects were studied in waking cats of brief stimulation (20 stimuli at a rate of 400 Hz) of the central gray matter (CGM) and dorsal raphe nucleus (DRN) on high-threshold jaw-opening reflex (HJOR) evoked by tooth pulp stimulation during blockade of serotonin synthesis produced by application of 300 mg/kg parachlorophenylalanine (PCPA) i.p. Inhibitory effects of CGM and DRN stimulation had already declined in comparison with post-stimulation (but pre-PCPA) level within 24 h after PCPA application; 96 h afterwards, inhibition of HJOR induced by CGM and DRN stimulation had become only minimal: amplitude of the reflex had declined to 30–35% and duration of inhibitory effects ws 200–250 msec. It is therefore deduced that serotonin contributes to the HJOR depression induced by CGM and DRN stimulation and the possible involvement of other neuromodulators in this effect is discussed.A. A. The effect of stimulation of the mesencephalic central gray matter and raphe nuclei on jaw opening reflexes evoked by excitation of high-threshold (dental pulp) and low-threshold (A-alpha) fibers of the infraorbital nerve afferents was studied in cats anesthetized with chloralose and pentobarbital. The jaw opening reflex evoked by stimulation of the dental pulp was shown to be effectively suppressed by conditioning stimulation of the central gray matter and raphe nuclei. The reflex evoked by stimulation of low-threshold infraorbital nerve afferents also was depressed (but less deeply and for a shorter period than the reflex evoked by stimulation of the dental pulp) during stimulation of the raphe nuclei and caudal zone of the central gray matter, but was unchanged after stimulation of the points located in the rostral zone of the central gray matter. Application of single stimuli or bursts of five stimuli with a frequency of 100 Hz had no effect on the reflexes studied. Short-term stimulation with a burst of 10–20 stimuli with a following frequency of 200–400 Hz led to inhibition of the reflexes, which lasted 450–1000 msec. Long-term stimulation of the central gray matter and raphe nuclei for 30 sec with a frequency of 50 Hz caused inhibition of jaw opening reflexes evoked by stimulation of both high- and low-threshold afferents for 60 min. Impulses from the central gray matter and raphe nuclei thus have a mainly inhibitory action on the jaw opening reflex evoked by stimulation of high-threshold afferents, but they act less effectively on the reflex evoked by stimulation of low-thres-hold afferents. It is shown by the use of Golgi's method in Antonova's modification that the neuronal structure of the periaqueductal gray matter (PGM) in the frontal plane is characterized by the presence of small and medium-sized cells of "reticular type," which can be subdivided into three types: fusiform, triangular, and multipolar. On the basis of the visual distribution of these types of neurons and also of statistical analysis of 800 identified neurons, two regions can be distinguished: medial, directly surrounding the aqueduct of Sylvius, containing small neurons, among which the fusiform kind predominate significantly (P<0.001), and a lateral region with larger neurons, with significantly (P<0.001) more triangular cells. Neurons in the medial region show a characteristic and strong (P<0.001) tendency for their dendrites to be oriented toward the lumen of the aqueduct, and through them the physiologically active substances of the CSF may influence the functional activity of neurons of PGM.Central Research Institute of Reflex Therapy, Moscow City Council Main Health Board, Moscow. Electrical stimulation of the periaqueductal gray matter (PGM) bringing about inhibition of high-threshold mouth-opening reflex on somatosensory response of neurons belonging to the pontine caudal reticular nucleus (CN) and the reticular gigantocellular nucleus (GN) were investigated in cats under light chloralose-induced anesthesia. It was found that inhibitory effects on CN and GN neurons adhered to the principle of control over their main synaptic input: in GN neurons and particularly in high-threshold cells, inhibition mainly affected response to nociceptive stimuli, in contrast to CN neurons, and low-threshold cells in particular, where response to non-nociceptive stimulation was inhibited. Stereotaxic microinjections of horseradish peroxidase (HP) were made into different parts of the rostral and caudal periaqueductal gray (PAG) in cats to study corticofugal projections to the PAG. The method of retrograde axonal transport of HP demonstrated labeled neurons in the I and II somatosensory areas, frontal, cingular and insular cortex of the brain. It was shown that the II somatosensory cortex projects to all the areas of the rostral and caudal PAG. The frontal cortex projects to the dorsolateral quadrant of the PAG. The findings obtained enabled the detection of the morphological substrate of the corticofugal effects on one of the antinociceptive brain structures--the PAG. 相似文献

