The marine diatoms Lauderia annulata and Synedra planctonicaharvested during exponential growth were exposed to differentdoses of u.v.-B (286, 439 and 710 J m–2 d–1) for2 days. Uptake of 15N-nitrate was estimated before, during andafter u.v.-B radiation over 2 days. Exposure to high levelsof u.v.-B (710 J m–2 d–1) caused irreversible damageat the second daily irradiance. Lauderia cells were less affectedby u.v.-B stress than Synedra cells. 15N-nitrate uptake wasreduced under u.v.-B irradiance but could be reactivated within1 day following exposure to a low dose (286 J m–2 d–1).Higher levels of u.v.-B radiation (710 J m–2 d–1)led to irreversible damage. The pattern of 15N-incorporationinto several amino acids of Lauderia varied after 2 days ofu.v.-B radiation. 15N enrichment of glutamine increased markedlyafter u.v.-B stress (717 J m–2 d–1) whereas I5Nexcess of aspartic acid was significantly reduced. Results arediscussed with reference to the u.v.-B damage of the nitratetransport system.  相似文献

Field and laboratory study of zooplankton has been hamperedby the lack of standardized and reliable culture methods. Thispaper describes a zooplankton bioassay chamber for both laboratoryand in situ use that is easy to construct, inexpensive, andsuitable for even very small zooplankton species such as Bosmina.The chamber, which allows easy access to animals for examinationor counting, may be particularly useful in toxicity bioassays. 1Financial Support was provided by NSF Grant # DDP 75-12949-A01and DDP 76-80652 to W.J. The investigation was carried out at one station in Korsfjorden,a typical deep silled fjord of western Norway. During 14 cruisesfrom 4 February to 30 June 1977 ATP, chlorophll a, phaeopigmenta, and in situ 14C-assimilation were measured in the net (>30µm), nano and ultraplankton (<5 µm). Sampleswere collected from five light depths within euphotic zone.The impact of hydrographical conditions and light regime onthe bloom dynamics was also studied. In the periods 4 February-7 March and 13 April-30 June, ultraplankton contributed >60%to the total primary production while net and nanoplankton dominatedfrom 7 March to 13 April. The diatoms Skeletonema costatum,Chaetoceros compressits and C. debilis, and Rhizosolenia hebetatavar. semispina made up the main part of the biomass on 21 March,28 March and 4 April respectively. A shade adapted diatom societywas located at the top of the nutricline in late June with S.costatum, Chaetoceros spp., and Thalassiosira spp. as the dominantspecies. The highest assimilation number of eight for the netplankton and four for the ultraplankton were found at the depthof 32% light intensity on 28 March and 24 May respectively.Linear relationships were found between chlorophyll a and ATPfor the different size fractions with regression slopes rangingfrom 4.3 to 5.8. The total primary production for the periodof investigation was calculated to 74 g C m–2. Light regimeand water column stability were decisive factors for the outburstof the first diatom bloom in late March. Grazing on net planktondiatoms increased during late March-early April. Changes inthe longshore wind-stress component were found to be essentialfor the understanding of the bloom dynamics.  相似文献

The two parameters of the hyperbolic tangent equation, Pm and, were estimated from in situ vertical profiles of primary productionusing mesocosm data along a nutrient gradient. The parameters,derived from 4-h (around noon) 14C incubations, were used togetherwith the photosynthesis-light curve and hourly solar radiationdata to calculate daily primary production rates (Pd). Approximately40% of the daily production occurred in the 4 h around noon.Considering parameter uncertainty, there was no indication ofan increase in variation in production with increased nutrientloading, nor did biomass-specific P-I parameters increase. Annualproduction ranged from 82 to 901 g C m–2 year–1and was highest in the highest nutrient treatment tank. Dailyproductivity ranged from 0.02 to 9.1 g C m–2 day–1and was significantly correlated, in all treatments, with acomposite parameter BI0/k (where B is phytoplankton biomass;I0 is daily radiation and k is the extinction coefficient).Linear regressions of Pd against BI0/k indicated that much ofthe variability (86%) in productivity was explained by lightavailability and phytoplankton biomass. Two approaches for predictingproductivity were compared: (i) predicting production directlyfrom environmental variables (i.e. BI0/k) and (ii) predictingthe parameters of the P-I curve from environmental variablesand using these to calculate daily production.  相似文献

A high-rate oxidation pond is studied as a model system forcomparing 14C and oxygen evolution methods as tools for measuringprimary productivity in hypertrophic aquatic systems. Our resultsindicate that at very dense algal populations (up to 5 mg chl.a l–1) and high photosynthetic rates, 14C based resultsmay severely underestimate primary productivity, unless a wayis found to keep incubation times very short. Results obtainedwith our oxygen electrode were almost an order of magnitudehigher than those obtained by all 14C procedures. These highervalues correspond fairly well with a field-tested computer-simulationmodel, as well as with direct harvest data obtained at the samepond when operated under similar conditions. By manipulation of various growth regulators and physical conditions,plants have been regenerated from excised roots, stem segments,cotyledons, leaves, and callus cultures of red cabbage (Brassicaoleracea var. capitata) grown under in vitro conditions. Shootbuds were induced on isolated root segments (1 cm long) culturedon Murashige and Skoog's medium and the frequency of bud formationwas greatly enhanced by the addition of kinetin (0.5 part 10–6).Callus obtained from the seeds, cotyledons, and hypocotyl segmentscultured on a medium fortified with 2,4-D (1 part 10–6),kinetin (0.1 part 10–6), and coconut milk (10%, v/v) hasbeen repeatedly subcultured. The callus is slow growing, andon transference to a kinetin (2 parts 10–6) and IAA (2parts 10–6) medium underwent morphogenesis to give riseto plants. The significance of the propagation of red cabbageby in vitro culture is pointed out.  相似文献

Photosynthetic carbon metabolism was studied with Chroomonassp. cells in which the rate of photosynthesis was inhibitedunder both an anaerobic condition and high concentrations ofoxygen. The time course of 14C-incorporation into photosyntheticproducts showed that 3-phosphoglycerate was the initial productof photosynthetic CO2 fixation in Chroomonas sp. cells. During5-min photosynthesis, a considerable amount of 14C was incorporatedinto the insoluble fraction (mostly cryptomonad starch), andoxygen predominantly affected 14C-incorporation into this fraction.Although 14C-incorporation into intermediates of the photorespiratorypathway increased with increasing O2 concentration, the amountswere much less than expected from the degree of oxygen inhibition.It is noteworthy that 14C-dihydroxyacetone phosphate accumulatedduring photosynthesis only under the anaerobic condition, whereasthe levels of the other phosphate esters were scarcely affectedby the oxygen concentration. Ribulose-1,5-bisphosphate carboxylase from Chroomonas sp. wascompetitively inhibited by oxygen, and its Km(CO2) value wassimilar to those of terrestrial C3 plant enzymes. Incorporation of 14C-phenylalanine, 14C-carbon dioxide, 14C-glucose,and 14C-glycine into the protein of Chlorella is inhibited bycycloheximide. A concentration of 2.5 µg per ml inhibitsincorporation by about 80 per cent; increasing the concentrationup to 10 µg per ml does not increase the degree of inhibition.The incorporation of 14C-adenine into ribonucleic acid (RNA)and deoxyribonucleic acid (DNA), and of 14C-glucose into polysaccharideis also inhibited. Unlike inhibition of protein synthesis, thatof nucleic acid and polysaccharide synthesis is observed onlyafter some delay. The delay is shortest for DNA synthesis andlongest for polysaccharide synthesis. Inhibition of 14C-glycineincorporation into DNA and RNA follows a similar pattern tothat obtained with 14C-adenine but the delay is much shorter.Cycloheximide also inhibits the formation of isocitrate lyasc(isocitrate-glyoxylate lyase, EC 4.1.3.1 [EC] ) when autotrophicallygrown cells are supplied with acetate.  相似文献

Microswards of white clover (Trifolium repens L.) were grownin controlled environments at 10/7, 18/13 and 26/21 °C day/nighttemperatures. The vertical distribution of leaves of differentages and their rates of 14CO2-uptake in situ were studied. Extending petioles carried the laminae of young leaves throughthe existing foliage. A final position was reached within 1/4to 1/3 of the time between unfolding and death. Newly unfoldedleaves had higher rates of 14CO2-uptake per leaf area than olderones at the same height in the canopy. At higher temperatures,the decrease with age was faster. However, the light-photosynthesisresponse of leaves which were removed from different heightsin the canopy varied much less with leaf age than did the ratesof 14CO2-uptake in situ. The comparison of the rates of 14CO2-uptake in situ with thelight-photosynthesis response curves suggests that young leavesreceive more light than older ones at the same height in thecanopy. This would imply that young white clover leaves havethe ability to reach canopy positions having a favourable lightenvironment. This ability may improve the chances of survivalof white clover in competition with other species. Community respiration (R) was determined in Bransfield Straitfrom oxygen changes in water samples incubated in borosilicatebottles maintained at in situ temperature. The respiratory electrontransport system (ETS) activity of seawater communities wasalso measured from the same samples. Both data sets were relatedby the regression equation: log R (mg O2 m–3 day–1)=0.462+0.730xlogETS activity mg O2 m–3 day–1) (r=0.80, n=23). Fromthis equation and 37 ETS activity depth profiles, we calculatedthe integrated (0–100 m) community respiration as beingin the range 1.2–4.5 g O2 m–2 day–1 (mean=2.2).These values do not differ significantly from other publishedresults for the Arctic and Antarctic Oceans. Assuming a respiratoryquotient of unity, the areal respiration ranges between 0.45and 1.69 g C m–2 day–1 (mean=0.8). This would representan important sink for the primary production reported for BransStrait. The spatial distribution of community respiration showedhigher values associated with the warmer and phytoplankton-richwaters outflowing from Gerlache Strait into Bransfield Strait,and with the front that separates Bellingshausen Sea watersfrom Weddell Sea waters. We suggest that this pattern of distributionmay be related to the transport of organic matter by the BransfieldCurrent along the front.  相似文献