In chronic cat experiments, bicucullin-induced GABA receptor blockade in the dorsal and ventral parts of the rostral caudate nucleus resulted in complicated changes in spontaneous activity of 19 of 55 neurons, primarily in the cells with low spike frequency. Changes were observed also in 8 to 31 neurons of VA-VL nuclei of the thalamus. The blockade of striatal GABA receptors was accompanied by changes in motor behavior and emotional motivational state of the animals. The character and latency of unit activity rearrangements in the caudate nucleus and modifications of cat's behavior depended on the localization of bicucullin injection point within different parts of the caudate nucleus. 相似文献

The effects induced on neuronal firing by microiontophoretic application of the biological amines noradrenaline (NA) and 5-hydroxytryptamine (5-HT) were studied "in vivo" in ventral-anterior (VA) and ventrolateral (VL) thalamic motor nuclei of anaesthetized rats. In both nuclei the amines had a mostly depressive action on neuronal firing rate, the percentage of units responsive to NA application (88%) being higher than to 5-HT (72%). Short-lasting (less than 2 min) and long lasting (up to 20 min) inhibitory responses were recorded, the former mostly evoked by NA and the latter by 5-HT ejection. In some cases 5-HT application had no effect on the firing rate but modified the firing pattern. NA-evoked responses were significantly more intense in VL than in VA neurons. Short-lasting inhibitory responses similar to NA-induced effects were evoked by the alpha2 adrenergic receptor agonist clonidine and to a lesser extent by the beta adrenergic receptor agonist isoproterenol. Inhibitory responses to 5-HT were partially mimicked by application of the 5-HT(1A) receptor agonist 8-hydroxy-2(di-n-propylamino)tetralin (8-OH-DPAT) and of the 5-HT2 receptor agonist alpha-methyl-5-hydroxytryptamine (ALPHA-MET-5-HT). The latter evoked excitatory responses in some cases. Both 5-HT agonists were more effective on VA than on VL neurons. The effects evoked by agonists were at least partially blocked by respective antagonists. These results suggest that although both 5-HT and NA depress neuronal firing rate, their effects differ in time course and in the amount of inhibition; besides aminergic modulation is differently exerted on VA and VL. 相似文献

Of 103 neurons in the rostral part of the posterior sigmoid gyrus of the cat cortex 30 responded to stimulation of the ventro-posterolateral and ventrolateral nuclei of the thalamus (VPL and VL), 42 responded to stimulation of VL only, and 31 to stimulation of VPL only. It was shown by intracellular recording that stimulation of VPL induces a spike response with or without subsequent IPSPs in some neurons and an initial IPSP in others. The spike frequency of single neurons reached 60/sec, but the IPSP frequency never exceeded 10–20/sec. Stimulation of VL was accompanied by: a) antidromic spike responses; b) short-latency monosynaptic EPSPs and spikes capable of following a stimulation frequency of 100/sec; c) long-latency polysynaptic EPSPs and spikes appearing in response to stimulation at 4–8/sec; d) short-latency IPSPs; e) long-latency IPSPs increasing in intensity on repetition of infrequent stimuli. It is concluded that the afferent inputs from the relay nuclei to neurons of the somatosensory cortex are heterogeneous. An important role is postulated for recurrent inhibition in the genesis of the long-latency IPSPs arising in response to stimulation of VL, and for direct afferent inhibition during IPSPs evoked by stimulation of VPL. It is shown that the rostral part of the posterior sigmoid gyrus performs the role of somatic projection and motor cortex simultaneously.A. A.