The effect of oxygen on photosynthetic 14CO2 fixation in theair-grown freshwater flagellate Chroomonas sp. (Cryptophyta)was studied. Considerable inhibition by anaerobiosis was observedonly under light-saturated conditions with no effect from theCO2 concentration. This inhibition was reversed by 2% O2. With,more than 2% O2, the rate of 14CO2 fixation was inhibited; 100%O2 caused about 20% inhibition which could be reversed by 2%O2. The degree of inhibition was only slightly higher at lowconcentrations (less than 0.43 mM NaHCO3) than at high CO2 concentrations,indicating that photorespiration is not a main cause of thisinhibition. A study has been made of photosynthetic 14CO2 fixation by isolated‘mature’ internodes of Nitella translucens. Experimentalconditions were similar to those used in studies of the ionicrelations of these cells. Maximum rates of photosynthesis were33–40µµmoles CO2, fixed per cm2 of surfacearea per second (equivalent to 12–15 /xmoles fixed permg chlorophyll per hour). l4CO2 fixation was inhibited to thedark level by 3(3,4,dichlorophenyl)-1, 1-dimethylurea (at 0-6µM or 10µM) and by the uncoupler carbonyl cyanide-m-chlorophenylhydrazone(SµM). The presence of imidazole or ammonium sulphate(both of which uncouple ATP production in vitro) did not resultin an inhibition of 14CO2 fixation. These results are discussedin relation to published work on solute uptake by Nitella translucens.During photosynthesis there was rapid movement of 14C-labelledorganic compounds out of the chloroplasts. 14C-labelled sucrose,ammo-acids, and sugar phosphates were found in samples of vacuolarsap.  相似文献

Diurnal series of fluorescence and photosynthesis assays wereconducted in high altitude (3803 m), tropical (16°), LakeTiticaca (Peru/Bolivia). Near-surface diurnal thermoclines formedon typical days of high photon flux density (PFD, {small tilde}2000 µE m–2 s–1). In the depth range of diurnalstratification profiles of in vivo fluorescence, both without(Fa and with (Fb DCMU, exhibited a mean decrease of 64% frommorning to mid-day, but little change (mean increase of 1.5%)through the afternoon. Three times during the day surface, mid-depth(3–5 m) and deep (15–20 m) phytoplankton sampleswere incubated with H14CO3 under short (<2 h) exposuresto a range of in situ PFDs. Comparison of phytoplankton in differentsamples (ANOVA) showed identical photosynthetic response insunrise (isothermal) samples but a significant drop in surfaceand mid-depth photosynthesis at all PFDs during times of diurnalstratification. Similarly, both low-light () and light-saturated(P2 max photosynthetic parameters were lower in mid-day surfacesamples compared to deep samples. In addition, previously photoinhibitedsamples had a higher threshold intensity for photoinhibition,IT. These results, together with diurnal time series of fluorescencefrom in situ incubations, demonstrate that recovery from extendedepisodes of photoinhibition during diurnal stratification isslower than suggested by previous observations in vitro. Photosynthesisby near-surface phytoplankton is different in light increasingup to IT than light decreasing from IT. Algae (mainly Euglena sp.) from a high rate oxidation pond (HROP)were used for studying the afternoon depression in primary productivity.The phenomenon was observed on the same date by laboratory measurementsof photosynthesis and respiration (oxygen evolution method)as well as by in situ determinations of 14C incorporation. Thefollowing values of were calculated: morning, 0.014 µmolO2/mg Chi a/min//mol quanta.m2.s; afternoon, 0.008. Assuminga constant kc of 0.006 m2/mg Chi a we found the quantum requirement(–1) in the morning sample to be considerably lower thanin the afternoon sample (surface: morning 44 mol quanta/molO2; afternoon, 71). Besides this reduction in photosyntheticefficiency the afternoon sample also exhibited reduced lightsaturated photosynthetic rates (Pmax) and enhanced dark respirationrates. The combination of these three effects led to considerablylower areal primary productivity in the HROP in the afternoon.We suggest that this phenomenon is brought about by carbon limitationand cell overloading by photosynthetic products.  相似文献

In "air-grown" Chroomonas sp. cells, low concentrations of DCMU(less than 0.1 µM) could prevent the inhibition of 14CO2fixation by anaerobiosis under light-saturating conditions (morethan 40 W.m–2), with phenazine methosulfate showing asimilar effect. Antimycin A, carbonyl cyanide m-chlorophenylhydrazone(CCCP), and N,N'-dicyclohexylcarbodiimide strongly inhibitedanaerobic photosynthesis at concentrations which did not significantlyinhibit the rate under 2% O2 at high light intensity (200 W.m–2),although 0.2 µM CCCP stimulated the rate under 2% O2 tosome extent. On the other hand, KCN inhibited the rate muchmore strongly under 2% O2 than N2, although it inhibited therate very strongly at concentrations above 5 µM both underN2 and 2% O2. These results suggest that the inhibition of photosynthetic14CO2 fixation by anaerobiosis in this alga result from ATPdeficiency caused by over-reduction of electron carriers ofthe cyclic electron flow and that oxygen can prevent the over-reduction.Cyclic electron flow seems to be necessary to provide additionalATP for CO2 reduction under anaerobic conditions, although itseems to be less necessary under aerobic conditions. In Daucus carota cells cultivated in vitro, the ammonium ionstimulates the incorporation of radioactivity from labelledglucose and labelled pyruvate into CO2 and into the residueinsoluble in 60 per cent (v/v) ethanol. There is a higher 14CO2production from [6-14C2] glucose than from [6-14C] glucose.These results suggest a possible stimulation of glycolysis bythe ammonium ion.  相似文献

A launch, suspension and recovery technique for oceanic, insitu, 14C productivity measurements is described. Ship's timefor deployment of 125 ml incubation bottles at several depthsdown to 100 m is approximately 10 minutes but another half houris required for water sampling and laboratory manipulations.Relocation from up to 9 nautical miles (16.7 km) is by inexpensiveVHF telemetry transmitters and recovery is by grappling fromthe ship. The method was tested in the Scotia Sea, The Antarctic,during the cruise John Biscoe 2 (1980). It was found feasibleto launch experiments in winds of up to 30 knots (15.4 m s–1)and recover in a wind of 40 knots (20.6 m s) on occasion.Eleven experiments were performed in 15 days.  相似文献

Histiobalantium sp. was found regularly in the pelagic zoneof Lake Constance, FRG, over five annual cycles. Maxima of upto 6400 cells l–1 were recorded in late summer, with similarnumbers in the 0–8 and 8–20 m depth intervals. Onan annual average, the population accounted for 10–17%of the total biomass of planktonic ciliates. In the laboratory,Histiobalantium grew well on a diet of the cryptophyte Rhodomonassp. Maximum growth rates obtained in batch cultures were 0.21and 0.33 day–11 at 9 and 18°C, respectively. In situexperiments using diffusion chambers yielded positive growthrates in autumn and winter. The highest values recorded at theambient temperatures 5, 14 and 17°C were 0.17, 0.32 and0.40 day–1, respectively. Surfacing and water-leaving behaviour (quitting) of aquaticpulmonate snails was investigated in the laboratory. Regressioncoefficients of surfacing rate per hour for 16 snails in 200cm3 of water were 1.28 for L. peregra, 1.35 for B. glabrata(albino form), 0.47 for B. glabrata (pigmented form) and 0.17for B. jousseaumei. Disturbance, snail size and conditioningof the water were relatively unimportant factors; water volumeadjusted by depth was also unimportant up to 50 mm depth. Importantfactors controlling surfacing appeared to be population density,time of year, temperature, water volume adjusted by perimeterand dissolved oxygen concentration. The response to the latterwas not clear and both B. glabrata and L. peregra orientatedalmost as well to a nitrogen/water interface as to an air/waterinterface. Water-quitting regressed on volume, which had beenadjusted by perimeter variation, revealed a curvilinear relationshipfor B. glabrata, with maximum quitting at 1 snail per 7cm3 (1snail per 14cm of climbable perimeter). L. peregra showed anegative exponential curve, with maximum quitting at 1 snailper 3 cm3 of water (1 snail per 8 cm of climbable perimeter).These data could be relevant to design and management of irrigationchannels where bilharzia might occur.
